目的:我们最近的实验发现大鼠侧脑室注射氨甲酰胆碱引起显著的促钠排泄作用,本工作同时还观察了下丘脑内不同脑区的儿茶酚胺能神经元活性的变化。方法和结果:氨甲酰胆碱注射后40min,下丘脑室旁核的腹侧和内侧小细胞部、内侧视前区、尾核、苍白球的酪氨酸羟化酶免疫反应(thyrosinehydroxylaseimmunoreactivity,THIR)阳性细胞数减少,免疫反应染色强度降低;下丘脑室旁核的后部,下丘脑前区的后部、下丘脑室周核、弓状核、下丘脑外侧区的THIR阳性细胞数增多,免疫反应染色强度增强。结论:侧脑室注射氨甲酰胆碱对脑内不同脑区的内源性儿茶酚胺能神经元分别有兴奋或抑制作用,其与促钠排泄的关系将在本文中讨论  相似文献

The distribution of growth hormone releasing factor (GHRF) immunoreactive structures in the rat hypothalmus was studied after colchicine treatment with PAP immunocytochemistry in vibratome sections using an antiserum directed to rat hypothalamic GHRF. The majority of the GHRF-immunoreactive cell bodies were found in the arcuate nucleus, the medial perifornical region, and the ventral premammillary nuclei of the hypothalamus. Scattered cells were seen in the lateral basal hypothalamus, the medial and lateral portions of the ventromedial nucleus, and the dorsomedial and paraventricular nuclei. Immunoreactive fibers were observed in all the regions mentioned above. GHRF terminals were located in the central region of the median eminence. In addition, GHRF-immunoreactive neuronal processes were seen in the ventral region of the dorsomedial nucleus, the medial preoptic and suprachiasmatic regions, dorsal portion of the suprachiasmatic nucleus, bed nucleus of the stria terminals and the hypothalamic portion of the stria terminals. The localization of GHRF-immunoreactive terminals in the median eminence reinforces the view that GHRF plays a physiological role in the regulation of pituitary function. In addition, the localization of GHRF-immunoreactive structures in areas not usually considered to project to the median eminence suggest that GHRF may act as a neuromodulator or neurotransmitter.  相似文献

Monosodium glutamate (MSG) was given to neonatal male rats to determine its effects on neurons containing beta-endorphin (beta-END) and alpha-melanocyte stimulating hormone (alpha-MSH) within the basal hypothalamus (arcuate nucleus) and caudal medulla [nucleus tractus solitarius (NTS)] and on the levels of beta-END and alpha-MSH within these areas. Immunocytochemical studies demonstrated a reduction in the number of cells within the medial hypothalamic area (arcuate nucleus) among MSG-treated animals versus saline controls. MSG did not reduce the number of cell bodies within the caudal medulla (NTS). MSG significantly reduced beta-END and alpha-MSH immunoreactive levels in the basal hypothalamus as determined by radioimmunoassay. Whereas a significant reduction in the level of beta-END occurred in the ventral caudal medulla (VCM), none occurred in the dorsal caudal medulla (DCM). In contrast, levels of alpha-MSH increased significantly in the DCM among animals receiving MSG compared to control animals. This study documents the contribution of beta-endorphin containing neurons of the basal hypothalamus to areas of the caudal medulla. The effect of MSG on beta-endorphin and alpha-MSH neurons in these areas and their differential effects on levels in the caudal medulla areas raises questions about the sites of origin of these peptides.  相似文献

Double-label immunocytochemistry was used to determine whether estradiol-induced progestin receptors and either beta-endorphin or leucine-enkephalin are colocalized in female guinea pig brain. Ovariectomized, adult guinea pigs were implanted with capsules containing estradiol-17 beta to induce high levels of progestin receptors, and injected intracerebroventricularly with colchicine to improve visualization of the opiate peptides. Sections through the hypothalamus and preoptic area were processed for progestin receptor, followed by beta-endorphin or leucine-enkephalin immunocytochemistry. As reported previously, high concentrations of progestin receptor-immunoreactive (PR-IR) cells were found in the preoptic area (medial and periventricular portions, medial preoptic nucleus) and hypothalamus (anterior hypothalamic and arcuate nuclei, ventrolateral area). Many beta-endorphin-IR cells contained PR-IR in the arcuate nucleus and its surroundings (33%) and in the dorsomedial area of the hypothalamus (64%). Scattered enkephalin-IR cells were found in the septal nucleus, medial and lateral preoptic area, bed nucleus of the stria terminalis, and the arcuate nucleus. The ventromedial nucleus of the hypothalamus and dorsolateral magnocellular nucleus, respectively, contained moderate and heavy concentrations of enkephalin-IR cells. Although some of these areas also contained PR-IR, enkephalin-IR was colocalized consistently with PR-IR only in a small number of cells in the arcuate nucleus and ventromedial/ventrolateral area of the hypothalamus. These data, taken together with earlier observations that virtually all cells containing estradiol-induced PR-IR also contain estrogen receptor-IR, provide neuroanatomical evidence that hypothalamic actions of progesterone and estradiol may be mediated by beta-endorphin and/or enkephalin.  相似文献

