BACKGROUND: In the cortical region of motile cells, the actin network rapidly reorganizes as required for movement in various directions and for cell-to-substrate adhesion. The analysis of actin network dynamics requires the combination of high-resolution imaging with a specific fluorescent probe that highlights the filamentous actin structures in live cells. RESULTS: Combining total internal reflection fluorescence (TIRF) microscopy with a method for labeling actin filaments, we analyze the dynamics of actin patterns in the highly motile cells of Dictyostelium. A rapidly restructured network of single or bundled actin filaments provides a scaffold for the assembly of differentiated actin complexes. Recruitment of the Arp2/3 complex characterizes stationary foci with a lifetime of 7-10 s and traveling waves. These structures are also formed in the absence of myosin-II. Arp2/3-actin assemblies similar to those driving the protrusion of a leading edge form freely at the inner face of the plasma membrane. CONCLUSIONS: The actin system of highly motile cells runs far from equilibrium and generates a multitude of patterns within a dynamic filamentous network. Traveling waves are the most complicated patterns based on recruitment of the Arp2/3 complex. They are governed by the propagated induction of actin polymerization. We hypothesize that the actin system autonomously generates primordia of specialized structures such as phagocytic cups or lamellipodia. These primordia would represent an activated state of the actin system and enable cells to respond within seconds to local stimuli by chemotaxis or phagocytic-cup formation.  相似文献

Studies were conducted in opposing gradients of oxygen and sulfide in microslide capillaries to (i) characterize the chemical microenvironment preferred by Candidatus Arcobacter sulfidicus, a highly motile, sulfur-oxidizing bacterium that produces sulfur in filamentous form, and (ii) to develop a model describing the mechanism of filamentous-sulfur formation. The highly motile microorganisms are microaerophilic, with swarms effectively aggregating within oxic-anoxic interfaces by exhibiting a chemotactic response. The position of the band was found to be largely independent of the sulfide concentration as it always formed at the oxic-anoxic interface. Flux calculations based on steady state gradients of oxygen and sulfide indicate that sulfide is incompletely oxidized to sulfur, in line with the formation of filamentous sulfur by these organisms. It is proposed that Candidatus Arcobacter sulfidicus effectively competes with other sulfur-oxidizing bacteria in the environment by being able to tolerate higher concentrations of hydrogen sulfide (1-2 mM) and by possessing the ability to grow at very low oxygen concentrations (1-10 muM). The formation of mat-like structures from filamentous sulfur appears to be a population mediated effort allowing these organisms to effectively colonize environments characterized by high sulfide, low oxygen and dynamic fluid movement.  相似文献

The motile properties of intermediate filament (IF) networks have been studied in living cells expressing vimentin tagged with green fluorescent protein (GFP-vimentin). In interphase and mitotic cells, GFP-vimentin is incorporated into the endogenous IF network, and accurately reports the behavior of IF. Time-lapse observations of interphase arrays of vimentin fibrils demonstrate that they are constantly changing their configurations in the absence of alterations in cell shape. Intersecting points of vimentin fibrils, or foci, frequently move towards or away from each other, indicating that the fibrils can lengthen or shorten. Fluorescence recovery after photobleaching shows that bleach zones across fibrils rapidly recover their fluorescence. During this recovery, bleached zones frequently move, indicating translocation of fibrils. Intriguingly, neighboring fibrils within a cell can exhibit different rates and directions of movement, and they often appear to extend or elongate into the peripheral regions of the cytoplasm. In these same regions, short filamentous structures are also seen actively translocating. All of these motile properties require energy, and the majority appear to be mediated by interactions of IF with microtubules and microfilaments.  相似文献

Freeze-fracture and thin-section methods were used to study tight junction formation between confluent H4-II-E hepatoma cells that were plated in monolayer culture in media with and without dexamethasone, a synthetic glucocorticoid. Three presumptive stages in the genesis of tight junctions were suggested by these studies: (1) “formation zones” (smooth P-fracture face ridges deficient in intramembranous particles), apparently matched across a partially reduced extracellular space, develop between adjacent cells; (2) linear strands and aggregates of 9–11 nm particles collect along the ridges of the formation zones. The extracellular space was always reduced when these structures were found matched with pits in gentle E-face depressions; (3) the linear arrays of particles on the ridges associate within the membranes to form the fibrils characteristic of mature tight junctions. The formation zones resemble tight junctions in terms of size, complexity and the patterns of membrane ridges. Although some of the beaded particle specialization may actually be gap junctions, it is unlikely that all can be interpreted in this way. No other membrane structures were detected that could represent developmental stages of tight junctions. Dexamethasone (at 2 × 10?6 M) apparently stimulated formation of tight junctions. Treated cultures had a greater number of formation zones and mature tight junctions, although no differences in qualitative features of the junctions were noted.  相似文献

