Summary From a large number of rice varieties tested, no variety was identified as resistant to tungro bacilliform virus (RTBV). Only in Utri Merah was the RTBV multiplication restrictive, whereas other varieties such as Kataribhog and Pankhari 203 were identified as tolerant. These varieties were crossed with a susceptible variety. TN1, to study the inheritance of restrictive multiplication and tolerance to RTBV. After 3 weeks of inoculation with RTBV, F1; F2, and F3 progenies were assessed by enzyme-linked immunosorbent assay (ELISA). The RTBV concentration in all F1 populations was intermediate between parents. The frequency distribution of F2 seedlings with various levels of RTBV concentration indicated that the RTBV tolerance is controlled by multiple genes. The RTBV concentrations in F1 and F2 progenies from the Utri Merah x TN1 cross revealed that restrictive multiplication of RTBV in Utri Merah is a polygenic character. The continuous variation observed in F2 populations from crosses between tolerant varieties and Utri merah indicated no allelic relationships between tolerant and restrictive multiplication traits.Part of PhD thesis submitted by senior author to University of Kebangsaan Malaysia, Bangi  相似文献

Summary The effect of heating on the properties of Apomu (Psammentic Usthorthent), Egbeda (Oxic Paleustalf) and Gambari (Typic Plinthustalf) surface soils were studied under laboratory conditions. Heating at low temperatures (100°C) have no detrimental effects on soil properties, on the contrary it increased the soil extractable P, Mg, Fe, Mn and Zn levels. Pronounced reductions in total N, Org. C, Org. P and extractable Ca and Mg levels and marked increases in extractable P, Zn, Mn and Fe were observed by heating to 200°C. Heating to 500° had an adverse effect on soil chemical and physical properties.Plant height and dry matter yeild of rice plants were higher when grown on Egbeda soil previously heated to 100°C. With addition of N, P and K there was no observed beneficial effect of the heating treatment. Rice plants grown on Egbeda soil previously heated to 200°C showed high uptake of Mn. Plants grew badly in soil previously heated to 500°C.  相似文献

Background  

Plant genomes contain various kinds of repetitive sequences such as transposable elements, microsatellites, tandem repeats and virus-like sequences. Most of them, with the exception of virus-like sequences, do not allow us to trace their origins nor to follow the process of their integration into the host genome. Recent discoveries of virus-like sequences in plant genomes led us to set the objective of elucidating the origin of the repetitive sequences. Endogenous rice tungro bacilliform virus (RTBV)-like sequences (ERTBVs) have been found throughout the rice genome. Here, we reconstructed putative virus structures from RTBV-like sequences in the rice genome and characterized to understand evolutionary implication, integration manner and involvements of endogenous virus segments in the corresponding disease response.  相似文献

在江西双季稻田进行长期田间定位试验,分析了多年保护性耕作对水稻产量、土壤理化性状及生物学性状的影响。连续8a稻田保护性耕作处理的平均产量高于传统耕作4.46%—8.79%,各处理的有效穗数、每穗粒数和结实率均高于对照,而各处理间穗长和千粒重差异不显著。实行稻田保护性耕作处理的土壤容重低于传统耕作3.6%—5.6%,而总孔隙度和毛管孔隙度分别高出传统耕作1.6%—17.4%、2.4%—16.7%。与传统耕作相比,连续8a保护性耕作显著提高了土壤有机质(2.9%—10.0%)、有效磷(4.8%—31.6%)、速效钾(9.7%—25.7%)。在2005年免耕+插秧的土壤真菌数量最多,显著高于对照处理51.6%,免耕+抛秧在2008年达到最大,显著高于对照处理54.1%。2012年免耕+抛秧、免耕+插秧显著高于对照126.1%、121.1%;另外,各处理间过氧化氢酶、脲酶活性均差异不显著。8a间土壤转化酶活性变化范围在0.292—0.451 mg/g之间,其中2005—2007、2012年均是免耕+抛秧达到最大,与对照相比,增加范围为72.7%—137.7%,且差异显著(P0.05)。因此,实行稻田保护性耕作是适合江南丘陵区双季稻区农业可持续发展的有效模式之一,其中免耕+抛秧和免耕+插秧两种方式效果最为显著。  相似文献