A study was made of the formation of catecholaminergic system in the medial basal hypothalamus of Wistar rat fetuses using histofluorescent method modified by de la Torre. In the periventricular region of 16-day fetuses the cells with green fluorescence characteristic for catecholamines were found. In 18-day fetuses catecholaminergic cells were found in paraventricular, arcuate and dorsomedial hypothalamic nuclei. In all the nuclei studied the number of catecholaminergic fibers and terminals of various size is greatly increased. The data obtained suggest that medial basal hypothalamus of 18-day fetuses has a complex catecholaminergic system.  相似文献

用NADPH-d组织化学方法观察了大白鼠下丘脑内一氧化氮合酶(NDS)阳性神经元的分布及形态特征。结果显示:在视上核、室旁核的大细胞部、环状核、穹窿周核、下丘脑外侧区、下丘脑腹内侧核、下丘脑背内侧核、乳头体区大部分核团均可见一氧化氮合酶阳性神经元聚集成团。在视前内侧区、视前外侧区、下丘脑前区、下丘脑背侧区、下丘脑后区、室周核、室旁核小细胞部及穹窿内可见散在的一氧化氮合酶阳性神经元。室周核内可见呈阳性反应的接触脑脊液神经元的胞体及突起。一氧化氮合酶阳性神经元大多可见突起,有的突起上可见1~2级分支,并可见膨体。下丘脑大部分区域内可见阳性神经纤维。弓状核内可见许多弧形纤维连于第三脑室室管膜和正中隆起。  相似文献

The distribution of opiocortin (OR-ir) immunoreactive fibers was examined immunocytochemically throughout the brain in rats following surgical isolation of the arcuate opiocortin-ir neuronal pool in the medial basal hypothalamus (MBH). Fibers which emanate from this pool were completely severed and thus eliminated from the rest of the brain, leaving intact those which can be identified immunocytochemically as opiocortin-ir projections from the medullary pool located in the nucleus tractus solitarius (NTS). These studies reveal a unique organizational pattern of proopiomelanocortin (POMC) peptidergic neuronal systems and demonstrate that several pontine and medullary regions receive projections from both the hypothalamic (arcuate) and medullary (NTS) opiocortin-ir perikarya. Comparative analyses of deafferented and control brains reveal that certain brainstem autonomic centers such as parabrachial (PB), locus coeruleus (LC), nucleus paragiganticellularis (PGi) are recipients of fibers which emanate from both arcuate and NTS opiocortin-ir perikarya. Areas which receive projections from arcuate opiocortin-ir neurons alone include forebrain and hypothalamic nuclei as well as the periaqueductal grey.  相似文献

An antiserum which has previously been thought to be specific for LHRH-like immunoreactive material was shown to contain two populations of antibodies, one demonstrating anti-LHRH activity and the other anti-ACTH(1-24) activity. In rat and mouse, ACTH(1-24)-like immunoreactive substance is present in perikarya within the basal hypothalamus and in fibers in arcuate, periventricular and dorsomedial nuclei. LHRH-like immunoreactivity is present in fibers within the median eminence and arcuate nucleus, in a few fibers running along the ventral border of the hypothalamus, and in a small number of cell bodies within the medial basal hypothalamus.  相似文献

