Immunization of ewes against growth differentiation factor 9 (GDF9) or bone morphogenetic protein 15 (BMP15) can lead to an increased ovulation rate; however, it is not known whether normal pregnancies occur following such treatments. The aims of the present study were to determine the effects of a short-term immunization regimen against BMP15 and GDF9 on ovulation rate, fertilization of released oocytes, the ability of fertilized oocytes to undergo normal fetal development, and the ability of immunized ewes to carry a pregnancy to term. Ewes were given a primary and booster immunization against keyhole limpet hemocyanin (KLH; control, n = 50), a GDF9-specific peptide conjugated to KLH (GDF9, n = 30), or a BMP15-specific peptide conjugated to KLH (BMP15, n = 30). The estrous cycles of all ewes were synchronized, and ewes were joined with fertile rams approximately 14 days after the booster immunization. The number of corpora lutea was determined by laparoscopy 3-4 days following mating. Subsequently, about one-half of the ewes in each group underwent an embryo transfer procedure 4-6 days following mating, with the embryos being transferred to synchronized, nonimmunized recipients. The remaining ewes were allowed to carry their pregnancies to term. Short-term immunization against either BMP15 or GDF9 peptides resulted in an increase in ovulation rate with no apparent detrimental affects on fertilization of released oocytes, the ability of fertilized oocytes to undergo normal fetal development, or the ability of the immunized ewes to carry a pregnancy to term. Therefore, regulation of BMP15, GDF9, or both is potentially a new technique to enhance fecundity in some mammals.  相似文献

The objective of this study was to determine the effects of timely injections of flunixin meglumine (FM) or vaginal application of prostaglandin E2 (PgE2) on pregnancy, fertility, fecundity, and prolificacy rates in Saanen goats. One hundred and sixty-three nonlactating Saanen does were treated with a flugestone acetate (20 mg)-containing intravaginal sponge for 12 days. They also received eCG (400 IU) and a PGF analogue (50 μg) 10 days after progestagen priming. Does detected in estrus were mated and assigned randomly to one of three treatment groups. The PgE2 group (N = 40) received PgE2 (2.5 mg) intravaginally 15 days after mating. The FM group (N = 54) received flunixin meglumine (total dose, 100 mg) intramuscularly 15 days after mating. Flunixin meglumine was administered at 9:00 AM. Animals in the control group (N = 69) received no treatment. Pregnancy was diagnosed using transrectal ultrasonography (B-mode at 8 MHz) 30 days after mating. The pregnancy rate was significantly greater (P < 0.01) after 30 days in goats treated with PgE2 and also in the control group than in those treated with FM (67.5%, 59.4%, and 42.5%, respectively). The pregnancy rate did not differ between the PgE2 and the control group. The pregnancy and fertility rate were lowest in the FM group compared with the other groups. There was no significant difference in the prolificacy rate among experimental groups. In conclusion, our results showed that FM administration during a late luteal phase is detrimental to early pregnancy in goats.  相似文献

A non-surgical uterine flushing technique was employed to recover rhesus monkey preimplantation embryos during April–-September, a period thought to be associated with reduced fertility. A total of 22 females of proven fertility, maintained indoors under strict light and temperature control, were employed for the study in which 72 menstrual cycles were monitored. The average length of their menstrual cycle was 27.9 ± 3.8 days. The percentages of cycles that showed normal cyclic patterns of estrogen, LH, and sex-skin color were 84.7%, 91.7%, and 90.3%, respectively. Following natural mating, uterine flushing was performed on days 4–7 of pregnancy. Of 58 attempts, 27 (46.6%) resulted in the recovery of embryonic materials. Two recoveries produced unfertilized oocytes; 20 yielded embryos which were morphologically normal, and 7 yielded damaged and/or degenerate embryos. Luteal function in cycles involving uterine flushings was evaluated by examining ovaries by laparoscopy, ovulation being confirmed in 34 of 35 examinations. Serum progesterone was > 2 ng/ml in 39 of 45 cycles. The conception rate of 46.6% noted in this study was similar (P > 0.4) to those observed from other natural matings in our colony, either during the same period (49.3%; n = 67) or during October–-March (46.9%; n = 113). These results show that rhesus monkeys, maintained under appropriate environmental conditions, can experience normal fertile menstrual cycles throughout the summer months, and extend previous observations to demonstrate, for the first time, that preimplantation embryos can be recovered from this species year-round. © 1993 Wiley-Liss, Inc.  相似文献

