Heterosis for horticultural traits in Broccoli   总被引:4,自引:0,他引:4  
Over the last three decades, broccoli (Brassica oleracea L., Italica Group) hybrids made by crossing two inbred lines replaced open-pollinated populations to become the predominant type of cultivar. The change to hybrids evolved with little or no understanding of heterosis or hybrid vigor in this crop. Therefore, the purpose of the present study was to determine levels of heterosis expressed by a set of hybrids derived by crossing relatively elite, modern inbreds (n = 9). An additional objective was to determine if PCR-based marker derived genetic similarities among the parents can be useful to predict heterosis in this crop. Thirty-six hybrids derived from a diallel mating design involving nine parents were evaluated for five horticultural characters including the head characteristics of head weight, head stem diameter, and maturity (e.g., days from transplant to harvest), and the plant vigor characteristics of plant height, and plant width in four environments. A total of 409 polymorphic markers were generated by 24 AFLP, 23 SRAP and 17 SSR primer combinations. Euclidean distances between parents were determined based on phenotypic traits. About half of the hybrids exhibited highparent heterosis for head weight (1–30 g) and stem diameter (0.2–3.5 cm) when averaged across environments. Almost all hybrids showed highparent heterosis for plant height (1–10 cm) and width (2–13 cm). Unlike other traits, there was negative heterosis for maturity, indicating that heterosis for this character in hybrids is expressed as earliness. Genetic similarity estimates among the nine parental lines ranged from 0.43 to 0.71 and were significantly and negatively correlated with highparent heterosis for all traits except for stem diameter and days from transplant to harvest. Euclidean distances were not correlated with heterosis. With modern broccoli inbreds, less heterosis was observed for head characteristics than for traits that measured plant vigor. In addition, genetic similarity based on molecular markers was more highly correlated with plant vigor characteristics than head traits. Unlike with molecular marker-based estimates of genetic similarity, euclidean distance determined using phenotypic trait data was not predictive of heterosis. In conclusion, this study has documented heterosis in Brassica oleracea L., and the ability to predict heterosis in this crop using molecular marker-based estimates of genetic similarity among parents used in producing the hybrid. The objective of the present study was to correlate the genetic distances (GD) of single cross hybrids with yield, heterosis and specific combining ability (SCA) in the double cross hybrid synthesis. For this, 10 single cross commercial hybrids were used from different companies, and all the possible double hybrids were synthesized by a complete dialell. The hybrids were assessed in 15 locations in the 2005/2006 agricultural season, using the randomized complete block design with three repetitions. DNA was extracted from the single cross hybrids and 20 simple sequence repeat primers were used, nine of which were linked to the quantitative trait loci. It was ascertained that the single hybrids were superior in general to the double cross hybrids and that yield was highly correlated with heterosis and SCA (r = 0.75 and 0.82, respectively). There was no significant correlation between yield and GD (r = 0.25), but this index was at the limit of significance. There was a medium correlation between GD and heterosis (r = 0.40) and GD and SCA (r = 0.38). The intergroup hybrids placed by genetic grouping were generally more productive than intragroup hybrids, and the hybrids with GD greater than 0.84 had the maximum heterosis and SCA. It was concluded that the markers were efficient in placing hybrids in different heterosis groups and were also useful in eliminating the most negative heterosis and SCA.  相似文献

