In the Elbe hardly anything is known about the actual fate of phytoplankton and the resultant pigment composition of suspended matter. As part of a longterm study on the role of suspended matter in the cycles of nitrogen and associated oxygen consumption processes in the Elbe estuary in northern Germany (‘Sonderforschungsbereich 327 Elbe’ project) and the characterization of estuarine and coastal water bodies with regard to the transport of pollutants such as heavy metals, we have started to characterize suspended matter with regard to the pigments present therein. This will allow us to study phytoplankton turnover and to determine the role of phytoplankton decay mechanisms in the mobilization, binding, and transport of pollutants associated with particulate matter. Our first data on the pigment composition of suspended matter from the Elbe and particularly over the region of Hamburg harbour, obtained using a High Performance Liquid Chromatography (HPLC) method, is presented. The pigment concentrations varied considerably over the summer months. Values up to 250 mg l−1 of chlorophyll a were observed upstream of Hamburg. We found that on its course through Hamburg the concentrations (μg g−1 of particulate matter) of all pigments in the Elbe drop to under half those found upstream of Hamburg. The pigment concentrations in the turbidity maximum were significantly lower than in the rest of the estuary due to the decline of algae as a result of inadequate light conditions and the salinity gradient. The highest particulate matter concentrations were found in the turbidity maximum and just downstream of Hamburg. The highest particulate organic carbon values were found just downstream of Hamburg. The presence of of fucoxanthin and the chlorophyllsc andb, and the changes in pigment patterns were indicative for the dominant algal classes and the phytoplankton succession in the suspended matter. The ratio of chlorophylla to lutein was found to be a possible indicator of phytoplankton breakdown.  相似文献

Heterotrophic bacterial and phytoplankton biomass, production, specific growth rates and growth efficiencies were studied in July 2001 and January 2002 during both spring and neap tides, along a tidal cycle, at three sites in a subtropical estuary. Major freshwater inputs located in the Northern region led to differences in both phytoplankton and bacterioplankton biomass and activity along the estuary. While in the Northern region phytoplankton is light-limited, with mean phytoplankton production (PP) between 1.1 and 1.9 μg C l−1 h−1 and mean specific growth rates (PSG) between 0.14 and 0.16 d−1, the Southern region registered values as high as 24.7 μg C l−1 h−1 for PP and 2.45 d−1 (mean PP between 3.4 and 7.3 μg C l−1 h−1; mean PSG between 0.28 and 0.57 d−1). On the other hand, maximum bacterial production (BP: 63.8 μg C l−1 h−1) and specific growth rate (BSG: 32.26 d−1) were observed in the Northern region (mean BP between 3.4 and 12.8 μg C l−1 h−1; mean BSG between 1.98 and 6.67 day−1). These bacterial activity rates are among the highest recorded rates in estuarine and coastal waters, indicating that this system can be highly heterotrophic, due to high loads of allochthonous carbon (mainly derived from mangrove forest). Our results also showed that, despite that BP rates usually exceeded PP, in the Southern region BP may be partially supported (∼45%) by PP, since a significant regression was observed between BP and PP (r = 0.455, P < 0.001). Typical features of particulate phosphorus in the Seine estuary (France)   总被引:1,自引:1,他引:0  
During the 2001–2002 hydrological cycle, 8,000 tons P year−1 (44% as particulate phosphorus) originating from the Seine basin entered the Seine estuary. P content in suspended sediments (SS) is 2.9 g P kg−1 (80% as inorganic form) at Poses (the upstream limit of the Seine estuary). During the transfer from Poses to Caudebec (the limit of saline water intrusion), Particulate Inorganic Phosphorus (PIP) in SS decreases by 40% whereas Particulate Organic Phosphorus (POP) remains stable. This decrease is explained as the result of (i) a dilution by SS poorer in P, originating from storage zones (mudflats) within the estuary, and (ii) a loss of P by sedimentation, especially in the Rouen harbour where 15% to 20% of SS are yearly trapped and extracted. Downstream, in the turbidity maximum of the estuary, P content in SS is twice as low as at Poses (1.5 g P kg−1, 70% as inorganic form). PIP content is fairly homogeneous both vertically and throughout an annual survey. On the other hand, POP varies by season. Higher POP content is observed during vernal period with phytoplankton accumulation. In a salinity gradient from 0 to 30 (PSS78), PIP content further decreases by 30%. POP does not vary much in this gradient, leading to the assumption that PIP is submitted to desorption in response to the dilution by marine waters (poor in orthophosphates). Using 32P isotopic method, we establish a mathematical formulation of P exchangeable between the suspended solid and water phases of the Seine estuary. Particulate P in the turbidity maximum zone is shown to represent a possible source of dissolved P, available for algal growth in the Seine Bight. Instead of playing a role of nutrient retention, the turbidity maximum zone of the Seine estuary could favour coastal eutrophication.  相似文献

