A very small fragment of a branch leaf of Sphagnum imbricatum sensu lato has been recovered from a sample of food residue taken from the Tyrolean Iceman’s colon. On grounds of present phytogeography, the fragment is probably S. affine rather than S. austinii. Neither of these species is known to grow in South Tyrol at present. No matter how it was that the Iceman came into contact with the species, the place this happened was most likely to have been somewhere in the valley bottom of Vinschgau at less than 1,500 m above sea level.  相似文献

Isozyme electrophoresis was used to study the morphologically similar sister speciesSphagnum rubellum andS. capillifolium from a sample of 1313 plants representing 37 populations from Scandinavia, Great Britain and S Germany. The mean pairwise genetic identities (I) among conspecific populations were 0.976 forS. rubellum and 0.969 forS. capillifolium, versus 0.627 between populations of the two species. Interspecific gene flow was indicated by the observation of occasional plants in sympatric populations with alleles otherwise unique to the other species. Populations of bisexualS. capillifolium were significantly more variable than populations of unisexualS. rubellum. Alpine populations ofS. rubellum andS. capillifolium were dominated by few genotypes, and differentiation among populations was pronounced, indicating a low level of sexual recombination. InS. rubellum, maximum variability was found in western areas with high annual precipitation. Distribution of alleles inS. rubellum indicated restricted gene flow between Great Britain and Scandinavia. Postglacial migration from separate refugia may explain large-scale variation inS. rubellum.  相似文献

Abstract

Genetic diversity in eight populations of Sphagnum capillifolium from different Bulgarian mountains was investigated by means of isozyme electrophoresis. High levels of allelic diversity were found (HS = 0.119), comparable to earlier estimates for northern European populations (HS = 0.116). Strong differentiation among populations and a low number of widespread genotypes suggest a high degree of isolation and restricted gene flow between populations, which is consistent with generally small and scattered populations. The large proportion of distinguishable genotypes (mean 0.498) suggests high levels of out-crossing either currently or in the past. Introgression between S. capillifolium and S. rubellum, a species not found in Bulgaria, was suggested by the occurrence of rubellum-alleles in five populations from different mountains. This could be explained by an ancient hybridization event in a sympatric population. Based on (1) the high genetic diversity, (2) the fairly wide distribution of alien alleles, and (3) the isolated distribution of populations even within one mountain, a possible survival of S. capillifolium in the Balkan area during the Quaternary ice periods is hypothesized.  相似文献

Genetic variability within and among 10 geographically distinct populations of Greenfinches (Carduelis chloris) was assayed by directly sequencing a 637 BP part of the mtDNA control region from 194 individuals. Thirteen variable positions defined 18 haplotypes with a maximum sequence divergence of 0.8%. Haplotype (h = 0.28–0.77) and nucleotide (π = 0.058–0.17%) diversities within populations were low, and decreased with increasing latitude (h:rs = –0.81; π: rs = –0.89). The distribution of pairwise nucleotide differences fit better with expectations of a “sudden expansion” than of an “equilibrium” model, and the estimates of long term effective population sizes were considerably lower than current census estimates, especially in northern European samples. Selection is an unlikely cause of observed patterns because the distribution of variability conformed to expectations of neutral infinite alleles model and haplotype diversity across populations was positively correlated with heterozygosity (HE) in nuclear genes (rs = 0.74, P < 0.05). Hence, a recent bottleneck, followed by serial bottlenecking during the process of post-Pleistocene recolonization of northern Europe, together with recent population expansion provide a plausible explanation for the low genetic diversity in the north. Genetic distances among populations showed a clear pattern of isolation-by-distance, and 14% of the haplotypic variation was among populations, the rest being distributed among individuals within populations. In accordance with allozyme and morphological data, a hierarchical analysis of nucleotide diversity recognized southern European populations as distinct from northern European ones. However, the magnitude of divergence in mtDNA, allozymes and morphology were highly dissimilar (morphology > mtDNA > allozymes).  相似文献

