Oocyst characteristics and histological features of the endogenous development of bile-bladder coccidia of the genus Choleoeimeria Paperna and Landsberg, 1989 are described and the main features for species differentiation are discussed for the following new species: C. allogamae n. sp. from Agama sp., Cameroon, West Africa; C. allogehyrae n. sp. from Gehyra australis, Magnetic Island (type) and mainland N Queensland, Australia; C. boulii n. sp. from Gehyra variegata, SW Queensland, Australia; C. calotesi n. sp. from Calotes mystaceus, Xiang-Mai, Thailand; C. heteronotis n. sp. from Heteronotia binoei, N Queensland, Australia; C. lygosomis n. sp. from Lygosoma buringi, Kon-Kaen, Thailand; C. sylvatica n. sp. from Carlia rhomboidalis, N Queensland, Australia, and C. xiangmaii n. sp. from Hemidactylus frenatus, Xiang-Mai, Thailand. Oocyst characteristic of Choleoeimeria are also reported from Oedura castelnaui, N Queensland. The described species demonstrate a diversity of associations with the bile-bladder epithelial lining, from a single parasite in a single hypertrophic host cell to multiple infections inducing the hypertrophied cells to form stratified layers, or merge into branched clumps.  相似文献

Choleoeimeria ghaffari n. sp. is described from the gallbladder of Eryx jayakari Boulenger in Saudi Arabia. Oöcysts are tetrasporocystic, cylindroidal, 23 × 14 μm, with a smooth bi-layered wall and length/width ratio of 1.5, without micropyle, oöcyst residuum and polar granule. Sporocysts are subspheroidal to ellipsoidal, 8 × 6 μm, with length/width ratio of 1.4, without Stieda, sub-Stieda and para-Stieda bodies but with sporocyst residuum. Sporozoites are banana-shaped measuring 10 × 1.5 μm. The endogenous development was found to occur in the gallbladder epithelium and the extrahepatic bile ducts. Mature meronts are spheroidal, c.10 μm wide, and suspected to produce 12–16 merozoites. Microgamonts are irregular in shape, 13 × 10 μm, whereas macrogamonts are mostly subspheroidal, c.12 μm wide, with a prominent centrally-located nucleus. Based on oöcyst morphology and the site of endogenous development (epithelium of the gallbladder and bile ducts) the new eimeriid coccidian was placed in the genus Choleoeimeria Paperna & Landsberg, 1989.  相似文献

The examination of gill parasites from Hemichromis fasciatus Peters, 1858 (Pisces, Cichlidae) in Africa revealed the presence of nine species of Monogenea, three belong to Onchobdella Paperna, 1968 and the others to Cichlidogyrus Paperna, 1960. Seven have been previously described (O. aframae Paperna, 1968; O. bopeleti Bilong Bilong & Euzet, 1995; O. voltensis Paperna, 1968; C. dageti Dossou & Birgi, 1984; C. euzeti Dossou & Birgi, 1984; C. falcifer Dossou & Birgi, 1984 and C. longicirrus Paperna, 1965); two, belonging to the latter genus, are considered as new species: C. sanseoi n. sp. and C. teugelsi n. sp. As parasitic species are not present in all the host distribution area, we think that H. fasciatus is made up of two distinct populations (or even sister species).  相似文献

The encystment of Laurenliella acuminata was divided into five stages: stage A (precystic semitransparent cell with dark-globules), stage B (precystic transparent cell), stage C (precystic pigmented cell), stage D (spherical shape without cyst wall) and stage E (young resting cyst), on the basis of observations of changes in morphology and pigmentation during encystment. The duration of these stages was also established. Observations by electron microscopy confirmed that the cyst wall, composed of four layers, is derived from different kinds of precursors which are synthesized “de novo.” The ectocyst precursors are composed of stacks of between 5 and 12 small thin plates or discs; these stacks are about 0.9 μm in length and 0.06 μm in height. The mesocyst precursors are fibrillar bodies of variable shapes, about 2.4 μm in maximum length and 0.12–0.16 μm in diameter. These precursors appear in the cytoplasm of the precystic cell during the first precystic stage (stage A). The endocyst precursors are rounded bodies surrounded by a fine membrane, and their contents appeared similar to the endocyst. The granular layer precursors are spherical bodies about 0.1–0.2 μm in diameter, surrounded by a double membrane presenting ribosomes adhering to its outer membrane. Both endocyst and granular layer precursors are observed in the precystic cytoplasm from stage B. On the basis of ultrastructural studies, a formation and growth model of the cyst wall of the hypotrichous ciliate Laurentiella acuminata is proposed.  相似文献