The pigment fingerprints, determined by HPLC, of suspended matter from different areas of the German Bight and from the Elbe Estuary are presented. These include areas with suspended matter concentrations varying between 10 mg l–1 in the deeper waters in the middle of the Bight and 150 mg l–1 in the Elbe mouth. Pigment data allows the identification of three groups of suspended matter (marine, turbidity zone and Elbe) for the numerous different locations. The changes in pigment concentrations in the suspended matter over tidal cycles are considered. The presence of pigments and pigment breakdown productsi.e. lutein and chlorophyllides is used to assess the possible condition of the phytoplankton present in the suspended matter. These pigment characteristics are compared with the loading of the heavy metals cadmium, copper, manganese, iron, zinc and lead in the particulate matter. It has been demonstrated that the pigments of the planktonic material can be used very effectively to identify different types of suspended matter and that the heavy metal loadings of the suspended matter were significantly correlated with chlorophyll-a.  相似文献

During January 1989, phytoplankton biomass and species composition were studied in a north / south transect at the Weddell / Scotia Confluence (47°W), between 57° and 61°30′S. Results showed a diatom bloom in the Scotia Sea (chlorophyll a 1.9 μg l−1, particulate organic carbon 239 μg l−1), dominated by Fragilariopsis cylindrus, Dactyliosolen antarcticus and Chaetoceros dichaeta. Low chlorophyll a / phaeopigments ratios (about 1.4) and silicate concentrations (15 μmol l−1) suggested that this was an advanced bloom phase, probably linked to high grazing pressure. Minimum chlorophyll a values of 0.1–0.2 μg l−1 and particulate organic carbon 46 μg l−1 were found at the Weddell / Scotia Front and in a subsurface layer of the Weddell Sea Water. In the southern part of the transect (61°30′S), in the Weddell Sea, a second surface maximum was found (chlorophyll a 0.9 μg l−1, particulate organic carbon 120 μg l−1), but with a different species composition, with Cryptomonas sp. dominant. Our results show a succession within the diatom community in the Weddell / Scotia Confluence Waters when comparing the three EPOS legs. In the Weddell Sea from spring to summer, nanoflagellates, with only a minor contribution from diatoms, persist over a long period with little change in the community structure. We suggest that the frontal system, together with the receding ice edge and the grazing pressure of either krill or protozooplankton, are mainly responsible for the phytoplankton distribution patterns found. Eutrophication has become increasingly serious and noxious algal blooms have been of more frequent occurrence in the Yangtze River Estuary and in the adjacent East China Sea. In 2003 and 2004, four cruises were undertaken in three zones in the estuary and in the adjacent sea to investigate nitrate (NO3–N), ammonium (NH4–N), nitrite (NO2–N), soluble reactive phosphorus (SRP), dissolved reactive silica (DRSi), dissolved oxygen (DO), phytoplankton chlorophyll a (Chl a) and suspended particulate matter (SPM). The highest concentrations of DIN (NO3–N+NH4–N+NO2–N), SRP and DRSi were 131.6, 1.2 and 155.6 μM, respectively. The maximum Chl a concentration was 19.5 mg m−3 in spring. An analysis of historical and recent data revealed that in the last 40 years, nitrate and SRP concentrations increased from 11 to 97 μM and from 0.4 to 0.95 μM, respectively. From 1963 to 2004, N:P ratios also increased from 30–40 up to 150. In parallel with the N and P enrichment, a significant increase of Chl a was detected, Chl a maximum being 20 mg m−3, nearly four times higher than in the 1980s. In 2004, the mean DO concentration in bottom waters was 4.35 mg l−1, much lower than in the 1980s. In comparison with other estuaries, the Yangtze River Estuary was characterized by high DIN and DRSi concentrations, with low SRP concentrations. Despite the higher nutrient concentrations, Chl a concentrations were lower in the inner estuary (Zones 1 and 2) than in the adjacent sea (Zone 3). Based on nutrient availability, SPM and hydrodynamics, we assumed that in Zones 1 and 2 phytoplankton growth was suppressed by high turbidity, large tidal amplitude and short residence time. Furthermore, in Zone 3 water stratification was also an important factor that resulted in a greater phytoplankton biomass and lower DO concentrations. Due to hydrodynamics and turbidity, the open sea was unexpectedly more sensitive to nutrient enrichment and related eutrophication processes.  相似文献

