The effect of water deficit on nodulation, N2 fixation, photosynthesis, and total soluble sugars and leghemoglobin in nodules was investigated in cowpea and groundnut. Nitrogenase activity completely ceased in cowpea with a decrease in leaf water potential ( leaf) from –0.4 MPa to –0.9 MPa, while in groundnut it continued down to –1.7 MPa. With increasing water stress, the acetylene reduction activity (ARA) declined very sharply in cowpea, but ARA gradually decreased in groundnut. Even with mild water stress ( leaf of 0.2 MPa), nodule fresh weight declined 50% in cowpea partly due to a severe nodule shedding whereas nodule fresh weight declined in groundnut only when leaf decreased by 1.0 MPa. No nodule shedding was noticed even at a higher stress level in groundnut. Photosynthesis and stomatal conductance were also more stable in groundnut than in cowpea under water stress. There was a sharp increase in total soluble sugars and leghemoglobin in the nodules of groundut with water stress, but no definite trend could be found in cowpea.  相似文献

Studies on the in vitro propagation of Alnus crispa, A. glutinosa, A. incana, A. japonica, A. rubra, A. sinuata and A. viridis indicated interspecific as well as intraspecific variations in their requirements for in vitro culture. The WPM and Blaydes media supported, respectively, growth of A. glutinosa and A. crispa but not that of both species, while the MS medium induced equal or significantly better growth than WPM and Blaydes media for both species. The optimum type and concentration of sugar to be used in the multiplication medium varied with species. Only A. glutinosa showed good growth on sucrose while glucose was optimum for all other species but at different concentrations. All species rooted in 3 weeks on half-strength MS medium including 1 M IBA. All clones of A. glutinosa and A. rubra rooted 100%, whereas easy-to-root and difficult-to-root clones were observed in the other species. In the rooting medium, glucose promoted rooting of the difficult-to-root clones better than sucrose. Survival following transfer to an artificial substrate was 100% for all species. Nodulation tests using pure cultures of two Frankia strains showed 100% nodulation on all Alnus clones.  相似文献

Cotyledonary explants of 4-day-oldCucumis melo cv. Hale's Best Jumbo in vitro seedlings showed maximum initiation of shoot buds when cultured onto a revised Murashige & Skoog medium supplemented with 5 M indole-3-acetic acid and 5 M benzylaminopurine and cultured at 25–29°C under low light intensity (5–30 mol m-2 s-1). Subculture of the shoot buds onto the same medium without auxin and supplemented with 3 M benzylaminopurine caused the development of shoots from 30% of the buds. The presence of abscisic acid significantly increased the number of explants producing shoot buds. Bud initiation was affected by genotype, seedling age, light intensity, and temperature. Addition of gibberellic acid, thidiazuron or silver nitrate to regeneration medium did not improve either bud initiation or shoot regeneration.  相似文献

The influence of different Al concentrations, (0, 60 and 120 M Al) on growth and internal concentrations of Al, Si and selected organic acids was analysed in plants of teosinte (Zea mays L. ssp. mexicana), a wild form of maize from acid soils from Mexico. The plants were grown in nutrient solutions (pH 4.0) with or without 4 M silicon. Analysis with the GEOCHEM speciation program did not reveal differences between free activities of Al3+ in solutions with and without 4 M Si, but solutions with Si yielded lower concentrations of monomeric Al species, [Al]mono, when analysed by a modified aluminon method. Plants grown on solutions with similar [Al]mono, but differing in silicon, showed highly significant differences in growth and tissue concentrations of Al and organic acids. Silicon prevented growth inhibition at [Al]mono concentrations as high as 35 M, while plants grown without Si suffered severe growth reductions with 33 M [Al]mono. In solutions with similar [Al]mono concentrations plants with Si had lower tissue Al concentrations and higher concentrations of malic acid than plants without Si. In view of both the significant influence of Si on the response of plants to Al toxicity and the fact that some soluble Si is always present in soil solutions, the addition of low Si concentrations to nutrient solutions used for Al-tolerance screening is recommended.  相似文献

