Power transmission along trichomes of filamentous cyanobacteria Phormidium uncinatum has been studied with the use of ethylrhodamine fluorescence as a probe for the transmembrane electric potential difference (delta psi). It is found that agents preventing the light-induced delta psi formation (photosynthetic redox chain inhibitor dibromothymoquinone) or dissipating delta psi (uncoupler tetrachlorotrifluoromethylbenzimidazole) strongly decrease the fluorescence of the ethyl-rhodamine-stained trichomes. K+-H+ antiporter nigericin converting delta pH to delta psi increases the fluorescence. These relationships are in agreement with the assumption that ethylrhodamine electrophoretically accumulates inside the cyanobacterial cells. Illumination of a single cell in the P. uncinatum trichome gives rise to quenching of the fluorescence in this cell and usually in one or two neighbor cells, whereas the rest of trichome remains fluorescing. A small light spot (5% of the trichome length) causes an increase in the ethylrhodamine fluorescence not only in the illuminated but also in the nonilluminated parts of the trichome up to the laser-treated cell or its neighbor(s). It is concluded ethylrhodamine can be used to monitor the power transmission which was previously demonstrated by microelectrode studies of the cyanobacterial trichomes. In certain trichomes, several "dark" cells appear during the storage of the trichomes without energy sources. Illumination for several minutes results in dark cells becoming fluorescing. Thus some cells or cell clusters can be reversibly excluded from the lateral delta psi-transmitting system of the trichome, the rest being still electrically connected. This means that filamentous cyanobacteria possess mechanisms to transmit power along the trichome and to switch off this transmission.  相似文献

The light-induced electric potential changes brought about local illumination of trichomes of cyanobacteria Phormidium uncinatum have been studied by means of extracellular electrodes. Responses of several electrodes located at various distances from the illuminated area of the trichome were monitored simultaneously. They turned out to be similar in shape: a rapid increase to the maximum value was followed by a slow decay toward a nonzero residual level. The results offer strong evidence in favor of power transmission along the trichome. The computerized experimental data lend support to the notion of a unified system of coupling membranes acting as a passive cable for electrical propagation, the cable parameters being tau C = 440 sec cm-2 and gamma = 0.07 cm.  相似文献

A mutant of Halobacterium halobium which contains halorhodopsin was isolated from strain S9. An absorbance change at 380 nm caused by steady orange light illumination (λ ?530 nm) was observed. This change depended upon the intensity of the actinic light. The bleached envelope vesicles and vesicles derived from nicotine-grown cells showed a small or no absorbance change at 380 nm, suggesting that the change stemmed from the photochemical intermediate of halorhodopsin (referred to as P-380). When blue light was superimposed on orange background illumination, the membrane potential (Δψ) of the envelope vesicles decreased. Δψ was determined from the tetraphenylphosphonium cation (TPP+) distribution by means of a TPP+ electrode. When blue light intensity was increased, both Δψ and the amount of P-380 were decreased. An equation was derived which showed that Δψ is proportional to the concentration of P-380 formed by illumination under the assumption that the ionic composition is not significantly changed upon illumination. This equation was checked experimentally from the following three points: The blue light effect, the relationship between Δψ and light intensity, and the effect of gramicidin. The data obtained accorded well with the theoretical relationship.  相似文献

Abstract The front portion ('head') of a Phormidium uncinatum trichome responds to a step-down in light intensity [10], whereas the rear end ('tail') responds to step-up stimuli [11]. We studied this phenomenon further and found that: (i) illumination of the head caused a reversal within 1 min in only 6% of the trichomes, whereas illumination of the tail produced a reversal in 56% of trichomes; (ii) if a light spot trained on the head of a trichome was moved together with the trichome, there were no reversals for > 20 min, while the normal rate of spontaneous reversals was once per 3–5 min. Shifting the light spot from the head to the tail caused a reversal within 1–2 min; (iii) both the step-up response of the tail and phototaxis were suppressed by an inhibitor of methylation, ethionine, but not by inhibitors of photosynthesis (DCMU, DBMIB); phototaxis and the step-up response of the tail were absent in red light ( λ > 670 nm). It was concluded that trichomes of P. uncinatum possess a one-instant mechanism of phototaxis, which involves a simultaneous comparison of light intensities between two parts of the organism.  相似文献

