Isoprene is a highly reactive gas, and is emitted in such large quantities from the biosphere that it substantially affects the oxidizing potential of the atmosphere. Relatively little is known about the control of isoprene emission at the molecular level. Using transgenic tobacco lines harbouring a poplar isoprene synthase gene, we examined control of isoprene emission. Isoprene synthase required chloroplastic localization for catalytic activity, and isoprene was produced via the methyl erythritol (MEP) pathway from recently assimilated carbon. Emission patterns in transgenic tobacco plants were remarkably similar to naturally emitting plants under a wide variety of conditions. Emissions correlated with photosynthetic rates in developing and mature leaves, and with the amount of isoprene synthase protein in mature leaves. Isoprene synthase protein levels did not change under short-term increase in heat/light, despite an increase in emissions under these conditions. A robust circadian pattern could be observed in emissions from long-day plants. The data support the idea that substrate supply and changes in enzyme kinetics (rather than changes in isoprene synthase levels or post-translational regulation of activity) are the primary controls on isoprene emission in mature transgenic tobacco leaves.  相似文献

Boreal and subarctic peatlands have been extensively studied for their major role in the global carbon balance. However, study efforts have so far neglected the contribution of these ecosystems to the non-methane biogenic volatile organic compound (BVOC) emissions, which are important in the atmospheric chemistry and feedbacks on climate change. We aimed at estimating the BVOC emissions from a subarctic peatland in northern Finland. Furthermore, our aim was to assess how these emissions are affected by enhanced UV-B radiation, the amount of which has increased especially at high latitudes as a result of stratospheric ozone depletion. The contribution of BVOC emissions to the total net carbon exchange and correlations between the emission of different BVOCs and net ecosystem CO2 exchange, CH4 emission, total green leaf area, and abiotic factors were also studied. The UV-B exposure, simulating a 20% depletion of stratospheric ozone, was started in 2003, and measurements were performed during the growing seasons of 2006 and 2008. The subarctic peatland proved to be a small source of BVOCs and the dominant moss, Warnstorfia exannulata, emitted a diverse compound spectrum. The water table level exerted a major influence on the BVOC emissions surpassing the effect of enhanced UV-B. In fact, no overall UV-B effect was established on the BVOC emissions, apart from toluene and 1-octene, emissions of which were doubled and tripled by enhanced UV-B in 2008, respectively. Isoprene (2‐methyl‐1,3‐butadiene) is emitted from many plants and it appears to have an adaptive role in protecting leaves from abiotic stress. However, only some species emit isoprene. Isoprene emission has appeared and been lost many times independently during the evolution of plants. As an example, our phylogenetic analysis shows that isoprene emission is likely ancestral within the family Fabaceae (= Leguminosae), but that it has been lost at least 16 times and secondarily gained at least 10 times through independent evolutionary events. Within the division Pteridophyta (ferns), we conservatively estimate that isoprene emissions have been gained five times and lost two times through independent evolutionary events. Within the genus Quercus (oaks), isoprene emissions have been lost from one clade, but replaced by a novel type of light‐dependent monoterpene emissions that uses the same metabolic pathways and substrates as isoprene emissions. This novel type of monoterpene emissions has appeared at least twice independently within Quercus, and has been lost from 9% of the individuals within a single population of Quercus suber. Gain and loss of gene function for isoprene synthase is possible through relatively few mutations. Thus, this trait appears frequently in lineages; but, once it appears, the time available for evolutionary radiation into environments that select for the trait is short relative to the time required for mutations capable of producing a non‐functional isoprene synthase gene. The high frequency of gains and losses of the trait and its heterogeneous taxonomic distribution in plants may be explained by the relatively few mutations necessary to produce or lose the isoprene synthase gene combined with the assumption that isoprene emission is advantageous in a narrow range of environments and phenotypes.  相似文献

