WRKY转录因子是植物响应病原菌胁迫最重要的转录因子之一,且参与抗病反应及信号传导通路的调控。为研究辣椒WRKY基因的生物学特征,以辣椒高抗疫病材料CM334为试材,克隆获得响应疫霉菌诱导的转录因子CaWRKY14。生物信息学分析表明,该基因DNA全长2 530 bp,cDNA全长1 662 bp,含有5个内含子,编码553个氨基酸,含有1个WRKY保守结构域,属于Group Ⅱ(b)。实时荧光定量表达分析表明,CaWRKY14不仅受ABA和疫霉菌胁迫诱导表达,且表达量分别在12 h和24 h时达到峰值,分别是对照的8.54和8.04倍,同时也受高盐、热激和干旱胁迫诱导。利用VIGS技术对CaWRKY14转录因子进行沉默后发现,抗病材料CM334接种疫霉菌后趋于发病。研究表明,CaWRKY14基因在辣椒响应疫霉菌胁迫进程中可能发挥着重要作用。  相似文献

Chylomicrons (CM), secreted by the intestine in response to fat ingestion and to a lesser extent during the postabsorptive state (lipid poor CM), are the major vehicles whereby ingested lipids are transported to and partitioned in energy-storing and energy-utilizing tissues of the body. CM contribute significantly, although not exclusively, to postprandial lipemia. Intestinal CM production is upregulated in humans under conditions of insulin resistance and CM overproduction in such conditions contributes to the highly prevalent dyslipidemia of these conditions. In addition, CM remnants possess direct atherogenic properties. CM assembly and secretion is regulated by many factors apart from ingested fat (the primary stimulus for their secretion), including a number of nutritional, hormonal, metabolic and genetic factors. Understanding the mechanisms that regulate CM production in health and disease may lead to treatments and prevention of atherosclerosis and cardiovascular disease. Contrast media (CM)-induced nephropathy is a common cause of iatrogenic acute renal failure. The aim of the present review was to discuss the mechanisms and risk factors of CM, to summarize the controlled studies evaluating measures for prevention and to conclude with evidence-based strategies for prevention. A review of the relevant literature and results from recent clinical studies as well as critical analyses of published systematic reviews used MEDLINE and the Science Citation Index. The cytotoxicity induced by CM leads to apoptosis and death of endothelial and tubular cells and may be initiated by cell membrane damage together with reactive oxygen species (ROS) and inflammation. Cell damage may be aggravated by factors such as tissue hypoxia, properties of individual CM such as ionic strength, high osmolarity and/or viscosity. Clinical studies indeed support this possibility, suggesting a protective effect of ROS scavenging with the administration of N-acetylcysteine, ascorbic acid erdosteine, glutathione and bicarbonate infusion. The interaction between extracellular Ca2+, which plays a central role in intercellular contacts and production of ROS, and the in vitro toxicity of CM was also reviewed. The current review addresses the role of oxidative stress in the pathogenesis of CM in the kidney as well as current and potential novel treatment modalities for the prevention of neutrophil activation and CM-induced kidney degeneration in patients. ROS production through CM-induced renal hypoxia may exert direct tubular and vascular endothelial injury. Preventive strategies via antioxidant supplementation include inhibition of ROS generation or scavenging.  相似文献

Summary Cochlear microphonic (CM) and evoked neural (N-1) potentials were studied in two species of Doppler shift compensating bats with the aid of electrodes chronically implanted in the scala tympani. Potentials were recorded from animals fully recovered from the effects of anesthesia and surgery. InPteronotus p. parnellii andRhinolophus rouxi the CM amplitude showed a narrow band, high amplitude peak at a frequency about 200 Hz above the resting frequency of each species. InPteronotus the peak was 25–35 dB higher in amplitude than the general CM level below or above the frequency of the amplitude peak. InRhinolophus the amplitude peak was only a few dB above the general CM level but it was prominent because of a sharp null in a narrow band of frequencies just below the peak. The amplitude peak and the null were markedly affected by body temperature and anesthesia. InPteronotus high amplitude CM potentials were produced by resonance, and stimulated cochlear emissions were prominent inPteronotus but they were not observed inRhinolophus. InPteronotus the resonance was indicated by a CM afterpotential that occurred after brief tone pulses. The resonance was not affected by the addition of a terminal FM to the stimulus and when the ear was stimulated with broadband noise it resulted in a continual state of resonance. Rapid, 180 degree phase shifts in the CM were observed when the stimulus frequency swept through the frequency of the CM amplitude peak inPteronotus and the frequency of the CM null inRhinolophus. These data indicate marked differences in the physiological properties of the cochlea and in the mechanisms responsible for sharp tuning in these two species of bats.  相似文献

