【目的】苏铁(Cycassp.)是珍稀濒危树种,能在干热河谷中长期稳定生存的原因可能与珊瑚状根内生微生物有密切关系。不同种苏铁在同一生境下其珊瑚状根内生微生物种类和群落组成存在怎样的差异性是本研究的科学问题。【方法】采用宏基因测序技术对四川省攀枝花公园内5种同属不同种苏铁珊瑚状根进行了分子鉴定,分析了苏铁间微生物类型、功能基因和代谢通路过程的差异性。【结果】公园内不同苏铁的珊瑚状根内生微生物的优势类群在门水平上基本相同,但相对丰度有差异性。在真菌界水平上的优势类群为担子菌门(Basidiomycota)、子囊菌门(Ascomycota)、球囊菌门(Glomeromycota)和隐真菌门(Cryptomycota),在细菌界水平上的优势类群为蓝细菌门(Cyanobacteria)、变形菌门(Proteobacteria)、厚壁菌门(Firmicutes)、螺旋体门(Spirochaetes)和放线菌门(Actinobacteria)。不同种的苏铁在真菌界和细菌界的微生物群落相对丰度存在一定差异。蓝细菌门在篦齿苏铁、攀枝花苏铁、华南苏铁和贵州苏铁中的相对丰度远高于宽叶苏铁,而放线菌门和球囊...  相似文献

This paper describes the frequency of occurrence and biodiversity of fungi from mangroves of the Godavari and Krishna deltas, on the east coast of India. Seventy three species were identified from Godavari and 67 from the Krishna mangroves. Fifty five species were common to both sites, 18 were found only at Godavari and 12 at Krishna mangroves. Verruculina enaliawas found to be very frequent at both sites with a higher frequency of occurrence at Godavari. Eutypa bathurstensis was very frequent at Godavari but only frequent at Krishna. Cirrenalia pygmea and Cryptosphaeria mangrovei were frequent at the Godavari mangrove, but were recorded occasionally at Krishna. Decaying samples of Rhizophora and Avicennia were studied in detail. Forty three species were common to both hosts, while 22 species were recorded only from Avicennia and 20 only from Rhizophora. Verruculina enalia was the only very frequent fungus recorded on both hosts with a lower percentage occurrence (14.8%) on R. apiculata as compared to Avicennia spp. (24.3%). Eutypa bathurstensis was the next most frequent fungus on Avicennia, while Rhizophila marina was next most frequent on Rhizophora. Desmids from 12 freshwater resources in the northern part of Thailand were investigated during 2002 to 2003. A total of 91 taxa were found. They belonged to 17 genera: Actinotaenium, Spirotaenia, Netrium, Gonatozygon, Pleurotaenium, Closterium, Euastrum, Micrasterias, Cosmarium, Cosmocladium, Stuarastrum, Staurodesmus, Xanthidium, Teilingia, Spondylosium, Hyalotheca and Desmidium. The water qualities in all the water resources were classified as oligotrophic to meso-eutrophic by trophic status. The taxa that could possibly be used as bioindicators of trophic state were Staurastrum gutwinskii, Spondylosium pandurifoemae, Cosmarium capitulum, C. mediosrobiculatum var. egranutum, S. tortum, Closterium gracile var. elongatum, C. kuetzingii and Closterium dianae var. dianae. The most frequently found taxa were Staurastrum limneticum var. burmense, S. tetracerum var. tetraerum, Pleurotaenium trabecula, Closterium ehrenbergii var. ehrenbergii and C. kuetzingii. The rare taxa in this study were Actinotaenium sp. Spirotaenia condensata, Pleurotaenium burmense var. dacchense and Micrasterias apiculata. Cross-compatibility relationships in almond are controlled by a gametophytically expressed incompatibility system partly mediated by stylar RNases, of which 29 have been reported. To resolve possible synonyms and to provide data for phylogenetic analysis, 21 almond S-RNase alleles were cloned and sequenced from SP (signal peptide region) or C1 (first conserved region) to C5, except for the S 29 allele, which could be cloned only from SP to C1. Nineteen sequences (S 4 , S 6 , S 11 S 22 , S 25 S 29 ) were potentially new whereas S 10 and S 24 had previously been published but with different labels. The sequences for S 16 and S 17 were identical to that for S 1 , published previously; likewise, S 15 was identical to S 5 . In addition, S 4 and S 20 were identical, as were S 13 and S 19 . A revised version of the standard table of almond incompatibility genotypes is presented. Several alleles had AT or GA tandem repeats in their introns. Sequences of the 23 distinct newly cloned or already published alleles were aligned. Sliding windows analysis of Ka/Ks identified regions where positive selection may operate; in contrast to the Maloideae, most of the region from the beginning of C3 to the beginning of RC4 appeared not to be under positive selection. Phylogenetic analysis indicated four pairs of alleles had ‘bootstrap’ support > 80%: S 5 /S 10 , S 4 /S 8, S 11 /S 24 , and S 3 /S 6 . Various motifs up to 19 residues long occurred in at least two alleles, and their distributions were consistent with intragenic recombination, as were separate phylogenetic analyses of the 5′ and 3′ sections. Five mutants of Escherichia coli K12 (lam 24, lam 25, lam 26, lam 27 and lam 646) that block head formation of λ are described. In vitro complementation tests and electron microscopy demonstrated that in these bacteria phage tails were produced normally, whereas head formation was abnormal, aberrant head-related structures being produced. In lysates prepared from lam 24, lam 25 and lam 26, monsters and empty heads without tail were the predominant structures, whereas in lysates from lam 27 and lam 646, petit λ and empty heads were the most common structures. The five lam mutations were located in two regions on the bacterial chromosome; lam 24, lam 25 and lam 26 were near the dnaB gene and lam 27 and lam 646 near the lac gene. It was suggested that the former three mutants are new isolates that belong to GroE mutants, whereas the latter two comprise a new group of mutants. Analyses of phage mutants (ov mutants) that overcome the interference by the lam 646 mutation revealed that this mutation blocks normal expression of the gene E of λ.  相似文献