The Baltic coast of Northern Poland is an interesting region for palaeoclimatic studies because of the mixed oceanic and continental climatic influences and the fact that the dominance of one or the other of these two influences might have changed over time. Also, unlike many more intensively studied regions of Europe, human impact in the region was rather limited until the 19th century. We present a 1200-year high-resolution record from Stążki mire, an ombrotrophic bog located 35 km from the Baltic Sea coast. Using testate amoebae, stable isotopes (δ13C) of Sphagnum stems, pollen, plant macrofossils and dendroecological analyses, our aims were to: 1) reconstruct the last millennium palaeoenvironment in the study site and its surroundings, 2) identify the major wet–dry shifts, 3) determine if those events correlate with climate change and human impact, 4) assess the resilience of the Baltic bog ecosystem following human impact, and 5) compare the palaeo-moisture signal from the Baltic coast with records from more oceanic regions. Two dry periods are inferred at AD 1100–1500 and 1650–1900 (–2005). The first dry shift is probably climate-driven as pollen record shows little evidence of human indicators. The second dry shift can be related to local peat exploitation of the mire. In the 20th century additional limited drainage took place and since ca. AD 1950 the mire has been recovering. From 1500 AD onwards all proxies indicate wetter condition. The beginning of this wet shift occurred during the Little Ice Age and may therefore be a climatic signal. The macrofossil data show that Sphagnum fuscum dominated the pristine mire vegetation but then declined and finally disappeared at ca. AD 1900. This pattern is comparable with the timing of extinction of Sphagnum austinii (= Sphagnum imbricatum) in the UK. This study illustrates the value of high-resolution multi-proxy studies of peat archives to assess the magnitude of anthropogenic land-use changes. This study presents the first direct comparison of testate amoebae and stable isotope data from the same core. The two proxies correlate significantly throughout the record and most strongly for the latter part of the record when most of the variability was recorded.  相似文献

The Pô River basin of northern Italy is the home of distinctive and endemic morphological forms of brown trout Salmo trutta. We used PCR-direct sequencing and RFLP techniques to study variation in the mitochondrial control region of 225 trout in order to assess genetic relatedness among 18 populations from that region. The distribution analysis of these genotypes among north Italian populations confirmed the phylogenetic differentiation of marbled trout Salmo trutta marmoratus populations and the postglacial origin of S. t. fario. Extensive genetic heterogeneity was observed among morphologically identical S. t. fario populations. Introgression with domestic strains of Atlantic basin origin was detected in all forms. In order to assess the phylogenetic congruence detected in coding and noncoding regions of the mitochondrial genome, we also analysed sequence variation in segments of the cytochrome b and ATPase subunit VI genes among representatives of all variants detected in the analysis of the control region. Variation in protein coding genes was only slightly less than that observed in the control region of the same individuals, both in terms of number of variants detected and of pairwise sequence divergence estimates among variants. Phylogenetic analysis based on protein coding genes sequences identified the same phylogenetic groupings defined by the control region analysis and also allowed a partial resolution of their phyletic relationships that was previously unresolved. However, coding and noncoding segments differed substantially in the transition-transversion ratio (17:0 in coding segments vs. 17:6 in control region segments).  相似文献

Noninvasively collected genetic data can be used to analyse large‐scale connectivity patterns among populations of large predators without disturbing them, which may contribute to unravel the species’ roles in natural ecosystems and their requirements for long‐term survival. The demographic history of brown bears (Ursus arctos) in Northern Europe indicates several extinction and recolonization events, but little is known about present gene flow between populations of the east and west. We used 12 validated microsatellite markers to analyse 1580 hair and faecal samples collected during six consecutive years (2005–2010) in the Pasvik Valley at 70°N on the border of Norway, Finland and Russia. Our results showed an overall high correlation between the annual estimates of population size (Nc), density (D), effective size (Ne) and Ne/Nc ratio. Furthermore, we observed a genetic heterogeneity of ~0.8 and high Ne/Nc ratios of ~0.6, which suggests gene flow from the east. Thus, we expanded the population genetic study to include Karelia (Russia, Finland), Västerbotten (Sweden) and Troms (Norway) (477 individuals in total) and detected four distinct genetic clusters with low migration rates among the regions. More specifically, we found that differentiation was relatively low from the Pasvik Valley towards the south and east, whereas, in contrast, moderately high pairwise FST values (0.91–0.12) were detected between the east and the west. Our results indicate ongoing limits to gene flow towards the west, and the existence of barriers to migration between eastern and western brown bear populations in Northern Europe.  相似文献

