In queen honey bees the free amino acid content in the haemolymph clearly depends on the physiological function and social environment of the individual. While in drones and workers the content of free amino acids increases after emergence until it reaches a peak in 5-day-old animals and decreases afterwards, the amino acid content in queens reaches its highest level (>60 nmol/ microl haemolymph) with the onset of egg laying (10 d of age). This level is about 2.5 times more than the highest level found in workers. Queens maintain this high level also when they are older (>30 d) and continue to lay eggs in average colonies. As in drones and workers, in queens the predominant amino acid is proline, which accounts for more than 50% of the total content of free amino acids in egg-laying individuals. When 10-day-old queens are prevented from mating and do not lay eggs, their amino acid content is significantly lower compared to laying queens of the same age. Also the social environment influences the contents of free amino acids in queens. When virgin queens were kept for 6 days with 20 worker bees and sufficient honey and pollen in an incubator, they had significantly lower concentrations of amino acids than virgin queens living for the same period with about 8000 workers in a colony. Most probably, the high amino acid concentration in the haemolymph is the basis for the high protein synthesis activity of laying queens.  相似文献

Summary In honeybee workers proline was the predominant amino acid and comprised from 50% in newly emerged bees up to 80% of total amino acids from the 3rd day on. The overall concentration averaged at about 20 mM in newly emerging bees, rose to a maximum of about 25mM at the 3rd–5th day and decreased in older bees. Essential amino acids decreased by 40% during the first 3 days and thereafter stayed constant. The bulk of amino acids with a lower concentration (from traces to about 2mM) showed either no change in concentration or was higher in newly emerging bees and decreased during the lifespan of the insects. Forager bees, collected after flight, had significantly lower proline concentrations as compared to 22 day old bees collected from the colony, while the concentrations of the bulk of all other amino acids did not change significantly. There was a great variance in the concentration of all amino acids between different colonies but we could not prove dependency on relatedness.  相似文献

In the two-fuel system for flight of the migratory locust, the haemolymph carbohydrate concentration falls during flight periods of up to 1 hr, the decrease being greater in case the pre-flight carbohydrate level is higher. The increase in the lipid concentration from the onset of flight is virtually independent of the initial lipid concentration. Flight intensity affects these changes in substrate concentrations: the carbohydrate level decreases more rapidly if flight speed is higher, whereas the increase in lipid concentration is delayed at higher flight speeds. Respiratory carbon dioxide production is elevated rapidly during flight and reaches over eight times the resting level. From the rate of 14CO2 production after labelling of the haemolymph diglyceride pool it is concluded that diglycerides contribute to providing the energy for flight from the earliest stage of flying activity; diglyceride oxidation increases until maximum utilization is attained after some 45 min of flight. The decline in haemolymph carbohydrate concentration due to flying activity results in a decrease of haemolymph osmolarity. Free amino acids, particularly taurine, increase markedly in the haemolymph during flight; yet their concentration only partially counterbalances the fall in haemolymph osmolarity.  相似文献

中华卵索线虫Ovomermis sinensis感染棉铃虫Helicoverpa armigera 1天后,棉铃虫幼虫血淋巴中游离氨基酸总量大幅度下降,各种游离氨基酸的含量也是如此变化。感染2~4天后的棉铃虫血淋巴中游离氨基酸总量变化不大,各种游离氨基酸的含量有升有降。感染5~6天后的棉铃虫血淋巴中游离氨基酸总量和各种氨基酸的含量都急剧上升。研究表明,中华卵索线虫寄生棉铃虫时,棉铃虫血淋巴中游离氨基酸含量的变化朝着有利于线虫生长发育的方向进行。  相似文献

In this work we examined the time course and the amount released, by hyposmolarity, for the most abundant free amino acids (FAA) in rat brain cortex astrocytes and neurons in culture. The aim was to evaluate their contribution to the process of cell volume regulation. Taurine, glutamate, andd-aspartate in the two types of cells, -alanine in astrocytes and GABA in neurons were promptly released by hyposmolarity, reaching a maximum within 1–2 min. after an osmolarity change. A substantial amount of the intracellular pool of these amino acids was mobilized in response to hyposmolarity. The amount released in media with osmolarity reduced from 300 mOsm to 150 mOsm or 210 mOsm, represented 50%–65% and 13%–31%, respectively, of the total amino acid content in cells. In both astrocytes and neurons, the efflux of glutamine and alanine was higher under isosmotic conditions and increased only marginally during hyposmotic conditions.86Rb+, used as tracer for K+, was released from astrocytes, 30% and 11%, respectively, in hyposmotic media of 150 mOsm or 210 mOsm but was not transported in neurons. From these results it was calculated that FAA contribute 54% and inorganic ions 46% to the process of volume regulation in astrocytes exposed to a 150 mOsm hyposmotic medium. This contribution was 55% for FAA and 45% for K+ and Cl in cells exposed to 210 mOsm hyposmotic solutions. These results indicate that the contribution of FAA to the process of cell volume regulation is higher in astrocytes than in other cell types including renal and blood cells.Special issue dedicated to Dr. Claude Baxter.  相似文献

