SYNOPSIS. The classical Tetrahymena pyriformis and T. vorax strains have been classified in antigenic subgroups based on immobilization and agar double diffusion in the presence of specific antisera. The present study utilized disc electrophoresis of proteins and lactate dehydrogenase isozymes for comparison of 11 strains. In agreement with similarities reported for agar diffusion data, close electrophoretic relationships were observed between strains GL and H, and V2 and W with aniline black staining. Altho LDH isozyme production was more variable and related to cultural growth phase, similarities were recognized between GL and H, and F and PR.  相似文献

Four strains of ballistoconidium-forming yeast-like fungi (K-95, K-125, K-132 and K139), isolated from plants collected in Bangkok, Thailand, were assigned to the genus Tilletiopsis based on morphological and chemotaxonomical characteristics. On the basis of sequence data of 18S rDNA and the D1/D2 region of 26S rDNA, strains K-95, K-125 and K-132 were close to T. flava and T. fulvescens, and strain K-139 each formed related to T. minor. DNA-DNA reassociation experiments with related species revealed that strains K-125, K-132 and K-139 each formed a new and distinct species whereas strain K-95 was identified as T. flava. Tilletiopsis derxii Takashima et Nakase sp. nov. (JCM 10217T; K-125), Tilletiopsis oryzicola Takashima et Nakase sp. nov. (JCM 10218T; K-132), and Tilletiopsis penniseti Takashima et Nakase sp. nov. Two strains, which are the free-living conchocelis of Porphyra yezoensis Ueda and Porphyra haitanensis T. J. Chang et B. F. Zheng, and four “interspecific hybridization” strains of these two species are investigated. The rbcL genes of 11 samples were amplified and sequenced, each of which were about 1,400 bp with a good specificity. The results of pair-wise distance matrix and multi-alignment showed that pair-wise distance were small while homologous index was large between strains Y-2066, Y-k2001, and Y-H001. These two indexes showed the same level as the above between strains H-2001, Y-hyC1, and Y-hyC2; however, the distances were larger between three former strains and three latter strains. Cluster trees showed the same trend. Fertile strains appeared after 2 years of culture of unfertile interspecific hybridization conchocelis and were separated from that based on different colors. Our finding that these fertile strains can bear offspring is important for understanding the interspecies hybridization of Porphyra. Molecular analysis of the fertile strains based on rbcL gene showed maternal inheritance. We inferred that somatic recombination was one of the reasons making interspecific hybridization strains fertile.  相似文献

The objective of this study was to determine the effect of freezing on the function in Atlantic salmon Salmo salar spermatozoa. The semen was frozen in Cortland's medium + 1.3M dimethyl sulphoxide + 0.3M glucose + 2% bovine serum albumin (final concentration) in a ratio of 1:3 (semen:cryoprotectant) as the treatment (T) and fresh semen as the control (F). Straws of 0·5 ml of sperm suspension were frozen in 4 cm of N2L. They were thawed in a thermoregulated bath (40° C). After thawing, the percentage of spermatozoa with fragmented DNA [transferase dUTP (deoxyuridine triphosphate) nick‐end labelling (TUNEL)], plasma membrane integrity (SYBR‐14/PI) and mitochondrial membrane potential (ΔΨMMit, JC‐1) were evaluated by flow cytometry and motility was evaluated by optical microscope under stroboscopic light. The fertilization rates of the control and treatment semen were tested at a sperm density of 1·5 × 107 spermatozoa oocyte?1, by observation of the first cleavages after 16 h incubation at 10° C. In the cryopreserved semen (T), the mean ± s.d . DNA fragmentation was 4·8 ± 2·5%; plasma membrane integrity 75·2 ± 6·3%; mitochondrial membrane potential 51·7 ± 3·6%; motility 58·5 ± 5·3%; curved line velocity (VCL) 61·2 ± 17·4 µm s?1; average‐path velocity (VAP) 50·1 ± 17·3 µm s?1; straight‐line velocity (VSL) 59·1 ± 18·4 µm s?1; fertilization rate 81·6 ± 1·9%. There were significant differences in the plasma membrane integrity, mitochondrial membrane potential, motility, fertilization rate, VCL, VAP and VSL compared with the controls (P < 0·05). Also the mitochondrial membrane potential correlated with motility, fertilization rate, VCL and VSL (r = 0·75; r = 0·59; r = 0·77 and r = 0·79, respectively; P < 0·05); and the fertilization rate correlated with VCL and VSL (r = 0·59 and r = 0·55, respectively).  相似文献