In response to treatment with 17β-estradiol, MCF-7 human breast carcinoma cells undergo a marked rearrangement of the F-actin cytoskeleton. The most conspicuous aspect of this rearrangement is the formation of an extensive array of lamellipodial structures which are situated beneath cell clusters. Treatment of cells with 17β-estradiol in the presence of the anti-estrogen ICI182,780 suppressed the development of the lamellipodial structures, indicating that this cytoskeletal rearrangement is mediated by the estrogen receptor. Time-lapse, video-enhanced, differential interference contrast microscopy reveals that the lamellipodial structures are actively motile beneath cell clusters. Furthermore, the lamellipodial structures form few focal contacts with the underlying substrate of the coverslip, as evidenced by either interference reflection microscopy or staining for the focal contact protein talin, indicating that these structures are not strongly adhered to the substratum. Immunofluorescence localization of E-cadherin indicates that this cell–cell adhesion receptor is present within these structures as either adhesion plaque- or point contact-like depositions. These findings implicate the cadherin-based cell–cell adhesion system in supporting tumor cell motility over adjacent cell surfaces via discrete adhesive structures which are associated with motile lamellipodia. The organization and development of cell coverings in two alternate phases of the life cycle in a marine dinoflagellate, Scrippsiella hexapraecingula Horiguchi et Chihara, were investigated by thin sectioning and freeze‐fracture electron microscopy. In one of these phases, the motile phase, cells have an outermost plasma membrane that is lined with flattened amphiesmal vesicles. Groups of microtubules lie beneath these vesicles. In mature motile cells, thecal plates are completely enclosed in individual amphiesmal vesicles. After settling, the cells enter the second, non‐motile phase. Here, ecdysis occurs, resulting in several steps including formation of the first pellicle layer (PI), fusion of the inner amphiesmal vesicle membranes to form the new plasma membrane, deposition of the second pellicle layer (PM) under PI, and the appearance and fusion of juvenile amphiesmal vesicles to form new territories, which eventually give rise to new thecal plates in the next motile phase. Thus, the pattern in which thecal plates are arranged in motile cells is determined at the time when the amphiesmal vesicles develop into non‐motile cells.  相似文献

The human telomere is known to form the G-quadruplex structure to inhibit the activity of telomerase. Its detailed structure has been of great interest. Recently, two kinds of the parallel-antiparallel hybrid structures have been specified in K(+) solution. However, the G-quadruplex structure is generally thought to be in equilibrium among different structures. Here, we describe the single-pair fluorescence resonance energy transfer (sp-FRET) experiments on telomere samples with bromoguanine (BrG)-substitutions, which control the G-quadruplex structures, at different positions and one without any substitution. The observed FRET distributions were decomposed into five components and the relative population of these components depended on the BrG-substitution positions. In order to consistently explain the variety of conformations, we proposed a novel structural model, the so-called triple-strand-core model. On the basis of this model, the components of the FRET distributions were attributed to the mixed-chair hybrid structures, which were reported recently, and chair-type antiparallel structures, which can be predicted from this model. The FRET efficiencies of these structures were explained in terms of partially broken structures due to steric hindrance and inappropriate capping. This basic model also consistently explains experimental results reported previously. Furthermore, using this model, the folding pathway of the hybrid structures and T-loop formation can be predicted.  相似文献

During development, dynamic changes in the actin cytoskeleton determine both cell motility and morphological differentiation. In most mature tissues, cells are generally minimally motile and have morphologies specialized to their functions. In metastatic cancer, cells generally lose their specialized morphology and become motile. Therefore, proteins that regulate the transition between the motile and morphologically differentiated states can play important roles in determining cancer outcomes. AFAP120 is a neuronal-specific protein that binds Src kinase and protein kinase C (PKC) and cross-links actin filaments. Here we report that expression and tyrosine phosphorylation of AFAP120 are developmentally regulated in the cerebellum. In cerebellar cultures, PKC activation induces Src kinase-dependent phosphorylation of AFAP120, indicating that AFAP120 may be a downstream effector of Src. In neuroblastoma cells induced to differentiate by treatment with a PKC activator, tyrosine phosphorylation of AFAP120 appears to regulate the formation of the lamellar actin structures and subsequent neurite initiation. Together, these results indicate that AFAP120 plays a role in organizing dynamic actin structures during neuronal differentiation and suggest that AFAP120 may help regulate the transition from motile precursor to morphologically differentiated neurons.  相似文献