Efflux and tissue content of N-acetylaspartate (NAA) and amino acids were evaluated from cultured and acutely prepared hippocampal slices in response to changes in osmolarity. The osmoregulator taurine, but not NAA, was lost from both types of slices after moderate reductions in extracellular osmolarity (−60 mOsm) for 10–48 h. Hypoosmotic shock (−166 mOsm) for 5 min resulted in unselective efflux of several amino acids from acutely prepared slices. Notably, the efflux of taurine, but not NAA, was prominent also after the shock. Efflux of NAA was markedly enhanced by NMDA and high K+, in particular after the stimulation period. The high K+-mediated efflux was decreased by high extracellular osmolarity and a NMDA-receptor antagonist. The results indicate that NAA efflux can be induced by a sudden non-physiological decrease in extracellular osmolarity but not by prolonged more moderate changes in osmolarity. The mechanisms behind the efflux of NAA by high K+ are complex and may involve both swelling and activation of NMDA-receptors.  相似文献

The microclimate in the habitat of the New Zealand alpine weta Hemideina maori is very variable with winter temperatures down to −6 °C under the rocks where the insects are found. Subfreezing temperatures may in winter prevail for up to 17 days but diurnal cycles of freezing and thawing are common, as is also the case in summer. Rates of temperature change can be very high and up to −7.20 °C/h. During winter, humidity was high for extended periods ranging from 70% to 100% relative humidity (RH). In the summer, humidity ranged from 30% RH during the day to 100% RH at night. The supercooling point of the haemolymph was approximately −8 °C year round, caused by a heat labile substance. The supercooling point of the haemolymph of an insect of the same genus, Hemideina femorata not regularly exposed to subfreezing temperatures, was ca. −16.5 °C. Thermal hysteresis was not detected in the haemolymph of H. maori. Haemolymph osmolality varied from 380 mOsm (summer) to 700 mOsm (winter). Body water content was ca. 75% all year round. Total concentrations of sodium, potassium and chloride in haemolymph varied from 170 mM (winter) to 250 mM (summer). The total concentration of free amino acids varied from 58 mM (summer) to 263 mM (winter). This variation was mostly due to proline which varied from ca. 15 mM (summer) to ca. 100 mM (winter). The freeze-tolerant weta H. maori is exposed to a highly variable and cold environment all year round and several properties of its haemolymph composition can be attributed to these climatic conditions, e.g. the presence of ice-nucleating agents and an increase in the concentration of proline during cold hardening in the autumn. The concentrations of free amino acids were measured in whole claw muscle, single fibres and haemolymph of Australian freshwater crayfish, Cherax destructor, during the intermoult stage. The average total pool of amino acids in short-sarcomere fibres (179 mmol kg(-1)) was 60% greater than in long-sarcomere fibres, due to higher concentrations of alanine, cysteine, glutamate, leucine and proline. The two fibre types exhibited differences in the banding pattern of the isoforms of troponin using gel electrophoresis. The average pool of amino acids in haemolymph was 2.7 mmol kg(-1). Cherax has symmetrical claws and the total pool of amino acids from whole muscles (approx. 79 mmol kg(-1)) was similar in left and right claw muscles. In animals acclimated to osmotic environments between 0 and 220 mOsm, the osmotic pressure of the haemolymph increased from 356 to 496 mOsm, but no systematic changes were observed in the amino acid profiles of muscles or haemolymph. The major findings were that (a) concentrations of amino acids differed between the two major fibre types in claw muscle and (b) amino acids in the muscle fibres did not play a major part in intracellular osmoregulation in Cherax, suggesting this species is an anisosmotic regulator.  相似文献

