通过体外NBT光化还原系统证明,乙酰水杨酸(ASA) 有直接清除O-·2 的作用。ASA 浓度大于150 mg·L- 1 时可完全清除体系内的O-·2 。水分亏缺下,0 .05 % ASA 明显降低小麦幼苗中O-·2 的含量。ASA 既有直接清除O-·2 的作用,又可通过提高SOD 活性来清除O-·2 。  相似文献

应用脱氧核糖降解法研究了CuZn-SOD对几种·OH产生系统的作用机理.结果证明:SOD对Fe(3+)·O·H2O2系统中·OH的产生有明显的抑制作用,而失活SOD或BSA对它的抑制作用不大;在Fe(2+)·H2O2和CU(2+)·H2O2系统中,SOD、失活SOD和BAS均能抑制·OH的产生;在Fe(2+)·O系统中,SOD对·OH产生作用不大,而失活SOD或BSA对它有明显的抑制作用.由此推测SOD对·OH形成可能有三方面的影响:1.对O的清除作用,阻断Haber-Weiss反应;2.对金属离子的络合作用,降低·OH的产额;3.促进H2O2的积累,加快Fenton反应.  相似文献

用四氮唑蓝光化学还原法对所合成的KCu(IDA)(Ser)·2H2O、KCu(IDA)(Ala)·H2O、Cu(IDA)(en)、KCu(IDA)(Gly)·H2O和Cu(IDA)·2H2O(IDA=N(羧基甲基)-甘氨酸,Ser=丝氨酸,Ala=丙氨酸,en=乙二胺,Gly=甘氨酸)等5种氨基酸─铜(Ⅱ)配合物进行了活性测定,发现它们均具有天然超氧化物歧化酶活性,其活性依次为0.34、0.45、0.50、0.54、0.72Cuμmol·L-1。  相似文献

应用脱氧核糖降解法研究了离体条件一Cu,Zn-SOD与H2O2反应产生.OH,并对其机理进行了探讨。H2O2可使Cu,Zn-SOD失活,在失活过程中有.OH产生。甲酸钠和苯甲酸钠均能不同程度地保护Cu,Zn-SOD和降低H2O2与CuZn-SOD反应中。OH的产额,热失活SOD也可和H2O2反应生成OH,且效能高于活性Cu,Zn-SOD;  相似文献

关于血清 Cu,Zn及 CuZn-SOD 与恶性肿瘤关系的探讨   总被引:4,自引:0,他引:4  
本文用原子吸收光度法,Oyangui法及AdachiY法分别测定了正常人与五种肿瘤患者血清Cu,Zn含量,铜锌超氧化物歧化酶(CuZn-SOD)的活性以及谷胱甘肽硫转移酶(GST)的活性。结果表明:恶性肿瘤患者血清中铜含量升高,锌含量降低,CuZn-SOD活性降低,而GST活性在正常人与肿瘤患者间无显著差异(P>0.05)。提示人血清中高铜,低锌,高铜/锌比值,以及CuZn-SOD活性降低与恶性肿瘤的发生有关。  相似文献

研究了生长在正常大气CO2和CO2倍增环境中的盐生植物碱蓬(Suaedasalsa)幼苗呼吸酶活性对KCl和NaCl的反应.结果表明,在CO2倍增(700μl·L-1)和正常大气CO2(350μl·L-1)下,300mmol·L-1KCl和NaCl均能抑制琥珀酸脱氢酶(SDH)和苹果酸脱氢酶(MDH)活性,而异柠檬酸脱氢酶(IDH)活性为NaCl抑制、KCl促进;NaCl和KCl明显抑制细胞色素氧化酶(CO)和光呼吸中乙醇酸氧化酶(GO)、羟基丙酮酸还原酶(HPR)活性;并指出在KCl胁迫下,CO2使三羧酸循环(TCAC)的运行变慢,NaCl胁迫下使其加快,TCAC运行限速步骤与MDH无关,CO为盐对呼吸代谢影响的重要位点.另外,K+、Na+对蛋白表达的影响有差异,CO2可使盐胁迫下的碱蓬幼苗蛋白表达降低.  相似文献