Levels and distribution of genetic variation were analyzed in 15 populations of the homosporous fern Gymnocarpium dryopteris ssp. disjunctum to evaluate the mating system and the genetic structure of this taxon. Estimated rates of intragametophytic selling were 0.000 in all populations, signifying that all sporophytes examined arose via intergametophytic matings. Furthermore, values of F, the fixation index, ranged from –0.171 to 0.038, indicating that all populations approach Hardy-Weinberg equilibrium. Therefore, not only is intragametophytic selling rare in G. dryopteris ssp. disjunctum, but intergametophytic selling is also uncommon. Factors promoting outcrossing in G. dryopteris ssp. disjunctum include an antheridiogen system, an ontogenetic sequence that favors intergametophytic matings, and high levels of genetic load. Analysis of F-statistics indicated statistically significant genetic differentiation among populations despite a mean genetic identity of 0.973 and high levels of interpopulational gene flow (Nm = 3.41 to 4.09). These patterns may reflect levels of gene flow that prevent significant interpopulational divergence while permitting slight genetic differentiation among populations.  相似文献

 Isozyme markers and morphological characters were studied in four populations of Sphagnum capillifolium and S. quinquefarium. Recombinant plants were found in three populations, where the two species occur sympatrically. All recombinants possessed different haplotypes and combinations of morphological characters, which show that they are results of independent hybridization events. Strongly male-biased sex ratios were found for Sphagnum capillifolium in all populations where it grew sympatrically with S. quinquefarium. Most of the recombinants were also male fertile. These observations suggest that S. quinquefarium is the female parent in the primary crosses and in subsequent backcrosses. The aim of this study was to analyse the genetic structure of populations for seven common cyprinid fish species within a 120‐km‐long stretch of the lowland Elbe River, northern Germany. The results are needed for habitat modelling to estimate the proportion that environmentally based variance has of the total variances of home range, species distribution, habitat use and fish assemblage structure. Polymerase chain reaction (PCR)‐fingerprinting offers a rapid, efficient method for generating genetic markers and was therefore used to obtain an overview on population‐genetic structures of the following seven fish species: asp (Aspius aspius), bleak (Alburnus alburnus), blue bream (Abramis ballerus), common bream (Abramis brama), gudgeon (Gobio gobio), ide (Leuciscus idus) and roach (Rutilus rutilus). Of the 20 random primers, between eight (ide) and 18 (roach) produced polymorphic bands. The mean levels of genetic similarity between samples, estimated as bandsharing frequencies, varied between 76% in bleak and 98% in asp. The corresponding genetic distances among samples varied between 0.02 ± 0.01 in asp and 0.24 ± 0.09 in bleak. The genetic distances among samples were not significant in all of the pairwise comparisons, and correlated only weakly with the geographic distances among sampling sites. It was therefore concluded that the stretch of the Elbe surveyed was inhabited by single, panmictic populations of the species studied and thus that the observed habitat preferences, fish distribution, home range and ecological performance of species within this area will depend on stochastic environmental factors or result from biotic interactions.  相似文献

The relative effect of diffusional resistance due to water films (rwf) and leaf anatomy (rp) on rates of net photosynthesis and on-line measures of carbon isotope discrimination (Δ=Δδ13C) was investigated in Sphagnum. Sphagnum species differ in the exposure of photosynthetic cells at the leaf surface. In S. affine, photosynthetic cells are widely exposed at the surface, whereas in S. magellanicum, photo-synthetic cells are enclosed within water-filled hyaline cells. This difference is expected to lead to variation in diffusive resistance within leaves (rp). Net photosynthesis and on-line Δ were measured at two water contents: greenhouse water content (wet) and blotted dry (dry). Without correcting for respiration, on-line Δ values differed significantly between wet (23.7%o) and dry (30.9%o) plants. However, there was no significant difference between species means and no species × water content interaction. Corrections for respiration lowered Δ values by approximately 8.1%o and reduced the mean difference to 3.1%o, but did not alter the rank order of treatments. Net photosynthesis also decreased by 16% in wet plants, but there was no significant difference between the two species. In addition, five populations of S. affine and S. magellanicum grown in a common garden were analysed for their organic matter carbon isotope composition (δ13C). These values varied more within each species (0.9–1.2%o) than between the two species (0.6%o). Therefore, we conclude that variation in surface water films leads to a greater difference in resistance to CO2 uptake and carbon isotope discrimination than that due to variation in leaf anatomical properties in Sphagnum.  相似文献