Comparative transmission by leafhoppers of three tungro isolates obtained from the Philippines, India and Malaysia, and of an infectious clone of the Philippine isolate of rice tungro bacilliform virus (RTBV) by agroinoculation, was conducted on 12 rice cultivars. The symptoms, including height of inoculated plants were recorded and the efficiency of RTBV and rice tungro spherical virus (RTSV) transmission was determined by enzyme-linked immunosorbent assay. In most cases, the reduction of height and leaf symptoms of plants infected with RTBV and/or RTSV by the three isolates were similar in any given cultivar. On cultivar ASD 7 , the Malaysian isolate showed more severe yellow orange leaf discolouration symptoms than the Indian isolate which in turn had more severe leaf discolouration than the Philippine isolate. On the other hand, cultivars ASD 7 and Ptb 18 produced the most severe yellow orange leaf discolouration when agroinoculated with an infectious RTBV clone of the Philippine isolate. There was some variation in the transmission profile of the two tungro viruses among the three isolates. However, there was no one clear set of characteristics by which one could use cultivars to distinguish isolates. The amount of viral DNA in agroinfected plants of cultivars Utri merah, Balimau putih, Utri Rajapan and ARC 11554 was low, while the amount was high in cultivars TN1, ASD7, Ptb 18 and TKM 6. There was high correlation between the amount of viral coat protein by ELISA and viral nucleic acid by DNA hybridisation on 10 agroinoculated rice cultivars; this might indicate that similar proportions of the total RTBV DNA are encapsidated in each cultivar.  相似文献

在江苏淮北东海县季节性干旱严重的岭沙土上,开展了施用保水剂和稻草覆盖对保持土壤水分和作物产量的效应的比较试验,保水剂和稻草的用量各分4级,分别为1,2,3,6g.kg^-1土和1500,3000,4500,6000kg/hm^-2,结果表明,两者均可促进小麦生长,提高当季及后茬作物产量,覆盖稻草和施用保水剂分别比对照增产小麦12.5%和10%,其作用机制在于两者均能减少土壤水分蒸发,提高土壤持水能力,增加有效水供应,并对土壤容重,温度及养分状况有一定的改善作用。  相似文献

Rice flour is becoming very attractive as raw material, but there is lack of information about the influence of particle size on its functional properties and starch digestibility. This study evaluates the degree of dependence of the rice flour functional properties, mainly derived from starch behavior, with the particle size distribution. Hydration properties of flours and gels and starch enzymatic hydrolysis of individual fractions were assessed. Particle size heterogeneity on rice flour significantly affected functional properties and starch features, at room temperature and also after gelatinization; and the extent of that effect was grain type dependent. Particle size heterogeneity on rice flour induces different pattern in starch enzymatic hydrolysis, with the long grain having slower hydrolysis as indicated the rate constant (k). No correlation between starch digestibility and hydration properties or the protein content was observed. It seems that in intact granules interactions with other grain components must be taken into account. Overall, particle size fractionation of rice flour might be advisable for selecting specific physico-chemical properties.  相似文献

Effects of different cellulose derivatives and carrageenans on the pasting, rheological, and textural properties of normal (NRS) and waxy (WRS) rice starches were investigated. When suspensions of both NRS and WRS were heated in a Rapid Visco Analyser (RVA) in the presence of the hydrocolloids, increases in apparent pasting temperatures and peak and final viscosities in the following decreasing order were observed: methylcellulose (MC) > carboxymethylcellulose (CMC) for cellulose derivatives and λ- > í- > κ-carregeenan for carrageenans. Slight decreases in peak and final viscosities were observed when hydroxypropylmethylcellulose (HPMC) was the hydrocolloid. Dynamic viscoelasticity measurements indicated that NRS–hydrocolloid pastes were less solid-like than the control, as evidenced by their higher tan δ values, whereas tan δ values of WRS–hydrocolloid pastes were the same as that of the control. Steady shear rheological measurements showed that addition of the different hydrocolloids used increased the apparent viscosity (ηa,100) and consistency coefficient (K) values of both starches with the same trend as that observed during pasting, whereas the opposite trend was observed for the flow behavior index (n) values. The hardness and adhesiveness of NRS pastes were significantly increased by addition of κ- and í-carrageenans, but were unaffected by the other hydrocolloids. A similar effect was observed for WRS, with the exception of κ-carregeenan, in which the hardness of the mixed paste was decreased. The starch–hydrocolloid pastes exhibited a phase-separated microstructure in which amylose- and amylopectin-rich domains were dispersed in a hydrocolloid-rich continuous phase.  相似文献