The purpose of our work was to observe the influence of probiotic preparation BioPlus 2B on average daily gains of weaned pigs, feed efficiency and to evaluate some metabolic indices. The weaned pigs, at the age of 42 days, were included into the trial and divided into four groups. Pigs in groups A (n = 5) and B (n = 4) received BioPlus 2B also before weaning. Only group A received BioPlus 2B, at the concentration of 3.2 × 109 CFU per kg of feed, after weaning continually. Groups C (n = 5) and D (n = 4) did not receive BioPlus 2B until the start of the trial, but group C was given BioPlus 2B at the same concentration as group A during the experiment. Blood samples for determination of metabolic indices were collected at the start of experiment, i.e. on 42nd day of pigs life, and then on 56th, 70th, 84th and 91st days of pigs life. The following biochemical indices were evaluated within the trial: serum levels of total proteins, albumin, urea, total cholesterol and total lipids. Total serum protein level (p < 0.0001) and serum albumin level (p = 0.0024) in groups A and B were significantly higher in comparison with groups C and D on day 56 of pigs life. Serum urea level in group D was significantly (p = 0.049) higher than in group A on 70th day of pigs life. Serum level of total cholesterol in group B on day 56 and 84 of pigs life was significantly (p = 0.0004) higher than in groups C and D. Total serum lipid level was significantly (p < 0.05) higher in B group compared to other groups on 56th, 70th, 84th days of pigs life. Average daily gains (ADG) in A group, even if non-significantly, were about 10% better than in groups B, C, D between 57th and 77th days of pigs life. ADG in groups A and B were 14% better in comparison with that in groups C and D between 78th and 91st days of pigs life (p = 0.036). The best feed efficiency in the trial was in group A, approximately 13%, 16% and 21% better than that in the groups B, C and D, repectively. Immuno-diffusion tests show that worker and queen haemolymph contains a protein fraction which does not occur in the haemolymph of drones. Its immunological and electrophoretical properties are identical with those of the main soluble fraction in the ovaries of queens in oviposition. It therefore is a vitellogenin.The titre of this vitellogenin in the haemolymph of 0- to 28-day-old workers was determined by rocket-immunoelectrophoresis. It attains a maximum on day 12. Its changes seem to be positively correlated with the volume of the corpora allata during the first 12 days of adult life.The hypopharyngeal, mandibular, and salivary glands and the content of the honey stomach of workers were immunologically examined. Vitellogenin could not be found in these organs nor in worker or royal jelly. It is also absent from the digestive tract of queens.14C-labelled amino acids were injected into 5-day-old workers. Later the uptake of radioactive proteins by the queen was examined. Autoradiography of immuno-diffusion plates showed that within 72 hr active material passed from the injected workers into the eggs laid by the queen. The soluble proteins were extracted from the ovaries and the thorax of the queens and their radioactivity determined. The ratio of ovary to thorax radioactivity of queens directly injected was significantly different from that of queens kept with injected workers.Several proteins of the homogenized hypopharyngeal glands of workers showed precipitation reactions with the antiserum against homogenate of queen ovaries. This together with the results of the tracer experiments indicates that the proteins of the worker hypopharyngeal glands may be precursors of queen yolk components.  相似文献

This study describes the effects of desiccation and rehydration on the water content, haemolymph volume (per cent), osmolarity, and concentrations of Na, K, Mg, and Ca in the haemolymph of the lone star tick, Amblyomma americanum.The water content percentages of ‘severely desiccated’, ‘moderately’ and ‘fully hydrated’ ticks were 46·0, 52·8, and 60·3 per cent respectively. The lowest and highest of these were near the minimum and maximum possible.The haemolymph volume (per cent) of ‘severely desiccated’ ticks was regulated near the level of ‘moderately hydrated’ ticks despite significant decreases in total body water content and increases in osmolarity and concentration of sodium. Conversely, the change from ‘severely desiccated’ to ‘moderately hydrated’ ticks can be viewed as causing an increase in total body water, decrease in blood osmolarity and sodium, but little change in haemolymph volume (per cent).Most of the water taken up by ‘moderately hydrated’ ticks (while becoming ‘fully hydrated’) was added to the haemolymph. At the same time, there was little change in the blood osmolarity or haemolymph concentration of sodium. Conversely, the change of ‘fully’ to ‘moderately hydrated’ ticks was marked by a substantial loss of haemolymph volume (per cent) but little change in osmolarity and concentration of sodium.The concentration of potassium was regulated over the full range of desiccating and hydrating conditions. The lone star tick appeared less able to regulate its haemolymph concentrations of Ca and Mg; both fluctuated at the same rate, but inversely as the haemolymph volume (per cent).It appears that a carefully controlled movement of solutes (Na the predominant cation) between haemolymph and non-haemolymph tissue is intimately linked with haemolymph volume regulation and movement of water into the haemolymph during hydration.  相似文献

