Chytrid fungi are highly potent parasites of phytoplankton. They are thought to force phytoplankton organisms into an evolutionary arms race with high population diversity as the outcome. The underlying selection regime is known as Red Queen dynamics. However, our study suggests a more complex picture for chytrid parasitism in the cyanobacterium Planktothrix. Laboratory experiments identified a “cold thermal refuge”, inside which Planktothrix can grow without chytrid infection. A field study in two Norwegian lakes underlined the ecological significance of this finding. The study utilized sediment DNA as a biological archive in combination with existing monitoring data. In one lake, temperature and light conditions forced Planktothrix outside the thermal refuge for most of the growing season. This probably resulted in Red Queen dynamics as suggested by a high parasitic pressure exerted by chytrids, an increase in Planktothrix genotype diversity over time, and a correlation between Planktothrix genotype diversity and duration of bloom events. In the second lake, a colder climate allowed Planktothrix to largely stay inside the thermal refuge. The parasitic pressure exerted by chytrids and Planktothrix genotype diversity remained low, indicating that Planktothrix successfully evaded the Red Queen dynamics. Episodic Planktothrix blooms were observed during spring and autumn circulation, in the metalimnion or under the ice. Interestingly, both lakes were dominated by the same or related Planktothrix genotypes. Taken together, our data suggest that, depending on environmental conditions, chytrid parasitism can impose distinct selection regimes on conspecific phytoplankton populations with similar genotype composition, causing these populations to behave and perhaps to evolve differently.  相似文献

Phomopsis and related taxa comprise important endophytic and plant pathogenic species, and are known for the production of a diverse array of secondary metabolites. Species concepts within this group based on morphological characters and assumed host specificity do not reflect phylogenetic affinities. Additional phenotypic characters, such as profiles of secondary metabolites, are needed for practical species recognition. We investigated 36 strains of Phomopsis spp. and Cytospora-like fungi, obtained as endophytes of different host plants in Brazil, using metabolite profiling based on HPLC-UV/liquid chromatography -mass spectrometry (LC-MS) combined with cluster analysis of the results. Strains were also subjected to phylogenetic analyses based on internal transcribed spacer (ITS) rDNA. Six chemotypes were identified. Chemotypes 1-5 contained Phomopsis strains, while Cytospora-like strains formed the chemotype 6. Strains of chemotype 1 typically produced alternariols, altenusin, altenuene, cytosporones, and dothiorelones. Alternariol and seven unknown compounds were consistently produced by strains of chemotype 2. Members of chemotypes 3-5 produced poor metabolite profiles containing few chemical markers. Cytospora-like endophytes (chemotype 6) produced a characteristic set of metabolites including cytosporones and dothiorelones. Bayesian and Maximum Parsimony (MP) trees classified strains of each chemotype into single phylogenetic lineages or closely related groups. Strains of chemotypes 1 and 2 formed a monophyletic group along with Diaporthe neotheicola. The remaining Phomopsis strains formed monophyletic (chemotype 4) or polyphyletic (chemotypes 3 and 5) lineages inside a large and well supported clade. Cytospora-like strains formed a monophyletic lineage located at an intermediary position between Diaporthe/Phomopsis and Valsa/Cytospora clades. The combined results show that the production of secondary metabolites by Phomopsis and related Diaporthales may be species-specific, giving support to the use of metabolite profiling and chemical classification for phenotypic recognition and delimitation of species.  相似文献

In populations of Planktothrix, microcystin-producers and non-producers, which are morphologically identical, coexist. In order to develop a basis for the reliable detection of microcystin producers in field samples with polymerase chain reaction (PCR) based methods, we studied the presence and variability of eight regions of the mcy gene cluster in 46 Planktothrix strains, including both microcystin-producing and non-producing ones. PCR-amplification products for two mcy gene regions were also found in non-microcystin-producing strains, indicating the existence of natural mutants. PCR-products of the other regions studied were only detected in microcystin-producing strains. Two of these mcy-amplicons were variable in sequence and length. Four gene regions remained that were conserved and specific for microcystin-producing Planktothrix strains, and thus qualified to detect the respective chemotypes in environmental samples.  相似文献