We synthesized one V3 peptide each from HTLV-IIIB, Thai A and Thai B, conjugating then to the T cell epitope of the env region, and we also synthesized a p17 protein peptide of the gag region (HGP-30). These peptide were then coupled to 8-lysine copolymers using N-succinimidyl maleimido carboxylate (Mr = ca 60 000). We designated this the branched lysine oligopeptide method. The large peptide complexes constructed from these four macromolecular peptide were used with aluminium hydroxide or complete Freund's adjuvant to immunize mice and rabbits four times. ELISA assay with aluminium hydroxide or complete Freund's adjuvant to immunize mice and rabbits four times. ELISA assay showed high titres of anti-peptide antibodies to each V3loop peptide and the HGP-30 peptide. Strong inhibition of CD4+ dependent cell fusion was obtained with these antisera when IIIb, Thai A and Thai B strains of human immunodeficiency virus (HIV) were used. Strong anti-fusion inhibition was also observed with two other HIV strains. In addition, an increase of the anti-HIV effect was observed when we used sera obtained by multicomponent vaccine immunization. The same kind of inhibition was also observed in p24 assay systems using these immunized antisera. Activation of IL-2 production in lymphocytes was observed in p24 assay systems using these immunized antisera. Activation of IL-2 production in lymphocytes was observed in mice immunized with this vaccine. These results suggest that immunization with macromolecular peptide complexes can result in strong immunogenicity towards HIV-1.  相似文献

 Breeders desire populations with a high mean performance and a large genetic variance. Theory and methods are lacking for predicting additive variance (V A ) and testcross variance (V T ) in biparental populations. Breeders have unsuccessfully attempted to predict V A based on the coefficient of coancestry ( f ) or molecular-marker similarity between parents. In this paper, we derive the expected values of V A and V T in biparental populations, examine the variability of V A among biparental crosses, and discuss how V A and V T may be predicted in applied breeding programs. Suppose i is a recombinant inbred derived from the cross between inbreds P 1 and P 2, and inbred j is not a direct descendant of i. Let V A(i,j) be the additive variance in the F2 of the (i×j) biparental cross. Let V T(i, j) be the variance among testcrosses of F2 individuals with a specific unrelated inbred or population. Assuming linkage equilibrium and the absence of epistasis, V A(i, j) V A(P1, j) +(1−λ) V A(P2, j) , where λ= parental contribution of P 1 to i. Similarly, V T(i, j) = λV T(P1, j) +(1−λ) V T(P2, j) . Additive variance in crosses between recombinant inbreds cannot be modelled as a function of  f if, as indicated in the literature, V A differs among crosses of founder inbreds. If molecular-marker similarity between parents is used as an estimate of f, then a strong linear relationship is likewise not expected between V A and marker similarity. Differences between the actual and expected λ led to variation in V A . In applied breeding programs, modelling V A or V T in biparental crosses may be feasible with estimates of V A or V T in prior crosses and information on λ obtained from molecular-marker data. A mathematical model for single and multi step deep-jet bioreactors is presented. A stagewise approach based on macroscopic mechanistic model which divides the reactor into compartments with good quality of mixing and plug flow regions (macromixer), was used. For the mathematical representation of this model a system of differential equations, describing the concentration of tracer in structural elements based on mass balance, and the Runge-Kutta-Fehlberg numerical method of integration, was applied. An indirect enzyme-linked immunoadsorption assay (ELISA) was developed for cytochrome c3 using antisera to the cytochromes fromDesulfovibrio africanus Benghazi, Desulfovibrio vulgaris Hildenborough andDesulfovibrio salexigens British Guiana. The ELISA system was used to test for cross-reactions between these antisera and the heterologous antigens. In contrast to previous experiments using the Ouchterlony technique, all of the cytochromes c3 tested exhibited some degree of cross-reaction. Considerable variation was seen in cross-reactions for cytochromes c3 from differing strains ofD. desulfuricans. This observation raises questions about the taxonomic relatedness of these strains. No cross-reaction was seen with eukaryotic cytochrome c or withD. vulgaris cytochrome c553. A new technique is presented for treating the ground state of an heteropolymer with a random sequence of components. An exact system of equations is found for determining the ground state energy E which is equal to the polymer free energy f in the lowest-order approximation in T/V (V/2 is the large “surface” energy arising at the boundaries between coiled and “helical” sections: V ? T, Uk; U1 and –U2 are the free energies of the components counted from the corresponding coiled state energies). These equations are essentially simplified at certain fixed values of the ratio U1/U2. For integer values of U2/U1 and U1/U2 a solution is obtained with an accuracy exp(–V/Uk). The ground-state energy as a function of U1 and U2 is shown to be highly irregular: its derivatives have jumps at an infinite number of points. These jumps provide a fine structure of the melting curves. A smoothed over the jumps function E′ is found by way of analytic continuation from the integer values of U1/U2 and U2/U1. The accuracy of the approximation fE is estimated and the correctional term of order T/V is determined.  相似文献