The external layer of the median eminence (ELME) and the arcuate nucleus of male rats were studied with the Falck-Hillarp technique and electron microscopy of aldehyde-OsO4 or KMnO4 fixed material after various types of hypothalamic deafferentation experiments with the Haláz knife. Special reference was paid to the monoamine systems and the results can be summarized as follows. 1. The main monoaminergic input to the ELME comes from the arcuate nucleus-periventricular area via a dorsal approach. A horizontal transection through the arcuate nucleus decreases the percentage of monoamine boutons i.e. boutons with small granular vesicles, from 31.6% in the controls to 4.4% in the lesion group, whereas only a small effect is seen after anterior (or complete) deafferentations. 2. A major input to the ELME enters the basal hypothalamus at the anterior-lateral aspects (see Réthelyi and Halsáz, 1970). The fibers cut after anterior deafferentations in all probability mainly come from cell bodies localized in the anterior hypothalamus or even further rostrally but some may represent Na axons ascending from the lower brain stem. 3. The degeneration course of nerve endings in the ELME both after anterior deafferentations as well as after lesions in the arcuate nucleus is rapid (within 2-3 days) and morphologically characterized by an initial aggregation of large dence cored vesicles seemingly to electron dense bodies within the boutons and probably also to a closer spacing of the small electron lucent synaptic vesicles (see Raisman, 1972). This type of degeneration seems to take place both in monoamine and non-monoamine neurons. 4. Degenerating boutons are found in the arcuate nucleus after anterior and complete deafferentations. Thus, the anterior hypothalamus may exert an "indirect" control of the pituitary gland via synapses on arcuate neurons although quantitavely the direct influence through the projection to the ELME is of more importance. 5. After anterior deafferentations enlarged axons containing large amounts of large dense cored vesicles and other organelles are found caudally of the cut indicating the existence of rostral projections from the medial hypothalamus.  相似文献

The variations of matrix and ependyma in the hypothalamus was histologicaly investigated in 120 human brains of both sexes from the 13 week old fetuses up to the end of the first year of postnatal life. The plane of cuts were in the region of the anterior hypothalamic nucleus, of the ventromedial nucleus, and the arcuate nucleus. We have found that the exhaustion of the matrix begins in the 14 week old embryo in the anterior and medial hypothalamus. A subependymale zone is present in these areas in the 17 week old embryo and exists up to 23 week. The one-cell-layer ependyma appears in the anterior are of the ventromedial nucleus at first in the 24 week week old embryo. After this the ependyma appears from the 25 to the 28 week old embryo in the regions of the anterior hypothalamic nucleus and the other areas of the ventromedial nucleus. In the immediate surrounding of the arcuate nucleus we observed the first alterations in the 15 week old embryo. The exhaustion of matrix is a continuous process up to the 23rd week. The boundary of the third ventricle remained to be a multilayer and in all stages of development cells were found, which reach into the lumen. Only after birth the multilayer boundary was reduced. At the end of the first year of postnatal life the one-layer ependyma was developed at a few sites. We could not find any significant variations of matrix exhaustion in the different sexes.  相似文献

Estrogen receptor alpha (ERalpha) participates in the neuroendocrine regulation of male sexual behavior, primarily in brain areas located in the limbic system. Males of many species present a long-term inhibition of sexual behavior after several ejaculations, known as sexual satiety. It has been shown that androgen receptor density is reduced 24 h after a single ejaculation or mating to satiety, in the medial preoptic area, nucleus accumbens and ventromedial hypothalamus. The aim of this study was to analyze if the density of ERalpha was also modified 24 h after a single ejaculation or mating to satiety. Sexual satiety was associated with an increased ERalpha density in the anteromedial bed nucleus of the stria terminalis (BSTMA), ventrolateral septum (LSV), posterodorsal medial amygdala (MePD), medial preoptic area (MPA) and nucleus accumbens core (NAc). A single ejaculation was related to an increase in ERalpha density in the BSTMA and MePD. ERalpha density in the arcuate (Arc) and ventromedial hypothalamic nuclei (VMN), and serum estradiol levels remained unchanged 24 h after one ejaculation or mating to satiety. These data suggest a relationship between sexual activity and an increase in the expression of ERalpha in specific brain areas, independently of estradiol levels in systemic circulation.  相似文献

Using the India ink double-perfusion technique, the blood vessels of the rat's medial hypothalamus were reconstructed from serial sections. The area studied comprised the ventromedial, dorsomedial and perifornical nuclei. The arterial supply of this territory comes from the middle hypothalamic and the anterior, middle and posterior tuberal arteries. The drainage is strictly undirectional: ventralward by the anterior, middle and posterior ventromedial, the posteromedial and posterolateral hypothalamic veins, all ending in the basal vein. The arteries of the ventromedial and dorsomedial nuclei are distinct from those of the arcuate nucleus and median eminence, and their drainage is not connected with the portal vessels. The nuclei studied, even at the levels of their subdivisions, possess own arteries whose territories of supply can well be distinguished with a minimum of overlap. The topography of these arteries is described in detail. The medial hypothalamus has no vascular connections with other regions of the diencephalon including the thalamus.  相似文献

