目的: 构建α1亚基诱导表达、β2和γ2L亚基稳定表达的人源α1β2γ2L-GABAAR-CHO(Chinese hamster ovary)细胞株。方法: 从人cDNA文库中扩增α1、β2、γ2L亚基编码基因,分别构建亚基表达载体;将三个亚基表达载体共转染CHO-K1细胞,通过抗性筛选、膜电位检测法进行稳定表达克隆筛选;通过qPCR、Western blot对亚基表达进行鉴定;以激动剂GABA、阳性变构调节剂地西泮(diazepam,Dia)、拮抗剂荷包牡丹碱(bicuculine)为工具药,采用全细胞膜片钳方法及膜电位检测法对稳定表达细胞的药理学功能进行鉴定。结果: 经克隆筛选获得表达量较高的α1β2γ2L-GABAAR-CHO并对其亚基表达鉴定,结果显示该细胞稳定表达α1、β2、γ2L亚基,构建的α1β2γ2L-GABAAR-CHO细胞仅在加入四环素(tetracyclin)诱导的情况下表达α1亚基并与β2、γ2L组装成具有功能活性的α1β2γ2L-GABAAR;对其进行全细胞膜片钳检测研究发现,GABA可对其产生激动效应,引起α1β2γ2L-GABAAR-CHO细胞产生氯离子通道特征性电流变化,Dia可剂量依赖性地增强GABA对α1β2γ2L-GABAAR的激动效应;在膜电位检测研究中,获得GABA激动效应EC50为(177.72 ± 15.92)nmol/L,Dia变构效应EC50为(3.63±0.52)μmol/L,拮抗剂Bicuculine拮抗效应IC50为(538.83±29.55)nmol/L。结论: 通过采用诱导表达策略,成功构建了α1β2γ2L-GABAAR-CHO稳定表达细胞株,该细胞株具有对激动剂、阳性变构剂、拮抗剂特异性检测的药理学功能。  相似文献

Chronic ethanol treatment is known to alter gene expression and function of γ-aminobutyric acid type-A (GABAA) receptors. Here we focus on the β2 subunit which is widely expressed in the mammalian brain, and plays a key role in the GABA binding site. Previous studies using rodent models of ethanol dependence show either increased or no change of β2 subunit mRNA and peptide content following chronic ethanol administration. In humans, polymorphism at the β2 subunit is associated with ethanol dependence in some, but not all, populations. In the present study we measured mRNA content in the cerebellum and cerebral cortex using ethanol-naive and ethanol-dependent DBA/2J and C57BL/6J mice. The DBA/2J strain displays severe ethanol withdrawal severity, while the C57BL/6J strain shows milder withdrawal reactions. RNase protection analysis demonstrated that the DBA/2J strain is more sensitive to ethanol-induced increases in β2 subunit mRNA content in the cerebellum, showing significant increases at lower blood ethanol concentrations than C57BL/6J mice. The ethanol-induced regulation in C57BL/6J mice appears to be more complex, with decreases in β2 subunit mRNA content at low blood ethanol concentrations, and increases at higher concentrations. These data suggest that differences between C57BL/6J and DBA/2J mice in the degree of physical dependence (withdrawal) on ethanol may be related to differential sensitivity to ethanol regulation of β2 subunit expression.  相似文献