Water samples and particulate materials settling under the pack ice were collected in an ice-covered area near the Terra Nova Bay Italian Station during late summer 1995, in order to study short-term changes in the biochemical composition of particulate organic matter. At the end of the study period the phytoplankton biomass increase (up to >3.0 μg chlorophyll-a l−1) was probably related to the intrusion under the pack ice of chlorophylls-enriched surface waters coming from the near ice-free area. Such increase was associated also with a notable increase in particulate organic matter concentrations, as well as in particulate organic matter vertical fluxes (up to >100 mg C m−2 day−1). Proteins were the most abundant biochemical class of particulate organic matter (on average about 49%), followed by lipids (29%) and carbohydrates (22%). By contrast, organic matter collected in the sediment trap was characterized by the dominance of lipids (about 55% of the total biopolymeric carbon flux) over carbohydrates (28%) and proteins (17%). The hydrolizable particulate biopolymeric carbon accounted for about 23% of total biopolymeric carbon. This value was about one-half of that found in ice-free waters, suggesting that the suspended particulate organic material under the pack ice was less digestible than in ice-free waters or was already partially digested. Despite this, and the decay of labile organic compounds in the sediment trap during the deployment, material settling towards the sea bottom under the pack ice in Terra Nova Bay, owing to its high lipid content, might represent an important high-quality food source for benthic consumers. Finally, assuming as possible the intrusion under sea ice of primary organic matter-enriched waters, we hypothesize the occurrence of a “fertilization” effect deriving from ice-melting areas towards under-ice waters, supplying the latter with an additional rate of primary organic matter. The biogeochemical controls on particulate phosphorus distribution in the Humber estuary have been investigated with regard to salinity and season. Surveys of the Humber were conducted in August 1991 and January, March and June 1992. Samples were collected for particulate phosphorus, dissolved phosphorus, suspended load and salinity analysis. Particulate phosphorus forms 16 to 93% of the total phosphorus present in the water column of the Humber estuary. Total particulate phosphorus concentrations in the Humber ranged from 1 to 45 μmol l−1. This range is comparable to that observed in similar sized industrialized estuaries of the U.S.A. and northern Europe. Flocculation at low salinities was found to be a major influence on particulate phosphorus concentrations. Highest concentrations of particulate phosphorus were found in reaches below a salinity of 5 and may result in phosphorus being trapped in the sediments of the estuary. There is evidence that this phosphorus may be released as oxygen concentrations decrease during the summer. Suspended load, adsorption/desorption equilibrium and external inputs were also found to influence the particulate phosphorus concentrations.  相似文献

In June 1992, an extensive investigation programme was carried out in the turbidity zone of the Elbe river. Special attention was paid to salinity and suspended particulate matter (SPM) and their influence on bacteria, phytoplankton, zooplankton, mysids and fish. SPM was separated into three fractions of different settling velocities. Mean settling velocity (ws) was 0.05 cm s−1. The major part of SPM belonged to the slow settling fraction (ws<0.02 cm s−1). Bacterial exoenzymatic activity showed a positive correlation with SPM and chlorophyll-a content, and also to total dissolved free amino acids. Phytoplankton biomass reached maximum values of 5.7 μg chlorophyll-a l−1 at ebb tide. Chlorophyll-a correlated negatively with salinity, indicating riverine input of phytoplankton. A positive correlation was found between chlorophyll-a and dissolved oxygen. Abundance of zooplankton species and their developmental stages varied over the tidal cycles; abundance of cirriped larvae and copepodite stages of the dominant speciesEurytemora affinis (Copepoda, Crustacea) was positively correlated with salinity. Individual filtering rates (IFR) ofEurytemora affinis were negatively affected by the SPM content of the water. Maximum IFR for adults was 7.2 ml h−1. Community grazing reached maximum rates of 30.3 ml l−1 h−1 (i.e. 72.7% d−1). The dominant mysidNeomysis integer showed maximum abundance at night, possibly resulting from diel vertical migration. Abundance ofN. integer was positively correlated with SPM content. The fish community, consisting of 17 fish species, was characterised by high densities of smelt (Osmerus eperlanus). A positive correlation was found between salinity and abundance of typical marine fish species, such as sprat (Sprattus sprattus). Similar temporal variation of abundance of smelt, sprat, andEurytemora copepodites indicates processes of habitat preference of these planktivorous fish in relation to optimal food supply. SPM was the factor controlling both distribution of organisms and the turnover of nutrients. Salinity only was important for the distribution of organisms.  相似文献

