Sensitivity to photoinhibition was assessed in sorghum infectedwith the angiosperm root parasite Striga her-monthica and inuninfected sorghum plants, at four times during the developmentof the host-parasite association. Photoinhibition was inducedby exposing either leaf discs or intact leaves to a photosyntheticphoton flux density of 2000 µmol m–2 s–1 for4 h. The inhibition of apparent quantum yield (a) and photosynthesisin high light (A1500) were assessed in leaf discs using an oxygenelectrode and the recovery of these from photoinhibition wasfollowed in intact leaves using an infra-red gas analyser. Fromsoon after attachment of the parasite, infected sorghum plantshad a lower A1500. During the period when Striga induced a loweringof A1500, a was more sensitive to photoinhibition in Striga-infectedplants. However, at the same time, the high-light-induced inhibitionof A1500 was similar in Striga-infected and uninfected plants.Recovery of both a and A1500 was incomplete after 6 h and thetime-course of recovery was similar in Striga-infected and uninfectedplants. The results indicate that Striga-infected plants weremore sensitive to photoinhibition and that photoinhibition wasprimarily due to damage to electron transport/photo-phosphorylationand not disablement of the recovery processes. Striga hermonthica (Del.) Benth. is an obligate hemiparasiticangiosperm which can cause severe losses of yield in cerealcrops in the semi-arid tropics. The effects of this parasiteon the growth and stomatal conductance of three varieties ofmaize (Zea mays L.) during the first 6 weeks of the associationhave been studied. From 24 d after planting (DAP), infectedplants were significantly shorter than uninfected controls.When the plants were harvested 45 DAP, infected plants had fewerfully expanded leaves, less leaf biomass and less pseudo-stembiomass than uninfected controls. However, the parasitized plantshad more root biomass and hence a higher root:shoot ratio thanuninfected controls. The stomatal conductance of infected hostswas severely inhibited by comparison with that in uninfectedplants. The possibility that abscisic acid (ABA) may be involved inthe regulation of the parasitic association was investigated.ABA concentrations in leaf tissue of maize (cv. Cargimontana)and S. hermonthica were determined by radioimmunoassay. Whilethere was a difference between cultivars in the extent of theresponse, the concentrations of ABA were significantly higherin infected maize plants than in the uninfected controls. InS. hermonthica, leaf tissue ABA concentration was found to bean order of magnitude higher than in the host leaf tissue. Detachedleaves of S. hermonthica which were dehydrated at room temperatureuntil they had lost 10–20% of their fresh weight containedthree times the ABA concentration of control leaves. This suggeststhat leaves of S. hermonthica can synthesize or re-mobilizeABA in response to water deficit. It is not yet known whetherthis contributes to the higher concentration in infected hosts,but the results suggest that ABA has a role in this parasiticassociation. The uptake and partitioning of nitrogen (N) by maize infectedwith the parasitic angiosperm,Striga hermonthicawas investigatedin sand culture in a glasshouse. The purpose was to determinethe effect ofStrigaon N uptake and partitioning in maize. Maizewas grown at 22, 66 and 133 mg N per plant and sampled fivetimes. There was no significantStrigaxN interaction in any measuredresponse. Leaf dry matter ofStriga-infected maize, averagedover all N treatments, was 92% that of uninfected maize at thefour-leaf stage but by the 18-leaf stage it had decreased to58%. Similarly, stem dry matter of infected maize which was91% that of uninfected maize at the four-leaf stage was only42% at the 18-leaf stage. Root dry matter was similar for infectedand uninfected maize. N concentration in the leaf, stem androot declined asymptotically from the first to the last samplingdate for both infected and uninfected maize. The asymptoticvalue of N concentration inStriga-infected maize was 16% greaterin the leaf, 55% in the stem, and 21% in the root than in uninfectedmaize. The concentration of N inStrigawas higher than in maizeat the 16- and 18-leaf stages. Uptake of N was similar for infectedand uninfected plants at the four–eight leaf stage butat the eight–12 leaf stage, N uptake by infected maizewas 52% that of uninfected maize. However, the proportion oftotal plant nitrogen partitioned to the root was greater (P<0.001)forStriga-infected maize. 