Effects of commonly used purification procedures on the yield and specific activity of (Na+ + K+)-ATPase (Mg2+-dependent, Na+ + K+-activated ATP phosphohydrolase, EC 3.6.1.3), the turnover number of the enzyme, and the kinetic parameters for the ATP-dependent ouabain-enzyme interaction were compared in canine brain, heart and kidney. Kinetic parameters were estimated using a graphical analysis of non-steady state kinetics. The protein recovery and the degree of increase in specific activity of (Na+ + K+)-ATPase and the ratio between (Na+ + K+)-ATPase and Mg2+-ATPase activities during the successive treatments with deoxycholate, sodium iodide and glycerol were dependent on the source of the enzyme. A method which yields highly active (Na+ + K+)-ATPase preparations from the cardiac tissue was not suitable for obtaining highly active enzyme preparations from other tissues. Apparent turnover numbers of the brain (Na+ + K+)-ATPase preparations were not significantly affected by the sodium iodide treatment, but markedly decreased by deoxycholate or glycerol treatments. Similar glycerol treatment, however, failed to affect the apparent turnover number of cardiac enzyme preparations. Cerebral and cardiac enzyme preparations obtained by deoxycholate, sodium iodide and glycerol treatments had lower affinity for ouabain than renal enzyme preparations, primarily due to higher dissociation rate constants for the ouabain enzyme complex. This tissue-dependent difference in ouabain sensitivity seems to be an artifact of the purification procedure, since less purified cerebral or cardiac preparations had lower dissociation rate constants. Changes in apparent association rate constants were minimal during the purification procedure. These results indicate that the presently used purification procedures may alter.  相似文献

The rates of association of [3H]ouabain to Na+,K+-ATPase and the rates of dissociation of the enzyme-ouabain complexes were determined for enzymes isolated from dog skeletal muscle, beef heart muscle, and lamb kidney medulla. The rates of association were strongly influenced by the presence of ligands such as magnesium, sodium, potassium, ATP, and inorganic phosphate. For a particular set of binding ligands, the rates of association did not vary much amongst the three enzymes studied, although enzyme from skeletal muscle was the fastest. In contrast, the rates of dissociation were relatively independent of the ligand conditions. The rates of dissociation also varied greatly amongst the enzyme sources, with skeletal muscle Na+,K+-ATPase being the fastest. Although the major determinant of the affinity of the Na+,K+-ATPase for ouabain is the rate of dissociation, the rate of association also plays a role. Since the binding of ouabain to the Na+,K+-ATPase in the presence of magnesium, ATP, sodium, and potassium is very slow, it is difficult to obtain an I50 (equilibrium) value for the inhibition of hydrolytic activity by ouabain. If measurements of activity are made after a long period of time (3 h), the affinity of the enzyme for ouabain, estimated from inhibition of Na+,K+-ATPase activity, approached the value calculated from [3H]ouabain binding. The ratio of the I50 value for ouabagenin to that for ouabain for the skeletal muscle enzyme was the same as that for cardiac muscle enzyme, indicating that the sugar moiety of ouabain was interacting with the receptor of both enzymes. It is apparent, therefore, that the absence of a sugar binding site in skeletal Na+,K+-ATPase is not the reason for the faster dissociation rate of this enzyme.  相似文献