Cyanobacteria form a very large and diverse phylum of prokaryotes that perform oxygenic photosynthesis. Many species of cyanobacteria live colonially in long trichomes of hundreds to thousands of cells. Of the filamentous species, many are also motile, gliding along their long axis, and display photomovement, by which a trichome modulates its gliding according to the incident light. The latter has been found to play an important role in guiding the trichomes to optimal lighting conditions, which can either inhibit the cells if the incident light is too weak, or damage the cells if too strong. We have developed a computational model for gliding filamentous photophobic cyanobacteria that allows us to perform simulations on the scale of a Petri dish using over 10(5) individual trichomes. Using the model, we quantify the effectiveness of one commonly observed photomovement strategy--photophobic responses--in distributing large populations of trichomes optimally over a light field. The model predicts that the typical observed length and gliding speeds of filamentous cyanobacteria are optimal for the photophobic strategy. Therefore, our results suggest that not just photomovement but also the trichome shape itself improves the ability of the cyanobacteria to optimize their light exposure.  相似文献

Possible energy transmission along conjugating membranes was investigated. It was found by extracellular electrodes that under local illumination of blue-green algae Phormidium uncinatum the electrical current propagated along trichomes. The form and sign of the responses show that e. m. f. appears in the illumination region, which generates on the cellular membrane the potential difference with a negative charge inside the cell. The initiated transmembrane potential difference propagates along the filament of Ph, uncinatum like along a cable. The hypothesis concerning a cable structure of Ph, uncinatum was supported in the experiments with intersection of algae, and with intracellular injection of fluorescein showing the existence of direct diffusion exchange through cellular interfaces.  相似文献

Electrical events after a light-dark stimulus were studied in the multicellular organism Phormidium uncinatum. Normally, such a stimulus causes the gliding trichome to reverse direction. By directing a large light spot on the end of a batch of trichomes and then switching it off, we achieved synchronization of the trichomes, since the "head" is much more sensitive than the "tail." The abrupt disappearance of a uniform light produced a depolarization wave which initiated at the head, as registered by externally applied electrodes. The second stimulus produced a depolarization of the opposite direction, reflecting the reorientation of the trichomes. No electrical response was observed at Ca2+ concentrations less than or equal to 10(-8) M. Factors causing oscillatory reversals, i.e., a combination of Ca2+ and A23187, or a viscous environment also abolished the electrical signal. Changes in an externally applied electrical field (4 V/cm2) had little effect on the motile behavior of P. uncinatum or Oscillatoria princeps. However, in the presence of 5 microM Ca2+-1 microM A23187, all the trichomes reversed synchronously to the anode after a change in polarity of an externally applied electrical field. We suggest that an increased Ca2+ concentration together with a change in delta psi (or delta mu H+) represents the taxis signal in cyanobacteria.  相似文献

Extracellular recordings of electrical potential in leaves of different species by means of band-pass amplifiers showed the occurrence of fast, small changes (spikes) with an amplitude below 1 mV. Local illumination of leaves induced temporal patterns of spikes outside the illuminated region. The light-induced patterns recorded by a given electrode in a particular experimental setup were similar for successive illuminations. The patterns recorded at different sites on the same leaf were different. Locally repetitive patterns of spikes at the electrode outside the illuminated region indicate the occurrence of some signals transmitted from this region to the cells in the neighborhood of the electrode.  相似文献

Hypothesis on long-distance power transmission along extended energy-transducing membranes (Skulachev, 1969, 1971, 1980), has been experimentally proven in four different systems, namely, (i) trichomes of filamentous cyanobacterium Phormidium uncinatum; (ii) filamentous mitochondria and mitochondrial network in fibroblasts; (iii) clusters of roundish heart muscle mitochondria interconnected with mitochondrial junctions; (iv) mixed animal cell cultures interconnected with gap junctions. In all cases, energy was shown to be transmitted in the form of a transmembrane electric potential difference. The transmission occurred for distances as long as several tens of micrometers. Since the (a) delta-muH-bearing cytoplasmic membrane of cyanobacteria and the inner mitochondrial membrane and (b) delta-muNa-bearing outer animal cell membrane were found to be competent in such an effect, one may assume that the power transmission is a fundamental function of extended membrane systems. This mechanism can be used at the intracellular level (mitochondrial) as well as at the supracellular level (cytoplasmic and outer cell membranes). Studies on the possible involvement of membranes in lateral transport of oxygen, ions, fatty acids and membrane proteins seem to hold good promise.  相似文献

