Phosphoenolpyruvate carboxylase (PEPC; EC 4-1-1-31) plays a paramount role in providing carbon for synthesis of malate and aspartate in alfalfa (Medicago sativa L.) root nodules. PEPC protein and activity levels are highly enhanced in N2-fixing alfalfa nodules. To ascertain the relationship between the cellular location of PEPC and root nodule metabolism, enzyme localization was evaluated by immunogold cytochemistry using alfalfa nodule PEPC antibodies. Gold labelling patterns in effective nodules showed that PEPC is a cytosolic enzyme and is distributed relatively equally in infected and uninfected cells of the nodule symbiotic zone. A high amount of labelling was also observed in pericycle cells of the nodule vascular system. Labelling was also detected within inner cortical cells, but the density was reduced by 60%. When Lotus corniculatus was transformed with a chimeric gene consisting of the 5′-upstream region of the PEPC gene fused to β-glucuronidase (GUS), GUS staining in nodules was consistent with immunogold localization patterns. The occurrence of PEPC in both infected and uninfected cells of the symbiotic zone of effective nodules coupled to the reduced amounts in ineffective nodules suggests a direct role for this enzyme in supporting N2-fixation. PEPC localization in the uninfected, interstitial cells of the symbiotic zone indicates that these cells may also have a role in nodule carbon metabolism. Moreover, the association of PEPC with the nodule vascular system implies a role for the enzyme in the transport of assimilates to and from the shoot.  相似文献

The contribution of carbon and nitrogen reserves to regrowth following shoot removal has been studied in the past. However, important gaps remain in understanding the effect of shoot cutting on nodule performance and its relevance during regrowth. In this study, isotopic labelling was conducted at root and canopy levels with both 15N2 and 13C‐depleted CO2 on exclusively nitrogen‐fixing alfalfa plants. As expected, our results indicate that the roots were the main sink organs before shoots were removed. Seven days after regrowth the carbon and nitrogen stored in the roots was invested in shoot biomass formation and partitioned to the nodules. The large depletion in nodule carbohydrate availability suggests that root‐derived carbon compounds were delivered towards nodules in order to sustain respiratory activity. In addition to the limited carbohydrate availability, the upregulation of nodule peroxidases showed that oxidative stress was also involved during poor nodule performance. Fourteen days after cutting, and as a consequence of the stimulated photosynthetic and N2‐fixing machinery, availability of Cnew and Nnew strongly diminished in the plants due to their replacement by C and N assimilated during the post‐labelling period. In summary, our study indicated that during the first week of regrowth, root‐derived C and N remobilization did not overcome C‐ and N‐limitation in nodules and leaves. However, 14 days after cutting, leaf and nodule performance were re‐established.  相似文献

Summary Soybeans (Glycine max (L.) Merr.) have a high N requirement which is fulfilled by soil N uptake and N2-fixation. This study was concerned with the effects of past yield selection on N2-fixation in soybeans.The soybean cultivars, Lincoln, Shelby, and Williams, which represent successive improvements in the Lincoln germplasm, and a non-nodulating control were planted in a soil containing15N labelled organic matter. Two replications occurred on soil previously cropped to alfalfa and two on soil previously cropped to soybeans. Plants were harvested at five growth stages and leaf area, plant weight, total N, and atom percent15N were determined. Mature grain was harvested and yield components were also determined, as well as the total N and15N content.Cultivar differences in total dry matter were only evident at physiological maturity, when Williams contained the greatest dry matter. Williams exhibited the longest period of seed formation and seed fill and also had the highest grain yield which resulted from a larger weight per seed.The N content of the cultivars did not vary until physiological maturity when Williams contained the highest percent N. The quantity of N fixed at physiological maturity was highest for Williams and lowest for Lincoln. Fixed N contained in the harvested grain was greater for Williams than for the other two cultivars. The fraction of the total plant N derived from fixation was not greatly affected by cultivar and all cultivars acquired an average of 50% of their total N through N2-fixation.Previous cropping history greatly affected the quantity of N fixed and the fraction of the total plant N derived from fixation. Soybeans following soybeans were more dependent upon N2-fixation than soybeans following alfalfa with the former deriving 65% of the total plant N from fixation and the latter only 32%. These soybean cultivars apparently utilized soil N first and then used N2-fixation to satisfy their N requirement.The past selection for higher yield has resulted in soybean cultivars with improved capacities to fix atmospheric N2 and an improved ability to take up available soil N.  相似文献