Summary Antibodies which were raised against highly purified membrane-bound (Na+–K+)-ATPase from the outer medulla of rat kidneys inhibit the (Na+–K+)-ATPase activity up to 95%. The antibody inhibition is reversible. The time course of enzyme inhibition and reactivation is biphasic in semilogarithmic plots.In the purified membrane-bound (Na+–K+)-ATPase negative cooperativity was observed (a) for the ATP dependence of the (Na+–K+)-ATPase activity (n=0.86), (b) for the ATP binding to the enzyme (n=0.58), and (c) for the ouabain inhibition of the (Na+–K+)-ATPase activity (n=0.77). By measuring the Na+ dependence of the (Na+–K+-ATPase reaction, a positive homotropic cooperativity (n=1.67) was found.As reactivation of the antibody-inhibited enzyme proceeds very slowly (t 0.5=5.2hr), it was possible to measure characteristics of the antibody-(Na+–K+)-ATPase complex: The antibodies exerted similar effects on the ATP dependence of the (Na+–K+)-ATPase reaction and on the ATP binding of the enzyme.V max of the (Na+–K+)-ATPase reaction and the number of ATP binding sites were reduced whileK 0.5 ATP for the (Na+–K+)-ATPase activity and for the ATP binding were increased by the antibodies. The Hill coefficients for the ATP binding and for the ATP dependence of the enzyme activity were not significantly altered by the antibodies. The antibodies increased theK 0.5 value for the Na+ stimulation of the (Na+–K+)-ATPase activity, but they did not alter the homotropic interactions between the Na+-binding sites. The negative cooperativity which was observed for the ouabain inhibition of the (Na+–K+)-ATPase activity was abolished by the antibodies.The data are tentatively explained by the following model: The antibodies bind to the (Na+–K+)-ATPase from the inner membrane side, reduce the ATP binding symmetrically at the ATP binding sites and reduce thereby also the (Na+–K+)-ATPase activity of the enzyme. The antibodies may inhibit the ATP binding by a direct interaction or by means of a conformational change at the ATP binding sites. This may possibly also lead to the alteration of the Na+ dependence of the (Na+–K+)-ATPase activity and to the observed alteration of the dose response to the ouabain inhibition.  相似文献

Ouabain activation of the phosphatase associated with Na+,K+-ATPase is a time-dependent process which is stimulated by ATP and other nucleotides. Further stimulation by Na+ is observed under certain conditions. The stimulatory effect of ATP was found to be due to an increase in the affinity of the enzyme for ouabain. The time required for maximal ouabain activation to be achieved was decreased by ATP and further decreased by ATP + Na+.These conditions for maximal activation by ouabain are similar to those required for maximal ouabain binding and suggest that the same ouabain site is responsible for activation of Mg2+-dependent phosphatase and for inhibition of Na+,K+-ATPase and K+-phosphatase.  相似文献

It is not known whether ouabain injected into the kidney in vivo is bound exclusively to the (Na+ + K+)-ATPase and whether the reduction of sodium pumping capacity is large enough to account for the reduction in sodium reabsorption. In the present study on dogs the total amount of parenchymal ouabain was therefore estimated and the specific renal binding compared to the reduction in (Na+ + K+)-ATPase activity. Ouabain, 120 nmol/kg body weight, was injected into the renal artery in vivo reducing the (Na+ + K+)-ATPase activity by 3lmost 80%. After nephrectomy, tissue ouabain could be quantified by radioimmunoassay after heating the homogenate to 70°C for 30 min; negligible amounts were detectable without heating. No correlation between ouabain binding and tissue volume, protein content, DNA content or Mg2+-ATPase content could be found when comparing the following four fractions of the kidney: outer cortex, inner cortex, outer medulla and papilla. For the whole kidney, mean parenchymal tissue concentration of ouabain equalled 0.58 ± 0.03 μmol/100 g wet tissue. Only 21.3 ± 1.2% of the ouabain was confined to the outer medulla corresponding to 54 ± 4 nmol giving a tissue concentration of 1.08 ± 0.05 μmol/100 g wet tissue. The renal ouabain concentrations were highly correlated to the reduction in (Na+ + K+)-ATPase activity, giving a ratio between the reduction in hydrolysis rate and bound ouabain (turnover number) of 6105 min?1 which is close to the value of 7180 min?1 found by in vitro Scatchard analysis. No ouabain seems to be bound to other tissue components than the (Na+ + K+)-ATPase and the present method is therefore a simple way of measuring the number of inhibited (Na+ + K+)-ATPase molecules after in vivo injection of ouabain.  相似文献