Summary Immunohistochemically, nerve fibers and terminals reacting with anti-N-terminal-specific but not with anti-C-terminal-specific glucagon antiserum were observed in the following rat hypothalamic regions: paraventricular nucleus, supraoptic nucleus, anterior hypothalamus, arcuate nucleus, ventromedial hypothalamic nucleus and median eminence. Few fibers and terminals were demonstrated in the lateral hypothalamic area and dorsomedial hypothalamic nucleus. Radioimmunoassay data indicated that the concentration of gut glucagon-like immunoreactivity was higher in the ventromedial nucleus than in the lateral hypothalamic area. In food-deprived conditions, this concentration increased in both these parts. This was also verified in immunostained preparations in which a marked enhancement of gut glucagon-like immunoreactivity-containing fibers and terminals was observed in many hypothalamic regions. Several immunoreactive cell bodies were found in the ventromedial and arcuate nuclei of starved rats. Both biochemical and morphological data suggest that glucagon-related peptides may act as neurotransmitters or neuromodulators in the hypothalamus and may be involved in the central regulatory mechanism related to feeding behavior and energy metabolism.  相似文献

To further understand the functions of the orexin/hypocretin system, we examined the expression and regulation of the orexin/hypocretin receptor (OX1R and OX2R) mRNA in the brain by using quantitative in situ hybridization. Expression of OX1R and OX2R mRNA exhibited distinct distribution patterns. Within the hypothalamus, expression for the OX1R mRNA was largely restricted in the ventromedial (VMH) and dorsomedial hypothalamic nuclei, while high levels of OX2R mRNA were contained in the paraventricular nucleus, VMH, and arcuate nucleus as well as in mammilary nuclei. In the amygdala, OX1R mRNA was expressed throughout the amygdaloid complex with robust labeling in the medial nucleus, while OX2R mRNA was only present in the posterior cortical nucleus of amygdala. High levels of OX2R mRNA were also observed in the ventral tegmental area. Moreover, both OX1R and OX2R mRNA were observed in the hippocampus, some thalamic nuclei, and subthalamic nuclei. Furthermore, we analyzed the effect of fasting on levels of OX1R and OX2R mRNA in the hypothalamic and amygdaloid subregions. After 20 h of fasting, levels of OX1R mRNA were significantly increased in the VMH and the medial division of amygdala. An initial decrease (14 h) and a subsequent increase (20 h) in OX1R mRNA levels after fasting were observed in the dorsomedial hypothalamic nucleus and lateral division of amygdala. Levels of OX2R mRNA were augmented in the arcuate nucleus, but remained unchanged in the dorsomedial hypothalamic nucleus, paraventricular hypothalamic nucleus, and amygdala following fasting. The time-dependent and region-specific regulatory patterns of OX1R and OX2R suggest that they may participate in distinct neural circuits under the condition of food deprivation.  相似文献

The results of studies of the localization of the hypothalamic hypophysiotropic factors based on their direct determination in sections or nuclear punches are described. Luteinizing hormone-releasing hormone was found in high concentrations in the median eminence-arcuate nucleus complex, in lower concentrations in the mediobasal zone of the preoptic area. In addition to these hypothalamic sites, it is present in all four periventricular organs, especially in the organum vasculosum laminae terminalis. Thyrotropin releasing hormone has a widespread distribution. High concentrations are in the median eminence, arcuate nucleus, dorsomedial nucleus, and anterior part of the ventromedial nucleus. Lower concentrations are in several other structures of the hypothalamus, preoptic area and septum, and low but measurable quantities are found in most of the structures of the brain. Somatostatin is also present in most structures of the central nervous system, with highest concentrations in the median eminence, arcuate nucleus, ventromedial nucleus and periventricular nucleus. There are indications that the ventromedial nucleus or its immediate vicinity contains growth hormone releasing factor. Prolactin releasing activity was present in the median eminence and mediobasal parts of the anterior hypothalamus, whereas prolactin inhibitory activity was in the dorsolateral parts of the anterior hypothalamus and/or preoptic area.  相似文献