The aim of this research was to describe the temperature responseof gas exchange in grain sorghum (Sorghum bicolor L. Moench)leaves as temperature was increased in a fashion similar toits daily increase in a hot environment. The hypothesis thatsorghum hybrids manifested heterosis for carbon dioxide exchangerate on a wider temperature range as compared with their parentallines was also evaluated. Gas exchange of detached turgid leaveswas measured in four sorghum hybrids and their parents as leaftemperature rose steadily from 32 °C to 43 °C in 4 h.CER was maximal at about 37–40 °C depending on thegenotype. In one genotype (Tx378), CER was maximal at 42 °C.In three out of four hybrids significant heterosis was foundfor CER at the lower, the higher or at the full range of temperaturestested depending on the hybrid. Consequently, all three heterotichybrids displayed a greater temperature range for high CER ascompared with their parents. Heterosis in CER was largely explainedby heterosis in stomatal conductance, though the effects ofnon-stomatal components could not be elucidated here. Heterosisin transpiration was revealed in the same three hybrids, especiallyat moderate temperatures. Since heterosis in CER was relativelygreater than heterosis in transpiration, significant heterosisin transpiration ratio at moderate temperatures was seen intwo of the four hybrids. An effect of heat hardening on CERwas observed in parental line Tx430 and its two hybrids as seenin the reduction of CER at 36–37 °C and its subsequentrecovery as temperatures rose slowly to 43 °C. The effectof heat hardening on photosynthesis was also observed in someof the genotypes as a hysteresis in the association betweenstomatal conductance and CER as temperatures increased. Thishysteresis indicated that, for the same stomatal conductance,CER was greater after than before leaves were exposed to hightemperature. Multi-environmental experimentation made it possible to compare 311 hybrids and their 59 parental lines for five characters: yield, two yield components (number of grains per m2 and 1000-grain weight), height and date of heading. At all three sites, average high-parent heterosis exceeded 6%, and our results confirm that hybrids in general head as early as the earlier parent does and are as high as the taller parent. For the five characters studied ecovalences were significantly lower in the F1 than in the parents, indicating a higher level of homeostasis for hybrids. The experimental model, based on a ”top-cross” design comprised of 55 lines used as females and four testers, made it possible to implement different hybrid-value prediction models. For yield, by far the most important character to predict, the predictor based on the general combining ability and the per se value of the parents, appears to be the most interesting. The prediction remains, however, imperfect as specific combining ability (SCA) has a strong impact for this character. Attempting to take account of SCA in the prediction, by using a multiplicative model for ”male×female” interaction modelling, did not prove convincing. The hypothesis that heterosis in biomass production of sorghum(Sorghum bicolor L. Moench) may be ascribed to stability incarbon exchange rate (CER) over a wide range of environmentalconditions was evaluated. This hypothesis was based on previousresults from detached leaves that hybrids sustained greaterCER over a wider temperture range than their parents. Two grain sorghum hybrids (ATx378/RTx430 and ATx378/RTx434)and their parental lines were grown in the greenhouse in a gradientof ambient temperatures under two water regimes (well-irrigatedand drought up to heading). Plant water-use (estimated by weighingpots), leaf area, leaf gas exchange, grain yield, and above-groundbiomass were determined. Significant heterosis was found for biomass, grain yield perplant, and grain number per panicle. No heterosis occurred forharvest index, indicating that heterosis in grain yield wasdue to heterosis in biomass. Neither growth duration nor leafarea could explain heterosis in biomass. CER and stomatal conductancefor hybrid ATx378/RTx430 in the controls were greater than forboth its parents at leaf temperatures above 38 °C. This,however, was not observed in the other hybrid which was lessheterotic for biomass and grain yield in the controls. WhenCER data were subjected to a stability analysis by joint linearregression, the two hybrids had greater CER than their respectiveparents especially under conditions favouring high CER. Heterosis often occurs in offspring derived from a cross between inbred or divergent parents and can be observed as the superior performance of these hybrids for a wide variety of characters. Heterosis was compared in maize lines at two ploidy levels, diploid and tetraploid, to gain a better understanding of the interaction of heterosis and ploidy level. Employing genetically identical diploid and tetraploid maize derived from four different inbred lines, we investigated heterosis for 11 morphological traits, including several plant height measures, as well as flowering time for both silks and anthers. We find that the heterotic response of a certain hybrid differs between diploid and tetraploid lines, and that the response at one ploidy cannot serve as a predictor for the other. Also, progressive heterosis was found for several of the characters in the tetraploid double-cross hybrid, which can have four different alleles at one locus, compared to the double-cross diploid hybrids, which can only possess two alleles per locus. Overall, the results indicate that the heterotic response of tetraploid maize lines differs significantly from that of the diploid. Progressive heterosis, i.e., the additional hybrid vigor in double-cross tetraploid hybrids not found in their single-cross tetraploid parents, has been documented in a number of species including alfalfa,potato, and maize. In this study, four artificially induced maize tetraploids, directly derived from standard inbred lines, were crossed in pairs to create two single-cross hybrids. These hybrids were then crossed to create double-cross hybrids containing genetic material from all four original lines. Replicated fieldbased phenotyping of the materials over four years indicated a strong progressive heterosis phenotype in tetraploids but not in their diploid counterparts. In particular, the above ground dry weight phenotype of double-cross tetraploid hybrids was on average 34% and 56% heavier than that of the single-cross tetraploid hybrids and the double-cross diploid counterparts, respectively. Additionally,whole-genome resequencing of the original inbred lines and further analysis of these data did not show the expected spectrum of alleles to explain tetraploid progressive heterosis under the complementation of complete recessive model. These results underscore the reality of the progressive heterosis phenotype,its potential utility for increasing crop biomass production, and the need for exploring alternative hypothesis to explain it at a molecular level.  相似文献