Summary The thyroid gland of adult salamanders, Hynobius nebulosus, in the breeding season was studied by electron microscopy. The follicular cells are different in cell height and fine structures; the taller cells with many cell organelles and granules and the lower cells with a few cell organelles and granules are both present in the same follicle. In the cytoplasm, three types of membrane-bounded granules, namely, cytosomes, colloid droplets, and vacuolar bodies and circular membrane complexes occur. The vacuolar bodies are subdivided into two types; the ordinary type having loosely distributed particles and the specific type containing tubules and/or closely packed filaments, crystalloid structures, except for the particles. The chromophobe colloids within the Bensley-cells correspond to extremely large, ordinary type vacuolar bodies, while the Langendorff-colloid cells possess increased numbers of granular cisternae of endoplasmic reticulum and a ribosome-rich, dense cytoplasmic matrix but not extremely large colloid. The intracytoplasmic circular membrane complexes appear in the Golgi area of cytosome-rich cells. It is suggested that they originate from the Golgi apparatus which was activated to produce many cytosomes. Intranuclear inclusions consisting of microtubules and filaments and tight junctions between two adjacent lateral plasma membranes are occasionally encountered.  相似文献

Seventy-seven specimens of seven freshwater fish species harvested in four sites from rivers and ponds of the Gauteng, North West and Limpopo Provinces (South Africa) were surveyed for coccidian infections. Two fish species were infected with apicomplexans belonging to Goussia Labbé, 1896. In banded tilapia Tilapia sparrmanii Smith unsporulated oöcysts of G. vanasi (Landsberg &; Paperna, 1987) were found which became sporulated in tap-water within 24 hours. Another species in the gut of chubbyhead barb Barbus anoplus Weber harboured sporulated oöcysts in the faeces and in the intestinal epithelium. Within 48h following the induction of mycelial to yeast-like phase conversion of Histoplasma farcininosum, randomly occurring hyphal cells were observed to contain multiple nuclei and markedly increased numbers of mitochondria. Yeast-like cells arose as buds from swollen tips of terminal hyphae, as sessile buds along the hyphae, and as buds from chlamydospores. Yeast-like cells were characterized by the presence of numerous buds over the surface of the mother cell. Bud scars were evident in the cell wall of the mother cell following abscission of the bud cell. Little similarity was noted between the fine structure of yeast-like H. farciminosum and that reported for H. capsulatum. The yeast-like cells of H. frciminosum underwent rapid transformation to the mycelial phase at 25 degrees C. The hyphal cell wall originated from the inner layer of cell wall of the yeast-like form. The cytoplasm of the hyphal cell usually contained a single nucleus, scattered mitochondria and occasional lipid storage bodies. Occasionally, Woronin bodies were observed at the septal pore.  相似文献

The fine structure of each of the blood cell types of Perophora viridis has been characterized and strong evidence for localization of vanadium in two of these types is given. There are eight cell types; phagocytes which may contain completely engulfed cells, lymphocytes with a prominant nucleolus and scanty cytoplasm packed with clustered ribosomes, and six other cell types each with distinctive granules. Morula cells contain a central nucleus and cytoplasm filled by wedged bodies, about five of which are seen in section. These bodies contain regularly spaced electron dense foci. Green cells have the same organization but contain bodies which are electron dense throughout. Granular amoebocytes contain many smaller lightly staining oval bodies and much glycogen. Another cell type (probably orange cells of light microscopy) contains numerous granular rounded bodies. Compartment cells have vacuoles containing electron dense particles and signet ring cells have usually one large vacuole which is electron dense lined and may contain electron dense particles. Developmental stages of these cell types show involvement of endoplasmic reticulum and Golgi bodies in granule formation. After glutaraldehyde fixation alone the only extremely electron dense components are particles in the compartment cells and signet ring cells implicating these as sites of vanadium localization, although not excluding other cell types.  相似文献

