A guinea pig kidney membrane preparation was incubated with thimerosal and then thoroughly washed. Comparison of the properties of the native and the modified membranes showed that (a) Na++K+-dependent activity is substantially inhibited by thimerosal; (b) thimerosal does not diminish Na+-dependent ATPase activity; and (c) the thimerosal treated enzyme, like the native enzyme, is phosphorylated in the presence of Na+ and ATP, and dephosphorylated upon the addition of K+. It is suggested that thimerosal does not affect the binding of ATP to the high-affinity catalytic site, but that it blocks the binding of ATP to a low affinity modifying site the occupation of which is essential for the dissociation of the stable K+-dephosphoenzyme and the recycling of the enzyme.  相似文献

The addition of LiCl stimulated the (Na++K+)-dependent ATPase activity of a rat brain enzyme preparation. Stimulation was greatest in high Na+/low K+ media and at low Mg. ATP concentrations. Apparent affinities for Li+ were estimated at the α-sites (moderate-affinity sites for K+ demonstrable in terms of activation of the associated K+-dependent phosphatase reaction), at the β-sites (high-affinity sites for K+ demonstrable in terms of activation of the overall ATPase reaction), and at the Na+ sites for activation. The relative efficacy of Li+ was estimated in terms of the apparent maximal velocity of the phosphatase and ATPase reactions when Li+ was substituted for K+, and also in terms of the relative effect of Li+ on the apparent KM for Mg· ATP. With these data, and previously determined values for the apparent affinities of K+ and Na+ at these same sites, quantitative kinetic models for the stimulation were examined. A composite model is required in which Li+ stimulates by relieving inhibition due to K+ and Na+ (i) by competing with K+ for the α-sites on the enzyme through which K+ decreases the apparent affinity for Mg·ATP and (ii) by competing with Na+ at low-affinity inhibitory sites, which may represent the external sites at which Na+ is discharged by the membrane NA+/K+ pump that this enzyme represents. Both these sites of action for Li+ would thus lie, in vivo, on the cell exterior.  相似文献

Summary The influence of K+ ions on the net Na+ fluxes in cells of excised barley roots (Hordeum distichon L.) and roots of whole barley plants was investigated. The fluxes were determined by flame photometry in the external solution. In both cases a transient net Na+ efflux against the external Na+ concentration was observed upon addition of K+. The results stress the effectiveness of the K+-dependent Na+ efflux mechanism residing at the plasmalemma, and its involvement in K–Na-selectivity in whole barley plants.  相似文献

The uptake of glycine in rabbit renal brush border membrane vesicles was shown to consist of glycine transport into an intravesicular space. An Na+ electrochemical gradient (extravesicular>intravesicular) stimulated the initial rate of glycine uptake and effected a transient accumulation of intravesicular glycine above the steady-state value. This stimulation could not be induced by the imposition of a K+, Li+ or choline+ gradient and was enhanced as extravesicular Na+ was increased from 10 mM to 100 mM. Dissipation of the Na+ gradient by the ionophore gramicidin D resulted in diminished Na+-stimulated glycine uptake. Na+-stimulated uptake of glycine was electrogenic. Substrate-velocity analysis of Na+-dependent glycine uptake over the range of amino acid concentrations from 25 μM to 10 mM demonstrated a single saturable transport system with apparent Km = 996 μM and Vmax = 348 pmol glycine/mg protein per min. Inhibition observed when the Na+-dependent uptake of 25 μM glycine was inhibited by 5 mM extravesicular test amino acid segregated dibasic amino acids, which did not inhibit glycine uptake, from all other amino acid groups. The amino acids d-alanine, d-glutamic acid, and d-proline inhibited similarly to their l counterparts. Accelerative exchange of extravesicular [3H]glycine was demonstrated when brush border vesicles were preloaded with glycine, but not when they were preloaded with l-alanine, l-glutamic acid, or with l-proline. It is concluded that a single transport system exists at the level of the rabbit renal brush border membrane that functions to reabsorb glycine independently from other groups of amino acids.  相似文献