Self-incompatibility (SI) is reported to play a key role in the evolution of species as it promotes their outcrossing through the recognition and rejection of self-pollen grains. In Brassica, two S-locus genes expressed in the stigma, S-locus glycoprotein (SLG) gene and S-locus receptor kinase (SRK) gene, and one expressed in the pollen, S-locus protein 11 (SP11) gene, were linked as an S haplotype. In order to analyze the evolutionary relationships of S haplotypes in Brassica, a total of 39 SRK, 37 SLG, and 58 SP11 sequences of Brassica oleracea, Brassica rapa and Brassica napus were aligned. Two phylogenetic trees with similar pattern were constructed based on the nucleotide sequences of SRK/SLG and SP11, respectively. Class I and class II alleles were clustered into two distinct groups, and alleles from different species, including all the interspecific pairs of S haplotypes, were closely related to each other. The S-locus genes identified in B. napus were intermingled in phylogenetic trees. All these observations showed that class I and class II S haplotypes diverged ahead of the species differentiation in Brassica. The evolution and the genetic diversity of S haplotypes in Brassica were discussed. Moreover, the relationships between S haplotypes and SI phenotypes in Brassica, especially in B. napus, were also discussed.  相似文献

 A new approach for selecting sampling sites of ectomycorrhizal roots is presented in order to describe ectomycorrhizas of Salix herbacea. Based on sporocarp mapping and statistical evaluation of the mapping data, sites for ectomycorrhizal root sampling were chosen underneath sporocarps of ectomycorrhizal Cortinarius (Myxacium) favrei, Hebeloma repandum, Laccaria montana, Entoloma alpicola, and Russula norvegica. Only in the samples beneath C. favrei, E. alpicola, and L. montana were corresponding ectomycorrhizas predominant and therefore described. Cenococcum geophilum ectomycorrhizas occurred throughout most samples and were also described. Numerous carpophores of the five selected ectomycorrhizal fungi were sampled for isolation purposes. Pure cultures were obtained of H. repandum and C. favrei, but laboratory syntheses of ectomycorrhizas were successful only with H. repandum and seedlings or cuttings of S. herbacea. 