Rhizodeposition has been proposed as one mechanism for the accumulation of significant amounts of N in soil during legume growth. The objective of this experiment was to directly quantify losses of symbiotically fixed N from living alfalfa (Medicago sativa L.) roots to the rhizosphere. We used 15N-labeled N2 gas to tag recently fixed N in three alfalfa lines [cv. Saranac, Ineffective Saranac (an ineffectively nodulated line), and an unnamed line in early stages of selection for apparent N excretion] growing in 1-m long polyvinylchloride drainage lysimeters in loamy sand soil in a greenhouse. Plants were in the late vegetative to flowering growth stage during the 2-day labelling period. We determined the fate of this fixed N in various plant organs and soil after a short equilibration period (2 to 4 days) and after one regrowth period (35 to 37 days). Extrapolated N2 fixation rates (46 to 77g plant–1 h–1) were similar to rates others have measured in the field. Although there was significant accretion of total N in rhizosphere compared to bulk soil, less than 1% was derived from newly fixed N and there were no differences between the excreting line and Saranac. Loss of N in percolate water was small. These results provide the first direct evidence that little net loss of symbiotically-fixed N occurs from living alfalfa roots into surrounding soil. In addition, these results confirm our earlier findings, which depended on indirect 15N labelling techniques. Net N accumulation in soil during alfalfa growth is likely due to other processes, such as decomposition of roots, nodules, and above ground litter, rather than to N excretion from living roots and nodules.  相似文献

以福鼎大白茶(FD)、保靖黄金茶1号(HJ1)、白毫早(BHZ)为材料,设置不施氮N_0(0 g)、低氮N1(11 g)、中氮N_2(22 g)和高氮N_3(33 g)4个氮素水平的盆栽实验,研究了铵态氮对3个品种茶树的根系活力、氮代谢关键酶及非结构性碳水化合物(NSC)的影响。结果表明:随着施氮水平的提高,N_2、N_3处理的茶树根系活力较对照N0显著增加(P0.05),但二者间无显著差异;叶片谷氨酰胺合成酶(GS)、谷氨酸合成酶(GOGAT)活性总体呈上升趋势;与对照比较,茶树叶片全氮和可溶性蛋白含量增加,其中HJ1在N_2和N_3处理后显著增加(P0.05);在3个茶树品种中,非结构性碳水化合物中可溶性总糖含量均呈上升趋势,淀粉含量具有品种特异性,施氮处理后3个茶树品种氮代谢关键酶活性及NSC含量变化存在差异,以HJ1的氮同化关键酶GS、GOGAT酶活性较高、根系活力较强,氮代谢产物显著增加,表明其具有较高的氮同化速率。施氮后HJ1的总NSC的含量及碳氮比的变化幅度较另外2个品种小,能够更好的保持碳氮平衡,游离氨基酸含量增幅较高,品质更优。因此,通过茶树氮代谢关键酶活性及非结构性化合物的研究能为茶树品种的品质评价以及提高茶树的品质和氮素利用效率提供依据。  相似文献