1. Complete sequences of 1140 base pair of the cytochrome b gene from 133 specimens were obtained from nine localities including the inflow drainage system, isolated lakes and outflow drainage system in Qinghai–Tibetan Plateau to assess genetic diversity and to infer population histories of the freshwater fish Schizopygopsis pylzovi. 2. Nucleotide diversities (π) were moderate (0.0024–0.0045) in populations from the outflow drainage system and Tuosuo Lake, but low (0.0018–0.0021) in populations from Qiadam Basin. It is probable that the low intra‐population variability is related with the paleoenvironmental fluctuation in Qiadam Basin, suggesting that the populations from Qiadam Basin have experienced severe bottleneck events in history. 3. Phylogenetic tree topologies indicate that the individuals from different populations did not form reciprocal monophyly, but the populations from the adjacent drainages cluster geographically. Most population pairwise FST tests were significant, with non‐significant pairwise tests between Tuosu Lake and Tuosuo Lake in the north‐west of the Qinghai–Tibetan Plateau. Analysis of molecular variance (amova ) indicates that the significant genetic variation was explained at the levels of catchments within and among, not among specific boundaries or inflow and outflow drainage systems. 4. The nested clade phylogeographical analysis indicates that historical processes are very important in the observed geographical structuring of S. pylzovi, and the contemporary population structure and differentiation of S. pylzovi may be consistent with the historical tectonic events occurred in the course of uplifts of the Qinghai–Tibetan Plateau. Fluctuations of the ecogeographical environment and major hydrographic formation might have promoted contiguous range expansion of freshwater fish populations, whereas the geological barriers among drainages have resulted in the fragmentation of population and restricted the gene flow among populations. 5. The significantly large negative Fs‐value (−24.91, P < 0.01) of Fu's Fs‐test and the unimodal mismatch distribution indicate that the species S. pylzovi underwent a sudden population expansion after the historical tectonic event of the Gonghe Movement. 6. The results of this study indicate that each population from the Qinghai–Tibetan Plateau should be managed and conserved separately and that efforts should be directed towards preserving the genetic integrity of each group.  相似文献

Abstract 1 Seven organically grown cabbage fields were surveyed for entomopathogenic nematodes in the autumn by baiting. Nematodes were obtained from three fields with bait larvae infection ranging from 1.3–4.0%. 2 Inoculation of Steinernema feltiae (Rhabditida: Steinernematidae) in the spring increased bait larvae infection to 16.0–32.0%. 3 Four different species were found (Steinernema affine, Steinernema bicornutum, S. feltiae and Steinernema C1). 4 The number of Delia radicum (L.) (Diptera: Anthomyiidae) puparia was significantly reduced at plants where S. feltiae had been inoculated. 5 Delia radicum parasitoids were also affected by S. feltiae. The results indicated that Aleochara species (Coleoptera: Staphylinidae) were more negatively affected than Trypliographa rapae (Westw.) (Hymenoptera: Cynipidae).  相似文献