The histochemistry and fine structure of typical colonies of six species of Candida were studied, using a total of 31 clinical isolates. The colonies consisted of viable and degenerate cells which lay in an intercellular matrix. This matrix was made up of amorphous, granular, and fibrillar components, the relative proportions and total amount of which varied from species to species. The cells of all species were surrounded by a zone of homogeneous amorphus material, which may be a highly cross-linked carbohydrate. This separated intact cells from irregularly distributed granular debris derived from the cytoplasm of degenerate cells. Focal cellular degeneration and associated granular debris were present within the colonies of all species and were most common in the surface layers of cells of colonies of C. albicans and C. tropicalis. The large amounts of intercellular matrix in this region formed a surface coat on colonies of these two species. Intercellular strands of cell wall material, and to a lesser extent other membranous elements from degenerate cells, formed a prominent fibrillar meshwork in the colonies of C. albicans and C. tropicalis, but were less common in those of C. pseudotropicalis and C. guilliermondii and seldom seen in those of C. parapsilosis and C. krusei.  相似文献

The effect of subinhibitory concentrations of monomycin on the submicroscopic structures of 2 strains of S. typhi, 5 and 799 was studied. It was shown that formation of filamentoue forms, separation of the cell wall, thinning out of the cytoplasm granular component, increasing of the size of the matter with low electron optic density of fine granular structure were common in the cell response of both strains. Formation of vacuoli containing the thinned out granular component, filterable elements and complex membrane structures followed by their liberation into the medium and formation of the forms devoid of the cell walls was a characteristic peculiar property of the cell response in strain 799. The cells of strain 5 were characterized by formation of large granular osmiefilic matters and their excretion from the cells.  相似文献

The endodermis in the needles of Pinus pinaster was examined with light and electron microscopy. The endodermis is composed of very long, radially flattened cells, filled with a large central vacuole, which contains spherical dense bodies whose concentration decreases from the ends of the cell to the middle part. They are individually surrounded by a fine granular matrix. The central vacuole is bounded by a thick tonoplast. Other small, clear vacuoles are limited by a thin tonoplast. The parietal cytoplasm contains relatively few ribosomes, long slender chloroplasts, and lipid bodies. The smooth endoplasmic reticulum is highly developed along the tangential walls and frequently connected, or apposed, to the plasma membrane. Numerous primary pit fields are seen in the radial walls which are lignified and in the tangential walls; the latter exhibit a characteristic loosening of the outer layer of the wall. The lipid bodies are connected to endoplasmic reticulum tubules. The role of the endodermis in the active transport of water inside the needle is discussed in reference to previous physiological studies. The chemical composition of the vacuolar dense bodies is as yet unknown.  相似文献

ABSTRACT Sarcocystis dubeyella n. sp. and S. phacochoeri n. sp. from muscle fibers of the skeletal musculature of two warthogs in South Africa are described by light and and electron microscopy. Sarcocystis dubeyella sarcocysts are macroscopic (up to 12 mm long and 1 mm wide), with a parasite-induced encapsulation of the host muscle fiber in which the plasma membrane of the latter remained unaltered. The sarcocyst wall is characterized by evenly arranged, irregularly semicircular or rectangular villar protrusions (5.0 T. 2.8-11.0 μm) with indented margins and no specific content. Sarcocystis phacochoeri formed filiform microcysts (up to 4 mm long and 0.13 mm wide). Its cyst wall is provided with tightly packed, molarlike villar protrusions (1.6-3.3 T. 1.7-3.3 μm), with smooth margins, hollow on one side, and with longitudinal condensations of the fine granular matrix at various locations in the interior.  相似文献

The micro-anatomy of the corpuscles of Stannius of the toadfish, Opsanus tau, an aglomerular marine teleost, has been studied by light and electron microscopy. The corpuscles are composed of extensively anastomosed cords of epithelial cells which maintain intimate contact with blood capillaries. Most of the epithelial cells contain acidophilic granules which also show a positive reaction with the periodic acid-Schiff technique and aldehyde fuchsin. On the basis of fine structural criteria, three cell types can be recognized. The granular cells contain abundant quantities of granular endoplasmic reticulum, ribosomes, Golgi apparatus with prosecretory granules, coated vesicles, polymorphic mitochondria with lamellar cristae, filaments, microtubules, a cilium, a variety of lysosome-like dense bodies, glycogen particles, lipid droplets, secretory granules and intranuclear lipid-like inclusions. One variety of agranular cell (type I) is characterized by the total absence of secretory granules, but it contains large amounts of granular endoplasmic reticulum and ribosomes, conspicuous profiles of Golgi apparatus, coated vesicles and sometimes an abundance of glycogen. Another variety of agranular cell (type II) has poorly developed cytoplasmic organelles. The perivascular space between the capillary and parenchyma contains connective tissue cells and abundant nerve fibers. The different types of epithelial cells observed in the corpuscles of Stannius of this fish may represent functional stages of the secretory cycle in a single cell type.  相似文献

