Spermatozoa and spermiogenesis in the deep-water cephalopodSpirula sprirula (L.) are examined using transmission electron microscopy. Mature spermatozoa (taken from spermatophores) are elongate cells 115–120 μm long, composed of a conical acrosomal vesicle, cylindrical nucleus (6.8–7 μm long), flagellum and a loose mitochondrial sleeve — the latter concealing the proximal 6–8 μm of the flagellum. The acrosomal vesicle is 2.8 μm long with fibro-granular contents and an electron-lucent apical zone. Subacrosomal material, organized as closely packed granules, fills a basal invagination of the acrosomal vesicle. In early spermatids the flagellum is derived from a triplet substructure centriole positioned close to the developing nuclear invagination. As flagellum formation proceeds, the acrosomal vesicle (produced evidently through Golgi secretion) attaches to the condensing nucleus. Spermatids are connected by cytoplasmic bridges throughout their development, and exhibit a perinuclear sheath of microtubules from the onset of the fibrous stage of nuclear condensation (mid-, late spermatids). In mid-spermatids, mitochondria collect posterior to the nucleus and subsequently are packed into a cylindrical extension of the plasma membrane to form the periflagellar mitochondrial sleeve. These features of spermiogenesis and mature spermatozoa ofSpirula clearly associate the Spirulidae with the Sepiida, Teuthida and Sepiolida — particularly with the latter order. However, pending results of a thorough review of coleoid sperm morphology, the Spirulidae are here included in their own order — Spirulida (of Reitner & Engeser, 1982) — rather than in either the Sepiida or Sepiolida.  相似文献

A short flagellum bearing unusual appendages inMallomonas guttata is described. These appendages, ca. 0.2 μm in diameter and 10–20 μm long, 7–10 in number, originate from the distal end of the short flagellum. Their membrane is continuous with the flagellar membrane and no particular structure was observed in appendage lumen.  相似文献

Cryothecomonas longipes Schnepf and Kühn sp. nov. is a colourless biflagellate organism, 9–14 μm long and 7–9 μm wide when not filled with food vacuoles. It was detected in the North Sea, feeding with pseudopodia on diatoms. It penetrates the host shell, while the main body of the flagellate remains outside the frustule. Cells are covered with a multilayered theca. The pseudopodium protrudes through a preformed slit in the theca. Each flagellum also emerges through a pit in which the theca forms a funnel of complex structure that girdles each flagellum. The anterior flagellum is 9–15 μm long and oriented forward; the ventral flagellum, posteriorly directed, is 20–24 μm long and bears fine hairs. The flagellar roots consist of microtubules that emerge at satellites around the basal bodies and run along the flagellar pits. In addition, the ventral flagellum is accompanied by a band of six microtubules. It is proximally attached to a small fibrillar band, which interconnects the basal bodies. Cryothecomonas longipes has two or three types of extrusomes which pierce the theca when discharged. Their mode of discharge is discussed. Microbody-like vesicles containing small tubules are closely associated with older digestion vacuoles. Cryothecomonas longipes is compared with other species of the genus and a diagnosis is given. Summary The cytoarchitecture and the pattern of cytoplasmic streaming change during the development of root hairs ofMedicago truncatula and after a challenge with nodulation (Nod) factors. We measured the speed and orientation of movement of 1–2 μm long organelles. The speed of organelle movement in cytoplasmic strands in the basal part of growing root hairs is 8–14 μm/s and is of the circulation type like in trichoblasts, bulges before tip-growth initiation, and full-grown hairs. In the subapical area of growing hairs, reverse-fountain streaming occurs discontinuously at a slower net speed. The reason for the slower speed is the fact that organelles often stop and jump. Reverse-fountain streaming is a pattern in which the main direction of organelle transport reverses 180 degrees before the cell tip is reached. Within minutes after their application to roots,Rhizobium leguminosarum-derived Nod factors, cause an increase and divergence in the subapical cytoplasmic strands. This phenomenon can best be observed in the growth-terminating hairs, since in hairs of this developmental stage, subapical cytoplasmic strands are transvacuolar. First, the tips of these hairs swell. The organelle movement in the swelling tip increases up to the level normal for circulation streaming, and the number of strands with moving organelles increases. When a new polar outgrowth emerges, reverse-fountain streaming is set up again, with all its characteristics like those seen in growing hairs. This outgrowth may obtain a new full root hair length, by which these hairs may become twice as long as nonchallenged hairs. A new spirochetal strain was isolated from the rumen of a black-and-white Holstein cow and preliminarily characterized. The sugar fermentation tests and morphological observations indicated this organism to be a member of a novel, as yet undescribed spirochetal rumen species. The small subunit ribosomal RNA genes were amplified and the PCR products were cut with the restriction endonucleasesTaql,Ddel,Hhal andSau3Al. The comparison of the observed RFLP with the hypothetical fragment lengths of the computer analyzed 16S rRNA sequences from the type strains of the ruminal spirochetesTreponema bryantii andT. saccharophilum confirmed the tentative novel identification. Transmission electron microscopy showed that the bacterium has the typical spirochetal structures,i.e. the outer sheath, the protoplasmic cylinder and the axial filament (it is not yet clear how many flagella compose the filament). An additional extracellular structure was observed which appeared as an exocytoplasmic polar flagellum, approximately 2 μm long and protruding from one tip of the cell. The average size of the cells was 0.5×10–15 μm and the wavelengths and the amplitudes of the primary coils were 2.9 and 1.3 μm, respectively.  相似文献

