【目的】本研究旨在克隆绿盲蝽Apolygus lucorum的气味受体基因,明确这些气味受体基因在绿盲蝽成虫不同组织中的表达水平及对主要寄主植物挥发物的电生理反应,为揭示绿盲蝽对寄主植物的识别机制提供理论基础。【方法】在绿盲蝽成虫触角转录组测序与分析的基础上,通过PCR技术克隆得到8个具有完整ORF的气味受体基因序列。用qPCR研究这8个基因在雌雄成虫不同组织(触角、无触角的头、胸、腹、足和翅)中的表达水平。然后通过爪蟾卵母细胞体外表达结合双电极电压钳技术测试这些气味受体对56种气味化合物的电生理反应。【结果】克隆得到了绿盲蝽8个气味受体基因AlucOR9, AlucOR16,AlucOR38, AlucOR53, AlucOR55, AlucOR56, AlucOR57和AlucOR58的cDNA全长序列(GenBank登录号:MN905538-MN905545)。这些气味受体含有7个跨膜结构域,且N末端位于胞内,C末端位于胞外,符合昆虫气味受体的典型特征。qPCR结果表明,这8个气味受体基因均在绿盲蝽成虫触角中高表达,而在其他组织中低表达,其中除AlucOR38外的其他7个气味受体基因在雌雄成虫触角间存在显著的表达差异。双电极电压钳记录结果显示,AlucOR57对其中15种气味化合物(苯甲醛、氧化石竹烯、庚醛、反-2-己烯醛、乙酸苯甲酯、桃金娘烯醛、4-乙基苯甲醛、乙酸壬酯、四氢芳樟醇、十三烷、反-3-己烯醇、2-丙烯酸丁酯、丙酸丁酯、乙酸辛酯和乙酸戊酯)有反应,其余7个气味受体对测试的全部气味化合物均无反应。【结论】AlucOR57对测试的一些气味化合物有反应,推测其在绿盲蝽的寄主识别过程中发挥重要作用;其他7个气味受体对测试气味化合物均无反应,其功能有待进一步研究。  相似文献

Host preference and blood feeding are restricted to female mosquitoes. Olfaction plays a major role in host-seeking behaviour, which is likely to be associated with a subset of mosquito olfactory genes. Proteins involved in olfaction include the odorant receptors (ORs) and the odorant-binding proteins (OBPs). OBPs are thought to function as a carrier within insect antennae for transporting odours to the olfactory receptors. Here we report the annotation of 32 genes encoding putative OBPs in the malaria mosquito Anopheles gambiae and their tissue-specific expression in two mosquito species of the Anopheles complex; a highly anthropophilic species An. gambiae sensu stricto and an opportunistic, but more zoophilic species, An. arabiensis. RT-PCR shows that some of the genes are expressed mainly in head tissue and a subset of these show highest expression in female heads. One of the genes (agCP1588) which has not been identified as an OBP, has a high similarity (40%) to the Drosophila pheromone-binding protein 4 (PBPRP4) and is only expressed in heads of both An. gambiae and An. arabiensis, and at higher levels in female heads. Two genes (agCP3071 and agCP15554) are expressed only in female heads and agC15554 also shows higher expression levels in An. gambiae. The expression profiles of the genes in the two members of the Anopheles complex provides the first step towards further molecular analysis of the mosquito olfactory apparatus.  相似文献

Testosterone-dependent olfactory signals emitted by male are well known to accelerate female puberty in mice (Vandenbergh effect). However, it remains unclear whether these chemosignals also influence adult expression of male-directed odor preference. Therefore, we exposed female mice to intact or castrated male bedding (vs clean bedding as control) during the peripubertal period (postnatal day (PD) 21–38) and measured male-directed odor preference in adulthood. At PD45 or PD60, females exposed to intact male odors, and thus showing puberty acceleration, preferred to investigate odors from intact males over females or castrated males. Females exposed to castrated male odors did not show puberty acceleration but preferred male (intact or castrated) over female odors. Finally, control females did not show any odor preference when tested at PD45, although a preference for male odors emerged later (PD60). In a second experiment, females that were exposed to intact male odors after pubertal transition (PD36–53) also preferred intact male over castrated male odors. In conclusion, our results indicate that peripubertal exposure to male odors induced early expression of male-directed odor preference regardless of puberty-accelerating effect and that induction of male-directed odor preference is not specific to the peripubertal period.  相似文献

