Objective: Peptide YY (PYY) 3‐36 has recently been recognized as an important gut hormone that influences food intake. Peripheral injections of PYY 3‐36 in rats inhibit food intake in experimental animals as well as in lean and obese human subjects. This hormone has been suggested as an attractive therapeutic option for obesity. The aim of this study was to assess the influence of age, sex, thyroid status, growth hormone (GH), pregnancy, and food restriction on PYY levels in rat. Research Methods and Procedures: We determined plasma PYY levels in all experimental sets. Results: PYY levels were influenced by age, with the highest hormone levels achieved in early postnatal life (day 10) and decreasing thereafter. PYY levels were also dependent on thyroid hormone status being decreased in hyperthyroid rats. Exogenous GH administration led to a clear‐cut decrease in PYY levels in both normal and GH‐deficient rats. Acute food deprivation or chronic food restriction led to decreased PYY levels in virgin and pregnant rats. In pregnant rats with food available ad libitum, PYY levels were enhanced at late gestation. Discussion: Our observations indicate that PYY levels are influenced by age, thyroid hormones, and GH. These data indicate that PYY could be involved in the changes of food intake associated with these conditions. The PYY levels observed in acute and chronic food‐restricted rats indicate that, in situations of decreased energy intake, the lower PYY levels could serve to disinhibit central pathways and facilitate food intake.  相似文献

Objective: Resistin was recently identified as a hormone secreted by adipocytes that is under hormonal and nutritional control. This hormone has been suggested to be the link between obesity and type 2 diabetes. The aim of this study was to assess the influence of gender, gonadal status, thyroid hormones, pregnancy, and food restriction on resistin mRNA levels in adipose tissue of rats. Research Methods and Procedures: We have determined resistin mRNA expression by Northern blot analysis in all experimental sets. Results: Resistin mRNA expression is influenced by age, with the highest hormone levels existing at 45 days after birth and decreasing thereafter. Resistin mRNA expression is higher in men than in women. Moreover, we studied the effect of orchidectomy and ovariectomy in rats of different ages and showed that gonadal hormones increase adipose tissue resistin mRNA expression in male rats. Resistin is also regulated by thyroid hormones; it is severely decreased in hyperthyroid rats. Our results clearly show that chronic food restriction (30% of ad libitum food intake) led to a decrease in adipose tissue mRNA levels in normal cycling female rats and pregnant rats. In pregnancy, resistin mRNA levels were enhanced particularly at midgestation. Discussion: Our observations indicate that resistin is influenced by gender, gonadal status, thyroid hormones, and pregnancy. These findings suggest that resistin could explain the decreased insulin sensitivity during puberty and could be the link between sex steroids and insulin sensitivity. Moreover, resistin could mediate the effect of thyroid hormones on insulin resistance and the state of insulin resistance present during pregnancy.  相似文献

Many experiments show that serotonin (5-HT) controls thyroidal function at hypothalamic level, inhibiting the TRH secretion. The majority of experiments are done in an acute way, consisting of a single serotonin dose injected intraperitoneally (ip) or intracerebroventricularly (ic) with the effect registered after a short time (usually 1 h) as in normal environmental conditions similar to the TSH stimulation test, that consists of transfer of the experimental animals from 30°C to 4°C for 30 min, thus inducing stimulation of the hypothalamus-hypophysis-thyroid axis. The aim of the present research was to study the correlation between 5-HT and the thyroidal function, measuring plasmatic thyroid hormone levels in rats ip treated in chronic (injected daily for 10 days with different doses of 5-HT), and in acute way (after 1 h from a single 2.0 mg/kg bw 5-HT dose) in normal environmental conditions to evidence the serotonin site action activity outside the blood-brain barrier. The results of the chronic experiment show an inhibitory effect of 5-HT, on T3 and T4 plasmatic level, only when it is injected at medium doses (0.2 and 0.4 mg/kg bw for T3, and 0.2 for T4), while the results of the acute experiment do not evidence any modification. These results show that in normal environmental conditions the outside 5-HT site action is active only when the 5-HT is injected chronically at defined doses, probably for a down-regulation phenomenon.  相似文献

