Current tree biology related to tree genetics and breeding has two important developments that have not well been integrated in the literature. The first is the physiological and biochemical dissection of plant yield, whereas the second is the genetic mapping based on molecular markers, such as RFLPs, RAPDs, AFLPs, and microsatellites. Genetic mapping has revolutionized traditional quantitative genetic analysis by which the genetic variation of a character is described in terms of its mean and (co)variance without the knowledge of the underlying genes. By integrating physiological and developmental studies of yield traits, genetic mapping can provide a unique means for detecting key QTL that play important roles in affecting tree growth and metabolism. The incorporation of these QTL into commercial populations through gene transformation or marker-assisted selection will move current breeding programs strictly based on an empirism to an approach that is mechanistically oriented. In this review, we discuss how plant physiology and development are merged with genetic mapping to formulate the strategy of molecular breeding in which superior forest crops are selected at the gene level. It is anticipated that this novel breeding strategy can potentially provide major breakthroughs for tree breeding.  相似文献

In light of impending water and arable land shortages, population growth and climate change, it is more important than ever to examine how forest tree domestication can be accelerated to sustainably meet future demands for wood, biomass, paper, fuel and biomaterials. Because of long breeding cycles, tree domestication cannot be rapidly achieved through traditional genetic improvement methods alone. Integrating modern genetic and genomic techniques with conventional breeding will expedite tree domestication. Breeders will only embrace these technologies if they are cost-effective and readily accessible, and forest landowners will only adopt end-products that meet with regulatory approval and public acceptance. All parties involved must work together to achieve these objectives for the benefit of society.  相似文献

Climate change and the increasing demand for sustainable energy resources require urgent strategies to increase the accuracy of selection in tree breeding (associated with higher gain). We investigated the combined pedigree and genomic-based relationship approach and its impact on the accuracy of predicted breeding values using data from 5-year-old Eucalyptus grandis progeny trial. The number of trees that can be genotyped in a tree breeding population is limited; therefore, the combined approach can be a feasible and efficient strategy to increase the genetic gain and provide more accurate predicted breeding values. We calculated the accuracy of predicted breeding values for two growth traits, diameter at breast height and total height, using two evaluation approaches: the combined approach and the classical pedigree-based approach. We also investigated the influence of two different trait heritabilities as well as the inclusion of competition genetic effects or environmental heterogeneity in an individual-tree mixed model on the estimated variance components and accuracy of breeding values. The genomic information of genotyped trees is automatically propagated to all trees with the combined approach, including the non-genotyped mothers. This increased the accuracy of overall breeding values, except for the non-genotyped trees from the competition model. The increase in the accuracy was higher for the total height, the trait with low heritability. The combined approach is a simple, fast, and accurate genomic selection method for genetic evaluation of growth traits in E. grandis and tree species in general. It is simple to implement in a traditional individual-tree mixed model and provides an easy extension to individual-tree mixed models with competition effects and/or environmental heterogeneity.  相似文献

Plant tissue culture techniques are of tremendous potential value to forest tree improvement. The technology is envisaged as playing a complementary role to traditional methods through exploiting spontaneous or induced genetic and epigenetic variability in culture, by use of haploidy and by the use of protoplasts. Haploids and protoplasts will aid in shortening breeding cycles and allow for unconventional crosses respectively. Clonal propagation is an integral part of any tree improvement program, and in addition can play an independent role in reforestation, clonal orchard establishment and in energy foresting. The goals, problems and limitations of these applications of tissue culture technology to forest tree improvement are indicated and assessed.  相似文献

以浙江省淳安县姥山林场的6×6半双列遗传交配设计的三代测定幼林为对象,分析幼林期(2、3和5 a)的生长性状的遗传参数和育种值,及2 a生时生物量的累积与分配。结果表明,马尾松幼林期不同杂交组合间的树高、地径、冠幅和活枝数均存在显著的遗传差异,且生物量在2 a生亦存在显著的遗传差异。杂交组合2 a生的地上生物量占植株总生物量的87.17%。幼林期生长性状以树高的增长量最大,5 a生较2 a生增长了4.23倍。采用综合育种值法,依据预测的树高(H)育种值为主,结合地径(D0)和2 a生茎干生物量(Bs)指标的配合选择方式,以入选率20%优选出杂交组合分别为22×44、33×22、40×44。以5 a生时树高、地径的单株育种值为依据,2%的入选率优选出16个单株,其树高、地径、冠幅及活枝数平均增益分别为1.07 m、1.58 cm、0.32 m和4.67个。  相似文献

