通过添加鸡肉浸液改良经典PCA培养基获得模拟鸡肉营养条件的近自然的CEA培养基,用于检测鸡肉中对营养要求苛刻的污染细菌。根据添加不同浓度的新鲜鸡肉浸液所获得菌落总数的变化获得最佳添加浓度,并利用生化特性及16SrRNA序列对仅生长于CEA培养基的细菌进行了鉴定。在CEA培养基上获得的细菌数量和多样性都显著高于PCA培养基(P0.05),且分离到3株无法在PCA培养基上生长的细菌,经鉴定其与Enterococcus faecalis、Rothia mucllaginosa,Staphylococcus saprophyticus subsp.saprophyticus的相似性分别为99%、96%、99%。E.faecalis因产生生物胺而被认为是肉品中的腐败菌,后两者则均是条件致病菌株,他们的存在对鸡肉产品的安全可能造成隐患。该方法中的近自然培养法能提高对微生物数量和种类的检测灵敏度,特别适于对营养要求苛刻的污染细菌的检出。  相似文献

蕙兰根内可培养细菌的物种多样性   总被引:1,自引:0,他引:1  
以MS基本培养基添加蕙兰菌根浸出液制成的培养基进行分离培养的方法,从野生蕙兰(Cymbidium faberi)根部首次分离到内生细菌。经过分离纯化培养获得纯菌株27株。经过16S rDNA基因序列测序,并与GenBank数据比对,其相似性均在98%以上,分析鉴定结果表明,存活的22株菌可分为8属14种。分别隶属于伯克氏菌属(Burkholderia)、假单胞菌属(Pseudomonas)、芽胞杆菌属(Bacillus)、Leifsonia属、贪食菌属(Variovorax)、欧文氏菌属(Erwinia)、Duganella属和不动杆菌属(Acinetobacter)。对这些菌株进行分离培养及鉴定有助于理解兰花与微生物之间的相互作用关系,为开发利用这些微生物开辟新的思路。  相似文献

摘要:【目的】为了了解塔河废弃古河道胡杨可培养内生细菌的多样性。【方法】从2棵胡杨树干部抽出其内存液,采用三种不同的培养基对样品的内生细菌进行了分离纯化;对它们进行16S rDNA测定和系统进化分析。【结果】分离纯化不同表型的细菌62株,对它们的16S rDNA序列分析表明,62株菌分别属于四个大类群;厚壁菌门(Firmicutes)、放线菌门(Actinobacteria)、α-变形菌纲(Alpha Proteobacteria) 、γ-变形菌纲(Gamma Proteobacteria),18个属,32个种;芽孢杆菌属和假单胞菌属是胡杨可培养内生细菌的优势细菌种群,它们分别占已测种群的40.32%、16.13%。其中菌株KTH-63为葡萄球菌科的潜在的新属新种,它与最近源菌株的16S rDNA序列相似率为92.491%;9株菌KLH-21、KLH-1、KTH-8、KTH-14、KNA-26、KLH-18、KTH-20、KNA-3、KLH-25是潜在的新种(16S rDNA相似率为96.089 %-97.769 %),胡杨树干内存液中潜在新种的发现率高达总分离检测菌株的16.13 % 。本研究获得的胡杨可培养内生细菌的群落结构数据给植物内生细菌新增了10个属,18个种。【结论】胡杨具有多样性极其丰富的可培养内生细菌菌种资源,土著新种的发现频率超出了预期,胡杨可培养内生细菌的群落结构极大地刷新了植物内生细菌的种群记录,极具进一步发掘的潜力。  相似文献

东海表层沉积物纯培养与非培养细菌多样性   总被引:2,自引:0,他引:2  
于2011年7月采集了浙江舟山沿岸海底沉积物样品(122°10′41″E,29°49′7″N),通过埋片原位观察、荧光显微计数、纯培养菌分离以及非培养细菌构建克隆文库的方法,分析和研究了东海表层沉积物细菌群落结构及其多样性.埋片原位观察和荧光显微计数法的结果表明:沉积物样品中细菌的细胞数量为(9.30±3.44)×107 cells/g;通过分离纯化共获得313株细菌,分属于20个属,4种培养基对细菌的分离效果依次为RO>M l>Zobell 2216>MR2A;常规形态学与生理生化鉴定结果显示芽孢杆菌属(Bacillus)和海球菌属(Marinococcus)从数量和种类上皆为优势菌属,分别占分离菌株总数的21.08%和17.25%;对73株典型海洋细菌进行16S rDNA分子鉴定发现,厚壁菌门(57.5%)、γ-变形杆菌纲(32.9%)、黄杆菌纲(4.1%)和放线菌纲(5.5%)等为主要类群.非培养细菌克隆文库序列分析发现:细菌克隆子主要属于厚壁菌门和变形杆菌门,而芽孢杆菌纲和γ-变形杆菌纲是上述两个门的优势类群.综合纯培养与非培养数据得出东海海域表层沉积物细菌多样性丰富,具有进一步开发研究的价值.  相似文献