Sex pheromone mating disruption (MD) is an approach used to control several moth pest species of pome fruit by disrupting the ability of the males to find females and consequently prevent mating. The following experiments were performed to determine the effectiveness of several new and experimental sex pheromone MD technologies, and dispenser densities for simultaneous control of the codling moth (CM), Cydia pomonella (L.), and the oriental fruit moth (OFM), Grapholita molesta (Busck) (both Lepidoptera: Tortricidae), in Pennsylvania apple orchards. In one study, three MD approaches to control CM and oriental fruit moth – CM and OFM Disrupt Micro‐Flakes, Isomate CM/OFM TT, and both a CideTrak OFM and a CideTrak CM dispenser containing both codlemone and pear ester – and an insecticides‐only treatment were compared over the course of 2 years. In the other studies, the efficacy of several CheckMate Duel dispenser densities (i.e., 250, 375, 425, and 500 dispensers ha?1) were compared against Isomate CM/OFM TT, and an insecticides‐only treatment. The CideTrak CM/pear ester combination and Isomate CM/OFM TT treatments both substantially reduced CM captures in traps in 2007 and 2008. Meanwhile, OFM trap shutdown was highest in the CheckMate Duel densities of 375 (99.9 ± 0.08%) and 500 dispensers ha?1 (98.9 ± 0.07%) and the Isomate CM/OFM TT treatment (98.0 ± 1.13%), and lowest in the 250 dispensers ha?1 density treatment (94.3 ± 3.23%). In orchards where OFM is the dominant pest species, a CheckMate Duel dispenser density of 375 ha?1 is necessary for effective control, whereas higher densities are needed to control CM.  相似文献

Summary Cassava (Manihot esculenta Crantz) is a staple food for a large sector of human population in the tropics. It is widely produced for its starchy roots by small farmers over a range of environments on poor infertile soils with virtually no inputs. It is highly productive under favorable conditions and produces reasonably well under adverse conditions where other crops fail. The crop, once established, cansurvive for several months without rain. There is a wide variation within the cassava germplasm for tolerance to prolonged drought and the possibility to breed and select for stable and relative high yields under favorable and adverse conditions does indeed exist. Research with several cassava clones at CIAT has shown that high root yield under mid—term stress is not incompatible with high yield under nonstress conditions. Plant types with high yield potential under both conditions (e.g. the hybrid CM 507-37) are characterized by having slightly higher than optimum leaf area index under nonstress conditions, higher leaf area ratio and more intensive and extensive fine root system.  相似文献

COPI vesicles serve for transport of proteins and membrane lipids in the early secretory pathway. Their coat protein (coatomer) is a heptameric complex that is recruited to the Golgi by the small GTPase Arf1. Although recruited en bloc, coatomer can be viewed as a stable assembly of an adaptin‐like tetrameric subcomplex (CM4) and a trimeric ‘cage’ subcomplex (CM3). Following recruitment, coatomer stimulates ArfGAP‐dependent GTP hydrolysis on Arf1. Here, we employed recombinant coatomer subcomplexes to study the role of coatomer components in the regulation of ArfGAP2, an ArfGAP whose activity is strictly coatomer‐dependent. Within CM4, we define a novel hydrophobic pocket for ArfGAP2 interaction on the appendage domain of γ1‐COP. The CM4 subcomplex (but not CM3) is recruited to membranes through Arf1 and can subsequently recruit ArfGAP2. Neither CM3 nor CM4 in itself is effective in stimulating ArfGAP2 activity, but stimulation is regained when both subcomplexes are present. Our findings point to a distinct role of each of the two coatomer subcomplexes in the regulation of ArfGAP2‐dependent GTP hydrolysis on Arf1, where the CM4 subcomplex functions in GAP recruitment, while, similarly to the COPII system, the cage‐like CM3 subcomplex stimulates the catalytic reaction.  相似文献

