A subtidal seaweed collected in antarctic waters, Plocamium cartilagineum (L. Dix.), displayed induction of mRNAs encoding the 70 kDa heat shock protein (HSP70) and the ubiquitin polyprotein (UBI) when incubated at 5°C. Maximal induction of HSP70 mRNA was observed when the alga was incubated at 10°C for 1 h. Incubations at higher temperatures or for longer periods reduced the amount of HSP70 mRNA detected. Incubations at 20°C or greater resulted in cell death. These data indicate that dispite the unusually low temperature of induction, this macrophyte exhibits a heat shock response similar to that of other organisms at temperatures 5 to 10°C above usual growth conditions.  相似文献

The effects of cryoprotectant alone and cryoprotectant plus additives on the preservation of polymorphonuclear neutrophils (PMNs) at cryogenic temperatures (?80 °C) were studied.A considerable difference among cryoprotective agents was observed in their protective effect against freeze injury of neutrophils. Based upon degree of chemotactic inhibition and impairment of trypan blue exclusion, Me2SO proved to be superior to ethylene glycol and glycerol as a cryoprotectant and to exhibit the best protective effect at a concentration of 4.2%. When PMNs were frozen in Me2SO alone, the ability of PMNs to exclude dye was retained after 3 days of cryopreservation, while chemotaxis was inhibited markedly. One-week preservation produced the death of 50% of the cells. To improve the protective effect of Me2SO against chemotactic inhibition by cryopreservation, additives such as glucose, ATP, and albumin were included in the freezing medium. Addition of albumin displayed the most distinct improvement in the recovery of chemotaxis, although ATP also exhibited a protective effect, especially during short-term storage. Studies on the combined effect of these additives with ethylene glycol or glycerol showed that only albumin had a considerably better protective effect against dye exclusion injury but not against chemotactic inhibition. Phagocytosis and adhesion were less inhibited by freezing than was chemotaxis. A combination of Me2SO and ATP markedly protected phagocytosis and adhesion from freeze injury. However, cyanide-insensitive oxygen uptake during phagocytosis, as well as chemotaxis, were considerably inhibited.  相似文献

A thermotolerant Aspergillus sp. strain ST-01 degrading poly(-caprolactone) films was isolated. The polyester was degraded and assimilated giving 36 mg of cell from 100 mg sample and 10 mg yeast extract after 6 days at 50 °C. The degradation products were identified as succinic acid, butyric acid, valeric acid, and caproic acid. The isolate also degraded more than 90% film samples of polyhydroxybutyrate (PHB) and poly(tetramethylene succinate-co-tetramethylene adipate) at 50 °C.  相似文献

The growth of all microorganisms is limited to a specific temperature range. However, it has not previously been determined to what extent global protein profiles change in response to temperatures that incrementally span the complete growth temperature range of a microorganism. As a result it has remained unclear to what extent cellular processes (inferred from protein abundance profiles) are affected by growth temperature and which, in particular, constrain growth at upper and lower temperature limits. To evaluate this, 8-plex iTRAQ proteomics was performed on the Antarctic microorganism, Methanococcoides burtonii. Methanococcoides burtonii was chosen due to its importance as a model psychrophilic (cold-adapted) member of the Archaea, and the fact that proteomic methods, including subcellular fractionation procedures, have been well developed. Differential abundance patterns were obtained for cells grown at seven different growth temperatures (-2°C, 1°C, 4°C, 10°C, 16°C, 23°C, 28°C) and a principal component analysis (PCA) was performed to identify trends in protein abundances. The multiplex analysis enabled three largely distinct physiological states to be described: cold stress (-2°C), cold adaptation (1°C, 4°C, 10°C and 16°C), and heat stress (23°C and 28°C). A particular feature of the thermal extremes was the synthesis of heat- and cold-specific stress proteins, reflecting the important, yet distinct ways in which temperature-induced stress manifests in the cell. This is the first quantitative proteomic investigation to simultaneously assess the response of a microorganism to numerous growth temperatures, including the upper and lower growth temperatures limits, and has revealed a new level of understanding about cellular adaptive responses.  相似文献