广西水牛瘤胃中的细菌多样性   总被引:1,自引:0,他引:1  
[目的]了解广西水牛瘤胃中细菌的组成及其可能的降解纤维素细菌的主要类群。[方法]提取水牛瘤胃内容物和高效降解滤纸的水牛瘤胃内容物的富集培养物的宏基因组DNA,以宏基因组DNA为模板,扩增16S rRNA基因序列,构建该两种样品的细菌的16S rRNA基因文库。通过对16S rRNA基因序列的分析,了解这两种样品的细菌群体种类及数量。 [结果] 水牛瘤胃内容物与其富集培养物中均主要含有LGCGPB (low G+C Gram-Positive Bacteria)、CFB (Cytophaga-Flexibacter-Bacteroides)两大类菌群和少数的螺旋体菌(Spirochaetes),且LGCGPB所占的比例都是最高的,LGCGPB在水牛瘤胃内容物细菌中的比例为56.66%,而在富集培养物细菌中的比例升高为73.33%。在水牛瘤胃内容物中,丝状杆菌(Fibrobacteres)占3.33%,但在富集培养物中未被检测到。而在富集培养物中占13.33%的变形杆菌(Proteobacteria),在水牛瘤胃内容物中未被检测到。本研究还发现了分类地位尚未明确的一菌群(R46)。[结论]细菌类群LGCGPB、Proteobacteria可能在水牛瘤胃中的纤维素降解过程中起重要作用。此外,水牛瘤胃中的细菌组成和牦牛、牛、羊瘤胃中的细菌组成较相似但比例有所不同。  相似文献

玉溪地区自然陈化烟叶表面可培养细菌多样性研究   总被引:1,自引:0,他引:1  
目的为了较全面的了解陈化烟叶的细菌多样性,进一步挖掘其中的有益微生物。方法以种植于玉溪元江县的红大烤烟品种为材料,利用16SrDNA克隆测序技术,系统研究了云南玉溪红塔区和元江县两仓库中不同自然陈化时期烟叶表面可培养微生物的种群结构。结果陈化初期,烟叶表面细菌数量较少,元江县低水分烟叶陈化30d时细菌数量达到高峰;红塔区高水分烟叶陈化4.5个月时细菌数量达最大;经过16SrDNA鉴定,烤烟叶面细菌包括芽孢杆菌属(Bacillus sp.)、肠杆菌属(Enterobacter sp.)、类芽孢杆菌属(Paenibacillus sp.)和泛菌属(Pantoeasp.)等8个细菌属。结论不同时期、不同地点陈化烟叶表面细菌的优势种群不尽相同,而芽孢杆菌属始终为优势种群。另外,研究中分离到的大量可培养细菌,为进一步筛选烟叶表面有益细菌提供了重要线索。  相似文献

【目的】为较系统地了解宜宾浓香型白酒酿造过程中可培养细菌的多样性,得到一些潜在的微生物资源。【方法】采用改良的NA培养基和高氏I号培养基分离、去除冗余,测定所得细菌纯培养物的16S rRNA基因,进行系统发育分析。【结果】分离得到603株细菌,4株菌的序列与GenBank中典型菌株序列相似性低于97%,代表着潜在新类群;599株菌与GenBank中34个属、101个种的典型菌株序列相似性大于97%,其中以Bacillus为绝对优势菌(315株),Streptomyces(121株)、Lysinibacillus(35株)、Staphylococcus(45株)为次优势菌,其余各属菌株均在10株以下。而且有16个属均只检测到1株菌。【结论】宜宾浓香型白酒发酵过程中的细菌呈现出较为丰富的多样性和一定的稳定性。  相似文献