Na+-dependent leucine uptake was greater in potassium loaded brush-border membrane vesicles compared with controls. This effect was not mediated by an electrical potential difference, since it was still present in voltage-clamped conditions. Inhibition experiments indicate the same Na+-dependent leucine transport activity in the presence or in the absence of potassium. The affinity of sodium for the cotransporter was identical at 10 or 100 mM potassium. Leucine kinetics at different potassium concentrations showed a maximum 2.4-fold increase in Vmax, while Km was unaffected. The secondary plots of the kinetic results were not linear. This kinetic behaviour suggests that K+ acts as a non-essential activator of Na+-dependent leucine cotransport. A charge compensation of sodium-leucine influx is most probably a component of the potassium effect in the presence of valinomycin.  相似文献

PbCl2 in micromolar concentrations stimulates phosphorylation of electroplax microsomal protein in the absence of Na+. Other divalent cations showed little or no such effect. The (Mg2+ + Pb2+)- and (Mg2+ + Na+)-dependent membrane-bound protein kinase activities in electroplax particulate preparations exhibit properties in common, including their acid stability, ouabain sensitivity, ATP specificity, and molecular size. It is concluded that the (Mg2+ + Pb2+)-dependent phosphoprotein is part of the Na+-, K+-dependent adenosine triphosphatase [(NaK)ATPase]. The Pb2+-dependent product, in contrast to the Na+-dependent one, is insensitive to K+ and the hydrolysis of ATP is thus inhibited.  相似文献

The specific activity of (Na+ + Mg2+)-dependent ATPase is three times greater in the microsomes of sea-water eels than in freshwater eels; the specific activity is one quarter of that of (Na+ + K+ + Mg2+)-dependent ATPase in both cases.(Na+ + Mg2+)-dependent ATPase is optimally active in a medium containing 8 mM NaCl, 4 mM MgCI2, 4 mM ATP, pH 8.8 and at 30 °C; the enzyme is inhibited by ouabain, by NaCl concentrations > 100 mM and by treatment with urea.It is concluded that the (Na+ + Mg2+)-dependent ATPase activity of gills arises from the presence of a (Na+ + K+ + Mg2+)-dependent ATPase.  相似文献