Summary Using intracellular microelectrode technique, we investigated the changes in membrane voltage (V) of cultured bovine pigmented ciliary epithelial cells induced by different extracellular solutions. (1)V in 213 cells under steady-state conditions averaged –46.1±0.6 mV (sem). (2) Increasing extracellular K+ concentration ([K+] o ) depolarizedV. Addition of Ba2+ could diminish this response. (3) Depolarization on doubling [K+] o was increased at higher [K+] o (or low voltage). (4) Removing extracellular Ca2+ decreasedV and reduced theV amplitude on increasing [K+] o . (5)V was pH sensitive. Extra-and intracellular acidification depolarizedV; alkalinization induced a hyperpolarization.V responses to high [K+] o were reduced at acidic extracellular pH. (6) Removing K o + depolarized, K o + readdition after K+ depletion transiently hyperpolarizedV. These responses were insensitive to Ba2+ but were abolished in the presence of ouabain or in Na+-free medium. (7) Na+ readdition after Na+ depletion transiently hyperpolarizedV. This reaction was markedly reduced in the presence of ouabain or in K+-free solution but unchanged by Ba2+. It is concluded that in cultured bovine pigmented ciliary epithelial cells K+ conductance depends on Ca2+, pH and [K+] o (or voltage). An electrogenic Na+/K+-transport is present, which is stimulated during recovery from K+ or Na+ depletion. This transport is inhibited by ouabain and in K+-or Na+-free medium.  相似文献

We have generated a new series of monoclonal antibodies recognizing allotypic determinants on mouse IgG1, IgG2a, and IgG2b. In this communication we describe their reactivities with immunoglobulins of the inbred mouse strains. Comparison with serology charts indicates that many of these monoclonal antibodies detect allotypic specificities previously defined by conventional antisera; others define previously undescribed specificities. Strain and isotype distribution allows us to assign five new allotypic specificities to Igh-1 and three new specificities to Igh-3. The effect of the sinking rate, or rate of medium flow (φ) on the rate of phosphate incorporation (V) by the planktonic diatoms Thalassiosira fluviatilis Hust. and T. pseudonana Hasle & Heimdal in batch and chemostat cultures was determined by passing medium at defined flow rates (0.5–25.0 mm·min?1) over algae on membrane filters. At concentrations from 1 to 100 μg phosphorus·l?1 V, increases with increasing velocity of flow, approaching a maximum value (Vm) as described by the empirical relationship: where Kφ is the sinking rate value when V = 1/2 Vm+ Vo and Vo is the uptake at 0 rate of flow. By comparing uptake at controlled flow with uptake in a vigorously stirred medium, the phosphate concentration in the cell boundary layer can be determined. The sinking rate that reduces the phosphate concentration in the boundary layer to half of nominal concentration in the medium is much lower for the larger T. fluviatilis than for T. pseudonana. For both diatoms, it is inversely related to the nominal concentration.  相似文献