We employed genetically modified mice to examine the role of 1,25-dihydroxyvitamin D3 [1,25(OH)2D3] on skeletal and calcium homeostasis. In mice expressing the null mutation for 25-hydroxyvitamin D 1 hydroxylase (1OHase−/−), or the vitamin D receptor (VDR−/−), 1,25(OH)2D3 and calcium were both required for optimal epiphyseal growth plate development, serum calcium and phosphorus alone were sufficient to mineralize skeletal tissue independent of 1,25(OH)2D3 and the VDR, and endogenous 1,25(OH)2D3 and the VDR were essential for baseline bone formation. In 2-week-old 1OHase−/− mice and in 2-week-old mice homozygous for the PTH null mutation(PTH−/−), PTH and 1,25(OH)2D3 were each found to exert independent and complementary effects on skeletal anabolism, with PTH predominantly affecting appositional trabecular bone growth and 1,25(OH)2D3 influencing both endochondral bone formation and appositional bone growth. Endogenous 1,25(OH)2D3 maintained serum calcium homeostasis predominantly by modifying intestinal and renal calcium transporters but not by producing net bone resorption. Administration of exogenous 1,25(OH)2D3 to double mutant PTH−/−1OHase−/− mice produced skeletal effects consistent with the actions of endogenous 1,25(OH)2D3. These studies reveal an important skeletal anabolic role for both endogenous and exogenous 1,25(OH)2D3 and point to a potential role for 1,25(OH)2D3 analogs in the treatment of disorders of bone loss.  相似文献

The lithiation of indole, using a slight excess of n-butyl lithium in THF, followed by methylation and reaction with [Cr(CO)6] in refluxing dibutyl ether, resulted in the formation of [Cr(η6-N-methylindole)(CO)3] (1a) and [Cr(η6-N-methyl-2-methylindole)(CO)3] (1b). In contrast, lithiation of quinoline in THF, silylation and the subsequent reaction with [Cr(CO)6] under similar reaction conditions, afforded [Cr(η6-N-trimethylsilyl-2-butyl-1,2-dihydroquinoline)(CO)3] (2) and [Cr(η6-{2-butyl-1,2,3,4-tetrahydroquinoline})(CO)3] (3). The formation of [Cr(η6-2,2′-bis{N-methylindolyl})(CO)3] (4) implied lithiation at the 2-position of 1a. However, metallation at the 7-position was also indicated during the same reaction. In the presence of [Mn(CO)5Br], product 4 and the transmetallation product [Cr(η6-{7-(N-methylindolyl)Mn(CO)5})(CO)3] (5) were isolated. Reaction with titanocene dichloride gave [Cr(η6-{2-(N-methylindolyl)TiCp2Cl})(CO)3] (6), which slowly converted into [TiCp2{Cr(η6-2-(N-methylindolyl)(CO)3}2] (7).  相似文献

The preparation and structure–activity relationships (SARs) of potent agonists of the human β3-adrenergic receptor (AR) derived from a 4-aminopiperidine scaffold are described. Examples combine human β3-AR potency with selectivity over human β1-AR and/or human β2-AR agonism. Compound 29s was identified as a potent (EC50=1 nM) and selective (greater than 400-fold over β1- with no β2-AR agonism) full β3-AR agonist with in vivo activity in a transgenic mouse model of thermogenesis.  相似文献

The new organometallic cluster (η24-CO)2(CO)136-C6Me6) has been prepared by the thermolysis of Ru3(CO)12 with hexamethylbenzene in octane and characterised by a single crystal X-ray diffraction study. It is isostructural with the known cluster Ru624-CO)2(CO)136-C6H3Me3) and the metal core constitutnts the same tetrahedral Ru4 unit with two edge-bridging Ru atoms. The mesitylene derivative has been shown to undergo rearrangement to afford the octahedral carbido cluster Ru6C(CO)146-C6H3Me3), but this conversion is not observed for the new hexamethylbenzene derivative.  相似文献