1-Aminocyclopropane-1-carboxylate (ACC) oxidase (ethylene-formingenzyme) was isolated from wounded mesocarp tissue of Cucurbitamaxima (winter squash) fruit, and its enzymatic properties wereinvestigated. The enzyme required Fe2+ and ascorbate for itsactivity as well as ACC and O2 as substrates. The in vitro enzymeactivity was enhanced by CO2. The apparent Km value for ACCwas 175 µM under atmospheric conditions. The enzyme activitywas inhibited by sulfhydryl inhibitors and divalent cationssuch as Co2+, Cu2+, and Zn2+. ACC oxidase activity was induced at a rapid rate by woundingin parallel with an increase in the rate of ethylene production.The exposure of excised discs of mesocarp to 2,5-norbornadiene(NBD),an inhibitor of ethylene action, strongly suppressed inductionof the enzyme, and the application of ethylene significantlyaccelerated the induction of the activity of ACC oxidase inthe wounded mesocarp tissue. Fourth leaves of healthy leek plants exhibit very low ratesof net photosynthesis which appear to be related to the veryslow growth rates of young leeks early in the season. A stimulation in the rate of net photosynthesis was found tooccur in uninfected leaves of leek plants infected with therust, Puccinia porri. This was accompanied by a reduction inthe rate of photorespiration. Although the mechanisms underlyingthis response are not known, it may be instructive to comparethese results with those obtained from partial defoliation experiments.Increased photosynthetic activity in uninfected leaves may enablethe infected plant to maintain a functional shoot: root equilibrium. Kinetin retarded the decrease in chlorophyll content in leafdiscs from 5 species of plants with amphistomatous leaves, wherethe upper surface was exposed to air, but not in Rumex acetosera.When leaf discs were floated so that the lower surface was exposed,the effect of kinetin was less evident. Kinetin also stimulatedtranspiration in leaf discs from Nicotiana tabacum (amphistomatous),but not in leaf discs from Paederia chinensis (hypostomatous).Nor kinetin did retard chlorophyll breakdown in this specieswhen leaf discs were floated so that the stomatal surface wasin contact with the solution. The ineffectiveness of cytokininsin chlorophyll retention in leaf discs from hypostomatous leaveswas not due to reduced uptake of benzylaminopurine-14C. Chlorophyll retention was severely inhibited by coating theleaf surface with vaseline either in the presence or absenceof kinetin. Leaf discs floated on a solution exposed to CO2-lessair retained more chlorophyll than those in normal air. Thereis thus a close relationship between stomatal opening (as measuredby stimulation of transpiration) and chlorophyll retention,as influenced by cytokinins. Maize seedlings were grown in pots either with or without preconditionedseeds of the parasitic weed, Striga hermonthica. After between4 and 8 weeks, net photosynthesis in the leaves of maize plantsinfected with Striga decreased compared to leaves of uninfectedcontrol plants. The activities of four enzymes of photosyntheticmetabolism were, however, little affected by infection. A pulse-chaseexperiment using 14CO2 showed that C4 acids were the main earlyproducts of assimilation even when the rate of photosynthesiswas much decreased by infection, but more radio-activity appearedin glycine and serine than in leaves of healthy maize plants.Leaves of infected maize required longer to reach a steady rateof photosynthesis upon enclosure in a leaf chamber than leavesof uninfected plants after similar treatment. Electron microscopy of transverse sections of the leaves ofinfected maize indicated that the cell walls in the bundle sheathand vascular tissue were less robust than in leaves of healthyplants. The results suggest that infection with Striga causesan increase in the permeability of cell walls in the bundlesheath, leakage of CO2 from the bundle sheath cells and decreasedeffectiveness of C4 photosynthesis in host leaves. Single plants of white clover (Trifolium repens) were establishedfrom stolon cuttings rooted in acid-washed silver sand. Allplants were inoculated with Rhizobium trifolii, and receivednutrient solution containing 0·5 mg 15N as either ammoniumor nitrate weekly for 12 weeks (i.e. 6 mg 15N in