A rat heart plasma membrane preparation isolated in a sucrose medium and some of its enzymatic properties have been investigated.It has been shown that a rat heart plasma membrane fraction contains high creatine phosphokinase activity which can not be diminished by repeated washing with sucrose solution. Creatine phosphokinase extracted from a plasma membrane fraction with potassium chloride and 0.01% deoxycholate solution is electrophoretically identical to MM isoenzyme of creatine phosphokinase. Under the conditions where (Na+,K+)-ATPase is activated by addition of Na+,K+ and MgATP, creatine phosphokinase of plasma membrane fraction is able to maintain a low ADP concentration in the medium if creatine phosphate is present. The rate of creatine release is dependent upon MgATP concentration in accordance with the kinetic parameters of the (Na+,K+)-ATPase and is significantly inhibited by ouabain (0.5 mM). The rate of creatine release is also dependent on creatine phosphate concentration in conformance with the kinetic parameters of MM isozyme of creatine phosphokinase,It is concluded that in intact heart cells the plasma membrane creatine phosphokinase may ensure effective utilization of creatine phosphate for immediate rephosphorylation of ADP produced in the (Na+,K+)-ATPase reaction.  相似文献

The involvement of membrane (Na+ + K+)-ATPase (Mg2+-dependent, (Na+ + K+)-activated ATP phosphohydrolase, E.C. 3.6.1.3) in the oxygen consumption of rat brain cortical slices was studied in order to determine whether (Na+ + K+)-ATPase activity in intact cells can be estimated from oxygen consumption. The stimulation of brain slice respiration with K+ required the simultaneous presence of Na+. Ouabain, a specific inhibitor of (Na+ + K+)-ATPase, significantly inhibited the (Na+ + K+)-stimulation of respiration. These observations suggest that the (Na+ + K+)-stimulation of brain slice respiration is related to ADP production as a result of (Na+ + K+)-ATPase activity. However, ouabain also inhibited non-K+-stimulated respiration. Additionally, ouabain markedly reduced the stimulation of respiration by 2,4-dinitrophenol in a high (Na+ + K+)-medium. Thus, ouabain depresses brain slice respiration by reducing the availability of ADP through (Na+ + K+)-ATPase inhibition and acts additionally by increasing the intracellular Na+ concentration. These studies indicate that the use of ouabain results in an over-estimation of the respiration related to (Na+ + K+)-ATPase activity. This fraction of the respiration can be estimated more precisely from the difference between slice respiration in high Na+ and K+ media and that in choline, K+ media. Studies were performed with two (Na+ + K+)-ATPase inhibitors to determine whether administration of these agents to intact rats would produce changes in brain respiration and (Na+ + K+)-ATPase activity. The intraperitoneal injection of digitoxin in rats caused an inhibition of brain (Na+ + K+)-ATPase and related respiration, but chlorpromazine failed to alter either (Na+ + K+)-ATPase activity or related respiration.  相似文献

(Na++K+)-ATPase is a target receptor of digitalis (cardiac glycoside) drugs. It has been demonstrated that the H1-H2 domain of the α-subunit of the (Na++K+)-ATPase is one of the digitalis drug interaction sites of the enzyme. Despite the extensive studies of the inhibitory effect of digitalis on the (Na++K+)-ATPase, the functional property of the H1-H2 domain of the enzyme and its role in regulating enzyme activity is not completely understood. Here we report a surprise finding: instead of inhibiting the enzyme, binding of a specific monoclonal antibody SSA78 to the H1-H2 domain of the (Na++K+)-ATPase elevates the catalytic activity of the enzyme. In the presence of low concentration of ouabain, monoclonal antibody SSA78 significantly protects enzyme function against ouabain-induced inhibition. However, higher concentration of ouabain completely inactivates the (Na++K+)-ATPase even in the presence of SSA78. These results suggest that the H1-H2 domain of the (Na++K+)-ATPase is capable of regulating enzyme function in two distinct ways for both ouabain-sensitive and -resistant forms of the enzyme: it increases the activity of the (Na++K+)-ATPase during its interaction with an activator; it also participates in the mechanism of digitalis or ouabain-induced inhibition of the enzyme. Understanding the dual activity of the H1-H2 domain will help better understand the structure-function relationships of the (Na++K+)-ATPase and the biological processes mediated by the enzyme.  相似文献

