Steady-state kinetic equations for isotope exchange are derived for a number of one substrate-one product enzymic mechanisms in which two molecules of substrate or product can be combined with an enzyme molecule at the one time (e.g. allosteric mechanisms). The usual assumption, that the radioactive material is distributed among the substrate and product components according to a first-order law, is not valid. One can recognize whether isotope-exchange kinetics of an enzyme reaction follows first-order behaviour by using various initial concentrations of the labelled substance added to a mixture.  相似文献

In classic biological control using natural enemies, the question of whether a single species or multiple species should be introduced has been a matter of debate. The introduction of two parasitoids, Aphytis yanonensis and Coccobius fulvus (Hymenoptera: Aphelinidae), to control the arrowhead scale, Unaspis yanonensis (Hemoptera: Diaspididae), which is a serious pest in Japanese citrus orchards, has been one of the most successful biological control projects in Japan. The success of this program may be explained by two alternative hypotheses: (1) the parasitoid species work complementarily, or (2) only one of them plays a major role. To test which hypothesis is applicable to this host-parasitoid system, we conducted caging experiments and observed temporal changes in the proportion of the parasitisms and the densities of arrowhead scales enclosed with one of the following combinations of parasitoids: (1) A. yanonensis and C. fulvus together, (2) A. yanonensis alone, (3) C. fulvus alone, or (4) neither parasitoid. Parasitisms in the cohorts with A. yanonensis and C. fulvus together and C. fulvus alone rapidly increased to approximately 70%; parasitism with A. yanonensis alone also increased slightly, although it remained consistently lower that those with A. yanonensis and C. fulvus together and C. fulvus. At the end of the experiment, parasitisms with A. yanonensis and C. fulvus together and C. fulvus alone were significantly higher than that with A. yanonensis alone. Parasitism by C. fulvus constituted most of (74%) the parasitism in the cohort with A. yanonensis and C. fulvus together. Further, only C. fulvus suppressed the population growth rates of scales significantly. These results suggest that C. fulvus alone successfully suppresses scale populations as efficiently as both species together do.  相似文献

Plasma-cell tumour 5563 forms a single molecular species of immunoglobulin IgG(2)a, i.e. one variant of heavy chain and one variant of light chain. The molecules formed are labile and undergo alterations in charge properties, which rapidly lead to heterogeneity of the myeloma protein after synthesis. The single immunoglobulin species originally formed is found only after the shortest time-intervals tested, i.e. 10min incubation. Two types of changes in charge properties take place: (1) The originally formed protein (component o) is converted via an intermediate o' into the most basic form of 5563 myeloma protein found in serum (component a). Charge differences between these components are suppressed at pH8.9, but can be studied by chromatography at pH6.5 or by analysis of isoelectric points by isoelectric focusing in polyacrylamide gel. The conversion of components o and o' into component a apparently commences soon after assembly of the molecules and proceeds to completion extracellularly. (2) The second type of charge difference that distinguishes components a, b, c and d is exhibited over the pH range 6.0-8.9, but not at acid pH, and has been studied by electrophoresis at pH8.9, by chromatography and by isoelectric focusing. Conversion of component a into components b, c, d and e is only partial so that all five components can be found at decreasing concentrations in serum. Both types of charge alteration can be effected in vitro in the presence of serum, with optimum pH8.0. None of the charge differences could be attributed to the secretion process, since a component with the same isoelectric point as component o was found in secreted myeloma protein (1h). We have found no evidence to support the idea that the first type of change from component o to component a is due to ring formation of N-terminal [(14)C]glutamine into pyrrolid-2-one-5-carboxylic acid; however, our findings do not exclude this process happening very rapidly to a precursor of component o, possibly the polypeptide chain during or immediately after synthesis. In studying this point we noted that not only the heavy chains but also the kappa-type light chain of mouse 5563 myeloma protein have a blocked N-terminus.  相似文献