通过大田试验和室外盆栽试验,采用人工增加紫外辐射的方法模拟UV-B辐射增强,用静态箱-气相色谱法测定N_2O排放通量,研究地表UV-B辐射增强对土壤-大豆系统N_2O排放的影响.结果表明:在相同的气象条件和田间管理措施下,UV-B辐射增强没有改变土壤-大豆系统N_2O排放通量的季节性变化规律.但从植株结荚到成熟,UV-B辐射增强降低了土壤-大豆系统N_2O排放通量,进而降低了N_2O的累积排放量.收割实验发现,在分枝开花期,UV-B辐射增强对土壤N_2O排放影响明显,降低了土壤N_2O排放通量;从结荚至鼓粒期,UV-B辐射增强主要通过降低植株地上部分N_2O排放通量来降低土壤-大豆系统的N_2O排放.UV-B辐射增强显著降低了植株的生物量,并影响到植株的氮代谢和土壤NH_4~+-N与微生物氮.UV-B辐射增强可能会导致农田生态系统N_2O排放量降低.  相似文献

Increasing surface levels of UV-B resulting from stratospheric ozone reduction directly affect tropospheric photochemistry. There may also be indirect tropospheric effects due to changes in emission of organic compounds from vegetation. We treated woody and herbaceous isoprene-emitting species in the field with supplemental UV-B simulating 30% ozone depletion. For Quercus gambelii, photosynthesis and isoprene emission were significantly greater in elevated UV-B treatments when expressed on a leaf area basis, but not on a leaf mass basis. Leaves of Mucuna pruriens, however, showed no significant differences in photosynthesis or isoprene emission between treatments, nor when exposed for 45 min to acute high levels of UV-B. Elevated UV-B during growth did not elicit significant isoprene emission from Acer platanoides, a non-emitting species. Other potential UV-B effects, such as changes in leaf area or species composition, which may influence regional isoprene emissions, should be examined.  相似文献

Many hours of sunlight in the midnight sun period suggest that significant amounts of biogenic volatile organic compounds (BVOCs) may be released from arctic ecosystems during night-time. However, the emissions from these ecosystems are rarely studied and limited to point measurements during daytime. We measured BVOC emissions during 24-hour periods in the field using a push-pull chamber technique and collection of volatiles in adsorbent cartridges followed by analysis with gas chromatography- mass spectrometry. Five different arctic vegetation communities were examined: high arctic heaths dominated by Salix arctica and Cassiope tetragona, low arctic heaths dominated by Salix glauca and Betula nana and a subarctic peatland dominated by the moss Warnstorfia exannulata and the sedge Eriophorum russeolum. We also addressed how climate warming affects the 24-hour emission and how the daytime emissions respond to sudden darkness. The emissions from the high arctic sites were lowest and had a strong diel variation with almost no emissions during night-time. The low arctic sites as well as the subarctic site had a more stable release of BVOCs during the 24-hour period with night-time emissions in the same range as those during the day. These results warn against overlooking the night period when considering arctic emissions. During the day, the quantity of BVOCs and the number of different compounds emitted was higher under ambient light than in darkness. The monoterpenes α-fenchene, α -phellandrene, 3-carene and α-terpinene as well as isoprene were absent in dark measurements during the day. Warming by open top chambers increased the emission rates both in the high and low arctic sites, forewarning higher emissions in a future warmer climate in the Arctic.  相似文献

The effects of global change on the emission rates of isoprene from plants are not clear. A factor that can influence the response of isoprene emission to elevated CO2 concentrations is the availability of nutrients. Isoprene emission rate under standard conditions (leaf temperature: 30°C, photosynthetically active radiation (PAR): 1000 μmol photons m?2 s?1), photosynthesis, photosynthetic capacity, and leaf nitrogen (N) content were measured in Quercus robur grown in well‐ventilated greenhouses at ambient and elevated CO2 (ambient plus 300 ppm) and two different soil fertilities. The results show that elevated CO2 enhanced photosynthesis but leaf respiration rates were not affected by either the CO2 or nutrient treatments. Isoprene emission rates and photosynthetic capacity were found to decrease with elevated CO2, but an increase in nutrient availability had the converse effect. Leaf N content was significantly greater with increased nutrient availability, but unaffected by CO2. Isoprene emission rates measured under these conditions were strongly correlated with photosynthetic capacity across the range of different treatments. This suggests that the effects of CO2 and nutrient levels on allocation of carbon to isoprene production and emission under near‐saturating light largely depend on the effects on photosynthetic electron transport capacity.  相似文献