Trichomes have been implicated as a mechanism which can confer resistance to both plant pests and drought. A study was conducted to provide information regarding genetic variability for trichome distribution and density among three diverse dry bean (Phaseolus vulgaris L.) cultivars, and to characterize the types of trichomes present among the cultivars. Trichomes on the leaf surfaces were micrographed with a scanning electron microscope (SEM) and counted using a stereomicroscope on both the abaxial and adaxial leaf surfaces of the cultivars ‘Bill Z’, ‘Pompadour Checa’ and ‘Diacol Calima’. Straight, hooked, and glandular trichomes were observed on the leaf surfaces of each cultivar. SEM micrographs are presented for the leaf surfaces of each cultivar and trichome type. The abaxial leaf surface had more straight trichomes than the adaxial leaf surface for ‘Pompadour Checa’ and ‘Diacol Calima’, however ‘Bill Z’ had more on the adaxial surface. The opposite relationship existed among the cultivars and leaf surfaces for the hooked trichomes.  相似文献

By the conversion of sunlight to chemical energy, photoelectrolysis was carried out using two different chlorophyll-redox compound lining electrodes. These electrodes were prepared by covering platinum plates with chlorophyll and naphthoquinone or anthrahydroquinone and a conducting adhesive. These electrodes exhibit a photoexciting property. The potential was found to shift to a less noble state when the system of the chlorophyll-naphthoquinone electrode was inserted into NAD solution with illumination. On the other hand, the photoexcitation of the system of the chlorophyll-anthrahydroquinone electrode inserted into ferrocyanide solution made the potential more noble. (If the potential in the dark is in the positive region of the scheme
and the potential moves in the positive direction when the light is turned on, it can be said to be more positive or more noble. But if the potential moves in the negative direction in the light, but remains in the positive region, it can be said to become less noble, but it is not suitable to say that it becomes more negative.) To make the potential difference between two electrodes as big as possible, various factors such as intensity of illumination, molar ratio of chlorophyll to naphthoquinone or anthrahydroquinone and concentration of redox compound in electrolyte were examined.A cell was set up by combining the system of the chlorophyll-naphthoquinone electrode in NAD solution with that of the chlorophyll-anthrahydroquinone electrode in ferrocyanide solution, and photoelectrolysis was carried out by closing the external circuit with illumination. The photovoltage between the two electrodes was 0.25 V and the current density was 8 μA/cm2. It was found that NAD was reduced to form NADH at the chlorophyll-naphthoquinone electrode and ferrocyanide was oxidized to form ferricyanide at the chlorophyll-anthrahydroquinone electrode.  相似文献

This paper reports the results of a study of the morphology and development of glandular trichomes in leaves ofCalceolaria adscendensLidl. using light and electron microscopy. Secretory trichomes started as outgrowths of epidermal cells; subsequent divisions gave rise to trichomes made up of a basal epidermal cell, a stalk cell and a two-celled secretory head. Ultrastructural characteristics of trichome cells were typical of terpene-producing structures. Previous phytochemical studies had revealed thatC. adscendensproduces diterpenes. Comparison withC. volckmanni,which produces triterpenes, and has trichomes with eight-celled secretory heads, suggests that there could be a relationship between the type of glandular trichome and the class of terpene produced. Further work is needed to test the hypothesis and to develop trichome characters as taxonomic tools.  相似文献

The effects of varying dark interval on the kinetics of light-induced formation of the membrane potential were studied on individual chloroplasts of Anthoceros with the use of capillary microelectrodes. Illumination of the chloroplast with 1 s light pulse after 3 min dark period induced the photoelectrical response with two peaks of the potential that were located at 20 and 500 ms after the onset of illumination. The position of the second peak was shifted along the time-scale depending on the preceding dark interval. The repeated illumination of the chloroplast with 1 s light pulse after 30 s dark interval induced the electrical response with only one maximum and a monotonous decay of the potential in the light. Distinctions in the electrical responses induced by the first and the second light pulses were eliminated by the addition of 50 μM dicyclohexylcarbodiimide (DCCD). The results show that the photoinduction kinetics of the membrane potential in chloroplasts is affected by functioning of H+-ATPase. The delayed peak of the membrane potential in the photoinduction kinetics is interpreted as a consequence of the photoactivated electron transport supported by Photosystem I.  相似文献

