Malaria and ovalocytosis--molecular mimicry?   总被引:1,自引:0,他引:1  
Two recently published reports have described findings which will have a profound impact on the understanding of molecular mechanisms of human resistance to malaria infection. In Melanesian ovalocytosis, a genetic polymorphism found in Papua New Guinea and parts of South East Asia, the red cells are highly resistant to invasion by various species of malaria parasite. The molecular nature of the defect in ovalocytic erythrocytes was not known. Recent reports by Liu et al. (Liu, S.-C., Zhai, S., Palek, J., Golan, D., Amato, D., Hassan, K., Nurse, G., Babona, D., Coetzer, T., Jarolim, P. Zaik, M. and Borwein, S. (1990) N. Engl. J. Med. 323, 1530-1538.) and Jones et al. (Jones, G.L., Edmundson, H.M., Wesche, D. and Saul, A. (1991) Biochim. Biophys. Acta 1096, 33-40.) have now identified the abnormality in the band 3 protein of ovalocytic red cell membranes. A major discovery in the Jones et al. study is the presence of an extended peptide at the N-terminus of ovalocyte band 3 protein. This novel 13 amino acid extended sequence is not found in the primary structure of normal band 3 protein and was suggested to be the cause of band 3 defect in ovalocytes. We have analyzed this extended sequence through Genbank using SWISS-PROT database and found that an almost identical sequence exists in a malaria parasite protein called RESA.  相似文献

Bacterial infections in patients with hematologic malignancies still represent a severe and life-treating problem. Several observational studies during the last decade have revealed that neutropenic patients with fever are a heterogeneous population with various differences regarding response to initial therapy, development of serious complications and mortality. The role of neutropenia as main risk factor for infections in hematologic patients and the definition of different level of risk related to neutrophils count and duration of neutropenia have been extensively studied and different categories of patients based on the risk of infection, mostly the condition of neutropenia, have been clearly defined. The strategies on antimicrobial therapy and supportive care in hematologic patients need to be continuously assessed, in fact new conditions favouring the occurrence of infectious complications in patients with hematologic malignancies have progressively emerged. The use of oral prophylactic antibiotics in neutropenic cancer patients is still a matter of debate. Before 2005, several trials showed how the prevention of infection can be extremely important in this setting of patients but none was conclusive. In 2005 two meta-analysis and two large randomized clinical trials gave new evidence that antibacterial prophylaxis can reduce in neutropenic patients several important outcomes including mortality. The use of the empiric antibacterial therapy represents the cornerstone of the antimicrobial strategies in the febrile neutropenic patients leading, over the span of 20 years, to a dramatic decrease of deaths: Actually beta-lactam monotherapy is commonly used for the empiric treatment of febrile neutropenia. Recently, large randomized clinical trials and meta-analysis showed that the addition of an aminoglycoside and/or a glycopeptides results in a more favourable outcome only in selected severe infections. The use of antibiotics should be prudent and safe also in neutropenic hematologic patients to prevent emergence of microbial resistance, to save costs, to reduce toxicity. For this reasons, according to the evidence, antibacterial prophylaxis should be restricted to high risk hematologic patients and empiric parenteral antibiotic monotherapy should be recommended in case of febrile neutropenia limiting the use of amynoglicosides and glycopeptides. In the next future, a major effort should be made to state in hematologic patients new risk factors which could more accurately define subgroups for targeted anti-infective strategies.  相似文献

Learning and memory in mimicry: II. Do we understand the mimicry spectrum?   总被引:4,自引:0,他引:4  
The evolution of mimicry is driven by the behaviour of predators. However, there has been little systematic testing of the sensitivity of evolutionary predictions to variations in assumptions about predator learning and forgetting. To test how robust mimicry theory is to such behavioural modifications we combined sets of rules describing ways in which learning and forgetting might operate in vertebrate predators into 29 computer predator behaviour systems. These systems were applied in simulations of simplified natural mimicry situations, particularly investigating the nature of density-dependence and the benefits and losses conferred by mimicry across a spectrum of payabilities. The classical Batesian-Muellerian spectrum was generated only by two of our 29 predator behaviour systems. Both of these ‘classical predators' had extreme asymptotes of learning and fixed rate, time dependent forgetting. All edible mimics were treated by them as Batesian in that they parasitized their model's protection and had positive monotonic effects of density on model-mimic attack rates. All defended mimics were treated as Muellerian (Mullerian) in that their presence benefited their Model's protection, and showed negative monotonic density effects on attack rates. With the remaining 27 systems Batesian or Muellerian relationships extended beyond their conventional edibility boundaries. In some cases, Muellerian mimicry extended into the edible region of the ‘palatability spectrum’ (we term this quasi-Muellerian mimicry), and in others Batesian mimicry extended into the ‘unpalatable’, defended half of the spectrum (quasi-Batesian mimicry). Although most of the 29 behaviour systems included at least some regions of true Batesian and Muellerian mimicries, if forgetting was triggered by avoidance events (as suggested by J.E. Huheey) rather than by the passage of time then the mimicry spectrum excluded Mullerian mimicry altogether, and was composed of Batesian and quasi-Batesian mimicries. In addition the classical prediction of monotonic density-dependent predation was shown not to be robust against variations in the forgetting algorithm. Time based forgetting which is retarded by observations of prey, or which varies its rate according to the degree of pleasantness or unpleasantness of a prey generates non-monotonic results. At low mimic densities there is a positive effect on attack rates and at higher densities a negative effect. Overall, the mode of forgetting has a more significant effect on mimetic relationships than the rate of learning. It seems to matter little whether learning and forgetting are switched or gradual functions. Predictions about mimetic evolution are therefore sensitive to assumptions about predator behaviour, though more so to variations in forgetting than learning rate. Based on findings from animal psychology and mimetic populations, we are able to rule out a number of predator behaviour systems. We suggest that the most credible of our 29 predators are those which generate results which incorporate Batesian, quasi-Batesian and Muellerian mimicries across the ‘palatability spectrum’.  相似文献

