Activities of both Hg2+-reductase and organomercurial lyase of Hg-resistant aquatic bacteria were stable at 20°C for several days within whole cells. Organomercurial lyase activity degrading specifically thimersol, an organomercurial, was more stable than the corresponding enzyme activity degrading other organomercurials like phenylmercuric acetate (PMA) and methoxyethyl mercuric chloride (MEMC). Hg2+-reductases of Gram-negative bacteria in cell-free extract were more heat resistant than those of Gram-positive bacteria, except for those of two species of Bacillus, which were as heat resistant as those of the Gram-negative bacteria. Differences in the heat resistances of organomercurial lyases of two different broad-spectrum Hg-resistant bacteria indicate the presence of two enzyme systems for degrading thimersol, PMA and MEMC.  相似文献

溶磷菌和固氮菌溶解磷矿粉时的互作效应   总被引:13,自引:0,他引:13  
采用4株溶磷菌(Lx81、Dm84、Jm92、Lx191)、和3株固氮菌(ChW5、ChW6、ChO6)单独和混合接种后测定培养液有效磷含量、pH值及总有机酸含量的方法,研究溶磷菌和固氮菌溶解磷矿粉时的互作效应。结果表明,相对于单独接种溶磷菌:Lx81与3株固氮菌分别混合培养能提高磷矿粉的溶解能力,4株溶磷菌与ChW6,Lx81、Dm84、Lx191与Ch06分别混合培养及Jm92+ChW5组合溶磷量极显著增加(P〈0.01);Dm84+ChW5、Lxl91+ChW5、Jm92+Ch06组合的溶磷量下降(P〈0.01)。除Lx81+ChW6、Lx81+Ch06培养液pH值降低外,混合培养的其它组合培养液pH值均较单独接种溶磷菌时升高。有机酸测定结果表明,Lx81、Jm92与ChW5、Ch06分别混合培养、ChW6+Lx81组合有机酸含量升高(P〈0.01),其它7种组合的有机酸含量均较单独接种溶磷菌的值下降(P〈0.01)。溶磷菌和固氮菌单菌培养时溶磷量与pH值、溶磷量与总有机酸含量及pH值与总有机酸含量之间呈现线性相关;Dm84、Lx191与3株固氮菌分别混合培养溶磷量与pH值之间、Lx81与3株固氮菌分别混合培养溶磷量与总有机酸含量之间呈现线性相关,其它组合的溶磷量与pH值、总有机酸含量间没有相关性。溶磷菌和固氮菌混合培养对溶解磷矿粉既有协同作用也有拮抗作用。  相似文献

Minimal inhibitory concentration values of HgCl2 and 5 organomercurials were determined against 24 mercury-resistant N2-fixing soil bacteria previously isolated from soil and identified in our laboratory. These bacterial strains also displayed multiple antibiotic resistant properties. Typical growth pattern of a highly mercury-resistantBeijerinckia sp (KDr2) was studied in liquid broth supplemented with toxic levels of mercury compounds. Four bacterial strains were selected for determining their ability to volatilize mercury and their Hg-volatilizing capacity was different. Cell-free extracts prepared from overnight mercury-induced cells catalyzed Hg2+-induced NADPH oxidation. Specific activities of Hg2+-reductase which is capable of catalyzing conversion of Hg2+ →Hg(o) of 10 Hg-resistant bacterial strains are also reported.  相似文献

New strains of Beijerinckia mobilis and Clostridium sp. isolated from the pea rhizosphere were studied with respect to their promoting effect on the growth and development of some agricultural crops. Seed soaking in bacterial suspensions followed by the soil application of the suspensions or their application by means of foliar spraying was found to be the most efficient method of bacterization. The application of B. mobilis andClostridium sp. cultures in combination with mineral fertilizers increased the crop production by 1.5–2.5 times. The study of the population dynamics of B. mobilis by the method of genetic marking showed that this bacterium quickly colonized the rhizoplane of plants and, therefore, had characteristics of an r-strategist. At the same time, Clostridiumsp. was closer to K-strategists, since this bacterium slowly colonized the econiches studied. The introduction of the bacteria into soil did not affect the indigenous soil bacterial complex. The presence of Clostridium sp. slowed down the colonization of roots by the fungal mycelium. The possible mechanisms of the plant growth–promoting activity of B. mobilisand Clostridiumsp. are discussed.  相似文献

