Erythritol is a natural sweetener commonly used in the food and pharmaceutical industries. Produced by microorganisms as an osmoprotectant, it is an ideal sucrose substitute for diabetics or overweight persons due to its almost zero calorie content. Currently, erythritol is produced on an industrial scale through the fermentation of sugars by some yeasts, such as Moniliella sp. However, the popularity of erythritol as a sweetener is still small because of its high retail price. This creates an opportunity for further process improvement. Recent years have brought the rapid development of erythritol biosynthesis methods from the low-cost substrates, and a better understanding of the metabolic pathways leading to erythritol synthesis. The yeast Yarrowia lipolytica emerges as an organism effectively producing erythritol from pure or crude glycerol. Moreover, novel erythritol producing organisms and substrates may be taken into considerations due to metabolic engineering. This review focuses on the modification of erythritol production to use low-cost substrates and metabolic engineering of the microorganisms in order to improve yield and productivity.  相似文献

To improve the erythritol productivity ofPenicillium sp. KJ81, mutants were obtained using UV irradiation and NTG treatment. Among these mutants,Penicillium sp. KJ-UV29 revealed no morphological changes, yet was superior to the wild strain in the following three points: (1)Penicillium sp. KJ-UV29 produced more erythritol than the wild strain under the same conditions, (2) no foam was produced during cultivation, unlike the wild strain, and (3) the mutant produced a significantly lower amount of glycerol.Penicillium sp KJ-UV29 produced as much as 15.1 g/L of erythritol, whereas the wild-typePenicillium sp. KJ-UV29 produced as much as 15.1 g/L of erythritol, whereas the wild-typePenicillium sp. KJ81 only produced 11.7 g/L.Penicillium sp. KJ-UV29 only generated 6.1 g/L of glycerol, compared to 19.4 g/L produced by the wild strain. When investigating the optimal culture conditions for erythritol production by the mutant strainPenicillium sp. KJ-UV29, sucrose was idetified as the most effective carbon source, and the mutant was even able to produce erythritol in a 70% sucrose-containing medium, although a 30% sucrose medium exhibited the highest productivity. The production of erythritol byPenicillium sp. KJ-UV29 was also significantly increased by the addition of ammonium carbonate, potassium nitrate, and sodium nitrate. Accordingly, under optimal conditions,Penicillium sp. KJ-UV29 produced 45.2 g/L of erythritol in a medium containing 30% sucrose, 0.5% yeast extract, 0.5% (NH4)2C2O4 0.1% NaNO3, and 0.01% FeSO4 with 1 vvm aeration and 200 rpm agitation at 37°C for 7 days in a 5-L jar fermentor.  相似文献

The penetration of 14C-labeled erythritol, mannitol, and sucrose through the axolemma was determined in medium sized paired axons, one at rest and the other stimulated 25 times per sec. The resting permeabilities, in 10-7 cm/sec, are erythritol, 2.9 ± 0.3 (mean ± SEM); mannitol, 2.3 ± 0.4; and sucrose 0.9 ± 0.1. In the stimulated axons they are: erythritol, 5.2 ± 0.3; mannitol, 4.0 ± 0.5; and sucrose, 1.8 ± 0.3. Thus, the calculated permeabilities during activity (1 msec per impulse), in the same units, are: 100, 75, and 38, respectively. These changes in permeability are reversible. The effects of external potassium and sodium concentrations on erythritol penetration were also studied. At rest, erythritol penetration is independent of potassium and sodium concentrations. In the stimulated axons, erythritol penetration decreases when the extracellular sodium is diminished. Sodium influx (not the efflux) decreases during rest and activity when the extracellular sodium is diminished. The diminution during activity of erythritol and sodium entries in low sodium solutions may be related to a decrease of a drag effect of sodium ions on the nonelectrolyte molecules or to independent effects of the sodium concentration on sodium influx and the nonelectrolyte pathways. The axolemma discriminates among erythritol, mannitol, sucrose, and the different ionic species during rest and activity.  相似文献

A total of 1752 strains of osmophilic yeasts were isolated from honey and pollens. Forty-three strains of osmophilic yeasts produced polyols, among which 6 strains produced erythritol in good yields. On the other hand, 52 osmophilic yeasts converted sucrose to fructooligosaccharides, among which 8 strains produced both extra and intracellular β-fructofuranosidase, which converted sucrose to fructooligosaccharides. This investigation concluded that osmophilic yeasts converted sucrose not only to polyols, but also to fructooligosaccharides in good yields.  相似文献