The effect of NERP-1 and NERP-2, two recently discovered VGF-derived peptides, on feeding and penile erection was studied after injection into the lateral ventricles, the lateral hypothalamus, the arcuate nucleus or the paraventricular nucleus of the hypothalamus. NERP-2 (1-5 nmol), but not NERP-1 (2-4 nmol), increased feeding in a dose-dependent manner when injected into the lateral ventricles or bilaterally into the lateral hypothalamus but not into the arcuate or the paraventricular nucleus. The effect of NERP-2 given into the lateral ventricles was found in the first, but not in the second 60 min after treatment, and was antagonized by SB-408124, an orexin-1 receptor antagonist given into the lateral ventricles or the arcuate nucleus, but not into the paraventricular nucleus. However, SB-408124 was unable to reduce NERP-2-induced feeding when injected bilaterally into the lateral hypothalamus before NERP-2 given also bilaterally into the lateral hypothalamus. In contrast, NERP-1, but not NERP-2, induced penile erection in a dose-dependent manner when injected into the lateral ventricles or the arcuate nucleus, but not into the paraventricular nucleus or the lateral hypothalamus. The pro-erectile effect of NERP-1 was not prevented by the prior injection of d(CH(2))(5)Tyr (Me)(2)-Orn(8)-oxytocin or SB-408124 into the lateral ventricles. The present results suggest that while NERP-2 facilitates feeding by acting in the lateral hypothalamus, possibly by increasing orexin activity in the arcuate nucleus, NERP-1 facilitates penile erection by acting in the arcuate nucleus with a mechanism not related to orexin or oxytocin.  相似文献

We have previously reported that administration of insulin into the arcuate nucleus of the hypothalamus decreases motivation for sucrose, assessed by a self-administration task, in rats. Because the pattern of central nervous system (CNS) activation in association with sucrose self-administration has not been evaluated, in the present study, we measured expression of c-Fos as an index of neuronal activation. We trained rats to bar-press for sucrose, according to a fixed-ratio (FR) or progressive-ratio (PR) schedule and mapped expression of c-Fos immunoreactivity in the CNS, compared with c-Fos expression in handled controls. We observed a unique expression of c-Fos in the medial hypothalamus (the arcuate, paraventricular, retrochiasmatic, dorsomedial, and ventromedial nuclei) in association with the onset of PR performance, and expression of c-Fos in the lateral hypothalamus and the bed nucleus of stria terminalis in association with the onset of FR performance. c-Fos expression was increased in the nucleus accumbens of both FR and PR rats. Our study emphasizes the importance of both hypothalamic energy homeostasis circuitry and limbic circuitry in the performance of a food reward task. Given the role of the medial hypothalamus in regulation of energy balance, our study suggests that this circuitry may contribute to reward regulation within the larger context of energy homeostasis.  相似文献

The role of the suprachiasmatic nucleus/medial preoptic area region of the hypothalamus in the expression of rat hypothalamic growth hormone-releasing factor-induced feeding in the rat was examined. Rats were tested for their 90-min food intake following microinjections of growth hormone-releasing factor (0.0, 0.01, 0.1 or 1.0 pmol) aimed at the suprachiasmatic nucleus/medial preoptic area region. It was found that growth hormone-releasing factor dose-dependently stimulated food intake with the 1.0 pmol dose being the most effective, increasing food intake by approximately 200%. Injections outside the suprachiasmatic nucleus/medial preoptic area region were ineffective. These data are taken to suggest that the suprachiasmatic nucleus/medial preoptic area region of the hypothalamus is important for the central stimulatory effects of growth hormone-releasing factor on feeding.  相似文献

Projections of different parts of the orbito-frontal cortex, the basal temporal cortex, and the hippocampus on hypothalamic nuclei were studied by recording focal responses in acute experiments on cats anesthetized with pentobarbital and chloralose. The proreal gyrus was shown to have local projections in the latero-dorsal zones of the preoptic region, in the rostral parts of the medial forebrain bundle, and also in the region of the lateral and posterior hypothalamus with the mammillary bodies. The orbital gyrus projects mainly to the latero-dorsal portions of the forebrain bundle, the latero-ventral part of the preoptic region, and the region of the lateral and latero-dorsal hypothalamic nuclei; projections from the orbital gyrus are relatively diffuse in character. The basal temporal cortex has diffuse projections in the central part of the preoptic region, in the latero-ventral parts of the medial forebrain bundle, and in the lateral mammillary body. No marked foci of activity were found in the hypothalamic structures during hippocampal stimulation. Diffuse projections of the hippocampus were traced in the ventral part of the preoptic region and the ventral regions of the medial forebrain bundle, and also in the lateral hypothalamus and in the lateral mammillary nucleus.A.