total). Plantswere then leniently defoliated or left intact, and the labelledN supply was replaced with unlabelled N. Lenient defoliationremoved fully expanded leaves only, leaving immature leaveswhich accounted for 50–55% of the total; growing pointnumbers were not reduced. Nodules, leaves and growing pointswere counted over the following 21 d period, and d. wts, N contents,and 15N enrichments of individual plant organs were determined. Defoliated plants had fewer nodules, but numbers of growingpoints were unaffected by defoliation. The rates of both leafemergence and expansion were accelerated in defoliated plants;in consequence the number of young leaves remained less thanin intact plants until day 21. Total dry matter (DM) and N accumulationwere less in defoliated plants, and a greater proportion oftotal plant DM was invested in roots. About 97 % of plant totalN was derived from fixed atmospheric N, but there was incompletemixing of fixed and mineral N within the plant. Relatively moremineral N was incorporated into roots, whereas there was relativelymore fixed N in nodules. There was isotopic evidence that Nwas remobilized from root and stolon tissue for leaf regrowthafter defoliation; approximately 2 % of plant N turned overdaily in the 7-d period after defoliation, and this contributedabout 50% of the N increment in leaf tissue. Two aspects of metabolic adaptation to increased terrestrialismare considered: (1) respiratory adaptations as reflected bycomparative cytochrome c oxidase activity in tissues of crabsfrom aquatic and terrestrial habitats, and (2) thermal acclimationpatterns in cytochrome c oxidase activity in tissues from thesecrabs. Enzymatic assays were done spectrophotometrically ongill, muscle, and mid-gut gland tissues from two aquatic species,Libinia emarginata and Callinectes sapidus, and the terrestrialOcypode quadrata. Cytochrome c oxidase was chosen for this studysince it is generally believed that the more aerobic the cellsor tissues become, the more fully developed the cytochrome systemwill be. This enzyme is also thought to have a role in thermalacclimation. In gill tissue the activity of cytochrome c oxidase is enhancedwith the advent of aerial respiration. Enzymatic activity ofgill tissue from Ocypode quadrata was significantly greaterthan it was in tissue from the aquatic species. No correlationwas observed with increased terrestrialism and enzymatic activityof muscle or mid-gut gland tissue. The thermal acclimation patternsof tissues of these three species of crabs indicate a clear-cuttendency for less enzymatic adaptation to temperature at thetissue level as these crabs evolve toward a land habitat.  相似文献

Two methods for measuring the turgor pressures of cells in discsof storage tissue of red beet (Beta vulgaris L.) were compared,and a centrifugation method for extracting sap from frozen andthawed tissue was evaluated. Turgor pressures were measureddirectly using a pressure probe, or indirectly using a vapourpressure osmometer. With the latter, discs were placed directlyin the osmometer chamber and turgor was calculated as the differencein osmotic pressure before and after freezing and thawing. Turgorin freshly cut discs, measured with the pressure probe, wasbetween 0-012 MPa and 0.118 MPa with a mean ±s.d. of0.092±;0.032 MPa (n = 24). That measured with the osmometervaried between 0.08 MPa and 0.12 MPa with a mean ±s.d.of 0.09±0.10 MPa (n = 54). After vacuum infiltrationof discs with distilled water, the turgor measured with thepressure probe increased to 1.05–1.12 MPa. Turgor measuredwith the osmometer also increased after vacuum infiltrationbut was, on average, 12% lower than that measured with the pressureprobe. Overall, the results suggest that for routine measurements,the osmometer can provide reasonable estimates of the turgorof cells in beet discs. This is because a number of factorsthat, potentially, could interfere with this method have onlya small effect in this tissue. None of the measured turgorsis indicative of that occurring in intact storage roots becauseboth excision and vacuum infiltration of discs alter the concentrationsof solutes in the extracellular space. The osmotic pressureof sap extracted by centrifugation from frozen and thawed discswas not significantly different from that measured by placingfrozen and thawed discs directly in the osmometer. Inactivation of 1-aminocyclopropane-1-carboxylate (ACC) oxidase   总被引:1,自引:0,他引:1  
