Withanolide-type steroids, withametelin Q (1) and 12α-hydroxydaturametelin B (2) along with three known withanolides, were isolated from leaves of Datura metel L. (Solanaceae). The respective structures, characterized mainly by NMR spectroscopy, were identified as (20R,22R,24R)-21,24-epoxy-1α,3β-dihydroxywitha-5,25(27)-dienolide-3-O-β-d-glucopyranoside (1) and (20R,22R,24R)-12α,21,27-trihydroxy-1-oxowitha-2,5,24-trienolide-27-O-β-d-glucopyranoside (2). The cytotoxicity of isolated compounds was evaluated against human lung carcinoma cells (A549) and human colorectal adenocarcinoma cells (DLD-1), respectively. Compound 2 exhibited cytotoxicity against A549 and DLD-1 cell lines, with IC50 values of 7 and 2.0 μM, respectively. However, for compounds 6 and 7, cytotoxicities were higher against DLD-1 cells with IC50 values of 0.6 and 0.7 μM. Both compounds blocked the cell cycle in the S-phase and induced apoptosis.  相似文献

A predominantly haploid, approx. 57%, suspension culture of Datura innoxia was obtained and conditions for the isolation, culture and regeneration to shoots of protoplasts from these cells are described.Chromosome counts of protoplast-derived cultures showed that no haploids were present among 15 regenerated shoots, and only 3 of 30 calli contained any haploid tissue. Approx. 9.5% of mitoses examined in a protoplast-derived suspension culture were haploid.The presence of haploid sets of diplochromosomes in the early divisions of protoplasts suggests that endomitosis may be one cause for the observed ploidy increase.  相似文献

Pectin methylesterases (PME; EC 3.1.1.11) involved in de-esterification of pectin and have applicability in food, textiles, wines, pulp, and paper industries. In the present study, we compared PME activity of different parts of 3 Datura species and found that fruit coat showed maximum PME activity followed by leaf and seed. PME from leaves of D. stramonium (DsPME) was purified and characterized. DsPME showed optimum activity at 60 °C and pH 9 in the presence of 0.3 M NaCl. DsPME was stable at 70 °C and retained more than 40% activity after 60 min of incubation. However, enzyme activity completely abolished at 80 after 5 min of incubation. It follows Michaelis-Menten enzyme kinetics. Km and Vmax with citrus pectin were 0.008 mg/ml and 16.96 µmol/min, respectively. DsPME in combination with polygalactourenase (PGA) increased the clarity of orange, apple, pomegranate and pineapple juices by 2.9, 2.6, 2.3, and 3.6 fold, respectively in comparison to PGA alone. Due to very high de-esterification activity, easy denaturation and significant efficacy in incrementing clarification of fruit juice makes DsPME useful for industrial application.  相似文献

A pathogen-elicitor-inducible acyltransferase [tyramine hydroxycinnamoyltransferase (THT); EC 2.3.1], which catalyzes the transfer of hydroxycinnamic acids from hydroxycinnamoyl-CoA esters to tyramine in the formation of N-hydroxycinnamoyltyramine, was purified to apparent homogeneity from cell-suspension cultures of potato (Solanum tuberosum L. cv. Datura), with a 1400-fold enrichment, a 5% recovery and a final specific activity of 208 mkat·(kg protein)–1. Affinity chromatography on Reactive Yellow-3-Agarose using the acyl donor (feruloyl-CoA) as eluent was the decisive step in the purification sequence. The purified protein showed a native molecular mass of ca. 49 kDa. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis in the presence and in the absence of a reducing agent (2-mercaptoethanol) indicated that THT is a heterodimer in which the protein subunits (ca. 25 kDa) are non-covalently associated. The enzyme was stimulated fivefold by 10 mM Ca2+. The apparent K m value for tyramine was dependent on the nature of the hydroxycinnamoyl-CoA present. Isometric virus-like particles (VLPs) have been purified from cucumber leaf tissue. Three dsRNA segments with estimated molecular weights of 1.8, 1.1 and 1.0 × 106d have been isolated from VLPs occurring in CsCl density gradient fractions but were also readily detected in preparations from as little as 1 g of fresh leaf tissue. The VLPs resemble dsRNA containing cryptic viruses and have been named Cucumis sativus cryptic virus (CsCV).  相似文献

Stolbur phytoplasma infection has been reported, for the first time, in a new host, Solanum malacoxylon, growing in the Botanical Garden of Milan University. This shrub, native of South America, synthesizes vitamin D compounds, important for biomedical and biotechnological purposes. Pathogen detection was performed by light and transmission electron microscopy, and confirmed by molecular diagnosis, based on PCR and restriction fragment length polymorphic (RFLP) analysis of the phytoplasmal 16S rRNA and tuf genes. By means of enzymatic restriction and phylogenetic analysis on these genes, it was found that the phytoplasma belongs to the Stolbur group, taxonomic subgroup 16SrXII‐A, thus indicating S. malacoxylon as an additional host for this pathogen. Solanum malacoxylon could be then involved in the natural Stolbur phytoplasma spreading throughout South American areas, where this wild plant grows endemically.  相似文献

d-Gluconate dehydrogenase isolated from Pseudomonas fluorescens was immobilized on the surfaces of carbon and gold electrodes by irreversible adsorption. The electrodes with the adsorbed enzyme produced anodic currents in solutions containing d-gluconate. The currents were attributable to the electro-enzymic oxidation (direct bioelectrocatalytic oxidation) of d-gluconate; the electrochemical system required no external redox molecules serving as mediators of electron transfer between the electrode and the adsorbed enzyme. A model of the direct bioelectrocatalysis at the enzyme-modified electrodes is presented.  相似文献

Two new withanolides baimantuoluoside H (1) and baimantuoluoline K (2), and one known withanolide glycoside (3) were isolated and identified from the ethyl acetate-soluble fraction of ethanol extract of Datura metel seeds. The structures of the new compounds were determined using 1D and 2D NMR spectroscopy, and mass spectrometry. All isolated compounds were evaluated for their antiproliferative activity against human gastric adenocarcinoma (SGC-7901), human hepatoma (Hepg2), and human breast cancer (MCF-7) cells, as well as their immunosuppressive properties. It was determined that compounds 13 exhibited medium antiproliferative and potential immunosuppressive effects.  相似文献

A new solanocapsine-type alkaloid named aculeamine has been isolated from roots of Solanum aculeatum and its structure elucidated by physical methods including X-ray analysis as 22,26-epimino-22β-methoxy- 16α,23-epoxy-5α,22αH,25βH-cholestane-3β-ol.
