The changes in length on the two opposite sides of etiolated radish (Raphanus sativus) hypocotyls prior to, and following gravitropic stimulation, were measured using an infrared-imaging system. It was observed that the growth suppression on the upper side began first at least 10 min after the onset of gravitropic stimulation, and after 30 min the acceleration in growth on the lower side started. The gravitropic curvature was steadily induced from 10 min. When radish hypocotyls were switched from a vertical to horizontal position for different durations and then replaced to the vertical position, the growth suppression on the gravity-stimulated (upper) side was observed in all cases, but the acceleration in growth on the opposite (lower) side appeared only in continuously gravity-stimulated seedlings, although it occurred later than the growth suppression on the upper side. These results suggest that the suppression in growth on the upper side of the hypocotyls is a direct effect of gravitropic stimulation, but not the acceleration on the lower side. When 4-methylthio-3-butenyl isothiocyanate (4-MTBI), which has an inhibitory activity against radish hypocotyl growth, was applied on the one side of radish hypocotyls and then the 4-MTBI-applied side or opposite side was placed in a horizontal position, the former showed greater bending than the control, suggesting that the growth suppression on the upper side is enhanced and maintained with MTBI application there. In the latter case, the seedlings showed less bending than the control, suggesting a decrease in growth on the lower side with MTBI application. All the results suggest that gravitropism of radish hypocotyls may be caused by an increase in growth-inhibiting substance(s) induced with gravitropic stimulation in the upper side, inducing growth inhibition there.  相似文献

The variation of indole-3-acetic acid (IAA) transport along Lupinus albus L. hypocotyls was studied using decapitated seedlings and excised sections. To confirm that the mobile species was IAA and not IAA metabolites, dual isotope-labeled IAAs, [5-3H]IAA + [1-14C]IAA, were used. After apical application to decapitated seedlings, the longitudinal distribution of both isotopes at different transport periods showed that the velocity of IAA transport was higher in the apical, elongating region than in the basal, non-growing region. This variation in velocity was not a traumatic consequence of decapitation because after application of IAA to the basal region of decapitated seedlings, both the velocity and intensity of IAA transport were lower than in the apical treatment. The variation in IAA transport down the hypocotyl was confirmed when it was measured in excised sections located at different positions along the hypocotyl. The velocity and, to a greater extent, the intensity of IAA transport decreased from the apical to the basal sections. Consequently, if the amount of IAA reaching the apical zones of lupin hypocotyl were higher than the IAA transport capacity in the basal zones, accumulation of mobile IAA might be expected in zones located above the basal region. In fact, an IAA accumulation occurred in the elongating region during the first 4-h period of transport after apical treatment with IAA. It is proposed that the fall in IAA transport along the hypocotyl might be responsible for the IAA distribution and, consequently, for the growth distribution reported in this organ. An indirect proof of this was obtained from experiments that showed that the excision of the slowly transporting basal zones strongly reduced the growth in the remaining part of the organ, whereas excision of the root caused no significant modification in growth during a 20-h period.  相似文献

Abstract. The growth response of etiolated cucumber ( Cucumis sativus L.) hypocotyls to gravitropic stimulation was examined by means of time-lapse photography and high-resolution analysis of surface expansion and curvature. In comparison with video analysis, the technique described here has five- to 20-fold better resolution; moreover, the mathematical fitting method (cubic splines) allows direct estimation of local and integrated curvature. After switching seedlings from a vertical to horizontal position, both upper and lower surfaces of the stem reacted after a lag of about 11 min with a two- to three-fold increase in surface expansion rate on the lower side and a cessation of expansion, or slight compression, on the upper surface. This growth asymmetry was initiated simultancously along the length of the hypocotyl, on both upper and lower surfaces, and did not migrate basipetally from the apex. Later stages in the gravitropic response involved a complex reversal of the growth asymmetry, with the net result being a basipetal migration of the curved region. This secondary growth reversal may reflect oscillatory and or self-regulatory behaviour of growing cells. With some qualifications, the kinetics and pattern of growth response are consistent with a mechanism involving hormone redistribution, although they do not prove such a mechanism. The growth kinetics require a growth mechanism which can be stimulated by two-to three-fold or completely inhibited within a few minutes.  相似文献