The objective of the study was to compare the efficiency of two ovarian follicular wave synchronization protocols coupled with fixed-time natural mating with that of random mating in dromedary camels. Dromedaries were assigned randomly to one of the three treatment groups. Group 1 animals (RM; n = 46) were mated randomly. Group 2 camels (1×GnRH-FTM; n = 46) were given a GnRH analog (Buserelin, 20 μg/animal, i.v.; Receptal, Intervet, Holland) at random, then were mated 14 days later. In Group 3 animals (2×GnRH-FTM; n = 41), random GnRH analog was followed by repeated GnRH injection 14 days later and fixed-time natural mating on Day 28. Transrectal examination and ultrasonography were performed at weekly intervals to evaluate ovarian follicular status, diagnose ovulation and pregnancy. Blood samples were collected for progesterone determination by ELISA to confirm ovulation and pregnancy. All female dromedaries were assigned randomly to one of thirteen fertile bulls and were bred once on Days 1, 14 and 28 in Groups 1-3, respectively. Ovarian follicular status and ovulation rate was similar among groups at the start of the study. Seventy-five of the 133 dromedaries (56.4%) ovulated after random natural mating or random GnRH treatment. Mean length of mating was 386 ± 17.8 (±SEM) seconds. There was no significant difference in mating time among groups and in pregnancy rate among dromedary bulls. In Group 3 (2×GnRH-FTM), ovarian follicular status before mating (P < 0.05), ovulation rate (n = 37, 90.2%, P < 0.001) and pregnancy rate at 21 and 60 days (PR 21 days n = 22, 53.7% and PR 60 days n = 19, 46.3%, P < 0.05) were greater compared to random natural mating (Group 1: OR n = 25, 54.3%, PR 21 days n = 13, 28.3% and PR 60 days n = 12, 26.1%). In Group 2 dromedaries (1×GnRH-FTM), treatment tended to improve follicular status before mating, ovulation rate (n = 34, 73.9%) and pregnancy rate at 21 and 60 days (PR 21 days n = 21, 45.7% and PR 60 days n = 16, 34.8%), but the effect was not significant compared to random natural mating. In conclusion, this is the first study demonstrating that favorable pregnancy rate can be achieved following ovarian follicular wave synchronization with repeated GnRH analog and fixed-time natural mating at 14 days intervals in dromedary camels.  相似文献

This paper reports the results of reproduction with 45 wild African green monkeys (Cercopithecus aethiops) (36 females and 9 males) during the nine years from 1981 to 1989 under indoor individually-caged conditions. In 206 cases of menstruation observed, menstrual discharge lasted for 2.5 +/- 1.2 days in cycles of 22-48 days, and the length of each menstrual cycle was 31.2 +/- 6.5 days. Females who had regular menstrual cycles were subjected to "one-to-one timed mating"; females and males were put together on a one-to-one basis daily only for a certain period of time on and after the day of ovulation. Females who had irregular menstrual cycles or had no menstruation were subjected to "every-other-day mating"; females and males were put together on a one-to-one basis every other day for at least 16 weeks. The pregnancy rate (No. of pregnant females/No. of mated females) by one-to-one timed mating was 48.9% (116/237); 2.0 mating trials were needed to obtain one case of pregnancy. On the other hand, the pregnancy rate (No. of pregnant females/No. of mating trials) by every-other-day mating was 96% (48/50). Females who delivered normally totaled 129. The mean gestation period was 165 days when males, weighing 343 g on average at birth, were delivered, and 166 days when females, weighing 318 g on average at birth, were delivered. The male and female newborns were nursed for 131 and 138 days, respectively, on average. Details are summarized in Table 3. This paper also reports 23 cases of abortion, 6 stillbirths, and 6 cases of Caesarean section, by which three live fetuses and three dead fetuses were obtained.  相似文献