Experimental studies of the evolutionary biology of lichen fungi have been hampered by massive difficulties of in vitro culture and artificial crosses are still not possible. Gene flow in these organisms is demonstrated here for the first time by the analysis of secondary products in the progeny of individuals from natural populations of mixed chemotypes of the Cladonia chlorophaea complex. All of the chemotypes in this study have been interpreted as distinct sibling species. In the Appalachian Mountains of North Carolina, however, the grayi and merochlorophaea chemotypes are found to belong to a single interbreeding populations that is reproductively isolated from the cryptochlorophaea chemotype. In the Coastal Plain, the cryptochlorophaea chemotype hybridizes with the local endemic perlomera chemotype. This study has major impact for species concept in lichens because consideration of neither morphologial tendencies nor biogenetic relationships of the secondary products could have predicted its result.  相似文献

Previous studies into the population structure of Melaleuca alternifolia by both isozyme and microsatellite analysis revealed little evidence for genetic structuring within genetic provenances. In contrast, analysis of the oil composition within these same regions showed distinct clustering of chemotypes within the provenances suggesting either that chemotype was not under genetic control, or that there is strong environmental selection for plant chemotypes. To investigate the level of genetic control of monoterpene composition in the essential oil of M. alternifolia, individuals representing the three extreme chemotypes of high terpinen-4-ol, high 1,8-cineole and high terpinolene were crossed with an individual with the commercially desirable high terpinen-4-ol oil profile. The progeny resulting from these crosses displayed oil profiles that were intermediate to that of the parent. Further analysis of the survey of oil chemotypes within the natural population also suggests that these intermediate chemotypes may arise naturally between regions containing high proportions of the extreme chemotypes. These results imply that there is a level of genetic structure for chemotype determination within the genetic provenance that is undetected by isozyme and microsatellite analysis. This information could play a vital role in the selection of appropriate genetic material to be used in future essential oil selection and breeding programs.  相似文献

The Plankthotrix Anagn. et Komárek population in the mesotrophic Lake Steinsfjorden has been intensively studied over several decades. This Planktothrix population produces a number of different classes of oligopetides. However, over the study period, only four main oligopeptide profiles (chemotypes) have been associated with the strains isolated from the lake. The chemotypes show distinct interactions with the environment, demonstrated by shifts in abundance along time series and vertical profiles. Here, we present genetic analysis of nonribosomal peptide synthetase (NRPS) gene regions in strains representing the four Planktothrix chemotypes in Lake Steinsfjorden. On the basis of phylogenetic analyses, we show that the NRPS genes for microcystin (mcy) and cyanopeptolin (oci) display the same clustering as do the chemotypes. Nucleotide diversity in mcy and oci was significantly higher between strains of different chemotypes than between strains of the same chemotype. Ka/Ks (nonsynonymous vs. synonymous mutations) values indicated positive selection in several polymorphic regions of the mcy and oci genes. Notably, incongruence between the phylogenetic trees for different gene segments and split decomposition analyses for segments of oci suggested horizontal gene transfer (HGT) events between strains showing different oligopeptide profiles. The oci HGT region encodes a module responsible for incorporating a variable amino acid in cyanopeptolin and is one of the regions suggested to be under positive selection. Taken together, our data suggest that there are four genetically distinct sympatric subpopulations—displayed as distinct chemotypes—in Lake Steinsfjorden. The diversification process of the chemotypes, and consequently the subpopulations, is driven by HGT and reinforced by positive selection of the corresponding NRPS gene regions.  相似文献

The sensitivity of smooth and rough mutants ofSalmonella typhimurium belonging to different chemotypes to bactericidal and bacteriolytic action of antibodies, complement and lysozyme was investigated. By the action of specific antibodies and complement the majority of strains was killed and in the presence of sufficient amount of lysozyme even lysed. Sera of newborn piglet which do not contain antibodies however, were able to kill regularly only strains of lower chemotypes (Rc and Rb). Smooth strains were usually resistant to the action of piglet serum whereas strains of Ra chemotype were killed only by sera of some piglets. Using the liver clearance method a significant correlation between the lipopolysaccharide chemotype of the particular strain and the degree of phagocytosis was found.  相似文献

