Stevioside is a natural sweetener extracted from leaves of Stevia rebaudiana (Bertoni) Bertoni. The literature about Stevia, the occurrence of its sweeteners, their biosynthetic pathway and toxicological aspects are discussed. Injection experiments or perfusion experiments of organs are considered as not relevant for the use of Stevia or stevioside as food, and therefore these studies are not included in this review. The metabolism of stevioside is discussed in relation with the possible formation of steviol. Different mutagenicity studies as well as studies on carcinogenicity are discussed. Acute and subacute toxicity studies revealed a very low toxicity of Stevia and stevioside. Fertility and teratogenicity studies are discussed as well as the effects on the bio-availability of other nutrients in the diet. The conclusion is that Stevia and stevioside are safe when used as a sweetener. It is suited for both diabetics, and PKU patients, as well as for obese persons intending to lose weight by avoiding sugar supplements in the diet. No allergic reactions to it seem to exist.  相似文献

Metalworking fluids (MWFs) are complex mixtures of chemicals and are indispensable materials in industry. They are used as cooling and lubricating agents in different machining process such as grinding, milling, and cutting. The quality of MWFs is affected by physical, chemical, and microbial contaminates. In particular, MWFs are highly vulnerable to microbial contamination, which may act both as potential pathogens and deteriorgens. Microbial contamination is of major concern due to potential health hazards such as skin dermatitis and hypersensitivity pneumonitis. The contaminated MWFs can exhibit high degrees of microbial loading, ranging from 10(4) to 10(10) colony-forming units (CFU)/ml. Wide varieties of microorganisms are reported to colonize MWFs. Traditional culturing techniques are not only laborious and time consuming but also underestimate the actual distribution of the microorganisms present in the contaminated MWFs. Therefore, rapid molecular methods such as real-time PCR and fluorescent in situ hybridization are implemented to monitor the microbial load. In industry, biocides are presently used to control microbial contamination. However, it has its own disadvantages and therefore, in recent years, alternative methods such as UV irradiation were evaluated to reduce microbial contamination in MWFs. Microbes inhabiting the MWF are also capable of forming biofilm which is detrimental to the MWF system. Biofilm is resistant to common disinfectant methods, and thus further research and development is required to effectively control its formation within MWF systems. This review is intended to discuss the overall microbiological aspects of MWF.  相似文献

Currently, the mainly used characteristics to predict outcome or treatment response in patients with breast cancer are tumor size, N-status, histological grade and receptor status (ER/PgR). However, these conventional clinico-pathological characteristics are of limited value. More accurate determinators are needed to select patients who are most likely to benefit from treatment in terms of prognosis as well as treatment response. Proliferation and apoptosis are assumed to play a key role in tumor progression as well as response to treatment. Currently, an increasing number of molecular factors controlling apoptosis as well as proliferation is known. The clinical relevance of apoptotic tumor markers in the treatment strategy of patients with breast cancer is the subject of this review. One of the Golgi's main functions is the glycosylation of secreted proteins. A large variety of glycan chains can be synthesized in the Golgi, and it is increasingly clear that these are critical in basic cellular functions as well as the development of multicellular organisms. The structurally best-documented glycans are N-glycans, yet these are also the most enigmatic in their function. In contrast, O-glycan function is far better understood, but here the structures and biosynthetic pathways are very incomplete. The critical importance of glycans is highlighted by the broad spectrum of diseases they are associated with, such as a number of inherited diseases, but also cancers or diabetes. The molecular clues to these, however, are only just being elucidated. Although some glycan structures are known to be involved in signaling or adhesion to the extracellular matrix, for most the functions are not yet known. This review aims at summarizing current knowledge as much as to point out critical areas key for future progress.  相似文献