ABSTRACT: BACKGROUND: We here report the first investigation of exclusively elective in vitro fertilization (IVF) cycles in women with no apparent history of infertility. Since IVF outcome in women with infertility are always influenced by underlying causes of infertility, a study on non-infertile women may offer new insights. METHODS: We investigated 88 females without history of infertility in 109 consecutive elective IVF cycles, almost exclusively performed for purposes of preimplantation genetic screening (PGS; i.e., elective gender selection). The following questions were addressed: (i) impact of PGS on IVF pregnancy chances; (ii) impact of transfer of 1 vs. [greater than or equal to]2 embryos on IVF pregnancy chances; (iii) correlation of anti-Mullerian hormone (AMH) levels to embryo ploidy (iv) effect of gonadotropin dosage used in stimulation on available embryos for transfer; and (v) in form of a 1:1 case control study, compared 33 elective PGS cycles with matched control cycles without PGS, performed in couples with either prior tubal ligations and/or severe male factor infertility as indication of IVF. RESULTS: The overall clinical pregnancy rate for the group was 36.7%; pregnancy was associated with number of euploid (P = 0.009) and number of embryos transferred (P = 0.001). Odds of pregnancy were 3.4-times higher if [greater than or equal to]4 euploid embryos were produced in comparison to <4 (95% CI 1.2 to 9.2; P = 0.019), and odds of pregnancy were 6.6-times higher if greater than or equal to 2 rather than <1 euploid embryos were transferred (95% CI 2.0 to 21.7; P = 0.002). Increasing AMH (P = 0.001) and gonadotropin dosage used in ovarian stimulation (P = 0.024), was, independently, associated with number of available euploid embryos. Increasing AMH, but not follicle stimulating hormone (FSH), was associated with number of embryos available for biopsy and PGS (P = 0.0001). Implantation rates were 26.4% with PGS and 9.5% without (P=0.008). Women undergoing PGS, demonstrated 4.58-times higher odds of pregnancy than matched controls (95% CI 1.102 to 19.060, Exp 4.584, P = 0.036). CONCLUSIONS: This study suggests that outcomes of elective IVF cycles may significantly deviate from infertility-associated cycles. Affirming proof of concept for PGS, utilizing day-3 embryo biopsy and fluorescence in-situ hybridization (FISH), both widely held responsible for earlier failures to establish such proof, suggests that the principal cause of prior failures were likely not insufficient laboratory techniques but poor patient selection for PGS. Such a conclusion questions the current reintroduction of PGS with improved techniques and technologies in absence of prior determination of suited patient populations.  相似文献

Ninety-three beef bulls and 2316 females were used to determine the relationships between breeding assessments of bulls and subsequent mating performance. Each bull was given a breeding soundness examination (BSE) and two 10-min libido/serving capacity (L/SC) tests. Breeding potential of each bull was classified as satisfactory (BSE score = 60 to 100) or questionable (BSE score = 30 to 59); libido was classified as either high (mean score = 9.0 to 10) or medium (mean score = 6.0 to 8.5). Bulls were then joined, single-sire, with groups of females which had received one of two treatments to synchronize estrus. Bull-to-female ratios ranged from 1:7 to 1:51. Continuous observations were conducted on the mating activity of each group. One bull was removed from the experiment due to a severe breeding problem. Bulls of satisfactory breeding potential (n = 80) did not differ (P > 0.10) from bulls of questionable breeding potential (n = 12) in measurements of mating activity. However, by the end of the synchronized breeding period, bulls classified as satisfactory breeders achieved approximately a 9% higher (P < 0.10) pregnancy rate than did bulls of questionable breeding status (45.6 +/- 2.1% vx 36.5 +/- 5.3%). Bulls with a high libido (n = 69) serviced more (P < 0.01) estrous females (81.3 +/- 3.1% vs 63.5 +/- 4.2%) than bulls with a medium libido (n = 23). However, pregnancy rates achieved by bulls of either libido classification did not differ significantly. Individual components of the BSE as well as mean libido score were poorly correlated with pregnancy rates (r = -0.22 to 0.18). It was concluded that classification of bulls by mean libido score can aid in identifying groups of bulls that service more estrus synchronized females, whereas classification by BSE score aids in identifying groups of bulls that impregnate more females.  相似文献