Effects of two triterpene glycosides, isolated from the holothurian Psolus fabricii, on rat brain Na+,K+-ATPase (Na,K-pump; EC 3.6.1.3) were investigated. Psolusosides A and B (PsA and PsB) inhibited rat brain Na+,K+-ATPase with I50 values of 1×10−4 M and 3×10−4 M, respectively. PsA significantly stimulated [3H]ATP binding to Na+,K+-ATPase, weakly increased [3H]ouabain binding to the enzyme, and inhibited K+-phosphatase activity to a smaller degree than the total reaction of ATP hydrolysis. In contrast, PsB decreased [3H]ATP binding to Na+,K+-ATPase, and had no effect on [3H]ouabain binding to the enzyme. K+-Phosphatase activity was inhibited by PsB in parallel with Na+,K+-ATPase activity. The fluorescence intensity of tryptophanyl residues of Na+,K+-ATPase was increased by PsA and decreased by PsB in a dose-dependent manner. The excimer formation of pyrene, a hydrophobic fluorescent probe, was decreased by PsA only. The different characteristics of inhibition mode for these substances were explained by peculiarities of their chemical structures and distinctive affinity to membrane cholesterol.  相似文献

IN an earlier paper1 we have presented a model for a sodium pump based on the operation of the adenosine triphosphatase component of membranes which is sensitive to ouabain and is activated by sodium and potassium; that is (Na++K+)-ATPase. We attempted to correlate the biochemical properties of this enzyme system as they were then known with the essential properties of Na+ transport systems. The model suggested further experiments which could clarify the role of (Na+ + K +)-ATPase in ion transport and some experimental evidence is now available for the stoichiometry of ouabain binding to isolated enzyme preparations2,3 although differences in the experimental techniques which have been used make the data equivocal.  相似文献

The effect of various mitogens was studied on sodium (Na+) potassium (K+) transport, 3H-ouabain binding, and adenosine triphosphatase (ATPase) activity in human and sheep peripheral lymphocytes. Concanavalin A (ConA), phytohemagglutinin (PHA), horse anti-lymphocyte serum (ALS), and anti-IgG antisera, in order of decreasing potency, stimulated in particular the ouabain-sensitive K+ pump influx, while the cardiac glycoside-insensitive K+ leak flux was only slightly affected. Sheep lymphocytes primed in vivo with human IgG as antigen also responded with K+ pump flux activation when exposed to the antigen in vitro. Both PHA and ConA also stimulated active Na+ efflux in human lymphocytes. Apparently these mitogens activate the Na+K+ pump system in the lymphocyte membrane—an assumption supported by the finding of a significant activation of the ouabain-sensitive Na+K+-ATPase. From rate studies of 3H-ouabain binding carried out at 37 °C in presence and absence of sodium azide, and at 0 °C, it is concluded that PHA alters the rate of ouabain uptake to these cells. Thus PHA may alter the affinity of the pump for ouabain, equivalent to an increased cation turnover per pump site. However, our findings do not completely discount the possibility that PHA also increases the total number of ouabain molecules bound and therefore of Na+K+ pumps.  相似文献

Liposomes containing either purified or microsomal (Na+,K+)-ATPase preparations from lamb kidney medulla catalyzed ATP-dependent transport of Na+ and K+ with a ratio of approximately 3Na+ to 2K+, which was inhibited by ouabain. Similar results were obtained with liposomes containing a partially purified (Na+,K+-ATPase from cardiac muscle. This contrasts with an earlier report by Goldin and Tong (J. Biol. Chem. 249, 5907–5915, 1974), in which liposomes containing purified dog kidney (Na+,K+)-ATPase did not transport K+ but catalyzed ATP-dependent symport of Na+ and Cl?. When purified by our procedure, dog kidney (Na+,K+)-ATPase showed some ability to transport K+ but the ratio of Na+ : K+ was 5 : 1.  相似文献

3H-ouabain binding and ouabain-inhibitable 86Rb+ (K+) uptake were investigated as a means to identify a third isoform of Na+, K+-ATPase in crude synaptosome preparations. The specific binding of low concentrations (10 nM and 1 uM) of 3H-ouabain, in crude synaptosome preparations, was markedly inhibited by K+ (0.5–5 mM). Accordingly, 86Rb+ (K+) uptake, in the presence of 5 mM K+ was not sensitive to inhibition by low concentrations (10−11–10−7 M) of ouabain. Higher concentrations (10−6–10−2.6 M) of ouabain resulted in a biphasic inhibition of K+ uptake, which distinguished the activities of the presumed alpha 2 and alpha 1 isozymes of Na+, K+-ATPase. Reduction of K+ (1.25 mM and 0.5 mM) in the incubation, resulted in the observation of a third component of ouabain- sensitive K+ uptake. This Na+, K+-ATPase activity, which was defined, pharmacologically, as very sensitive (VS) to ouabain, exhibited IC50s of 3.6 nM and 92 nM at 1.25 mM K+ and 0.5 mM K+, respectively. Inhibition of ouabain binding and VS-dependent K+ uptake, at a high, physiological cocentration (5 mM) of K+, suggests that VS may be an inactive isoform of brain Na+, K+-ATPase under resting conditions.  相似文献