The enzyme 1-aminocyclopropane-1-carboxylate (ACC) oxidase,which catalyses the final step in the biosynthesis of ethylene,showed a non-linear time-course in vitro and activity decayedwith a half-life of around 14 min. This loss of activity wasstudied using tomato ACC oxidase purified from Escherichia coiltransformed with the cDNA clone pTOM13. Inactivation was notdue to end-product inhibition by dehydroascorbic acid or cyanide.Preincubatlon of enzyme in the combined presence of Fe2+ ascorbateand ACC, which together allowed catalytic turnover, resultedin almost total loss of ACC oxidase activity. Enzyme Inactivatedby catalysis could not be reactivated by passage through SephadexG-25 or by treating with combina tions of DTT and CO2 A non-lineartime-course and inactivation in the presence of all substratesand cofactors was also shown for the enzyme assayed in vivowith melon fruit discs. Using the purified tomato enzyme a distinctascorbate-dependent inactivation was also observed, which occurredIn the absence of catalysis and was prevented, although notreversed, by catalase. Percentage germination, and growth of hyphae from single conidia of Erysiphe cichoracearum DC., were measured on leaf discs from topped and intact tobacco plants, grown in aerated nutrient solutions consisting of basal medium plus large or small amounts of potassium. The effect of supplying sodium was also studied. Discs were incubated on water and on 10% sucrose solution. Changes in free amino nitrogen and carbohydrate in comparable uninfected leaf discs, before and after incubation, were also measured. Potassium deficiency resulted in more free amino nitrogen and soluble carbohydrate and less insoluble carbohydrate, per cm.2 of leaf. Spore germination was not greatly affected by treatments, though it was usually less on discs from potassium-deficient leaves. The pathogen grew slower on potassium-deficient leaf discs, whether they were incubated on water or on sucrose. Incubating discs from some leaves on sucrose, compared with water, gave greatly increased sugar content and less fungal growth; discs from other leaves had a much smaller increase in sugar, and hyphal length was similar to that on discs incubated on water. Sodium, when potassium was scarce, increased potassium deficiency symptoms, free amino nitrogen and sugar content, and resistance to powdery mildew.  相似文献

Cucumber (Cucumis sativus L. cv. Poinsett 76) seedlings withfully expanded cotyledons, and excised cotyledons, first trueleaves, hypocotyl segments and fruit mesocarp discs were exposedto vapours from a series of aqueous alcohol solutions of 0 to320 mM methanol, ethanol, n-propanol, n-butanol, and n-pentanolduring chilling at 2.5C for 5 d. Certain concentrations ofeach alcohol reduced subsequent chilling-induced ion leakagefrom the cotyledons and leaves. Exposure of cotyledons to certainmethanol or ethanol solutions also reduced chilling-inducedethylene production, but not carbon dioxide production. In contrast,exposing cucumber seedlings with fully expanded cotyledons tothe same series of alcohol concentrations that resulted in reducedchilling-induced ion leakage and ethylene production of excisedcotyledons actually increased chilling injury of the seedlings.The hypocotyl region directly below the cotyledons was the siteof chilling-induced injury and contained the most chilling-sensitivehypocotyl tissue. Exposing hypocotyl segments excised from thissensitive region to alcohol solutions did not significantlyreduce chilling-inducedion leakage. Exposing excised cucumbercotyledons or hypocotyl segments to equivalent osmotic nonvolatilesolutions of mannitol and glycerol at 2.5C or to alcohol solutionsat 12.5C had no significant effect on the rate of ion leakage.For the series of alcohols used, the relationship between thelog of the alcohol concentration that minimized chilling-inducedion leakage from cucumber cotyledon discs held at 2.5C for5 d and the log of the partition coefficient of the alcoholinto olive oil or the log of the molecular weight of the alcoholswas highly significant. The same concentrations of alcoholsthat reduced chilling-inducedion leakage also reduced stomatalaperture as measured as decreased porosity of excised cotyledons.The correlation between reduced chilling injury and stomatalconductance of cotyledons exposed to a series of ethanol