Several complexes of species in Sphagnum (peat mosses) originated through hybridization and allopolyploidy, suggesting that these processes have played a major evolutionary role in this genus. The Sphagnum subsecundum complex includes gametophytically haploid and diploid species in North America. Analyses of 12 microsatellite loci and sequences from two plastid DNA markers show that the evolutionary history of this group is substantially more complex than previously thought. Two taxonomic species, Sphagnum lescurii and Sphagnum inundatum, include both haploid and diploid populations. Within each ploidal level, S. lescurii and S. inundatum are not genetically differentiated. The diploid taxa show patterns of fixed heterozygosity for the microsatellite markers, consistent with an allopolyploid origin. Diploid S. lescurii is an allopolyploid between haploid S. lescurii and (haploid) S. subsecundum. Sphagnum carolinianum is an allopolyploid between haploid S. lescurii and an unknown parent. We detected homoploid hybridization between the haploids Sphagnum contortum and S. subsecundum. Chinese mitten crab (Eriocheir sinensis), a native species in China, has populated Continental Europe and North America since 1912. In this paper, the nucleotide variation in the fragments of mitochondrial COII (693 bp), Cytb (766 bp), and nucleotide ITS (706 bp) was analyzed in native (Yantgze, Yellow, and Liaohe Rivers in China) and colonized (Elbe, Rhine, and Thames Rivers in Europe, and San Francisco Bay in North America) populations of the Chinese mitten crab. The major findings are as follows. First, the genetic variability in the native populations is higher than that in the colonized European and North American populations, with the exception of the Elbe River population, which possesses a similar level of variability with the native populations. Second, a remarkable loss of singletons has been associated with the colonization of Chinese mitten crabs. Third, the AMOVA and F ST results demonstrate that there are no significant genetic differentiations among the populations from the three continents, but there is a significant differentiation between pairwise populations within and among continents. Fourth, it is found that expansion-drift and gene flow pattern are involved in the European populations. The neutrality test and R 2 statistics suggest that a moderate founder population exists in the colonized populations, and only the Yangtze River population has undergone a recent population expansion. Finally, the results demonstrate that the European populations originate from multiple rivers in China on multiple occasions. The San Francisco population originates from both the native Chinese populations and the colonized European populations, most likely the Thames population.  相似文献

In Central Europe, salt-influenced habitats are restricted mainly to the coastlines of the North Sea and the Baltic Sea. The few natural inland salt sites suffer from size reduction and loss in biodiversity, while anthropogenic salt sites around potash mining dumps experienced recent and massive de novo colonization by numerous halophytes. Our study aimed to elucidate the general patterns of gene flow among coastal and inland salt habitats. We used amplified fragment length polymorphisms to assess the genetic diversity and genetic structure of 18 European populations of the halophyte Spergularia media (Caryophyllaceae), sampled from inland and coastal salt sites of Germany, the Netherlands, Denmark, Austria, France and Italy. Estimates of genetic diversity on the population level were generally low, especially within inland salt habitats. Analyses of molecular variance showed comparatively strong differentiation among populations. Multivariate ordination (principal coordinate analysis) and a NeighborNet analysis revealed four distinct groups of populations that showed good correspondence to their geographic origins. A Bayesian analysis performed with the program Structure as well as high pairwise Φ ST values supported the results of the phenetic analyses. The observed patterns of diversity and differentiation among inland populations of S. media are best explained by the isolated nature of suitable salt habitats, with concomitant reduction of gene flow to and among these sites. Our data support the hypothesis that the colonization of anthropogenic salt sites by S. media originated from natural inland habitats.  相似文献

Bryophytes dominate some ecosystems despite their extraordinary sensitivity to habitat quality. Nevertheless, some species behave differently across various regions. The existence of local adaptations is questioned by a high dispersal ability, which is thought to redistribute genetic variability among populations. Although Sphagnum warnstorfii is an important ecosystem engineer in fen peatlands, the causes of its rather wide niche along the pH/calcium gradient are poorly understood. Here, we studied the genetic variability of its global populations, with a detailed focus on the wide pH/calcium gradient in Central Europe. Principal coordinates analysis of 12 polymorphic microsatellite loci revealed a significant gradient coinciding with water pH, but independent of geography; even samples from the same fens were clearly separated along this gradient. However, most of the genetic variations remained unexplained, possibly because of the introgression from phylogenetically allied species. This explanation is supported by the small heterogeneous cluster of samples that appeared when populations morphologically transitional to S. subnites, S. rubellum, or S. russowii were included into the analysis. Alternatively, this unexplained variation might be attributed to a legacy of glacial refugia with recently dissolved ecological and biogeographic consequences. Isolation by distance appeared at the smallest scale only (up to 43 km). Negative spatial correlations occurred more frequently, mainly at long distances (up to 950 km), implying a genetic similarity among samples which are very distant geographically. Our results confirm the high dispersal ability of peatmosses, but simultaneously suggested that their ability to cope with a high pH/calcium level is at least partially determined genetically, perhaps via specific physiological mechanisms or a hummock‐forming ability.  相似文献