Class I and class II CPD photolyases are enzymes which repair pyrimidine dimers using visible light. A detailed characterization of class I CPD photolyases has been carried out, but little is known about the class II enzymes. Photolyases from rice are suitable for functional analyses because systematic breeding for long periods in Asian countries has led to the selection of naturally occurring mutations in the CPD photolyase gene. We report the biochemical characterization of rice mutant CPD photolyases purified as GST-form from Escherichia coli. We identified three amino acid changes, Gln126Arg, Gly255Ser, and Gln296His, among which Gln but not His at 296 is important for complementing phr-defective E. coli, binding UV-damage in E. coli, and binding thymine dimers in vitro. The photolyase with Gln at 296 has an apoenzyme:FAD ratio of 1 : 0.5 and that with His at 296 has an apoenzyme:FAD ratio of 1 : 0.12-0.25, showing a role for Gln at 296 in the binding of FAD not in the binding of thymine dimer. Concerning Gln or Arg at 126, the biochemical activity of the photolyases purified from E. coli and complementing activity for phr-defective E. coli are similarly proficient. However, the sensitivity to UV of cultivars differs depending on whether Gln or Arg is at 126. The role of Gln and Arg at 126 for photoreactivation in rice is discussed.  相似文献

The growing number of rice microsatellite markers warrants a comprehensive comparison of allelic variability between the markers developed using different methods, with various sequence repeat motifs, and from coding and non-coding portions of the genome. We have performed such a comparison over a set of 323 microsatellite markers; 194 were derived from genomic library screening and 129 were derived from the analysis of rice-expressed sequence tags (ESTs) available in public DNA databases. We have evaluated the frequency of polymorphism between parental pairs of six inter- subspecific crosses and one inter-specific cross widely used for mapping in rice. Microsatellites derived from genomic libraries detected a higher level of polymorphism than those derived from ESTs contained in the GenBank database (83.8% versus 54.0%). Similarly, the other measures of genetic variability [the number of alleles per locus, polymorphism information content (PIC), and allele size ranges] were all higher in genomic library-derived microsatellites than in their EST-database counterparts. The highest overall degree of genetic diversity was seen in GA-containing microsatellites of genomic library origin, while the most conserved markers contained CCG- or CAG-trinucleotide motifs and were developed from GenBank sequences. Preferential location of specific motifs in coding versus non-coding regions of known genes was related to observed levels of microsatellite diversity. A strong positive correlation was observed between the maximum length of a microsatellite motif and the standard deviation of the molecular-weight of amplified fragments. The reliability of molecular weight standard deviation (SDmw) as an indicator of genetic variability of microsatellite loci is discussed. Received: 5 May 1999 / Accepted: 16 August 1999  相似文献

1991-2010年宁夏水稻病虫害发生特征与经济损失分析   总被引:2,自引:0,他引:2  
水稻病虫害发生种类繁多、暴发频繁,是威胁宁夏水稻稳产、高产的重要因素,但对其变化趋势与实际危害损失不清楚.基于宁夏水稻统计数据、稻田覆盖类型遥感数据和水稻产量数据,分析1991年至2010年宁夏水稻病虫害发生特征与经济损失情况.结果表明:1991年到2010年期间,宁夏水稻病害、虫害年均发生面积分别为5.3万hm2和2.0万hm2,其年均防治面积分别为8.4万hm2和1.6万hm2;水稻病害的发生程度下降了23.10%,防治程度上升了77.30%.20年间宁夏水稻虫害的发生面积、发生程度、防治面积和防治程度均显著增加.防治水稻病害、虫害后,分别挽回稻谷为2.67万吨、0.28万吨,其挽回损失量在20年期间分别增加了55.15%、2775.0%,表明水稻病虫害防治意义重大.由于气候变化等诸多因子,导致1991年到2010年宁夏水稻病害、虫害年均造成的实际稻谷损失量分别为0.71万吨与0.13万吨,水稻病害实际损失量在0值附件波动、虫害呈现波动增加趋势,说明水稻病害防控效果好、虫害的防控还有提升的空间.从全区各市县分布来看,水稻病虫害主要分布在宁夏的银北地区.为有效地防止或减少病虫害对水稻产量的损失,应加强农田景观变化和气候变化等对水稻病虫害发生与灾变的风险评估和监测预警,开展区域性水稻病虫害综合治理研究,并建立相应的防控新对策与技术体系.  相似文献