Pigeon erythrocytes expelled preloaded 45Ca2+ in response to a low dose of A23187 at 0° C. We call this phenomenon ‘paradoxical’ expulsion. Within the first minute, 1.85 ± 0.38 μmol/l cell water was expelled; after that the internal 45Ca2+ began to rise. The rises in Ca2+ uptake with and without A23187 addition were essentially paralleled. No premonitory rise of 45Ca2+ upon the addition of A23187 was observed. Expulsion of 45Ca2+ in response to A23187 was probably by the action of the Ca2+ pump and not by Na+-Ca2+ exchange since vanadate inhibited, but K+ replacement of Na+ in the medium had no effect. Lysophosphatidylcholine (lysoPC) caused an abrupt increase in 45Ca2+ influx by cells at 0° C and was dose dependent. However, a very low dose of lysoPC induced expulsion of preloaded 45Ca2+ similar to that by A23187, the response was fast and transitory, without any premonitory rise in 45Ca2+ uptake. The results lend support to the suggestion that the signal to which cells respond may be a sudden change in Ca2+ influx per se rather than a change in internal Ca2+ concentration. These features of ‘paradoxical’ 45Ca2+ expulsion induced by A23187 and lysoPC are not expected from mass-action equilibria but, instead, agree with the characteristics of an energy-dissipating control mechanism.  相似文献

When a pepper cultivar (Capsicum chinense cv. Seychelles-2, Sy-2) native to the Seychelles was grown in Japan, all seedlings showed seasonal developmental abnormalities such as development of abnormally shaped leaves. Other pepper cultivars grew well in all seasons while the growth of cv. Sy-2 was stunted. In this study, we first examined the effects of various changes in temperature and photoperiod on the cv. Sy-2 phenotype. The results showed that temperatures lower than 24°C led to the formation of abnormal leaves. Second, morphological and anatomical analyses of cotyledons and true leaves developed at 28 and 20°C were conducted. The narrower and thicker cotyledons developed at 20°C had fewer palisade cells in the leaf-length direction, and more cells in the leaf-thickness direction. True leaves developed at 20°C were irregularly shaped, thicker and had smaller leaf area. In addition, true leaves developed at 20°C had fewer palisade cells in the leaf-length and leaf-width directions and had more cells in the leaf-thickness direction. Furthermore, abnormal periclinal cell divisions in the mesophyll and/or epidermal cell layers were observed during leaf blade development at 20°C. These results suggest that the observed changes in cell proliferation and abnormal periclinal cell divisions were related, at least in part, to abnormal leaf development of cv. Sy-2 at temperatures below 24°C.  相似文献

Ten strains representing a novel genus of marine thermophilic archaebacteria growing at between 70 and 103°C with an optimal growth temperature of 100°C and a doubling time of only 37 min were isolated from geothermally heated marine sediments at the beach of Porto di Levante, Vulcano, Italy. The organisms are spherical-shaped, 0.8 to 2.5 m in width and exhibit monopolar polytrichous flagellation. They are strictly anaerobic heterotrophs, growing on starch, maltose, peptone and complex organic substrates. Only CO2 and H2 could be detected as metabolic products, the latter being inhibitory to growth at high concentrations. Hydrogen inhibition can be prevented by the addition of So, whereupon H2S is formed in addition, most likely as the result of a detoxification reaction. The GC-content of the DNA of isolate Vc 1 is 38 mol%. The new genus is named Pyrococcus, the fireball. Type species and strain is Pyrococcus furiosus Vc 1 (DSM 3638).  相似文献

