In this treatise the author describes two species from the Lias, and four from the Dogger. One liassic species is already known (Reinholdella macfadyeni [Ten Dam]) from the Upper Lias (Bifrons-Zone), England, the other one was found by Dr.Bartenstein in Luxemburg, Lias δ Grenze ε, and has been namedReinholdella pachyderma nov. spec. The species from the Dogger areReinholdella dreheri (Bartenstein) and three new species, viz.Reinholdella brandi nov. spec.,Reinholdella ornata nov. spec. andReinholdella epistominoides nov. spec. The genusReinholdella has obviously derived from the genusConorboides Hofker (1951) and the speciesR. epistominoides proves that the genusEpistomina is related withReinholdella. It may be thatReinholdella is related withAsterigerina; but the large gap in the Cretaceous, where noReinholdella is found nor any species ofAsterigerina, contradicts this view.  相似文献

Tests ofEpistomaria semi-marginata (D’ORBIGNY) are analysed; a complicated toothplate is present and the marginal foramen is homologised with that ofEpistomina, whereas the supplementary sutural foramina are connected with the toothplate also. The taxonomy of the Epistominidae can be established on the basis of the development of the toothplate inReinholdella, Epistomina, Epistomaria, Cushmanella, Pseudobulimina, Robertina andRobertinoides. Species with coarse pores, double septal walls, spaces between which form supplementary chambers closed ventrally by porous plates, and lacking a toothplate in the chambers do not belong toEpistomaria but are closely allied toGavelinella from the Upper Crateceous. They are joined in the genusEpistomaroides Uchio. Two of them,E. separans (LE CALVEZ) andE. punctata (SAID) are analysed in this paper. Pycnotic degeneration of neoplastic and normal nuclei of mice has been studied cytologically and cytochemically, in pieces of tissues removed some time after subcutaneous transplantation. The cytological changes in pycnosis were found to be about as they have often been described earlier: the nuclei become spherical, shrink progressively, the nucleoli become lost, the chromatin becomes homogeneous. The relative changes in total desoxyribose nucleic acid were followed by the Feulgen reaction; the methylgreen stain was used as an index of nuclei acid polymerization; the Millon reaction was used for the detection of the protein changes. In order to measure the amounts of the Feulgen and methylgreen dye, respectively the color developed by the Millon reaction within individual nuclei, the photometric microscopic method after Pollister and Ris was used, which allows the estimation of relative amounts of colored precipitates within individual nuclei of fixed and stained sections. Using these methods the nucleoprotein composition of resting nuclei of a viable tumortissue was compared with that of three pycnotic stages (Stage I, II, III) in nuclei of necrotic areas of the same tumor tissue (Sarcoma 180). In Sarcoma 180 the change from a fresh tumor cell to Pycnosis I involves loss of nearly half the protein, no significant decrease in desoxyribose nucleic acid, and depolymerisation of over half of the desoxyribose nucleic acid. Later these processes continue, and there is added progressive loss of DNA. It is pointed out that at any pycnotic stage there is a total protein equivalent to about 20 times the highly polymerised (methylgreen positive) DNA, which is the ratio found in non-pycnotic nuclei of the type from which these were derived. It is suggested that this part of the pycnotic chromatin represents the unaltered nucleoprotein. In transplanted liver nuclei pycnosis is much more rapid than in neoplastic tissue, but otherwise the two processes are similar both cytologically and chemically. The discussion points out that: a) there is no real increase in chromatin stainability in pycnotic nuclei; b) the change of shape in pycnosis may be due to lower nuclear viscosity accompanying the DNA depolymerisation; c) the highly polymerised state of the DNA may depend upon the presence of some particular protein, possibly histone; and d) that pyenosis can be interpreted as showing high proteolytic activity in the nucleus and a delayed nuclease activity.  相似文献

The P.D. across the protoplasm of Valonia macrophysa has been studied while the cells were exposed to artificial solutions resembling sea water in which the concentration of KCl was varied from 0 to 0.500 mol per liter. The P.D. across the protoplasm is decreased by lowering and increased by raising the concentration of KCl in the external solution. Changes in P.D. with time when the cell is treated with KCl-rich sea water resemble those observed with cells exposed to Valonia sap. Varying the reaction of natural sea water from pH 5 to pH 10 has no appreciable effect on the P.D. across Valonia protoplasm. Similarly, varying the pH of KCl-rich sea water within these limits does not alter the height of the first maximum in the P.D.-time curve. The subsequent behavior of the P.D., however, is considerably affected by the pH of the KCl-rich sea water. These changes in the shape of the P.D.-time curve have been interpreted as indicating that potassium enters Valonia protoplasm more rapidly from alkaline than from acidified KCl-rich sea water. This conclusion is discussed in relation to certain theories which have been proposed to explain the accumulation of KCl in Valonia sap. The initial rise in P.D. when a Valonia cell is transferred from natural sea water to KCl-rich sea water has been correlated with the concentrations of KCl in the sea waters. It is assumed that the observed P.D. change represents a diffusion potential in the external surface layer of the protoplasm, where the relative mobilities of ions may be supposed to differ greatly from their values in water. Starting with either Planck''s or Henderson''s formula, an equation has been derived which expresses satisfactorily the observed relationship between P.D. change and concentration of KCl. The constants of this equation are interpreted as the relative mobilities of K+, Na+, and Cl- in the outer surface layer of the protoplasm. The apparent relative mobility of K+ has been calculated by inserting in this equation the values for the relative mobilities of Na+ (0.20) and Cl- (1.00) determined from earlier measurements of concentration effect with natural sea water. The average value for the relative mobility of K+ is found to be about 20. The relative mobility may vary considerably among different individual cells, and sometimes also in the same individual under different conditions. Calculation of the observed P.D. changes as phase-boundary potentials proved unsatisfactory.  相似文献