Periodontal infections and atherosclerosis: mere associations?   总被引:5,自引:0,他引:5  
PURPOSE OF REVIEW: Several lines of evidence from the last few decades suggest that periodontitis is an important risk factor for cardiovascular diseases. In this review we discuss the recent findings on the systemic effects of periodontitis, which may contribute to the pathogenesis of atherosclerosis, with a special emphasis on lipoproteins. RECENT FINDINGS: In addition to the epidemiological studies exploring the direct or indirect relationship between clinical periodontitis and cardiovascular diseases, studies utilizing serology, animal models, cell cultures, and biochemistry of lipoproteins have been published. Local infection in the periodontal pockets triggers a systemic inflammatory response releasing inflammatory mediators and awakens a strong immune response against periodontal pathogens. Elevated systemic antibody levels especially to Porphyromonas gingivalis are associated with an increased risk for atherosclerosis. Periodontitis is also accompanied by proatherogenic changes in both low and high density lipoproteins, which lead to enhanced cholesteryl ester uptake by and reduced cholesterol efflux from macrophages. Vesicles and lipopolysaccharide isolated from P. gingivalis activate macrophages to convert into foam cells. Moreover, animal studies have demonstrated that infection by P. gingivalis enhances progression of atherosclerosis. SUMMARY: Recent studies have clarified the mechanisms by which periodontitis may contribute to the development of atherosclerosis. Serological, animal, and cell culture studies provide evidence that infection by P. gingivalis may promote atherosclerosis. The influence of periodontitis on lipoprotein metabolism has emerged as a new, important factor. Recent studies provide experimental proof that periodontitis may predispose to atherosclerosis.  相似文献

Microorganisms induce strong immune responses, most of which are specific for their encoded antigens. However, microbial infections can also trigger responses against self antigens (autoimmunity), and it has been proposed that this phenomenon could underlie several chronic human diseases, such as type 1 diabetes and multiple sclerosis. Nevertheless, despite intensive efforts, it has proven difficult to identify any single microorganism as the cause of a human autoimmune disease, indicating that the 'one organism-one disease' paradigm that is central to Koch's postulates might not invariably apply to microbially induced autoimmune disease. Here, we review the mechanisms by which microorganisms might induce autoimmunity, and we outline a hypothesis that we call the fertile-field hypothesis to explain how a single autoimmune disease could be induced and exacerbated by many different microbial infections.  相似文献

Abstract The intracellular development of chlamydiae in McCoy cells incubated in Eagle's minimal essential medium lacking all 13 amino acids was examined both by fluorescence and electron microscopy and by infectivity titration. Aberrant development occurred in almost all inclusions of strains of Chlamydia trachomatis and C. psittaci with the production of abnormal forms which differed in size, shape and internal structure from normal reticulate and elementary body forms. Detailed analysis of the response of C. trachomatis L2 strain 434 to graded reductions in amino acid level showed that infectivity was reduced and morphological abnormality increased as amino acid concentrations were lowered from 33 to 0% of amino acids present in minimal essential medium. Reversion of inclusions to normal and reappearance of infectious forms occured on restoration of amino acids and further incubation. It is suggested that aberrant development may account for the presence in vivo of non-cultivable chlamydiae and that such development can arise via tryptophan deprivation mediated by local release of interferon gamma.  相似文献