Witte et al. (1992) described two distinct chemotypes of Senecio jacobaea L. Asteraceae, a chemotype with jacobine as one of the major pyrrolizidine alkaloids (PAs) and a chemotype with erucifoline as one of the major PAs. We hypothesized that the presence of erucifoline might be the factor responsible for the lack of success of the cinnabar moth on Senecio erucifolius L. Asteraceae and the S. jacobaea erucifoline chemotype. We performed a survey of the distribution of the two chemotypes in the Netherlands and compared this with the distribution map of Tyria jacobaeae L. Lepidoptera, Arctiidae. The distribution of the two chemotypes in the Netherlands is poorly correlated with the distribution of the cinnabar moth. The jacobine chemotype occurs along the coast and the erucifoline chemotype predominantly inward.An oviposition experiment showed that the cinnabar moth did not discriminate between the two chemotypes of S. jacobaea and S. erucifolius. Larval performance did not differ between the two chemotypes and species. Although the distribution of S. jacobaea jacobine chemotype is loosely associated with the abundance of the cinnabar moth the oviposition and growth experiments indicate that other factors than the presence of erucifoline play a role in this association.The absence of recordings of S. erucifolius as a foodplant for the cinnabar moth might be explained by the phenology of the foodplant. Ovipositing females of the univoltine cinnabar moth prefer flowering plants for oviposition. S. erucifolius starts flowering about 1–2 month later than S. jacobaea just after the peak density of moths.  相似文献

The contents of five ginsenosides (Rg1, Re, Rb1, Rc and Rd) were measured in American ginseng roots collected from 10 populations grown in Maryland. Ginsenoside contents and compositions varied significantly among populations and protopanaxatriol (Rg1 and Re) ginsenosides were inversely correlated within root samples and among populations. The most abundant ginsenoside within a root and by population was either Rg1 or Re, followed by Rb1. Ginseng populations surveyed grouped into two chemotypes based on the relative compositions of Rg1 and Re. Four populations, including the control population in which plants were grown from TN and WI seed sources, contained roots with the recognized chemotype for American ginseng of low Rg1 composition relative to Re. The remaining 6 populations possessed roots with a distinctive chemotype of high relative Rg1 to Re compositions. Chemotype did not vary by production type (wild versus cultivated) and roots within a population rarely exhibited chemotypes different from the overall population chemotype. These results provide support for recent evidence that relative Rg1 to Re ginsenoside contents in American ginseng roots vary by region and that these differences are likely influenced more by genotype than environmental factors. Because the physiological and medicinal effects of different ginsenosides differ and can even be oppositional, our findings indicate the need for fingerprinting ginseng samples for regulation and recommended usage. Also, the High Rg1/Low Re chemotype discovered in MD could potentially be used therapeutically for coronary health based on recent evidence of the positive effects of Rg1 on vascular growth.  相似文献

For Thymus pulegioides L. (Lamiaceae), occurring in almost entire Europe, about 20 different essential oil chemotypes are described in approximately 25 studies. However, only few studies mention chemotype diversity on population level, describing up to five chemotypes growing together. The aim of the study was to investigate the chemotype diversity within one T. pulegioides population of a limestone grassland in Germany. Essential oil compounds from samples of 93 thyme cushions were extracted with solid phase extraction (SPE) and analysed by GC-MS. Cluster analysis and nonmetric multi-dimensional scaling (NMDS) of the samples’ essential oil composition revealed the existence of seven different chemotypes: linalyl acetate-chemotype (56 individuals), geraniol-citral-linalyl acetate-chemotype (5), geraniol-citral-chemotype (7), thymol-chemotype (8), carvacrol-chemotype (2), linalool-chemotype (1), and β-caryophyllene-germacrene D-β-bisabolene-chemotype (14). The pattern of major monoterpenes from all chemotypes could be explained by the inhibition of specific steps in monoterpene biosynthesis. The sesquiterpene-dominant chemotype might be caused by a block in a very early step of monoterpene production or a regulatory/channeling mechanism. While the geraniol-citral-linalyl acetate-chemotype has been found for the first time, the other chemotypes have been found in other regions and the β-caryophyllene-germacrene D-β-bisabolene-type is similar to other sesquiterpene chemotypes rarely found in other areas. The large intra-population chemical polymorphism in this study raises questions about the chemotype diversity of thyme in other regions of Germany and potential correlations between essential oil composition and abiotic factors or biotic interactions.  相似文献