随着世界肉牛产业科技的快速发展,我国肉牛产业的整体水平得到明显提高并取得丰硕成果。肉牛育种技术实现了由常规杂交育种向分子标记辅助育种、全基因组选择育种和基因组修饰育种的技术跨越,揭示出大量与生长发育、肉质品质、繁殖与疾病等相关的候选基因与分子标记,并逐步应用于肉牛育种实践。与生长发育性状相关的基因或分子标记主要集中在生长激素基因、生肌调节因子家族、肌肉生长抑制因子和胰岛素样生长因子等;参与肉质形成的基因主要集中在脂肪酸运输与沉积相关信号通路、钙蛋白酶信号通路、生肌调节因子家族与肌肉生长抑制因子等;繁殖性状相关基因或分子标记主要集中在GnRH-FSHR-LH、生长分化因子9、催乳素受体和FoxO1等;抗病相关基因主要有MHC基因家族、TOLL样受体4基因等。目前,利用精准基因编辑技术已培育出促生长发育与提高肉品质的肉牛育种新材料。本文总结了近年来我国在肉牛分子与基因组修饰育种领域取得的研究进展,以期为我国肉牛遗传育种技术研究提供参考和借鉴。  相似文献

1.  Governmental authorities in many countries financially support the implementation of habitat connectivity measures to enhance the exchange of individuals among fragmented populations. The evaluation of the effectiveness of such measures is crucial for future management directions and can be accomplished by using genetic methods.
2.  We retraced the population history of the European tree frog in two Swiss river valleys (Reuss and Thur), performed comprehensive population sampling to infer the genetic structure at 11 microsatellite markers, and used first-generation migrant assignment tests to evaluate the contemporary exchange of individuals.
3.  Compared with the Thur valley, the Reuss valley has lost almost double the number of breeding sites and exhibited a more pronounced genetic grouping. However, similar numbers of contemporary migrants were detected in both valleys. In the Reuss valley, 81% of the migration events occurred within the identified genetic groups, whereas in the Thur valley migration patterns were diffuse.
4.  Our results show that the connectivity measures implemented in the Reuss valley facilitated effective tree frog migration among breeding sites within distances up to 4 km. Nevertheless, the Reuss valley exhibited high genetic differentiation, which reflected the impact of barriers to tree frog movement such as the River Reuss. By contrast in the Thur valley, a larger number of breeding sites have been preserved and high admixture indicated exchange of individuals at distances up to 16 km.
5.   Synthesis and applications . We show that genetic methods can substantiate the effectiveness of connectivity measures taken in conservation management at the landscape scale. We urge responsible authorities from both river valleys to continue implementing connectivity measures and to create a dense network of breeding sites, as spatial gaps of 8 km are rarely traversed by tree frogs.  相似文献

Fish genetic breeding is a process that remolds heritable traits to obtain neotype and improved varieties.For the purpose of genetic improvement,researchers can select for desirable genetic traits,integrate a suite of traits from different donors,or alter the innate genetic traits of a species.These improved varieties have,in many cases,facilitated the development of the aquaculture industry by lowering costs and increasing both quality and yield.In this review,we present the pertinent literatures and summarize the biological bases and application of selection breeding technologies(containing traditional selective breeding,molecular marker-assisted breeding,genome-wide selective breeding and breeding by controlling single-sex groups),integration breeding technologies(containing cross breeding,nuclear transplantation,germline stem cells and germ cells transplantation,artificial gynogenesis,artificial androgenesis and polyploid breeding)and modification breeding technologies(represented by transgenic breeding)in fish genetic breeding.Additionally,we discuss the progress our laboratory has made in the field of chromosomal ploidy breeding of fish,including distant hybridization,gynogenesis,and androgenesis.Finally,we systematically summarize the research status and known problems associated with each technology.  相似文献