K+-dependent Na+-Ca2+ exchangers (NCKXs) play an important role in Ca2+ homeostasis in many tissues. NCKX proteins are bi-directional plasma membrane Ca2+-transporters which utilize the inward Na+ and outward K+ gradients to move Ca2+ ions into and out of the cytosol (4Na+:1Ca2+ + 1 K+). In this study, we carried out scanning mutagenesis of all the residues of the highly conserved α-1 and α-2 repeats of NCKX2 to identify residues important for K+ transport. These structural elements are thought to be critical for cation transport. Using fluorescent intracellular Ca2+-indicating dyes, we measured the K+ dependence of transport carried out by wildtype or mutant NCKX2 proteins expressed in HEK293 cells and analyzed shifts in the apparent binding affinity (Km) of mutant proteins in comparison with the wildtype exchanger. Of the 93 residue substitutions tested, 34 were found to show a significant shift in the external K+ ion dependence of which 16 showed an increased affinity to K+ ions and 18 showed a decreased affinity and hence are believed to be important for K+ ion binding and transport. We also identified 8 residue substitutions that resulted in a partial loss of K+ dependence. Our biochemical data provide strong support for the cation binding sites identified in a homology model of NCKX2 based on crystal structures reported for distantly related archaeal Na+-Ca2+ exchanger NCX_Mj. These experiments examined effects of several ligands on the K+ p-nitrophenylphosphatase activity of the (Na+,K+)-ATPase in membranes of a rat brain cortex synaptosomal preparation. K+-independent hydrolysis of this substrate by the synaptosomal preparation was studied in parallel; the rate of hydrolysis in the absence of K+ was approximately 75% less than that observed when K+ was included in the incubation medium. The response to the H+ concentrations was different: K+-independent activity showed a pH optimum around 6.5–7.0, while the K+-dependent activity was relatively low at this pH range. Ouabain (0.1 mM) inhibited K+-dependent activity 50%; a concentration 10 times higher did not produce any appreciable effect on the K+-independent activity. Na+ did not affect K+-independent activity at all, while the same ligand concentration inhibited sharply the K+-dependent activity; this inhibition was not competitive with the substrate,p-nitrophenyl phosphate. K+-dependent activity was stimulated by Mg2+ with low affinity (millimolar range), and 3 mM Mg2+ produced a slight stimulation of the activity in absence of K+, which could be interpreted as Mg2+ occupying the K+ sites. Ca2+ had no appreciable effect on the activity in the absence of K+. However, in the presence of K+ a sharp inhibition was found with all Ca2+ concentrations studied. ATP (0.5 mM) did not affect the K+-independent activity, but this nucleotide behaved as a competitive inhibitor top-nitrophenylphosphate. The uptake of l-glutamic acid into brush-border membrane vesicles isolated from rat renal proximal tubules is Na+-dependent. In contrast to Na+-dependent uptake of d-glucose, pre-equilibration of the vesicles with K+ stimulates l-glutamic acid uptake. Imposition of a K+ gradient ([Ki+] > [Ko+]) further enhances Na+-dependent l-glutamic acid uptake, but leaves K+-dependent glucose transport unchanged. If K+ is present only at the outside of the vesicles, transport is inhibited. Intravesicular Rb+ and, to a lesser extent, Cs+ can replace intravesicular K+ to stimulate l-glutamic acid uptake. Changes in membrane potential incurred by the imposition of an H+-diffusion potential or anion replacement markedly affect Na+-dependent glutamic acid uptake only in the presence of K+. Experiments with a potential-sensitive cyanine dye also indicate that, in the presence of intravesicular K+ a charge movement is involved in Na+-dependent transport of l-glutamic acid.The data indicate that Na+-dependent l-glutamic acid transport can be additionally energized by a K+ gradient. Furthermore, intravesicular K+ renders Na+-dependent l-glutamic acid transport sensitive to changes in the transmembrane electrical potential difference.  相似文献

Inactivation of the (Na+ + K+)-dependent ATPase by 50 μm BeCl2 occurred during brief incubations in the presence of both Mg2+ and K+. Inactivation followed, initially, a first-order time course, with rate constants sensitive to the concentration of K+ (other components held constant). From these data dissociation constants can be calculated for K+ binding to sites controlling inactivation. Comparisons of relative affinities for K+ analogs (T1+ and NH4+), and of sensitivity to reagents altering K+ activation (phlorizin and dimethylsulfoxide) indicate that the same K+ sites operate for both Be2+ inactivation and enzyme activation. With 3 mm MgCl2 the dissociation constant, KD, for K+ was 1.4 mm, but decreased 20-fold on addition of both Na+ and CTP. Alone, Na+ increased the apparent KD for K+, either by direct competition or indirectly from its own site, with a KD of 7 mm. The data suggest a model for K+ transport with K+ sites on the outer membrane surface that increase in affinity after formation of the phosphorylated enzyme intermediate, sufficiently to bind K+ in a high Na+ environment. Translocation may occur by an “oscillating pore” mechanism discharging K+ at the inner surface, while leaving demonstrable sites of moderate affinity at the outer end of the pore (which preclude attempts to document low-affinity discharge sites).  相似文献

