The incorporation of Tweens (1 %, 2 %, 5 %) or olive oil (1 %, 2 %) in soil or in soil-containing substrate strongly stimulated mycelial growth of the edible ectomycorrhizal mushroom Tricholoma matsutake (Matsutake) after 1 or 3 months, respectively. The growth responses to Tween 40 and Tween 80 were dose-dependent. Fungal biomass increased up to 15-fold as a result of olive oil incorporation. After 4 months of Matsutake/pine co-culture in the presence of olive oil (2 %), compact aggregates of substrate, hyphae, and surface-colonized roots were observed, recalling in some ways the mycelial mat structure of Matsutake in the field, i.e. Shiro. Olive oil did not prevent formation of well-developed Hartig net palmettis although those seemed rather less abundant than without oil addition. The incorporation of Tween 80 or olive oil (2 %) into nutrient agar induced the proliferation of peripheral hydrophilic-like hyphae penetrating the medium. Tricholoma matsutake growth stimulation, possibly related to the presence of fatty acids in surfactants and oil, could be a consequence of the higher hydrophilicity of treated hyphae, or of enhanced lytic enzyme excretion and activity. Parameters such as adjuvant type, concentration, and growth conditions will be further optimised to formulate culture substrates adapted to the co-culture of T. matsutake and its host plants.  相似文献

One type of soil collected from Maoer-shan in Heilongjiang Province,China was selected to induce hyphal growth of Tricholoma matsutake by a soil screening experiment.It was confirmed that hyphal growth of all the tested T.matsutake isolates was significantly stimulated in soil by supplemented with 0.5%~2.0% olive oil.The aggregation of hyphae and soil resembled natural Shiro.The biomass of hyphae in the soil increases with increasing olive oil concentrations.Moreover,seedlings of Pinus densiflora grew well in the soil containing 0.5%~1.0% olive oil and were also successfully infected by T.matsutake isolate A in the soil containing 1.0% olive oil.This study established a culture system of artificial Shiro formation and also provided a premise for formulation of culture substratum for fruit body formation of T.matsutake.  相似文献

While it has been suggested that Matsutake mycorrhizae might not be functional and that Matsutake may behave as a saprobic fungus in soil or even have some pathogenic activity on seedlings, we investigated the consequences of Matsutake inoculation on Pinus densiflora growth. Seventy-five days after inoculation, hyphae were anchored on short roots and well-developed Hartig net palmettis were observed. Compared to both control treatments—seedlings treated with distilled water and seedlings treated with autoclaved mycelium—inoculation significantly stimulated seedling total dry weight by 70.9% and 98.0%, respectively. These findings attest that some type of symbiotic relationship must be functional and favour host growth, ruling out claims of pathogenicity under the sterile conditions used here.  相似文献

本研究通过土壤筛选找到适合松口蘑菌丝体生长的土壤,并在其土壤中添加适当浓度的橄榄油(0.5%-2.0%),有效地促进了菌丝体在土壤中的生长,菌丝生长状况酷似野外的松口蘑菌塘,且随着添加浓度的增加,菌丝体生物量呈明显的正相关。当土壤中橄榄油浓度为0.5%和1.0%时,松口蘑共生植物赤松苗的生长不但没有受到抑制,而且还能与菌丝体在这种“菌塘”中形成菌根。本研究成功地建立了中国松口蘑人工菌塘诱导体系,同时也为其人工子实体的诱导研究提供了必要的前提条件。  相似文献

Tricholoma matsutake (matsutake) is an ectomycorrhizal (ECM) fungus that produces economically important mushrooms in Japan. Here, we use microsatellite markers to identify genets of matsutake sporocarps and below-ground ECM tips, as well as associated host genotypes of Pinus densiflora. We also studied ECM fungal community structure inside, beneath and outside the matsutake fairy rings, using morphological and internal transcribed spacer (ITS) polymorphism analysis. Based on sporocarp samples, one to four genets were found within each fairy ring, and no genetic differentiation among six sites was detected. Matsutake ECM tips were only found beneath fairy rings and corresponded with the genotypes of the above-ground sporocarps. We detected nine below-ground matsutake genets, all of which colonized multiple pine trees (three to seven trees per genet). The ECM fungal community beneath fairy rings was species-poor and significantly differed from those inside and outside the fairy rings. We conclude that matsutake genets occasionally establish from basidiospores and expand on the root systems of multiple host trees. Although matsutake mycelia suppress other ECM fungi during expansion, most of them may recover after the passage of the fairy rings.  相似文献