Ten elite inbred lines (four japonica, six indica), chosen from those widely used in the hybrid rice breeding program at Human Hybrid Rice Research Center in China, were crossed to produce all possible hybrids excluding reciprocals. The 45 F1 hybrids along with the ten parents were evaluated for eight traits of agronomic importance, including yield potential, in a replicated field trial. The ten parents were analyzed with 100 arbitrary decamer oligonucleotide primers and 22 microsatellite (simple sequence repeats, SSRs) primer sets via polymerase chain reaction (PCR). Out of the 100 random primers used, 74 were informative and amplified 202 non-redundant bands (variants) with a mean of 2.73 bands per polymorphic primer. All 22 microsatellite primer sets representing 23 loci in the rice genome showed polymorphisms among the ten parents and revealed 90 alleles with an average of 3.91 per SSR locus. Cluster analysis based on Nei's genetic distance calculated from the 291 (202 RAPDs, 89 SSRs) non-redundant variants separated the ten parental lines into two major groups that corresponds to indica and japonica subspecies, which is consistent with the pedigree information. Strong heterosis was observed in hybrids for most of the traits examined. For the 43 diallel crosses (excluding 2 crosses not heading), yield potential, its components (including panicles per plant, spikelets per panicle and 1000-grain weight) and their heterosis in F1 hybrids showed a significant positive correlation with genetic distance. When separate analyses were performed for the three subsets, yield potential and its heterosis showed significant positive correlations with genetic distance for the 15 indica x indica crosses and the 6 japonica x japonica crosses; however, yield potential and its heterosis were not correlated with genetic distance for the 22 indica x japonica crosses. Results indicated that genetic distance measures based on RAPDs and SSRs may be useful for predicting yield potential and heterosis of intra-subspecific hybrids, but not inter-subspecies hybrids.  相似文献

To understand the genetic basis of inbreeding depression and heterosis in rice, main-effect and epistatic QTL associated with inbreeding depression and heterosis for grain yield and biomass in five related rice mapping populations were investigated using a complete RFLP linkage map of 182 markers, replicated phenotyping experiments, and the mixed model approach. The mapping populations included 254 F(10) recombinant inbred lines derived from a cross between Lemont (japonica) and Teqing (indica) and two BC and two testcross hybrid populations derived from crosses between the RILs and their parents plus two testers (Zhong 413 and IR64). For both BY and GY, there was significant inbreeding depression detected in the RI population and a high level of heterosis in each of the BC and testcross hybrid populations. The mean performance of the BC or testcross hybrids was largely determined by their heterosis measurements. The hybrid breakdown (part of inbreeding depression) values of individual RILs were negatively associated with the heterosis measurements of their BC or testcross hybrids, indicating the partial genetic overlap of genes causing hybrid breakdown and heterosis in rice. A large number of epistatic QTL pairs and a few main-effect QTL were identified, which were responsible for >65% of the phenotypic variation of BY and GY in each of the populations with the former explaining a much greater portion of the variation. Two conclusions concerning the loci associated with inbreeding depression and heterosis in rice were reached from our results. First, most QTL associated with inbreeding depression and heterosis in rice appeared to be involved in epistasis. Second, most ( approximately 90%) QTL contributing to heterosis appeared to be overdominant. These observations tend to implicate epistasis and overdominance, rather than dominance, as the major genetic basis of heterosis in rice. The implications of our results in rice evolution and improvement are discussed.  相似文献