Abstract. A system is described whereby seedling development can be analysed in terms of growth rates of specific 1 mm regions of the hypocotyl. The technique involves time-lapse photography of marked hypocotyls in a specially designed chamber which accommodates seedlings in various orientations with respect to gravity, and under irradiation regimes differing in light quality, quantity and direction. The results of a preliminary study of the upward growth of etiolated or green cress seedlings in darkness or overhead while light are reported. Highest growth rates in etiolated seedlings were observed in zones in the upper one-third of ihe hypocotyl. In green seedlings, growth was more prominent within the subapical zones. Light further restricted growth of the median and basal zones in both types of seedling. However, in their immediate responses to the onset of irradiation, green and etiolated seedlings differed markedly. In etiolated seedlings, recovery of growth at the apex was accompanied by the development of inhibition in the median-basal regions; green seedlings showed a transient inhibition of growth in the apical zone together with a strong immediate inhibition in the median-basal regions.  相似文献

Both temperature and soil moisture vary greatly in the surface layers of the soil through which seedlings grow following germination. The work presented studied the impact of these environmental variables on post-germination carrot growth to nominal seedling emergence. The rapid pre-crook downward growth of both the hypocotyl and root was consistent with their requirement for establishment in soil drying from the surface. At all temperatures, both hypocotyl and root growth rates decreased as water stress increased and there was a very distinct temperature optimum that tended to occur at lower temperatures as water stress increased. A model based on the thermodynamics of reversible protein denaturation was adapted to include the effects of water potential in order to describe these growth rate responses. In general, the percentage of seedlings that reached the crook stage (start of upward hypocotyl growth) decreased at the extremes of the temperature range used and was progressively reduced by increasing water stress. A model was developed to describe this response based on the idea that each seedling within a population has lower and upper temperature thresholds and a water potential threshold which define the conditions within which it is able to grow. This threshold modelling approach which applies growth rates within a distribution of temperature and water potential thresholds could be used to simulate seedling growth by dividing time into suitable units.  相似文献

Photosynthetic CO2 uptake rate and early growth parameters of radish Raphanus sativus L. seedlings exposed to an extremely low frequency magnetic field (ELF MF) were investigated. Radish seedlings were exposed to a 60 Hz, 50 microT(rms) (root mean square) sinusoidal magnetic field (MF) and a parallel 48 microT static MF for 6 or 15 d immediately after germination. Control seedlings were exposed to the ambient MF but not the ELF MF. The CO2 uptake rate of ELF MF exposed seedlings on day 5 and later was lower than that of the control seedlings. The dry weight and the cotyledon area of ELF MF exposed seedlings on day 6 and the fresh weight, the dry weight and the leaf area of ELF MF exposed seedlings on day 15 were significantly lower than those of the control seedlings, respectively. In another experiment, radish seedlings were grown without ELF MF exposure for 14 d immediately after germination, and then exposed to the ELF MF for about 2 h, and the photosynthetic CO2 uptake rate was measured during the short-term ELF MF exposure. The CO2 uptake rate of the same seedlings was subsequently measured in the ambient MF (control) without the ELF MF. There was no difference in the CO2 uptake rate of seedlings exposed to the ELF MF or the ambient MF. These results indicate that continuous exposure to 60 Hz, 50 microT(rms) sinusoidal MF with a parallel 48 microT static MF affects the early growth of radish seedlings, but the effect is not so severe that modification of photosynthetic CO2 uptake can observed during short-term MF exposure.  相似文献