Reactions of [(PPh3)2Pt(η3-CH2CCPh)]OTf with each of PMe3, CO and Br result in the addition of these species to the metal and a change in hapticity of the η3-CH2CCPh to η1-CH2CCPh or η1-C(Ph)=C=CH2. Thus, PMe3 affords [(PMe3)3Pt(η1-C(Ph)=C=CH2)]+, CO gives both [trans-(PPh3)2Pt(CO)(η1-CH2CCPh)]+ and [trans-(PPh3)2Pt(CO)(η1-C(Ph)=C=CH2)]+, and LiBr yields cis-(PPh3)2PtBr(η1-CH2CCPh), which undergoes isomerization to trans-(PPh3)2PtBr(η1-CH2CCPh). Substitution reactions of cis- and trans-(PPh3)2PtBr(η1-CH2CCPh) each lead to tautomerization of η1-CH2CCPh to η1-C(Ph)=C=CH2, with trans-(PPh3)2PtBr(η1-CH2CCPh) affording [(PMe3)3Pt(η1-C(Ph)=C=CH2)]+ at ambient temperature and the slower reacting cis isomer giving [trans-(PPh3)(PMe3)2Pt(η1-C(Ph)=C=CH2)]+ at 54 °C . All new complexes were characterized by a combination of elemental analysis, FAB mas spectrometry and IR and NMR (1H, 13C{1H} and 31P{1H}) spectroscopy. The structure of [(PMe3)3Pt(η1-C(Ph)=C=CH2)]BPh4·0.5MeOH was determined by single-crystal X-ray diffraction analysis.  相似文献

Metathesis of [(η33−C10H16)Ru(Cl) (μ−Cl)]2 (1) with [R3P) (Cl)M(μ-Cl)]2 (M = Pd, Pt), [Me2NCH2C6H4Pd(μ-Cl)]2 and [(OC)2Rh(μ-Cl)]2 affords the heterobimetallic chloro bridged complexes (η33-C10H16) (Cl)Ru(μ-Cl)2M(PR3)(Cl) (M = Pd, Pt), (η33-C10H16) (Cl)Ru(μ-Cl)2PdC6H4CH2NMe2 and (η33-C10H16) (Cl)Ru(μ-Cl)2Rh(CO)2, respectively. Complex 1 reacts with [Cp*M(Cl) (μ-Cl)]2 (M = Rh, Ir), [p-cymene Ru(Cl) (μ-Cl]2 and [(Cy3P)Cu(μ-Cl)]2 to give an equilibrium of the heterobimetallic complexes and of educts. The structures of (η33-C10H16)Ru(μ-Cl)2Pd(PR3) (Cl) (R = Et, Bu) and of one diastereoisomer of (η33-C10H16)Ru(μ-Cl)2IrCp*(Cl) were determined by X-ray diffraction.  相似文献

The phosphinoalkenes Ph2P(CH2)nCH=CH2 (n= 1, 2, 3) and phosphinoalkynes Ph2P(CH2)n C≡CR (R = H, N = 2, 3; R = CH3, N = 1) have been prepared and reacted with the dirhodium complex (η−C5H5)2Rh2(μ−CO) (μ−η2−CF3C2CF3). Six new complexes of the type (ν−C5H5)2(Rh2(CO) (μ−η11−CF3C2CF3)L, where L is a P-coordinated phosphinoalkene, or phosphinoalkyne have been isolated and fully characterized; the carbonyl and phosphine ligands are predominantly trans on the Rh---Rh bond, but there is spectroscopic evidence that a small amount of the cis-isomer is formed also. Treatment of the dirhodium-phosphinoalkene complexes with (η−CH3C5H4)Mn(CO)2thf resulted in coordination of the manganese to the alkene function. The Rh2---Mn complex [(η−C5H5)2Rh2(CO) (μ−η11−CF3C2CF3) {Ph2P(CH2)3CH=CH2} (η−CH3C5H4)Mn(CO)2] was fully characterized. Simi treatment of the dirhodium-phosphinoalkyne complexes with Co2(CO)8 resulted in the coordination of Co2(CO)6 to the alkyne function. The Rh2---Co2 complex [(η−C5H5)2Rh2(CO) (μ−η11−CF3C2CF3) {Ph2PCH2C≡CCH3}Co2(CO)2], C37H25Co2F6O7PRh2, was fully characteriz spectroscopically, and the molecular structure of this complex was determined by a single crystal X-ray diffraction study. It is triclinic, space group (Ci1, No. 2) with a = 18.454(6), B = 11.418(3), C = 10.124(3) Å, = 112.16(2), β = 102.34(3), γ = 91.62(3)°, Z = 2. Conventional R on |F| was 0.052 fo observed (I > 3σ(I)) reflections. The Rh2 and Co2 parts of the molecule are distinct, the carbonyl and phosphine are mutually trans on the Rh---Rh bond, and the orientations of the alkynes are parallel for Rh2 and perpendicular for Co2. Attempts to induce Rh2Co2 cluster formation were unsuccessful.  相似文献