Sclerocarya birrea (marula) is an indigenous South African tree with highly valued medicinal and nutritional properties. Induction of nodular meristemoids from leaf explants was achieved on Murashige and Skoog (MS) and woody plant medium (WPM) supplemented with 6-benzyladenine (BA) in combination with naphthalene acetic acid (NAA), indole-3-butryric acid (IBA) and indole-3-acetic acid (IAA). Induction of nodular meristemoids from 86% of the leaf cultures was achieved on MS medium with 4.0 μM BA and 1.0 μM NAA. High levels (78–100%) of induction were also achieved on WPM with different concentrations of BA (1.0–4.0 μM) and IBA (1.0–4.0 μM). The highest conversion of meristemoids into shoots was only 22% for 4.0 μM BA and 1.0 μM NAA on MS initiation medium. This was improved to 62% when nodular clusters were cultured in a MS liquid medium. Histological studies revealed the globular stage of the nodular meristemoids. This protocol has potential for application in mass micropropagation and plant breeding of S. birrea.  相似文献

A new species of Hirsutella, H. proturicola, isolated from a subterranean proturan (Baculentulus densus; Protura, Hexapoda), is described and illustrated. Hirsutella proturicola is characterized by producing monoblastic phialides of 24–51.5 × 2.5–5 μm with a slightly roughened neck, fusiform and curved conidia of 9–18 × 2.5–4 μm that have a truncate base and a papillate projection often capped with sheath-like mucilage, and pluricellular, globose to subglobose chlamydospores of 21–48 × 21–41.5 μm. This species is morphologically and phylogenetically close to H. rostrata, an acaropathogenic species, but can be distinguished from the size of the phialides and the size and shape of the conidia.  相似文献

Summary Callus of Phalaenopsis Nebula was induced from seed-derived protocorms on 1/2 Murashige and Skoog (MS) basal medium plus 0–1.0 mg l−1 (0–4.52 μM) N-phenyl-N′-1,2,3,-thiadiazol-5-yl urea (TDZ) and/or 0–10 mg l−1 (0–45.24 μ M) 2,4-dichlorophenoxyacetic acid (2,4-D). Protocorms 2 mo. old performed better than 1-mo.-old protocorms for callus induction. More calluses formed on 1/2 MS basal medium supplemented with 0.1–1.0 mg l−1 (0.45–4.52 μM) TDZ. These calluses could be maintained by subculturing every month with basal medium supplemented with 0.5 mg l−1 (2.27 μM) TDZ and 0.5 mg l−1 (2.26 μM) 2,4-D. Protocorm-like bodies were formed, and plants regenerated from these calluses on 1/2 MS basal medium alone or supplemented with 0.1–1.0 mg l−1 (0.45–4.52 μM) TDZ. Plantlets were then potted on sphagnum moss in the greenhouse and grew well. No chromosomal abnormalities were found among the root-tip samples of 21 of the regenerated plantlets that were successfully acclimatized.  相似文献