Flagellar-mediated motility is an indispensable function for cell types as evolutionarily distant as mammalian sperm and kinetoplastid parasites, a large group of flagellated protozoa that includes several important human pathogens. Despite the obvious importance of flagellar motility, little is known about the signalling processes that direct the frequency and wave shape of the flagellar beat, or those that provide the motile cell with the necessary environmental cues that enable it to aim its movement. Similarly, the energetics of the flagellar beat and the problem of a sufficient ATP supply along the entire length of the beating flagellum remain to be explored. Recent proteome projects studying the flagella of mammalian sperm and kinetoplastid parasites have provided important information and have indicated a surprising degree of similarities between the flagella of these two cell types.  相似文献

Alarm calling has been interpreted from the viewpoint of parental investment and nest defence. Calling should increase with an increasing value of the brood to the caller or with increasing vulnerability of the brood. Parental alarm calling might also be viewed from assessment–management with callers using vocalisations to elicit affective responses in others, e.g. in silencing offspring. I examined parental alarm calling in the Cyprus Wheatear (Oenanthe cypriaca) at different developmental stages. The Cyprus Wheatear produces two alarm calls, but one is emitted only during a short period of the year (type II alarm call). The commoner alarm call (type I alarm call), however, is used year-round. Predation was simulated using consistent approach by human observers. The brood cycle was divided into (I) incubation, (II) nestling, (III) early fledging, and (IV) late fledging. Type I calls increased from phases I to III, and decreased afterwards. Type II calls were absent during incubating and occurred during nestling and early fledging stages. As a conclusion, I suggest that the type I alarm call might be used to alert the mate, deter the predator and signal strength of nest defence, while the type II alarm call is addressed to offspring.  相似文献

Learning, and hence memory, is ubiquitous not only throughout the animal kingdom, but apparently throughout many regions of the brain. Is all learning reducible to a single common form? Neuropsychological dissociations suggest that the mammalian brain possesses a number of different and potentially independent memory systems, with different mechanisms and anatomical dispositions, some of which are neurally widely dispersed and others of which are narrowly organized. Among the types considered are: (i) short-term memory; (ii) knowledge and skills; (iii) stable associative memory; (iv) event memory; and (v) priming. As double or multiple dissociations do not lead to logically inevitable conclusions, it has been argued that an alternative to multiple memory systems is variable modes of processing. But these, too, would be dissociable on the same lines of evidence. Dissociations, if strong and absolute, have strong pragmatic power when they are combined with evolutionary and neuroscientific evidence. Multiple memory systems may possibly share some common cellular mechanisms, but such mechanisms do not define the separate properties at the systems level.  相似文献

Attention and motor preparation are two intimately linked processes. However, they can be dissociated in the laboratory in order to study their neuronal basis. Behavioral neurophysiology has thus shown that neurons that discharge in relation with attention or with motor preparation (or intention) exist in a variety of brain regions in the monkey, especially the prefrontal and premotor cortices. When examined more carefully, these two regions appear different in both the proportion of cells that respond during attention versus intention, and in the information coded in the so-called "preparatory activity". This activity reflects sensory selection in the prefrontal cortex (spatial attention/memory), motor selection in the premotor cortex. Furthermore, two regions in the dorsal aspect of premotor cortex can be distinguished on the basis of their relative involvement in attention: a rostral (anterior) region, functionally close to prefrontal cortex, and a caudal one, which appears functionally close to motor cortex. Using an experimental design derived from monkey experiments, a functional magnetic resonance imaging (fMRI) study recently indicated that the functional specialization within the premotor cortex is similar in monkey and man.  相似文献

To study the pattern of gene regulation of the plastid chaperonin 60 gene family a chimaeric gene was constructed fusing the 5-flanking region of the chaperonin 60 B3 gene to the -glucuronidase reporter gene. Histochemical and fluorometric analysis of the GUS activity present in transgenic plants harbouring this gene construct showed that the B3 promoter is expressed in leaves, stem, petioles and several flower tissues. The pattern of cell type-specific expression in stems and flowers was found to be developmentally regulated. Expression of the B3 promoter was found not to be heat-inducible, but highly repressed by wounding. The rapid decay in GUS activity upon wounding indicates that, at least under some physiological conditions, the gene product of this reporter gene is not as stable as has been previously thought.  相似文献