Abstract. The patterns of growth in organs curving under the influence of gravity were analysed by time-lapse photography of cress and cucumber hypocotyls which were delimited into 1 mm zones by ion-exchange beads. Geotropic curvature resulted from changes in growth rate on both sides of the organ. Growth inhibition of varying degrees of intensity occurred in all the previously growing zones of the upper (concave) side. An absolute reduction in length due to compression frequently occurred in some zones. Also, in both species growth stimulation was observed on the lower (convex) side. The disparity in growth rate between the upper and lower surfaces varied with time, being more apparent in the subapical region in the first hour of curvature. A later promotion of growth rate on the lower surface subsequently increased the curvature of the more basal zones. Autotropic straightening occurred as a consequence of growth changes, both inhibitory and stimulatory, in the apical zones. These events indicate a polarity of response in which apical zones have precedence over basal zones.  相似文献

花青素是植物体内广泛存在的一类天然色素,具有重要的生理功能。花青素合成途径可受多种因素调控,其中植物生长激素赤霉素(gibberellic acid,GA)对其的调控作用报道较少。本文用不同浓度的赤霉素处理心里美萝卜幼苗,以探讨它对花青素含量的影响。结果表明,外源GA3处理显著增加了萝卜幼苗的下胚轴长度,并提高了下胚轴中α-淀粉酶活性;显著降低下胚轴中花青素的含量。1 μmol/L GA3处理效果较好;处理后第3 d和第5 d,花青素合成的关键酶查尔酮合酶、查尔酮异构酶和花青素还原酶编码基因的表达水平均低于对照组。同时,外源GA3显著诱导过氧化物酶活性的升高。上述结果表明,外源赤霉素可能通过下调花青素合成基因的表达,提高过氧化物酶活性和促进下胚轴伸长生长降低花青素的水平。  相似文献

The changes in osmotic potential and cell wall composition of hypocotyl cell walls from different hypocotyl regions were investigated during growth of etiolated seedlings of Pinus pinaster Aiton. The osmotic potential in the subapical 5 mm part was minimum when hypocotyl growth rate was low, and increased when the fast growth phase began. The main non-cellulosic sugars of the cell wall from pine hypocotyl were arabinose, galactose, xylose, glucose and uronic acids, although their relative proportions were different from those found for angiosperm cell walls. Non-cellulosic glucose was the sugar showing the most important changes during hypocotyl growth as well as along the hypocotyl, suggesting that a glucose-rich polysaccharide is involved in a very active turnover during growth. A partial degradation of a xyloglucan during growth is suggested.  相似文献

Phototropic sensitivity of forest wood seedlings to lateral illumination was proposed as an early assay for distinquishing various ecotypes of woody plants of the same species. Statistical analysis showed that results were significantly influenced by heterogenity of experimental material caused by an interference of phototropic movements and natural oscillations of hypocotyl. Both movements of pine seedlings (Pinus silvestris L.) were registered by phase photography and their mechanism was analyzed. The apical part of growing hypocotyl illuminated from above oscillates in a space spiral with frequency 3.3 h at mean growth rate 0.66 mm h?1. The mean size of spiral amplitudes is 2.9 mm. The oscillation rhythm is disturbed after the lateral illumination and a phase shift was observed as a result. A new oscillation rhythm with frequency 3.9 h and mean growth rate 0.69 mm h?1 was stabilized after a period of time equal to one nutation turn. Oscillation amplitudes were increased to 4.3 mm. In addition to the radiation intensity the appearance of the phototropic response to light pulse is first of all effected by the actual position of the apical part of hypocotyl in the course of endogenous circumnutations. A uniform plant material for the early assay may be obtained by selection of seedlings which are at the beginning of lateral illumination in the same phase of nutation rhythm. Under such conditions the deviations of longitudinal axis of oscillating spirals characterize the actual phototropic curvature.  相似文献

The variations in length and fresh and dry mass of etiolated hypocotyls of lupin during the growth have been studied. The growth exhibited by the different zones delimited along the hypocotyl was dependent on the localization of the zone as well as on the age of seedlings, but in both cases the pattern of growth was similar. During the period of growth studied (seedlings 7 to 21 d old), the growth of hypocotyl was basically due to cell elongation, since the relative elongation of cells was positively correlated with the relative elongation of the hypocotyl.  相似文献

