Infections caused by mucorales, with an increasing incidence after candidiasis and aspergillosis, are characterized by the fast angioinvasion of blood vessels and invasion of neighboring organs or structures. Mucorales most commonly cause rhinocerebral, pulmonary, cutaneous, digestive or disseminated infections, and their spread is favored by certain underlying diseases (diabetes, kidney failure) and risk factors (neutropenia, immunosuppression, iron overload). These infections have a high mortality rate, over 40% in many series, and the key to their cure depends on both an early diagnosis and an antifungal treatment, associated in most cases with extensive surgical debridement and other adjunctive therapies. Currently, there are international guidelines, not only local ones, for the management of mucormycosis, in which it is considered by consensus and with a strong recommendation that first-line treatment with high-dose liposomal amphotericin B is the best choice. The combined antifungal treatment of polyene agents with triazoles or candins remains in open debate.  相似文献

The design of new proteins that expand the repertoire of natural protein structures represents a formidable challenge. Success in this area would increase understanding of protein structure and present new scaffolds that could be exploited in biotechnology and synthetic biology. Here we describe the design, characterization and X-ray crystal structure of a new coiled-coil protein. The de novo sequence forms a stand-alone, parallel, six-helix bundle with a channel running through it. Although lined exclusively by hydrophobic leucine and isoleucine side chains, the 6-? channel is permeable to water. One layer of leucine residues within the channel is mutable, accepting polar aspartic acid and histidine side chains, which leads to subdivision and organization of solvent within the lumen. Moreover, these mutants can be combined to form a stable and unique (Asp-His)(3) heterohexamer. These new structures provide a basis for engineering de novo proteins with new functions.  相似文献

Through the human infantile stage, the progressive acquisition of the orthograde posture and bipedal locomotion is responsible for important morphostructural bony changes at the pelvis and the coxofemoral joint. This study uses digital processing techniques applied to a set of radiographic and microtomographic images to characterise the textural properties of the trabecular network and the topographic variation of the cortical shell in an ontogenetic series of 15 ilia from individuals aged between 0 and 40 years. The results suggest that the main structures of the trabecular architecture which are functional to the distribution and dissipation of the postural- and locomotor-related loads, are already recognizable at the age of 1–2 years, in association with the adoption of bipedal walking.  相似文献

De novo prediction of protein structures, the prediction of structures from amino acid sequences which are not similar to those of hitherto resolved structures, has been one of the major challenges in molecular biophysics. In this paper, we develop a new method of de novo prediction, which combines the fragment assembly method and the simulation of physical folding process: structures which have consistently assembled fragments are dynamically searched by Langevin molecular dynamics of conformational change. The benchmarking test shows that the prediction is improved when the candidate structures are cross-checked by an empirically derived score function.  相似文献

Through an examination of business relationships in the textile industry of Surat, a city in western India, this article evaluates different theories about the importance of social trust in economic development. The case study of Surat raises questions about models that posit a necessary expansion of broad civic ties or legal‐bureaucratic structures at the expense of close personal ties as a basis for industrial development. Surat's business people rely most strongly upon relationships to particular kins‐people, caste‐mates, and co‐religionists rather than broader civic bonds, formal legal controls, or bureaucratic systems as foundations for business trust. Although the expansion of the city's textile industry has created a greater need among the local population for impersonal, universalizing types of social trust, this article argues that these forms, show little sign of replacing personalized trust as the primary basis for local economic activity.  相似文献

The mystery of how a protein sequence specifies a unique structure has intrigued chemists, leading to the design and study of foldamers, non-natural oligomeric molecules that adopt well-defined structures. Recently, the sequence specificity of the various regular repeating structures has been revealed for bioinspired foldamers and such foldamers have been created to adopt helical bundle tertiary structures. One major strategy for the generation of abiotic foldamers has involved molecular design of the monomer geometry. These advances in foldamer research may lead to future applications in biomedical and materials science.  相似文献