The time-course of thyroliberin transfer to the nucleus of GH3/B6 rat pituitary prolactin cells was studied by both autoradiography and cell fractionation of intact cells exposed to [3H]thyroliberin at 4°C or 37°C. It was previously shown that thyroliberin is not degraded in these conditions. It is found by autoradiography that [3H]-thyroliberin is transferred to the nucleus of GH3/B6 cells within 5 min at least at both 37° C and 4°C. Consistent results are obtained by fractionation of cells exposed to [3H]thyroliberin at 37°C. However after binding at 4°C 50% of the cell radioactivity is extractible by glutaraldehyde and after fractionation the isolated nuclei retain only 1–1.5% of the cell radioactivity. This suggests the existence of both tightly bound and loosely bound internalized thyroliberin molecules.  相似文献

A novel group of hyperthermophilic rod-shaped motile methanogens was isolated from a hydrothermally heated deep sea sediment (Guaymas Basin, Gulf of California) and from a shallow marine hydrothermal system (Kolbeinsey ridge, Iceland). The grew between 84 and 110°C (opt: 98°C) and from 0.2% to 4% NaCl (opt. 2%) and pH 5.5 to 7 (opt: 6.5). The isolates were obligate chemolithoautotrophes using H2/CO2 as energy and carbon sources. In the presence of sulfur, H2S was formed and cells tended to lyse. The cell wall consisted of a new type of pseudomurein containing ornithin in addition to lysine and no N-acetylglucosamine. The pseudomurein layer was covered by a detergent-sensitive protein surface layer. The core lipid consisted exclusively of phytanyl diether. The GC content of the DNA was 60 mol%. By 16S rRNA comparisons the new organisms were not related to any of the three methanogenic lineages. Based on the physiological and molecular properties of the new isolates, we describe here a new genus, which we name Methanopyrus (the methane fire). We have analyzed the structure of genes encoding the glyoxylate cycle enzyme isocitrate lyase from Brassica napus L. and their expression during embryogeny and postgermination. Restriction mapping, nucleotide sequence, and DNA gel blot hybridization analyses of cDNA and genomic clones indicated that there are approximately six isocitrate lyase genes in the B. napus genome that can be divided into at least two subfamilies based upon their divergence in 5′ and 3′ untranslated regions. We showed previously that isocitrate lyase mRNA accumulates during late embryogeny and postgermination. Here, we present results which indicate that several isocitrate lyase genes are expressed at both stages of development. First, gene-specific probes were used to show that mRNAs encoded by representatives of both gene subfamilies accumulated in both late maturation stage embryos and in seedlings of B. napus. Second, a single B. napus isocitrate lyase gene, together with 3.5 kb and 1.4 kb of 5′ and 3′ flanking regions, respectively, was expressed in both embryos and seedlings of transgenic tobacco plants. The results indicated that accumulation of isocitrate lyase in late embryogeny and postgermination does not result from the alternate expression of distinct members of the gene family.  相似文献

We examined whether a hyperthermophilic microbial fuel cell (MFC) would be technically feasible. Two-chamber MFC reactors were inoculated with subsurface microorganisms indigenous to formation water from a petroleum reservoir and were started up at operating temperature 80 °C. The MFC generated a maximum current of 1.3 mA 45 h after the inoculation. Performance of the MFC improved with an increase in the operating temperature; the best performance was achieved at 95 °C with the maximum power density of 165 mWm?2, which was approximately fourfold higher than that at 75 °C. Thus, to our knowledge, our study is the first to demonstrate generation of electricity in a hyperthermophilic MFC (operating temperature as high as 95 °C). Scanning electron microscopy showed that filamentous microbial cells were attached on the anode surface. The anodic microbial consortium showed limited phylogenetic diversity and primarily consisted of hyperthermophilic bacteria closely related to Caldanaerobacter subterraneus and Thermodesulfobacterium commune.  相似文献