The aluminium (Al) tolerance of 34 temperate legume species (143 genotypes, including 57 from Trifolium repens) was determined in 60 experiments over a 3 year period in a low ionic strength (2.7 × 10-3 M) solution culture. For each genotype, the relationship between solution Al3+ activity (M) and relative yield was determined and the Al3+ activity associated with a 50% reduction in yield (AlRY50) calculated. In addition, plant chemical concentrations were determined in at least one genotype from most species. For white clover, AlRY50 over all genotypes had an approximately normal distribution with mean of 1.31 M for the tops and 1.51 M for the roots, and a standard deviation of about 0.4. This suggested that Al tolerance had a polygenic inheritance. For the other species tested, AlRY50 ranged from 0.15 to 4.53 M in the tops and from 0.21 to 4.89 M in the roots. In the tops and roots, 37% and 26% respectively of the genotypes had an AlRY50 less than 1 M, including all species tested in the genera Melilotus and Medicago. Only 8% or 23% of the genotypes, based on the tops and roots respectively, had an AlRY50 greater than 2, including all genotypes in the species Lotus pedunculatus. Except for Lotus, there were no consistent differences between genera in plant chemical concentrations. In Lotus, concentrations of Ca, Zn, Mn and Cu in the tops and of all elements except B in the roots were lower than that of the other species. The AlRY50 of the species was not related to plant chemical concentrations in the absence of Al. Depending on the plant element, increasing solution Al concentrations had no significant effect on plant chemical concentrations for 56–94% of the species. When a significant effect did occur, increasing Al in solution generally decreased S and K concentrations and increased Mn, Zn, Cu Fe, B and Al concentrations in the tops and roots and decreased Ca concentrations in the tops. Plant P concentrations decreased in the tops but increased in the roots. Increasing Al in solution increase plant Al at the average rate of 44 g g-1 M -1 (range 20–87) in the tops and 333 g M -1 (range 162–616) in the roots.  相似文献

In vitro plant regeneration was achieved from eightsweet pepper varieties (Capsicum annuum L.). The effect ofvarious explant types (cotyledons, leaves, cotyledonary nodes and shoot-tip from25-day-old seedlings and embryonic cotyledons, embryonic hypocotyls and woundedseedlings) on bud and shoot regeneration and shoot elongation was evaluated.Differences in ability for in vitro shoot regeneration andelongation depended on the variety and explant type. In general, highregeneration frequency was obtained from all varieties. Agridulcedisplayed the highest regeneration response: an average of 3.45 elongated shootsper explant using embryonic cotyledons. Elongated shoots were excised and rootedon Murashige and Skoog (MS) basal medium either without plant growth regulatorsor with 0.5 IBA (indole-3-butyric acid) or 0.05 NAA (-naphthaleneacetic acid). Plantlets weretransplanted to soil and acclimatised in the greenhouse showing normaldevelopment and growing to maturity bearing normal fruits with seeds.  相似文献

Concentrations of ions were measured in soil solutions from beech (Nothofagus) forests in remote areas of New Zealand and in solutions from beech (Fagus sylvatica) and Norway spruce (Picea abies) forests in North-East Bavaria, West Germany, to compare the chemistry of soil solutions which are unaffected by acid deposition (New Zealand) with those that are affected (West Germany). In New Zealand, soil solution SO4 2– concentrations ranged between <2 and 58 mol L–1, and NO3 concentrations ranged between <1 and 3 mol L–1. In West Germany, SO4 2– concentrations ranged between 80 and 700 mol L–1, and NO3 concentrations at three of six sites ranged between 39 and 3750 mol L–1, but was not detected at the remaining three sites. At all sites in New Zealand, and at sites where the soil base status was moderately high in West Germany, pH levels increased, and total Al (Alt) and inorganic monomeric Al (Ali) levels decreased rapidly with increasing soil depth. In contrast, at sites on soils of low base status in West Germany, pH levels increased only slightly, and Al levels did not decline with increasing soil depth.Under a high-elevation Norway spruce stand showing severe Mg deficiency and dieback symptoms in West Germany, soil solution Mg2+ levels ranged between 20 and 60 mol L, and were only half those under a healthy stand. Alt and Ali levels were substantially higher the healthy stand than under the unhealthy stand, indicating that Al toxicity was not the main cause of spruce decline.  相似文献