Distribution of net assimilated C in meadow fescue (Fectuca pratensi L.) was followed before and after cutting of the shoots. Plants were continuously labelled in a growth chamber with 14C-labelled CO2 in the atmosphere from seedling to cutting and with 13C-labelled CO2 in the atmosphere during regrowth after the cutting. Labelled C, both 14C and 13C, was determined at the end of the two growth periods in shoots, crowns, roots, soil and rhizosphere respiration. Distribution of net assimilated C followed almost the same pattern at the end of the two growth periods, i.e. at the end of the 14C- and the 13C-labelling periods. Shoots retained 71–73% of net assimilated C while 9% was detected in the roots and 11–14% was released from the roots, determined as labelled C in soil and as rhizosphere respiration. At the end of the 2nd growth period, after cutting and regrowth, 21% of the residual plant 14C at cutting (14C in crowns and roots) was found in the new shoot biomass. A minor part of the residual plant 14C, 12%, was lost from the plants. The decreases in 14C in crowns and roots during the regrowth period suggest that 14C in both crowns and roots was translocated to new shoot tissue. Approximately half of the total root C at the end of the regrowth period after cutting was 13C-labelled C and thus represents new root growth. Summary Two experiments were carried out with two nodulating and non-nodulating soybean isolines, with three different levels of N as (15NH4)2SO4 at the equivalent of 0, 25 and 50 kg N/ha. In the first experiment three seeds were sown in each pot and the plants harvested at 35, 55 and 75 days. In the second experiment only one seed was sown per pot and harvested at 75 days.Isotope dilution technique and in certain cases natural isotope variation (15N) was used to determine directly the origin of nitrogen in the plant, whether from soil, fertilizer or biological N2-fixation. The use of nodulating and non-nodulating isolines enabled comparison with the classical method of estimating N2-fixation by difference from total plant N. Results at the 75 day harvest were similar for either method, but at the earlier harvests, particularly at 35 days, the total-N method was inadequate. The isotope method appeared more sensitive while the total-N method suffered from greater variability with correspondingly high standard errors and significant differences.It was found that by the 35 and 55 day harvests hardly any N2-fixation had taken place, plant nitrogen being almost entirely derived from soil or fertilizer N. Plants in competition used up soil fertilizer N more rapidly, thus stimulating symbiotic nitrogen fixation. When only one plant was grown in each pot it had a greater proportion of N derived from soil or fertilizer, and less N derived from fixation. In general the15N data showed that only about 25% of the applied fertilizer N was absorbed by the plant.The nodulating isoline absorbed more N than the non-nodulating plants. This suggests a possible synergistic effect of N2-fixation on N derived from other sources, giving an increase in total-N content of nudulated plants. The N derived from N2-fixation was scarcely detectable in the roots but appeared to be translocated almost entirely to shoots and pods.With 25 kg N/ha the greater proportion of the nitrogen in the pods was derived from N2-fixation. Even with 50 kg N/ha the nitrogen in the pods derived from fixation remained high, that being derived from fertilizer being less than 15%. About 80% of the nitrogen in the nodules was due to fixation.In the present experiment the application of 25 kg N/ha appeared sufficient to give maximum N absorption by both isolines. Summary White clover (Trifolium repens L.) plants grown in pots and supplied with the same concentration x days of15N labelled nitrate, but in contrasting patterns and doses had similar N concentrations but differed in the proportions devived from N2 fixation and nitrate. N2-fixation and nodule dry weight responded rapidly (2–3 days) to changes in nitrate availability. Plants exposed frequently to small doses of nitrate took up more nitrate (and hence relied less on N2-fixation) and had greater dry weights and shoot: root ratios than those exposed to larger doses less often. In mixed ryegrass (Lolium perenne L.)/clover communities clover's ability to either successfully compete for nitrate or fix N2 gave it consistently higher N concentrations than grass whether they were given high or low nitrate nutrient. This higher N concentration was accompanied by greater dry weights than grass in the low nitrate swards but not where high levels of nitrate were applied.  相似文献

A S-deficient soil was used in pot experiments to investigate the effects of S addition on growth and N2-fixation in pea (Pisum sativum L.). Addition of 100 mg S pot−1 increased seed yield by more than 2-fold. Numbers of pods formed were the most sensitive yield component affected by S deficiency. Sulphur addition also increased the concentration of N in leaves and stems, and the total content of N in the shoots. The amounts of N fixed by pea were determined at four growth stages from stem elongation to maturity, using the 15N dilution technique. Sulphur addition doubled the amount of N fixed at all growth stages. In contrast, leaf chlorophyll content and shoot dry weight were increased significantly by S addition only after the flowering and pod fill stage, respectively. Pea roots were found to have high concentrations of S, reaching approximately 10 mg g−1 dry weight and being 2.6–4.4 times the S concentration in the shoots under S-sufficient conditions. These results suggest that roots/nodules of pea have a high demand for S, and that N2-fixation is very sensitive to S deficiency. The effects of S deficiency on pea growth were likely to be caused by the shortage of N, due to decreased N2-fixation. Phosphoenolpyruvate carboxylase (PEPC) plays a key role in N2 fixation and ammonia assimilation in legume root nodules. The enzyme can comprise up to 2% of the soluble protein in root nodules. We report here the isolation and characterization of a cDNA encoding the nodule-enhanced form of PEPC. Initially, a 2945 bp partial-length cDNA was selected by screening an effective alfalfa nodule cDNA library with antibodies prepared against root nodule PEPC. The nucleotide sequence encoding the N-terminal region of the protein was obtained by primer-extension cDNA synthesis and PCR amplification. The complete amino acid sequence of alfalfa PEPC was deduced from these cDNA sequences and shown to bear striking similarity to other plant PEPCs. Southern blots of alfalfa genomic DNA indicate that nodule PEPC is a member of a small gene family. During the development of effective root nodules, nodule PEPC activity increases to a level that is 10- to 15-fold greater than that in root and leaf tissue. This increase appears to be the result of increases in amount of enzyme protein and PEPC mRNA. Ineffective nodules have substantially less PEPC mRNA, enzyme protein and activity than do effective nodules. Maximum expression of root nodule PEPC appears to be related to two signals. The first signal is associated with nodule initiation while the second signal is associated with nodule effectiveness. Regulation of root nodule PEPC activity may also involve post-translational processes affecting enzyme activity and/or degradation.  相似文献