We investigated seasonal variation of grazing impact of the pigmented nanoflagellates (PNF) with different sizes upon Synechococcus in the subtropical western Pacific coastal waters using grazing experiments with fluorescently labeled Synechococcus (FLS). For total PNF, conspicuous seasonal variations of ingestion rates on Synechococcus were found, and a functional response was observed. To further investigate the impact of different size groups, we separated the PNF into four categories (<3, 3–5, 5–10, and >10 μm). Our results indicated that the smallest PNF (<3 μm PNF) did not ingest FLS and was considered autotrophic. PNF of 3–5 μm in size made up most of the PNF community; however, their ingestion on Synechococcus was too low (0.1–1.9 Syn PNF−1 h−1) to support their growth, and they had to depend on other prey or photosynthesis to survive. The ingestion rate of the 3–5 μm group exhibited no significant seasonal variation; by contrast, the ingestion rates of 5–10 and >10 μm PNFs showed significant seasonal variation. During the warm season, 3–5 μm PNF were responsible for the grazing of 12% of Synechococcus production, 5–10 μm PNF for 48%, and >10 μm PNF for 2%. Taken together, our results demonstrate that the PNF of 3–10 μm consumed most Synechococcus during the warm season and exhibited a significant functional response to the increase in prey concentration.  相似文献

Using numerical methods, the initial rates of oxygen uptake by the red blood cell have been computed. The methods accommodate both a water layer and membrane which may act as diffusive impedance to gas influx. The differential solubilities of the gas in these two layers have also been incorporated in the methods. The presence of a 0.50–0.65 μm deoxygenated water layer has been calculated to simulate the experimental results by Roughton (1959). Experimental studies of CO and NO uptake by the red cell could also be simulated. Although a membrane-only model with given parameters can also account for the observed rates of oxygenation of the red cell (Weingardenet al., submitted for publication), the additional incorporation of differential solubilities of oxygen in the different layers of the RBC yields results that indicate a three layer model to be more plausible. Using a thin layer-red cell oxygenation system, the rates of oxygenation were determined for red cells surrounded by a 4.2 μm deoxygenated water layer. The rates were found to compare favorably to the results of the theoretical model.  相似文献

We stained sarcomere thin filaments with fluorescently labeled phalloidin, measured sarcomere and muscle length, and calculated sarcomere number in pyloric and gastric mill muscles. A wide range of sarcomere lengths (3.25–12.29 μm), muscle lengths (5.9–21.1 mm), and sarcomere numbers (648–3,036) were observed. Sarcomere number differences occurred both because of changes in sarcomere length and muscle length, and sarcomere and muscle length varied independently. This independence, the wide range of sarcomere numbers present, and the muscles being all ‘slow’, graded muscles allowed us to use these data to test Huxley and Neidergerke’s (1954) hypothesis that muscle dynamics depend on sarcomere number. The time constants of exponential fits to contraction relaxations were used to measure muscle dynamics, and comparison of theoretical predictions and experimental results quantitatively confirm the predicted dependence. The differing dynamics of the various pyloric muscles are likely functionally important, and the dependence of muscle dynamics on sarcomere number implies that sarcomere number is likely closely regulated in these muscles. The stomatogastric system may thus be an excellent model system for studying the mechanisms regulating muscle sarcomere number.  相似文献

Purpose of the present study was to evaluate antioxidant, antibacterial, antifungal, and antiviral activities of the petroleum ether, chloroform, ethyl acetate and methanol extracts as well as the alkaloid fraction of Lycopodium clavatum L. (LC) from Lycopodiaceae growing in Turkey. Antioxidant activity of the LC extracts was evaluated by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging method at 0.2 mg/ml using microplate-reader assay. Antiviral assessment of LC extracts was evaluated towards the DNA virus Herpes simplex (HSV) and the RNA virus Parainfluenza (PI-3) using Madin-Darby Bovine Kidney (MDBK) and Vero cell lines. Antibacterial and antifungal activities of the extracts were tested against standard and isolated strains of the following bacteria; Escherichia coli, Pseudomonas aeruginosa, Proteus mirabilis, Acinobacter baumannii, Klebsiella pneumoniae, Staphylococcus aureus, Bacillus subtilis as well as the fungi; Candida albicans and C. parapsilosis. All of the extracts possessed noteworthy activity against ATCC strain of S. aureus (4 μg/ml), while the LC extracts showed reasonable antifungal effect. On the other hand, we found that only the chloroform extract was active against HSV (16–8 μg/ml), while petroleum ether and alkaloid extracts inhibited potently PI-3 (16–4 μg/ml and 32–4 μg/ml, respectively). However, all of the extracts had insignificant antiradical effect on DPPH. In addition, we also analyzed the content of the alkaloid fraction of the plant by capillary gas chromatography-mass spectrometry (GC-MS) and identified lycopodine as the major alkaloid.  相似文献