Domesticated horses are increasingly kept under semi-natural housing conditions, whereas their adaptation capacity is not fully investigated. In all, 10 Shetland pony mares were held under semi-extensive conditions for 1 year. In winter animals were allocated into two feeding groups (60% and 100% of maintenance energy requirement, respectively). Triiodothyronine, thyroxine, non-esterified fatty acids (NEFA), total bilirubin, total protein, triglyceride, glucose, insulin and hair length were measured at monthly intervals, whereas BW, body condition score, cresty neck score and resting heart rate were recorded every 2 weeks. From summer to winter all Ponies showed a reduction in resting heart rate (P<0.001) and triiodothyronine (P<0.001) but an increase in NEFA (P<0.001), thyroxine (P<0.001) and triglyceride (P<0.001) concentrations. Feed restriction led to a reduced resting heart rate (P=0.009), increased NEFA (P<0.001) and total bilirubin (P=0.008) concentrations. Thyroid hormones did not differ between feeding groups (P>0.05). Refeeding of restrictively fed ponies resulted in a rapid increase in resting heart rate and BW and a return of blood parameters to reference values. Adequately supplied animals adapted without difficulty to varying environmental conditions, whereas feed restriction in ponies during winter resulted in reduced resting heart rates suggesting a reduced basal metabolic rate. The energy restriction was compensated by mobilizing body fat reserves which led to changes in blood parameters. Refeeding in feed restricted animals revealed a remarkably quick recovery of physiological and blood parameters to reference values. We therefore suggest that year round-outdoor housing can be a suitable housing system for robust horse breeds provided that an adequate food supply is available.  相似文献

Resistin as an adipokine identified from rodents in 2001 is involved in many biological processes. However, little is known about this gene in fish. We cloned Siberian sturgeon (Acipenser baerii) resistin cDNA of 795 base pairs, encoding 107 amino acids, which showed 38–40% identity to human and rodents. Real-time quantitative PCR showed that the resistin gene was widely distributed in tissues of Siberian sturgeon, with the highest expression in liver. After fasting for 1, 3, 6 and 10 days, the expression of the resistin gene in the liver of Siberian sturgeon decreased significantly, and after refeeding on the 10 days of fasting the resistin mRNA expression increased rapidly, suggesting that resistin may play an important role in liver in response to starvation. Taken together, these results suggest that resistin may be involved in the regulation of energy homeostasis in liver.  相似文献

Objective : To examine the effect of age on body protein losses occurring during severe energy restriction in obesity. Research Methods and Procedures : Weanling (young) Sprague-Dawley rats (YR) were fed a high fat (35% energy) diet (HFD) until mean body weight approached that of a group of chowfed retired breeder (aged) rats (AR). Both groups were then fed HFD for an additional 2 weeks, after which selected controls from YR and AR groups were killed for baseline carcass analysis. Remaining rats were fed a very-low-energy diet (VLED, 33% kcal of HFD) for 3 weeks and then killed for carcass analysis. Results : YR had greater fat stores before VLED, and lost proportionately more fat and less protein during VLED than did AR. Weight loss composition during VLED was 66.7% fat, 11.1% protein, and 22.2% water in YR, and 39.4% fat, 26.2% protein, and 34.3% water in AR. Greater YR fat loss during VLED* (70.6 ± 30.4 vs. 32.6 ± 29.1 g in AR; mean ± SD) was paralleled by significantly larger decreases in epididymal and retroperitoneal fat pad weights, mean adipocyte size, and lipoprotein lipase activity. Greater protein loss in AR (21.6 ± 13.9 g vs. 11.8 ± 10.7 g in YR) coincided with larger decreases in visceral organ weights and serum thyroxine and triiodothyronine. Energy expenditure changes during VLED were similar between groups. Discussion : Dietary obese young rats appear better able than aged rats to conserve body protein while losing body fat during severe energy restriction.  相似文献

Transthyretin (TTR) is a visceral protein, which facilitates the transport of thyroid hormones in blood and cerebrospinal fluid. The homotetrameric structure of TTR enables the simultaneous binding of two thyroid hormones per molecule. Each TTR subunit provides a single cysteine residue (Cys10), which is frequently affected by oxidative post‐translational modifications. As Cys10 is part of the thyroid hormone‐binding channel within the TTR molecule, PTM of Cys10 may influence the binding of thyroid hormones. Therefore, we analysed the effects of Cys10 modification with sulphonic acid, cysteine, cysteinylglycine and glutathione on binding of triiodothyronine (T3) by molecular modelling. Furthermore, we determined the PTM pattern of TTR in serum of patients with thyroid disease by immunoprecipitation and mass spectrometry to evaluate this association in vivo. The in silico assays demonstrated that oxidative PTM of TTR resulted in substantial reorganization of the intramolecular interactions and also affected the binding of T3 in a chemotype‐ and site‐specific manner with S‐glutathionylation as the most potent modulator of T3 binding. These findings were supported by the in vivo results, which indicated thyroid function‐specific patterns of TTR with a substantial decrease in S‐sulphonated, S‐cysteinylglycinated and S‐glutathionylated TTR in hypothyroid patients. In conclusion, this study provides evidence that oxidative modifications of Cys10 seem to affect binding of T3 to TTR probably because of the introduction of a sterical hindrance and induction of conformational changes. As oxidative modifications can be dynamically regulated, this may represent a sensitive mechanism to adjust thyroid hormone availability.  相似文献