In a synthetic route that varies from the standard procedure requiring irradiation, the (η6-C6H5Cl)Cr(CO)2PPh3 complex is obtained upon reacting (η6-C6H5Cl)Cr(CO)3 with tetrakis(triphenylphosphine)palladium(0), CuI, and trimethylsilylphenylacetylene in triethylamine. The X-ray crystal structure of the yellow–orange crystals of (η6-C6H5Cl)Cr(CO)2PPh3 allows structural comparisons to related (arene)Cr(CO)2PR3 complexes.  相似文献

In the present report, we investigated the possible importance of β1 integrins in the growth and metastasis of a murine mammary carcinoma, SP1, and a metastatic variant, SP1-3M in vivo. CBA/J female mice bearing SP1 tumor transplants were injected with anti-β1 integrin IgG or control nonimmune IgG (200 μg per mouse; i.p.) every two days. Animals received anti-CD4 antibody (100 μg per mouse) at time zero to suppress immunity against rabbit IgG. Outgrowth of macroscopic metastases from SP1, but not from SP1-3M primary tumors, was markedly inhibited in animals receiving anti-β1 integrin IgG but not nonimmune IgG. To assess the stage(s) in the metastatic cascade affected, we examined the number and diameter of micrometastatic nodules in treated and untreated groups. The diameter of micrometastases was significantly reduced in SP1-tumor-bearing mice treated with anti-β1 integrin IgG compared to control IgG, although the number of nodules per cm2 of lung sections examined remained unchanged. No change in the number or size of micrometastases in SP1-3M tumor-bearing mice was observed. No difference in the binding, or complement-mediated and antibody-dependent cell-mediated cytotoxicity of anti-β1 integrin IgG with SP1 and SP1-3M cells was detected. The results suggest that under these conditions anti-β1 integrin inhibits metastatic tumor growth in lung tissue, but has minimal effect on intravasation, adhesion to target organs and extravasation.  相似文献

The horse EE and human β1β1 alcohol dehydrogenase isoenzymes have almost identical protein backbone folding patterns and contain 2 tryptophans per subunit (Trp-15 and Trp-314). Tyr-286, which had been proposed to quench the fluorescence of Trp-314 by resonance energy transfer at alkaline pH in EE, is substituted by Cys in β1β1. The proposed role of Tyr-286 in pH-dependent quenching of EE is confirmed by our observation that tryptophan fluorescence of β1β1 is not substantially quenched at alkaline pH. Tyr-286 had also been implicated in the quenching of Trp-314 upon formation of the EE-NAD+-trifluoroethanol ternary complex. However, β1β1 exhibits the same extent of tryptophan fluorescence quenching as EE upon complexation, which strongly suggests that Tyr-286 is not involved in ternary complex quenching.  相似文献