为探究适用于马鞭草科植物的DNA条形码及该类群的系统分类关系,对豆腐柴(Premna microphylla)的叶绿体基因ycf6-psbM、trnV-atpE、rbcL、trnL-F、psbM-trnD、atpB-rbcL、trnC-ycf6、trnH-psbA、rpl36-infA-rps8和核基因ITS序列进行了PCR扩增和测序,结果表明仅rbcL、trnl-F、trnH-psbA序列的PCR扩增以及测序效果较好,而ITS不能得到明显的扩增条带,ycf6-psbM不能成功测序,其它序列存在有部分双峰或噪值高等问题。根据DNA条形码标准,rbcL序列是所有测试条码中相对最适合的。应用rbcL和matK序列对马鞭草科(Verbenaceae)豆腐柴属、牡荆属(Vitex L.)、马鞭草属(Verbena L.)和大青属(Clerodendrum L.)等4属与唇形科宝盖草属(Lamium L.)、水苏属(Stachys L.)、鼠尾草属(Salvia L.)和香科科属(Teucrium L.)等4属的分类和系统发育关系进行分析,以紫草科Lithospermum multiflorum L.为外群,最大简约法对2个片段的单独和联合矩阵分别构建系统发育树。豆腐柴属和大青属应从马鞭草科划入唇形科,马鞭草属仍归于马鞭草科,而牡荆属的系统学位置还需更多的证据。  相似文献

为揭示固氮树种土壤养分转化的酶学机制,对固氮树种[厚荚相思(Acacia crassicarpa)、黑木相思(A. melanoxylon)、卷荚相思(A.cincinnata)、大叶相思(A.auriculiformis)和马占相思(A.mangium)]及非固氮树种尾巨桉(Eucalyptusurophylla×E.grandis)人工林的土壤养分含量、酶活性及其相关性进行研究。结果表明,相思林40~60cm土层的pH均高于尾巨桉林;5种相思林土壤各土层的TP、TK含量均低于尾巨桉林,而20~40 cm土层的TC、TN含量均高于尾巨桉林,黑木相思林和马占相思林各土层的有效养分均显著高于尾巨桉林(P0.05)。0~10 cm土层中,相思林的土壤酸性磷酸酶和纤维素酶活性均高于尾巨桉林,大叶相思林的土壤脲酶、蔗糖酶、纤维素酶和芳基硫酸酯酶活性显著高于尾巨桉林(P0.05),卷荚相思林的土壤脲酶、纤维素酶、几丁质酶和淀粉酶活性显著高于尾巨桉林(P0.05)。相关分析结果表明,土壤脲酶、蔗糖酶和几丁质酶活性与AP显著负相关(P0.05),蔗糖酶和纤维素酶活性与NH4+-N显著负相关(P 0.05),脲酶、纤维素酶、芳基硫酸酯酶与土壤TK显著负相关(P0.05),几丁质酶活性与TN含量呈显著正相关(P0.05),土壤淀粉酶活性与NH4+-N呈显著正相关(P 0.05),过氧化氢酶活性与土壤TK含量呈显著正相关。可见,与尾巨桉人工林相比,在我国南方退化山地引种相思树可提高土壤关键酶的活性,对土壤有效养分具有明显改良作用,有利于退化地土壤的生态修复及人工林长期生产力的维持。  相似文献

林火是森林生态系统的一种主要干扰因子,以青岛市三标山林火迹地为研究对象,采集荆条(Vitex negundo)、胡枝子(Lespedeza bicolor)、花木蓝(Indigofera kirilowii)、青花椒(Zanthoxylum schinifolium)和野青茅(Deyeuxia arundinacea)5种优势植物根围土壤,研究不同林火强度对丛枝菌根(AM)真菌多样性的影响。结果表明,AM真菌侵染率和孢子密度随火灾强度的加强而降低;非过火区植物根围土壤中,分离鉴定出AM真菌3属11种,轻度过火区分离鉴定出AM真菌3属10种,中度过火区分离鉴定出AM真菌3属9种,重度过火区分离鉴定出AM真菌3属8种。过火区AM真菌种丰度低于非过火区。过火区和非过火区AM真菌的重要值和优势种不同,非过火区植物根围的优势种是地球囊霉(Glomus geosporum)、台湾球囊霉(G.taiwanensis)、分支巨孢囊霉(Gigaspora ramisporophora)、极大巨孢囊霉(Gi.gigantean)、福摩萨球囊霉(G.formosanum)、悬钩子球囊霉(G.rubiforme)、柯氏无梗囊霉(Acaulospora koskei)和松蜜无梗囊霉(A.thomii);轻度过火区植物根围的优势种是地球囊霉和台湾球囊霉;中度过火区的是台湾球囊霉和地球囊霉(野青茅除外);重度过火区植物根围的优势种是地球囊霉。不同强度的过火区对AM真菌群落组成有不同程度的影响。认为林火降低植物根围土壤中AM真菌多样性。  相似文献