Fungal parasitism is recurrent in plankton communities, especially in the form of parasitic chytrids. However, few attempts have been made to study the community structure and activity of parasites at the natural community level. To analyse the dynamics of zoosporic fungal parasites (i.e. chytrids) of phytoplankton, samples were collected from February to December 2007 in two freshwater lakes. Infective chytrids were omnipresent in lakes, with higher diversity of parasites and infected phytoplankton than in previous studies. The abundance and biomass of parasites were significantly higher in the productive Lake Aydat than in the oligomesotrophic Lake Pavin, while the infection prevalence in both lakes were similar and averaged about 20%. The host species composition and their size appeared as critical for chytrid infectivity, the larger hosts being more vulnerable, including pennate diatoms and desmids in both lakes. The highest prevalence (98%) was noted for the autumn bloom of the cyanobacterium Anabaena flosaquae facing the parasite Rhizosiphon crassum in Lake Aydat. Because parasites killed their hosts, this implies that cyanobacterial blooms, and other large size inedible phytoplankton blooms as well, may not totally represent trophic bottlenecks because their zoosporic parasites can release dissolved substrates for microbial processes through host destruction, and provide energetic particles as zoospores for grazers. Overall, we conclude that the parasitism by zoosporic fungi represents an important ecological driving force in the food web dynamics of aquatic ecosystems, and infer general empirical models on chytrid seasonality and trophodynamics in lakes.  相似文献

The composition and yield of oil in 615 trees representing the natural populations of Melaleuca alternifolia, or tea tree, was investigated. A sixth distinct oil chemotype was identified. Of the six chemotypes, one chemotype is dominated by terpinen-4-ol, one by 1,8-cineole, one by terpinolene and the remaining three chemotypes are all dominated by 1,8-cineole and differ in either terpinen-4-ol or terpinolene content. Whilst most chemotypes are present throughout the distribution range, a definite correspondence of oil types with geographic location was found. Terpinen-4-ol types predominate in and around the Bungawalbin basin in the Casino area of northern New South Wales (NSW), high 1,8-cineole types predominate toward the southern end of the distribution around Grafton and terpinolene types predominate in southern Queensland. Preliminary formulae have been developed to allow comparisons of oil data obtained by steam distillation with a static headspace gas chromatography method.  相似文献

Further detailed experiments under laboratory conditions havebeen carried out using clones of Fragilaria crotonensis Kitton.Two morphologically distinct types (rod- and flared-cell forms)were investigated in relation to their infection by a chytridreferred to as species 3. The pathogenicity of the chytrid Rhizophydiumfragilariae Canter towards four newly isolated clones of F.crotonensis was also studied. The results in general confirmedthat clones of the diatom composed of flared-type cells werecompatible with the development and increase of species 3 butremained more or less resistant to R. fragilariae. In contrast,rod-celled type clones showed exactly the opposite host parasiterelationships. Further study has shown that slight differencesin susceptibility towards a particular chytrid can occur withindiatom clones of similar morphological type. The presence ofhypersensitive algal cells was noted in relation to parasitismby both fungi. Some observations are recorded on the developmentof mucilage around filaments of the Fragilaria clones. The ability of cyanobacteria to produce toxins and other secondary metabolites is patchily distributed in natural populations, enabling the use of cellular oligopeptide compositions as markers to classify strains into ecologically-relevant chemotypical subpopulations. The composition and spatiotemporal distribution of Microcystis chemotypes within and among waterbodies was studied at different time scales by analyzing (i) Microcystis strains isolated between 1998 and 2007 from different Spanish reservoirs and (ii) individual Microcystis aeruginosa colonies collected from pelagic and littoral habitats in Valmayor reservoir (Spain) during a bloom. No agreement between chemotypes and both morphotypes and genotypes (based on cpcBA-IGS, 16S–23S rRNA ITS and mcyB genes) was found, suggesting that oligopeptide profiles in individual strains evolve independently across morphospecies and phylogenetic genotypes, and that the diversity of microcystin variants produced cannot be explained by mcyB gene variations alone. The presence of identical chemotypes in spatially-distant reservoirs with dissimilar trophic state, lithology or depth indicate that waterbody characteristics and geographical boundaries weakly affect chemotype composition and distribution. At smaller spatiotemporal scales (i.e. during bloom), M. aeruginosa populations showed high number of chemotypes, as well as marked differences in chemotype composition and relative abundance among the littoral and pelagic habitats. This indicates that the factors influencing chemotype composition, relative abundance and dynamics operate at short spatial and temporal scales, and supports emerging hypotheses about interactions with antagonistic microorganisms as possible drivers for widespread chemical polymorphisms in cyanobacteria.  相似文献