A yeastlike fungus, probably belonging to the genus Torula, was isolated from fresh pollen and was shown to produce erythritol in yields of 35 to 40% of the sugar utilized. The ability to produce erythritol is an inherent characteristic of the isolate, but unfavorable fermentation conditions can lead to the production of glycerol at the expense of erythritol. By the use of a synthetic medium, it was shown that the concentrations of both nitrogen and phosphorous in the medium must be closely controlled to obtain satisfactory erythritol yields.  相似文献

Summary Immobilized cells of Aspergillus niger needed a lower initial sucrose concentration than free cells in order to obtain maximal yields of citric acid production. High sucrose concentrations led to reduced yields and increased polyol formation (glycerol, erythritol, arabitol). Continuous fermentation with media containing low sugar concentrations prevented the formation of polyols. The change from nitrogen-limited to phosphate-limited precultivation of immobilized spores significantly increased the productivity of the mycelium. The ratio of citric acid to residual sugar in the effluent distinctly lay in the direction of citric acid. Inside the alginate beads mainly large bulbous cells were observed.  相似文献

Serosal-to-mucosal and mucosal-to-serosal diffusion of 14C-labelled inulin, sucrose, erythritol and propionamide was compared with 3HHO diffusion in mucosae incubated with isosmotic solutions at both surfaces, as well as isosmotic solution at serosal surface and hyperosmotic solution at the mucosal surface. The use of a hyperosmotic solution at the mucosal surface significantly increases unidirectional diffusion fluxes of inulin and of sucrose. To a nonsignificant extent, it affects the fluxes of erythritol and propionamide and significantly reduces the 3HHO diffusion. A size increment of the diffusion path utilized by the larger molecules is proposed.  相似文献

Previous feeding studies showed the polyalcohol erythritol was toxic when ingested by adult laboratory fruit flies (Drosophila melanogaster). We asked whether erythritol could additionally affect fly population growth either through larval toxicity or through effects on adult reproduction. Females did not avoid laying on food substrates with 1M erythritol; laying rate on 1M erythritol food was similar to control food when females were given free‐choice access. Eggs laid or placed on 0.5 M to 2.5 M erythritol foods hatched at normal rates, suggesting erythritol was not toxic to eggs upon contact. Drosophila melanogaster larvae readily consumed food containing 1 M erythritol, but none of these larvae reached pupation. Longevity of larvae feeding on in 1 M erythritol food was significantly reduced relative to controls, and mean ± SE larval lifespan on erythritol was 1.54 ± 0.10 days (max. = 3 days). Exposing cohorts of second‐instar larvae to food with varying concentrations of erythritol showed the LD50 (at 24 hr) concentration was approximately 0.6 M. Taken together, these results suggest erythritol could be employed in effective larval‐sink baits. Adults flies fed with erythritol produced significantly fewer eggs on days when they fed on 1 M erythritol, and egg production was significantly reduced for one additional day after the adults were moved to control food. These findings suggest erythritol is rapid and effective at temporarily suppressing D. melanogaster reproduction, increasing its potential for use in effective insect population control.  相似文献

Objective

Erythritol (1,2,3,4-butanetetrol) is a 4-carbon sugar alcohol that occurs in nature as a metabolite or storage compound. In this study, a multiple gene integration strategy was employed to enhance erythritol production in Y. lipolytica.

Results

The effects on the production of erythritol in Y. lipolytica of seven key genes involved in the erythritol synthesis pathway were evaluated individually, among which transketolase (TKL1) and transaldolase (TAL1) showed important roles in enhancing erythritol production. The combined overexpression of four genes (GUT1, TPI1, TKL1, TAL1) and disruption of the EYD1 gene (encoding erythritol dehydrogenase), resulted in produce approximately 40 g/L erythritol production from glycerol. Further enhanced erythritol synthesis was obtained by overexpressing the RKI1 gene (encoding ribose 5-phosphate isomerase) and the AMPD gene (encoding AMP deaminase), indicating for the first time that these two genes are also related to the enhancement of erythritol production in Y. lipolytica.

Conclusions

A combined gene overexpression strategy was developed to efficiently improve the production of erythritol in Y. lipolytica, suggesting a great capacity and promising potential of this non-conventional yeast in converting glycerol into erythritol.