Plasma thyroid hormone levels were measured in pink salmon, Oncorhynchus gorbuscha , during their spawning migration in the Fraser River, British Columbia. The plasma levels of both l-thyroxine (T4) and triiodo-l-thyronine (T3) were significantly higher in males than in females. In both sexes the hormone levels were maintained, or increased somewhat, during the early stages of migration, but fell thereafter. In females the plasma T4 and T3 levels of salmon collected on the spawning grounds were at or below detectable levels of the assays. The changes in thyroid hormone levels are correlated with changes in plasma insulin, gonadotropin, gonadal steroid hormones, cortisol and vitellogenin levels measured in the same specimens.  相似文献

Food restriction (FR) retards animals' growth. Understanding the underlying mechanisms of this phenomenon is important to conceptual problems in life-history theory, as well as to applied problems in animal husbandry and biomedicine. Despite a considerable amount of empirical data published since the 1930s, there is no relevant general theoretical framework that predicts how animals vary their energy budgets and life-history traits under FR. In this paper, we develop such a general quantitative model based on fundamental principles of metabolic energy allocation during ontogeny. This model predicts growth curves under varying conditions of FR, such as the compensatory growth, different age at which FR begins, its degree and its duration. Our model gives a quantitative explanation for the counterintuitive phenomenon that under FR, lower body temperature and lower metabolism lead to faster growth and larger adult size. This model also predicts that the animals experiencing FR reach the same fraction of their adult mass at the same age as their ad libitum counterparts. All predictions are well supported by empirical data from mammals and birds of varying body size, under different conditions of FR.  相似文献

Serum T4 and T3 in wild Atlantic cod Gadus morhua ranged from 1 to 12 ng ml−1 and from 2 to 27 ng ml−1 respectively over a 3-year period. In general, the concentrations increased from summer (T3) or early autumn (T4) to maxima in mid-winter and declined abruptly during spring. The T4/T3 monthly means were lowest in summer and highest in winter. The seasonal patterns of thyroid hormones were weakly correlated with changes in water temperature. However, both T4 and T3 co-varied simultaneously with photoperiod. In addition, T3 was correlated with the hepatosomatic index and condition factor during summer and autumn. It is suggested that the seasonal changes in the release of T4 from the thyroid were photoperioddriven, and that the course of T3 was regulated by the metabolic state of the fish during the somatic growth period.  相似文献

Objective : To investigate, in young obese male Zucker rats, the effects of chronic food restriction and subsequent refeeding on: 1) parameters of nonadipose and adipose growth, 2) regional adipose depot cellularity [fat cell volume (FCV) and number], and 3) circulating leptin levels. Research Methods and Procedures : Obese (fa/fa) and lean (Fa/?) male Zucker rats were studied from age 5 to 19 weeks. After baseline food intake monitoring, 10 obese rats were subjected to 58 days of marked caloric restriction from ad libitum levels [obese‐restricted (OR)], followed by a return to ad libitum feeding for 22 days. Ten lean control rats and 10 obese control rats were fed ad libitum for the entire experiment. All rats were fed using a computer‐driven automated feeding system designed to mimic natural eating patterns. Results : After food restriction, OR rats weighed significantly less than did lean and obese rats and showed a significant diminution in body and adipose growth as compared with obese rats. Relative adiposity was not different between obese and OR rats and was significantly higher than that of lean rats. The limitation in growth of the adipose tissue mass in OR rats was due mostly to suppression of fat cell proliferation because the mean FCV in each of the four depots was not affected. Serum leptin levels of OR and obese rats were not different from each other but were significantly higher than those of lean rats. Discussion : Marked caloric restriction affects obese male Zucker rats in a manner different from that of nongenetic rodent models (i.e., Wistar rats). In comparison with the response to caloric deprivation of Wistar rats, these calorically restricted obese male Zucker rats appeared to defend their relative adiposity and mean FCV at the expense of fat cell number. These findings indicate that genetic and/or tissue‐specific controls override the general consequences of food restriction in this genetic model of obesity.  相似文献