In the present study, we identified l-erythro-β-hydroxyasparagine (l-β-EHAsn) found abundantly in human urine, as a novel substrate of Zn2+-dependent d-serine dehydratase (DSD). l-β-EHAsn is an atypical amino acid present in large amounts in urine but rarely detected in serum or most organs/tissues examined. Quantitative analyses of urinary l-β-EHAsn in young healthy volunteers revealed significant correlation between urinary l-β-EHAsn concentration and creatinine level. Further, for in-depth analyses of l-β-EHAsn, we developed a simple three-step synthetic method using trans-epoxysuccinic acid as the starting substance. In addition, our research revealed a strong inhibitory effect of l-β-EHAsn on mammalian serine racemase, responsible for producing d-serine, a co-agonist of the N-methyl-d-aspartate (NMDA) receptor involved in glutamatergic neurotransmission.  相似文献

The action curve in Chara seems to depend (as in Nitella) on the outward movement of K+ from the sap. Presumably the increase in permeability in the inner protoplasmic surface and the outward movement of K+ destroy the concentration gradient of K+ across the inner protoplasmic surface. Hence the outwardly directed P.D. disappears, causing the rise (spike) of the action curve. The outer protoplasmic surface is normally insensitive to K+. But when it is made sensitive to K+ by treatment with guanidine the outwardly moving K+ sets up a positive P.D. on reaching the outer surface and this causes the action curve to fall, producing a peak. Then the curve has 2 peaks, the second being due to the process of recovery. The action curve thus comes to resemble that of Nitella in which the outer protoplasmic surface is normally sensitive to K+.  相似文献

If only one substance S passes through a membrane, the nature of this membrane is not of importance with respect to the direction of the diffusion; this is namely determined only by the O.S.A. of the two liquids. If, however, more substances pass through a membrane, the nature of this membrane is of great importance. If n substances diffuse through a membrane, we can distinguish 2n cases, when we take into consideration only the direction in which each of these substances passes through the membrane; if we call each of these cases a D.T. (diffusion-type), 2n D.T.''s may be conceived. Now we can deduce: one of these D.T.''s is not possible, the other 2n x 1 D.T.''s are thermodynamically admissible. Abstract

The shallow continental shelf in the Cape Fear Region of southwestern Onslow Bay, North Carolina, contains lag deposits with an abundance of megatoothed shark teeth belonging to Otodus megalodon (Agassiz 1835) and Otodus chubutensis (Ameghino 1906) that derive from the Pliocene Yorktown and Miocene Pungo River formations, respectively. These teeth exhibit different frequencies and orientations of macroborings identified as Gastrochaenolites torpedo Kelly and Bromley (1984 Kelly, S., and R. Different frequencies and orientations of macroborings seen in lag deposits containing O. megalodon and O. chubutensis teeth are the result of repeated exhumation and reworking in response to bathymetrically controlled wave-based erosion during storm events and glacioeustatically driven sea-level cyclicity across Onslow Bay. Chronological ranges of O. megalodon and O. chubutensis teeth that contain macroborings indicate that these lag deposits may have been forming since the late early Miocene.  相似文献

Fluoride is everywhere in the environment, yet it is toxic to living things. How biological organisms detoxify fluoride has been unknown until recently. Fluoride-specific ion transporters in both prokaryotes (Fluoride channel; Fluc) and fungi (Fluoride Exporter; FEX) efficiently export fluoride to the extracellular environment. FEX homologs have been identified throughout the plant kingdom. Understanding the function of FEX in a multicellular organism will reveal valuable knowledge about reducing toxic effects caused by fluoride. Here, we demonstrate the conserved role of plant FEX (FLUORIDE EXPORTER) in conferring fluoride tolerance. Plant FEX facilitates the efflux of toxic fluoride ions from yeast cells and is required for fluoride tolerance in plants. A CRISPR/Cas9-generated mutation in Arabidopsis thaliana FEX renders the plant vulnerable to low concentrations (100-µM) of fluoride at every stage of development. Pollen is particularly affected, failing to develop even at extremely low levels of fluoride in the growth medium. The action of the FEX membrane transport protein is the major fluoride defense mechanism in plants.