The microscopic study and mineralogic determination of two triassic Sphinctozoa show that the spherolitic sclerodermites are exclusively build by fibro-cristalline aragonite. It is suggested that these structures have a significant systematic character.It is also shown, in the second part of the paper, that other organisms present preciseanalogies with typical Sphinctozoa regarding histology and mineralogy.  相似文献

Modulation of the xylem response to different types of vascular aggressions The evolution of cells lining the vessels in the stem xylem of carnation was compared in sham-stressed (s. e.), V. dabliae-infected (i V. d.) and P. cinerescens-infected (i P. c.) plants. All experimental stresses triggered ultrastructural changes leading to the appearance of new cell types. In parallel, vacuolar polyphenols were metabolized and dense-core vesicles appeared. These were early and independant phenomena. In all cases the reactions of carnation xylem were stronger against Verticillium than against Phialophora. Differences in vascular gummosis were disclosed whatever the criterion used (speed of installation, relative importance of the phenomenon, level of structural complexity, dynamics of appearance of the different structures). The possible relations between cell structures and gummous secretions were investigated by means of comparison of an efficient defence system (i V. d.) with a deficient one (i P. c.). All the criteria selected for analysis confirm that faced with a specific parasite the host response was disrupted. In other words, the biosynthetic pathways normally used by the plant after a fungal aggression were blocked, slowed or diverted. The differences in the responses of carnation to Verticillium and Phialophora give a picture of parasite specificity in agreement with the model recently proposed by de Wit (1981) for tomato/Cladosporium fulvum.  相似文献

Summary Aspergillus fasciculatum n. sp. has been found as contaminant of Lab. cultures, on 30.10.1952 and again on 29.9.1954. It grows well in Czapek neutral agar, as well as in Malt agar and Potato-dextroseagar, less intensively in Corn steep agar.Other media have not been tried so far.It presents conidiophores smooth or rough, yellowish, septated or not, undivided or branching at the apex, supporting then 2 vesicles or yet with the apex trifurcated or turning into branches, each swelling to form vesicles, side by side, with single head conidiophores.Abnormal structures in which the sterigma becomes a second conidiophore with a vesicle, were found very common. Normal structures present vesicles with two and one series of sterigmata.Having its connections within the Flavus-oryzae group, by these outstanding characteristics not reported before, it was considered a new species.

The thermal decomposition of methyl 4,6-O-benzylidene-2,3-di-O-[(methylthio)-thiocarbonyl]-α-d-glucopyranoside afforded methyl 4,6-O-benzylidene-2-thio-α-d-mannopyranoside 3-O,2-S-(S,S-dimethyl trithioorthocarbonate) and methyl 4,6-O-benzylidene-3-thio-α-d-allopyranoside 2-O,3-S-(S,S-dimethyl trithioorthocarbonate) in good yield. This decomposition can be generalized to 1,3-diols derived from sugars. Thus methyl 2,3-di-O-methyl-4,6-di-O-[(methylthio)thiocarbonyl]-α-d-glucopyranoside afforded in the same way the corresponding trithioorthocarbonates, following a regioselective process. The structures of these trithioorthocarbonates are confirmed by spectral and chemical proofs.  相似文献

In Pachycerianthus fimbriatus McMurrich, collar cells have been found in the ectoderm layer of marginal and labial tentacles. Villi, nematocytes, spirocysts, two types of mucous gland cells, and trypsinogen gland cells have been described. We have failed to discover special apical structures linked with the uptake of ferritin in solution. The rôle of apical villi and collar cells is discussed.  相似文献

The development of glycan-related databases and bioinformatics applications is considerably lagging behind compared with the wealth of available data and software tools in genomics and proteomics. Because the encoding of glycan structures is more complex, most of the bioinformatics approaches cannot be applied to glycan structures. No standard procedures exist where glycan structures found in various species, organs, tissues or cells can be routinely deposited. In this article the concepts of the GLYCOSCIENCES.de portal are described. It is demonstrated how an efficient structure-based cross-linking of various glycan-related data originating from different resources can be accomplished using a single user interface. The structure oriented retrieval options-exact structure, substructure, motif, composition and sugar components-are discussed. The types of available data-references, composition, spatial structures, nuclear magnetic resonance (NMR) shifts (experimental and estimated), theoretically calculated fragments and Protein Database (PDB) entries-are exemplified for Man(3.) The free availability and unrestricted use of glycan-related data is an absolute prerequisite to efficiently share distributed resources. Additionally, there is an urgent need to agree to a generally accepted exchange format as well as to a common software interface. An open access repository for glyco-related experimental data will secure that the loss of primary data will be considerably reduced.  相似文献