Hemolymph osmolarity has great effect on honey bee health, especially in arid and semi-arid zones. It regulates water and nutrients in stressed tissues. Osmotic concentration in three races (Apis mellifera ligustica, A. m. carnica and A. m. jemenitica) of Apis mellifera was tested in central Saudi Arabia during spring and summer seasons in 2015. Newly emerged bee workers were first marked and later their hemolymph was extracted after intervals of 1, 5, 10, 15, 20 and 25 days. A significant positive correlation between age and osmolarity was found in all three races during spring and summer seasons. The lowest combined osmotic concentration for all three races was found after 1 day interval, while the highest osmotic concentration was recorded after 25 days. Among all races, A. m. ligustica showed significantly high osmotic concentration after 25 days in spring and summer seasons as compared to the other two races. Only A. m. jemenitica showed similar osmotic concentration after 10 and 15 days in both spring and summer seasons compared to other two races. Mean osmotic concentration of all three races was significantly different after 20 and 25 days in spring and summer seasons. Overall mean recorded during summer was significantly higher than the mean of spring season. Combined osmotic concentration in young drones of all races was significantly lower than that of old drones during spring and summer seasons.  相似文献

Summary Toads (Bufo viridis) were kept on soil without access to free water (simulated terrestrial conditions) for over 12 weeks. Body water compartments were estimated using the dilution method (inulin and Evans Blue). They were found to remain fairly constant after a period of adjustment which lasted 1–2 weeks. In particular, plasma volume was closely controlled. Plasma osmolarity increased to over 1000 mOsm · 1–1 accompanying a large increase in its urea concentration. NaCl also increased, while potassium remained constant. Tissue (liver and skeletal muscle) water content did not change much and electrolytes were kept constant. Tissue water urea concentration seemed to equilibrate with that of the plasma. Urine osmolarity, which was hypotonic during water access, became nearly isosmotic and correlated with the plasma following transfer onto soil. Urine urea concentration correlated with the plasma in the terrestrial conditions, potassium was greatly elevated, sodium increased to some extent, and chloride hardly changed. The efficient osmoregulatory mechanisms for the control of distribution of body water sustain normal physiological functions.  相似文献

The concentrations of sodium, potassium, chloride, bicarbonate, total phosphate, labile phosphate, inorganic phosphate, protein, polypeptides, amino acids, trehalose, and glucose, as well as pH and osmotic pressure of larval-pupal moulting fluid and haemolymph were measured in the tobacco hornworm, Manduca sexta, during the secretion and resorption of moulting fluid. Moulting fluid is a mildly alkaline (pH 7.2), iso-osmotic (330 mOsm) potassium bicarbonate salt solution containing within it the sol form of moulting gel. Bicarbonate is the principal anion in moulting fluid. It is only a minor component of haemolymph. Significant differences in the concentrations of potassium, chloride, bicarbonate, non-labile phosphate, labile phosphate, inorganic phosphate, protein, polypeptide, amino acids, trehalose, and glucose indicate that the integumentary epithelium does not act as a semi-permeable membrane, and that free diffusion between moulting fluid and haemolymph can account for only a small fraction of the molecular species movement between these two fluids.  相似文献

Late instar larvae starved at 53% r.h. maintained constant haemolymph osmotic pressure (O.P.) for 12 days with only a small rise from 353 to 363 mOsm at day 17 when haemolymph volume was nearly zero. Total body water was also nearly constant for the first 12 days and then dropped from 62 to 58·5%. At low r.h. for 7 days, starved larvae lost more water than those at high r.h., but haemolymph O.P. ranged from 351 to 363 mOsm, and total body water remained nearly constant. Measured values were lower than expected from actual water losses, requiring that solutes be removed from the blood. Larvae starved at 53% r.h. for 7 days and then given distilled water took in 60 per cent of the starved weight and increased haemolymph water volume by 55 per cent. O.P. dropped only to 326 mOsm as against the expected 210 mOsm. More solute was mobilized than had been apparently sequestered during starvation. Thus body fluids are closely regulated despite wide internal and external changes.  相似文献