Putrescine uptake and translocation were studied by feeding [3H] putrescine to roots of tomato seedlings ( Lycopersicon esculentum Miller, cv. Earlypak 7) at the stage of expanded cotyledons, of maize seedlings ( Zea mais L.) at the coleoptile stage, and of one year old pines ( Pinus pinea L.). Putrescine translocation was rapid as radioactivity appeared in the upper part of the seedlings within 30 min, continuing to increase up to 24 h, while it decreased in roots. The putrescine supplied was partly metabolized to spermidine and spermine in the course of 24 h. The transport was temperature-dependent as it increased with increasing temperature from 4°C to 30°C. In plants kept in 100% relative humidity the transport decreased by 27% compared to controls kept in 50% relative humidity. The existence of basipetal transport was assessed by feeding labeled putrescine to cotyledons or to a primary leaf of tomato plants at different stages of growth. The influence of ringing at the hypocotyl level on polyamine translocation in pine plants was studied in order to exclude cortical parenchyma and phloem from transport. Radioactivity decreased in the hypocotyl just above the ring and in the upper parts (epicotyls with needles), but long-distance transport was low affected indicating xylem transport. It is suggested that polyamine transport is not polar, and that it occurs mainly through xylem vessels.  相似文献

When flax seedlings are decapitated above cotyledons and three days later one of the two cotyledons is removed then the remaining cotyledon stimulates in four to five days growth of its axillary bud. It has been found that content of endogenous cytokinins was higher in the stimulated bud as compared with the other one already 12 h after the cotyledon removal. Flax seedlings decapitated under cotyledons regenerate adventitious buds on thy hypocotyl stump during 5–6 days. The endogenous fytohormonal preparation of this regeneration was investigated in the 20 mm apical part of the hypocotyl stump. Decrease in auxin and increase in gibberellins was already found during the first day after decapitation while the level of cytokinins increased as late as three days after the apex removal.  相似文献

Polar transport of kinetin-8-14C occurred in segments of petioles, hypocotyls, and roots of radish (Raphanus sativus L.). The polarity was basipetal in petioles and hypocotyls and acropetal in roots. In segments excised from seedlings with fully expanded cotyledons, indole-3-acetic acid was required for polarity to develop. In hypocotyl segments isolated at this stage, basipetal and acropetal movements were equal during the first 12 hours of auxin treatment after which time acropetal movement declined. Pretreatment with auxin eliminated this delay in the appearance of polarity. In hypocotyl segments excised from seedlings with expanding cotyledons, exogenous auxin was unnecessary for polarity. Potassium cyanide abolished polarity at both stages of growth by allowing increased acropetal movement. The rate of accumulation of kinetin in receiver blocks was greater than the in vivo increase in cytokinin content of developing radish roots.  相似文献

Abstract. The rate of curvature of etiolated cress ( Lepi-dium sativum L. ) hypocotyls in response to gravity (negative geotropism) was retarded by red or blue light; far-red irradiation was without effect. The timing of the irradiation period in relation to the presentation for geostimulus markedly affected the response. When seedlings were irradiated during the 1–2 h period of geostimulus, blue light was more effective than red at retarding curvature; when seedlings were irradiated prior to geostimulus, only red light affected geocurvature. These results are interpreted as a further example of the kinetically distinct effects of red and blue light on hypocotyl development. Blue light elicited a rapid, immediate response effective only during the period of irradiation; red light induced a response characterized by a lag period and persistence in subsequent darkness. Etiolated mustard seedlings showed similar responses to light and gravity. The results are discussed in relation to the possibility that two photosystems operate in hypocotyl growth.  相似文献