Exploitation of heterosis in rice(Oryza sativa L.) has contributed greatly to global food security.In this study,we generated three sets of reciprocal F1 hybrids of indica and japonica subspecies to evaluate the relationship between yield heterosis and the circadian clock.There were no differences in trait performance or heterosis between the reciprocal hybrids,indicating no maternal effects on heterosis.The indica-indica and indica-japonica reciprocal F1 hybrids exhibited pronounced heterosis for chlorophyll and starch content in leaves and for grain yield/biomass.In contrast,the japonica-japonica F1 hybrids showed low heterosis.The three circadian clock genes investigated expressed in an above-high-parent pattern(AHP)at seedling stage in all the hybrids.The five genes downstream of the circadian clock,and involved in chlorophyll and starch metabolic pathways,were expressed in AHP in hybrids with strong better-parent heterosis(BPH).Similarly,three of these Research Arfive genes in the japonica-japonica F1 hybrids showing low BPH were expressed in positive overdominance,but the other two genes were expressed in additive or negative overdominance.These results indicated that the expression patterns of circadian clock genes and their downstream genes are associated with heterosis,which suggests that the circadian rhythm pathway may be related to heterosis in rice.  相似文献

研究了杂交水稻青优159(母本青A,父本R159)和广优四号(母本广A,父本青六矮)及其亲本功能叶片的希尔反应活性、光合磷酸化、ATP含量及ATP酶活性等。实验结果表明了两组杂交水稻功能叶片的希尔反应活性高于其亲本,其超亲优势分别为13.44%、13、93%,平均优势分别为26.44%、1774%;功能叶片的光合磷酸化活性亦有杂种优势,其超亲优势分别为21.35%、18.81%,平均优势分别为34.06%、22.71%;杂种F1的两种ATP酶(Cd -ATP酶和Mg -ATP酶)活性和叶组织中ATP含量均高于其亲本,亦表现出明显的杂种优势。另外从我们的试验结果中还可以见到,希尔反应活性、光合磷酸化活性、ATP酶活性及ATP含量与光合速率的大小有密切的正相关关系,说明这些生理生化特性可以作为高光合速率杂交水稻鉴别的指标。  相似文献

Early-generation hybrid fitness is difficult to interpret because heterosis can obscure the effects of hybrid breakdown. We used controlled reciprocal crosses and common garden experiments to distinguish between effects of heterosis and nuclear and cytonuclear epistasis among morphotypes and advanced-generation hybrid derivative populations in the Piriqueta caroliniana (Turneraceae) plant complex. Seed germination, growth, and sexual reproduction of first-generation hybrids, inbred parental lines, and outbred parental lines were compared under field conditions. Average vegetative performance was greater for hybrids than for inbred lines, and first-season growth was similar for hybrids and outbred parental lines. Hybrid survival surpassed that of inbred lines and was equal to or greater than outbred lines' survival, and more F(1) than parental plants reproduced. Reductions in hybrid fitness due to Dobzhansky-Muller incompatibilities (epistasis among divergent genetic elements) were expressed as differences in vegetative growth, survival, and reproduction between plants from reciprocal crosses for both F(1) and backcross hybrid generations. Comparing performance of hybrids against parental genotypes from intra- and interpopulation crosses allowed a more robust prediction of F(1) hybrids' success and more accurate interpretations of the genetic architecture of F(1) hybrid vigor.  相似文献