Chiari-like malformation (CM) is a developmental abnormality of the craniocervical junction that is common in the Griffon Bruxellois (GB) breed with an estimated prevalence of 65%. This disease is characterized by overcrowding of the neural parenchyma at the craniocervical junction and disturbance of cerebrospinal fluid (CSF) flow. The most common clinical sign is pain either as a direct consequence of CM or neuropathic pain as a consequence of secondary syringomyelia. The etiology of CM remains unknown but genetic factors play an important role. To investigate the genetic complexity of the disease, a quantitative trait locus (QTL) approach was adopted. A total of 14 quantitative skull and atlas measurements were taken and were tested for association to CM. Six traits were found to be associated to CM and were subjected to a whole-genome association study using the Illumina canine high density bead chip in 74 GB dogs (50 affected and 24 controls). Linear and mixed regression analyses identified associated single nucleotide polymorphisms (SNPs) on 5 Canis Familiaris Autosomes (CFAs): CFA2, CFA9, CFA12, CFA14 and CFA24. A reconstructed haplotype of 0.53 Mb on CFA2 strongly associated to the height of the cranial fossa (diameter F) and an haplotype of 2.5 Mb on CFA14 associated to both the height of the rostral part of the caudal cranial fossa (AE) and the height of the brain (FG) were significantly associated to CM after 10 000 permutations strengthening their candidacy for this disease (P = 0.0421, P = 0.0094 respectively). The CFA2 QTL harbours the Sall-1 gene which is an excellent candidate since its orthologue in humans is mutated in Townes-Brocks syndrome which has previously been associated to Chiari malformation I. Our study demonstrates the implication of multiple traits in the etiology of CM and has successfully identified two new QTL associated to CM and a potential candidate gene.  相似文献

The cultivation of bone marrow was used to quantitate the levels of eosinophil differentiation factors (EDF) produced in conditioned medium (CM) by incubation of mesenteric lymph node cells (MLNC) with mitogens or specific antigens from the intestinal nematode parasite, Trichostrongylus colubriformis. In liquid cultures with 20 units ml−1 recombinant murine interleukin-5 (IL-5), bone marrow cells (BMC) from either normal or infected donors contained <5% eosinophils and differentiated to > 50% eosinophils over 2–3 weeks. Conditioned medium from 3–4 week infected donors produced between 20 and 50% eosinophils when donor MLNC were stimulated with the specific antigen preparation SP3, but macrophages predominated when using CM from MLNC incubated with Concanavalin A (ConA). CM from MLNC of challenged donors incubated with SP3 produced 30–70% eosinophils in BMC assays, with highest levels induced by CM from high responder (HR) donors. Marrow from parasitized or normal donors gave rise to comparable proportions of eosinophils. CM was also produced from LNC of donors given protein or parasite antigens in adjuvant where between 28 and 35% eosinophils were produced in culture. There were no differences between activities attributable to the antigen, but Freund's complete adjuvant induced earlier differentiation of BMC than alum-induced CM. The results confirm that high levels of EDF activity are specifically induced by parasitic infection, and can also be produced by intraperitoneal and subcutaneous inoculation of adjuvanted antigens. Consistent with the greater eosinophilia exhibited by HR guinea pigs to infection with T.colubriformis L3, their MLNC also produced the highest levels of EDF activity.  相似文献

