To investigate the incorporation of essential fatty acids into myelin components, 24-day-old rabbits were injected intracerebrally with [14C]linoleate, [14C]linolenate, or [3H]Myristate for comparison. Animals were killed 22 hr later and myelin was isolated. [3H]myristate labeled all myelin lipids including monogalactosyl diglyceride, with the exception of sulfatides. With14C-essential fatty acids, only glycerophospholipids were efficiently labeled and their specific activities were in the following decreasing orders: PC>PI>PE>PS with [14C]linoleate, and PE>PC>PI=PS with [14C]linolenate. Among myelin proteins, PLP and DM-20 were labeled with all 3 precursors. PLP was purified from myelin labeled with14C-essential fatty acids. The label was then cleaved from the protein by alkaline methanolysis and was identified as a dienoic ([14C]linoleate) or a tetraenoic ([14C]linolenate) fatty acid. MBP was not labeled with [3H]myristate, but was slightly labeled with both14C-essential fatty acids. [1-14C]-Acetate incorporation into total and polar lipids was studied in the growing pollen tubes of Crotalaria juncea. Ungerminated pollen had phosphatidyl inositol, phosphatidyl serine, phosphatidyl choline, phosphatidyl ethanolamine, phosphatidyl glycerol, monogalactosyl diglyceride, digalactosyl diglyceride, sulpholipid and steryl glycosides. In the growing pollen tubes considerable [1-14C]-acetate incorporation was observed into the individual polar lipids. The exogenous carbon source significantly influenced lipid biosynthesis. Boric acid (20mg/l.) promoted both pollen tube growth and acetate incorporation into phospholipids. In comparison to 5′-adenosine monophosphate, cyclic-3′,5′-adenosine monophosphate (cAMP) promoted tube growth and also enhanced phospho-and glycolipid biosynthesis. The regulation of membrane component biosynthesis by cAMP is suggested.  相似文献

Axonal regulation of Schwann cell glycolipid biosynthesis   总被引:2,自引:0,他引:2  
Schwann cell biosynthesis of glycolipids was studied by in vitro incorporation of [3H]galactose into neonatal rat sciatic nerves before and after endoneurial explant culture and in culture of purified Schwann cells. In neonatal nerves prior to culture, [3H]galactose was actively incorporated into galactocerebrosides (GalCe), monogalactosyl diacylglycerol (MGDG), and the sulfatides (Su). In contrast, the incorporation of [3H]galactose into MGDG, GalCe, and Su was nearly undetected in endoneurial explants after 4 days in vitro (div). Instead, there was increased3H-labeling of glucocerebrosides (GlcCe) and its homologues, with tetrahexosylceramides (GL-4) being a major product, which continued through 8 div. This shift in glycolipid biosynthesis was further demonstrated in the purified Schwann cell cultures. Synthetic diglycerides which differed in unsaturation of fatty acids gave the same incorporation of [14C]galactose from UDP-[14C]galactose when added to acetone powders of spinach chloroplasts up to about 0·6 mg diglyceride/20 mg acetone powder. Diolein and the endogenous diglyceride isolated from the acetone extract of chloroplasts stimulated galactolipid biosynthesis to a similar extent. With all diglycerides used, monogalactosyl diglyceride was the main product with little accompanying synthesis of digalactosyl diglyceride. The radioactivity in the monogalactosyl diglyceride synthesized from UDP-[14C]galactose by whole chloroplasts was distributed widely among the monogalactosyl diglycerides with different fatty acid composition. It is concluded that the enzyme which catalyses the transfer of galactose from UDP-galactose to diglyceride is not specific for polyunsaturated diglycerides and that the polyunsaturated monogalactosyl diglycerides arise either by desaturation of the fatty acyl residues after monogalactolipid synthesis or by transacylation. Acetone powders of chloroplasts prepared from several Gramineae did not exhibit transferase activity although whole chloroplasts were active.  相似文献

