The main objective of this investigation was to study the enzymatic activities of Actinomycetales strains isolated from an agricultural soil amended with different amounts of municipal solid waste compost (MSWC) or farmyard manure (FM). For this purpose, the hydrolytic activities of carboxymethyl cellulase, xylanase, pectinase, amylase, chitinase and protease were tested for 75 isolates of Sterptomyces, Amycolatopsis and Nocardioides from different sources (unamended soil, amended soil with FM or MSWC, FM and MSWC) at temperature ranging between 30 and 50°C. It was shown that the highest rate of enzymes producer’s strains was registered at 30°C, and decreased gradually to annul at 50°C, with the exception of the MSWC strains origin. It was also shown that the percentage of strains producers of enzymes isolated from soil amended with MSWC appeared higher than the one registered for those isolated from control and amended with FM soils. Application of MSWC increases the number of enzymes producer-actinomycetes in the soil and then it improves its fertility.  相似文献

Bacterial community structure and diversity of Tunisian agricultural soil treated with different amounts of municipal solid waste compost (MSWC) and other fertilizers were studied using DGGE and ARISA fingerprinting methods. Sequence analysis of dominant DGGE bands revealed the presence of three major clusters, Cytophaga/Flexibacter/Bacteroides (CFB) group, Proteobacteria and Acidobacteria group. Using ARISA profiles, dominant populations were assigned to low and high GC Gram positive bacteria, Cyanobacteria, Spirochetes and Cytophagales. The two methods revealed the absence of significant bacterial community shifts related to the different MSWC applications. Moreover, indigenous bacterial population of the used loam-clayey soil was observed to limit proliferation and survival of Proteobacteria, initially dominant in MSWC and farmyard manure. Effectiveness of the two methods for soil bacterial community studying was shown. While DGGE was more accurate for bacterial identification, ARISA was more practical for handling and rapid estimation of dominant bacteria.  相似文献

花生属豆科固氮作物,具较强的抗旱耐盐性,土壤微生物在盐碱土生态系统中具有重要的生态功能。以花生平作、花生/棉花间作为对象,通过16S rRNA基因克隆文库技术分析了黄河三角洲滨海盐碱地花生旺盛生长期不同含盐量盐碱地和非盐碱地0—40cm根层非培养土壤微生物群落组成及其多样性,分析了盐碱地花生根层土壤细菌群落与非盐碱地花生根层土壤细菌群落的差异,为揭示盐碱地花生根层土壤微生物的多样性以及土地利用变化与生态环境效应间的关系奠定基础。利用免培养技术直接从土壤样品提取总DNA,针对细菌基因组16S rRNA基因的V3高变区进行PCR扩增;利用焦磷酸测序的方法对V3高变区PCR产物进行高通量测序,并对测序数据进行生物信息学分析。结果表明,(1)黄河三角洲滨海盐碱土较高含盐量土壤中根层土壤微生物种类、优势种群数量和群落功能多样性较非盐碱土壤较为丰富。(2)盐碱土花生平作或花生//棉花间作两种种植方式基本不影响二者0—40cm根层土壤微生物优势类群;不同土壤类型和种植模式下,花生和棉花根层土壤中优势菌群均为变形菌门(Proteobacteria)、放线菌门(Actinobacteria)、绿弯菌门(Chloroflexi)和酸杆菌门(Acidobacteria) 4种菌群,其总丰度为80%—90%。非盐碱土壤中花生根层的酸杆菌门(Acidobacteria)丰度是盐碱土壤中的3倍以上,嗜热油菌纲(Thermoleophilia)和放线菌纲(Actinomycetales)丰度远高于各种盐碱土壤花生平作和花生//棉花间作两种植模式下的花生根层土壤;非盐碱土平作花生0—40cm土层中Rubellimicrobium、Pontibacter和Lamia细菌则显著缺失。(3)土壤类型对土壤微生物菌群类型影响较大,聚类分析表明,10个土壤样本依据土壤含盐量高低和根系分布深度聚为3类,即非盐碱土壤归为1类,盐碱土壤根系密集分布层0—20cm、20—40cm各归为1类。  相似文献