During vegetative regrowth of Medicago sativa L., soil N, symbiotically fixed N2 and N reserves meet the nitrogen requirements for shoot regrowth. Experiments with nodulated or non-nodulated plants were carried out to investigate the changes in N flows originating from the different N sources and in xylem transport of amino acids during regrowth. Exogenous N uptake, N2 fixation and endogenous N remobilization were estimated by 15N labelling and amino acids in xylem sap were analysed. Removal of shoots resulted in great declines of exogenous N flows derived either from N2 or from NH4NO3 during the first week of regrowth, thereafter recovery increased linearly. Mineral N uptake as well as N2 fixation occurred mainly between the 10th and 18th day after removal of shoots while exogenous N assimilation in intact plants remained at a steady level. Nitrogen remobilization rates in defoliated plants increased by at least three to five-fold, especially during the first 10 days following shoot removal. Compared to control plants, contents of amino acids in xylem sap, during the first 10 days of regrowth, were reduced by about 72% and 82% in NH4NO3 grown and in N2 fixing plants, respectively. Asparagine was the main amino acid transported in xylem sap of both treated plants. Its relative contents during this period significantly decreased from 75% to 59% and from 67% to 36% respectively in non-nodulated plants and in nodulated ones. The addition of combined nitrogen to substrate at an appropriate rate can stimulate N2-fixation thus inreasing the efficiency of the Alnus-Frankia symbiosis. To examine how nitrogen additions can effect the peformance of different pairs of symbionts, growth and time course of N2-fixation were studied in plants supplied with NH4NO3. Two cloned ofAlnus glutinosa (L.) Gaertn., propagatedin vitro, were inoculated with two strains ofFrankia (AVP3d and ACN14a) and grown in a greenhouse. Calcined montmorillonite (TotfaiceR) was used as growth substrate. Six N treatments were made up of varied amounts of NH4NO3 supplied in one single addition shortly before inoculation. Weekly measurements of shoot height and repeated measurements of nitrogenase activity (acetylene reduction) performed on intact root systems were used to monitor the development of the symbioses. Nitrogen treatments containing from 0.10 to 0.68 mg N g−1 dry substrate stimulated N2-fixation as well as growth. The relative performance of the two clones was different according to N treatment; one clone showed a greater benefit from the nitrogen input. Our results support the recommendation that selection of symbionts according to performance should be carried out with an input of combined nitrogen. This can provide optimum conditions for the development of each pair of symbionts.  相似文献