Because intracellular [Na+] is kept low by Na+/K+-ATPase, Na+ dependence is generally considered a property of extracellular enzymes. However, we found that p94/calpain 3, a skeletal-muscle-specific member of the Ca2+-activated intracellular “modulator proteases” that is responsible for a limb-girdle muscular dystrophy (“calpainopathy”), underwent Na+-dependent, but not Cs+-dependent, autolysis in the absence of Ca2+. Furthermore, Na+ and Ca2+ complementarily activated autolysis of p94 at physiological concentrations. By blocking Na+/K+-ATPase, we confirmed intracellular autolysis of p94 in cultured cells. This was further confirmed using inactive p94:C129S knock-in (p94CS-KI) mice as negative controls. Mutagenesis studies showed that much of the p94 molecule contributed to its Na+/Ca2+-dependent autolysis, which is consistent with the scattered location of calpainopathy-associated mutations, and that a conserved Ca2+-binding sequence in the protease acted as a Na+ sensor. Proteomic analyses using Cs+/Mg2+ and p94CS-KI mice as negative controls revealed that Na+ and Ca2+ direct p94 to proteolyze different substrates. We propose three roles for Na+ dependence of p94; 1) to increase sensitivity of p94 to changes in physiological [Ca2+], 2) to regulate substrate specificity of p94, and 3) to regulate contribution of p94 as a structural component in muscle cells. Finally, this is the first example of an intracellular Na+-dependent enzyme.  相似文献

A rapid loss of accumulated Ca2+ is produced by addition of H+ to isolated heart mitochondria. The H+-dependent Ca+ efflux requires that either (a) the NAD(P)H pool of the mitochondrion be oxidized, or (b) the endogenous adenine nucleotides be depleted. The loss of Ca2+ is accompanied by swelling and loss of endogenous Mg2–. The rate of H+-dependent Ca2+ efflux depends on the amount of Ca2+ and Pi taken up and the extent of the pH drop imposed. In the absence of ruthenium red the H+-induced Ca2+-efflux is partially offset by a spontaneous re-accumulation of released Ca2+. The H+-induced Ca2+ efflux is inhibited when the Pi transporter is blocked withN-ethylmaleimide, is strongly opposed by oligomycin and exogenous adenine nucleotides (particularly ADP), and inhibited by nupercaine. The H+-dependent Ca2+ efflux is decreased markedly when Na+ replaces the K+ of the suspending medium or when the exogenous K+/H+ exchanger nigericin is present. These results suggest that the H+-dependent loss of accumulated Ca2+ results from relatively nonspecific changes in membrane permeability and is not a reflection of a Ca2+/H+ exchange reaction.  相似文献

Inside-out membrane vesicles have been prepared from sheep reticulocytes. With these vesicles, Na+-dependent glycine uptake and net accumulation have been demonstrated to occur in reverse, i.e., from extravesicular (normal cytoplasmic) to intravesicular (normal extravesicular) surface. Uptake and accumulation are inhibited by energization of the sodium pump by ATP whereby the Na+ electrochemical gradient is dissipated. Glycine-dependent Na+ uptake was also observed, providing evidence that Na+-dependent glycine influx into these vesicles, equivalent to normal efflux, is characterized by Na+-glycine co-transport.  相似文献

External treatment of human erythrocytes with the diazonium salt of sulfanilic acid does not inhibit the Mg2+-dependent ATPase but does markedly inhibit the Ca2+-stimulated ATPase activity. Inhibition of the (Na+ + K+)-dependent activity is dependent upon the concentration of diazonium salt used. Treatment of membrane fragments does not irreversibly inhibit the (Na+ + K+)-dependent ATPase even though the diazonium salt binds covalently to membrane components. However, the Mg2+-dependent and Ca2+-stimulated ATPase activities are irreversibly inhibited. ATP and Mg-ATP will completely protect the (Na+ + K+)-dependent ATPase when present during treatment of membrane fragments with the diazonium salt, but only Mg-ATP will protect the Mg2+-dependent ATPase from inhibition. The Ca2+-stimulated ATPase activity is not protected.  相似文献