We studied the variation in pyrrolizidine alkaloid (PA) patterns of lab-grown vegetative plants of 11 European Senecio jacobaea populations. Plants were classified as jacobine, erucifoline, mixed or senecionine chemotypes based on presence and absence of the PAs jacobine or erucifoline. Due to the presence of jacobine, total PA concentration in jacobine chemotypes was higher than in erucifoline chemotypes. Both relative and absolute concentrations of individual PAs differed between half-sib and clonal families, which showed that variation in PA patterns had a genetic basis. Within most populations relative abundance of PAs varied considerably between individual plants. Most populations consisted either of the jacobine chemotype or of the erucifoline chemotype, sometimes in combination with mixed or senecionine chemotypes.  相似文献

The authors studied antigens obtained by Grasset's method from 13 strains of Citrobacter of the International collection. The strains possessed O- and H-antigens whose behaviur in the electric field differed. All the strains under study were divided into two groups (by the number of serologically-active components of their O-antigens); representatives of the second group had no cathode O-antigen component. Chemical composition of specific lipopolysaccharides (LPS) obtained by Westphal's method was determined. Fourteen different sugars were revealed. The strains under study were referred to the known chemotypes. Strain 16/52 (8a, 8c) was for the first time studied in respect to the monosaccharide composition of specific LPS, and was referred to chemotype designated as CC-L.  相似文献

A parasitic wasp, Aenasius bambawalei, was studied for its biological parameters and parasitism levels in the cotton mealybug (Phenacoccus solenopsis). Biological parameters including parasitism efficiency, time to pupation, time to eclosion and adult sex ratio were studied under lab conditions. Parasitism levels in field collected mealybug were determined using PCR. Results showed an increase in parasitism over the study period, with higher parasitism levels in 2009 compared to the preceding 2 years.  相似文献

Although the cosmopolitan marine bryozoan Bugula neritina is recognized as a single species, natural products from this bryozoan vary among populations. B. neritina is the source of the anticancer drug candidate bryostatin 1, but it also produces other bryostatins, and different populations contain different bryostatins. We defined two chemotypes on the basis of previous studies: chemotype O contains bryostatins with an octa-2,4-dienoate substituent (including bryostatin 1), as well as other bryostatins; chemotype M lacks bryostatins with the octa-2,4-dienoate substituent. B. neritina contains a symbiotic gamma-proteobacterium "Candidatus Endobugula sertula," and it has been proposed that bryostatins may be synthesized by bacterial symbionts. In this study, B. neritina populations along the California coast were sampled for genetic variation and bryostatin content. Colonies that differ in chemotype also differ genetically by 8% in the mitochondrial cytochrome c oxidase subunit 1 (CO I) gene; this difference is sufficient to suggest that the chemotypes represent different species. Each species contains a distinct strain of "E. sertula" that differs at four nucleotide sites in the small subunit ribosomal RNA (SSU rRNA) gene. These results indicate that the chemotypes have a genetic basis rather than an environmental cause. Gene sequences from an Atlantic sample matched sequences from the California chemotype M colonies, suggesting that this type may be cosmopolitan due to transport on boat hulls.  相似文献

A large number of Fusarium isolates was collected from blighted wheat spikes originating from 175 sampling sites, covering 15 provinces in China. Species and trichothecene chemotype determination by multilocus genotyping (MLGT) indicated that F. graminearum s. str. with the 15-acetyl deoxynivalenol (15ADON) chemotype and F. asiaticum with either the nivalenol (NIV) or the 3-acetyl deoxynivalenol (3ADON) chemotype were the dominant causal agents. Bayesian model-based clustering with allele data obtained with 12 variable number of tandem repeats (VNTR) markers, detected three genetic clusters that also show distinct chemotypes. High levels of population genetic differentiation and low levels of effective number of migrants were observed between these three clusters. Additional genotypic analyses revealed that F. graminearum s. str. and F. asiaticum are sympatric. In addition, composition analysis of these clusters indicated a biased gene flow from 3ADON to NIV producers in F. asiaticum. In phenotypic analyses, F. asiaticum that produce 3ADON revealed significant advantages over F. asiaticum that produce NIV in pathogenicity, growth rate, fecundity, conidial length, trichothecene accumulation and resistance to benzimidazole. These results suggest that natural selection drives the spread of a more vigorous, more toxigenic pathogen population which also shows higher levels of fungicide resistance.  相似文献