Phosphoenolpyruvate carboxylase (PEPC) genes from Corynebacterium glutamicum (cppc), Escherichia coli (eppc) or Flaveria trinervia (fppc) were transferred to Solanum tuberosum. Plant regenerants producing foreign PEPC were identified by Western blot analysis. Maximum PEPC activities measured in eppc and fppc plants grown in the greenhouse were doubled compared to control plants. For cppc a transgenic plant line could be selected which exhibited a fourfold increase in PEPC activity. In the presence of acetyl-CoA, a known activator of the procaryotic PEPC, a sixfold higher activity level was observed. In cppc plants grown in axenic culture PEPC activities were even higher. There was a 6-fold or 12-fold increase in the PEPC activities compared to the controls measured in the absence or presence of acetyl-CoA, respectively. Comparable results were obtained by transient expression in Nicotiana tabacum protoplasts. PEPC of C. glutamicum (PEPC C.g.) in S. tuberosum leaf extracts displays its characteristic K m(PEP) value. Plant growth was examined with plants showing high expression of PEPC and, moreover, with a plant cell line expressing and antisense S. tuberosum (anti-sppc) gene. In axenic culture the growth rate of a cppc plant cell line was appreciably diminished, whereas growth rates of an anti-sppc line were similar or slightly higher than in controls. Malate levels were increased in cppc plants and decreased in antisense plants. There were no significant differences in photosynthetic electron transport or steady state CO2 assimilation between control plants and transformants overexpressing PEPC C.g. or anti-sppc plants. However, a prolonged dark treatment resulted in a delayed induction of photosynthetic electron transport in plants with less PEPC. Rates of CO2 release in the dark determined after a 45 min illumination period at a high proton flux density were considerably enhanced in cppc plants and slightly diminished in anti-sppc plants. When CO2 assimilation rates were corrected for estimated rates of mitochondrial respiration in the light, the electron requirement for CO2 assimilation determined in low CO2 was slightly lower in transformants with higher PEPC, whereas transformants with decreased PEPC exhibited an appreciably elevated electron requirement. The CO2 compensation point remained unchanged in plants (cppc) with high PEPC activity, but might be increased in an antisense plant cell line. The dynamics of biomass and N accumulation following defoliation of alfalfa and the application of N fertilization has rarely been studied under field conditions, particularly in the seeding year. Our objectives were to determine the effect of N fertilization on the dynamics of biomass and N accumulation during the first regrowth of alfalfa in the seeding year, and to determine if a model describing critical N concentration developed for established stands could be used in the seeding year. In two separate experiments conducted in 1992 and 1993, the biomass and N accumulation of alfalfa grown with three N rates (0, 40 and 80 kg N ha-1) were determined weekly. Maximum shoot growth was reached with 40 kg N ha-1 in 1992, and maximum shoot growth was not reached with the highest N fertilization rate in 1993. Nitrogen fixation, root N reserves and soil inorganic N uptake when no N was applied were, therefore, not sufficient to ensure non-limiting N conditions, particularly when growth rates were the highest between 14 to 21 d after defoliation. Nitrogen fertilization increased shoot biomass accumulation in the first 21 d of regrowth, biomass partitioning to the shoots and shoot and taproot N concentrations. The model parameters of critical N concentration developed by Lemaire et al. (1985) for established stands of alfalfa were not adequate in the seeding year. The N requirements per unit of shoot biomass produced are greater in the seeding year than on established stands, and this was attributed to a greater proportion of leaves in the seeding year.  相似文献

Dominant canopy tree species have strong effects on the composition and function of understory species, particularly bryophytes. In boreal forests, bryophytes and their associated microbes are a primary source of ecosystem nitrogen (N) inputs, and an important process regulating ecosystem productivity. We investigated how feather moss-associated N2-fixation rates and contribution to N budgets vary in time and space among coniferous and broadleaf deciduous forests. We measured N2-fixation rates using stable isotope (15N2) labeling in two moss species (Pleurozium schreberi and Hylocomium splendens) in broadleaf deciduous (Alaska paper birch—Betula neoalaskana) and coniferous (black spruce—Picea mariana) stands near Fairbanks, interior Alaska, from 2013 to 2015. N2-fixation rates showed substantial inter-annual variation among the 3 years. High N2-fixation was more strongly associated with high precipitation than air temperature or light availability. Overall, contribution of N2-fixation to N budgets was greater in spruce than in birch stands. Our results enhance the knowledge of the processes that drive N2-fixation in boreal forests, which is important for predicting ecosystem consequences of changing forest composition.  相似文献