Abstract. We show that the anterior, prestalk region of the Dictyostelium slug contains cells which express, or have expressed, a prespore-specific marker. We term these cells "prespore-like cells" (PLC). In newly formed slugs there is a sharp prespore/prestalk boundary, with very few PLC, but after several days of migration the clear demarcation between prespore and prestalk zones breaks down because the number of PLC increases dramatically. This is consistent with previous observations showing there to be rapid interchange of cells between the prestalk and prespore regions. This is not, however, their only source, as a scattering of PLC appear when separate prestalk and prespore regions first become apparent at the time of tip formation. Also, at culmination, there is respecification of "prespore" cells at the pre-stalk/prespore boundary to form part of the mature stalk. The existence of these cells, and of PLC, may explain why we find prespore-specific mRNAs in mature stalk cells.  相似文献

A review of theoretical ideas on the physics of structurization instability of a homogeneous dusty plasma, i.e., the formation of zones with elevated and depressed density of dust grains and their arrangement into different structures observed in laboratory plasma under microgravity conditions, is presented. Theoretical models of compact dust structures that can form in the nonlinear stage of structurization instability, as well as models of a system of voids (both surrounding a compact structure and formed in the center of the structure), are discussed. Two types of structures with very different dimensions are possible, namely, those smaller or larger than the characteristic mean free path of ions in the plasma flow. Both of them are characterized by relatively regular distributions of dust grains; however, the first ones usually require external confinement, while the structures of the second type can be self-sustained (which is of particular interest). In this review, they are called dust clusters and self-organized dust structures, respectively. Both types of the structures are characterized by new physical processes that take place only in the presence of the dust component. The role of nonlinearities in the screening of highly charged dust grains that are often observed in modern laboratory experiments turns out to be great, but these nonlinearities have not received adequate study as of yet. Although structurization takes place upon both linear and nonlinear screening, it can be substantially different under laboratory and astrophysical conditions. Studies on the nonlinear screening of large charges in plasma began several decades ago; however, up to now, this effect was usually disregarded when interpreting the processes occurring in laboratory dusty plasma. One of the aims of the present review was to demonstrate the possibility of describing the nonlinear screening of individual grains and take it into account with the help of the basic equations for the equilibrium between plasma components when analyzing equilibrium structures. The effect of plasma screening nonlinearity on both the diffusion processes and the forces of dust drag by plasma fluxes is analyzed. It is shown how self-organized dust structures form in these processes. In the limit of very small dust grain charges, the forces acting on the dusty plasma components and the set of basic equations for stationary dust structures (with allowance for nonlinear screening) take a standard form. New qualitative effects, such as the suppression of diffusion due to ion scattering from dust grains and the formation of structures of different configurations, are described. A detailed comparison with previous results is performed. It is shown that the solution of basic nonlinear equations for dust structures yields new qualitative effects. A number of new effects to be studied in future dusty plasma experiments with the formation of structures in spherical chambers are predicted (it is assumed that diffusion will play a significant role under microgravity conditions). Recent ground-based experiments, as well as experiments carried out onboard the International Space Station, directly confirm the nonlinear character of screening and the significant role played by this nonlinearity in the structurization of dusty plasma. Experiments on the formation of structures consisting of smaller dust grains within structures formed of larger grains are discussed. It is shown that those experiments can be interpreted only using the concept of nonlinear screening.  相似文献

Male-killing bacteria are bacteria that are transmitted vertically through the females of their insect hosts. They can distort the sex ratio of their hosts by killing infected male offspring. In nature, male-killing endosymbionts (male killers) often have a 100% efficient vertical transmission, and multiple male-killing bacteria infecting a single population are observed. We use different model formalisms to study these observations. In mean-field models a male killer with perfect transmission drives the host population to extinction, and coexistence between multiple male killers within one population is impossible; however, in spatially explicit models, both phenomena are readily observed. We show how the spatial pattern formation underlies these results. In the case of high transmission efficiencies, waves with a high density of male killers alternate with waves of mainly wild-type hosts. The male killers cause local extinction, but this creates an opportunity for uninfected hosts to re-invade these areas. Spatial pattern formation also creates an opportunity for two male killers to coexist within one population: different strains create spatial regions that are qualitatively different; these areas then serve as different niches, making coexistence possible.  相似文献