Two congenic strains of mice were bred in Konstanz that bear theIg heavy chain allotype gene of the C57BL/6 (Ig-1 b ) in a BALB/c background genome. One line (CB-8K) underwent eight backcross generations to BALB/c before sister-brother mating was initiated. For the other line (CB-16KN) backcrossing was continued for eight further generations, then a homozygousIg-1 b /Ig-1 b strain was produced by sister-brother mating. Both lines were tested for four VH markers of the BALB/c and one of the C57BL parent. The CB-16KN strain expressed the C57BL marker VHNPb together with the C57BL allotype marker, and failed to express the three BALB/c markers, VHDEX, VHS117, VHphOx and VHNPa. It thus resembled the CB-20 strain.Strain CB-8K expressed the VHNPb marker of C57BL but also all the four BALB/c markers that were tested. The strain appeared more heterogeneous than the CB-16KN strain, and a subline was bred from two exceptional mice that did not express the VHNPb marker. This subline (CB-8KN) expressed the BALB/c marker VHNPa regularly, and was negative for the VHNPb marker. It thus resembles the BAB-14 line.The crossing over event thus must have happened in one of the two meioses, which led to the CB-8K line. As BAB-14 is derived in an analogous manner to a branch of the backcross of Potter and Lieberman, which ended up in CB-20, the unexpected finding is discussed that two independent crossing over events (in CB-8KN and BAB 14) within theIg heavy chain gene region have taken place at approximately the same stage of two breeding programs.Recipient of an EMBO fellowship during part of the study.  相似文献

3-Iodothyronamine is considered as a derivate of thyroid hormone as a result of enzymatic deiodination and decarboxylation. The physiological role of thyronamine (T1AM) is not known. The aim of this study was to analyze the metabolic response to T1AM in the Djungarian hamster Phodopus sungorus. We measured the influence of T1AM (50 mg/kg) on metabolic rate (VO2), body temperature (T b) and respiratory quotient (RQ) in this species and in BL/6 mice. T1AM treated hamsters as well as the mice showed a rapid decrease in VO2 and T b, accompanied by a reduction of RQ from normal values of about ∼0.9 to ∼0.70 for several hours. This indicates that carbohydrate utilisation is blocked by the injection of T1AM and that metabolic pathways are rerouted from carbohydrate to lipid utilisation in response to T1AM. This assumption was further supported by the observation that the treatment of T1AM caused ketonuria and a significant loss of body fat. Our results indicate that T1AM has the potential to control the balance between glucose and lipid utilisation in vivo.  相似文献

Individual plants from the BC1F6 and BC1F8 backcross progenies of barley-wheat [H. marinum subsp. gussoneanum Hudson (=H. geniculatum All.) (2n = 28) × T. aestivum L. (2n = 42)] and the BC1F6 progeny of their amphiploids were used to obtain alloplasmic euploid (2n = 42) lines L-28, L-29, and L-49 and alloplasmic telocentric addition (2n = 42 + 2t) lines L-37, L-38, and L-50. The lines were examined by genomic in situ hybridization (GISH), microsatellite analysis, chromosome C-banding, and PCR analysis of the mitochondrial 18S/5S repeat. Lines L-29 and L-49 were characterized by substitution of wild barley chromosome 7H1 for common wheat chromosome 7D. In line L-49, common wheat chromosomes 1B, 5D, and 7D were substituted with homeologous barley chromosomes. Lines L-37, L-38, and L-50 each contained a pair of telocentric chromosomes, which corresponded to barley chromosome arm 7H1L. Summary Minute ventilation (V E), tidal volume (V T), respiratory frequency (f) and clavicular air sac gas composition were measured in conscious domestic fowl breathing air and hypoxic gas mixtures at neutral (18±1°C) and raised (33±1°C) air temperatures. Increases inV E caused by inhalation of 10%, 8% or 6.5% O2 in N2, respectively, were independent of temperature although at each level the absoluteV E was ca. 21·min−1 greater in the panting birds. Changes in respiratory pattern during hypoxia were markedly dependent on temperature. At 18°C almost all of the increasedV E resulted from increasedf. At 33°C hypoxia led to a strong suppression off and increase inV T. It is concluded that hyperthermia and hypoxia are additive and non-interactive in their effects on ventilatory drive, in agreement with previously reported effects of hypercapnia and physical exercise on breathing in panting fowl.  相似文献