Numerous and large vertically extensive specimens of Conichnus conicus are described from the Tortonian deposits of the central Guadalquivir Basin (SW Spain). These burrowing structures form a low bioturbation index of the ichnofabric, which allows one to infer the original depositional setting. Both ichnological and sedimentological features suggest a coastal to shallow marine environment dominated by high sediment supply and high-energy sedimentary processes. Conichnus exhibits a characteristic internal fabric that reflects changes in sedimentation rates linked to various processes: (1) initial substrate penetration by the tracemaker; (2) gradual retrusive movement in response to rapid but continuous sediment aggradation; 3) moderate to high rates of retrusive migration as a consequence of rapid to very rapid sediment aggradation and tracemaker escape; and, finally, (4) abandonment of the burrow or burial of the tracemaker subsequently in response to extremely high sediment accumulation rates. Conichnus has been also described from other areas with similar deposits of different ages, where its important value as a paleoenvironmental indicator similarly has been indicated.  相似文献

Autophagy - the degradation of organelles and cytoplasmic material - occurs through dynamic rearrangements of cellular membrane structures. Following the induction of autophagy, newly formed autophagosomes transfer cytosolic materials to the lysosome or vacuole for degradation. The autophagosome is an organelle destined for degradation, suggesting that the membrane structure is formed de novo many times. The autophagosome is formed through the nucleation, assembly and elongation of membrane structures. The concerted action of several Apg/Aut/Cvt proteins around a characteristic subcellular structure (the preautophagosomal structure) is the key to understanding this novel type of membrane-formation process.  相似文献

The de novo molecular design program SPROUT has been applied to the X-ray crystal structures of Plasmodium and human dihydroorotate dehydrogenase, respectively. Prometaphasic chromosomes of man and chimpanzee are compared, using R, Q, T and H-bands techniques. — Six pericentric inversions, one telomeric fusion-translocation, 4 intercalar deletions or insertions, 16 deletions or additions of terminal Q-bands, and an important variation of heterochromatin distinguish the 2 species. — The evolutive role of chromosomal rearrangements separating the 2 species is discussed with particular reference to the formation of human chromosome No 2.- The simultaneous analysis of human chromosomal pathology and of the chromosomal structures in primates should contribute to the understanding of accidental modifications of the genome and to the interpretation of their consequences.

De novo structure prediction can be defined as a search in conformational space under the guidance of an energy function. The most successful de novo structure prediction methods, such as Rosetta, assemble the fragments from known structures to reduce the search space. Therefore, the fragment quality is an important factor in structure prediction. In our study, a method is proposed to generate a new set of fragments from the lowest energy de novo models. These fragments were subsequently used to predict the next‐round of models. In a benchmark of 30 proteins, the new set of fragments showed better performance when used to predict de novo structures. The lowest energy model predicted using our method was closer to native structure than Rosetta for 22 proteins. Following a similar trend, the best model among top five lowest energy models predicted using our method was closer to native structure than Rosetta for 20 proteins. In addition, our experiment showed that the C‐alpha root mean square deviation was improved from 5.99 to 5.03 Å on average compared to Rosetta when the lowest energy models were picked as the best predicted models. A study of the chitin contents in Rhodotorula yeast walls indicates that the contents of this amino polysaccaharide may vary from 0.58 to 12% according to the yeast and depending on the culture conditions.The variations of the glucosamine concentrations, estimated by direct determinations on the cell walls, do not reflect the modifications of the chitin contents.Depending on the culture conditions, the yeast cell walls possess structures which are differently degraded by enzyme actions. Cell walls with high glucosamine content, but low amino-acids content, are the more easily degraded.  相似文献