Ascorbic acid (AA) is an essential cofactor for osteoblast differentiation both in vivo and in vitro. Before it can function, this vitamin must be transported into cells via a specific Na+-dependent AA transporter. In this study, we examine the regulation of this transport activity by glucocorticoids, a class of steroid hormones known to stimulate in vitro osteoblast differentiation. Dexamethasone stimulated Na+-dependent AA transport activity approximately twofold in primary rat calvarial osteoblasts. Effects of hormone on ascorbic acid transport were rapid (detected within 24 h) and were maximally stimulated by 25–50 nM dexamethasone. Similar effects of dexamethasone on transport activity were also observed in murine MC3T3-E1 cells. This preosteoblast cell line was used for a more detailed characterization of the glucocorticoid response. Transport activity was stimulated selectively by glucocorticoids (dexamethasone > corticosterone) relative to other steroid hormones (progesterone and 17-β-estradiol) and was blocked when cells were cultured in the presence of cycloheximide, a protein synthesis inhibitor. Kinetic analysis of AA transporter activity in control and dexamethasone-treated cells indicated a Km of approximately 17 μM for both groups. In contrast, dexamethasone increased Vmax by approximately 2.5-fold. Cells also contained an Na+-independent glucose transport activity that has been reported in other systems to transport vitamin C as oxidized dehydroascorbic acid. In marked contrast to Na+-dependent AA transport, this activity was inhibited by dexamethasone. Thus, glucocorticoids increase Na+-dependent AA transport in osteoblasts, possibly via up-regulation of transporter synthesis, and this response can be resolved from actions of glucocorticoids on glucose transport. Rat hindbrain NaI-enriched microsomal (Na+-K+)-ATPase activity, K+-pNPPase activity, and Na+-dependent steady-state phosphorylation levels all increase approx 10-fold relative to microsomal protein between 5 days prenatally and 60 days postnatally. These activities, as well as the mean wet weight of the hindbrain, are at half of their 60 day values shortly after the 10th postnatal day. For all ages, these hindbrain activities average over twice those found in the forebrain in a companion paper (Bertoni & Siegel , 1978). Increases during development in the amount of the related phosphorylatable polypeptide, estimated by densitometry of stained polyacrylamide gels containing fixed amounts of microsomal protein dissolved in SDS, are in agreement with increases in steady state levels of Na+-dependent phosphorylation. The fraction of total phosphorylation that is Na+-dependent rises steadily during development consistent with, but not obligatorily due to, a conversion of some of the previously Na+-independent portion. Mg2+-ATPase and Mg2+ -pNPPase activities and steady-state Na+,-independent phosphorylation levels do not increase in parallel during development. These observations add further support to the proposed partial reaction scheme for (Na+- K+)-ATPase. The major increments in (Na +-K+)-ATPase occur simultaneously with the deposition of specialized plasma membranes, particularly in the molecular layer of the cerebellum, as described in previous studies of rat hind brain.  相似文献

Amino acids stimulated the (Na+ + K+)-dependent ATPase activity of a rabbit kidney preparation without affecting the Mg2+-ATPase activity; the most effective was histidine, producing a 2-fold increase in activity. Similar stimulation was produced by the well-known chelators EDTA, EGTA, and 8-hydroxyquinoline, and by the chelating phospholipid phosphatidylserine. In the presence of maximally effective concentrations of one agent, the other agents were unable to produce additional stimulation. It is suggested that the amino acids, phosphatidylserine, and the conventional chelators all stimulate the ATPase by a common mechanism: the removal of inhibitory trace metal (s). From measurements of the metal content of the enzyme preparation and experiments with extracted reagents it was concluded that the chelatable inhibitor was in the reagents used in the incubation medium rather than being endogenous to the enzyme; attempts to identify the inhibitor (s) were unsuccessful. The chelators also stimulated the K+-dependent phosphatase activity in the preparation but had no major effect on Na+-dependent incorporation of 32P from [32P]ATP. On monovalent cation activation the chelators appeared to relieve an uncompetitive inhibition of Na32 activation and a noncompetitive inhibition of K32 activation, also suggesting an action of the chelatable inhibitor on the later stages of the ATPase reaction sequence.  相似文献