The rates of oxygen uptake by rat liver mitochondria (MC) (native coupled, freshly frozen, and uncoupled by FCCP) have been measured polarographically in the absence (V 0) or presence (V 1) of 0.11–0.25 mM sperm whale MbO2. Under the same standard conditions, the rate of sperm whale MbO2 deoxygenation (V 2) has been studied spectrophotometrically in the presence of respiring MC. For freshly frozen MC, the dependence of V 1 and V 2 on the overall charge of MbO2 has been investigated at pH 5.6–7.6, and the influence of other differently charged proteins (apomyoglobin, egg lysozyme, lactalbumin, and BSA) has been studied at pH 7.4. It is shown that the rate of mitochondrial respiration in the presence of MbO2 increases by 10–30% (V 1 > V 0). No myoglobin effect is observed for FCCP-uncoupled MC (V max does not change). The rate of MbO2 deoxygenation is equal to the rate of oxygen uptake by mitochondria (V 2/V 1 ∼ 1 at pH 7.2–7.5). At varying pH < 7.2, the V 2 values become markedly higher than V 1, evidently due to the increased MbO2 positive charge and its stronger interaction with negatively charged mitochondrial membrane. At pH 7.4, on the contrary, V 2 is twice lower than V 1 in the case of negatively charged CM-MbO2 (pI 5.2), which has carboxymethylated histidines. Positively charged lysozyme (pI 11) strongly inhibits MbO2 deoxygenation (V 2) without affecting oxygen uptake by MC (V 0 and V 1). At the same time, apomyoglobin (pI 8.5), which is structurally very similar to the holoprotein, and both negatively charged lactalbumin (pI 4.4) and BSA (pI 4.7) have no substantial influence on V 2 and V 1. The MC membrane evidently has no specific sites for the interaction with myoglobin. Rather, the protein contacts with phospholipids of the outer membrane during MbO2 deoxygenation, and electrostatic interactions are of great importance for this process.  相似文献

Sex differences in running economy (gross oxygen cost of running, CR), maximal oxygen uptake (VO2max), anaerobic threshold (Than), percentage utilization of aerobic power (% VO2max), and Than during running were investigated. There were six men and six women aged 20–30 years with a performance time of 2 h 40 min over the marathon distance. The VO2max, Than, and CR were measured during controlled running on a treadmill at 1° and 3° gradient. From each subject's recorded time of running in the marathon, the average speed (v M) was calculated and maintained during the treadmill running for 11 min. The VO2 max was inversely related to body mass (m b), there were no sex differences, and the mean values of the reduced exponent were 0.65 for women and 0.81 for men. These results indicate that for running the unit ml·kg–0.75·min–1 is convenient when comparing individuals with different m b. The VO2max was about 10% (23 ml·kg–0.75·min–1) higher in the men than in the women. The women had on the average 10–12 ml·kg–0.75·min–1 lower VO2 than the men when running at comparable velocities. Disregarding sex, the mean value of CR was 0.211 (SEM 0.005) ml·kg–1·m–1 (resting included), and was independent of treadmill speed. No sex differences in Than expressed as % VO2max or percentage maximal heart rate were found, but Than expressed as VO2 in ml·kg–0.75·min–1 was significantly higher in the men compared to the women. The percentage utilization of f emax and concentration of blood lactate at v M was higher for the female runners. The women ran 2 days more each week than the men over the first 4 months during the half year preceding the marathon race. It was concluded that the higher VO2max and Than in the men was compensated for by more running, superior CR, and a higher exercise intensity during the race in the performance-matched female marathon runners.  相似文献