研究不同有机肥配比下解磷固氮细菌对土壤养分和相关酶活性的动态影响,可为筛选合适的菌肥类型、有机肥配比和施肥周期提供参考。试验设置0%、4%、8%、12%(质量比)4种有机肥配比,选择巨大芽孢杆菌、荧光假单胞菌2种解磷菌和褐球固氮菌、巴西固氮螺菌2种固氮菌,以亚热带贫瘠黄棕壤为研究对象,在控制条件下(28 ℃、避光)持续土壤培养60 d,以探讨不同有机肥水平和不同取样时间(培养第3、8、16、30、45、60天)4种细菌单独或混合接种对土壤有效态养分和相关酶活性的影响。结果表明: 随着有机肥含量的增加,菌肥处理土壤的有效氮和有效磷含量逐渐提高,表现为12%>8%>4%>0%。随着培养时间的延长,各处理土壤养分基本呈现先增加后降低的趋势。相对于单施有机肥,配施有机肥和菌肥能够显著提高和延长菌肥对土壤养分含量和酶活性的增益。菌肥对土壤养分特性的影响在不同有机肥配比下呈现不同的规律: 有机肥含量较低(0%~4%)时,虽然菌肥显著提高了土壤有效养分含量,但在初期各菌肥间差异并不显著;随着有机肥含量的提高和培养时间的延长,解磷菌(尤其是巨大芽孢杆菌)显著提高了有效磷含量,固氮菌(尤其是巴西固氮螺菌)显著提高了有效氮含量。4种细菌混合菌肥主要呈现解磷功能,比单施解磷菌具有更明显的优势,而固氮功能未得到显著提高。相关分析表明,土壤养分含量和酶活性大小呈显著正相关关系,而酶活性受土壤培养时间和碳氮比的影响。因此,针对该土壤类型,菌肥能够在短期内显著提高土壤养分含量,但菌肥特定功能的发挥依赖于有机碳的输入和土壤碳氮比的大小,配施合适比例的有机肥(8%~12%)能够提高并延长菌肥的改良效果;每45~60 d需进行菌肥复施,以保证其存活率和持续效果。  相似文献

[目的]香根草(Vetiver zizanioides)是一种多年生禾本科草本植物,具有极强的生态适应性和抗逆能力,可作饲料和水土保持用.通过研究香根草联合固氮菌多样性,为进一步研究和应用打下基础.[方法]采用无氮培养基,首次从香根草中分离到47株联合固氮菌,分别应用SDS-PAGE全细胞蛋白质电泳、DNA指纹图谱、唯一碳源和16S rDNA全序列测定等方法,进行聚类和多样性分析.[结果]SDS-PAGE、IS-PCR和Bio-BIQA碳源利用的聚类结果基本一致,将供试菌株分为6个类群和4个单菌株;16S rDNA序列测定表明,从香根草中分离的菌株包括了佛莱辛草螺菌(Herbaspirillum frisingense)、中型假食酸菌(Pseudacidovorax intermedius)、恶臭假单胞菌(Pseudomonas putida)、荧光假单胞菌(Pseudomonas fluorescens)、越南伯克氏菌(Burkholderia vietnamiensis)、阴沟肠杆菌(Enterobacter cloacae)、路德维希肠杆菌(Enterobacter ludwigii)和松江壳聚糖降解菌(Mitsuaria chitosanitabida)等不同菌种.[结论]香根草联合固氮菌具有较大的资源多样性,对固氮菌资源的扩展和将来牧草上的应用具有重要意义.  相似文献

Structural variants of mercury reductase containing the N-terminal domain, which is easily cleaved by trypsin, have been found in Gram-positive bacteria with a low genomic G + C content (Bacillus, Staphylococcus and, possibly, some other genera). Mercury reductases without the N-terminal domain and relatively resistant to limited proteolysis are typical for Gram-positive bacteria with a high genomic G + C content (Arthrobacter, Citreobacterium, Micrococcus, Mycobacterium, Rhodococcus). According to existing data, mercury resistance operons (mer operons) are in general thought to be rare in bacteria, other than those from mercury-contaminated sites. We have found that a high proportion of strains in environmental isolates of Gram-positive bacteria express mercuric reductase (MerA protein): the majority of these strains are apparently sensitive to mercury. The expression of MerA was also inducible in all cases. These results imply the presence of phenotypically cryptic mer resistance operons, with both the merA (mercuric reductase) and merR (regulatory) genes still present, but the possible absence of the transport function required to complete the resistance mechanism. This indicates that mer operons or parts thereof are more widely spread in nature than is suggested by the frequency of mercury-resistant bacteria.  相似文献