Liver plasma membranes enriched in bile canaliculi were isolated from rat liver by a modification of the technique of Song et al. (J. Cell Biol. (1969) 41, 124–132) in order to study the possible role of ATPase in bile secretion. Optimum conditions for assaying (Na+ + K+)-activated ATPase in this membrane fraction were defined using male rats averaging 220 g in weight. (Na+ + K+)-activated ATPase activity was documented by demonstrating specific cation requirements for Na+ and K+, while the divalent cation, Ca2+, and the cardiac glycosides, ouabain and scillaren, were inhibitory. (Na+ + K+)-activated ATPase activity averaged 10.07 ± 2.80 μmol Pi/mg protei per h compared to 50.03 ± 11.41 for Mg2+-activated ATPase and 58.66 ± 10.07 for 5′-nucleotidase. Concentrations of ouabain and scillaren which previously inhibited canalicular bile secretion in the isolated perfused rat liver produced complete inhibition of (Na+ + K+)-activated ATPase without any effect on Mg2+-activated ATPase. Both (Na+ + K+)-activated ATPase and Mg2+-activated ATPase demonstrated temperature dependence but differed in temperature optima. Temperature induced changes in specific activity of (Na+ + K+)-activated ATPase directly paralleled previously demonstrated temperature optima for bile secretion. These studies indicate that (Na+ + K+)-activated ATPase is present in fractions of rat liver plasma membranes that are highly enriched in bile canaliculi and provide a model for further study of the effects of various physiological and chemical modifiers of bile secretion and cholestasis.  相似文献

The effects of chronic reserpine pretreatment (0.1 mg/kg/day, 7–9 days) on myocardial sodium pump activity, the binding of 3H-ouabain to Na+, K+-ATPase, and the positive inotropic effect of ouabain were studied in guinea pig hearts. Ouabain-sensitive 86Rb uptake, an estimate of sodium pump activity, was significantly decreased (33.0%) in papillary muscles of chronic reserpine-pretreated guinea pigs as compared to the saline-treated controls. Kinetic analyses of the interaction of 3H-ouabain with Na+, K+-ATPase indicated that chronic reserpine pretreatment resulted in a significant decrease (24.3%) in the maximum 3H-ouabain binding site concentration when the results were expressed as pmoles per mg protein. The maximum 3H-ouabain binding sites or the number of Na+, K+-ATPase units, however, were not significantly different between the two groups when they were expressed as pmoles per mg DNA. The affinity or the dissociation rate constant (Kd) of 3H-ouabain binding was not altered after chronic reserpine pretreatment. In isolated, electrically-driven left atrial preparations, the basal contractile force was slightly higher in the reserpine-pretreated animals; the subsequent development of the positive inotropic effect and the concentration of ouabain needed to produce half-maximal inotropic response, however, were not different from the controls. Thus, it is concluded that chronic reserpine pretreatment is accompanied by a significant reduction in myocardial sodium pump activity; however, the number of sodium pump sites per cell was unchanged. The sensitivity of the reserpine-pretreated myocardium to the inotropic action of ouabain as well as its affinity for 3H-ouabain binding in vitro are also unchanged.  相似文献

Trypsin treatment of the purified Na+, K+-ATPase from canine renal outer medulla causes loss of ADP-ATP exchange activity when digestion takes place in 0.1 M KCl. Activity surviving this treatment remains inhibitable by ouabain. Addition of ATP to such digestion mixtures stabilizes the Na+, K+-ATPase in a different conformation (Na+-form). Under these conditions ADP-ATP exchange activity is protected, and becomes ouabain-insensitive. Quantitative analysis of the cleavage products and rates of loss of ouabain binding and exchange activity suggest that catalytically inactive trypsinolysis products can bind ouabain, and that the 85,000 dalton fragment associated with ouabain-insensitive ADP-ATP exchange activity cannot bind ouabain. Cleavage to produce the 85,000 dalton fragment therefore destroys the ouabain binding site.  相似文献