Successful reproduction in vertebrates depends critically upon a suite of precopulatory behaviors that occur prior to mating. In Syrian hamsters (Mesocricetus auratus), these behaviors include vaginal scent marking and preferential investigation of male odors. The neural regulation of vaginal marking and opposite-sex odor preference likely involves an interconnected set of steroid-sensitive nuclei that includes the medial amygdala (MA), the bed nucleus of the stria terminalis (BNST), and the medial preoptic area (MPOA). For example, lesions of MA eliminate opposite-sex odor preference and reduce overall levels of vaginal marking, whereas lesions of MPOA decrease vaginal marking in response to male odors. Although BNST is densely interconnected with both MA and MPOA, little is known about the role of BNST in female precopulatory behaviors. To address this question, females received either bilateral, excitotoxic lesions of BNST (BNST-X) or sham lesions (SHAM), and were tested for scent marking and for investigatory responses to male and female odors. Whereas SHAM females vaginal marked more to male odors than female odors on two days of the estrous cycle, BNST-X females marked at equivalent levels to both odors. This deficit is not due to alterations in social odor investigation, as both BNST-X and SHAM females investigated male odors more than female odors. Finally, BNST lesions did not generally disrupt the cyclic changes in reproductive behaviors that occur across the estrous cycle. Taken together, these results demonstrate that BNST is critical for the normal expression of solicitational behaviors by females in response to male odor stimuli.  相似文献

A small cDNA library was constructed from antennae of 100 adult male Aedes aegypti yellow fever mosquitoes. Sequencing of 80 clones identified 49 unique gene products, including a member of the Odorant Binding Protein family (Aaeg-OBP10), a homologue of Takeout (Aaeg-TO), and transposable elements of the LINE, SINE and MITE classes. Aaeg-OBP10 encodes a 140 amino acid protein including a predicted 25 amino acid signal peptide. Aaeg-OBP10 expression was adult male enriched, increased with adult age, and greatest in antennae and wings but also present in maxillary palps, proboscis and leg. Aaeg-OBP10 is a likely orthologue of Agam-OBP10 of the malaria mosquito Anopheles gambiae and shares significant similarity with members of the OBP56 gene cluster of Drosophila melanogaster. These OBP genes may represent a unified class of OBPs with unique roles in chemodetection; the expression pattern of Aaeg-OBP10 suggests it may play a role in adult male chemosensory behavior. Aaeg-TO encodes a 248 amino acid protein including a predicted 22 amino acid signal peptide. Aaeg-TO is homologous with the circadian/feeding regulated D. melanogaster Takeout protein (Dmel-TO) and a subclass of Juvenile Hormone Binding Proteins (JHBP) characterized by Moling from Manduca sexta; both Dmel-TO and Moling are sensitive to feeding, suggesting Aaeg-TO might regulate the antennal response to food, host or pheromonal odors in a JH sensitive manner. Aaeg-TO was used to identify 25 D. melanogaster and 13 A. gambiae homologues by Blast analysis suggesting these may comprise a relatively large class of protein involved in the hormonal regulation of behavior.  相似文献

Odor-detection in the malaria mosquito Anopheles gambiae involves large families of diverse proteins, including multiple odorant binding proteins (AgOBPs) and olfactory receptors (AgORs). The receptors AgOR1 and AgOR2, as well as the binding protein AgOBP1, have been implicated in the recognition of human host odors. In this study, we have explored the expression of these olfactory proteins, as well as the ubiquitous odorant receptor heteromerization partner AgOR7, in the thirteen flagellomeres (segments) of female and male antenna. Expressing cells were visualized by adapting a whole mount fluorescence in situ hybridization method. In female mosquitoes, AgOR1-expressing olfactory receptor neurons (ORNs) were almost exclusively segregated in segments 3 to 9, whereas AgOR2-expressing ORNs were distributed over flagellomeres 2 to 13. Different individuals comprised a similar number of cells expressing a distinct AgOR type, although their antennal topography and number per flagellomere varied. AgOBP1-expressing support cells were present in segments 3 to 13 of the female antenna, with increasing numbers towards the distal end. In male mosquitoes, total numbers of AgOR- and AgOBP1-expressing cells were much lower. While AgOR2-expressing cells were found on both terminal flagellomeres, AgOR1 cells were restricted to the most distal segment. High densities of AgOBP1-expressing cells were identified in segment 13, whereas segment 12 comprised very few. Altogether, the results demonstrate that both sexes express the two olfactory receptor types as well as the binding protein AgOBP1 but there is a significant sexual dimorphism concerning the number and distribution of these cells. This may suggest gender-specific differences in the ability to detect distinct odorants, specifically human host-derived volatiles.  相似文献