氧化剂、还原剂处理前后,L-SOD的活性及紫外光谱发生变化,H2O2使Fe(Ⅲ)吸收增强,同时钝化L-SOD的活性;加入保险粉后,L-SOD重新活化,Fe(Ⅲ)吸收减弱.NEM封闭Cys后,L-SOD紫外吸收谱发生变化,且活性减弱.说明Fe辅基及Cys是活性发挥的必需基团.  相似文献

电离辐射能诱发超氧阴离子自由基(O-2·)的产生,同时也是细胞凋亡发生的诱因之一。锰超氧化物歧化酶(MnSOD)是一种存在于真核细胞线粒体、具有清除O-2·作用的抗氧化酶。为探讨MnSOD在辐射诱发细胞凋亡中的作用,采用基因转染技术分别将有义和反义MnSODcDNA真核表达载体直接导入CHO细胞,并用此细胞模型研究了MnSOD对X射线诱发细胞凋亡的调控。结果发现,稳定表达有义MnSOD的细胞克隆能明显抑制8GyX射线诱发的细胞凋亡,而表达反义MnSOD的细胞克隆对X射线诱发的细胞凋亡更加敏感,进一步的研究发现,线粒体膜电位的改变可能是MnSOD调控细胞凋亡的机制之一。  相似文献

用苯甲基磺酰氟(PMSF)和H_2Se相继处理铜锌超氧化物岐化酶(Cu,Zn-SOD),将酶分子中的丝氨酸(Ser)转化为硒代半胱氨酸(SeCys),从而引入了谷胱甘肽过氧化物酶(GPX)的催化基团,使其在SOD酶活性大部分保留的情况下,具有GPX活性,其GPX活力是PZ51活力的30倍。研究了双功能酶的最佳制备条件,包括PMSF的剂量、反应最适温度及H_2Se处理时间等,并用电子能谱、DTNB等方法测定了双功能酶的硒含量;测定了双功能酶对不同底物的米氏常数及双功能酶的荧光光谱、紫外吸收光谱及稳定性。  相似文献

Potentiometric and spectroscopic (UV-Vis, CD and EPR) studies were carried out on copper(II) complexes with chicken prion protein N-terminal fragments, Ac-(PHNPGY)4-NH2, and the mutated residue, Ac-(PHNPGF)4-NH2, to assess the role of tyrosine in the copper coordination. Both thermodynamic and spectroscopic results indicate that chicken prion fragments are not able to bind more than two copper ions and only with the involvement of side chain tyrosine groups. The prevailing complex shows one copper ion bound to four imidazole nitrogen atoms in the 1:1 metal to ligand ratio systems. The superoxide dismutase (SOD)-like activity of copper(II) complexes with the avian peptides and mammal analogue, Ac-(PHGGGWGQ)4-NH2, was also investigated by means of Pulse radiolysis. The copper(II) complexes with avian peptides do not display SOD-like activity, while very low activity has been detected for the copper(II) complexes with mammalian tetraoctarepeat.  相似文献

AIMS: Selection of suitable surfactants for enhancing and stabilizing alpha-amylase of Geobacillus thermoleovorans. METHODS AND RESULTS: Geobacillus thermoleovorans was cultivated in shake flasks containing 50 ml of starch-yeast extract-tryptone (SYT) medium with/without surfactants. Titres of the enzyme in media were monitored. The enzyme was also preserved at 4 degrees C with/without surfactants and enzyme activities were determined. Among polyethylene glycol (PEGs) of different molecular weights, PEG 8000 (0.5%, w/v) caused a slight increase in the enzyme titre, while Tween-20, Tween-40 and Tween-60 (0.03%, w/v) exerted a significant stimulatory effect on enzyme secretion. In the presence of SDS, Tween-80 and cholic acid (0.03%, w/v), the enzyme production was nearly twofold higher than that in the control. The anionic (SDS, cholic acid) and non-ionic (Tweens) detergents increased the cell membrane permeability, and thus, enhanced alpha-amylase secretion. Furthermore, anionic surfactants exhibited stabilizing effect on the enzyme during preservation at 4 degrees C. CONCLUSIONS: PEG 8000 and the ionic detergents (SDS, cholic acid and Tween-80) were more effective in the solubilization of cell membrane components, and enhancing enzyme yields than the cationic detergents such as CTAB (N,Cetyl-N,N,N-trimethyl ammonium bromide). Further, these surfactants were found to stabilize the enzyme at 4 degrees C. SIGNIFICANCE AND IMPACT OF THE STUDY: The secretion of Ca2+-independent hyperthermostable alpha-amylase was enhanced in the presence of certain anionic and non-ionic detergents in the medium. Furthermore, the surfactants stabilized the enzyme during preservation at 4 degrees C. The use of this enzyme in starch hydrolysis eliminates the addition of Ca2+ in starch liquefaction and its subsequent removal by ion exchange from sugar syrups.  相似文献