Heterosis is of utmost economic importance in plant breeding. However, its underlying molecular causes are still unknown. Given the numerous advantages of Arabidopsis thaliana as a model species in plant genetics and genomics, we assessed the extent of heterosis in this species using five hybrids derived from five ecotypes. Parents, F(1) and F(2), generations in both reciprocal forms were grown in a greenhouse experiment with four replications. Mid-parent heterosis (MPH) and best-parent heterosis (BPH) averaged across hybrids were surprisingly high for biomass yield (MPH: 60.3%; BPH: 32.9%) and rosette diameter (MPH: 49.4%; BPH: 34.8%), but smaller for flowering date (MPH: 27.5%; BPH: 18.5%), seed yield (MPH: 18.9%; BPH: 1.7%), and yield components. Individual hybrids varied considerably in their MPH and BPH values for all traits, one cross displaying 140.1% MPH for biomass yield. MPH was not associated with parental genetic distance determined from molecular markers. Reciprocal effects were significant only in a few cases. With a proper choice of hybrids, our results encourage the use of Arabidopsis as a model species for investigating the molecular causes of heterosis.  相似文献

Sixty one double haploid (DH) lines were evaluated for yield components and yield and compared with the three corresponding hybrids from which they were derived through anther culture. Analysis of a 6×6 diallel experiment led to the selection of these hybrids based on their high heterosis and revealed the nature of gene action of the characters under evaluation. The DH lines along with the hybrids and parents were planted following a simple lattice design with two replications. The results show that in DH lines the values of the characters expressing predominantly additive genetic effects could reach the heterotic level of the hybrids. For characters including yield showing predominantly dominance effects, values in the DH lines were significantly lower than those of the corresponding hybrids. The promising DH lines, however, possess a higher yield potential than the better parents.  相似文献

Four lines of Ephestia kühniella each homozygous for one of three different allozyme alleles at the Est-2 locus and two alleles at the Adh locus were crossed in order to study the extent of somatic, reproductive and adaptive heterosis in F1 hybrids in comparison with the mean performance of the simultaneously reared inbred parent lines. With regard to adult weight and wing length (somatic heterosis) hybrids exhibit maximally 20% (males 10%) and 9% heterosis, respectively. As concerns the production of eggs and hatched larvae (reproductive heterosis) hybrids exceed the parental mean by 90% and 200%. Adaptive heterosis is realized by a shorter development period of the hybrids (maximally by 30%) as well as by significant lower variance of all metrical characters studied. In the F2 the degree of heterosis diminishes. There is neither an excess of heterozygotes among the segregating allozyme genotypes nor superior performance of the heterozygotes concerning any one of the traits studied. Therefore, it is concluded that the pronounced heterosis in F1 is not a single-gene-heterosis operating at the Est-2 and the Adh-locus.  相似文献

利用红花大金元×青梗,红花大金元×中烟14号P1、P2、F1、F2、B1、B2 6个世代资料对7个农艺性状和4个品质性状进行了基因效应分析。结果表明,性状均不符合简单的加性-显性遗传模型,多数性状加性效应显著而显性效应不显著,在3种互作效应中,所有性状至少有一种显著。互作效应普遍存在,是烟草性状杂种优势表现的主要原因之一。 Abstract:Two tobacco F1 hybrids,F2s,backcrosses B1s and B2s and their parents P1 and P2 were used to estimate the gene effects for 7 agronomic and 4 quality characters.The additive-dominance genetic model was not fit for all characters.The additive effects and the epistatic effects of most characters were significant,but the dominant effect not.The epistatic effects could not be ignored in tobacco breeding.They were one of main causes of heterosis for most characters.  相似文献

以6个籼型三系不育系和5个籼型恢复系按不完全双列杂交设计配制的30个杂交稻组合及其亲本为材料,对其籽粒淀粉RVA谱各特征值进行了测定和分析。结果表明:(1)亲本间和杂交稻组合间的淀粉RVA谱各特征值均存在极显著的差异,其中变异最大的是消减值,最小的是糊化开始温度。(2)杂交稻组合淀粉RVA谱各特征值的变异系数均小于杂交稻亲本的变异系数;峰值粘度、崩解值、最低粘度和最终粘度的平均值杂交稻组合大于亲本,其他性状的平均值杂交稻组合小于亲本。(3)峰值粘度存在极显著的正向超亲优势;崩解值、最低粘度、最终粘度和回复值存在极显著的正向中亲优势;消减值和峰值时间存在极显著的负向中亲优势和正向低亲优势;糊化开始温度存在极显著的负向低亲优势;最终粘度、回复值和糊化开始温度表现为极显著的负向竞争优势,其他性状表现为正向竞争优势。(4)峰值粘度、崩解值、消减值和回复值等4个性状,各杂交稻组合与其母本、中亲值呈现极显著正相关,峰值时间与中亲值呈显著正相关。而与其父本的相关均未达显著水平。  相似文献