solutionswas highly significant. It appears that alcohols may reducechilling injury of cucumber cotyledons by inducing stomata closure.Sufficient endogenously synthesized ethanol accumulated in discsheld in N2 at 10C for 1 d to confer tolerance to chilling at2.5C for 5 d. IAA oxidase in Begonia evansiana leaves was partially purifiedby DEAE cellulose chromatography. The enzyme was identifiedas peroxidase from its elution pattern, its Michaelis constantand the effects of various inhibitors and high temperature. In B. evansiana, which requires SD treatment to form aerialtubers, the amount of IAA oxidase in leaves increased to a maximumafter 2 days of SD treatment. When SDs with interrupted nightwere given, a maximum was reached after 6 SDs. Thus, the changein IAA oxidase content had an inverse correlation to that forIAA content observed previously. Ethylene Release from Leaves of Xanthium strumarium L. and Zea mays L.   总被引:1,自引:0,他引:1  
The release of ethylene into sealed Erlenmeyer flasks by intactleaves and leaf discs of Xanthium strumarium L. a C3 plant andZea mays L. a C4 plant were compared both in white light andin darkness. The effects of the presence or absence of addedCO2 (in the form of sodium bicarbonate) the photosynthetic inhibitor3-[3,4-dichlorophenyl]-l, l-dimethyl urea (DCMU) and 1-aminocyclopropane-1-carboxylicacid (ACC), the precursor of ethylene in higher plants, werealso investigated. The rate of ethylene release from leaf tissue of Xanthium inthe absence of added CO2 was markedly reduced in the light (i.e.at the CO2 compensation point). Treatments that would enhancethe CO2 availability to the tissue (i.e. added bicarbonate,darkness, treatment with DCMU) allowed higher levels of ethylenerelease. Incubation of the tissue with ACC considerably enhancedthe release of ethylene compared to that from the correspondingcontrol tissue without ACC. However, the pattern of ethylenerelease induced by the various treatments was similar with orwithout added ACC. When tissue, in the absence of added CO2, was transferred fromlight to darkness, and back to light for 90 min periods, theethylene release rates Increased during the interposed darkperiod but resumed the lower rate during the final light period.The addition of CO2 in the light resulted in a similar rateof ethylene release to that found in the dark. The overall pattern of ethylene release from Zea leaf tissuesubjected to light and dark in the presence or absence of addedCO2 was similar to that of Xanthium. However, two or three timesmore ethylene was released from maize leaves in the light whenCO2 was added compared to that generated in the dark. This isin marked contrast to Xanthium, where, under the light conditionsused, the ethylene release rate in the dark equalled or exceededthat occurring in the light, even in the presence of high levelsof CO2. A very low rate of ethylene release was observed atthe CO2 compensation point of maize. A speculative model is presented to explain how photosyntheticactivity might act as a key factor in regulating ethylene evolutionfrom leaf tissue in these experiments. It invokes the conceptof an inhibition by CO2 of ethylene retention or breakdown thuspermitting more ethylene to be released from the leaves.  相似文献

The transport of sugars and amino acids into the mycelium of Erysiphe pisi DC. was investigated using two different systems, intact leaf discs and mycelial suspensions. Of the sugars tested, glucose was preferentially taken up by both uninfected and mildew-infected leaf discs, whereas glutamine was taken up by both tissues at a higher rate than lysine or aspartic acid. Leaf discs from infected tissue had a greater uptake capacity than those from healthy tissue for both sugars and amino acids. The uptake of glucose was inhibited more markedly than that of sucrose and fructose by 10 μ m carbonyl cyanide m -chlorophenylhydrazone (CCCP), 1 m m N -ethylmaleimide (NEM), 1 m m diethyl pyrocarbonate (DEPC) and 1 m m phenylglyoxal, whereas 1 m m PCMBS ( p -chloro-mercuribenzenesulphonic acid) inhibited sucrose uptake to the greatest extent. Uptake of glutamine, lysine and aspartic acid was inhibited similarly by CCCP (80%), NEM (20%), DEPC (70%) and PCMBS (60%). Additionally, leaf discs were used to determine which solutes could be taken up from leaf tissue by the fungus. The uptake of sugars into the mycelium was greater than that of amino acids.