氮是限制生态系统生产力的主要元素,生物固氮是自然生态系统中氮的主要来源.生物固氮包括共生、联合和自生固氮3种类型,其中联合固氮和自生固氮统称为非共生固氮.相对于共生固氮而言,非共生固氮速率虽然较低,但其不需要与其他生物形成共生体系就可以生存并进行固氮,在时空分布上更加广泛,因此对生态系统氮循环特别是素输入具有重要贡献.本文对近年有关非共生固氮微生物的多样性、土壤和叶际固氮微生物的分布特征及影响因素等研究进展进行了综述,并在此基础上阐述了现有研究中存在的问题和发展前景.  相似文献

利用氯仿熏蒸法和变性梯度凝胶电泳法(PCR-DGGE)研究了秸秆覆盖还田与施肥对灰棕冲积水稻土0—10cm和10—20cm土层土壤微生物生物量碳、氮和固氮菌群落结构的影响。结果表明:土壤微生物量碳、氮和固氮菌多样性从0—10cm土层到10—20cm土层均呈现降低趋势。无秸秆覆盖处理(对照组)的土壤微生物生物量碳(SMB-C)和微生物生物量氮(SMB-N)量最小。在秸秆覆盖还田处理中,低氮和无钾处理的SMB-C和SMB-N都显著低于全量氮磷钾肥处理。虽然无磷处理的SMB-N低于全量氮磷钾处理,但差异不显著。说明秸秆覆盖还田配施充足氮磷钾肥能显著提高土壤微生物生物量碳、氮。由DGGE图谱多样性指数分析得知,配施充足氮磷钾肥的处理土壤的固氮菌多样性最丰富。UPGMA聚类分析显示,10种不同处理的聚类图也不同,对照(无秸秆)处理0—10cm和10—20cm的微生物不同于其它处理单独聚在了一个群里。DGGE条带测序得知,14个条带的近缘种大部分为非培养细菌nifH基因片段,主要优势菌群其归属于变形菌门(Proteobacteria)的β-变形菌纲(Betaproteobacteria)。应用PCR-DGGE技术可以解释灰棕冲积水稻土秸秆覆盖不同肥料用量固氮菌分子群落结构特点。  相似文献

To understand the composition and structure of nitrogen-fixing bacterial communities from the Sanjiangyuan Nature Reserve on the Tibetan Plateau, the molecular diversity of nifH genes from soil obtained at six sites was examined using a PCR-based cloning approach. Six samples were collected from different regions at an altitude of 3907-4824 m above sea level, and a principal component analysis (PCA) showed that they had different biogeochemical properties. A total of 446 clones and 162 unique RFLP patterns were found. PCA of the RFLP patterns and their biogeochemical parameters showed that the content of soil organic carbon (C), total nitrogen (N) and altitude were the most important factors affecting the nitrogen-fixing bacteria community. Fifty-nine nifH clones were sequenced and their nucleotide identity varied from 64% to 98%, subdivisible into four groups in our phylogenetic tree. Some of the clone sequences were related to nifH genes belonging to four phylogenetic subdivisions (alpha, beta, gamma and delta subclasses of the Proteobacteria), while most of the clones were closely related to the genes of the uncultured bacteria. The tree also showed that the sequence distributions were not clearly related to the sample sites.  相似文献

为探讨间作大豆(Glycine max)对甘蔗(Saccharum officinarum)根际土壤细菌及固氮细菌多样性的影响, 收集和开发固氮菌资源, 筛选高效甘蔗联合固氮体系, 选用3个甘蔗栽培品种‘ROC22’、‘GT21’、‘B8’与大豆品种‘Guizao 2’进行间种栽培, 采用巢式PCR特异扩增细菌16S rRNA基因片段和固氮细菌nifH基因片段, 并结合变性梯度凝胶电泳(DGGE)技术, 对间作大豆的甘蔗根际土壤细菌及固氮细菌进行系统演化和多样性分析。聚类分析结果显示, 间作大豆改变了甘蔗根际土壤细菌及固氮细菌原来的群落组成结构, 尤其对固氮菌群落组成的改变更大, 但对群落物种的优势度影响较小。Shannon-Wiener多样性指数和Simpson多样性指数分析结果表明, 甘蔗-大豆间作显著影响甘蔗根际土壤中细菌和固氮菌的多样性, 其中对固氮细菌多样性的影响较大。不同甘蔗品种的根际土壤细菌和固氮菌在间作大豆条件下表现出不同的多样性, ‘ROC22’和‘GT21’间作处理甘蔗根际土壤固氮细菌的Shannon-Wiener多样性指数显著高于单作处理, 而‘ROC22’与大豆间作处理的甘蔗根际土壤固氮菌多样性最为丰富。在大豆生长盛期, 间作处理的甘蔗根际土壤细菌多样性最为丰富, 不同处理间的差异也最大, 随后下降。总体来看, 甘蔗-大豆间作显著地影响根际土壤细菌和固氮菌的群落结构和群落多样性, 有助于对甘蔗合理间作栽培模式的认识和筛选高效甘蔗联合固氮体系。  相似文献