The emission of isoprene from the biosphere to the atmosphere has a profound effect on the Earth's atmospheric system. Until now, it has been assumed that the primary short-term controls on isoprene emission are photosynthetically active radiation and temperature. Here we show that isoprene emissions from a tropical tree (oil palm, Elaeis guineensis) are under strong circadian control, and that the circadian clock is potentially able to gate light-induced isoprene emissions. These rhythms are robustly temperature compensated with isoprene emissions still under circadian control at 38 degrees C. This is well beyond the acknowledged temperature range of all previously described circadian phenomena in plants. Furthermore, rhythmic expression of LHY/CCA1, a genetic component of the central clock in Arabidopsis thaliana, is still maintained at these elevated temperatures in oil palm. Maintenance of the CCA1/LHY-TOC1 molecular oscillator at these temperatures in oil palm allows for the possibility that this system is involved in the control of isoprene emission rhythms. This study contradicts the accepted theory that isoprene emissions are primarily light-induced.  相似文献

Isoprene emission from plants is one of the principal ways in which plant processes alter atmospheric chemistry. Despite the importance of this process, few long-term controls over basal emission rates have been identified. Stress-induced changes in carbon allocation within the entire plant, such as those produced by defoliation, have not been examined as potential mechanisms that may control isoprene production and emission. Eastern cottonwood (Populus deltoides) saplings were partially defoliated and physiological and growth responses were measured from undamaged and damaged leaves 7 days following damage. Defoliation reduced isoprene emission from undamaged and damaged leaves on partially defoliated plants. Photosynthetic rates and leaf carbon and nitrogen pools were unaffected by damage. Photosynthetic rate and isoprene emission were highly correlated in undamaged leaves on undamaged plants and damaged leaves on partially defoliated plants. There was no correlation between photosynthetic rate and isoprene emission in undamaged leaves on partially defoliated plants. Isoprene emission was also highly correlated with the number of source leaves on the apical shoot in damage treatments. Increased carbon export from source leaves in response to defoliation may have depleted the amount of carbon available for isoprene synthesis, decreasing isoprene emission. These results suggest that while isoprene emission is controlled at the leaf level in undamaged plants, emission from leaves on damaged plants is controlled by whole-branch allocation patterns. Isoprene is a volatile compound emitted from leaves of many plant species in large quantities, which has an impact on atmospheric chemistry due to its massive global emission rate (5 x 10(14) carbon g year(-1)) and its high reactivity with the OH radical, resulting in an increase in the half-life of methane. Isoprene emission is strongly induced by the increase in isoprene synthase activity in plastids at high temperature in the day time, which is regulated at its gene expression level in leaves, while the physiological meaning of isoprene emission for plants has not been clearly demonstrated. In this study, we have functionally overexpressed Populus alba isoprene synthase in Arabidopsis to observe isoprene emission from transgenic plants. A striking difference was observed when both transgenic and wild-type plants were treated with heat at 60 degrees C for 2.5 h, i.e. transformants revealed clear heat tolerance compared with the wild type. High isoprene emission and a decrease in the leaf surface temperature were observed in transgenic plants under heat stress treatment. In contrast, neither strong light nor drought treatments showed an apparent difference. These data suggest that isoprene emission plays a crucial role in a heat protection mechanism in plants.  相似文献