对我国古老特有植物青檀叶片进行内生和附生真菌的研究,以了解青檀叶片内生和附生真菌的组成特点和探讨内生和附生真菌菌群之间的可能联系,为研究真菌资源多样性、植物附生和内生真菌的相互演化关系及真菌与宿主植物协同进化等提供有益参考资料。研究结果表明,从健康的青檀叶片获得可培养内生真菌839株,附生真菌1857株,共计2696株,鉴定其分属于4目,5科,43属。在目的分类水平上,内生和附生真菌均以丛梗孢目Moniliales为优势菌群,分别占90.23%和92.51%;在科的水平上,内生真菌以暗梗孢科Dematiaceae和丛梗孢科Moniliaceae为优势菌群,分别占47.56%和42.67%,附生真菌以丛梗孢科Moniliaceae和暗梗孢科Dematiaceae为优势菌群,分别占67.04%和25.47%;在属的水平上,内生真菌以黑团孢属Periconia和青霉属Penicillium为优势菌群,分别占31.47%和10.73%,附生真菌以小球霉属Glomerularia、膝葡孢属Gonatobotrys和青霉属Penicillium为优势菌群,分别占20.03%、13.95%和12.22%。青檀叶片内生真菌和附生真菌均存在的菌群数量达到23个属,占53.49%。内生真菌特有的属有6个,共分离19株,占0.70%,附生真菌特有的属有14个,共分离120株,占4.45%。内生真菌的Shannon-Wiener index(H')多样性指数(2.44)和Margalef index(R)丰富度指数(2.88)分别小于附生真菌ShannonWiener index(H')多样性指数(2.57)和Margalef index(R)丰富度指数(3.32),但两者的Evenness index(E)均匀度指数几乎相等。青檀叶片内生和附生真菌菌群组成具有较高的相似性,相似性系数达0.70。通过Fisher's exact test分析表明青檀叶片内生和附生真菌菌群组成无明显差异(P=0.072)。  相似文献

Summary We isolated strains of Escherichia coli K 12 in which the lac structural genes were fused to the structural genes of the guaBA operon. These strains were used to isolate regulatory mutations that increased the expression of the guaBA operon under normal repressing conditions as compared to the wild type parental fusion strain. Three classes of guaBA specific regulatory mutations were identified. Class I regulatory mutations were trans-acting and unlinked to the guaBA operon as shown by bacteriophage P1 transduction. Class II regulatory mutations were tightly linked to the guaBA operon, cis-dominant to the wild type allele in a cis-trans analysis and were regarded as control region mutations. Class III regulatory mutations were tightly linked to the guaBA operon and trans-recessive to the wild type allele in a cis-trans analysis. We have designated the locus responsible for the class III regulatory mutations as guaR. The guaR locus is tightly linked and was mapped to the counterclockwise side of the guaBA operon. The guaR locus is proposed to specify a trans acting regulatory element involved in the regulation of the guaBA operon.  相似文献

Summary The histidine utilization (hut) operons of Klebsiella aerogenes were cloned into pBR322. The hut genes are wholly contained on a 7.9 kilobase pair fragment bounded by HindIII restriction sites and expression of hut is independent of the orientation of the fragment with respect to pBR322. A restriction map locating the 27 cleavage sites within hut for the enzymes, HindIII, PvuII, SalI, BglII, KpnI, PstI, SmaI, AvaI, and BamHI was deduced. Several of the cleavage sites for the enzymes HaeIII and HinfI were also mapped. A set of deletion plasmids was isolated by removing various restriction fragments from the original plasmid. These deletions were characterized and were used to assist in mapping restriction sites. This physical characterization of hut DNA opens the way for genetic and molecular analysis of the regulation of hut gene expression in vitro as well as in vivo.  相似文献

