Blood lead levels of 634 healthy Japanese (422 males and 212 females), aged 0–87, living in the Kyushu and Okinawa Islands, were determined by the colorimetric dithizone method and atomic absorption spectrometry. The lead level in the atmospheric environment was not extremely high; it was below 1 μg/m3 in most of the places examined. The differences in the geometric means of blood lead levels between 0–5 yr and other age groups were statistically significant (P<0.01). The blood lead levels of Japanese seem to increase in late teens and then decrease slowly to a steady state. The accumulation of lead taken from food was calculated by assuming the amount of lead from food corresponds to the caloric intake recommended for each Japanese age group (e.g., assuming 150 μg/2700 kcal for boys in late teens). Lead concentration in the air was assumed to be 1 μg/m3. The pattern of lead accumulation in blood by age was similar to the theoretical curve for the accumulated total amount of lead obtained by simulation. The fact probably indicates that blood lead levels in Japanese are dependent on lead in daily foods rather than on lead in the atmospheric environment.  相似文献

Background, Aim and Scope  

By using recycled aluminium or by disposing used aluminium products for recycling, it is normal LCA practice to give a credit for the avoided production of primary or recycled aluminium. Lately, consequential approaches have been suggested to qualify and quantify this credit in terms of market mechanisms. Depending on supply, demand and price elasticity of primary products and scrap products, a mixed share of primary and recycled material may be credited. Aluminium, having high energy consumption for its primary production and low energy consumption for recycling, is very sensitive concerning whether production of primary or recycled aluminium is avoided. This paper includes presentations of aluminium products which are typically made from primary and from recycled aluminium. This is essential concerning which production may be avoided. Examples of market mechanism parameters of aluminium for consequential LCA are given.  相似文献

The mithramycin fluorescence procedure described by B. T. Hill and S. Whatley (1975, FEBS Lett., 56, 20–23) for DNA measurement tends to underestimate DNA concentrations in biological samples as compared to the results obtained by the diphenylamine reaction. This discrepancy disappears when DNA is first solubilized, by buffer containing heparin, from either cell homogenates or nuclear preparations. The optimal conditions for maximal fluorescence are 8 mm Mg2+, 10 μg/ml mithramycin, and heparin to DNA ratios ≥0.15 (ww). Background fluorescence is reduced 90% by dextran-coated charcoal adsorption of unbound mithramycin. The limit of sensitivity of the assay is 0.3 μg/ml and fluorescence is linear up to 30 μg DNA/ml.  相似文献

Summary Xylem cavitation has been studied in Ricinus plants using vibration detection to examine its induction by different factors. These observations provide considerable circumstantial evidence in justification of the new technique as already described and further developed. In general cavitation is induced only when the tissue water balance is reduced hydrostatically. Thus cavitation is promoted by intense radiation which enhances transpiration, or alternatively by the blockage of xylem conduits by suspended particles carried in the transpiration stream. In contrast a reduction in radiation, or prevention of transpiration tends to restrict cavitation. Thus cavitation can be prevented by immersing a leaf in liquid paraffin. This technique has been used to see if radioactive bombardment would trigger its induction but no detectable effect has been observed even when exposed to intense radiation.An excised leaf, losing water in air, produces a click total. On restoration to full turgor by standing the petiole in water it recovers very slowly and subsequently its click total is much reduced. If however the newly wilted leaf is allowed to recover in water following gas evacuation treatment the cavitation total often approaches the original and the rate of recovery is extremely rapid. Apparently gas emboli develop rapidly in conduits which have cavitated, but they can be removed by vacuum injection: the conduits refill and conduction is restored.  相似文献

* In thylakoids from Nicotiana benthamiana infected with the pepper mild mottle virus (PMMoV), a decreased amount of the PsbP and PsbQ proteins of photosystem II and different proteins of the Calvin cycle have been previously observed. We used thermoluminescence to study the consequences in vivo. * Measurements on unfrozen discs from symptomatic and asymptomatic leaves of plants infected by two tobamovirus PMMoV-S and PMMoV-I strains were compared with homologous samples in control plants. * Thermoluminescence emission did not reveal noticeable alteration of PSII electron transfer activity in infected symptomatic leaves. In these leaves, the relative intensity of the 'afterglow' emission indicated an increase of the NADPH + ATP assimilatory potential, contrasting with its decrease in asymptomatic leaves. High-temperature thermoluminescence, as a result of peroxides, increased in symptomatic and asymptomatic leaves. * In young infected leaves, PSII activity is preserved, producing a high assimilatory potential. Older asymptomatic leaves export more nutrients towards young infected leaves. This depresses their assimilatory potential and weakens their defence mechanisms against reactive oxygen species, resulting in higher peroxide content.  相似文献