Human mesenchymal stem cells (hMSCs) hold great promise in cell therapy and regenerative medicine. Various preclinical and clinical trials have been carried out to illustrate the therapeutic potential of these cells. However, one major challenge for manufacturing clinical grade hMSCs is the requisition of current good manufacturing practice (cGMP) grade practices in cell isolation, processing, storage, and distribution. Development of non-toxic and animal serum-free preservation medium is critical for storage and distribution of mesenchymal stem cells (MSCs). In this study, we developed a solution formula that could preserve MSCs at 4°C for up to 3 weeks. In the solution, trehalose is a key ingredient for maintaining survival of MSCs. Among the concentrations investigated, 40 mM trehalose showed the best outcome with the viability maintained more than 92.7 ± 1.5% for 7 days. Cells preserved in the solution formula for 3 weeks still remained about 70% viability, and produced results similar to those of freshly harvested hMSCs in terms of growth kinetics, expression profile of cell surface antigens, and differentiation potential. In summary, storage of MSCs in the medium makes it far easier for transporting the cells from processing units to clinical sites.  相似文献

The reaction rate and selectivity of the enzymatic kinetic resolution of ibuprofen and 1-phenylethanol with supercritical CO2 as solvent were studied in a batch reactor from 40 °C to 160 °C. The commercial enzyme, Novozym 435, remained partly active for at least 14 h up to 140 °C at 15 MPa. The maximum reaction rate for the esterification of 1-phenylethanol and ibuprofen was at about 90 °C. The enantiomeric excess for 1-phenylethanol exceeds 99% and was temperature independent. Selectivity for ibuprofen esterification reached a lower enantiomeric excess of 61% caused by equilibrium adjustment. The results show that with supercritical CO2 as reaction medium enzymes remain active above 100 °C.  相似文献

Preexposure to mild temperatures such as 40°C induces thermotolerance, whereby cells resist subsequent exposure to a toxic insult. This study investigates the protective effect of mild thermotolerance (3h, 40°C) against activation of death receptor-mediated apoptosis by H(2)O(2) in HeLa cells. H(2)O(2) (5-50μM) caused rapid activation (1-3h) of the Fas death receptor pathway of apoptosis, which was evident by up-regulation of the death ligand FasL and recruitment of the adaptor protein Fas-associated death domain to the plasma membrane. This resulted in activation of caspase-8 and caspase-2, which led to activation of the cross-talk pathway involving Bid cleavage, t-Bid translocation to mitochondria, and caspase-9 activation. These changes were all diminished in thermotolerant cells. Mild thermotolerance also protected cells against cytotoxicity from H(2)O(2) as well as execution-phase events of apoptosis such as caspase-3 activation and chromatin condensation. The antioxidant polyethylene glycol-catalase abolished FasL induction and caspase-8 activation due to H(2)O(2). FasL up-regulation; activation of caspases-8, -2, -9, and -3; and chromatin condensation were decreased by the p53 inhibitor pifithrin-α, implicating p53 as an upstream factor in the activation of death receptor-mediated apoptosis by H(2)O(2). This study advances knowledge about the protective effect of adaptive responses induced by mild stresses, such as fever temperatures, against induction of apoptosis by oxidative stress.  相似文献

An amoeba strain was isolated from marine sediment taken from the beach near a fumarole in Italy. The trophozoites of this new marine species transforms into flagellates with variable numbers of flagella, from 2 to 10. The strain forms round to oval cysts. This thermophilic amoeboflagellate grows at temperatures up to 54 °C. Molecular phylogenetic analysis of the small subunit ribosomal DNA (SSU rDNA) places the amoeboflagellate in the Heterolobosea. The closest relatives are Stachyamoeba sp. ATCC50324, a strain isolated from an ocean sample, and Vrihiamoeba italica, a recent isolate from a rice field. Like some other heterolobosean species, this new isolate has a group I intron in the SSU rDNA. Because of the unique place in the molecular phylogenetic tree, and because there is no species found in the literature with similar morphological and physiological characteristics, this isolate is considered to be a new genus and a new species, Oramoeba fumarolia gen. nov., sp. nov.  相似文献