Chinese hamster cell strains in the early passages in culture display wide variation in number of nucleolus-like bodies per cell, though such strains are characteristically euploid. A variety of criteria indicate that the nucleolus-like bodies are true nucleoli. Their Azure B- and fast green-staining properties indicate the presence of RNA and protein; they have typical nucleolar fine structure, including both fibrous and granular components; radioautography reveals that their patterns of uptake of uridine-3H into RNA are similar to those reported for nucleoli of other cell types; actinomycin D, at a level which selectively inhibits ribosomal RNA synthesis, greatly reduces their RNA synthesis and also causes segregation of fibrous and granular nucleolar components. Colchicine was used to experimentally fragment the nuclei of these cells into a number of separate karyomeres, each presumably containing some, or only one, of the chromosomes of the complement. Almost all the karyomeres contain nucleolus-like bodies which, by the same criteria applied to the multiple nucleolus-like bodies of uninuclear cells, appear to be true nucleoli. The nucleoli of individual karyomeres of the same cell often differ from each other in fine structure while the multiple nucleoli of a uninuclear cell generally resemble each other. The evidence presented in this study indicates that Chinese hamster cells contain many nucleolus-producing sites scattered through the genome.  相似文献

The fine structural localization of nonspecific alkaline phosphatase was studied in the granular pneumonocytes (type II alveolar epithelial cells) of hamster lung by incubating sections of glutaraldehyde-fixed tissues in a medium containing lead ions and sodium beta-glycerophosphate or alpha-naphthyl acid phosphate. The specificity of the reaction was tested by exposing the sections to inhibitors of alkaline phosphatase. The results showed that alkaline phosphatase activity was present in the inclusion bodies of granular pneumonocytes. The enzyme reaction was strong in the membrane lining the inclusion bodies and a weaker reaction was generally detectable in the inclusion contents. Although only a proportion of the inclusion bodies showed enzyme activity, there was no obvious correlation between the reactivity of the inclusions and their intracellular position or size. The other organelles were unreactive. The finding of alkaline phosphatase activity within the inclusion bodies of granular pneumonocytes is an enigma as these organelles are generally considered to be lyosomes.  相似文献

Stigmatic hairs of the cotton flower were studied through their developmental stages up to anthesis. Stigmatic hairs invariably develop from a densely straining band of epidermal cells opposite the transmitting tissue cells. At anthesis, these are single cell structures measuring up to 300 μm long. At the 5-mm stage of stylar length (7–10 days before anthesis), some stigmatic hair cells begin to accumulate an osmiophilic substance between the plasmalemma and the cell wall, possibly synthesized in the endoplasmic reticulum. This material is apparently never secreted outside the cell wall. Immediately following this secretory phase in some stigmatic hair cells a second secretory phase starts. A dense osmiophilic substance, different in appearance from the previous phase, accumulates in the vacuoles of each hair cell. Concomitantly, dimorphism develops in the cytoplasmic densities of stigmatic hair. Some stigmatic hair cytoplasm appears very dense and shows signs of degeneration while other cytoplasm appears normal. A third secretory phase, which begins at anthesis, occurs in the normal hair cells. This phase is characterized by enhanced activity in the cytoplasm of the endoplasmic reticulum and Golgi apparatus. Large vesicles containing granular material are seen fusing with the plasmalemma. Coincident with this activity there is dissolution of the middle layers of the cell wall and the cuticle is ruptured at various points. The dense osmiophilic substance that had accumulated in the vacuole breaks down into fine granular material. Significance of these changes is discussed in relation to the pollen germination mechanism on the dry type stigma of cotton.  相似文献