In vitro mother plants initiated from a mature tree of Sorbus aucuparia, produced numerous propagules on a medium containing 2 μM 6-benzylaminopurine (BAP) and 0.2 μM 1-naphthaleneacetic acid (NAA). These were rooted on a medium containing 0.25 μM NAA and 0.25 μM indole−3-butyric acid. Adventitious shoots were produced on excised leaves and internodes on media containing 10 μM thidiazuron and 0.3–1.0 μM NAA. They formed by direct regeneration in the axils of leaflets of intact leaves. They also developed indirectly, from callus that developed on the rachis of intact leaves, and the cut ends of petioles and internodes. Somatic embryos were produced on cotyledons of zygotic embryos on medium containing 1 μM BAP, 1 μM kinetin, 0.5 μM NAA, 500 mg l−1 casein hydrolysate and 250 mg l−1 glutamine. On basal medium, 69% developed cotyledons and 20% germinated after pre-treatment at 4 °C on medium containing 30 g l−1 maltose.  相似文献

Cell cultures of Cayratia trifolia (Vitaceae), a tropical lianas, were maintained in Murashige and Skoog’s medium containing 0.25 mg l−1 NAA, 0.2 mg l−1 kinetin and casein hydrolysate 250 mg l−1. Cell suspension cultures of C. trifolia accumulate stilbenes (piceid, resveratrol, viniferin, ampelopsin), which on elicitation by any of 500 μM salicylic acid, 100 μM methyl jasmonate, 500 μM ethrel and 500 mg l−1 yeast extract, added on the 7th day, were enhanced by 3- to 6-fold (5–11 mg l−1) by the 15th day.  相似文献

The effect of suspended sediment concentrations (SSC) on fluid turbulence in an annular flume was investigated. Flow speed was held constant at 0.57 m s−1, and the resulting turbulent conditions were recorded using a 3-D Acoustic Doppler Velocimeter (ADV) at height (z) of 8.5 cm above the bed. The suspended material was composed of a natural glacial clay made up of particles smaller than 6 μm. The SSC in the flume were increased from clear water to 4800 mg l−1 in nine discrete increments; temporal variations of SSC were monitored using three optical backscatter sensors (OBS) mounted in the flume wall at heights of 0.03, 0.10 and 0.20 m above the flume base. The results showed that turbulent intensity ( ) and energy dissipation rate (ɛ) did not change significantly between clear water and 200 mg l−1, but decreased by nearly 30% in the SSC range between 200 and 2400 mg l−1. Above 2400 mg l−1, no further decrease was observed. Analyses of the velocity variances over narrow frequency bands (0.2 Hz wide) from 0 to 12.5 Hz showed that most of the flow turbulent energy (~70–80%) was contained within the lower frequencies i.e. larger eddies, and that these eddies experienced the greatest decrease in energy due to turbidity. It is proposed that these patterns are the consequence of the increase in suspended sediment concentrations and of the vertical stratification of sediments for SSC >200 mg l−1.  相似文献