The reaction of TiCl4 with Li2[(SiMe2)25-C5H3)2] in toluene at room temperature afforded a mixture of cis- and trans-[(TiCl3)2{(SiMe2)25-C5H3)2}] in a molar ratio of 1/2 after recrystallization. The complex trans-[(TiCl3)2{(SiMe2)25-C5H3)2}] was hydrolyzed immediately by the addition of water to THF solutions to give trans-[(TiCl2)2(μ-O){(SiMe2)25-C5H3)2}] as a solid insoluble in all organic solvents, whereas hydrolysis of cis-[(TiCl3)2{(SiMe2)25-C5H3)2}] under different conditions led to the dinuclear μ-oxo complex cis-[(TiCl2)2)(μ-O){(SiMe2)25-C5H3)2}] and two oxo complexes of the same stoichiometry [(TiCl)2(μ-O){(SiMe2)25-C5H3)2}]2(μ-O)2 as crystalline solids. Alkylation of cis- and trans-[(TiCl3)2{(SiMe2)25-C5H3)2}] with MgCIMe led respectively to the partially alkylated cis-[(TiMe2Cl)2{(SiMe2)25-C5H3)2}] and the totally alkylated trans-[(TiMe3)2{(SiMe2)25-C5H3)2}] compounds. The crystal and molecular structure of the tetranuclear oxo complex [(TiCl)2(μ-O){(SiMe2)25-C5H3)2}]2(μ-O)2 was determined by X-ray diffraction.  相似文献

The dinuclear Pt---Si complex {(Ph3P)Pt{μ-η2-H---SiH(IMP)]}2 (trans-1a–cis-1b=3:1; IMP=2-isopropyl-6-methylphenyl) reacted with basic phosphines such as 1,2-bis(diphenylphosphino)ethane (dppe) and dimethylphenylphosphine (PMe2Ph) to afford different dinuclear Pt---Si complexes with loss of H2, {(P)2Pt[μ-SiH(IMP)]}2 [P=dppe, trans-2a (major), cis-2b (trace); PMe2Ph, 3 (trans only)]. Complexes 2 and 3 were characterized by multinuclear NMR spectroscopy and X-ray crystallography (2a). In contrast, the reaction of 1a,b with the sterically demanding tricyclohexylphosphine (PCy3) afforded {(Cy3P)Pt{μ-η2-H---SiH(IMP)]}2 (trans-4a–cis-4b 2:1) analogous to 1a,b where the central Pt2Si2(μ-H)2 core remains intact but the PPh3 ligands have been replaced by PCy3. Reactions of [Rh(COD)Cl]2 with the ligand RN(PX2)2 (1: R = C6H5; X = OC6H5) give mono- or disubstituted complexes of the type [Rh2(COD)Cl22−C6H5N(P(OC6H5)2)2}] or [RhCl{ν2−C6H5 N(P(OC6H5)2)2 }]2 depending on the reaction conditions. Reaction of 1 with [Rh(CO)2Cl]2 gives the symmetric binuclear complex, [Rh(CO)Cl{μ−C6H5N(P(OC6H5)2)2} 2, whereas the same reaction with 2 (R = CH3; X = OC6H5) leads to the formation of an asymmetric complex of the type [Rh(CO)(μ−CO)Cl{μ−CH3N(P(OC6H5)2)2}2 containing both terminal and bridging CO groups. Interestingly the reaction of 3 (R = C6H5, X = OC6H4Br−p with either [Rh(COD)Cl]2 or [Rh(CO)2Cl]2 leads only to the formation of the chlorine bridged binuclear complex, [RhCl{ν2−C6H5N(P(OC6H4Br−p)2)2}]2. The structural elucidation of the complexes was carried out by elemental analyses, IR and 31P NMR spectroscopic data.  相似文献

Recently, evidence has accumulated in support of the heterologous expression of functional membrane proteins and their complexes on extracellular baculovirus particles (budded virus, BV). In this study, we attempted to apply this BV display system to detect G-protein-coupled receptor (GPCR) signaling. We infected Sf9 cells with a combination of four recombinant baculoviruses individually encoding the dopamine D1 receptor (DR-D1), G-protein -subunit (Gs), G-protein β1γ2 subunit dimer (Gβ1γ2), and adenylyl cyclase type VI (ACVI). The recovered BV fraction produced cAMP in response to the stimulation with dopamine. Co-expression of all three G-protein subunits in addition to receptor and ACVI led to a maximal response. BV co-expressing DR-D1, Gs, Gβ1γ2, and ACVI also responded to dopamine agonists and an antagonist. Furthermore, BV expressing two other Gs-coupled receptors together with Gs, Gβ1γ2, and ACVI also produced cAMP in response to their specific ligands. These results indicate the functional coupling of receptor, Gs and ACVI is reconstituted on BV. Since BV is essentially free of endogenous GPCRs, this BV co-display system should prove highly useful for the development of functional assay systems for GPCRs.  相似文献