Mycelial growth of the Matsutake mushroom (Tricholama matsutake) was much slower than that of the other mushroom species. Fungal and actinobacterial communities were analyzed together with soil chemistry and enzyme activities in order to profile the microbial diversity associated with the economically important mushroom Tricholoma matsutake. Samples of mycelium-soil aggregation (shiro) were collected from three experimental sites where sporocarps naturally formed. PCR was used to confirm the presence and absence of matsutake in soil samples. PCR-denaturing gradient gel electrophoresis (DGGE) fingerprinting and direct sequencing were used to identify fungi and actinobacteria in the mineral and organic soil layers separately. Soil enzyme activities and hemicellulotic carbohydrates were analyzed in a productive experimental site. Soil chemistry was investigated in both organic and mineral soil layers at all three experimental sites. Matsutake dominated in the shiro but also coexisted with a high diversity of fungi and actinobacteria. Tomentollopsis sp. in the organic layer above the shiro and Piloderma sp. in the shiro correlated positively with the presence of T. matsutake in all experimental sites. A Thermomonosporaceae bacterium and Nocardia sp. correlated positively with the presence of T. matsutake, and Streptomyces sp. was a common cohabitant in the shiro, although these operational taxonomic units (OTUs) did not occur at all sites. Significantly higher enzyme activity levels were detected in shiro soil. These enzymes are involved in the mobilization of carbon from organic matter decomposition. Matsutake was not associated with a particular soil chemistry compared to that of nearby sites where the fungus does not occur. The presence of a significant hemicellulose pool and the enzymes to degrade it indicates the potential for obtaining carbon from the soil rather than tree roots.  相似文献

Tricholoma matsutake is an economically important ectomycorrhizal fungus of coniferous woodlands. Mycologists suspect that this fungus is also capable of saprotrophic feeding. In order to evaluate this hypothesis, enzyme and chemical assays were performed in the field and laboratory. From a natural population of T. matsutake in southern Finland, samples of soil-mycelium aggregate (shiro) were taken from sites of sporocarp formation and nearby control (PCR-negative) spots. Soil organic carbon and activity rates of hemicellulolytic enzymes were measured. The productivity of T. matsutake was related to the amount of utilizable organic carbon in the shiro, where the activity of xylosidase was significantly higher than in the control sample. In the laboratory, sterile pieces of bark from the roots of Scots pine were inoculated with T. matsutake and the activity rates of two hemicellulolytic enzymes (xylosidase and glucuronidase) were assayed. Furthermore, a liquid culture system showed how T. matsutake can utilize hemicellulose as its sole carbon source. Results linked and quantified the general relationship between enzymes secreted by T. matsutake and the degradation of hemicellulose. Our findings suggest that T. matsutake lives mainly as an ectomycorrhizal symbiont but can also feed as a saprotroph. A flexible trophic ecology confers T. matsutake with a clear advantage in a heterogeneous environment and during sporocarp formation.  相似文献

Abstract

Colonization of plant roots by arbuscular mycorrhizal fungi can greatly increase the plant uptake of phosphorus and nitrogen. The most prominent contribution of arbuscular mycorrhizal fungi to plant growth is due to uptake of nutrients by extraradical mycorrhizal hyphae. Quantification of hyphal nutrient uptake has become possible by the use of soil boxes with separated growing zones for roots and hyphae. Many (but not all) tested fungal isolates increased phosphorus and nitrogen uptake of the plant by absorbing phosphate, ammonium, and nitrate from soil. However, compared with the nutrient demand of the plant for growth, the contribution of arbuscular mycorrhizal fungi to plant phosphorus uptake is usually much larger than the contribution to plant nitrogen uptake. The utilization of soil nutrients may depend more on efficient uptake of phosphate, nitrate, and ammonium from the soil solution even at low supply concentrations than on mobilization processes in the hyphosphere. In contrast to ectomycorrhizal fungi, nonsoluble nutrient sources in soil are used only to a limited extent by hyphae of arbuscular mycorrhizal fungi. Side effects of mycorrhizal colonization on, for example, plant health or root activity may also influence plant nutrient uptake.  相似文献

