Influence of biological and ecological factors on hematological values in wild Little Penguins, Eudyptula minor

Clinical hematology and biochemistry are recognized as useful aids in health diagnosis in birds. The Little Penguin (Eudyptula minor) is endemic to Australia and numbers are declining due to a number of factors including declining health. This 2-year study on both Bowen and Lion Island populations aimed at assessing the potential causes of their declining health. Blood was collected from a total of 294 adult Little Penguins. R.B.C., Hb, P.C.V., M.C.V., M.C.H., M.C.H.C., W.B.C. and differential counts, thrombocytes and T.P.P. were measured. Multiple regression was used to identify relationships between hematological values and the following predictor variables: site, season, sex, time held prior to sampling and body condition. Values obtained showed some differences compared to the values for other penguin species reported in the literature. P.C.V., R.B.C., Hb, M.C.H.C. and T.P.P. showed major differences in comparison with a previous study on little penguins. Considering the influence of predictor variables, it appeared that: P.C.V., R.B.C., M.C.V., M.C.H., M.C.H.C. varied according to seasons; P.C.V., R.B.C., M.C.H., T.P.P. were correlated to body condition; P.C.V. was higher in birds on Bowen Island compared to birds on Lion Island; and T.P.P. was higher in females. These ecological and biological predictor variables have a strong influence upon hematological values. As a consequence, they must be taken into consideration when interpreting results of future studies.

Role of transmembrane segment M8 in the biogenesis and function of yeast plasma-membrane H(+)-ATPase

Of the four transmembrane helices (M4, M5, M6, and M8) that pack together to form the ion-binding sites of P(2)-type ATPases, M8 has until now received the least attention. The present study has used alanine-scanning mutagenesis to map structure-function relationships throughout M8 of the yeast plasma-membrane H(+)-ATPase. Mutant forms of the ATPase were expressed in secretory vesicles and at the plasma membrane for measurements of ATP hydrolysis and ATP-dependent H(+) pumping. In secretory vesicles, Ala substitutions at a cluster of four positions near the extracytoplasmic end of M8 led to partial uncoupling of H(+) transport from ATP hydrolysis, while substitution of Ser-800 (close to the middle of M8) by Ala increased the apparent stoichiometry of H(+) transport. A similar increase has previously been reported following the substitution of Glu-803 by Gln (Petrov, V. et al., J. Biol. Chem. 275:15709-15718, 2000) at a position known to contribute directly to Ca(2+) binding in the Ca(2+)-ATPase of sarcoplasmic reticulum (Toyoshima, C., et al., Nature 405: 647-655, 2000). Four other mutations in M8 interfered with H(+)-ATPase folding and trafficking to the plasma membrane; based on homology modeling, they occupy positions that appear important for the proper bundling of M8 with M5, M6, M7, and M10. Taken together, these results point to a key role for M8 in the biogenesis, stability, and physiological functioning of the H(+)-ATPase.

Biosynthesis of lasalocid A: biochemical mechanism for assembly of the carbon framework

Labeling experiments on the biosynthesis of the polyether antibiotic lasalocid A (1) using carboxylic acid precursors bearing 13C, 2H, and 3H labels at various positions established the following: (1) 2H or 3H at C-2 of propionate or 2H at C-2 of butyrate was partially retained at C-12 and C-14 of 1, respectively. (2) 2H at C-2 of propionate or at C-2 and C-3 of succinate did not label C-10. These and earlier data [Hutchinson, C. R., Sherman, M. M., Vederas, J. C., & Nakashima, T. T. (1981) J. Am. Chem. Soc. 103, 5956] are consistent with a hypothesis for the stereochemical control of lasalocid A biosynthesis, whose main tenets are that the configuration of C-12 and C-14 is determined by the stereoselectivity of the carbon chain forming condensation between acyl thio ester and 2-carboxyacyl thio ester intermediates and that the configuration of C-11 and C-15 results from the reduction of 2-keto thio ester intermediates with opposing stereospecificities.

Analyses of protein levels in these failing hearts reveal that the SR Ca-ATPase is down-regulated on average by 50% and that the Na/Ca exchanger is up-regulated on average by a factor of two. In this paper, we test the hypothesis that this altered pattern of expression of Ca handling proteins is sufficient to account for changes in excitation-contraction coupling properties measured experimentally at the cellular level. To do this, we present an integrated model of excitation-contraction coupling in the guinea pig ventricular cell. The model is used to determine the effects of SR Ca-ATPase down-regulation and Na/Ca exchanger up-regulation on action potential duration, Ca transient shape and amplitude, and isometric force. Model analyses demonstrate that changes in Ca handling proteins play a direct and critical role in prolongation of action potential duration, and in reduction of contractile force in heart failure.