There was greater incorporation of [2?14C] acetate and of [6?14C] glucose into phosphoglycerides than into triglycerides, of 1 1/2, 2, 3, 4 and 6 day old mycelial sample of the fungus Glomerella cingulata. Maximum incorporation into both classes of lipids occurred in young mycelial samples (2 to 3 days of age) which had a high content of total nitrogen. The five sets of mycelial samples all contained somewhat larger quantities of phosphoglycerides than triglycerides, and changes in content of both classes of lipids were similar in pattern to changes in content of total nitrogen. Incorporation, accumulation and total nitrogen of the mycelial samples, decreased at 4 days but increased again by 6 days. The apparent turnover of the triglycerides and phosphoglycerides was qualitatively similar although there was greater apparent turnover of phosphoglycerides than triglycerides; the similarity in patterns of apparent turnover was inferred to be a consequence of acyl exchange between labeled and unlabeled triglycerides and phosphoglycerides. There was greater incorporation of [2?14C] acetate into the acyl than into the glyceryl moieties of both classes of lipids but greater incorporation of [6?14C] glucose into the glyceryl than acyl moieties. With both precursors, the glyceryl and acyl moieties of the phosphoglycerides were more heavily labeled than corresponding moieties of the triglycerides.  相似文献

Abstract— Partially purified myelin from the brains of 17-day-old rats was separated into 4 subfractions on a three-step sucrose gradient by virtue of heterogeneity in density and particle size. Precursor-product relationships between different membrane fractions were investigated by determining the specific radioactivity of individual lipids in each subcellular fraction 15 min after intracranial injection of an appropriate precursor. Rats were injected with [2-3H]glycerol. myelin subfractions prepared, and individual lipids separated by TLC. For choline and ethanolamine phospholipids, specific radioactivity was highest in the densest fraction (D), intermediate in the next densest fraction (C), and lowest in the lighter fractions (B and A). Similar results were observed for cerebroside and sulphatide when [3H]galactose was the precursor. These data are consistent with (but do not prove) a precursor-product relationship for individual lipids from the densest to the lightest subfraction. Another experimental design involving time staggered injections of [3H] and [14C] precursors was developed which enables a more definitive result with regard to precursor-product relationships to be obtained. A precursor-product relationship between a given lipid in a dense myelin membrane fraction, and the same lipid in a lighter subfraction, would be indicated by a change in isotope ratio. If there is no precursor-product relationship. Ihe isotope ratio should be constant. Such experiments were done with [3H] and [14C]glycerol. The data indicated that phosphatidyl ethanolamine and its plasmalogen analog were added first to the densest subfraction and then in turn to the lighter subfractions. In contrast, phosphatidyl choline and its plasmalogen analog were added “simultaneously” (i.e. with delays of much less than 15min) to each of the subfractions. Similar experiments with [3H] and [14C]galactose showed that cerebroside, sulphatide and galactosyl diglyceride also entered the subfractions simultaneously rather than in sequential order. Thus the assembly of the myelin sheath involves an obligate order of addition of certain lipids. while other lipids are probably added in a random order.  相似文献

In barley, glycine betaine is a metabolic end product accumulated by wilted leaves; betaine accumulation involves acceleration of de novo synthesis from serine, via ethanolamine, N-methylethanolamines, choline, and betaine aldehyde (Hanson, Scott 1980 Plant Physiol 66: 342-348). Because in animals and microorganisms the N-methylation of ethanolamine involves phosphatide intermediates, and because in barley, wilting markedly increases the rate of methylation of ethanolamine to choline, the labeling of phosphatides was followed after supplying [14C]ethanolamine to attached leaf blades of turgid and wilted barley plants. The kinetics of labeling of phosphatidylcholine and betaine showed that phosphatidylcholine became labeled 2.5-fold faster in wilted than in turgid leaves, and that after short incubations, phosphatidylcholine was always more heavily labeled than betaine. In pulse-chase experiments with wilted leaves, label from [14C]ethanolamine continued to accumulate in betaine as it was being lost from phosphatidylcholine. When [14C]monomethylethanolamine was supplied to wilted leaves, phosphatidylcholine was initially more heavily labeled than betaine. These results are qualitatively consistent with a precursor-to-product relationship between phosphatidylcholine and betaine.  相似文献

The membrane structural lipids of somatic cells and gonidia isolated from Volvox carteri f. nagariensis spheroids have been characterized. The principal polar lipid components of both cell types are sulfoquinovosyl diglyceride, mono- and digalactosyl diglyceride, phosphatidylglycerol, phosphatidylethanolamine, and 1(3), 2-diacylglyceryl-(3)-O-4′-(N,N,N,-trimethyl)homoserine. Light-synchronized cultures of spheroids were shown to incorporate [14C]bicarbonate, [35S]sulfate, [14C]palmitic acid, and [14C]lauric acid into complex lipids. [14C]Palmitic acid was incorporated mainly into diacylglyceryltrimethylhomoserine and was not significantly modified by elongation or desaturation. In contrast, [14C]lauric acid was incorporated into a wider variety of complex lipids and was also converted into longer chain saturated and unsaturated fatty acids. Volvox is a promising system for studying the role of membranes in algal cellular differentiation.  相似文献