Summary Thirty mapped Indica rice genomic (RG) clones were partially sequenced from each end. From such sequence data, pairs of oligonucleotides were synthesized to act as primers for polymerase chain reaction (PCR) amplification of the corresponding loci in crude total DNA preparations. The PCR products from DNA of Indica varieties were of the sizes expected from the sizes of the corresponding RG clones. However, size polymorphisms were seen between PCR products from Indica and Japonica varieties, and among wildOryza species. Restriction fragment length polymorphism (RFLP) was observed between PCR products of Indica varieties simply by electrophoretic analysis of restricted products, without the need for Southern hybridization or radiolabelling. The RFLPs noted between varieties ARC6650 and Phalguna were inherited in recombinant inbred lines derived from a cross between them. The RFLPs were detectable in PCR products amplified from DNA extracted by a simple procedure from single seedlings or leaves, and revealed genetic heterogeneity in cultivated lines. An approach is described that is relevant to the acceleration of classical plant breeding through molecular techniques.  相似文献

通过培养试验研究了猪粪和稻草对Cd污染黄泥土生物活性的影响,结果表明,Cd污染土壤的生物活性下降,施用有机肥料后,土壤有效态Cd含量降低,降幅约为40%;微生物量C、N、P和脱氢酶、过氧化氢酶的活性增高,增幅为30%~100%,其中微生物量C、N与土壤有效态Cd之间有显著的负相关关系,可作为污染土壤的生物指标。  相似文献

水稻内生放线菌OsiRt-1的分离鉴定及对稻瘟病的防治作用   总被引:2,自引:0,他引:2  
【目的】从水稻中分离、筛选并鉴定出对稻瘟病真菌具有拮抗性能的内生放线菌,对高抗菌株进行稻瘟病生防效果评定,初步探讨其作用机制。【方法】采用4种分离培养基对水稻内生放线菌进行分离,用平板对峙法筛选出对稻瘟病菌拮抗性能最好的菌株,结合其菌落形态、生理生化及16S rRNA基因序列分析进行鉴定;用环境扫描电镜(SEM)观察拮抗菌对稻瘟病菌菌丝形态影响,用菌丝生长速率法对拮抗菌发酵滤液进行抗菌活性评价;大田条件下,喷洒拮抗菌孢子液于水稻,检测其稻瘟病控制效果;同时,对拮抗菌进行产酶和次级代谢产物分析,检测其聚酮合酶基因(PKSⅡ型)和非核糖体多肽合成酶基因(NRPS)。【结果】从1 800个水稻样品中分离出117株内生放线菌,从中筛选出拮抗性能最好的菌株Osi Rt-1,鉴定为米修链霉菌Streptomyces misionensis。该菌株使稻瘟病菌菌丝出现畸形,其无菌滤液对病菌菌丝生长的抑制率为28.06%;大田条件下,Osi Rt-1对水稻苗瘟、穗瘟均有较好防效,其中对苗瘟防效为7.76%,穗瘟防效高达25.65%,损失率降低了17.46%。与之对应,Osi Rt-1处理过的水稻地上部分,Osi Rt-1所占内生放线菌的比例明显高于未处理水稻。该菌株具有可能降解真菌细胞壁的纤维素酶、蛋白酶活性,同时可产生植物生长促进剂铁载体、IAA、ACC脱氨酶。菌株Osi Rt-1呈现PKSⅡ型和NRPS阳性。【结论】菌株Osi Rt-1是一株具生防潜力的内生放线菌,在农业上具有实际研发价值。  相似文献