It was found that ouabain and marinobufagenin, specific inhibitors of Na+,K+-ATPase, increased the contraction of the isolated rat diaphragm by ~15% (positive inotropic effect) at EC50 = 1.2 ± 0.3 nM and 0.3 ± 0.1 nM, respectively, which was indicative of the participation of the ouabain-sensitive Na+,K+-ATPase α2 isoform. Analysis of the dose-response curves for the effect of ouabain on the resting membrane potential of muscle fibers in the absence and in the presence of 100 nM acetylcholine (hyperpolarizing the membrane) showed the presence of two pools of Na+,K+-ATPase α2 that differed in affinity for ouabain. Only the high-affinity pool (IC50 ~ 9 nM) mediates the hyperpolarizing effect of nanomolar concentrations of acetylcholine. Most likely, it is this pool of that is involved in the positive inotropic effect of ouabain, which can be a mechanism of regulation of the muscle function by circulating endogenous inhibitors of Na+,K+-ATPase.  相似文献

De novo mutations in ATP1A3, the gene encoding the α3-subunit of Na+,K+-ATPase, are associated with the neurodevelopmental disorder Alternating Hemiplegia of Childhood (AHC). The aim of this study was to determine the functional consequences of six ATP1A3 mutations (S137Y, D220N, I274N, D801N, E815K, and G947R) associated with AHC. Wild type and mutant Na+,K+-ATPases were expressed in Sf9 insect cells using the baculovirus expression system. Ouabain binding, ATPase activity, and phosphorylation were absent in mutants I274N, E815K and G947R. Mutants S137Y and D801N were able to bind ouabain, although these mutants lacked ATPase activity, phosphorylation, and the K+/ouabain antagonism indicative of modifications in the cation binding site. Mutant D220N showed similar ouabain binding, ATPase activity, and phosphorylation to wild type Na+,K+-ATPase. Functional impairment of Na+,K+-ATPase in mutants S137Y, I274N, D801N, E815K, and G947R might explain why patients having these mutations suffer from AHC. Moreover, mutant D801N is able to bind ouabain, whereas mutant E815K shows a complete loss of function, possibly explaining the different phenotypes for these mutations.  相似文献

[3H]-Ouabain binding to muscle preparations was utilized to estimate the number of Na+,K+-ATPase enzyme units in hindlimbs from 8 week old lean and obese mice. Specific [3H]-ouabain binding per mg particulate protein was 36% lower in obese mice; whereas, the affinity of the binding sites for ouabain was similar in obese and lean mice. Since obese mice had less muscle than lean mice, the number of Na+,K+-ATPase enzyme units in hindlimbs from obese mice was less than half the number observed in lean mice.  相似文献

Cardiac glycosides are inhibitors of Na+,K+-ATPase, and K+-phosphatase activities of the transport enzyme. Previous studies have shown that when the sensitivities of these two activities to ouabain are compared by the addition of varying concentrations of the drug to the assay media, the K+-phosphatase is significantly less sensitive than Na+,K+-ATPase. This work was done to seek an explanation for this phenomenon. 3-O-Methyl-fluorescein phosphate was used as substrate for the continuous fluorimetric assay of K+-phosphatase obtained from human red cells. When ouabain was added to the assay medium, a time-dependent inhibition of K+-phosphatase was observed. The rate of inhibition was also influenced by the order of additions of K+ and ouabain. In view of these results, several enzyme samples exposed to ouabain for varying lengths of time were prepared, and their Na+,K+-ATPase and K+-phosphatase activities were then determined. A good correlation between the extent of inhibition of the two activities was obtained. These results prove that the previously observed discrepancies between the sensitivities of Na+,K+-ATPase and K+-phosphatase to ouabain are due to the different kinetics of drug interaction with the enzyme under the different conditions of the two assays and that once a certain level of ouabain binding to the enzyme is achieved, both activities are equally inhibited.  相似文献