1. The incorporation of methyl groups into histones from dimethylnitrosamine and from methionine was studied by injection of the labelled compounds, isolation of rat liver and kidney histones, and analysis of hydrolysates by column chromatography. 2. Labelled methionine gave rise to labelled in-N-methyl-lysine, di-in-N-methyl-lysine and an amino acid presumed to be omega-N-methyl-arginine. 3. Administration of labelled dimethylnitrosamine gave rise to labelled S-methylcysteine, 1-methylhistidine, 3-methylhistidine and in-N-methyl-lysine derived from the alkylating metabolite of dimethylnitrosamine. In addition, labelled formaldehyde released by metabolism of dimethylnitrosamine leads to the formation of labelled S-adenosylmethionine, and hence to labelling of in-N-methyl-lysine, di-in-N-methyl-lysine and omega-N-methylarginine by enzymic methylation. 4. The formation of in-N-methyl-lysine by alkylation of liver histones was confirmed by using doubly labelled dimethylnitrosamine to discriminate between direct chemical alkylation and enzymic methylation via S-adenosylmethionine. These experiments also suggested the possibility that methionine residues in the histones were alkylated to give methylmethionine sulphonium residues. 5. The extent of alkylation of liver histones was maximal at about 5h after dosing and declined between 5 and 24h. The methylated amino acids resulting from direct chemical alkylation were preferentially lost: this is ascribed to necrosis of the more highly alkylated cells. 6. Liver histones were about four times as alkylated as kidney histones; the extent of alkylation of liver histones was similar to that of liver total nuclear proteins. 7. Methyl methanesulphonate (120mg/kg) alkylated liver histones to a greater extent than did dimethylnitrosamine. Diethylnitrosamine also alkylated liver histones. 8. The results are discussed with regard to the possible effects of alkylation on histone function, and the possible role of histone alkylation in carcinogenesis by the three compounds.  相似文献

The development of somatic embryos is, in many plants, inhibited by 2,4-dichlorophenoxyacetic acid (2,4-D) and other auxins. The finding that difluoromethylornithine (DFMO) can counteract this inhibition has been used to test some of the hypotheses for the mechanism of inhibition.
Inhibition of somatic embryogenesis in carrot ( Daucus carota L.) by exogenous ethylene (from ethephon), antioxidants (ascorbic acid and glutathione), ethanol/acetaldehyde and abscisic acid was not counteracted by DFMO, indicating that the inhibitory effect of 2,4-D is not manifest through the formation of these compounds. Embryogenesis was abolished by micromolar concentrations of the polar auxin transport inhibitors 2, 3, 5-triiodobenzoic acid (TIBA), N-1-naphthylphthalamic acid (NPA) and 9-hydroxyfluorene-9-carboxylic acid (HFCA). This inhibition was counteracted to a considerable extent by DFMO. Inhibition by relatively high concentrations of the antiauxin 2-( p -chlorophenoxy)-isobutyric acid (CPIB), which does not affect polar auxin transport, was in contrast not counteracted by DFMO. These findings indicate that exogenous auxins may inhibit embryogenesis by interfering with the ability of postglobular embryos to set up internal auxin gradients necessary for polarized growth.  相似文献

The relative accuracy of two pregnancy testing methods for swine were compared in a field study. The procedures used were manual palpation and amplitude-depth ultrasonic scanning. A total of 369 sows were examined by both methods. Seven additional gilts were examined by ultrasound only and 46 sows by palpation per rectum only. The number of correct positive and negative diagnoses made by both methods were calculated, and determination of accuracy as well as comparison between the tests were made on this basis. The relative accuracy was 97.6% for the manual method and 96.8% for the ultrasound method. Both tests had a high sensitivity, 99.2 and 98.9%, respectively. The ability of the tests to detect the non-pregnant animals was not as high, which is reflected by a lower specificity. No significant differences were noted between the two methods. A lower specificity and a lower negative predictive value were provided by ultrasound scanning as compared with those acquired by manual palpation. Both procedures were considered to be quick and convenient to perform. It was concluded that in spite of the new pregnancy testing methods introduced in the swine industry, manual palpation remains the most practical in terms of its accuracy, ease, and the minimal requirement for equipment. In gilts, palpation is unsuitable and ultrasonography currently remains the best choice for the diagnosis of pregnancy.  相似文献