Müllerian mimicry, where two unpalatable species share a warning pattern, is classically believed to be a form of mutualism, where the species involved share the cost of predator education. The evolutionary dynamics of Müllerian mimicry have recently become a controversial subject, after mathematical models have shown that if minor alterations are made to assumptions about the way in which predators learn and forget about unpalatable prey, this textbook case of mutualism may not be mutualistic at all. An underlying assumption of these models is that Müllerian mimics possess the same defence chemical. However, some Müllerian mimics are known to possess different defence chemicals. Using domestic chicks as predators and coloured crumbs flavoured with either the same or different unpalatable chemicals as prey, we provide evidence that two defence chemicals can interact to enhance predator learning and memory. This indicates that Müllerian mimics that possess different defence chemicals are better protected than those that share a single defence chemical. These data provide insight into how multiple defence chemicals are perceived by birds,and how they influence the way birds learn and remember warningly coloured prey. They highlight the importance of considering how different toxins in mimicry rings can interact in the evolution and maintenance of Müllerian mimicry and could help to explain the remarkable variation in chemical defences found within and between species.  相似文献

African trypanosomiasis is endemic over much of sub-saharan Africa. But whereas domestic animals - especially cattle - often succumb to the infection, wild mammals generally show a high degree o f resistance. Many species o f wildlife living in tsetse-infested areas carry trypanosome infections, and so act as important reservoir hosts, but generally show no obvious ill-effects. How they survive the infection is an important question in understanding mechanisms o f trypanotolerance, yet relevant data are sparse and scattered. Host immune responses limit, and in some instances eliminate, nematode infections. There is considerable interest in enhancing these natural processes by the use of antinematode vaccines to achieve control of infection or disease. How nematodes are damaged is unclear. Worms might be damaged directly by effector cells and molecules of the immune system. Alternatively, they might be damaged by the physiological stress of their efforts to resist attack. Separating these possibilities could have important implications for approaches to the control of nematode infections and the disease that they cause.  相似文献

Immune systems face a daunting control challenge. On the one hand, they need to minimize damage from pathogens, without wasting energy and resources, but on the other must avoid initiating or perpetuating autoimmune responses. Finally, because pathogens interfere with immune function, immune systems must be robust against sabotage. We describe here how these challenges are met by two immune systems, the intracellular RNA interference system and the vertebrate CD8 T-cell response. BACKGROUND: Although the exact mechanisms mediating the initiation of autoimmune diseases are unknown, sequence similarity between infectious agents and self-proteins (epitope mimicry) has been proposed as the main trigger mechanism. Interestingly, this mechanism of epitope mimicry may also evoke potent tumor immunity. Indeed, experimental data support a beneficial role of autoimmunity in some patients with cancer. Additionally, autoimmunity induced via vaccination with xenogeneic antigens was found to be effective. Thus, the ability to manipulate the immune system via immunologic cross-reactions should have important potential in both preventive and therapeutic strategies for cancer. This strategy may down the friendly established relationship between tumor tissues and the cells of the immune system.  相似文献

Visual mimicry is a textbook case of natural selection because it is both intuitively understandable and has repeatedly evolved in a range of organisms: it is the ultimate example of parallel evolution. In many mimetic groups, particularly butterflies, a huge variety of colour patterns has arisen, even in closely related species. There has been much recent controversy over explanations of this variety. Mimicry is today a broad field of evolutionary study; here we discuss the evolution of its diversity in predator-prey systems.  相似文献

In this paper I argue that the nature of mimetic relationships remains contentious because there are insufficient data to enable full evaluation of theoretical models. There is, however, a growing appreciation of the need to draw together empirical studies to provide foundations for theoretical work. I review some recent data that considers the responses of predators to changing numbers of defended prey items and the nature of mimicry along a palatability spectrum. A simple model of predator behaviour is constructed which combines assumptions from Pavlovian learning studies with traditional ‘number dependent’ learning models. This model has two important properties. First it shows that Pavlovian assumptions can be represented in a simple model which generates interesting predictions. Second it indicates some areas that still need detailed empirical study – most importantly perhaps is the way that predators respond to prey with different levels of edibility. While mechanobiological processes employ diverse mechanisms, at their heart are force-induced perturbations in the structure and dynamics of molecules capable of triggering subsequent events. Among the best characterized force-sensing systems are bacterial mechanosensitive channels. These channels reflect an intimate coupling of protein conformation with the mechanics of the surrounding membrane; the membrane serves as an adaptable sensor that responds to an input of applied force and converts it into an output signal, interpreted for the cell by mechanosensitive channels. The cell can exploit this information in a number of ways: ensuring cellular viability in the presence of osmotic stress and perhaps also serving as a signal transducer for membrane tension or other functions. This review focuses on the bacterial mechanosensitive channels of large (MscL) and small (MscS) conductance and their eukaryotic homologs, with an emphasis on the outstanding issues surrounding the function and mechanism of this fascinating class of molecules.  相似文献

SLC11A1 is known to link infections, autoimmunity and cancers. A review is presented of the mechanisms by which a balance is maintained between infections caused by pathogens (viral, bacterial and protozoan; intracellular and extracellular) and disorders resulting from (acute or chronic) inflammation, and of the interactions that determine how the initial innate immune system directs subsequent acquired immune responses in human populations.  相似文献