Haptoglossa spp. (Lagenidiales, Oomycetes) have been known to parasitize microscopic animals by means of a "gun" cell that shoots an infection cell, named the sporidium, into the body of the animal. A thallus grown from the sporidium changes into a zoosporangium at maturation to produce a number of zoospores that encyst after a swarming period, and the resulting cysts germinate to produce gun cells. In Haptoglossa zoospora, endoparasitic in nematodes, the cysts of primary zoospores that swam for about 5 min did not develop gun cells but produced secondary zoospores that swam for about 3 h. After encystment of the secondary zoospores, each secondary cyst germinated to produce a gun cell. In the present study, the secondary zoospores of the genus Haptoglossa could be recorded with a videocassette recorder for the first time. The videocassette recording also revealed the infection of a nematodes by H. zoospora and H. heterospora to be composed of two steps of injection of a sporidium by the gun cell, in which the gun cell came in contact with the cuticle of a nematode and produced a spherical adhesorium on the tip of the cell in 0.07–0.1 s in both species. The adhesorium was ∼2 μm in H. zoospora and ∼4 μm in H. heterospora. When the adhesorium infiated to full size, it shot the sporidium into the nematode's body in 0.5–0.65 s and in 0.2–0.5 (or rarely 1.0) s in H. zoospora and H. heterospora, respectively. During the latest years medium-sized (15–30 μm), single-celled dinoflagellates have been reported to form blooms in the northern Baltic Proper and the Gulf of Finland in winter and spring. Recent studies (Kremp et al., 2003. Proceedings of the 7th International conference of Modern and Fossil Dinoflagellates, September 21–25, Nagasaki, Japan, 66 pp.) indicate that those blooms are caused by two isomorphic species – Scrippsiella hangoei (Schiller) Larsen, and a new species, tentatively belonging to the genus Woloszynskia. Until now there has been no report on how widely distributed these phytoplankton species are in the Baltic Sea. In this study, the occurrence of Scrippsiella/Woloszynskia complex in the entire Baltic Sea was investigated, by using monitoring data from 1997 to 2003. The species occurred in a salinity range from 2 to 8 PSU. Highest concentrations were observed at salinity 4.5–6.5 PSU. Maximum cell densities of Scrippsiella/Woloszynskia complex in the water column were mainly obtained in April or in the beginning of May by the water temperature <3 °C prior to stratification was formed. In the central Gulf of Finland, the second maximum was found in 1999 and 2002 by the temperature >6 °C. Bloom formations in the Baltic Proper and in the Gulf of Finland may not only be explained by optimum temperature and salinity, but also with other factors e.g. high nutrient concentrations and good seeding conditions from the sediments.  相似文献

The effect of design parameters and voltage of pulsed cesium sources with sapphire envelopes on their modulation characteristics are considered for the following combinations of operating parameters: the average repetition rate of current pulses up to f i = 1.0 kHz and the consumed electric power up to P l = 3 kW for the spectral range 3–5 μm; f i = 1.0–8.0 kHz and P l up to 1.0 kW for the spectral range 0.8–2.7 μm. Ampullary organs of Euristhmus lepturus occur in high densities along the head and in four parallel pathways along the trunk of the body. Large ampullary pores (125–130 μm) are easily distinguishable from other sensory epithelial pores due to the differences in size and the presence of a collar-like structure. Simple, singular ampullary organs of the head region consist of an ampullary pore connected to a long canal with a diameter of 115–175 μm before terminating as a simple ampulla with an external diameter of 390–480 μm. The ampullary canal is composed of 1–2 layers of flattened squamous epithelial cells, the basement membrane and an interlocking collagen sheath. The innermost cells lining the canal wall are adjoined via tight junctions and numerous desmosomes, as are those of the receptor and supportive cells. Canal wall tissue gives rise to a sensory epithelium containing between 242 and 285 total receptor cells, with an average diameter of 11.7 ± 5.3 μm, intermixed with medially nucleated supportive cells. Each receptor cell (21.38 ± 4.41 μm, height) has an apically positioned nucleus and a luminal surface covered with numerous microvilli. Neural terminals abut the basal region of receptor cells opposite multiple presynaptic bodies and dense mitochondria. Supportive cells extend from the ampullary lumen to the basement membrane, which is adjacent to the complex system of collagen fibres.  相似文献