Suspensions of powdery mildew mycelium accumulated glucose at about three times the rate of sucrose or fructose, and the amino acid glutamine was taken up at three times the rate of lysine or aspartic acid. Spores separated from the suspension had a low uptake capacity.
When the reducing sugar concentration of leaf apoplastic fluid was estimated, leaves infected by powdery mildew had much higher amounts in the apoplast, whereas the activity of acid invertase also appeared to be higher in apoplastic fluids from infected leaves. When apoplastic fluid samples were run on SDS gels, an invertase antibody detected two bands in samples from infected tissues that were not found in the uninfected samples.  相似文献

Structural, biochemical, and immunological comparisons of nodulesfrom ten species of plants were made to determine if a correlationexists between nodule structure, ureide production, urate oxidaseactivity, and antigenic similarity in urate oxidase. In specieswith high urate oxidase activity and cross-reaction with soybeananti-urate oxidase [soybean (Glycine max), green bean (Phaseolusvulgaris), mung bean (Vigna radiata), cowpea (Vigna unguiculata)],the nodules were determinate and contained numerous interstitialcells, interspersed among the infected cells. Within the interstitialcells of the ureide producing nodules numerous peroxisomes werenoted and these peroxisomes appear to be structurally similar,viz. a large electron opaque core surrounded by a less electronopaque rim. Although hemp sesbania (Sesbania exaltata) noduleswere similar in ultrastructure to other ureide producers withdetectable urate oxidase activity, no cross-reactivity was observedwith anti-soybean urate oxidase. Amide producing nodules eithercontained no interstitial cells [e.g. Indian jointvetch (Aeschynomeneindica), showy crotalaria (Crotalaria spectabilis)} or interstitialcells with few peroxisomes [e.g. alfalfa (Medicago saliva),broad bean (Vicia faba), pea (Pisum sativum)] with little urateoxidase activity, exhibiting no cross-reaction with soybeananti-urate oxidase. These data indicate that the urate oxidasein most ureide producing nodules is very similar and, structurally,ureide producing nodules are organized in a specialized wayto carry out ureide assimilation in the uninfected interstitialcells. Water Pathways in Leaves of Hedera helix L. and Tradescantia virginiana L.   总被引:2,自引:0,他引:2  
Hydraulic conductances of leaf tissues of Hedera helix and Tradescantiavirginiana leaves were measured. It was found that water couldflow most easily through the veins, but that the cell wallsof at least the ventral epidermis were more efficient at resupplyingwater lost from the epidermal tissue than was the mesophyllat rehydrating itself. Vein and bundle-sheath extensions, whichare characteristic of mesomorphic leaves (e.g. T. virginiana),seem to be important in maintaining a close hydraulic connectionbetween the epidermis and the vascular tissue. Inhibition of Light-Stimulated Leaf Expansion by Abscisic Acid   总被引:9,自引:2,他引:7  
Abscisic acid (ABA) applied to intact bean (Phaseolus vulgaris)leaves or to isolated leaf discs inhibits light-stimulated cellenlargement This effect may be obtained with 10–4 molm–3 ABA, but is more significant at higher concentrations.The inhibition of disc expansion by ABA is greater for discsprovided with an external supply of sucrose than for discs providedwith KC1, and may be completely overcome by increasing the KC1concentration externally to 50 mol m–3. Decreased growthrate of ABA-treated tissue is not correlated with loss of solutesfrom growing cells, but is correlated with a decrease in cellwall extensibility. ABA does not prevent light-stimulated acidificationof the leaf surface, and stimulates the acidification of theexternal solution by leaf pieces. However, the capacity of thecell walls to undergo acid-induced wall loosening is diminishedby ABA-treatment. The possibility that ABA acts directly byinhibiting growth processes at the cellular level, or indirectlyby causing stomatal closure, is discussed.