为了研究密林熊蜂Bombus patagiatus Nylander雄蜂出房后生殖系统发育变化过程, 本研究首次对密林熊蜂雄性成蜂(1-15日龄)生殖系统的精液量、 精子数、 性腺大小、 储精囊大小等进行了测定。结果显示: 雄性成蜂在1日龄时精液量最大, 达到5.95±0.39 μL; 镜检精子数在2日龄最多; 而精液中活动精子数出现波浪式的循环趋势, 9日龄达到最大值, 为25.93±1.06个; 精液中精子总数在9日龄达到最大值, 为160.67±17.11万个; 活动精子百分比以8、 9日龄最高, 分别达到86.38%±2.02%及86.45%±2.50%; 性腺长度在1日龄时最小, 为1 522.01±37.93 μm; 储精囊的中部宽度在7日龄最宽, 达86.38±2.96 μm, 但与9日龄相比无显著差异。由此可见, 8-9日龄雄性成蜂个体各指标较为稳定, 该日龄即为雄性成蜂性成熟且可以交配的时期。本研究结果为密林熊蜂人工繁育提供了理论依据。  相似文献

The present study aimed to elucidate the effects that osmotic shock exerts on equine spermatozoa. To achieve this goal, a retrospective study of the cellular volume of 40 equine ejaculates subjected to osmolarities ranging from 75 to 900 mOsm in Biggers-Whitten-Whittingham (BWW) media was performed using a Multisizer3 Coulter Counter®. The 300 mOsm BWW solution was used as control. The sperm volume ranged between 37.93 ± 0.6 (mean ± Standard Error of the Mean (SEM)) in 75 mOsm BWW to 21.61 ± 0.27 (mean ± SEM) for 900 mOsm BWW. Thus the spermatozoa behaved as linear osmometers when adjusted to the Boyle Van't Hoff equation (R2 = 0.9808). After the different osmotic challenges, spermatozoa were returned to 300 mOsm BWW and the cellular volume was measured again. The results showed that the spermatozoa were able to retrieve the isosmolar volume (20.81 ± 0.34; mean ± SEM). Also, an ultrastructural study of spermatozoa membrane and mitochondria was accomplished using Transmission Electron Microscopy (TEM) after the osmotic challenges in 2 ejaculates. As observed by TEM, sperm plasmalemma swelled and detached from the sperm head in hypotonic conditions (75 mOsm), with blebbing on return to isosmolarity. When subjected to 900 mOsm, the sperm plasmalemma shrank, with disarrangement and blebbing when returned to isosmolarity. Mitochondria were also found to change their volume; the main pathologic change was irreversible vacuolization and changes in their arrangement for all the osmotic challenges tested. The present work leads to a better understanding of how osmotic shock adversely affects equine spermatozoa structure.  相似文献

The concentration of free amino acids, total soluble protein, and haemoglobin in the haemolymph of fourth instar Chironomus tentans was investigated.The concentration of the free amino acid pool increases between the early (15.7 mM/l) and mid-(33.9 mM/l) fourth larval stages followed by a decline during the late (16.9 mM/l) fourth larval period. Alanine, serine, and the amides of aspartic acid and glutamic acid are the predominant free amino acids at all stages. Physiological fluid analysis of late fourth instar haemolymph detected 32 ninhydrin positive components including 18 common amino acids plus homoarginine, ornithine, citrulline, β-alanine, α-aminoadipic acid, α-aminoisobutyric acid, and sarcosine.The concentration of total soluble protein steadily increases during fourth instar larval development to a maximum of 9.3 g100 ml followed by a decline during the pharate pupal period. A similar pattern of variation occurs in haemoglobin content which comprises from 51 to 66% of Chironomus tentans haemolymph protein.The mM percentage of individual amino acids of total haemolymph protein varies little during the fourth instar. At all stages alanine and aspartic acid are the predominant amino acids.  相似文献

Abstract. Macropterous females of Pyrrhocoris apterus (L.) reared under short-day conditions (LD 12:12 h) were analysed for temporal patterns of feeding and drinking behaviour, activities of digestive enzymes in the gut, and lipid and glycogen content in the haemolymph and fat body. Peaks of drinking activity were recorded at days 3, 7 and 10 during the first 14 days after imaginal ecdysis. Feeding activity peaked on the third day, ceasing completely after the fourth day of adult life. Esterase, protease, amylase and aminopeptidase activities exhibited the highest overall activity in the first days after imaginal emergence; then enzyme activities decreased. In the fat body, the content of lipids was highest on day 5, then a decrease of about 40% was observed at day 14; the amount of glycogen was highest on day 1 at 11 μg of glucose equivalents/mg of fat body, then decreased to 2 μg at day 14 after the imaginal moult. In the haemolymph, the lipid content rose until day 8 when it reached almost 0.3 μmol/μl; at day 14 the value was slightly lower. The association of fasting with reproductive arrest in macropterous females of P. apterus, accompanied by a decrease in digestive enzyme activities and a mobilization of lipid reserves from the fat body, was demonstrated.  相似文献