From 1985 to 1989 five large scale surveys were made to document the spread of the cassava mealybug (CM) Phenacoccus manihoti Matile‐Ferrero (Hom., Pseudococcidae) and the releases and successful establishment of its exotic parasitoid Epidinocarsis lopezi (De Santis) (Hym., Encyrtidae) through most cassava‐growing areas of Malawi. In a multiple regression analysis involving 29 meteorological, agronomic and plant variables from 476 fields, the duration of E. lopezi's presence was the major factor influencing CM population densities. In the first year the CM was recorded in a particular place, 25% of all tips had more than 100 CM. Wherever E. lopezi had been present for two years or more, CM populations were reduced on average seven times and tips infested with more than 100 CM became rare (1%). In parallel, damage on cassava tips stabilized at a low level. Similarly, along Lake Malawi, 84.9% of all tips had more than 10 CM in 1986; this value stabilized at 3.3%‐4.0% in 1988–1989. Indigenous coccinellids were often abundant in the first year of the CM infestation, but their populations collapsed later, while the frequency of fields with E. lopezi increased. In 1987, farmers abandoned 28.4% of all fields where E. lopezi was just introduced as compared with 2.7% where the parasitoid had been present for two years or more. Overall, satisfactory control was achieved by E. lopezi in all but a few fields concentrated on extremely poor soils characterized by sand dune vegetation. The socioeconomic implications of this ongoing and apparently successful biological control program are discussed.  相似文献

We confirmed the existence of growth-promoting substances in the conditioned media (CM) from the rotifer Brachionus plicatilis at an early exponential growth phase and isolated a novel protein with a growth-promoting activity from the crude extract (CE) of rotifer cells. CM was prepared from the culture media where rotifers had been cultured at an early exponential growth phase and filtered through a 0.22-μm filter membrane. The growth-promoting activity was determined using rotifers in CM for 5 days. As a result, the increase of rotifers added with CM was significantly higher than that of the control in artificial seawater (P < 0.001). Moreover, the growth-promoting activity of CM was dose-dependent and inactivated by heat treatment at 80°C for 60 min. Meanwhile, CM filtered through a <10 kDa ultrafiltration membrane showed a low activity, whereas proteinase K treatment resulted in a complete inactivation. These results suggest that the rotifer secrets growth-promoting proteins into CM. CE also contained a protein with the activity and properties similar to those found in CM. Then, CE was subjected to purification of a growth-promoting protein for convenience using various types of chromatography after fractionation with 30–80% saturated ammonium sulfate. Subsequently, a protein with an approximate molecular weight of 25000 was isolated, and its N-terminal amino acid sequence was determined to be PAVVDFTAVWFGPLQMIKP. An orthologue was found in the EST database of B. plicatilis, the full sequence of which showed about 50% identity to the corresponding regions of thioredoxins from other organisms.  相似文献

Summary The linear programming model (LPM) of Belovsky (1978, 1986) and modifications of the classical or contingency model incorporating a digestive constraint (CM) were tested using foraging data recorded for kudus (Tragelaphus strepsiceros) browsing savanna vegetation over the late wet season. Food choice was between the herbaceous and woody plant components for LPM and among plant species or categories for CM. The constraints considered were consumption (cropping) rate, foraging time and digestive capacity. Woody communities dominated byBurkea andAcacia represented alternative habitat types. Following a minor adjustment, LPM represented the overall average diet and predicted the dietary differences between habitat types. However, the kudus failed to respond dietarily to variations among days and foraging sessions (meals) in the parameters constraining intake. The kudus accepted a wider dietary range than predicted to be optimal by CM. Evidence suggested that neither foraging time, nor digestive capacity, formed an effective constraint under the study conditions. Thermal tolerance and gut space may become limiting only towards the extremes of environmental variability that animals experience. LPM is vulnerable to circularity if average parameter values are used to estimate constraint settings. The energy maximizer—time minimizer dichotomy fails to take into account the fitness consequences of alternative foraging responses. CM is less cryptic in its application than LPM and so has greater heuristic value, despite its predictive failures. However, there may be no consistent ranking of food types where multiple constraints that are variable in their effectiveness apply. Dynamic programming models offer a solution, but pose a formidable challenge in complex natural environments.  相似文献