Transgenic herbicide tolerant Acacia sinuata plants were produced by transformation with the bar gene conferring phosphinothricin resistance. Precultured hypocotyl explants were infected with Agrobacterium tumefaciens strain EHA105 in the presence of 100 μM acetosyringone and shoots regenerated on MS (Murashige and Skoog, 1962, Physiol Plant 15:473–497) medium with 13.3 μM benzylaminopurine, 2.6 μM indole-3-acetic acid, 1 g l−1 activated charcoal, 1.5 mg l−1 phosphinothricin, and 300 mg l−1 cefotaxime. Phosphinothricin at 1.5 mg l−1 was used for the selection. Shoots surviving selection on medium with phosphinothricin expressed GUS. Following Southern hybridization, eight independent shoots regenerated of 500 cocultivated explants were demonstrated to be transgenic, which represented transformation frequency of 1.6%. The transgenics carried one to four copies of the transgene. Transgenic shoots were propagated as microcuttings in MS medium with 6.6 μM 6-benzylaminopurine and 1.5 mg l−1 phosphinothricin. Shoots elongated and rooted in MS medium with gibberellic acid and indole-3-butyric acid, respectively both supplemented with 1.5 mg l−1 phosphinothricin. Micropropagation of transgenic plants by microcuttings proved to be a simple means to bulk up the material. Several transgenic plants were found to be resistant to leaf painting with the herbicide Basta.  相似文献

Embryogenic cultures were induced from leaflets from new vegetative flushes of mature ‘Brewster’ litchi trees on B5 medium containing 400 mg l−1 glutamine, 200 mg l−1 casein hydrolysate, 30 g l−1 sucrose, 4.52 μM 2,4-D, 9.30 μM kinetin and 3 g l−1 gellan gum in darkness. Embryogenic cultures consisting of proembryonic cells and masses were maintained either on semi-solid MS medium supplemented with 4.52 μM 2,4-D and 0.91 μM zeatin or as embryogenic suspension cultures in liquid medium of the same composition. Maturation of somatic embryos occurred on semi-solid MS medium with 5–20% (v/v) filter-sterilized coconut water in darkness. Recovery of plants from somatic embryos was improved with 14.4 μM GA3 on half-strength MS medium with 0.2 g l−1 activated charcoal under a 16 h photoperiod provided by cool white fluorescent lights (60–80 μmol s−1 m−2). Plants have been successfully acclimatized in the greenhouse.  相似文献

Samples of suspended particulate matter (SPM) were collected from the north eastern Irish Sea, under different tidal conditions. The SPM was extracted with 1M HCl and concentrations of Pb and stable isotopic ratios of Pb (206/207Pb) determined using atomic absorption spectrometry and ICP-MS. Lead concentrations were found to be in the range 49 to 115 μg g−1 for spring tides and 72 to 148 μg g−1 for neap tides. The206/207Pb ratios, which were between 1.140 and 1.194, were used to establish possible sources of SPM during the tidal cycle. Samples of surface sediments from along the coastline showed evidence of anthropogenic Pb contamination in the vicinity of Liverpool Bay, with low206/207Pb ratios of about 1.144, which increased northwards away from the estuary. The results are discussed in terms of the tidally-induced transport of SPM in the coastal boundary zone of the Irish Sea.  相似文献

The concentrations of particulate matter, expressed as dry weight (DW), particulate organic (POM), and inorganic material were measured at regular intervals in Lake Constance between February 1980 and December 1982. Maximum particle concentrations were recorded for the euphotic zone in summer (7 mg l−1), while minima occurred during the early summer and in winter. Annual mean concentrations of DW within the entire water column varied between 0.6 and 0.7 mg l−1. In the euphotic zone nearly 70% of DW is organic material. The inorganic particles originate either from phytoplankton (diatomaceous silicon, biogenic decalcification) or from the tributaries. Although phytoplankton biomass only comprises a relatively small proportion (i.e. 30% at maximum) of organic matter, it is the primary source of POM. Therefore, seasonal variations in phytoplankton control epilimnetic concentrations of POM in Lake Constance. Inorganic material comprises smaller proportions of suspended matter. Seasonal variations are related predominantly to fluctuations in biomass and therefore particulate inorganic matter is suggested to originate mainly from autochthonous sources. At the sampling station concentrations of inorganic particles supported by the main tributary, the Alpenrhein, only occasionally vary concomitantly with runoff.  相似文献