An in vivo method of labelling lipid fractions in developing seeds of Brassica campestris using [1–14C] acetate has been developed. The “wick” method for introducing label into the intact plant is quite effective, safe and easy to use. The results obtained were reproducible and comparable to those reported earlier for seeds procured from greenhouse grown plants. The labelling pattern showed that rapid oil deposition began around 20 days after anthesis (DAA) and continued until about 45 DAA. The proportion of label in polar lipids declined and that in non-polar lipids increased during the phase of active oil synthesis. Among phospholipids, the label was incorporated mainly in phosphatidyl choline (PC), which was found to be the major fraction of phospholipids. During development, the two galactolipids i.e. monogalactosyl diglyceride (MGDG) and digalactosyl diglyceride (DGDG) followed patterns exactly opposite to each other. The content of the label in MGDG decreased, while that in DGDG increased, indicating the conversion of MGDG to DGDG during maturation.  相似文献

The major lipids in Cyanidium caldarium Geitler are monogalactosyl diglyceride, digalactosyl diglyceride, plant sulfolipid, lecithin, phosphatidyl glycerol, phosphatidyl inositol, and phosphatidyl ethanolamine. Fatty acid composition varies appreciably among the lipids, but the major ones are palmitic acid, oleic acid, linoleic acid, and moderate amounts of stearic acid. Trace amounts of other acids in the C14 to C20 range were also present. Moderate amounts of linolenic acid were found in two strains, but not in a third. The proportion of saturated acid is relatively high in all lipids ranging from about a third in monogalactosyl diglyceride to three-fourths in sulfolipid. This may be a result of the high growth temperature. Lipases forming lysosulfolipid, and lysophosphatidyl glycerol are active in ruptured cells; galactolipid is degraded with loss of both acyl residues. Thus the lipid and fatty acid composition of Cyanidium more closely resembles that of green algae than that of the blue-green algae, although there are differences of possible phylogenetic interest.  相似文献

A crude membrane fraction from Chlamydomonas reinhardii transferredradioactivity from UDP-[14C]galactose to endogenous lipids.Most of the radioactivity was detected in digalactosyl diacylglycerolmoieties in which only the distal galactose residue (-linkedto monogalactosyl diacylglycerol) was labeled. A second compoundwas identified as monogalactosyl diacylglycerol labeled in thegalactose moiety ß-linked to glycerol. In additionto these two galactolipid species, trace amounts of radioactiveglucose were detected in the aqueous phase after mild acid treatmentof the total lipid fraction. This demonstrates the presenceof a 4-epimerase and provides indirect evidence for the presenceof a small amount of polyprenyl-monophosphate-glucose whichwas presumably not detectable per se because of the bulk ofneutral galactolipids. The failure to detect polyprenyl-phosphate-galactoseor mild acid labile galactose at any time during the incubationsuggests that galactosylation of Chlamydomonas proteins mightoccur without the involvement of lipid intermediates. Suspensions of isolated pine needle chloroplasts were shown to incorporate galactose from UDP galactose-[14C] into galactolipids. The incorporation of the label among galactolipids was always considerably higher in the monogalactosyl diglycerides than in the digalactosyl diglycerides. The galactosyl incorporation into both galactolipid fractions was optimal at pH 8.0 and was inhibited by sulphydryl reagents (p-chloromercuribenzoate, N-ethyl maleimide and CdCl2). The chloroplast preparations were also able to biosynthesize various phospholipids and galactolipids from palmitoyl-[1-14C]-CoA; the major portion of the label appeared in phosphatidyl choline. The incorporation of palmitic-[1-14C] acid into various lipids was very poor compared to that of palmitoyl-[1-14C]-CoA. However, addition of ATP and CoA markedly stimulated lipid biosynthesis from palmitic-[1-14C] acid, suggesting the presence of activating enzymes. These chloroplast suspensions did not show any de novo fatty acid synthesis.  相似文献