Two species of Echinochloa are grown as cereals. Echinochloa crusgalli is native to temperate Eurasia and was domesticated in Japan some 4,000 yr ago. Echinochloa colona is widely distributed in the tropics and subtropics of the Old World. It was domesticated in India. Echinochloa colona is morphologically allied to E. crusgalli, but hybrids between them are sterile. Echinochloa colona differs consistently from E. crusgalli in having smaller spikelets with membran-aceous rather than chartaceous glumes. Hybrids between wild and cultivated taxa of E. colona and between those of E. crusgalli are fertile. Cultivated E. colona is variable. It is grown as a cereal across India, Kashmir and Sikkim. Four morphological races are recognized, although these do not have geographical, ecological or ethnological unity. Race laxa is confined to Sikkim where races robusta, intermedia and stolonifera are also grown. In India, races robusta, intermedia and stolonifera are often grown as mixtures, and Echinochloa is sometimes grown as a mixture with other cereals, particularly Setaria italica (foxtail millet) or Eleusine coracana (finger millet). The species is planted on poor soil, and some cultivars mature in less than 2 mo. They hold considerable promise as cereals for the semiarid tropics.  相似文献

Although optical resolution of alpha-methylphenylalanine (alpha-Me-Phe) has been achieved by the action of carboxypeptidase A on the N-trifluoroacetyl derivative of the amino acid (TFA-alpha-Me-Phe), it is improbable that an alpha-methyl substrate could bind in the same orientation as glycyl-L-tyrosine, due to steric interaction of the alpha-methyl group with an atom in the imidazole ring of zinc ligand His-196. The kinetic parameters for TFA-alpha-Me-Phe and for an ester substrate bearing an alpha-methyl group (beta-hippuryl-alpha-methylphenyllactic acid, HMPL) have been determined and compared to those for the appropriate nonmethylated control substrates. Both TFA-alpha-Me-Phe and HMPL appear to be bound nearly as well as are their respective controls, and HMPL is hydrolyzed nearly as rapidly as its control. TFA-alpha-Me-Phe, however, is hydrolyzed only about one-fiftieth as rapidly as is the nonmethylated substrate. These findings are consistent with the possibilities that: (1) the proposed substrate-induced conformational shift of Tyr-248 is hindered when the methylated substrates are bound; (2) the orientation in which alpha-methyl substrates are bound precludes optimal positioning of Tyr-248 and the scissile bond even after the rotation of Tyr-248 has occurred; (3) amides and esters are bound in different orientations, and in the amide orientation an alpha-methyl group is so directed as to interfere with the approach of Glu-270 to the scissile bond.  相似文献

An association of contemporaneous Jurassic mega- and miospores with reworked Carboniferous mega- and miospores is described from the Lower Lias rocks of northeast Scotland. The assemblage is compared with other accounts of reworked spores in Jurassic sediments. It is suggested that such reworking was particularly common in the Lower Jurassic of northern Europe and that four situations exist in the literature concerning probable cases of reworking: (1) the spores are recognised as reworked; (2) the identity of the spores as Carboniferous species is recognised and the “range” of these species is extended into the Jurassic; (3) the reworked spores are not recognised as such, and are incorrectly assigned to Jurassic genera and species; (4) the spores are recognised as Carboniferous, but their origin is attributed to contamination rather than reworking. In conclusion, the contrasting preservational features exhibited by the reworking and contemporaneous megaspores are discussed and interpreted as the result of differing depositional and postdepositional histories.  相似文献

Synopsis This review is devoted mainly to an evaluation of the status of microscopical cytochemistry seen as a discipline aiming at both the localization and the quantification of molecular processes in cells. Its relationships to ultramicrochemistry, as well as, in a broader sense, to biochemistry and cell biology, are discussed from both the historical and the methodological points of view. Recent developments in quantitative cytophysical techniques, such as automated cytophotometry using microscopes fitted with flying spot systems, TV cameras, or scanning stages, and the development of rapid flow cytometers are discussed. Analytical electron microscopy is touched upon too.The main part of the review is devoted to recent trends that strengthen the analytical basis of cytochemical staining methods. The special character of staining procedures as a kind of matrix chemistry is discussed and the potentialities of the use of matrixincorporated compounds for the fundamental study and calibration of microscopical staining procedures are elaborated. Parallel developments in the theory and practice of matrix chemistry in biochemistry are stressed. Growing interrelations between microscopical cytochemistry and related fields of investigation, such as the controlled fragmentation of cells, and methods like ultramicroanalysis of individual cells are indicated.  相似文献