Scanning electron microscopy of leaf trichomes of the forty two native species of oaks in eastern North America indicates five patterns of variability: 1) Eight trichome types are evident among the species and each species possesses a definite complement of trichome types. Certain trichomes are restricted to particular subgenera and series. 2) An obvious seasonal loss of trichomes occurs during leaf maturation. This loss may be both quantitative in terms of trichome density and qualitative in terms of trichome type. 3) There is an obvious difference between the adaxial and abaxial surfaces. The adaxial side of most oak leaves is dark green, lustrous, and glabrous or glabrate. The abaxial surface either remains pubescent, becomes glabrate or glabrous, or maintains trichomes along the midrib or in the axils of major secondary veins. There are also initial quantitative and qualitative trichome differences between the two sides. 4) Geographical and ecological variations are due in part to non-genetic ecophenic modifications, ecotypic differentiation, and random genetic differences not necessarily correlated with environmental conditions. Trichome types are considered to be less affected by environment than is trichome density. 5) Hybridization and introgression within a subgenus leads to localized variability. Trichomes of hybrids are usually a combination of the parental types. These five patterns of variation are predictable and appear to be held within rather narrow limits. The complement of foliar trichomes, therefore, is a reliable character in the taxonomy of the oaks.  相似文献

Adenylate concentrations were measured in intact chloroplasts under a variety of conditions. Energy charge was significant in the dark and increased in the light, but remained far below values expected from observed phosphorylation potentials in broken chloroplasts, which were 80 000 M?1 or more in the light. With nitrite as electron acceptor, phosphorylation potentials in intact chloroplasts were about 80 M?1 in the dark and only 300 M?1 in the light. Similar phosphorylation potentials were observed, when oxaloacetate, phosphoglycerate or bicarbonate were used as substrates. ΔGATP was ?42 kJ/mol in darkened intact chloroplasts, ?46 kJ/mol in illuminated intact chloroplasts and ?60 kJ/mol in illuminated broken chloroplasts. Uncoupling by NH4Cl, which stimulated electron transport to nitrite or oxaloacetate and decreased the proton gradient, failed to decrease the phosphorylation potential of intact chloroplasts. Also, it did not increase the quantum requirement of CO2 reduction. It is concluded that the proton motive force as conventionally measured and phosphorylation potentials are far from equilibrium in intact chloroplasts. The insensitivity of CO2 reduction and of the phosphorylation potential to a decrease in the proton motive force suggests that intact chloroplasts are over-energized even under low intensity illumination. However, such a conclusion is at variance with available data on the magnitude of the proton motive force.  相似文献

Floating-leaved rhizophytes and pleustophytes are the first barrier to Sun’s rays and significantly affect the light regime of the water column. To evaluate these effects on light attenuation, the reflectance and transmittance spectra variability were examined according to the leaf traits within three plant groups: (1) seed plants with green abaxial surfaces; (2) seed plants with red abaxial surfaces; and (3) ferns with trichomes. Specific leaf area (SLA), chlorophyll a and b, and UV-B and UV-A-absorbing substances differed between these three groups. The ‘spectral signatures’ of floating-leaved seed plants are similar to those of terrestrial seed plants, with a peak in the green region and a pronounced ‘red edge’. Ferns transmitted more light along the whole spectrum compared to other species. Most reflectance and transmittance spectra variability of the first group was explained by SLA. In the second group, 36% of the reflectance spectra variability was explained by tissue density and carotenoids, and 48% of the transmittance spectra variability by carotenoids, anthocyanins and SLA. In ferns, the reflectance spectra variability was mainly explained by chlorophylls, and partly by trichome length and mesophyll thickness, with the transmittance spectra variability significantly affected by chlorophyll b.  相似文献