Photo-, mixo- and heterotrophically grown cultures of Chlamydomonas reinhardi (wild type ss and 2 streptomycin-resistant mutants sr3 and sr35) have been analyzed for lipids and fatty acids. Ether-soluble lipids, chlorophyll, monogalactosyl diglyceride, digalactosyl diglyceride, sulfolipid, phosphatidyl ethanolamine, phosphatidyl choline, phosphatidyl glycerol and the relative amounts of fatty acids in total and individual lipids have been determined. The lipid and fatty acid compositions are very similar in the 3 strains and are not affected by the mutations. Fatty acids belong exclusively to the C16 and C18 series, 16:0, 16:4, 18:1, 18:2, 18:3 (6,9,12) and 18:3 (9,12,15) comprising about 90% of the total. 18:3 (6,9,12) is concentrated in phosphatidyl ethanolamine. In streptomycin-bleached sr3 cells, ether-soluble lipids increase from 7 to 11% of dry weight on greening, mostly due to synthesis of monogalactosyl diglyceride and chlorophyll. Monogalactosyl diglyceride of bleached cells exhibits the same fatty acid pattern before and after greening.  相似文献

The lipid composition of tomato fruit and its mitochondrial fraction were examined at various stages of fruit ripeness. Phosphatidyl choline, phosphatidyl ethanolamine, monogalactosyl diglyceride, digalactosyl diglyceride and phosphatidyl inositol were found to be the major lipids of tomato pericarp at all stages of ripeness. Mitochondrial lipids resembled those of the parent tissue except for the absence of monogalactosyl diglyceride and a greater percentage of diphosphatidyl glycerol and phosphatidic acid. Changes in the lipid-protein ratio of mitochondria were noted with ripening.  相似文献

Photosynthetically active chloroplasts retaining high rates of fatty acid synthesis from [1-14C]acetate were purified from leaves of both 16:3 (Solanum nodiflorum, Chenopodium album) and 18:3 plants (Amaranthus lividus, Pisum sativum). A comparison of lipids into which newly synthesized fatty acids were incorporated revealed that, in 18:3 chloroplasts, enzymic activities catalyzing the conversion of phosphatidate to diacylglycerol and of diacylglycerol to monogalactosyl diacylglycerol (MGD) were significantly less active than in 16:3 chloroplasts. In contrast, labeling rates of MGD from UDP-[14C]gal were similar for both types of chloroplasts.

The composition and positional distribution of labeled fatty acids within the glycerides synthesized by isolated 16:3 and 18:3 chloroplasts were similar and in each case only a C18/C16 diacylglycerol backbone was synthesized. In nodiflorum chloroplasts, C18:1/C16:0 MGD assembled de novo was completely desaturated to the C18:3/C16:3 stage.

Whereas newly synthesized C18/C18 MGD could not be detected in any of these chloroplasts if incubated with [14C]acetate after isolation, chloroplasts isolated from acetate-labeled leaves contained MGD with labeled C18 fatty acids at both sn-1 and sn-2 positions. Taken together, these results provide further evidence on an organellar level for the operation of pro- and eucaryotic pathways in the biosynthesis of MGD in different groups of plants.

Exogenous [1-14C]oleic acid and [1-14C]linoleic acid were taken up and esterified to complex lipids by greening cucumber (Cucumis sativus L.) cotyledons. Both 14C-labeled fatty acids were initially esterified to phosphatidylcholine prior to eventual accumulation in triacylglycerols and galactolipids. Kinetic data suggest that esterification occurs prior to desaturation and that phosphatidylcholine is the initial site of both [14C]-oleate and [1-14C]linoleate esterification and of [1-14C]oleate desaturation to [1-14C]linoleate. [1-14C]Linoleic acid was esterified more rapidly than [14C]oleic acid and its desaturation product, [1-14C]α-linolenate, occurred mainly on monogalactosyl diacylglycerol, although some was also observed on the other major acyl lipids, including phosphatidylcholine.  相似文献

We determined the expression of three myelin-typical lipids in the continuous CG-4 glial cell line of oligodendrocyte progenitor cells, as the cells differentiated into oligodendrocytes. On 6 different days during the first 9 days of oligodendrocyte development, cells were labeled for 24 h with [3H]ethanolamine to label ethanolamine plasmalogens or with [3H]galactose to label the galactocerebroside and sulfogalactocerebroside; and the amount of labeled lipid expressed on each day was determined. Each labeled lipid was expressed with its own specific time course and in a defined amount on each day of differentiation. Increased labeling of plasmalogens and sulfogalactocerebroside started at early developmental stages, and increased labeling of galactocerebroside started at later stages. The results indicate that the differentiating CG-4 cell line provides a valuable system to investigate factors affecting the early time course of myelin-lipid expression and the amounts expressed.  相似文献