摘要: 单核苷酸多态性(Single nucleotide polymorphisms, SNPs)是许多生物体最丰富的遗传变异形式。林木是重要的植物类群和陆地植物生态系统的重要组成部分, SNP作为新的分子标记已应用于松、杨、黄杉、桉和云杉等属的多个树种的遗传育种学研究, 获得了包括核苷酸多样性、连锁不平衡及群体结构等相关的遗传信息, 这些研究主要建立在对候选基因序列进行测序分析的基础上。基于SNP的关联遗传学分析或连锁不平衡(Linkage disequilibrium, LD)作图, 已成为研究林木复杂数量性状的理想工具, 对桉树和火炬松的关联遗传学研究发现, 多个基因内的SNP位点与不同的木材性状相关联。利用SNP标记对林木遗传参数的估算从不同程度上揭示了林木群体进化规律及其生态学意义。SNP标记在林木中应用的不断深入, 必将极大地推动林木遗传育种学研究的发展。  相似文献

? Premise of the study: Microsatellite markers were developed for the endemic Japanese species Thujopsis dolabrata var. hondai for studies on forest ecology and tree breeding. ? Methods and Results: We characterized 10 dinucleotide microsatellite loci by screening primers developed using a simple sequence repeat-enriched library. The number of alleles per locus ranged from eight to 44 with an average of 20.3. The observed and expected heterozygosities ranged from 0.326 to 0.854 and from 0.670 to 0.976, respectively. ? Conclusions: The development of these markers can be used to assess useful genetic information for ecological studies and tree breeding in T. dolabrata var. hondai.  相似文献

The eating and cooking qualities of rice are heavily emphasized in breeding programs because they determine market values and they are the appealing attributes sought by consumers. Conventional breeding has developed traditional varieties with improved eating and cooking qualities. Recently, intensive genetic studies have pinpointed the genes that control eating and cooking quality traits. Advances in genetic studies have developed molecular techniques, thereby allowing marker-assisted breeding (MAB) for improved eating and cooking qualities in rice. MAB has gained the attention of rice breeders for the advantages it can offer that conventional breeding cannot. There have been successful cases of using MAB to improve the eating and cooking qualities in rice over the years. Nevertheless, MAB should be applied cautiously given the intensive effort needed for genotyping. Perspectives from conventional breeding to marker-assisted breeding will be discussed in this review for the advancement of the eating and cooking qualities of fragrance, amylose content (AC), gel consistency (GC) and gelatinization temperature (GT) in rice. These four parameters are associated with eating and cooking qualities in rice. The genetic basis of these four parameters is also included in this review. MAB is another approach to rice variety improvement and development in addition to being an alternative to genetic engineering. The MAB approach shortens the varietal development time, and is therefore able to deliver improved rice varieties to farmers within a shorter period of time.  相似文献

The agronomic performance of fruit trees is significantly influenced by tree internal organization. Introducing architectural traits in breeding programs could thus lead to select new varieties with a regular bearing and lower input demand in order to reduce training and environmental costs. However, an interaction between tree ontogeny and genetic factors is expected. In this study, we investigated the genetic determinism of architectural traits in the olive tree, accounting for tree development over 5 years until first flowering occurrence. We studied an F1 progeny issued from a cross between two contrasted genotypes, ‘Olivière’ and ‘Arbequina’. Tree architecture was decomposed in quantitative traits, related to (1) growth and branching, (2) first flowering and fruiting. Models, including the year of growth, branching order and genotype effects, were built with variance function and covariance structure when necessary. After a model selection, broad sense heritabilities were calculated. During the first 3 years, both the mean values of vegetative traits and genetic factor significance depended on the shoot within-tree position. Dependencies between consecutive years were revealed for traits related to whole tree form. Whole tree form variables showed medium to high broad sense heritability values, whereas reproductive traits were highly heritable. This study demonstrates the existence of ontogenic trends in the olive tree, which result in traits heritable only at the tree periphery. A phenotyping strategy adapted to its architectural characteristics and a list of relevant traits, such as maximal internode length, is proposed. Transgressive effects suggest that genetic progress could be performed in future selection programs.  相似文献