Gastrodia elata, a fully mycoheterotrophic orchid without photosynthetic ability, only grows symbiotically with the fungus Armillaria. The mechanism of carbon distribution in this mycoheterotrophy is unknown. We detected high sucrose concentrations in all stages of Gastrodia tubers, suggesting sucrose may be the major sugar transported between fungus and orchid. Thick symplasm‐isolated wall interfaces in colonized and adjacent large cells implied involvement of sucrose importers. Two sucrose transporter (SUT)‐like genes, GeSUT4 and GeSUT3, were identified that were highly expressed in young Armillaria‐colonized tubers. Yeast complementation and isotope tracer experiments confirmed that GeSUT4 functioned as a high‐affinity sucrose‐specific proton‐dependent importer. Plasma‐membrane/tonoplast localization of GeSUT4‐GFP fusions and high RNA expression of GeSUT4 in symbiotic and large cells indicated that GeSUT4 likely functions in active sucrose transport for intercellular allocation and intracellular homeostasis. Transgenic Arabidopsis overexpressing GeSUT4 had larger leaves but were sensitive to excess sucrose and roots were colonized with fewer mutualistic Bacillus, supporting the role of GeSUT4 in regulating sugar allocation. This is not only the first documented carbon import system in a mycoheterotrophic interaction but also highlights the evolutionary importance of sucrose transporters for regulation of carbon flow in all types of plant‐microbe interactions.  相似文献

For the first time, the effects of varying osmotic and matric potential on fungal radial growth and accumulation of polyols were studied in three isolates of Pochonia chlamydosporia. Fungal radial growth was measured on potato dextrose agar modified osmotically using potassium chloride or glycerol. PEG 8000 was used to modify matric potential. When plotted, the radii of the colonies were found to grow linearly with time, and regression was applied to estimate the radial growth rate (mm day?1). Samples of fresh mycelia from 25-day-old cultures were collected and the quantity (mg g?1 fresh biomass) of four polyols (glycerol, erythritol, arabitol and mannitol) and one sugar (glucose) was determined using HPLC. Results revealed that fungal radial growth rates decreased with increased osmotic or matric stress. Statistically significant differences in radial growth were found between isolates in response to matric stress (P<0.006) but not in response to osmotic stress (P=0.759). Similarly, differences in the total amounts of polyols accumulated by the fungus were found between isolates in response to matric stress (P<0.001), but not in response to osmotic stress (P=0.952). Under water stress, the fungus accumulated a combination of different polyols important in osmoregulation, which depended on the solute used to generate the stress. Arabitol and glycerol were the main polyols accumulated in osmotically modified media, whereas erythritol was the main polyol that was accumulated in media amended with PEG. The results found that Pochonia chlamydosporia may use different osmoregulation mechanisms to overcome osmotic and matric stresses.  相似文献

Erythritol is produced in yeasts via the reduction of erythrose into erythritol by erythrose reductases (ERs). However, the genes codifying for the ERs involved in this reaction have not been described in any Saccharomyces species yet. In our laboratory, we recently showed that, during alcoholic fermentation, erythritol is differentially produced by Saccharomyces cerevisiae and S. uvarum species, the latter being the largest producer. In this study, by using BLAST analysis and phylogenetic approaches the genes GRE3, GCY1, YPR1, ARA1 and YJR096W were identified as putative ERs in Saccharomyces cerevisiae Then, these genes were knocked out in our S. uvarum strain (BMV58) with higher erythritol biosynthesis compared to control S. cerevisiae wine strain, to evaluate their impact on erythritol synthesis and global metabolism. Among the mutants, the single deletion of GRE3 markedly impacts erythritol production, although ΔYPR1ΔGCY1ΔGRE3 was the combination that most decreased erythritol synthesis. Consistent with the increased production of fermentative by-products involved in redox balance in the Saccharomyces uvarum strain BMV58, erythritol synthesis increases at higher sugar concentrations, hinting it might be a response to osmotic stress. However, the expression of GRE3 in the S. uvarum strain was found to peak just before the start of the stationary phase, being consistent with the observation that erythritol increases at the start of the stationary phase, when there is low sugar in the medium and nitrogen sources are depleted. This suggests that GRE3 plays its primary function to help the yeast cells to maintain the redox balance during the last phases of fermentation.  相似文献

Erythritol, a four-carbon polyol, is a biological sweetener with applications in food and pharmaceutical industries. It is also used as a functional sugar substitute in special foods for people with diabetes and obesity because of its unique nutritional properties. Erythritol is produced by microbial methods using mostly osmophilic yeasts and has been produced commercially using mutant strains of Aureobasidium sp. and Pseudozyma tsukubaensis. Due to the high yield and productivity in the industrial scale of production, erythritol serves as an inexpensive starting material for the production of other sugars. This review focuses on the approaches for the efficient erythritol production, strategies used to enhance erythritol productivity in microbes, and the potential biotechnological applications of erythritol.  相似文献