Leaves of fast‐growing, woody bioenergy crops often emit volatile organic compounds (VOC). Some reactive VOC (especially isoprene) play a key role in climate forcing and may negatively affect local air quality. We monitored the seasonal exchange of VOC using the eddy covariance technique in a ‘coppiced’ poplar plantation. The complex interactions of VOC fluxes with climatic and physiological variables were also explored by using an artificial neural network (Self Organizing Map). Isoprene and methanol were the most abundant VOC emitted by the plantation. Rapid development of the canopy (and thus of the leaf area index, LAI) was associated with high methanol emissions and high rates of gross primary production (GPP) since the beginning of the growing season, while the onset of isoprene emission was delayed. The highest emissions of isoprene, and of isoprene photo‐oxidation products (Methyl Vinyl Ketone and Methacrolein, iox), occurred on the hottest and sunniest days, when GPP and evapotranspiration were highest, and formaldehyde was significantly deposited. Canopy senescence enhanced the exchange of oxygenated VOC. The accuracy of methanol and isoprene emission simulations with the Model of Emissions of Gases and Aerosols from Nature increased by applying a function to modify their basal emission factors, accounting for seasonality of GPP or LAI.  相似文献

Isoprene emission rates from quaking aspen (Populus tremuloides Michx.) leaves were measured simultaneously with photosynthesis rate, stomatal conductance, and intercellular CO2 partial pressure. Isoprene emission required the presence of CO2 or O2, but not both. The light response of isoprene emission rate paralleled that of photosynthesis. Isoprene emission was inhibited by decreasing ambient O2 from 21% to 2%, only when there was oxygen insensitive photosynthesis. Mannose (10 millimolar) fed through cut stems resulted in strong inhibition of isoprene emission rate and is interpreted as evidence that isoprene biosynthesis requires either the export of triose phosphates from the chloroplast, or the continued synthesis of ATP. Light response experiments suggest that photosynthetically generated reductant or ATP is required for isoprene biosynthesis. Isoprene biosynthesis and emission are not directly linked to glycolate production through photorespiration, contrary to previous reports. Isoprene emission rate was inhibited by above-ambient CO2 partial pressures (640 microbar outside and 425 microbar inside the leaf). The inhibition was not due to stomatal closure. This was established by varying ambient humidity at normal and elevated CO2 partial pressures to measure isoprene emission rates over a range of stomatal conductances. Isoprene emission rates were inhibited at elevated CO2 despite no change in stomatal conductance. Addition of abscisic acid to the transpiration stream dramatically inhibited stomatal conductance and photosynthesis rate, with a slight increase in isoprene emission rate. Thus, isoprene emission is independent of stomatal conductance, and may occur through the cuticle. Temperature had an influence on isoprene emission rate, with the Q10 being 1.8 to 2.4 between 35 and 45°C. At these high temperatures the amount of carbon lost through isoprene emission was between 2.5 and 8% of that assimilated through photosynthesis. This represents a significant carbon cost that should be taken into account in determining midsummer carbon budgets for plants that are isoprene emitters.  相似文献

Isoprene emission from vegetation is the single most important source of photochemically active reduced compounds to the atmosphere. We present the first controlled-environment measurements of isoprene emission from leaves of tropical forest trees. Our studies were conducted in the Guanica State Forest in Puerto Rico. We report the effects of temperature and light variations on biogenic isoprene emissions during 1995. Maximum emission rates varied among species from 0 to 268 nmol m?2 s?1. Values at the upper end of this range of maximum emission rates are 2–3 times higher than values reported from any temperate taxa. Isoprene emission showed strong sensitivity to light and temperature variations. In contrast to temperate plants, whose emissions tend to saturate at a light intensity of ~1000 μmol m?2 s?1, emissions from the tropical species increased with light intensity up to 2500 μmol m?2 s?1. The temperature optima for emissions from these plants were similar to those previously reported for temperate plants: ~40 °C. The high maximum emission rates and lack of light saturation indicate that estimates of isoprene emission from tropical forests need to be revised upwards.  相似文献