Filtrates (conditioned medium) from high-density Chlorella vulgaris cultures in photobioreactors were obtained and tested for autoinhibitory activity under different conditions. Exponentially growing cells were inoculated at low initial cell concentration (2 × 105 cells/ml) in 90% conditioned medium (CM) supplemented with 10% fresh medium (FM) at low (atmospheric) CO2 levels. The time sequence of DNA histograms of cells in CM cultures showed that there is an accumulation of cells with two and four DNA equivalents in the culture over a period of time, signifying a blockage of cells at the division stage of the cell cycle. Examination of the chemical composition of CM showed the presence of high concentrations (> 10 mM) of bicarbonate. Adding similar bicarbonate concentrations to FM were found to have similar effects as CM cultures, causing blockage of cell division, though the intensity of the blocking effect was lower. The bicarbonate-free CM did not show any cell cycle modulating or inhibitory activity. The growth of cells cultivated at high (5%) CO2 levels in 90% CM supplemented with 10% FM was comparable to 10% FM cultures, indicating nutrient limitation in 90% CM culture. When the 90% CM culture was supplemented with 100% nutrients, the growth rate and final cell concentration was similar to 100% FM culture. Based on these results we conclude that C. vulgaris does not secrete any autoinhibitor(s) or cell cycle modulating compound(s) under the conditions from which the CM was obtained.  相似文献

The fungus Fusarium moniliforme is ubiquitous on corn throughout the world and is a likely co-contaminant on corn infested with aflatoxin-producing Aspergillus flavus. Ammoniation has been used to detoxify aflatoxin-contaminated commodities. To determine the effect of ammoniation on the toxic potential of Fusarium moniliforme, male Sprague-Dawley rats were fed either diets containing 10% sound corn, ammoniated corn, corn culture material of hepatotoxic F. moniliforme strain MRC 826 (CM), or ammoniated CM for four weeks. They were observed for signs of toxicity and hematological, serum chemical and histopathological evaluations were made. Groups of male Balb/c mice were fed diets fortified with 10% sound corn or CM for four weeks and evaluated by serum chemical and histopathological means to determine the suitability of mice as a model species for investigation of F. moniliforme-induced hepatotoxicity. Ammoniation was ineffective for detoxification of the CM. Hepatotoxicity and renal toxicity of CM and ammoniated CM were qualitatively similar, although renal tubular lesions appeared more advanced in rats fed ammoniated CM. Adrenal cortical cellular vacuolation was also found in CM and ammoniated CM-fed rats, while focal seminiferous tubular degeneration and aspermia were found only in the testes of ammoniated CM-fed rats. Fumonisin B1 concentrations of the CM and ammoniated CM diets averaged 99 and 75 ppm, respectively. CM containing 99 ppm fumonisin B1 also produced hepatotoxicity in mice similar to that found in CM-fed rats. Thus, mice may be useful for investigations of F. moniliforme-induced hepatotoxicity.Mention of a trademark, proprietary name or vendor does not imply its approval by the US Department of Agriculture to the exclusion of others that may also be suitable.  相似文献

Obesity is a multifactorial, chronic, inflammatory disease that involves different processes, such as adipose tissue hypoxia. The aim of the current study was to characterize the effects of conditioned medium (CM) from lipopolysaccharide (LPS)-activated macrophages on the regulation of hypoxia-inducible factor 1α (HIF-1α)-related genes in murine adipocytes. For the in vitro analyses, 3T3-L1 murine adipocytes (9 days postdifferentiation) were incubated either in CM (25% medium of RAW 264.7 murine macrophages with 24 hr 500 ng/ml LPS), LPS at 500 ng/ml, or hypoxia (Hx; 1% O2, 94% N2, 5% CO2) for 24 hr. For the in vivo experiments, mice were fed a high-fat diet. Both epididymal white adipose tissue (eWAT) and adipocytes in CM showed upregulation of Glut1, Mcp1, Il10, Tnf, and Il1b. The secretion of IL-6, TNF-α, and MCP-1 was also increased in CM-treated adipocytes. Moreover, increased levels of HIF-1α subunit and nuclear factor kappa B p65 were found after CM treatment, linking Hx, and inflammation. HIF-1α directly bound vascular endothelial growth factor A (Vegfa) and uncoupling protein 2 (Ucp2) genes, up- and downregulating its expression, respectively. Furthermore, the oxygen consumption rate was 30% lower in CM. The siRNA knockdown of mammalian target of rapamycin (Mtor) reversed the induction of HIF-1α found in CM. The macrophage infiltration simulated through CM seems to be a similar environment to an abnormally enlarged eWAT. We have evidenced that HIF-1α plays a regulatory role in the expression of Vegfa and Ucp2 in CM. Finally, the inhibition of the mTOR pathway prevented the HIF-1α activation induced by CM. The involvement of HIF-1α under proinflammatory conditions provides insight into the origins of Hx in obesity.  相似文献

