We derive a formula for the correlation of the three practical transportparameters Lp, , and appearing in Kedem-Katchalskyequations. It has a form = KLp/vs(1-), where K = 0.0306 is a universal constant independent ofthe choice of a membrane and a solute. It can be used to calculate the valueof any of these parameters, provided the other two and the molar volume of the solute are known. The formula couldbe very useful, in particular when measurements of the parameters aredifficult or even impossible.  相似文献

The plasma membrane vesicles were purified from soybean ( Glycine max L. ) hypocotyls by two-phase partitioning methods. The stimulatory effects of K+ on the coupling between ATP hydrolysis and proton transport by the plasma membrane H+-ATPase were studied. The results showed that the proton transport activity was increased by 850% in the presence of 100 mmol/L KC1, while ATP hydrolytic activity was only increased by 28.2%. Kinetic studies showed that Km of ATP hydrolysis decreased from 1.14 to 0.7 mmol/L, while Vmax of ATP hydrolysis increased from 285.7 to 344.8 nmol Pi·mg- l protein·min-1 in the presence of KC1. Experiments showed that the optimum pH was 6.5 and 6.0 in the presence and absence of KC1, respectively. Further studies revealed that K+ could promote the inhibitory effects of hydroxylamines and vanadates on the ATP hydrolytic activity. The above results suggested that K+ could regulate the coupling between ATP hydrolysis and proton transport of the plasma membrane H+ -ATPase through modulating the structure and function of the kinase and phosphatase domains of the plasma membrane H + -ATPase.  相似文献

Lathyrus sativus L. (Grass pea) is the source for cheap and nutritious food choice in drought and famine susceptible zones in greater part of North India and Africa. The non-protein amino acid β-N-oxalyl-L-α,β-diaminopropionic acid (β-ODAP) has been known for decades for its potent neurotoxic effect, causing irreversible neurodegenerative disease “neurolathyrism”, present in both seed and leaf of Lathyrus sativus L. and other species in varying proportions. It is crucial to establish a rapid as well as reliable detection methodology for β-ODAP content in various Lathyrus plants. Currently available HPLC based methods involve multi-step derivatization of the sample. To overcome this, we have developed β-ODAP analysis method by HPLC without any prior derivatization. This method is statistically significant in the range of 2 to 100μg/ml and exhibited linear response with r 2 > 0.99. Limit of detection and quantitation of the later method was determined to be 5.56 μg/ml and 16.86 μg/ml, respectively. In addition to this, a TLC based method has also been developed. The limit of detection of β-ODAP is 0.6μg and for its substrate, L-1,2-diaminopropionic acid is 5μg. Both HPLC and TLC methods were validated by conducting in-vitro bioconversion test to detect the presence of biocatalyst in plant extract. This method is economical, rapid and simple.  相似文献

The chromosome numbers of 5 tuberous sections of Chinese Dioscorea, including 23 species and varieties, are reported in the present paper as a continuation of the previous reports. They are all polyploids with the basic number x=10. On the basis of analysis of chromosome numbers of whole genus, the rhizomatous diploid species of Sect. Stenophora Uline are presumed to be primitive taxa, while the polyploids of chromosome numbers 40-142 are considered derived groups as a result of hybridization between their ancestral diploids followed by chromosome doubling. Sect. Lasiophyton Pr. et Burk., Sect. Opsophyton UIine, Sect. Shannicorea Pr. et Burk., Sect. Combilium Pr. et Burk. and Sect. EnantiophylIum Uline may be the advanced groups. The chromosomal evolution and geographical distribution suggest that the primitivediploid might have originated in Hengduan Mountains of Asia, an old highland.  相似文献

The Penicillium chrysogenum oat1 gene, which encodes a class III omega-aminotransferase, was cloned and characterized. This enzyme converts lysine into 2-aminoadipic semialdehyde, and plays an important role in the biosynthesis of 2-aminoadipic acid, a precursor of penicillin and other beta-lactam antibiotics. The enzyme is related to ornithine-5-aminotransferases and to the lysine-6-aminotransferases encoded by the lat genes found in bacterial cephamycin gene clusters. Expression of oat1 is induced by lysine, ornithine and arginine, and repressed by ammonium ions. AreA-binding GATA and GATT sequences involved in regulation by ammonium, and an 8-bp direct repeat associated with arginine induction in Emericella (Aspergillus nidulans and Saccharomyces cerevisiae, were found in the oat1 promoter region. Deletion of the oat1 gene resulted in the loss of omega-aminotransferase activity. The null mutants were unable to grow on ornithine or arginine as sole nitrogen sources and showed reduced growth on lysine. Complementation of the null mutant with the oat1 gene restored normal levels of omega-aminotransferase activity and the ability to grow on ornithine, arginine and lysine. The role of the oat1 gene in the biosynthesis of 2-aminoadipic acid is discussed.  相似文献