The effect of gibberellic acid (GA) and naphthylacetic acid (NAA) on hypocotyl elongation and cell wall polysaccharides was studied using Phaseolus vulgaris seedlings grown in light condition. The hypocotyl was demarcated into two segments — one near the root was called lower and the one near the cotyledon was called upper. The upper segment showed a typical sigmoidal growth curve while lower segment did not show any growth at all. GA promoted the growth of upper segment while NAA showed clear inhibition in both the segments. Xyloglucan content showed a clear inverse correlation with growth. Pectic polysaccharides did not show a clear trend, though showed an initial inverse correlation with growth. It is concluded that degradation of low and high molecular weight xyloglucans are involved in cell wall loosening which in turn may be responsible for the elongation growth of Phaseolus hypocotyls in light.  相似文献

Unilateral application of indole-3-acetic acid (IAA) in a lanolin base to hypocotyls of partially etiolated seedlings of wild-type Arabidopsis thaliana induced growth curvature in a dose-dependent manner. The effects of IAA in concentrations from 1 to 1000 microM were studied, with maximum IAA-induced curvature at 100 microM. Three IAA-insensitive mutants were isolated and are all in the same locus, massugu1 (msg1). They did not undergo hypocotyl growth curvature at any of the IAA concentrations tested. msg1 is recessive and is located on chromosome 5. msg 1 hypocotyl growth is resistant to 2,4-dichlorophenoxyacetic acid (2,4-D), but the roots are as sensitive to 2,4-D as the wild type. Growth of the hypocotyl was inhibited to essentially the same extent as the wild type by 6-benzylaminopurine, abscisic acid, and 1-aminocyclopropane-1-carboxylate, an ethylene precursor. The msg1 leaves were also resistant to 2,4-D-induced chlorosis. The gravitropic response of the msg1 hypocotyl takes much more time to initiate and achieve the wild-type degree of curvature, whereas the msg1 roots responded normally to gravity. The mature plants and the etiolated seedlings of msg1 were generally wild type in appearance, except that their rosette leaves were either epinastic or hyponastic. msg1 is the first auxin-insensitive mutant in which it effects are mostly restricted to the hypocotyl and leaf, and msg1 also appears to be auxin specific.  相似文献

A neutral growth inhibitor, for which the name raphanusanin is proposed, has been isolated in crystalline form from light-exposed Sakurajima radish (Raphanus sativus var. hortensis f. gigantissimus Makino) seedlings and identified as a new compound, 3-methoxy-4-methylthio-2-piperithione by spectrometric analyses.

Applied raphanusanin inhibited the hypocotyl growth of etiolated radish and lettuce seedlings at concentrations higher than 1.5 × 10−6 molar.

The endogenous raphanusanin contents in cotyledons and hypocotyls of radish seedlings increased more under red light, but decreased or maintained the initial level in the dark. Its content in roots showed almost no change between the light and dark materials.

A positive hydraulic signal in the form of a xylem pressure step was applied to the roots of intact seedlings of Cucumis sativus L. and Pisum sativum L. Surface electrodes at three positions along the epicotyl/hypocotyl recorded a propagating depolarization which appeared first in the basal, then the central and sometimes the apical electrode positions and fitted the characteristics of a slow wave potential (SWP). This depolarization differed between pea and cucumber. It was transient in cells of pea epicotyls but sustained in cucumber hypocotyls. It was not associated with a change in cell input resistance in pea epicotyls but preceded an increase in the input resistance of cucumber hypocotyl cells. With the increased xylem pressure the growth rate (GR) of cucumber hypocotyls and pea epicotyls underwent a transient increase, peaking after 5 min. If the depolarization reached the growing upper region, it preceded a sustained decrease in the GR of cucumber hypocotyls but only a transient decrease in the GR of pea epicotyls. A temperature jump in the root medium (heat treatment) induced a steep pressure spike in the xylem of the cucumber hypocotyl which showed similar electric and growth effects as the previously applied, non-injurious pressure steps. We suggest that the observed differences in the electric and growth responses between the species were caused by the closure of ion channels in depolarized cells of cucumber but not pea seedlings.  相似文献