Geometric models of vegetation (conceptual spaces) are reviewed. Spaces with samples or species as axes are termed flortistic spaces, as opposed to ecological space with environmental gradients as axes. The terms floristic and ecological relationships are defined as relationships in floristic and ecological spaces, respectively. Compositional turnover is pointed out as the essence of ecological gradients, and arguments in favour of measuring ecological distance in units of compositional turnover are given. The most important criteria for evaluation of ecological distance measures are considered to be linear response to separation along ecological gradients and robustness. Theoretical disadvantages of measures of floristic relationships used as ecological distance measures are discussed. A new measure of ecological distance, separation along a DCA ordination axis, is proposed. This measure and four measures of floristic relationships were tested on four simulated coenoclines (high and low beta diversity, high and low noise) using four weighting functions. The new measure was generally superior, particularly with noisy data. The distance measures generally performed best with intermediate weighting of a percentage cover scale. Application of DCA to calculation of ecological distances in multi-gradient systems is briefly discussed. The potential of DCA for rescaling of ecological gradients is emphasized, and some possible applications of rescaling are suggested.  相似文献

Species’ distributions are dynamic and are linked to the changing physical environment. Temperature is considered to be a major factor influencing biogeography, especially in ectotherms such as the family Lithodidae. Lithodids are rare amongst decapods in their ability to inhabit the higher latitudes of the Southern Ocean; however, they are usually found in locations where water temperature is above 0.5°C. This study, for the first time, provides a baseline indication of the limits of the lithodid distribution around Antarctica, which will be instrumental in any future work on range extensions in this group. The distribution of lithodids is likely to change as temperatures along the West Antarctic Peninsula continue to rise, and range extensions by durophagous predators, such as the lithodids, are regarded as a potential threat to the unique structure of Antarctic continental-shelf ecosystems.  相似文献

Glucosidic bonds from different non-soluble polysaccharides such as starch, cellulose and xylan are hydrolyzed by amylases, cellulases and xylanases, respectively. These enzymes are produced by microorganisms. They have a modular structure that is composed of a catalytic domain and at least one non-catalytic domain that is involved in polysaccharide binding. Starch-binding modules are present in microbial enzymes that are involved in starch metabolism; these are classified into several different families on the basis of their amino acid sequence similarities. Such binding domains promote attachment to the substrate and increase its concentration at the active site of the enzyme, which allows microorganisms to degrade non-soluble starch. Fold similarities are better conserved than sequences; nevertheless, it is possible to notice two evolutionary clusters of microbial starch-binding domains. These domains have enormous potential as tags for protein immobilization, as well as for the tailoring of enzymes that play a part in polysaccharide metabolism.  相似文献

Tracer clearance curves are conventionally extrapolated beyond times of observation by using monoexponential asymptotic forms. The inadequacy of the resulting predictions, especially as to the mean transit time and quantities derived from it, has been previously demonstrated experimentally. Here improvements in extrapolations and in the resulting predictions are derived theoretically and tested on previously published data, venous as well as externally recorded. First, secure lower bounds on the mean transit times are constructed, and shown to be much higher than conventional outright estimates for venous data (twice as high in some cases). Next, new asymptotic forms of tracer clearance curves from kinetically heterogeneous systems are derived; they are not monoexponential, but they are as robust, contain as few parameters and are as easily connected to data. It is shown theorectically that for real organs these new asymptotic forms should extrapolate and predict better than monoexponentials, and this is demonstrated on previously published venous data from perfused muscle. In particular, the resulting outright predictions of mean transit times are substantially better than the best lower bounds. Furthermore, a correction is derived to the standard estimate of the rate of regional cerebral blood flow. In an application to previously published data recorded externally, that correction reduces the estimated flow rate by 4%.  相似文献

One of the major aims of bioprocess engineering is the real-time monitoring of important process variables. This is the basis of precise process control and is essential for high productivity as well as the exact documentation of the overall production process. Infrared spectroscopy is a powerful analytical technique to analyze a wide variety of organic compounds. Thus, infrared sensors are ideal instruments for bioprocess monitoring. The sensors are non-invasive, have no time delay due to sensor response times, and have no influence on the bioprocess itself. No sampling is necessary, and several components can be analyzed simultaneously. In general, the direct monitoring of substrates, products, metabolites, as well as the biomass itself is possible. In this review article, insights are provided into the different applications of infrared spectroscopy for bioprocess monitoring and the complex data interpretation. Different analytical techniques are presented as well as example applications in different areas.  相似文献