Induction of chloramphenicol (CM) resistance in Staphylococcus aureus was investigated by using several CM derivatives. It was found that dl-threo-1-p-nitrophenyl-2-dichloroacetamino-1,3-dichloropropane has high antibacterial activity but low activity of induction for CM resistance. In spite of low antibacterial activity, induction of CM resistance occurred after prior treatment with dl-threo-1-p-nitrophenyl-2-dichloroacetamino-3-chloropropane-1-ol. It was found that dl-chloramphenicol di-acetate, dl-threo-1-p-nitrophenyl-2-dichloroacetamino-3-bromopropane-1-ol and dl-threo-1-phenyl-2-dichloroacetamino-1,3-propanediol have induction ability in spite of the absence of antibacterial activity. Other derivatives were classified into two groups; (1) low antibacterial activity and induction of CM resistance and (2) loss of both activities.  相似文献

Culture-dependent and independent approaches were used to understand the microbiota thriving in tertiary coalbed, located in Jammu and Kashmir, India. We observed changes in physicochemical properties of the surface sediment (CM1) and coalbed (CM2) which detailed the influence of environmental factors on the structure and capabilities of bacterial communities. A total of 316 bacterial isolates representing 35 genera were isolated. We noted comparable difference in uncultivable bacterial communities which revealed the predominance of Proteobacteria in both the study sites. Moreover, we observed differential abundance of phyla Actinobacteria (49.6%), Firmicutes (4.2%), and Bacteroidetes (0.8%) in CM1, whereas Actinobacteria (11%), Firmicutes (37.8%), and Bacteroidetes (2.3%) in CM2. Additionally, functional imputations using PICRUSt depicted ~30% higher assemblage of major gene families in CM1 in comparison to CM2. Bacterial communities residing at CM1 were predominantly involved in methane oxidation, whereas CM2 communities found to play a vital process of conversion of coal to biogenic-methane enabling microbes to survive under constraints of high sulfur content, salt precipitation, and low nutrients and also provide clues to understand the potential of methanogenesis.  相似文献

The fine structure of the upper cuticular membrane (CM) of Clivia miniata leaves was investigated using electron microscopy. The CM is made up of a thin (130 nm) lamellated cuticle proper (CP) and a thick (up to 7 m over periclinal walls) cuticular layer (CL) of marbled appearance. Evidence is presented to show that the electron lucent lamellae of the CP do not simply represent layers of soluble cuticular lipids (SCL). Instead, the lamellation is probably due to layers of cutin differing in polarity. It is argued that the SCL in the Cp are the main barrier to water. Thickening of the CM during leaf development takes place by interposition of cutin between the CM and the cellin wall. The cutin of young, expanding leaves has a high affinity for KMnO4 and is therefore relatively polar. 广西北仑河口区新造高程约220 cm 的潮间带裸滩,移植白骨壤与茳芏、沟叶结缕草、芦苇和南水葱四种盐沼草进行混种,研究盐沼草-红树混种减轻污损动物对人工红树林危害的生物防治效果。结果表明:沟叶结缕草和茳芏可快速生长和扩展,缓流、促淤能力高于长势较差的南水葱和芦苇。在盐沼和白骨壤上发现19种污损动物,其中潮间藤壶、白条地藤壶、黑口滨螺和粗糙滨螺为优势种。茳芏和沟叶结缕草受污损程度较南水葱和芦苇轻。四种盐沼草受污损程度均低于白骨壤。白骨壤+茳芏、白骨壤+沟叶结缕草混种均可有效减轻污损动物对白骨壤苗木危害,这两类树-草混种处理区苗木的高度、叶数、枝数、枝下高和存活率等指标均较其他处理区好。初步判断“盐沼草-红树林协同生态修复体系”对于营造人工红树林有较高应用价值。  相似文献