Random amplified polymorphic DNA (RAPD) analysis was used to characterize the genetic diversity and population genetic structure of Stipa krylovii populations in Inner Mongolia steppe of North China. Thirteen 10-bp oligonucleotide primers, which generated 237 RAPD bands, were used to analyze 90 plants of five populations from three regions, meadow steppe, typical steppe and desert steppe, from the east to the west. The genetic diversity of Stipa krylovii that was revealed by observed number of alleles (na), expected number of alleles (ne), Nei’s diversity index (h), Shannon’s diversity index (H), amplificated loci, polymorphic loci and the percentage of polymorphic loci (PPB) increased from the east to the west. The Pearson’s correlation analysis between genetic diversity parameters and ecological parameters indicated that the genetic diversity of Stipa krylovii was associated with precipitation and cumulative temperature variations along the longitude (humidity were calculated by precipitation and cumulative temperature). Dendrogram based on Jaccard’s genetic distance showed that the individuals from the same population formed a single subgroup. Although most variation (56.85%) was within populations, there was high genetic differentiation among populations of Stipa krylovii, high differentiation within and between regions by AMOVA analysis. Either Nei’s unbiased genetic distance (G ST) or gene flow (Nm) among pairwise populations was not correlated with geographical distance by Mantel’s test (P > 0.05), suggesting that there was no consistency with the isolation by distance model in these populations. Natural selection may have played a role in affecting the genetic diversity and population structure, but habitat destruction and degradation in northem grassland in China may be the main factor responsible for high genetic differentiation among populations, within and among regions. Tobacco blue mold, caused by Peronospora tabacina, is an oomycete plant pathogen that causes yearly epidemics in tobacco (Nicotiana tabacum) in the United States and Europe. The genetic structure of P. tabacina was examined to understand genetic diversity, population structure and patterns of migration. Two nuclear loci, Igs2 and Ypt1, and one mitochondrial locus, cox2, were amplified, cloned and sequenced from fifty‐four isolates of P. tabacina from the United States, Central America–Caribbean–Mexico (CCAM), Europe and the Middle East (EULE). Cloned sequences from the three genes showed high genetic variability across all populations. Nucleotide diversity and the population mean mutation parameter per site (Watterson's theta) were higher in EULE and CCAM and lower in U.S. populations. Neutrality tests were significant and the equilibrium model of neutral evolution was rejected, indicating an excess of recent mutations or rare alleles. Hudson's Snn tests were performed to examine population subdivision and gene flow among populations. An isolation‐with‐migration analysis (IM) supported the hypothesis of long‐distance migration of P. tabacina from the Caribbean region, Florida and Texas into other states in the United States. Within the European populations, the model documented migration from North Central Europe into western Europe and Lebanon, and migration from western Europe into Lebanon. The migration patterns observed support historical observations about the first disease introductions and movement in Europe. The models developed are applicable to other aerial dispersed emerging pathogens and document that high‐evolutionary‐risk plant pathogens can move over long distances to cause disease due to their large effective population size, population expansion and dispersal.  相似文献

Swertia przewalskii Pissjauk. (Gentianaceae) is a critically endangered and endemic plant of the Qinghai-Tibet Plateau in China. RAPD and ISSR analyses were carried out on a total of 63 individuals to assess the extent of genetic variation in the remaining three populations. Percentage of polymorphic bands was 94% (156 bands) for RAPD and 96% (222 bands) for ISSR. A pairwise distance measure calculated from the RAPD and ISSR data was used as input for analysis of molecular variance (AMOVA). AMOVA indicated that a high proportion of the total genetic variation (52% for RAPD and 56% for ISSR) was found among populations; pairwise Φ ST comparisons showed that the three populations examined were significantly different (p < 0.001). Significant genetic differentiation was found based on different measures (AMOVA and Hickory θB) in S. przewalskii (0.52 on RAPD and 0.56 on ISSR; 0.46 on RAPD and 0.45 on ISSR). The differentiation of the populations corresponded to low average gene flow (0.28 based on RAPD and 0.31 based on ISSR), whereas genetic distance-based clustering and coalescent-based assignment analyses revealed significant genetic isolation among populations. Our results indicate that genetic diversity is independent of population size. We conclude that although sexual reproduction and gene flow between populations of S. przewalskii are very limited, they have preserved high levels of genetic diversity. The main factors responsible for the high level of difference among populations are the isolation and recent fragmentation under human disturbance.  相似文献