Triaenorhina burti n. sp. (Cyclophyllidea: Paruterinidae) is described from Harpactes fasciatus (Trogoniformes: Trogonidae) from the Southern Province of Sri Lanka. The new species is characterised by: a body 24–32 mm long; 44 rostellar hooks alternating in two closely adjacent regular rows, with lengths of 63–65 μm (anterior row) and 39–41 μm (posterior row); regularly alternating genital pores; testes divided into two groups by the ovary and vitellarium; a gravid uterus forming a single oval sac; and a cylindrical paruterine organ not reaching the anterior proglottis margin. A key to the seven recognised species of Triaenorhina Spasskii & Shumilo, 1965 is presented.  相似文献

Neoskrjabinolepis (Neoskrjabinolepidoides) nuda n. sp. is described from the shrews Sorex unguiculatus (type-host), S. gracillimus, S. isodon and S. caecutiens on Sakhalin Island, Russia. The new species is characterised by: rostellar hooks 40–44 μm long and provided with small epiphyseal thickening of the handle; a long (95–100 μm) cirrus consisting of basal region with claw-shaped spines, a parabasal region with thin needle-shaped spines and an unarmed distal region; a cirrus-sac extending well into the median field; and 15–22 eggs per gravid uterus. A review of the species of Neoskrjabinolepis Spasskii, 1947 is presented. Currently, this genus includes nine species and is divided in two subgenera on the basis of strobilar development, which is gradual in the subgenus Neoskrjabinolepis (four species) and serial in the subgenus Neoskrjabinolepidoides Kornienko, Gulyaev & Mel’nikova, 2006 (five species). An amended generic diagnosis and an identification key to Neoskrjabinolepis spp. are presented.  相似文献

A halophilic nonpigmented rod-shaped (0.8–1.0 × 2.0–2.5 μm), gram-negative bacterium with a single polar flagellum (strain RS91) was isolated from acidic brines of flotation enrichment of potassium minerals (Silvinit Co., Solikamsk, Russia). The strain grew in the media with 2 to 25% NaCl (optimum at 10–12%), 20–45°C (optimum at 37°C), and pH 5.5–8.5 (optimum 6.5–7.5). It was an aerobe or facultative anaerobe incapable of fermentation. The strain was characterized by the absence of growth on glucose, fructose, and citrate, extensive aerobic growth on n-hexadecane and in the mineral medium with H2 + O2 + CO2 in the gas phase, anaerobic nitrate reduction with acetate or hydrogen (under H2 + CO2 + N2), and variable fatty acid composition. The DNA G+C content was 68.2 mol %. Phylogenetic analysis based on 16S rRNA gene sequencing revealed that while strain RS91 was most closely related to Arhodomonas aquaeolei HA-1T (98.3%) and Nitrococcus mobilis (98.1%), it was only remotely related to the halophilic phototroph Halorhodospira halophila (90.6%). Based on the combination of its phenotypic and genotypic characteristics, the organism was classified as a new species of the genus Arhodomonas, family Ectothiorhodospiraceae with the proposed name Arhodomonas recens sp. nov. This is the first report on the ascomycete Metschnikowia typographi from the adults and larvae of the great spruce bark beetle Dendroctonus micans in Turkey. In total, 910 of 1928 adults and 44 of 149 larvae investigated during the two years were infected by the pathogen. In a fresh smear the asci of the pathogen measure 18.5 ± 2.05 μm (14.7–22.3) in length and 2.1 ± 0.4 μm in width (n = 35). The ascospores are about 2 μm shorter than asci, having an average length of 16.4 ± 1.5 μm (14.2–18.0). The total infection rate of D. micans was 47.2 %. The prevalence of M. typographi infections differed between localities and years. Different infection rates of male and female beetles of D. micans were not recognized.  相似文献