The abundance of pelagic invertebrate predators in relation to turbidity and depth gradients in Lake Hiidenvesi (southern Finland) were studied. In the shallow (<5 m) and the most turbid (up to 75 NTU) part of the lake, the community of invertebrate predators consisted of cyclopoid copepods (max biomass >500 μg dw l−1) and Leptodora kindtiii (Focke) (17 μg dw l−1), while in the less turbid (10–40 NTU) stratifying area Chaoborus flavicans (Meigen) dominated (max 146 μg dw l−1). In the temporarily stratifying and moderately turbid basin Chaoborus and small-bodied invertebrate predators co-existed. Mysis relicta (Lovén) occurred only in the stratifying area (max 15 μg dw l−1). The results suggested that both water depth and turbidity contributed to the community structure of Chaoborus flavicans. Depth great enough for stratification was of special importance and its effect was amplified by elevated turbidity, while high turbidity alone could not maintain chaoborid populations. Mysis relicta also requires a hypolimnetic refuge but is more sensitive to low oxygen concentrations and may therefore be forced to the epilimnion where it is vulnerable to fish predation. Cyclopoids as rapid swimmers can take advantage at elevated turbidity levels and coexist in high biomass with fish even in shallow water. Leptodora kindtii can form high biomass despite planktivorous fish providing that turbidity exceeds 20 NTU. The results demonstrated that depth and water turbidity can strongly regulate the abundance and species composition of invertebrate predators. These factors must thus be taken into account when applying food web management, which aims to reduce phytoplankton biomass by depressing planktivorous fish.  相似文献

Shallow lakes often alternate between two possible states: one clear with submerged macrophytes, and another one turbid, dominated by phytoplankton. A third type of shallow lakes, the inorganic turbid, result from high contents of suspended inorganic material, and is characterized by low phytoplankton biomass and macrophytes absence. In our survey, the structure and photosynthetic properties (based on 14C method) of phytoplankton were related to environmental conditions in these three types of lakes in the Pampa Plain. The underwater light climate was characterized. Clear-vegetated lakes were more transparent (K d 4.5–7.7 m−1), had high DOC concentrations (>45 mg l−1), low phytoplankton Chl a (1.6–2.7 μg l−1) dominated by nanoflagellates. Phytoplankton productivity and photosynthetic efficiency (α ~ 0.03 mgC mgChla −1 h−1 W−1 m2) were relatively low. Inorganic-turbid lakes showed highest K d values (59.8–61.4 m−1), lowest phytoplankton densities (dominated by Bacillariophyta), and Chl a ranged from 14.6 to 18.3 μg l−1. Phytoplankton-turbid lakes showed, in general, high K d (4.9–58.5 m−1) due to their high phytoplankton abundances. These lakes exhibited the highest Chl a values (14.2–125.7 μg l−1), and the highest productivities and efficiencies (maximum 0.56 mgC mgChla −1 h−1 W−1 m2). Autotrophic picoplankton abundance, dominated by ficocianine-rich picocyanobacteria, differed among the shallow lakes independently of their type (0.086 × 105–41.7 × 105 cells ml−1). This article provides a complete characterization of phytoplankton structure (all size fractions), and primary production of the three types of lakes from the Pampa Plain, one of the richest areas in shallow lakes from South America.  Biomass, production and heterotrophic activity of bacterioplankton were determined for two weeks in the Great Astrolabe Reef lagoon, Fiji. Bacterial and Bacterial activities were distributed homogeneously throughout the water column (20 to 40 m deep) and varied little from site to site inside the lagoon. Bacterioplankton biomass and production also varied little over a diel period with coefficients of variation of 9 and 22%, respectively. On average, over the whole study, bacterial abundance was 0.77×109 cells l-1 and bacterial production averaged 0.36 μg-at. C l-1 d-1. Bacterial abundance and production were greater in the lagoon than in oceanic waters. Attachment to particles seems to provide an advantage for bacterioplankton growth because specific growth rates for attached bacterioplankton were, on average, significantly greater than that of the free community. Growth efficiency, determined by correlating the net increase of bacterial biomass and the net decrease of dissolved organic carbon (DOC) in dilution cultures, was very low (average 6.6%). Using carbon growth efficiency and bacterial production rates, heterotrophic activity was estimated to average 5.4 μg-at. C l-1 d-1. The turn-over rate of DOC (average 114 μg-at. C l-1) due to bacterial consumption was estimated to be 0.048 d-1 during the period of study. Accepted: 25 July 1998  相似文献