The paper’s overall goal is to offer some guidance and clarification about how aspects of the Cantor et al. (2018 Cantor, P., Osher, D., Berg, J., Steyer, L., & Rose, T. (2018). Maleability, plasticity, and individuality: How children learn and develop in context. Cellular RNA is decorated with over 170 types of chemical modifications. Many modifications in mRNA, including m6A and m5C, have been associated with critical cellular functions under physiological and/or pathological conditions. To understand the biological functions of these modifications, it is vital to identify the regulators that modulate the modification rate. However, a high‐throughput method for unbiased screening of these regulators is so far lacking. Here, we report such a method combining pooled CRISPR screen and reporters with RNA modification readout, termed CRISPR integrated gRNA and reporter sequencing (CIGAR‐seq). Using CIGAR‐seq, we discovered NSUN6 as a novel mRNA m5C methyltransferase. Subsequent mRNA bisulfite sequencing in HAP1 cells without or with NSUN6 and/or NSUN2 knockout showed that NSUN6 and NSUN2 worked on non‐overlapping subsets of mRNA m5C sites and together contributed to almost all the m5C modification in mRNA. Finally, using m1A as an example, we demonstrated that CIGAR‐seq can be easily adapted for identifying regulators of other mRNA modification.  相似文献

1.25 per cent gelatin solutions containing enough NaOH to bring them to pH 7.367 (or KOH to pH 7.203) were made up with various concentrations of NaCl, KCl and MgCl2, alone and in mixtures, up to molar ionic strength. The effects of these salts on the pH were observed. MgCl2 and NaCl alone lower the pH of the Na gelatinate or the K gelatinate, in all amounts of these salts. KCl first lowers the pH (up to 0.01 M K+), then raises the pH. Mixtures of NaCl and KCl (up to 0.09 M of the salt whose concentration is varied) raise the pH; then (up to 0.125 M Na+ or K+) lower the pH; and finally (above 0.125 M) behave like KCl alone. Mixtures of MgCl2 and NaCl raise the pH up to 0.10 M Na+, and lower it up to 0.15 M Na+ regardless of the amount of MgCl 2. Higher concentrations of NaCl have little effect, but the pH in this range of NaCl concentration is lowered with increase of MgCl2. Mixtures of MgCl2 and KCl behave as above described (for MgCl2 and NaCl) and the addition of NaCl plus KCl to gelatin containing MgCl2 produces essentially the same effect as the addition of either alone, except that the first two breaks in this curve come at 0.07 M and 0.08 M [Na+ + K+] and there is a third break at 0.12 M. In this pH range the free groups of the dicarboxylic acids and of lysine are essentially all ionized and the prearginine and histidine groups are essentially all non-ionized. The arginine group is about 84 per cent ionized. Hence we are studying a solution with two ionic species in equilibrium, one with the arginine group ionized, and one with it non-ionized. It is shown that the effect of each salt alone depends upon the effect of the cation on the activity of these two species due to combination. The anomalous effects of cation mixtures may be qualitatively accounted for if one or both of these species fail to combine with the cations in a mixture in proportion to the relative combination in solutions of each cation alone. Special precautions were taken to ensure accuracy in the pH measurements. The mother solutions gave identical readings to 0.001 pH and the readings with salts were discarded when not reproducible to 0.003 pH. All doubtful data were discarded.  相似文献

Abstract

Quercetin is an important flavonoid compound, usually extracted from plants, vegetables and fruits such as blueberries, apples, green tea, wine, onions and possessing broad range of pharmacological properties, in particular, powerful antioxidant, antitoxic, antiinflammation and antimicrobial effects due to its various reactive sites. The structure of this phenolic compound consists of three (A?+?C) and B rings, bearing five hydroxyl groups. Primarily, the chemical structure of quercetin determines its physico-chemical properties. Earlier, it was established that isolated quercetin molecule can acquire 48 stable conformations (24 planar and 24 non-planar) due to the mobility of its hydroxyl groups and (A?+?C) and B rings with relative Gibbs free energies in the range of 0.0–25.3?kcal·mol?1 under normal conditions (Brovarets’ et al., 2019c Brovarets’, O. In this work by quantum-mechanical calculations at the MP2/6-311++G(2df,pd)//B3LYP/6-311++G(d,p) level of theory and Bader’s ‘Quantum Theory of Atoms in Molecules’, we have theoretically modeled the interconversions in the 24 pairs of the conformers of the quercetin molecule, occuring via the rotation of its non-deformable (A+С) and B rings around the С2-С1' bond through the quasi-orthogonal transition state with low values of the imaginary frequencies (28–33/29–36?cm?1) and Gibbs free energies of activation in the range of 2.17–5.68/1.86–4.90?kcal·mol?1 in the continuum with dielectric permittivity ε?=?1/ε?=?4 under normal conditions. Also, we studied the changes of the number of physico-chemical characteristics of all intramolecular-specific contacts – hydrogen bonds and attractive van der Waals contacts during these conformational rearrangements.