The ectomycorrhizal basidiomycete Tricholoma matsutake associates with members of the Pinaceae such as Pinus densiflora (red pine), forming a rhizospheric colony or “shiro,” which produces the prized “matsutake” mushroom. We investigated whether the host specificity of T. matsutake to conifers is innately determined using somatic plants of Cedrela odorata, a tropical broad-leaved tree (Meliaceae) that naturally harbors arbuscular mycorrhizal fungi. We found that T. matsutake could form in vitro shiro with C. odorata 140 days after inoculation, as with P. densiflora. The shiro was typically aromatic like that of P. densiflora. However, this was a root endophytic interaction unlike the mycorrhizal association with P. densiflora. Infected plants had epidermal tissues and thick exodermal tissues outside the inner cortex. The mycelial sheath surrounded the outside of the epidermis, and the hyphae penetrated into intra- and intercellular spaces, often forming hyphal bundles or a pseudoparenchymatous organization. However, the hyphae grew only in the direction of vascular bundles and did not form Hartig nets. Tricholoma fulvocastaneum or “false matsutake” naturally associates with Fagaceae and was also able to associate with C. odorata as a root endophyte. With T. matsutake, C. odorata generated a number of roots and showed greatly enhanced vigor, while with T. fulvocastaneum, it generated a smaller number of roots and showed somewhat lesser vigor. We argue that the host–plant specificity of ectomycorrhizal matsutake is not innately determined, and that somatic arbuscular mycorrhizal plants have a great potential to form mutualistic relationships with ectomycorrhizal fungi.  相似文献

The role of mycorrhizal fungi in acquisition of mineral nutrients by host plants is examined for three groups of mycorrhizas. These are; the ectomycorrhizas (ECM), the ericoid mycorrhizas (EM), and the vesicular-arbuscular mycorrhizas (VAM). Mycorrhizal infection may affect the mineral nutrition of the host plant directly by enhancing plant growth through nutrient acquisition by the fungus, or indirectly by modifying transpiration rates and the composition of rhizosphere microflora. A capacity for the external hyphae to take up and deliver nutrients to the plant has been demonstrated for the following nutrients and mycorrhizas; P (VAM, EM, ECM), NH4 + (VAM, EM, ECM), NO3 - (ECM), K (VAM, ECM), Ca (VAM, EM), SO4 2- (VAM), Cu (VAM), Zn (VAM) and Fe (EM). In experimental chambers, the external hyphae of VAM can deliver up to 80% of plant P, 25% of plant N, 10% of plant K, 25% of plant Zn and 60% of plant Cu. Knowledge of the role of mycorrhiza in the uptake of nutrients other than P and N is limited because definitive studies are few, especially for the ECM. Although further quantification is required, it is feasible that the external hyphae may provide a significant delivery system for N, K, Cu and Zn in addition to P in many soils. Proposals that ECM and VAM fungi contribute substantially to the Mg, B and Fe nutrition of the host plant have not been substantiated. ECM and EM fungi produce ectoenzymes which provide host plants with the potential to access organic N and P forms that are normally unavailable to VAM fungi or to non mycorrhizal roots. The relative contribution of these nutrient sources requires quantification in the field. Further basic research, including the quantification of nutrient uptake and transport by fungal hyphae in soil and regulation at the fungal-plant interface, is essential to support the selection and utilization of mycorrhizal fungi on a commercial scale.  相似文献

Bidding Customs and Habitat Improvement for Matsutake (Tricholoma matsutake) in Japan. A brief history of Japan’s matsutake production, use, and management is presented, with emphasis on habitat improvement efforts and the land use tradition of iriai. Bidding systems for allotment of matsutake gathering rights are discussed. Three villages with different bidding systems are compared to see what effects the bidding systems have on village finances, matsutake production, and matsutake habitat enhancement. Contrary to expectations, habitat for matsutake was not improved when land owners were guaranteed the gathering and selling rights to matsutake growing on their own lands. Instead, habitat improvement was most successful and matsutake production was highest on community-owned lands in Oka Village, where the iriai tradition is strongest.  相似文献

There have been some scientific reports suggesting that dual inoculations with arbuscular mycorrhizal (AM) and saprophytic soil fungi may cause an additive or synergistic growth enhancement of the inoculated host plant. Some Trichoderma spp. have shown antagonistic potential against pathogenic fungi and a beneficial effect on plant growth. Joint inoculations of the mycorrhizal fungus Glomus intraradices Schenck and Smith, isolated from a citrus nursery (Tarragona, Spain) and a strain of Trichoderma aureoviride Rifai, isolated from an organic compost, were tested on a citrus rootstock, Citrus reshni Hort. ex Tan. The interactions between both microorganisms and their influence on mycorrhizal root colonization and plant growth enhancement, the changes produced in the soil microbial activity, like esterase, trehalase, phosphatase and chitinase activities, and on microbial populations were evaluated in three organic substrates: (1) sphagnum peat and autoclaved sandy soil (1/1, v/v), (2) sphagnum peat, quartz sand and perlite (1/1/1, v/v) and (3) pine bark compost (BVU, Prodeasa Product). Substrate characteristics were more important than the AM inoculation treatment in the determination of enzyme activity. In bark compost, the number of bacterial colonies obtained on soil-dilution plates was significantly higher than in peat and sand mixtures. Inoculation with T. aureoviride alone produced no significant effect on growth enhancement of C. reshni. However, dual inoculation with both, T. aureoviride and G. intraradices significantly increased plant growth in two of the substrates used and was the best treatment in pine bark amended compost. The inoculation with T. aureoviride did not affect the development of mycorrhizal root colonization. These results show a synergistic effect of G. intraradices and T. aureoviride on the growth of C. reshni in organic substrates and indicate the potential benefits of using combined inoculations.  相似文献