This programme provides a practical initiative promoting the conservation of rare and endangered conifer and associated temperate rainforest tree species, by providing a scientifically sound strategy to minimise further genetic erosion of an already fragmented wild tree resource. Using existing horticultural expertise and the tree-growing potential of the British climate, the programme provides for the development of permanent, scientifically structured, threatened conifer breeding populations, in a network of sites across the British Isles and elsewhere, as a long-term species rescue strategy. 由于树鼩是灵长类动物的近亲,且具有体型小、繁殖周期短、饲养管理成本低等优点,长期以来被认为有望替代灵长类动物用于人类疾病的动物模型研究.然而,目前对树鼩的群体遗传结构还知之甚少,这极大地限制了其在疾病动物模型研究的应用,也是其品系资源创制的瓶颈.本研究通过分析80只采自于云南省昆明周边地区的野生树鼩(Tupaia belangeri chinensis)线粒体DNA(mtDNA)多态性,结合国外报道的2个树鼩(Tupaia belangeri)序列比较后发现,在604 bp的mtDNA控制区片段中兵检测到29个核苷酸替代变异,这些变异共界定了13种单倍型,表现较高的群体遗传多样度.另外,昆明地区的树鼩与国外报道的2个树鼩间存在较大的遗传分化,mtDNA控制区单倍型之间的核苷酸替换数大于18个,远高于昆明地区树鼢群体内部不同单倍型之间的差异.选择含有代表性的mtDNA控制区单倍型的17个昆明地区树鼩个体进一步测定了细胞色素b基因片段(1134 bp),结合前人报道的数据分析,结果进一步支持mtDNA控制区数据反映的遗传格局及揭示的昆明地区树询与国外报道树鼩之间的明显差异.本研究结果提示,昆明地区树鼩与国外树鼩之间存在较大遗传差异,在将树鼩用于人类疾病动物模型研究中要注意这些遗传差别.昆明城郊的树鼩群体具有较高的遗传多样度,在开展近交系建立等工作时须考虑选取群体内部具有代表性的mtDNA世系.
Abstract:
Due to their special phylogenetic position in the Euarchontoglires and close affinity to primates, tree shrews have been proposed as an alternative experimental animal to primates in biomedical research. However, the population genetic structure of tree shrews has largely remained unknown and this has hindered the development of tree shrew breeding and selection. Here we sampled 80 Chinese tree shrews (Tupaia belangeri chinensis) in Kunming, China, and analyzed partial mtDNA control region sequence variation. Based on our samples and two published sequences from northern tree shrews (T. belangeri), we identified 29 substitutions in the mtDNA control region fragment (~ 604 bp)across 82 individuals and defined 13 hapiotypes. Seventeen samples were selected for sequencing of the cytochrome b (Cyt b; 1134 bp) gene based on control region sequence variation and were analyzed in combination with 34 published sequences to solidify the phylogenetic pattern obtained from control region data. Overall, tree shrews from Kunming have high genetic diversity and present a remarkable long genetic distance to the two reported northern tree shrews outside China. Our results provide some caution when using tree shrews to establish animal models because of this apparent genetic difference. In addition, the high genetic diversity of Chinese tree shrews inhabiting Kunming suggests that systematic genetic investigations should be conducted before establishing an inbred strain for medical and biological research.  相似文献

Birch (Betula platyphylla Suk.), distributed in Eurasia, North America, and Australia, is a kind of cold-resistant, fast-growing, and vital pulpwood tree species. It is also one of the most important ecological restoration tree species with high values of economic benefits in Northeast China. To improve the genetic gain and expand the economic benefit of B. platyphylla, many genetic improvements have been carried out. In China, B. platyphylla is widely distributed and varied, and there are many varieties with excellent genetic characteristics. In this paper, the genetic improvement of B. platyphylla was reviewed, and the previous research results were discussed from two aspects: conventional breeding and molecular breeding. Some problems and corresponding solutions in the genetic improvement were put forward to provide ideas for B. platyphylla breeding in the future.  相似文献