Aim of this work was to evaluate if long-term dietary supplementation of potassium iodide (KI) to dairy goats can influence metabolic and hormonal parameters. Thirty Sarda crossbred dairy goats were divided into three groups, which were orally administered 0 (control group; CON), 0.45 (low iodine group; LI) or 0.90 (high iodine group; HI) mg of KI/day, respectively. The daily dose of KI (76.5% of iodine) was administered as salt dissolved in water for 8 weeks. Plasma contents of nonesterified fatty acids (NEFA), urea, glucose, insulin, free triiodothyronine (FT3) and thyroxine (FT4) were determined weekly. Iodine supplementation increased significantly the FT3 hormone (P = 0.007) and FT3/FT4 ratio (P = 0.001) and tended to influence the FT4 hormone (P = 0.059). An iodine level × week of sampling interaction for NEFA (P = 0.013) evidenced a temporary concentration increase in supplemented groups. The ‘Revised Quantitative Insulin Sensitivity Check Index’ increased with KI supplementation (P ⩽ 0.01). Blood urea nitrogen (BUN) and insulin were lowered (P ⩽ 0.01) by iodine supplementation (groups LI and HI; P ⩽ 0.01). The glucose concentration evidenced an iodine level × week of sampling interaction (P = 0.025) due to an unexpected and temporary increase of its concentration in the CON group. Glucose concentration was decreased by KI supplementation only in LI group (P < 0.05). In conclusion, the daily supplementation of low doses of KI can improve insulin sensitivity and decrease BUN in dairy goats.  相似文献

The morphological alterations of hepatocytes of cave‐dwelling salamander Proteus anguinus anguinus after food deprivation periods of one and 18 months were investigated and the concentrations of glycogen, lipids, and proteins in the liver were determined. Quantitative analyses of the hepatocyte size, the lipid droplets, the number of mitochondria, and volume densities of M and P in the hepatocytes were completed. After one month of food deprivation, the cytological changes in the hepatocytes are mainly related to the distribution and amount of glycogen, which was dispersed in the cytoplasm and failed to form clumps typical of normal liver tissue. After 18 months of food deprivation hepatocytes were reduced in size, lipid droplets were less numerous, peroxisomes formed clusters with small, spherical mitochondria, and specific mitochondria increased in size and lost cristae. Lysosomes, autophagic vacuoles, and clear vacuoles were numerous. The liver integrity was apparently maintained, no significant loss of cytoplasmic constituents have been observed. Biochemical analysis revealed the utilization of stored metabolic reserves in the liver during food deprivation. Glycogen is rapidly utilized at the beginning of the starvation period, whereas lipids and proteins are utilized subsequently, during prolonged food deprivation. In the Proteus liver carbohydrates are maintained in appreciable amounts and this constitutes a very important energy depot, invaluable in the subterranean environment. J. This review discusses aspects of feeding ecology, nutrition, and dietary husbandry that are particularly relevant to reproductive success in wild animals. Emphasis has been placed on recently published literature. Special attention has been given to requirements for energy and protein and the unique roles of essential amino acids, essential fatty acids, calcium, phosphorus, zinc, copper, selenium, and vitamins A, C, D, and E. Information has been drawn from research with mammals, birds, reptiles, amphibians, and fish, including the domestic and laboratory species that play such a large role in elucidating mechanisms connecting nutrition and reproduction. Transthyretin (TTR), the major transporter of thyroid hormones and vitamin A in cerebrospinal fluid (CSF), binds the Alzheimer beta-peptide and thus might confer protection against neurodegeneration. In addition, altered TTR levels have been described in the CSF of patients with psychiatric disorders, yet its function in the CNS is far from understood. To determine the role of TTR in behaviour we evaluated the performance of TTR-null mice in standardized tasks described to assess depression, exploratory activity and anxiety. We show that the absence of TTR is associated with increased exploratory activity and reduced signs of depressive-like behaviour. In order to investigate the mechanism underlying these alterations, we measured the levels of catecholamines. We found that the levels of noradrenaline were significantly increased in the limbic forebrain of TTR-null mice. This report represents the first clear indication that TTR plays a role in behaviour, probably by modulation of the noradrenergic system.  相似文献