旨在探究表面活性剂对菌株降解溴氰菊酯(Deltamethrin,DM)的影响。以前期筛选到具备DM较高降解能力的Acinetobacter junii LH-1-1为目标菌株,通过荧光光谱法测定了3种不同类型表面活性剂(CTAB、AES和Tween-20)的临界胶束浓度(Critical Micelle Concentration,CMC),并探究不同表面活性剂各CMC对A.junii LH-1-1生长、DM增溶作用和菌株降解DM的影响。结果显示,测得CTAB、AES、Tween-20的CMC分别为0.793 mmol/L、0.547 mmol/L和0.031 mmol/L;CTAB对菌株的生长无明显影响,AES对菌株生长有抑制作用,Tween-20对菌株的生长有一定的促进作用;CTAB、AES和Tween-20对DM均具有较强的增溶作用;且CTAB可显著提高A.junii LH-1-1对DM的降解率,当其含量为2 CMC,72 h时DM降解率达到98.20%,较未添加表面活性剂的对照组(阳性)降解率提高了24.31%。阳离子表面活性剂CTAB可显著提高A.junii LH-1-1对DM的降解。  相似文献

The prion protein is a membrane attached glycoprotein that is involved in binding of divalent copper ions. In vivo human and chicken PrPs exhibit SOD-like activity associated with octarepeat and hexarepeat regions, respectively, when bind Cu(II) ions. However, the species of Cu(II)-PrP involved in the Cu(II) center which determines the highest SOD-like activity is still unknown. The data presented here clearly show that the single Cu(II) ion bound to PrP octapeptide repeat region of mammalian prion and hexapeptide repeat region of avian prion via 4 His side-chain imidazoles reveals the highest SOD activity.  相似文献

The N-terminal region of the prion protein PrP(C) contains a series of octapeptide repeats. This region has been implicated in the binding of divalent metal ions, particularly copper. PrP(C) has been suggested to be involved in copper transport and metabolism and in cell defense mechanisms against oxidative insult, possibly through the regulation of the intracellular CuZn superoxide dismutase activity (CuZn-SOD) or a SOD-like activity of PrP(C) itself. However, up to now the link between PrP(C) expression and copper metabolism or SOD activity has still to be formally established; particularly because conflicting results have been obtained in vivo. In this study, we report a link between PrP(C), copper binding, and resistance to oxidative stress. Radioactive copper ((64)Cu) was used at a physiological concentration to demonstrate that binding of copper to the outer plasma cell membrane is related to the level of PrP(C) expression in a cell line expressing a doxycycline-inducible murine PrP(C) gene. Cellular PIPLC pretreatment indicated that PrP(C) was not involved in copper delivery at physiological concentrations. We also demonstrated that murine PrP(C) expression increases several antioxidant enzyme activities and glutathione levels. Prion protein may be a stress sensor sensitive to copper and able to initiate, following copper binding, a signal transduction process acting on the antioxidant systems to improve cell defenses.  相似文献

应用平衡振荡法,研究了阴、阳和非离子表面活性剂在土壤上的吸附.结果表明,阳离子表面活性剂十六烷基三甲基溴化铵能强列吸附在6种不同性质的土壤上,吸附等温线为L型,分配常数Kd,为3.0×10^2~48×10^2L·kg^-1;阴离子表面活性剂十二烷基苯磺酸钠、非离子表面活性剂OP及Tween-20的吸附等温线随土壤类型不同而不同,有L、S等型,吸附强度远弱于阳离子表面活性剂,Kd分别大体处于5.3~39、0.13~0.44(Tween-20)和4.4~22.4L·kg^-1(OP).阳离子表面活性剂的土壤最大吸附量与土壤阳离子交换容量呈线性相关.低浓度范围内,阴离子表面活性剂的土壤分配常数与土壤粘粒含量呈正相关.同时土壤颗粒表面的电荷特性也影响吸附.非离子表面活性剂的Kd与土壤粘粒、砂粒、粉沙含量及表面积存在经验函数关系.  相似文献