Three types of population growth and development of chemotaxic motile bacteria Escherichia coli on semi-solid nutrient media are investigated: a) stable development – circular symmetrical waves; b) bursts; c) fractal-like self-organization. Experimental investigation of the burst formation is presented. The microscopic analysis of growing, fractal-like structures is carried out, and a mechanism for such structure formation is suggested. It is supposed that fractal-like bacterial structures growth is based on the principle of successively forming multiple micro-bursts. A mathematical model has been suggested to reproduce the experimental results. The structures obtained by numerical modeling of population growth in the parameter space substrate concentration - bacterial movement rate reproduce the corresponding experimental structures in the space nutrient concentration in the media – the density of the media.  相似文献

A new species of benthic marine dinoflagellate, Pyramidodinium spinulosum Horiguchi, Moriya, Pinto & Terada is described from the deep (36 m) seafloor off Mageshima Island, Kagoshima Prefecture, Japan in the subtropical region of the northwest Pacific. The life cycle of the dinoflagellate consists of a dominant, attached, dome‐shaped, vegetative form and short‐lasting, motile cell. Asexual reproduction takes place by the formation of two motile cells within each non‐motile cell. The released motile cells swim only for a short period and transform directly into the dome‐shaped vegetative form. The duration of the cell cycle varies and can be extremely long, ranging 5–38 days under culture conditions. The non‐motile cell is enclosed by a cell wall and its surface is covered with many (80 – 130) spines of various length. The dinoflagellate is photosynthetic and contains many (more than 50) discoidal chloroplasts. Phylogenetic analysis reveals that the dinoflagellate is closely related to the type species of the genus Pyramidodinium, P. atrofuscum which also possesses a dominant, attached, non‐motile form. However, P. spinulosum can be clearly distinguished from P. atrofuscum by the cell shape (dome‐shaped vs. pyramid‐shaped) and surface ornamentation (spines vs. wart‐like processes) of the non‐motile form. Based on these morphological differences together with molecular evidence, it was concluded that this organism from a deep water sand sample should be described as a second species of the genus Pyramidodinium, P. spinulosum.  相似文献

Gamma bodies, which are present in the sporangia and gametangia of Allomyces and in its spores, are interpreted as constituting vesicle generating structures. During spore cleavage the mobilization–decay of the gamma bodies leads to vesicle formation; the vesicles appear to fuse to form the axonemal and plasma membrane of the spore. Vesicle formation by the gamma bodies during spore cleavage can be perturbed by phosphate buffer which leads to the formation of myelin–figure arrays of membranes, or by colchicine and benomyl which give rise to large vacuolar structures after gamma body decay. During the motile period of the spores of Allomyces , mobilization of the gamma bodies leads to vacuole formation and the resulting vacuoles fuse with the plasma membrane of the spore and by this means maintain the osmotic balance of the spore. During spore encystment the gamma body decays and forms vesicles which fuse with the plasma membrane of the cyst; these vesicles appear to be instrumental in chitin wall synthesis.  相似文献

Two sorts of inverted micellar structures have previously been proposed to explain morphological and 31P-NMR observations of bilayer systems. These structures only form in systems with components that can adopt the inverse hexagonal (HII) phase. LIP (lipidic particles) are intrabilayer structures, whereas IMI (inverted micellar intermediates) are structures that form between apposed bilayers. Here, we calculate the formation rates and half-lives of these structures to determine which (or if either) of these proposed structures is a likely explanation of the data. Calculations for the egg phosphatidylethanolamine and the Ca+-cardiolipin systems show that IMI form orders of magnitude faster than LIP, which should form slowly, if at all. This result is probably true in general, and indicates that "lipidic particle" electron micrograph images probably represent interbilayer structures, as some have previously proposed. It is shown here that IMI are likely intermediates in the lamellar----HII phase transitions and in the process of membrane fusion in some systems. The calculated formation rates, half-lives, and vesicle-vesicle fusion rates are in agreement with this observation.  相似文献

It is shown that, during Perseid, Geminid, Orionid, and Leonid meteor showers, the excitation of low-frequency dust acoustic perturbations by modulational instability in the Earth’s ionosphere can lead to the generation of infrasonic waves. The processes accompanying the propagation of these waves are considered, and the possibility of observing the waves from the Earth’s surface is discussed, as well as the possible onset of acoustic gravitational vortex structures in the region of dust acoustic perturbations. The generation of such structures during Perseid, Geminid, Orionid, and Leonid meteor showers can show up as an increase in the intensity of green nightglow by an amount on the order of 10% and can be attributed to the formation of nonlinear (vortex) structures at altitudes of 110–120 km.  相似文献