Thirty adult Mule (Blue-faced Leicester x Swaledale) ewes were actively immunized against a synthetically produced peptide corresponding to the N-terminus of the alpha-subunit of bovine inhibin conjugated to tuberculin purified protein derivative (PPD). Primary immunization in the late anoestrous period was followed by two booster injections at 5 week intervals. Control groups were either not immunized (n = 15) or received PPD only (n = 15). Ten days after the second booster, oestrus was synchronized using progestagen sponges and ovulation rate was assessed by laparoscopy on days 9-10 of the cycle. Blood samples were taken at the time of each immunization and immediately before laparoscopy. Ewes were mated with fertile rams in mid-November and the resulting conception, pregnancy and lambing rates monitored. All inhibin-immunized ewes generated antibodies that bound 125I-labelled native bovine inhibin (M(r) 32,000), and their plasma follicle-stimulating hormone (FSH) concentrations after the second booster were significantly higher than the preimmunization values (30%; P less than 0.001) and the corresponding value in the controls (25%; P less than 0.025). Inhibin immunization was associated with a 90% increase in ovulation rate (P less than 0.005) and had no adverse effect on conception rate (100%), pregnancy rate (100%) or length of gestation (146 days). In Exp. 1 twice daily i.m. injections of 2 mg recombinant bovine IFN-alpha I1 (rboIFN-alpha I1) (N = 24) or placebo (N = 25) were administered to ewes from Day 12 to Day 16 during a normal oestrous cycle. Treatment did not increase (P greater than 0.10) oestrous cycle length (20.7 +/- 1.2 versus 18.5 +/- 1.4 days). In Exp. 2, ewes were injected twice daily with 2 mg IFN (N = 34) or placebo (N = 36) from Days 11 to 18 after natural mating. The rboIFN-alpha I1 significantly (P = 0.05) improved pregnancy rate (79% versus 58%) as determined by a failure of ewes to return to oestrus within 50 days. The number of ewes that lambed was greatest in the rboIFN-alpha I1-treatment group (71% versus 50%; P = 0.07), and no teratogenic effects were observed in the young born to IFN-treated ewes. The study was repeated a second year with a more fecund group of ewes (Exp. 3). More (P = 0.08) ewes injected with rboIFN-alpha I1 (58/65) than placebo-treated ewes (48/61) were judged pregnant by ultrasound. Again more ewes lambed (55 versus 45) and more lambs were born (98 versus 80) from the rboIFN-alpha I1-treated group. Combining the data from both studies revealed a significant (P = 0.01) effect of treatment. The amount of antiviral activity in jugular vein blood of ewes injected with rboIFN-alpha I1 (2 mg) was determined over time in Exp. 4. Activity rose to a maximum (approximately 450 IRU/ml) within 1-2 h and declined by over 75% in 24 h. Single injections of 1, 2 and 5 mg in buffer or 2 mg emulsified in sesame oil all gave similar profiles of antiviral activity in jugular blood over a 48-h period. In Exp. 5, antiviral activity was measured in uterine vein, ovarian artery and jugular vein serum of untreated pregnant (N = 7) and non-pregnant (N = 11) ewes at Day 15 after mating. Activity was detected in the uterine vein (58 +/- 19 IRU/ml) of all pregnant ewes. The observations in Exps 1-5 are consistent with a role for conceptus-derived IFN-alpha in maternal recognition of pregnancy and suggest that supplemental IFN-alpha might be useful in improving pregnancy success in sheep.  相似文献

Twelve mature Angus bulls possessing similar semen quality were used to study the effects of single and multiple services and single-sire and multiple-sire mating on pregnancy rate of beef heifers. Heifers were observed for natural estrus twice daily and mated within 6 hr of onset of estrus. Pregnancy rate was determined by rectal palpation at 60 days post-mating. Pregnancy rate of heifers receiving one service per estrus averaged 62.1% (n=124) and pregnancy rate of heifers receiving multiple services (single-sire) per estrus averaged 62.9% (n=105). Pregnancy rate per estrus of heifers mated to three sires (multiple-sire, one service per sire; 74.0%, n=123) was significantly higher than that of heifers mated to a single sire (62.9%). The range in pregnancy rate per bull for single-sire matings was large (0 to 95%), but this range was reduced among the same bulls used for multiple-sire matings (68 to 84%). These results indicate that pregnancy rate to one service did not differ (P>.10) from pregnancy rate to multiple services among single-sire matings, but multiple-sire matings increased (P<.05) pregnancy rate per estrus by 11% over that obtained with single-sire matings.  相似文献