Manganese tricarbonyl complexes (η5-C5H4CH2CH2Br)Mn(CO)3 (3) and (η5-C5H4CH2CH2I)Mn(CO)3 (4), with an alkyl halide side chain attached to the cyclopentadienyl ligand, were synthesized as possible precursors to chelated alkyl halide manganese complexes. Photolysis of 3 or 4 in toluene, hexane or acetone-d6 resulted in CO dissociation and intramolecular coordination of the alkyl halide to manganese to produce (η51-C5H4CH2CH2Br)Mn(CO)2 (5) and (η51-C5H4CH2CH2I)Mn(CO)2 (6). Low temperature NMR and IR spectroscopy established the structures of 5 and 6. Photolysis of 3 in a glass matrix at 91 K demonstrated CO release from manganese. Low temperature NMR spectroscopy established that the coordinated alkyl halide complexes are stable to approximately −20°C.  相似文献

Using RT-PCR we distinguished mRNAs for all known G-protein coupled serotonin receptors expressed in various rat and porcine blood vessels. Nearly all vessels expressed 5HT1 β, 5-HT2A, 5-HT2B, 5-HT4, and 5-Ht7 receptor mRNA to different extents. New splice variants of the porcine 5-HT4 receptor were observed. Similar PCR assays were performed with endothelial and smooth muscle cells from human pulmonary artery, aorta, and with endothelial cells from human coronary artery and umbilical vein. All endothelial cells expressed 5-HT1 β, 5-HT2b, and 5-HT4 receptor mRNA, whereas in smooth muscle cells 5-HT1 β, 5-HT2A, 5-HT7, and in some experiments 5-HT2B receptor mRNA were found. A model for the regulation of vascular tone by different 5-HT receptors is proposed.  相似文献

The reaction of RuCl3(H2O), with C5Me4CF3J in refluxing EtOH gives [Ru25-C5Me1CF2)2 (μ-Cl2] (20 in 44% yield. Dimer 2 antiferromagnetic (−2J=200 cm1). The crystal structures of 2 (rhombohedral system, R3 space group, Z=9, R=0.0589) and [Rh25-C5Me4CF3(2Cl2(μ-Cl)2] (3) (rhombohedral system. space group, Z = 9, R = 0.0641) were solved; both complexes have dimeric structures with a trans arrangement of the η5-C5Me4CF4 rings. Comparison of the geometry of 2 and 3 with those of the corresponding η5-C5Me5 complexes shows that lowering the ring symmetry causes significant distortion of the M2(μ-Cl)2 moiety. The analysis of the MCl3 fragment conformations in 2 and 3 and in the η5-C5ME5 analogues shows that they are correlated with the M---M distances. The Cl atoms are displaced by Br on reaction of 2 with KBr in MeOH to give the diamagnetic dimer [Ru25-C5Me4CF3)2Br2 (μ-Br2] (4). Complex 2 reacts with O2 in CH2Cl2 solution at ambient temperature to form a mixture of isomeric η6-fulvene dimers [Ru26-C5Me3CF3 = CH2)2Cl2(μ-Cl)2] (5). Reactions of 5 with CO and allyl chloride give Ru(η5-C5Me3CF3CH2Cl)(CO)2Cl (6) and Ru(η5-C5Me3CF3CF3CH2Cl)(η3-C3H5)Cl2 (7) respectively.  相似文献