Axillary shoot proliferation was induced in vitro from shoot explants of greenhouse grown candellila (Euphorbia antisyphilitica Zucc). Optimum shoot proliferation was obtained by supplementing a modified Murashige and Skoog [7] medium with 0.13 M naphthalene-acetic acid and 4.44 M 6-benzylaminopurine. Rooting occurred on 100% of shoots transferred to a medium containing half strength salts supplemented with 0.49 M indole-3-butyric acid. Fully rooted plants were transferred to potting soil and established under greenhouse conditions without special acclimatization techniques.  相似文献

This work studied the effects of P fertilization on nodulation of field-grown soybean by two Bradyrhizobium strains (SMGS1 and THA7), and checked if differences between strains were consistent with bacterial growth and growth pouch nodulation ability in response to P availability. In the field, nodule dry weight and nitrogen fixation activity of inoculated soybean were studied on typical acid soils of Thaïland at the flowering (R1) stage and at the end of grain filling. Grain yield, growth and phosphorus content were recorded. The bradyrhizobial strains were cultivated in culture medium, and growth parameters recorded. Nodulation patterns were observed during growth pouch experiments: infective root cells were inoculated with strains cultivated at two P concentrations in their culture media, namely 1 M and 1 mM. Ten days after inoculation, the position of each nodule was measured relative to the root tip (RT) mark, expressed relative to the smallest emerging root hairs-RT distance in the nodulation frequency profile, and the consistency of responses was tested. In the field, on P deficient soils, dry weight of nodules was higher with Bradyrhizobium japonicum strain SMGS1 than with strain THA7. P supply increased the number and dry weight of nodules for both strains, with a higher dry weight response for THA7 than for SMGS1. It also had a positive effect on tissue phosphorus status and grain yield at R8 stage. In growth media, significant differences were recorded between strains under P-limiting conditions: The growth rate was higher for strain SMGS1, as well as the maximal number of bacterial cells supported. With growth pouch, inoculating plants with bacteria grown in P-deficient medium resulted in a less intense nodulation of roots by THA7, and with nodules appearing earlier on roots than in the case of SMGS1. At 1 mM P, there was no significant difference between strains. Thus, strain THA7 is more affected by P deficiency than strain SMGS1. Although P was not supplied in the same way in the soil and in the growth pouch experiments, this consistency of behaviour between work scales indicates that phosphorus availability is a key component for a successful inoculation. TheN-linked carbohydrate chains of the-subunit of highly purified urinary human chorionic gonadotropin have been re-investigated. The oligosaccharides were released enzymatically by peptide-N 4-(N-acetyl--glucosaminyl)asparagine amidase-F, and fractionated by a combination of FPLC and HPLC. As a result of the application of improved fractionation methods, apart from the earlier reported carbohydrate chains, also small amounts of trisialo tri- and tri-antennary oligosaccharides were found. Summary In order to explore interrelations between S nutrition, soluble sugars, leaf area, nodulation and N2 fixation, greenhouse experiments were done with several levels of S added to perlite-sand cultures or to a moderately S-deficient soil. Sulfur had indirect effects on nodulation and N2 fixation, possibly by improving sugars supply and N metabolism.In perlite-sand culture, leaf area increased with concentrations of supplied S up to 50 and 200 M for symbiotic and N-treated plants respectively, then decreased at higher concentrations. Plant yield and total sugars content (mg per plant) for the N-treated plants behaved similar to leaf area in response to added S but in the symbiotic plants maximum values were obtained at 100 M S. In soil, Mo had no effect on growth but interacted significantly with S in affecting total sugars content. High levels of S depressed sugars content at low Mo but raised it at high Mo.Sulfur increased the N content of soil-grown plants. It increased the N content of plants grown in perlite-sand culture except at very high levels of S. There was little effect on concentration of N in the shoots. Nitrogen content correlated significantly with leaf area and sugar content, and highly significantly with S concentration in the shoots.  相似文献

Soybean plants cv. Corsoy were grown in greenhouse conditions on sterilized quartz sand. They were inoculated with Bradyrhizobium japonicum, strain 542. The plants were treated with different concentrations of quercetin (ranging from 10 nM to 1M) at regular intervals during the experiment. The experiment was terminated at flower development. The following parameters, important for symbiosis efficiency were determined: shoot, root and nodule weights, nodule number, total leghemoglobin in the nodules,total nitrogen and soluble protein concentrations in shoots and roots, as well as chlorophyll concentration in the leaves.The results obtained partly confirmed the earlier findings that quercetin inhibits nodulation since increasing quercetin concentration decreased the number of nodules. However, at very low concentrations, quercetin stimulated the number of nodules. Quercetin also exerted a stimulating influence on other characteristics of the plant and nodules which did not correlate with nodule number and quantity of N fixed. These are: nodule weight, leghemoglobin concentration, total soluble protein content in shoots and roots as well as shoot and root weight.  相似文献