Abstract— Tissue explants from frontal lobes of rat brain were used for the study of cerebral fatty acid metabolism. After tissues had been maintained in serum-supplemented medium, a lipid-free medium was substituted and metabolic studies were carried out. Under these conditions explants continued to take up lipid precursors for at least 48 h, as judged by incorporation of dl -[2-14C]mevalonic acid into cellular lipids. [l-14C]Stearic acid and [l-14C]palmitic acid were bound to cells as the free fatty acids, or incorporated into neutral lipids (particularly triglycerides), glycolipids and phospholipids. In the galactolipid fraction, cerebrosides were the principal radioactive lipids. Choline phosphoglycerides, ethanolamine phosphoglycerides, inositol phosphoglycerides and serine phosphoglycerides were the principal radioactive phospholipids. Fatty acids were incorporated into cellular lipids either unchanged or after desaturation, chain elongation, or both. Maximum incorporation of stearate occurred in tissues derived from 3-day-old animals. With increasing age the uptake of fatty acid dropped sharply. When the labelling of lipids as a function of time was followed in 3-day-old animals, triglycerides and choline phosphoglycerides were the first fractions to take up labelled stearate. Labelling of cerebrosides occurred slowly, only becoming evident after 24 h. These studies exemplify the usefulness of tissue explants for prolonged metabolic studies in normal and pathological specimens of brain.  相似文献

1. Fatty acid synthesis from [2-(14)C]acetate by Chlorella vulgaris cells grown and incubated in the dark is limited almost entirely to the production of saturated and monoenoic acids. 2. In light-incubated cells, both saturated and polyunsaturated fatty acids are rapidly synthesized. 3. Two groups of lipids can be distinguished in both dark- and light-incubated cells. The first group, consisting of phosphatidyl-glycerol, monogalactosyl diglyceride, lecithin and neutral glyceride, has a very high turnover rate for certain fatty acids. The second group, consisting of digalactosyl diglyceride, sulpholipid, phosphatidylethanolamine and phosphatidylinositol, has a slow turnover of fatty acids. 4. The lipids with rapid fatty acid turnover may be involved in the sequences of saturated and unsaturated fatty acid synthesis. A classification of lipids is made on the basis of their suggested functions.  相似文献

Comparative studies were undertaken on the in vivo and in vitro incorporation of [14C] ethanolamine, [3H] methionine and [14C] S-adenosyl-methionine into phosphatidylethanolamine (PhE) and phosphatidylcholine (PhC) of rat liver and brain. It was observed that brain can synthesize de novo PhC from PhE via the transmethylation pathway, however synthesis rates were (1) markedly lower than those of liver and (2) decreased significantly with age. In the choline-containing lipids more than 95% of the radioactivity was found in PhC. Studies on the localization of the radioactivity in PhC following the intracranial injection of [3H] methionine or [14C] ethanolamine revealed that both precursors are incorporated almost exclusively into the choline moiety of this phospholipid. There was significant labeling of PhC only when the precursors were administered intracranially and much less incorporation was observed with the systemic routes. Thus following the intravenous administration of [14C] ethanolamine, the specific radioactivities of liver PhE and PhC were up to 75 times as high as those of brain and 4 to 5 times as high in the organs of the 20-day old as those of the adult. In contrast, when this precursor was administered intracranially the specific radioactivities of both phospholipids in liver were only twice as high as those of brain. Although the short-and long-term time-course studies on the in vivo incorporation of [14C] ethanolamine and [3H] methionine into PhC of both organs could suggest a precursor-product relationship between the biosynthesis of this phospholipid in liver and brain, this apparent relationship could also be due to the high turnover of PhE in liver, with half-life of 2.87 hr, and its low turnover in brain, with half-life of 10.7 days. The present findings on the low rate of formation of PhC from PhE in brain coupled with the fact that this conversion declines sharply with age, especially when the isotopes are administered systemically, could explain the observation of previous investigators that the brain cannot synthesize its own choline and thus it must derive its choline from exogenous sources such as lipid-choline. It was concluded that the brain can synthesize its own choline; however it remains also dependent on liver and dietary choline which are probably transported into the brain as free choline.  相似文献