为探究岩溶植物的光合生理适应机制,采用Li-6400XT便携式光合作用测量系统,对广西平果市岩溶区8种适生植物的叶片净光合速率(Pn)、气孔导度(Gs)、胞间CO2浓度(Ci)、蒸腾速率(Tr)、水分利用效率(WUE)和气孔限制值(Ls)等光合特征参数进行了测定分析。结果表明:(1)6个光合特征参数在种内和种间均存在不同程度的变异,并且种内变异均大于种间变异。(2)Gs和Tr的变化主要来源于种间变异(46.72%~49.76%),而Pn、Ci、WUE和Ls变化主要来源于种内变异(48.66%~64.50%)。在生活型水平上,Pn、Gs和Tr的种内变异表现为常绿植物小于落叶植物,而Ci、WUE和Ls则相反。(3)各参数的种间变异均表现为落叶植...  相似文献

The oviposition preferences of Oscinella frit, O. vastator, O. nitidissima, O. albiseta andO. nigerrima for differenct Gramineae were investigated in the laboratory. O. frit, laid most eggs on oats, Lolium multiflorum and Festuca rubra, relatively few eggs were laid on barley and virtually none on Dactylis glomerata None of the other Osinella species oviposted on cereals. Of the other species, O. vastator appeared to be the most polyphagous and the preferred hosts were L. multiflorum, Lolium perenne and Festuca pratensis; few eggs were laid on either F. rubra, Agrostis tenuis or Poa pratensis. The host ranges of the remaining species were much more limited. Although a few eggs were laid by O. nitidissima on Lolium, the preferred host was A. tenuis. Oscinella albiseta oviposited only on D. glomerata whilst nearly all the eggs laid by O. nigerrima were deposited on shoots of Arrhenatherum elatius. The distribution of eggs by O. frit on L. multiflorum and A. tenuis was different from that on oats; on grasses most eggs were laid inside withered leaf sheaths whereas on oats most were deposited inside the coleoptile. The oviposition sites of O. nitidissima, O. nigerrima and O. albiseta on their respective host grasses were similar to those of O. frit on grasses. O. frit laid most eggs on grasses which were at the five-leaf stage and tailoring.  相似文献

The major histocompatibility (B) complex of a distinct commercial pure White Leghorn chicken line was characterized using serological, biochemical and restriction fragment length polymorphism (RFLP) typing. Line B chickens displayed a high recombination frequency within the B complex. Three recombinant haplo-types were identified. The influence of these haplotypes was determined in relation to the haplotypes Bl9 and B21 on their resistance to Marek's disease (MD) in an experimental infection with the virus. Offspring of sires with a recombinant haplotype in combination with B19 or B21, and dams, which were homozygous B19/B19 or B21/B21 were infected. The B type of the offspring had a significant effect upon survival. Animals with B complex types B21/B21, B134/B21 and B234/B21 were relatively resistant to MD (24–32% mortality), whereas B19/B19 birds were highly susceptible (68% mortality). Animals with a recombinant halpotype B19r21 (B-G21, B-F19) were equally susceptible to MD as birds with the complete B19 haplotype. In contrast to earlier publications, resistance was not inherited as a dominant trait. Apparently, B19 was associated with a dominant susceptibility. The gene(s) associated with the B complex and involved in resistance to MD were localized within the B-F/B-L region. However, the association with a presumably non-coding subregion of B-G could not be excluded.  相似文献

A quantitative trait locus (QTL) analysis was performed on the size and shape of the mandible in F2 mice between KK-A y and C57BL/6 J strains and the effect of the A y allele on the morphology of the mandible was analyzed. A total of 13 measurements were taken on each right mandible. By means of discriminant and canonical discriminant analyses, KK-A y males and KK males were exactly discriminated from each other. In contrast to its known effects on body weight, the A y allele reduced the overall size of the mandible. QTL analysis of the 13 measurements and on three principal components extracted from these measurements identified multiple QTLs. When F2 a/a and F2 A y /a were analyzed separately, 11 significant main-effect QTLs were identified in F2 a/a, whereas only two QTLs were identified in F2 A y /a. Although four significant interactions were identified, all were in F2 a/a. The A y allele thus made the difference in the size and shape of the mandible between strains obscure. Among mandible QTLs, those on Chrs 5 (Mssq6 and Mssq7) and 15 (Mssq14) were important. Mssq6 had an effect on the height of the posterior mandible. Mssq7 had an effect on mandible length. Mssq14 had an effect on the height of the anterior and posterior mandible. Mssq7 and Mssq14 also had an effect on the overall size. Thus, mandible QTLs have distinct and characteristic sites of action. Therefore, mandible morphology will be determined largely by the combination of these QTLs.  相似文献