In oviparous species like birds, eggs provide the direct environment in which embryos are developing. Mothers may adjust different egg components in different ways in reaction to environmental cues either to adjust offspring development or because of constraints. In this study, we investigated the effects of food quality and quantity before and during egg laying on three different aspects of egg quality: macro‐nutrients (egg and yolk mass), androgens (testosterone and androstenedione), and thyroid hormones (3,5,3′‐triiodothyronine, T3 and l ‐thyroxine, T4), using the rock pigeon (Columba livia). As expected, egg and yolk mass were significantly reduced for the eggs laid under the poor‐food condition, indicating a maternal trade‐off between offspring and self in allocating important resources. We did not find any significant change in yolk testosterone or their within‐clutch pattern over the laying sequence. This is consistent with the fact that, in contrast with nutrients, these hormones are not costly to produce, but does not support the hypothesis that they play a role in adjusting brood size to food conditions. In contrast, we found that T3 levels were higher in the egg yolks under the poor‐food condition whereas the total T4 content was lower. This change could be related to the fact that iodine, the critical constituent of thyroid hormones, might be a limiting factor in the production of this hormone. Given the knowledge that food restriction usually lead to reduction of circulating T3 levels, our results suggested that avian mothers can independently regulate its concentrations in their eggs from their own circulation. The study demonstrates that environmentally induced maternal effects via the egg can be a result of a combination of constrained resources and unconstrained signals and that thyroid hormones might be an interesting case of both. Therefore, this hormone and the interplay of different maternal effects on the offspring phenotype deserve much more attention.  相似文献

Female rats were fed either ad libitum or 30% energy-restricted diets from 5 weeks through 25 weeks of age. Genomic DNA was extracted from mammary tissue and liver at 7, 9, 14 (mid-pregnancy), 16.5 (mid-lactation), and 25 weeks of age. The 5-methyldeoxycytidine content of DNA was determined by high pressure liquid chromatography. As animals aged from 7–25 weeks, 5-methyldeoxycytidine increased in mammary tissue, whereas in liver it decreased. This suggests that 5-methyldeoxycytidine exhibits tissue specific patterns. No changes in 5-methyldeoxycytidine content due to 30% energy restriction were observed in either mammary tissue or liver.  相似文献

The effects of 17β-oestradiol (E2) on plasma kinetics of thyroid hormones (T4, l-thyroxine; T3, 3,5,3′-triiodo-l-thyronine) were studied in immature rainbow trout. E2-3-benzoate (0.5 mg/100 g) was injected intraperitoneally on days 0 and 3, and on the morning of day 4 each trout received an intracardiac injection of either [125I]T4 and Na 131I or [I25I]T3. Groups of trout were bled and killed from 5 min to 4 days post-injection of tracer. E2 did not alter the plasma T4 level but depressed the T4 plasma clearance rate, plasma-to-total tissue flux of T4 and thyroidal T4 secretion rate. Monodeiodination of T4 to T3 was also depressed, as judged from plasma [I25I]T3 and I25I ? levels in [125I]T4-injected trout. E2 had no major effect on T3 plasma clearance rate but depressed the plasma T3 level, plasma-to-total tissue flux of T3 and the T3 plasma appearance rate. E2 had no influence on biliary transport of [I25I]T4 or [125I]T3. The above results suggest that E2, at the dose range employed, depresses extrathyroidal T4 to T3 conversion, which may in turn decrease plasma T4 clearance and thyroidal T4 secretion.  相似文献

Serum thyroid hormone levels were determined in adult Atlantic salmon ( Salmo salar ) of both sexes caught in the ocean and at a sequence of locations on their return migration to spawn. Tri-iodothyronine (T3) levels were greatest in fish caught in coastal or estuarine waters or in a river near head-of-tide. T3 levels were lower in fish caught in rivers throughout the angling season and lowest in those captured entering a tributary near spawning. Thyroxine (T4) levels were lowest in immature fish captured in the ocean in winter but raised in fish captured in spring; many of the latter group showed endocrine evidence of their becoming sexually mature. T4 levels were greatest in fish captured in coastal waters and progressively lower in fish captured in an estuary, near head-of-tide and in rivers. T4 levels in fish captured at tributary entry near spawning exceeded those in fish caught in rivers earlier in the year. In general, these data support the hypothesis that motor activity level in migrant fish is a determinant of thyroid status.  相似文献