细菌作为微生物中的重要组成部分, 在有机质的形成与分解、维持生态系统平衡、促进动植物发育等多方面都有着重要作用。2014-2017年, 我们采用常见培养基分离纯培养法及16S rRNA基因序列分析方法对大小兴安岭地区土壤可培养细菌的群落结构和多样性进行了调查研究。结果表明: 从大小兴安岭地区的17个自然保护区内不同生境的土壤中分离获得3,180个菌株, 隶属于24属120种。其中, 芽孢杆菌属(Bacillus)的种数和株数最多, 分别为38种和2,419株, 是大小兴安岭地区可培养细菌的绝对优势类群(占总株数的76.1%); 其次是短杆菌属(Brevibacterium)(13.0%)。大兴安岭地区的物种数、Simpson多样性指数和Shannon-Wiener指数高于小兴安岭地区。优势类群芽孢杆菌属的枯草芽孢杆菌(B. subtilis)、苏云金芽孢杆菌(B. thuringiensis)、巨大芽孢杆菌(B. megaterium)等有重要的生产及科学研究价值。  相似文献

16S rDNA library-based analysis of ruminal bacterial diversity   总被引:13,自引:0,他引:13  
Bacterial 16S rDNA sequence data, incorporating sequences > 1 kb, were retrieved from published rumen library studies and public databases, then were combined and analysed to assess the diversity of the rumen microbial ecosystem as indicated by the pooled data. Low G+C Gram positive bacteria (54%) and the Cytophaga-Flexibacter-Bacteroides (40%) phyla were most abundantly represented. The diversity inferred by combining the datasets was much wider than inferred by individual studies, most likely due to different diets enriching for bacteria with different fermentative activities. A total of 341 operational taxonomic units (OTU) was predicted by the Chao1 non-parametric estimator approach. Phylogenetic and database analysis demonstrated that 89% of the diversity had greatest similarity to organisms which had not been cultivated, and that several sequences are likely to represent novel taxonomic groupings. Furthermore, of the 11% of the diversity represented by cultured isolates (> 95% 16S rDNA identity), not all of the bacteria were of ruminal origin. This study therefore reinforces the need to reconcile classical culture-based rumen microbiology with molecular ecological studies to determine the metabolic role of uncultivated species.  相似文献

本文比较分析了相同销售条件下的未包装冷却牛肉与保鲜膜包装冷却牛肉中的微生物群落结构。采用16S rDNA的V3区片段进行PCR-DGGE(变性梯度凝胶电泳), 同时利用16S rDNA全长序列, 通过克隆文库技术对两个样品的微生物群落结构进行研究。研究发现: 微生物群落结构表现出较大的差异。保鲜膜包装冷却牛肉共有6个OTU (Operational taxonomic unit), 主要为Lactococcus(乳球菌属, 28%), Lactobacillus(乳杆菌属, 26%), Carnobacterium(肉品杆菌属, 18%)和Brochothrix(索丝菌属, 10%); 未包装冷却牛肉则共得到18个OTU, 主要是Lactococcus(乳球菌属, 28%), Brochothrix(索丝菌属, 18%)和Acinetobacter(不动杆菌属, 11%)。结果表明, 保鲜膜包装对于葡萄球菌属以及冷杆菌属等一些细菌都起到了一定的抑制作用。本研究为肉类加工生产中微生物的控制提供一定的理论依据。  相似文献

通过构建16S rDNA克隆文库的方法,分析太岁样品中细菌的群落结构及多样性。太岁样品中的细菌归属于4个门9个目,优势类群依次是芽胞杆菌目(Bacillales,33.01%)、柄杆菌目(Caulobacterales,32.04%)和伯克霍尔德氏菌目(Burkholderiales,12.62%);优势属为短波单胞菌属(Brevundimonas,30.10%)、葡萄球菌属(Staphylococcus,29.13%)和食酸菌属(Acidovorax,7.77%)。并且其中的5个目中含有未培养的细菌,红杆菌目(Rhodobacterales)、伯克霍尔德氏菌目和红环菌目(Rhodocyclales)的11个克隆子的细菌16S rDNA序列同源性低于97%。研究表明太岁样品中细菌多样性较丰富,且蕴藏着许多未知的微生物资源。  相似文献

Abstract: Molecular phylogeny increasingly supports the understanding of organismal relationships and provides the basis for the classification of microorganisms according to their natural affiliations. Comparative sequence analysis of ribosomal RNAs or the corresponding genes currently is the most widely used approach for the reconstruction of microbial phylogeny. The highly and less conserved primary and higher order structure elements of rRNAs document the history of microbial evolution and are informative for definite phylogenetic levels. An optimal alignment of the primary structures and a careful data selection are prerequisites for reliable phylogenetic conclusions. rRNA based phylogenetic trees can be reconstructed and the significance of their topologies evaluated by applying distance, maximum parsimony and maximum likelihood methods of phylogeny inference in comparison, and by fortuitous or directed resampling of the data set. Phylogenetic trees based on almost equivalent data sets of bacterial 23S and 16S rRNAs are in good agreement and their overall topologies are supported by alternative phylogenetic markers such as elongation factors and ATPase subunits. Besides their phylogenetic information content, the differently conserved primary structure regions of rRNAs provide target sites for specific hybridization probes which have been proven to be powerful tools for the identification of microbes on the basis of their phylogenetic relationships.  相似文献