Alfalfa (Medicago sativa L.) N-sufficient plants were fed 1·5 mM N in the form of NO3, NH4+ or NO3 in conjunction with NH4+, or were N-deprived for 2 weeks. The specific activity of phosphoenolpyruvate carboxylase (PEPC) from the non-nodulated roots of N-sufficient plants was increased in comparison with that of N-deprived plants. The PEPC value was highest with NO3 nutrition, lowest with NH4+ and intermediate in plants that were fed mixed salts. The protein was more abundant in NO3-fed plants than in either NH4+- or N mixed-fed plants. Nitrogen starvation decreased the level of PEPC mRNA, and nitrate was the N form that most stimulated PEPC gene expression. The malate content was significantly lower in NO3-deprived than in NO3-sufficient plants. Root malate accumulation was high in NO3-fed plants, but decreased significantly in plants that were fed with NH4+. The effect of malate on the desalted enzyme was also investigated. Root PEPC was not very sensitive to malate and PEPC activity was inhibited only by very high concentrations of malate. Asparagine and glutamine enhanced PEPC activity markedly in NO3-fed plants, but failed to affect plants that were either treated with other N types or N starved. Glutamate and citrate inhibited PEPC activity only at optimal pH. N-nutrition also influenced root nitrate and ammonium accumulation. Nitrate accumulated in the roots of NO3- and (NO3 + NH4+)-fed plants, but was undetectable in those administered NH4+. Both the nitrate and the ammonium contents were significantly reduced in NO3- and (NO3 + NH4+)-starved plants. Root accumulation of free amino acids was strongly influenced by the type of N administered. It was highest in NH4+-fed plants and the most abundant amides were asparagine and glutamine. It was concluded that root PEPC from alfalfa plants is N regulated and that nitrate exerts a strong influence on the PEPC enzyme by enhancing both PEPC gene expression and activity.  相似文献

Future climatic conditions, including rising atmospheric CO2 and temperature may increase photosynthesis and, consequently, plant production. A larger knowledge of legume performance under the predicted growth conditions will be crucial for safeguarding crop management and extending the area under cultivation with these plants in the near future. N2 fixation is a key process conditioning plant responsiveness to varying growth conditions. Moreover, it is likely to increase under future environments, due to the higher photosynthate availability, as a consequence of the higher growth rate under elevated CO2. However, as described in the literature, photosynthesis performance is frequently down-regulated (acclimated) under long-term exposure to CO2, especially when affected by stressful temperature and water availability conditions. As growth responses to elevated CO2 are dependent on sink-source status, it is generally accepted that down-regulation occurs in situations with insufficient plant C sink capacity. Alfalfa management involves the cutting of shoots, which alters the source-sink relationship and thus the photosynthetic behaviour. As the growth rate decreases at the end of the pre-cut vegetative growth period, nodulated alfalfa plants show photosynthetic down-regulation, but during regrowth following defoliation, acclimation to elevated CO2 disappears. The shoot harvest also leads to a drop in mineral N uptake and C translocation to the roots, resulting in a reduction in N2 fixation due to the dependence on photosynthate supply to support nodule function. Therefore, the production of new shoots during the first days following cutting requires the utilization of reduced C and N compounds that have been stored previously in reserve organs. The stored reserves are mediated by phytohormones such as methyl jasmonate and abscisic acid and in situations where water stress reduces shoot production this potentially enables the enhancement of taproot protein levels in nodulated alfalfa, which may lead to these plants being in better condition in the following cut/regrowth cycle. Furthering our knowledge of legume performance under predicted climate change conditions will be crucial for the development of varieties with better adaptation that will achieve greater and more efficient production values. Furthermore, for this purpose it will be necessary to improve existing methodologies and create new ones for phenotype characterization. Such knowledge will provide key information for future plant breeding programs.  相似文献

Waterlogging of soybean plants (Glycine max L.) led to impaired symbiotic N2 fixation and a marked decline in glutamine (Gln) concentration in xylem bleeding sap. Xylem Gln concentration increased during the growth cycle of the plant and was correlated with nodule formation. Treatments known to impair N2 fixation, such as exposing the root system to pure N2 gas or a mixture of Ar and O2 (80:20; v/v), led to specific declines in xylem sap Gln. The decrease in Gln observed during waterlogging was also seen on transfer of nodulated plants to aerated hydroponics, where the decline was highly correlated with ureide content in the xylem sap. Upon flooding the nodulated root system, the specific decline in xylem sap Gln could be detected within 10 min and reached a minimum within 60 min, indicating that waterlogging has an immediate effect on N2 fixation. It is concluded that xylem Gln arises directly from N2-fixation and is a useful indicator of N2 fixation activity of symbiotic soybean plants.  相似文献