Interactions among shoots within plant modules could allow gall-insects to acquire resources from other plant parts near the feeding sites. As a result, nearby plant parts may act as a functional resource, or extended resource base. We tested for functional interconections between galls and adjacent ungalled shoots in Adelges cooleyi Gil. (Homoptera: Adelgidae) on Picea engelmanni, Engelmann spruce. Observations of gall and surrounding shoot weights showed that gall weights were twice normal shoot weights, but that surrounding shoot weights were unaffected. Reducing photosynthate availability by covering galls or surrounding shoots with opaque cloth suggested that functional interconnections exist among them; covering galls reduced surrounding current-year ungalled shoot weight, and covering surrounding shoots reduced their weight even further, but neither covering treatment affected gall or gall-insect weight. These patterns suggest that surrounding shoots constitute an extended and flexibly utilized resource base for adelgid galls. Resources made available via functional interconnections appeared to be equally available throughout adelgid galls. No differences were found in gall-insect performance in parts of the gall closer to connections with surrounding shoots compared to more distantly-located gall-insects. Further studies are required to examine patterns of resource movement among unmanipulated galls and shoots. Functional resources may be common features of plant/gall-insect interactions, potentially playing an important role in gall-insect reproductive success and habitat selection.  相似文献

Summary Juvenile plants ofPlantago lanceolata andP. major ssp.major were grown in a flowing solution system at 7.5 mM or 9.5 M NO3. The parameters investigated were: RGR, shoot weight percentage, leaf length, length of main root axis, shoot concentrations of major ions and organic N, and the specific uptake rate for NO3. At 9.5 M NO3 growth ofP. major was not hampered, whereas shoot growth and leaf length ofP. lanceolata were reduced. The NO3 concentration ofP. lanceolata decreased more than that ofP. major. The different performances of the species at 9.5 M NO3 were associated with different specific uptake rates. In both treatments the root system ofP. major was shorter than that ofP. lanceolata. P. lanceolata accumulated more NO3 in the leaves. The performance of thePlantago species is discussed in relation to the availability of nutrients in their habitats.Grassland Species Research Group. A factorial experiment was conducted to determine the effect of aluminium (0 and 600M) and media (sand, and 1:1 sand:soil) on mycorrhizal (M) and non-mycorrhizal (NM) highbush blueberry plantlets. There were no differences in nutrient uptake and total plant dry weight between M and NM plantlets. However, more root growth, as determined by dry weight, was observed in M than NM plantlets. The plantlets growing in sand had more dry weight than did those in the soil medium. Although the root growth and shoot growth were reduced by the 600M Al treatment, the direct effect of Al on plantlet growth was not clear due to Al and P interactions. Plant nutrient uptake was reduced by high concentrations of Al, suggesting that high Al concentration limited the ability of roots to acquire most of the nutrients. Mycorrhizal cortical cell infection levels of 15–20% wene maintained in the roots in soil medium but decreased to about 5% over the 6 weeks of the experiment in the sand medium. Although M plantlets accumulated more Al in their roots, Al was readily transported to the leaf tissues of M and NM plantlets.  相似文献

Summary Soybean (Glycine max {L.} Merr.) cultivars were inoculated withGigaspora gigantea andGlomus mosseae to determine mycorrhizal: cultivar relationships as affected by soil pH. The specific cultivarfungal response was dependent on soil pH. Overall cultivar responses in unlimed soil (pH 5.1) were greater forG. gigantea thanG. mosseae. The Bossier —G. gigantea combination was particularly responsive in unlimed soil and showed an increase of 10% in shoot length, 35% in shoot dry weight. 75% in root dry weight, and 397% in nodule dry weight over uninoculated controls. Little cultivar response was observed withG. mosseae inoculation in unlimed soil. In limed soil (pH 6.2), the larger responses were obtained withG. mosseae inoculated plants, although inoculation with eitherG. mosseae orG. gigantea appeared effective. In general, nodulation was greater on mycorrhizal roots than on control roots.  相似文献