The influence of temperature on total glycoalkaloid (TGA) synthesis in tubers exposed to light (250 jumol m“2 s”2 PAR, Photosynthetically Active Radiation) or dark environments for 96 h was examined in three potato cultivars. Following 96 h light or dark the tubers were stored without light at 5°C or 24°C and TGA concentrations monitored over the subsequent 30 and 90 days. Exposure to light and cultivar were found to be major factors influencing TGA concentrations; temperature had no significant effect. TGA content in illuminated tubers of cvs ‘Pentland Hawk’ and ‘Kerrs Pink’ were significantly higher (P < 0.01) compared with tubers placed in the dark. TGA concentrations in cv. ‘Desiree’ increased significantly only following exposure to light at low temperatures (P < 0.05). Removal of tubers from storage at 5°C and immediate illumination at 24°C altered the ratio of glycoalkaloids in cvs ‘Pentland Hawk’ and ‘Kerrs Pink’. Regardless of cultivar and storage temperature TGA concentrations were higher at the end of the storage period compared with initial TGA concentrations. During storage TGA concentrations fluctuated widely and gradual accumulation of glycoalkaloids with time was rarely demonstrated except in cv. ‘Desiree’. Tubers stored at 24°C accumulated higher TGA concentrations than those stored at 5°C in cv. ‘Kerrs Pink’ but not in cvs ‘Pentland Hawk’ and ‘Desiree’. Tubers of cv. ‘Kerrs Pink’ exposed to light prior to storage accumulated glycoalkaloids more rapidly than unexposed tubers during storage at 24°C and occasionally at 5°C. Light enhanced glycoalkaloids are not degraded over time.  相似文献

This study was initiated to characterize the distribution, morphology, secretion mode, histochemistry and ultrastructure of the glandular trichomes of Ceratotheca triloba using light and electron microscopy. Its leaves bear two morphologically distinct glandular trichomes. The first type has long trichome with 8-12 basal cells of pedestal, 3-14 stalk cells, a neck cell and a head of four cells in one layer. The second type has short trichome comprising one or two basal epidermal cells, a unicellular or bicellular stalk and a multicellular head of two to eight cells. There is a marked circular area in the upper part of each head cell of the long trichome. This area is provided with micropores to exudate directly the secretory product onto the leaf surface by an eccrine pathway. The secretory product has copious amount of dark microbodies arising from plastids which are positive to Sudan tests and osmium tetroxide for unsaturated lipids. The secretion mode of short trichomes is granulocrine and involves two morphologically and histochemically distinct vesicle types: small Golgi-derived vesicles which are positive to Ruthenium Red test for mucilaginous polysaccharides; the second type is dark large microbodies similar to that of long trichomes with low quantity. These two types are stored in numerous peripheral vacuoles and discharge their contents accompanied by the formation of irregular invaginations of the plasmalemma inside the vacuoles via reverse pinocytosis. These two secretion modes of long and short trichomes are reported for the first time in the family Pedaliaceae. The long trichomes have more unsaturated lipids, while the short trichomes contain more mucilaginous polysaccharides.  相似文献

An attempt was made to find evidence that morphologically distinct terminal cells of filamentous cyanobacterium Aphanizomenon gracile strain CCALA 8 are capable of dividing and forming trichomes. Based on our current knowledge, the division of morphologically diversified terminal cells is possible in nostocalean cyanobacteria. However, this process has been observed only in a few species. Terminal cells of A. gracile differ morphologically from other vegetative cells of a trichome, as they are not hyaline and can sometimes be found as solitary cells in cultures. Hence, it was reasonable for us to suspect that these cells are capable of dividing and forming trichomes. We observed terminal cells under a light and transmission electron microscope. Microscopic observations revealed that the septum formed in both solitary terminal cells and in terminal cells attached to trichomes. Our study is the first to demonstrate division and renewal of trichomes in terminal cells of A. gracile. Previously, such mode of reproduction was described only for another nostocalean cyanobacterium Raphidiopsis mediterranea. Moreover, our findings further emphasize the variability among members that belong to the genus Aphanizomenon , in which a type species (A. flos‐aquae) has hyaline cells incapable of dividing and renewing trichomes, while A. gracile can additionally propagate by solitary terminal cells division. This additional feature distinguishing A. gracile from typical species of Aphanizomenon, such as A. flos‐aquae, might be valuable for resolving taxonomic position of the species considering ambiguous genetic relationship between A. gracile and A. flos‐aquae.  相似文献