When maintained under a 14L:10D photoperiod, the duration of behavioural receptivity in female golden hamsters was about 18-21 h depending on age and/or parity. The effectiveness of mating stimuli in initiating pregnancy was shown to be a function of when in the receptive period (early, middle, late) that mating occurred. During the 9-h period before ovulation, 5 ejaculatory series were sufficient to produce a nearly 100% pregnancy rate and maximum litter size. During the ovulation period, however, high pregnancy rates were achieved only when mating continued to satiety (12-15 ejaculatory series plus 10-24 long intromissions). Late in the receptive period even mating to satiety failed to result in a pregnancy. In general, pregnancy rates were significantly higher for young virgin than for older multiparous females when mating occurred during or after the ovulation period. The reduced fecundity of females mating during or after ovulation was due to insufficient vaginocervical stimulation to induce functional luteal activity and not to lack of spermatozoa. Females mating late in the receptive period did not show a cessation of oestrous cycles which characteristically follows the induction of a luteal phase. Greater amounts of vaginocervical stimulation during this time increased the number of females which delivered litters but had no significant effect on litter size. These results suggest that levels of male copulatory behaviour considered 'excessive' when mating occurs early in the receptive period are essential for pregnancy initiation when mating occurs later.  相似文献

Mature nonlactating British Saanen and Toggenburg does with a body score 2 were fed 25% (n=24) and 100% (n=16) maintenance rations from about 19 days before mating until slaughter at approximately 60 days after mating. Estrus was synchronized using PGF(2alpha), and the ovulation rate was determined by laparoscopic examination of the ovaries once between Days 6 and 10 after mating. Pregnancy rate, potential kidding rate and embryo loss were determined by counts of viable fetuses at slaughter. The proportion of does in estrus within 96 hours of PGF(2alpha) administration was not different (P<0.5) between the feed-restricted and the maintenance groups (71.0% and 87.5%, respectively); however, the time of onset of estrus after PGF(2alpha) tended to be longer (P=0.12) in the feed-restricted group. Ovulation rate, incidence of multiple ovulations and proportion of does pregnant at 60 days were significantly lower (P=0.0004, P=0.025, P=0.05, respectively) in the restricted group. More embryos from single than multiple ovulations were lost in the restricted group (P=0.01). There was no difference in the overall ovulatory activity between right and left ovaries in the 2 groups. Transuterine migrations were observed in all does that had unilateral multiple ovulations. No migration was observed in does which had single ovulations. These data indicate that restricted feed intake in goats tended to delay the onset of estrus and lowered the ovulation rate, incidence of multiple ovulations, and pregnancy rate.  相似文献

On Day 9 of pregnancy (day of mating = Day 1), the number of corpora lutea in the right ovary was greater than that in the left (mean +/- s.e.m. 9.3 +/- 0.1 and 6.5 +/- 0.3 respectively; N = 70). Although the percentages of ova fertilized on the left and right side were not different (82% and 94%), the percentage wastage was higher on the left side (20%) than the right (14%). A significant difference in sperm numbers in the right (2.8 X 10(6] and the left (0.5 X 10(6] uterine horns were found 1.5 h after mating in 51 females. Morphometric measurements of the lower uterine luminal size showed that the right side (103.9 mm3) was larger than the left side (88.9 mm3; N = 5). It is obvious that there is structural and functional asymmetry in the ovary and uterus in the golden hamster.  相似文献