Adventitious shoot regeneration was compared among leaf, stem and petal explants of carnation (Dianthus caryophyllus L.) cv. Scania on MS medium containing different concentrations of 6-benzyladenine (BA) and -naphthaleneacetic acid (NAA). High frequency regeneration was obtained only from petal explants on the media containing 5 to 10 M BA with or without 5 M NAA. Among the cytokinins tested, N-2-chloro-4-pyridyl-N-phenylurea and N-1,2,3-thiadiazol-5-yl-N-N-phenylurea were more effective than BA, kinetin, N6-2-isopentenyl adenine and zeatin on regeneration from petal explants. Although, high frequency shoot regeneration was obtained from all petal explants harvested from various developmental stages of buds, a significant decrease in regeneration capacity was observed in the explants obtained from fully-opened flowers. High frequency shoot regeneration was also obtained from the petal explants of cvs. Coral. We developed a novel micropropagation method for garlic (Allium sativum L.) by the combination of initial shoot-tip culture, shoot multiplication and in vitro bulblet formation. Garlic shoot-tips were cultured on LS medium containing 1 M indole-3-acetic acid (IAA) and 1 M 6-benzyladenine (BA) to regenerate proliferative shoots. These shoot-tips produced multiple shoots when transferred to modified LS medium containing 5 M 1-naphthaleneacetic acid (NAA) and 10 M BA, and cultured at 20°C under 12-h light conditions. Higher ratios of KNO3/NH4Cl in the media promoted multiple shoot formation, together with suppressing vitrification of these shoots. The proliferated shoots of early maturing cultivars produced bulblets by culture on LS growth regulator-free medium at 25°C under 16-h light. On the other hand, the late maturing cultivar, Howaito-roppen, formed bulblets after a low temperature treatment of the proliferated shoots for 6 months followed by culture on LS medium containing 6 to 12% sucrose for two months. The dormancy of the bulblets of cv. Summary Copper-deficient cells ofPseudomonas stutzeri strain ZoBell synthesize catalytically inactive nitrous oxide (N2O) reductase which is activated by added Cu(II) in the absence of de novo protein synthesis. The apparentK m for the activation process is 0.13 M. Activation is temperature-dependent and is inhibited by Cd(II)(K i 1.27 M) and less strongly by Zn(II), Ni(II), and Co(II). The same metal ions at 20 M have little or no effect on N2O reduction of intact cells. Apo-N2O reductase of transposon Tn5-inducednos mutants with defective Cu-chromophore biosynthesis is not reactivated by Cu(II). N2O reductase of Cu-sufficient and Cu-deficient wild type, and ofnos mutants is localized in the periplasm, the latter providing the likely site of metal incorporation into the apoenzyme.  相似文献

A new genus of methanogenic bacteria is described, which was isolated from a mesophilic sewage digester. It is most probably the filamentous bacterium, earlier referred to asMethanobacterium soehngenii, fat rod or acetate organism. The single non-motile, non-sporeforming cells are rod-shaped (0.8×2 m) and are normally combined end to end in long filaments, surrounded by a sheath-like structure. The filaments form characteristic bundles.Methanothrix soehngenii decarboxylates acetate, yielding methane and carbon dioxide. Other methanogenic substrates (H2–CO2, formate, methanol, methylamines) are not used for growth or methane formation. Formate is split into hydrogen and carbon dioxide. The temperature optimum for growth and methane formation is 37°C and the optimal pH range is 7.4–7.8. Sulfide and ammonia serve as sulfur and nitrogen source respectively. Oxygen completely inhibits growth and methane formation, but the bacteria do not loose their viability when exposed to high oxygen concentrations. 100 mg/l vancomycin showed no inhibition of growth and methanogenesis. No growth and methane formation was observed in the presence of: 2-bromoethanesulfonic acid, viologen dyes, chloroform, and KCN. The bacterium has a growth yield on acetate of 1.1–1.4 g biomass per mol acetate. The apparent K S of the acetate conversion system to methane and carbon dioxide is 0.7 mmol/l. The DNA base composition is 51.9 mol% guanine plus cytosine. The nameMethanothrix is proposed for this new genus of filamentous methane bacterium. The type species,Methanothrix soehngenii sp. nov., is named in honor of N. L. Söhngen.  相似文献