采用分子生物学方法构建16SrDNA基因文库,研究鸡粪沼气池发酵液中产甲烷菌的菌群结构。随机分析文库中50个克隆的16SrDNA基因序列,结果发现,其中46个克隆属于产甲烷菌属,与Methanogenium marinum strain AK-1菌株的相似性为99%-100%,占总数的92%;3个克隆(KD525、KD526和KD567)属于甲烷袋状菌属,与Methanoculleus sp.dm2菌株的相似性均为99%,占总数的6%;1个克隆(KD519)属于甲烷粒菌属,与Methanocorpusculum bavaricum菌株的相似性为99%,占总数的2%。另外,同样分析了样品中甲基辅酶M还原酶alpha亚基mcrA基因氨基酸序列的差异。气相色谱法分析结果显示,发酵液中甲酸含量为28.85g/L,约占总有机酸含量的81.7%;沼气的主要成分为甲烷、二氧化碳和氢气,分别约占总气体量的55.5%、41.1%和3.2%。  相似文献

从连云港台南盐场海盐生产区中分离纯化到一株嗜盐古菌HBCC-2,该菌株经PCR扩增后,测定其16S rRNA基因序列,采用BLAST软件对基因库中基因序列进行同源性比较,选取其相似性序列,采用Clustalx1.8和MEGA3.1软件对其16S rDNA序列进行了系统发育分析研究,结果表明HBCC-2菌株与菌株Halorubrum sp.GSL5.48的相似性达99%,结合其形态观察及生理生化反应特性,初步确定该菌株属于嗜盐红菌属(Halorubrum),菌株HBCC-2的16S rDNA序列已登陆到GenBank,其序列号为EF687739.通过比较不同NaCl浓度、pH和培养温度对该菌株生长的影响情况,研究了该菌株的生长特性,结果表明NaCl浓度为4mol/L、温度为35℃和pH为7.0的培养条件下其生长最佳.  相似文献

The gut bacterial community from four species of feral locusts and grasshoppers was determined by denaturing gradient gel electrophoresis (DGGE) analysis of bacterial 16S rRNA gene fragments. The study revealed an effect of phase polymorphism on gut bacterial diversity in brown locusts from South Africa. A single bacterial phylotype, consistent with Citrobacter sp. dominated the gut microbiota of two sympatric populations of Moroccan and Italian locusts in Spain. There was evidence for Wollbachia sp. in the meadow grasshopper caught locally in the UK. Sequence analysis of DGGE products did not reveal evidence for unculturable bacteria and homologies suggested that bacterial species were principally Gammaproteobacteria from the family Enterobacteriaceae similar to those recorded previously in laboratory reared locusts.  相似文献

The chicken cecum contains a great many bacteria, most of which are strict anaerobes. A strictly anaerobe culture-based method was used in the present study, in conjunction with the 16S rDNA clone library, to elucidate bacterial diversity and the phylogenetic relationship of cecal microbiota in the chicken. A comparative 16S rDNA sequence analysis of cultivated strains and retrieved clones from cecal contents was performed. Approximately 90% of the bacterial cells detected by microscopy did not form colonies on a medium 10 in plate-in-bottle. The 19 isolated strains yielded 11 distinct rDNA sequences, 58% of which were classified as low G + C gram-positive bacteria, 26% were related to Bacteroides spp., and 16% were classified as Proteobacteria. Based on the sequence analysis of 164 clones, 24% were identified to belong to 8 known species and 76% were considered to be 65 novel phylotypes. Approximately 94% of cloned sequences were classified into low G + C gram-positive bacteria, 4% were related to Bacteroides spp., and 2% were classified into Proteobacteria. Clostridium subcluster XIVa (38%), Clostridium cluster IV (13%), Lactobacillus spp. (24%), and Bacteroides spp. (4%) were the major groups constituting the cecal microbiota in chicken, in which the Clostridium subcluster XIVa was the most phylogenetically diverse group in chicken cecum. The 16S rDNA sequences of Lactobacillus acidophilus, L. crispatus, L. salivarius, and L. reuteri were the most frequently found in the Lactobacillus group in chicken cecum.  相似文献