The phenomenon of heterosis describes the increased agronomic performance of heterozygous F(1) plants compared to their homozygous parental inbred plants. Heterosis is manifested during the early stages of root development in maize. The goal of this study was to identify nonadditive gene expression in primary roots of maize hybrids compared to the average expression levels of their parental inbred lines. To achieve this goal a two-step strategy was used. First, a microarray preselection of nonadditively expressed candidate genes was performed. Subsequently, gene expression levels in a subset of genes were determined via high-throughput quantitative real-time (qRT)-PCR experiments. Initial microarray experiments identified 1941 distinct microarray features that displayed nonadditive gene expression in at least 1 of the 12 analyzed hybrids compared to the midparent value of their parental inbred lines. Most nonadditively expressed genes were expressed between the parental values (>89%). Comparison of these 1941 genes with nonadditively expressed genes identified in maize shoot apical meristems via the same experimental procedure in the same genotypes revealed significantly less overlap than expected by pure chance. This finding suggests organ-specific patterns of nonadditively expressed genes. qRT-PCR analyses of 64 of the 1941 genes in four different hybrids revealed conserved patterns of nonadditively expressed genes in different hybrids. Subsequently, 22 of the 64 genes that displayed nonadditive expression in all four hybrids were analyzed in 12 hybrids that were generated from four inbred lines. Among those genes a superoxide dismutase 2 was expressed significantly above the midparent value in all 12 hybrids and might thus play a protective role in heterosis-related antioxidative defense in the primary root of maize hybrids. The findings of this study are consistent with the hypothesis that both global expression trends and the consistent differential expression of specific genes contribute to the organ-specific manifestation of heterosis.  相似文献

为探讨玉米杂种优势的分子机理,以10个玉米自交系及其组配的38个杂交种为材料,利用cDNA-AFLP技术,分析杂种与亲本在玉米雌穗小穗分化期功能叶片的基因差异表达类型与主要农艺性状的杂种表现及杂种优势的关系。研究表明:(1)杂种的基因相对于其双亲,存在质和量的表达差异,其中质的差异表达类型包括:单亲沉默表达,双亲沉默表达,亲本显性表达和杂种特异表达等类型。(2)在雌穗小穗分化期,同一差异表达类型中不同杂交组合间差异很大;从总体平均看,杂种特异表达类型占25.22%,亲本显性表达类型占21.46%,双亲沉默表达类型占8.27%,单亲沉默表达类型占33.49%。(3)单亲沉默表达与株高的杂种表现呈显著正相关;双亲沉默表达与穗粗的杂种优势呈显著负相关,显性表达与行粒数和单株粒重的杂种优势呈显著负相关,其余表达类型与所有农艺性状杂种表现及杂种优势均不相关,并对结果进行了讨论。  相似文献

The effects of each of four contrasting combinations of two day (30, 26°C) and two night (24, 19°C) temperatures were studied on four cowpea varieties and their F1 hybrids. Early vegetative development was enhanced by the higher day, higher night temperature and flowering occurred first in plants receiving this treatment. The hybrids showed marked heterosis for leaf area but not for time to flowering. The ten genotypes studied showed correlated responses to increases in night temperature and differed in the degree of their response. The extent of the response (or stability) of varieties was often determined by the character values in the environment with the highest yield and heterotic hybrids were the least stable. Grain yield was highest in the lower day temperature environments but there was variation between genotypes in whether this occurred at the lower or higher night temperature. The pattern of responses shown by hybrids agreed with those of their parents indicating that stability was an inherited character. The results confirmed and extended those in earlier studies and emphasised the importance of a greater understanding of the nature of responses to the environment in selecting new varieties.  相似文献