Summary A broad-spectrum mercury resistance locus (mer) from a spontaneous chloramphenicol-sensitive (Cms), arginine auxotrophic (Arg) mutant of Streptomyces lividan 1326 was isolated on a 6 kb DNA fragment by shotgun cloning into the mercury-sensitive derivative S. lividans TK64 using the vector pIJ702. The mer genes form part of a very large amplifiable DNA sequence present in S. lividans 1326. This element was amplified to about 20 copies per chromosome in the Cms Arg mutant and was missing from strains like S. lividans TK64, cured for the plasmid SLP3. DNA sequence analysis of a 5 kb region encompassing the whole region required for broad-spectrum mercury resistance revealed six open reading frames (ORFs) transcribed in opposite directions from a common intercistronic region. The protein sequences predicted from the two ORFs transcribed in one direction showed a high degree of similarity to mercuric reductase and organomercurial lyase from other gram-negative and gram-positive sources. Few, if any, similarities were found between the predicted polypeptide sequences of the other four ORFs and other known proteins.  相似文献

以腾格里沙漠东南缘固沙植被区和相邻天然植被区发育的藻类和藓类结皮为研究对象,采用不同规格的OTCs研究了冬季低温及短期模拟升温对其固氮活性的影响。结果表明:不同规格的OTCs装置冬季全天气温升温幅度在1℃左右,不同深度土层升温幅度更加明显,约为3.2℃;冬季试验期,湿润条件下藻类和藓类结皮均具有固氮活性,平均固氮活性分别为1.2×104和0.4×104nmolC2H4·m-2·h-1,藻类结皮的固氮活性显著高于藓类结皮(P<0.01);试验期藻类和藓类结皮的固氮活性均与培养期气温显著正相关(P<0.001),与试验前3d降水量也呈显著正相关(P<0.001)。低温湿润冷冻环境下,结皮生物体胞内冰晶形成而导致的固氮酶体系受损可能是造成冬季结皮固氮活性降低的主要原因,冬季升温能促进结皮固氮活性的提高。本研究表明,在未来全球变暖和降水格局变化背景下,冬季升温能促进生物土壤结皮对区域生态系统的氮贡献。  相似文献

This research aimed to study the diversity of purple nonsulfur bacteria (PNSB) and to investigate the effect of Hg concentrations in shrimp ponds on PNSB diversity. Amplification of the pufM gene was detected in 13 and 10 samples of water and sediment collected from 16 shrimp ponds in Southern Thailand. In addition to PNSB, other anoxygenic phototrophic bacteria (APB) were also observed; purple sulfur bacteria (PSB) and aerobic anoxygenic phototrophic bacteria (AAPB) although most of them could not be identified. Among identified groups; AAPB, PSB and PNSB in the samples of water and sediment were 25.71, 11.43 and 8.57%; and 27.78, 11.11 and 22.22%, respectively. In both sample types, Roseobacter denitrificans (AAPB) was the most dominant species followed by Halorhodospira halophila (PSB). In addition two genera, observed most frequently in the sediment samples were a group of PNSB (Rhodovulum kholense, Rhodospirillum centenum and Rhodobium marinum). The UPGMA dendrograms showed 7 and 6 clustered groups in the water and sediment samples, respectively. There was no relationship between the clustered groups and the total Hg (HgT) concentrations in the water and sediment samples used (<0.002–0.03 μg/L and 35.40–391.60 μg/kg dry weight) for studying the biodiversity. It can be concluded that there was no effect of the various Hg levels on the diversity of detected APB species; particularly the PNSB in the shrimp ponds.  相似文献