Tannic acid (TA) has well-described antimutagenic and antioxidant activities. The antioxidant activity of TA has been previously attributed to its capacity to form a complex with iron ions, interfering with the Fenton reaction [Biochim. Biophys. Acta 1472, 1999, 142]. In this work, we observed that TA inhibits, in the micromolar range, in vitro Cu(II) plus ascorbate-mediated hydroxyl radical (*OH) formation (determined as 2-deoxyribose degradation) and oxygen uptake, as well as copper-mediated ascorbate oxidation and ascorbate radical formation (quantified in EPR studies). The effect of TA against 2-deoxyribose degradation was three orders of magnitude higher than classic *OH scavengers, but was similar to several other metal chelators. Moreover, the inhibitory effectiveness of TA, by the four techniques used herein, was inversely proportional to the Cu(II) concentration in the media. These results and the observation of copper-induced changes in the UV spectra of TA are indications that the antioxidant activity of TA relates to its copper chelating ability. Thus, copper ions complexed to TA are less capable of inducing ascorbate oxidation, inhibiting the sequence of reactions that lead to 2-deoxyribose degradation. On the other hand, the efficiency of TA against 2-deoxyribose degradation declined considerably with increasing concentrations of the *OH detector molecule, 2-deoxyribose, suggesting that the copper-TA complex also possesses an *OH trapping activity.  相似文献

Superoxide dismutase (SOD) activity in rat liver cytosol and submitochondrial fractions was characterized as enzymatic and nonenzymatic (due to the SOD-like activity of copper) by four approaches: (i) aerobic NBT2+ (nitroblue tetrazolium) photoreduction in the absence of EDTA; (ii) aerobic NBT2+ photoreduction in the presence of 10?4m EDTA; (iii) anaerobic NBT2+ photoreduction; and (iv) o-dianisidine photooxidation. Under normal conditions nonenzymatic SOD activity has been observed only in the intermembrane space. The single subcutaneous injection of rats with CuSO4 solution (5 mg Cu/kg body wt) led to (i) an elevation of the copper level in all submitochondrial fractions; (ii) an increase in enzymatic SOD activity in only cytosol and intermembrane spaces; (iii) the appearance of a new electrophoretic SOD activity band in the intermembrane space preparations; and (iv) the appearance of nonenzymatic SOD-like activity in the outer and inner mitochondrial membranes, and a twofold increase in lipid hydroperoxides. This suggests that the increased nonenzymatic copper in vivo has a prooxidant effect, and does not catalyze the dismutation of O2? as it has been shown in in vitro experiments [E. M. Russanov S. G. Ljutakova, and S. I. Leutcher (1982) Arch. Biochem. Biophys.215, 220–229]. The peculiarities of the SOD activity in the intermembrane space are explained by the lysosomal localization of the granular CuZnSOD.  相似文献

Oxygen free radicals are the final or intermediate products of many metabolic reactions. Of greatest significance to the organism are superoxide anion radical (O2-), hydrogen peroxide (H2O2), hydroxyl radical (OH). singlet oxygen (lO2) etc. A proper ratio between both production and breakdown of oxy-radicals is essential for the maintenance of a dynamic equilibrium of vital processes. The superoxide dismutases protect cells against toxic influence of the superoxide. In addition, some square-pyramidally pentacoordinated copper(II) complexes, derived from tridentate Schiff bases of the N-salicylideneaminoal-canoate type, show remarkable SOD-like activity. A selected set of complexes of this type have been tested: potassium [aqua-(N-salicylideneglutamato) cuprate] (L-and D,L-form). potassium [(isothiocyanato)-(N-salicylideneglycinato) cuprate], potassium [(isothiocyanato)-(N-salicylidene-D,L-alaninato) cuprate]. potassium ((isothiocyanato)-(N-salicylidene-β-alaninato) cuprate] and potassium [(isocyanate)-(N-salicylideneglycinato) cuprate]. Our results suggest that the copper complexes are not only antioxidants, but may also possess anti-inflammatory, cytostatic and radioprotective properties.  相似文献