Cells exhibit propagating membrane waves which involve the actin cytoskeleton. One type of such membranal waves are Circular Dorsal Ruffles (CDR) which are related to endocytosis and receptor internalization. Experimentally, CDRs have been associated with membrane bound activators of actin polymerization of concave shape. We present experimental evidence for the localization of convex membrane proteins in these structures, and their insensitivity to inhibition of myosin II contractility in immortalized mouse embryo fibroblasts cell cultures. These observations lead us to propose a theoretical model which explains the formation of these waves due to the interplay between complexes that contain activators of actin polymerization and membrane-bound curved proteins of both types of curvature (concave and convex). Our model predicts that the activity of both types of curved proteins is essential for sustaining propagating waves, which are abolished when one type of curved activator is removed. Within this model waves are initiated when the level of actin polymerization induced by the curved activators is higher than some threshold value, which allows the cell to control CDR formation. We demonstrate that the model can explain many features of CDRs, and give several testable predictions. This work demonstrates the importance of curved membrane proteins in organizing the actin cytoskeleton and cell shape.  相似文献

During early development migratory mesenchymal cells navigate to distant sites where they aggregate to form a variety of embryonic organ rudiments. We present here a new model for mesenchymal cell morphogenesis based on the mechanical interaction between motile cells and their extracellular environment. The model is based on two properties of motile cells: (a) they are capable of generating large traction forces which can deform the extracellular matrix through which they move, and (b) the deformations they produce in their environment affect the direction of their movements. We derive field equations which describe the motion of cells in an elastic extracellular matrix and show that these equations can generate a variety of spatial patterns, such as the formations of skin organ primordia, especially feather germs, cartilage condensation patterns which presage bone formation in limb development, and melanocyte density patterns which form animal coat patterns.Support for this work was provided by NSF Grant # MCS-8110557 [GFO]  相似文献

The neural crest is a migratory population of cells that produces many diverse structures within the embryo. Trunk neural crest cells give rise to such structures as the dorsal root ganglia (DRG) and sympathetic ganglia (SG), which form in a metameric pattern along the anterior-posterior axis of the embryo. While static analyses have provided invaluable information concerning the development of these structures, time-lapse imaging of neural crest cells navigating through their normal environment could potentially reveal previously unidentified cellular and molecular interactions integral to DRG and SG development. In this study, we follow fluorescently labeled trunk neural crest cells using a novel sagittal explant and time-lapse confocal microscopy. We show that along their dorsoventral migratory route, trunk neural crest cells are highly motile and interact extensively with neighboring cells and the environment, with many cells migrating in chain-like formations. Surprisingly, the segregated pattern of crest cell streams through the rostral somite is not maintained once these cells arrive alongside the dorsal aorta. Instead, neural crest cells disperse along the ventral outer border of the somite, interacting extensively with each other and their environment via dynamic extension and retraction of filopodia. Discrete sympathetic ganglia arise as a consequence of intermixing and selective reorganization of neural crest cells at the target site. The diverse cell migratory behaviors and active reorganization at the target suggest that cell-cell and cell-environment interactions are coordinated with dynamic molecular processes.  相似文献

Formation of terminal synapses at sites such as the neuromuscular junction involves transformation of the motile growth cone into the nonmotile synaptic terminal. However, transformation does not need to be the mechanism when a neurite forms multiple widely spaced synaptic varicosities along a target in an en passant configuration. Synaptic varicosities could form here by specialization of the neurite after the growth cone has advanced past the site. We examined this issue by using cocultures of identified sensory (SN) and motor (L7) neurons from Aplysia. Living SNs were labeled with fluorescent dye and their neurites were observed at high resolution every few minutes growing along the axon of L7, allowing a fine-grained analysis of the behavior of the growth cone at the sites of synapse formation. All varicosities whose formation was observed indeed developed from the growth cone. Sensory varicosities were shown by electron microscopy to contain features characteristic of active zones for transmitter release within a day of their formation on the motor axon. Growth cone advance slowed or stopped transiently during varicosity formation, but the motile activity of the peripheral region of the growth cone (veils and filopodia) was maintained. These results suggest that target "stop signals" involved in the formation of synapses, at least of the en passant variety, may be of a different type from the growth inhibitory molecules, such as the collapsins, which guide axons to their targets.  相似文献