Isoprene (2-methyl-1,3 butadiene) is a low-molecular-weight hydrocarbon emitted in large quantities to the atmosphere by vegetation and plays a large role in regulating atmospheric chemistry. Until now, the atmosphere has been considered the only significant sink for isoprene. However, in this study we performed both in situ and in vitro experiments with soil from a temperate forest near Ithaca, N.Y., that indicate that the soil provides a sink for atmospheric isoprene and that the consumption of isoprene is carried out by microorganisms. Consumption occurred rapidly in field chambers (672.60 +/- 30.12 to 2,718.36 +/- 86.40 pmol gdw day) (gdw is grams [dry weight] of soil; values are means +/- standard deviations). Subsequent laboratory experiments confirmed that isoprene loss was due to biological processes: consumption was stopped by autoclaving the soil; consumption rates increased with repeated exposure to isoprene; and consumption showed a temperature response consistent with biological activity (with an optimum temperature of 30 degrees C). Isoprene consumption was diminished under low oxygen conditions (120 +/- 7.44 versus 528.36 +/- 7.68 pmol gdw day under ambient O(2) concentrations) and showed a strong relationship with soil moisture. Isoprene-degrading microorganisms were isolated from the site, and abundance was calculated as 5.8 x 10 +/- 3.2 x 10 cells gdw. Our results indicate that soil may provide a significant biological sink for atmospheric isoprene.  相似文献

异戊二烯(Isoprene)的排放具有特殊的生物学功能,对大气环境具有重要影响,另外,异戊二烯也是一种具有广泛用途的化合物。在生物体内,异戊二烯是由异戊二烯合成酶(Isoprene synthase,Isps)催化烯丙基二磷酸(Dimethylallyl diphosphate,DMAPP)脱去焦磷酸(Pyrophosphate)而生成的。因此,作为异戊二烯合成过程中的关键酶,Isps在异戊二烯的自然排放和生物合成过程都发挥着重要的作用,对Isps的研究具有非常重要的意义。到目前为止,已经在多种植物中发现了该酶,研究表明,来源于不同生物的异戊二烯合成酶具有保守的结构特征和相似的生化性质。文中就Isps的最新研究进展进行综述,包括比较分析不同生物来源Isps的生化特征和结构特征,探讨催化机制,并对该酶在生物工程领域的应用进行展望。  相似文献

? Depending on the atmospheric composition, isoprene emissions from plants can have a severe impact on air quality and regional climate. For the plant itself, isoprene can enhance stress tolerance and also interfere with the attraction of herbivores and parasitoids. ? Here, we tested the growth performance and fitness of Populus × canescens in which isoprene emission had been knocked down by RNA interference technology (PcISPS-RNAi plants) for two growing seasons under outdoor conditions. ? Neither the growth nor biomass yield of the PcISPS-RNAi poplars was impaired, and they were even temporarily enhanced compared with control poplars. Modelling of the annual carbon balances revealed a reduced carbon loss of 2.2% of the total gross primary production by the absence of isoprene emission, and a 6.9% enhanced net growth of PcISPS-RNAi poplars. However, the knock down in isoprene emission resulted in reduced susceptibility to fungal infection, whereas the attractiveness for herbivores was enhanced. ? The present study promises potential for the use of non- or low-isoprene-emitting poplars for more sustainable and environmentally friendly biomass production, as reducing isoprene emission will presumably have positive effects on regional climate and air quality.  相似文献

The response of tundra plants to enhanced UV-B radiation simulating 15 and 30% ozone depletion was studied at two high arctic sites (Isdammen and Adventdalen, 78° N, Svalbard).The set-up of the UV-B supplementation systems is described, consisting of large and small UV lamp arrays, installed in 1996 and 2002. After 7 years of exposure to enhanced UV-B radiation, plant cover, density, morphological (leaf fresh and dry weight, leaf thickness, leaf area, reproductive and ecophysiological parameters leaf UV-B absorbance, leaf phenolic content, leaf water content) were not affected by enhanced UV-B radiation. DNA damage in the leaves was not increased with enhanced UV-B in Salix polaris and Cassiope tetragona. DNA damage in Salix polaris leaves was higher than in leaves of C. tetragona. The length of male gametophyte moss plants of Polytrichum hyperboreum was reduced with elevated UV-B as well as the number of Pedicularis hirsuta plants per plot, but the inflorescence length of Bistorta vivipara was not significantly affected. We discuss the possible causes of tolerance of tundra plants to UV-B (absence of response to enhanced UV-B) in terms of methodology (supplementation versus exclusion), ecophysiological adaptations to UV-B and the biogeographical history of polar plants  相似文献