The solvatochromic solvent features of water (dipolarity/polarizability, π*, hydrogen bond donor acidity, α, and hydrogen bond acceptor basicity, β) of water have been determined in aqueous solutions of erythritol, glucose, inositol, sarcosine, xylitol and urea with concentrations from 0 to ~3 M and higher. The concentration effects of the osmolytes on the solvent features of water were characterized and compared with those reported previously for sorbitol, sucrose, trimethylamine N-oxide (TMAO), and trehalose. The solvent features of water in solutions of all osmolytes except TMAO and sarcosine were established to be linearly interrelated. It is shown that the concentration effects of essentially all nonionic osmolytes depend on osmolytes’ lipophilicity, molecular polarizability, and polar surface area. It is demonstrated that solubility of various compounds in aqueous solutions of glucose, sucrose, sorbitol, and urea of varied concentrations may be described in terms of solvent dipolarity/polarizability of water in these solutions. Surface tension of aqueous solutions of sucrose and sorbitol may also be described in the same terms. The relative permittivity of aqueous solutions of glucose and sucrose may be described in terms of the solvent hydrogen bond donor acidity of water. It is suggested that the effects of nonionic osmolytes on behavior of proteins and nucleic acids in aqueous media may be considered in terms of the altered solvent features of water instead of “nano-molecular crowding” effect.  相似文献

In response to osmotic stress, the yeast Yarrowia lipolytica produces erythritol, a four‐carbon sugar alcohol, from erythrose‐P, an intermediate of the pentose phosphate pathway. Under non‐stressing conditions (isotonic environment), the produced erythritol is subsequently recycled into erythrose‐P that can feed the pentose phosphate pathway. Herein, gene YALI0F01584g was characterized as involved in the erythritol catabolic pathway. Several experimental evidences suggested that it encodes an erythrulose‐1P isomerase that converts erythrulose‐1P into erythrulose‐4P. On the basis of our previous reports and results gathered in this study with genetically modified strains, including ΔYALI0F01584g and ΔYALI0F01628g disrupted mutants, the entire erythritol catabolic pathway has been characterized.  相似文献

Background  

Erythrose reductase (ER) catalyzes the final step of erythritol production, which is reducing erythrose to erythritol using NAD(P)H as a cofactor. ER has gained interest because of its importance in the production of erythritol, which has extremely low digestibility and approved safety for diabetics. Although ERs were purified and characterized from microbial sources, the entire primary structure and the corresponding DNA for ER still remain unknown in most of erythritol-producing yeasts. Candida magnoliae JH110 isolated from honeycombs produces a significant amount of erythritol, suggesting the presence of erythrose metabolizing enzymes. Here we provide the genetic sequence and functional characteristics of a novel NADPH-dependent ER from C. magnoliae JH110.  相似文献

Botrytis cinerea is a pathogenic filamentous fungus, which infects more than 200 plant species. The enzymes secreted by B. cinerea play an important role in the successful colonization of a host plant. Some of the secreted enzymes are involved in the degradation of pectin, a major component of the plant cell wall. A total of 126 proteins secreted by B. cinerea were identified by growing the fungus on highly or partially esterified pectin, or on sucrose in liquid culture. Sixty‐seven common proteins were identified in each of the growth conditions, of which 50 proteins exhibited a SignalP motif. Thirteen B. cinerea proteins with functions related to pectin degradation were identified in both pectin growth conditions, while only four were identified in sucrose. Our results indicate it is unlikely that the activation of B. cinerea from the dormant state to active infection is solely dependent on changes in the degree of esterification of the pectin component of the plant cell wall. Further, these results suggest that future studies of the B. cinerea secretome in infections of ripe and unripe fruits will provide important information that will describe the mechanisms that the fungus employs to access nutrients and decompose tissues.  相似文献

An erythritol-producing osmophilic yeast-like fungus, Moniliella sp. 440, was isolated from honey and then successively mutated with iterative rounds of N-methyl-N′-nitro-N-nitrosoguanidine (NTG) treatment and selection. Six generations of mutants, named N12115-6, N21105-6, N31074-3, N42208-2, N53199-9, and N61188-12, were selected for and produced erythritol at 151.0, 157.2, 177.8, 191.4, 196.6, and 237.8 g/L, respectively, while the wild type strain produced 113.0 g/L erythritol in media containing 40% glucose and 1% yeast extract. The mutant cells were found to have a short rod-like shape, while the wild type cells have a long rod-like shape. The most efficient erythritol producer, N61188-12, assimilated myo-inositol and weakly assimilated erythritol. However, the wild type strain did not assimilate myo-inositol and assimilated erythritol well. In 250-L and 2000-L pilot-scale fermentors, the erythritol production by N61188-12 was 151.4 g/L and 152.4 g/L, respectively. A simple fed-batch culture of strain N61188-12 in a 2000-L fermentor increased erythritol production to 189.4 g/L after 10 days fermentation.  相似文献