Clematis sect. Atragene is revised in this paper based on the examination of a large number of herbarium specimens, extensive field observations, and morphometric analyses. Brief taxonomic history and geographical distribution of the section are given, the relationships among the species are discussed, and the evolutionary trends of some characters in the section are evaluated. The staminodes of the plants in this section may have evolved from the outer stamens with petaloid filaments and gradually disappearing anthers. Subsequently, they may have evolved in two different ways. One possibility is that the staminodes elongate and become lanceolate, as long as sepals, and their apices turn into attenuate. The other is that the staminodes are spathulate, but not elongating, as long as stamens, and their apices turn into retuse from obtuse and rounded. The evolutionary trend of sepals may be from thin to thick in texture, and the veins from non-prominent to prominent. As a result, five new series are established and nine species, two subspecies and nine varieties (including three new ranks) are recognized in this section. An identification key is provided, and each taxon is described and illustrated. Clematis sibirica and Clematis ochotensis are treated as subspecies of Clematis alpina due to their subtle differences and lack of, or few, overlapping distributions. Clematis fusijamana and Clematisfauriei are recognized as varieties of C. alpina ssp. ochotensis for the continuous variation of the velutinous strips on the sepal margins. Clematis iliensis is treated as variety of C. alpina ssp. sibirica for the continuous variation of leaf division types. Extensive variations in sepal color and basal caruncle size support degrading Clematis chiisanensis as a variety of Clematis koreana. The North American ser. Occientales may be primitive, whereas ser. Macropetalae may be the most advanced taxon in this section. Ser. Alpinae and ser. Koreanae are closely related to each other. The isoelectric point of normal red cells cannot be measured but is certainly lower than that of any plasma protein. Red cells are easily damaged so that they will adsorb proteins from low concentrations. Normal red cells do not adsorb protein even from concentrated solutions, as is evidenced by the finding that the ratio of the mobility of the cells to that of the proteins themselves is at least as high in concentrated casein, albumin, gelatin, or fibrinogen solutions as in dilute. The finding that the observed mobility of red cells is unchanged or only slightly decreased when bulk viscosity is increased by added protein is interpreted as indicating that the red cell surfaces are hydrated. The aggregating effect of certain proteins has been determined and is assumed to be due to their dehydrating effect on the cells. Some types of cells, as beef, are not aggregated, presumably because they are resistant to this dehydrating effect. The difference in the behavior of different types of red cells demonstrates the importance of the nature of the cell as well as of the medium in determining the rate of aggregation and therefore of sedimentation.  相似文献

Multiple synaptonemal complexes (polycomplexes) (PC) are similar in structure to synaptonemal complexes (SC) and are also highly conserved through evolution. They have been described in over 70 organisms throughout all life forms. The appearance of PCs are restricted to meiotic and germ-line derived tissues and are most commonly present after SC formation. However, in a number of animals and plants, both extra- and intranuclear PCs are present during premeiotic and pre-pachytene stages. The structure and biochemical composition of PCs is similar to SCs that the basic unit is tripartite, consisting of two lateral elements and a central region (in which transverse elements are located), and the dimensions of such structures are equivalent. Stacking of SC subunits, while still maintaining equivalent SC dimensions, creates a problem since the lateral elements (LE) would then be twice as thick in the PC as compared to the SC. Recently, it has been shown that the LE of the SC is actually multistranded, thus the LE of each subunit of the PC is half as thick as its counterpart in the SC.  相似文献

The genus Agapornis consists of nine small African parrot species that are globally well known as pets, but are also found in their native habitat. Illegal trapping, poaching and habitat destruction are the main threats these birds face in the wild. In aviculture, Agapornis breeding is highly popular all across the globe. Birds are mainly selected based on their plumage colour variations but very little molecular research has been conducted on this topic. There are 30 known colour variations amongst the nine species and most of these are inherited as Mendelian traits. However, to date none of the genes or polymorphisms linked to these variations have been identified or verified. Due to unethical breeding practices, the need for the development of molecular tests such as identification verification tests or species identity tests is growing. Future research is paramount to ensure the conservation of wild populations as well as aiding breeders in improving breeding strategies.  相似文献