The use of photoperiod, progestagen, prostaglandin and hCG treatments was investigated to obtain mating of mares at predetermined times. The objectives were: (1) synchronization of oestrus at an early time of the year, (2) simplification of treatment schedules by use of vaginal sponges, and (3) use of several controlled cycles by successive synchronization. The following conclusions were reached. First, after a 16 h photoperiod was applied beginning on 25 November, hormonal synchronization of oestrus and ovulation followed by cyclicity were obtained on 1 February; i.e. 2 months of light are essential as hormonal synchronization of ovulation was not obtained by 10 January. Second onset of oestrus was well synchronized after vaginal application of progestagens (3.75 days +/- 0.98 s.d. after withdrawal) and in spite of vaginal irritation, fertility was high (71%, N = 24) after mating every 48 h of the induced oestrus. Third, for synchronization of return to oestrus in mated non-conceiving mares, oral progestagens were given from Days 7 to 21 after mating. Predetermined mating (Days 27 and 29) and hCG injection (day 28) for non-pregnant animals were decided after a progesterone assay of Day 21 blood plasma. After 3 controlled mating periods, the cumulative fertility was 88% (N = 24) in non-lactating mares and 58% (N = 19) in lactating mares. Two trials involving 578 mares were performed to investigate the effect of a single intramuscular treatment of 40 microg buserelin, an analog of gonadotrophin releasing hormone, on pregnancy rate in mares. All mares were bred by natural mating and were allocated into pairs One mare in each pair was injected with buserelin either on Day 10 or 11 (Trial 1) or on Days 8 to 10 (Trial 2) after ovulation. Pregnancy status of mares was determined by transrectal ultrasonographic examination on Day 14 or 15 after the day of ovulation and was repeated between Days 28 and 30 of pregnancy. In Trial 1, buserelin treatment increased the pregnancy rate at Days 14 and 15 (72.5 vs 66.6%, P < 0.01). At the second pregnancy examination, pregnancy losses were lower in the treated group of mares (4.1 vs 7.4%; P < 0.05). In Trial 2, buserelin also improved the pregnancy rate (57.2 vs 53 5%; P < 0.05) at Days 14 and 15 Pregnancy losses between the first and second examinations were lower in the treated group of mares (6.5 vs 12.0%; P < 0.05). Buserelin increased pregnancy rates after breeding at the first estrus in both trials. In addition, buserelin treatment increased the pregnancy maintenance rate at Days 28 to 30.  相似文献

Methods of artificial insemination (AI) for indoor breeding in the Japanese monkey and the Cynomolgus monkey were investigated. For the Japanese monkey AI was carried out in six females during the winter mating season and in six females during the summer non-mating season. During the mating season, semen was inseminated near ovulation time in natural menstrual cycles. In the mating season study, three females inseminated at the uterine cavity became pregnant. Three inseminated at the cervical canal failed to become pregnant. For the non-mating season study, ovulation was induced artificially by PMSG and hCG and AI was carried out near the induced ovulation time. In the non-mating season, no animals became pregnant. Of four Cynomolgus monkeys used, pregnancy occurred in two animals inseminated near ovulation time in natural menstrual cycles. AI occurred at the uterine cavity in one and cervical canal in the other. In both species ovulation was verified by laparoscopy. Semen was collected by penile electro-stimulation then diluted to 2.5 to 5.0×107/ml with Whitten's medium. Diluted semen of 0.2l was inseminated at the uterine cavity or cervical canal. Our results indicate the usefulness of vaginal AI as a method of artificial indoor breeding.  相似文献

摘要目的:探讨宫腔镜联合腹腔镜在女性不孕诊断及治疗中的临床应用价值。方法:回顾性分析60 例我院收治的采用宫腔镜联 合腹腔镜进行诊断和治疗的女性不孕症患者为研究对象,对其临床资料进行分析。结果:宫腔镜联合腹腔镜检查发现,60 例不孕 症患者中,56.7 %的患者患有慢性盆腔炎,16.7 %的患者为子宫内膜异位症,11.7 %的患者为多囊卵巢综合征;单纯腹腔镜检查的 阳性检出率为60.0 %,单纯宫腔镜检查的阳性检出率为28.3 %,宫腔镜联合腹腔镜检查的阳性检出率高达91.7 %,宫腔镜联合腹 腔镜镜检阳性发现率明显高于前二者(P < 0.05)。治疗前,双侧不通、一侧通畅和双侧输卵管通畅的患者分别为38.3 %、48.3 %和 13.3 %,经宫腔镜联合腹腔镜治疗后分别为11.7 %、50.0 %和38.3 %,差异均有统计学意义(P<0.05)。34 例原发性不孕患者,术后 13例妊娠,妊娠率38.2 %;26 例继发性不孕患者,术后15 例妊娠,妊娠率57.7 %;总妊娠率为46.7 %,其中宫外孕2例。结论:宫 腔镜联合腹腔镜检查可帮助明确女性不孕症患者明确原因及发病部位,并可针对病因进行治疗,提高女性不孕症的病因诊断准 确率及治愈率。  相似文献