Staphylococcus aureus is one of the major causes of mastitis in dairy animals and its resistance against multiple antimicrobials always remains crucial concern. Present investigation was carried out to detect the distribution of antibiotic-resistant genes of S. aureus isolates. Isolates (128) of S. aureus from mastitic milk were collected, tested for antibiotics with disc-diffusion method, and resistant genes mecA, linA, msrA msrB, vatA, vatB, vatC ermA, ermC tetK, tetM and aacA-D were detected by PCR. The phenotypic antibiotics resistance percent in S. aureus isolates was classified as tetracycline (36.7), gentamycin (30.5), streptomycin (26.6), kanamycin (25.8) and penicillin G (22.7). All the isolates were susceptible to vancomycin. Among isolates, 10.2% were observed as methicillin-resistant. The distribution of antibiotic-resistant genes was linA (51.6) followed by msrB (46.1), tetK + M (34.4), msrA and aacA-D (26.6%). Different antibiotic-resistant genes combinations (mecA/linA-2; mecA/aacA-D/tetK/linA/msrB-3; mecA/linA/msrA/msrB-3; aacA-D/linA/msrA/msrB-4; aacA-D/linA/msrB-7; linA/msrA/msrB-10; tetK/linA/msrA/msrB-11; aacA/tetK/linA/msrB-12 isolates) were observed. All the isolates lacked amplification of vatA, vatB, ermA and ermC genes. Molecular typing resulted genetic variation in protein A (6–12 repeats) and coagulase genes (A–E patterns) were observed. Coagulase A and D genotypes were more prevalent in antibiotic-resistant isolates, while E, B and C in susceptible ones. The significant observation was the prevalence of methicillin-resistant S. aureus, which were resistant to multiple antibiotics. Findings revealed the status of resistant isolates in herd that might be helpful in treatment, controlling of resistant strains and culling of cows for mastitis reduction.  相似文献

During a one year period, 944 dogs from the Municipal kennel of Barcelona were examined to detect animals with suspected dermatophytosis. Only a few animals (1.8%) presented skin lesions but none of them had dermatophytosis. A representative number of dogs without visible skin lesions (n=172), selected at random, were used to carry out a seasonal study of the mycobiota of their fur. Fifteen isolates belonging to the genera Microsporum and Trichophyton were isolated from 14 of the 172 (8.1%) dogs without lesions. The identity of these fungi was Microsporum gypseum (6/15), Trichophyton terrestre (4/15), M. canis (2/15), M. cookei (2/15) and Trichophyton ajelloi (1/15) (one strain each of M. gypseum and T. ajelloi were isolated from one dog). Species of Penicillium (% prevalence=89.5%), Alternaria (86.6%), Cladosporium (84.9%), Aspergillus (77.3%), Scopulariopsis (65.7%) and Chrysosporium (64.5%) were the most prevalent. No significant differences in the fungal biota were observed with respect to age, gender, hair length or between mixed and pure breed dogs. A large number of isolates, including species belonging to the genera Beauveria, Chrysosporium, Malbranchea and Scopulariopsis, that macroscopically and/or microscopically resemble dermatophytes and may be mistaken for them, produced a red color change in Dermatophyte Test Medium. No significant seasonal difference was detected among the isolates belonging to the the most frequently encountered genera, with the exception of Scopulariopsis (higher in summer and autumn) and Chrysosporium (higher in summer). Species from other genera, with lower occurrence also presented significant differences in their seasonal distribution. Arthrinium, Aureobasidium, Chaetomium and Phoma spp. presented maximum prevalence peaks in spring, Fusarium, Paecilomyces, Phoma and Rhizopus spp. in summer and Geotrichum and Mucor spp. in autumn. The Microsporum and Trichophyton species were more frequently isolated in summer.  相似文献