Matsutake are the edible fruiting bodies of a few species ofTricholoma. The principal species areT. matsutake, T. magnivelare, andT. caligatum. Related species areT. bakamatsutake, T. quercicola, T. fulvocastaneum andT. robustum. These occur naturally throughout the Northern Hemisphere associated mainly with softwood trees but also with members of the Fagaceae. However, none have ever been cultivated or successfully introduced into new areas. They are held in very high regard in Japan where annual consumption is approximately 3000 tons. Annual production of matsutake in Japan is less than 1000 tons with the balance imported from China, North and South Korea, Canada, the USA, Morocco and Taiwan. Matsutake only fruit in late summer and autumn and are not found in the Southern Hemisphere. Therefore, there is the opportunity for it to be introduced into Southern Hemisphere countries and fruiting bodies produced for out-of-season Japanese markets. Wholesale prices in Japan range from ¥3350 to ¥70 000/kg (US$27-560) with the price reflecting quality, origin and availability. Retail prices at the start of the season can be up to ¥160 000/kg (US$1275). This paper reviews published and our own unpublished information on matsutake, and highlights areas of research that might result in a method for the cultivation of these fungi.  相似文献

以采自云南丽江的松茸子实体为材料,进行组织分离后,利用一对ITS引物(ITS1-ITS4)对子实体(SR176B,SR172B)和分离物(SR176H,SR172H)进行了PCR扩增、琼脂糖凝胶电泳分析,得到了700bp左右的扩增条带,进一步对ITS序列进行同源性检索比对,结果表明SR176H与SR176B,SR172H与SR172B序列同源性均为100%,鉴定出该分离物就是松茸的纯培养物。  相似文献

本文采用液体培养的方法,对六个松口蘑菌种的碳素营养生理及培养液中的-葡萄糖苷酶的活性进行了分析。结果表明,纤维二糖能很好的支持所有菌株的生长,其效果不逊色于葡萄糖;木聚糖能够一定程度地维持各个菌种的生长,但效果不及葡萄糖和纤维二糖。这些数据为松口蘑具有一定的利用天然有机碳源的潜在能力提供了间接的证据,也为松口蘑人工栽培基质的调制提供了新的思路。  相似文献

Matsutake, the fruiting body of Tricholoma matsutake, is among the most economically important edible ectomycorrhizal (EM) mushrooms worldwide. This EM fungus develops “shiros”, which are mycelial aggregations that develop in association with the roots of EM coniferous trees and soil particles in well-drained and nutrient-poor forest soil. The fruiting bodies occur on the periphery of the outward-growing shiro. In spite of vast research, the cultivation of matsutake has been mostly unsuccessful. Commercial demand is therefore met by harvesting the fruiting bodies that naturally occur in forests of EM coniferous trees, mainly Pinus densiflora. Recent inoculation studies have produced mycorrhiza and shiro structures, and the host range of Tr. matsutake and associated species has been clarified. It has also become possible to identify strains of Tr. matsutake by DNA sequencing, which has also been useful to determine the origins of matsutake species in Asia and to elucidate the genetic structure of shiro. In this review, basic research and the outcomes of various trial of matsutake cultivation are discussed.  相似文献

The higher levels of nitrogen in ammonium form stimulated the growth of mycelia and increased the accessible as well as the total acid phosphatase activity of Paxillus involutus (Batsch) Fr. isolates grown in pure culture. Rates of mycelia growth and acid phosphatase activities varied widely from one isolate to another. Scots pine (Pinus sylvestris L.) seedlings were inoculated with different P. involutus isolates in axenic conditions. Shoots of pine seedlings with mycorrhizae contained more phosphorus than shoots of non-mycorrhizal seedlings. The relations between growth and phosphatase activity of P. involutus isolates and their efficiency in supplying the host plant with phosphate are discussed.  相似文献