Semen from 3 stallions was extended using 2 methods (Kenney extender and a modified Kenney extender), slowly cooled, and stored for 41 ± 6 (s.d.) h before insemination. An insemination dose (40 ml) contained 1.5-2 billion spermatozoa. In the experiment, 26 mares were inseminated in 30 cycles. The pregnancy rate per cycle obtained with sperm stored in the Kenney extender was 87% (n=15). When the semen was extended with the modified extender, centrifuged and stored, the pregnancy rate was 60% (n=15). Inseminations were done every other day until ovulation was detected. If a mare ovulated more than 24 h after the last insemination, she was inseminated also after ovulation. The single-cycle pregnancy rate was 58% when the mares were inseminated only before ovulation (n=19) but the rate was 100% when the inseminations were done both before and after ovulation (n=9) or only after ovulation (n=2). The difference in pregnancy rates was significant (p<0.05), indicating that postovula-tory inseminations probably serve to ensure the pregnancies. The extending and handling methods used in this study resulted in a combined pregnancy rate of 73%, and appear thus to be useful for storing stallion semen for approximately 2 days.  相似文献

Previous studies from this center have indicated that the cynomolgus macaque (Macaca fascicularis) may serve as a model for human sperm interaction with the cervix and uterus. In some macaque species, transcervical aspiration of the uterine contents carries a significant risk of disturbing the cervical milieu due to the serpentine nature of the cervix. The only alternatives have been surgical procedures such as laparotomy or laparoscopy. In this paper, we report our experience with a new technique for ultrasound-guided sampling of spermatozoa in the macaque uterus. Twenty adult female cynomolgus macaques were monitored for menses (first day of menses = day l), and one mating per cycle was allowed on day 10, 11, or 12. In one group of ten animals, cervical mucus was sampled at 3 or 18 hr postcoitus (pc) and ultrasound-guided uterine aspiration was performed at 24 hours pc. In a second group of ten monkeys, uterine aspiration was at six hr pc and sperm numbers and motility were counted in the uterine fluid. Uterine fluid was obtained from fourteen of twenty monkeys. Pregnancy occurred in ten of the twenty experimental cycles. Ultrasound-guided uterine aspiration appears to be a reliable method for the evaluation of sperm transport in female macaques. The correlations between uterine sperm recovery and cervical mucus sperm populations arc discussed. The high conception rate in treatment cycles indicates that this procedure can be performed without apparent risk to pregnancy.  相似文献

The effects of grazing endophyte-infected tall fescue on luteal function, pregnancy rates, and embryonic loss rates were compared between treated mares (n=18) and untreated controls (endophyte-free, n=12). Mares grazing endophyte-infected fescue demonstrated significantly (P<0.01) prolonged luteal function (22.9 vs 15.8 d) than those grazing endophyte-free fescue. Continuous grazing of endophyte-infected fescue resulted in a decreased (P=0.30) per cycle 14-d viable pregnancy rate (14 31 , 45.2%) compared with that of endophyte-free grazing (12 16 , 75.0%). Early embryonic death rates were higher (P=0.20) in the endophyte-infected group (6 20 , 30.0%) than the endophyte-free group (1 13 , 7.7%). Cumulative pregnancy rates after a 60-d breeding period did not differ between the 2 groups. Embryonic development based on mean vesicle height at 14-d was not significantly different between treatment groups for embryos that maintained viability. Embryos that underwent early embryonic death were smaller (P<0.10) at Day-14 than embryos that maintained viability. Mean plasma progesterone concentrations were significantly (P< 0.01) greater at Day-21 postovulation in endophyte-infected mares in which the embryo remained viable (15.8 ng/ml) than in endophyte-free mares that experienced early embryonic death (9.8 ng/ml) or that demonstrated prolongation of luteal function (11.2 ng/ml). The results of this study suggest that grazing endophyte-infected tall fescue can have a detrimental effect on reproductive efficiency in the mare due to an increase in cycles bred per pregnancy rate, increased early embryonic death rate and prolongation of luteal function.  相似文献