Incubation tests were used to assess the effectiveness of three different organic residues and three different liming materials, alone or in combination, in the remediation of a mine contaminated soil. The organic residues tested were sewage sludge from a municipal wastewater treatment plant (SS), compost from the organic fraction of unsorted municipal solid waste (MSWC), and garden waste compost (GWC), applied at 100 and 200 Mg ha? 1. The liming materials tested were agriculture limestone (6.4 Mg ha? 1), calcium oxide (3.7 Mg ha? 1), and sugar beet sludge (12.2 Mg ha? 1) from the sugar manufacturing process. The soil and mixtures of soil and amendments were adjusted to 70% of the maximum water holding capacity and incubated for 28 days in a controlled-temperature room at 20°C ± 1°C. At the end of the incubation, samples were analyzed for pH, electrical conductivity, organic matter content, CaCl2-extractable, and' NH4Ac/HAc+ EDTA–extractable metal fractions (Cu, Zn, and Pb). Correlations among the variables and/or similarities among the treatments were identified by principal component analysis and hierarchical cluster analysis. The amendments tested decreased the CaCl2-extractable Cu and Zn fractions, considered as mobile metal fractions, to below analytical detectable limits, providing organic matter to the soil with levels between 1% and 2% at the end of the experiment, significantly different relatively to the original soil. pH and electrical conductivity reached high values when using 200 Mg ha? 1 SS or 200 Mg ha? 1 MSWC, with any of the liming materials tested, making these application rates excessive for this particular situation. Furthermore, the treatments using MSWC increased the NH4Ac/HAc+ EDTA–extractable Cu, Pb, and Zn fractions, considered as mobilizable metal fractions, as did the 200 Mg ha? 1 SS for Pb and Zn. Considering the overall results, the compost made from garden waste decreased metal solubility in the soil and increased soil pH and organic matter content, without the addition of large amounts of soluble salts, and without increasing the mobilizable metal content. Of the organic materials tested, this was the only one that can be considered adequate for remediation of the contaminated soil under study, at the application rates tested.  相似文献

Leached spent mushroom compost (SMC), municipal solid waste compost (MSWC) and their extracts, were tested to suppress Phytophthora drechsleri in cucumber plants. The composts were mixed with sand-loam soil in sterile and non-sterile types and were used to assess suppressiveness against P. drechselri in greenhouse experiments. The extracts of composts, in both sterile and non-sterile types, were applied to evaluate their effect in suppression of pathogen in vitro. The results of the experiments showed that all applications rate of non-sterile SMC were significantly effective in the control of the pathogen. However the sterile SMC amendments did not have a positive effect on the pathogen suppression in vitro or in vivo, as it was expected. In greenhouse experiments, both composts were effective in controlling pathogen at the rate of 15%, but the treatments amended with higher rate of MSWC did not show a positive effect. The treatments amended with MSWC (15%) and SMC (25%) showed the most suppressive effect in controlling the pathogen. The extract of leached-SMC could inhibit P. drechselri in petri dish.  相似文献

The limits for loading soils with Tunisian urban compost for cultivating Medicago sativa were determined in a 6-month experiment in a greenhouse. Mature municipal solid waste compost (MSWC) from Tunis city was applied to clay and sandy soils from cultivated fields at rates equivalent to 40, 80, 120tha(-1). In the absence of MSWC, the shoot biomass (dry weight) cumulated over four cuts was 2-2.5 lower in sandy soil than in clay soil. It was 20-25% augmented upon MSWC addition in clay soil, independently of MSWC dose. The opposite trend was observed in sandy soil, the shoot yields being diminished by MSWC in a dose dependent manner. In MSWC-amended clay soil but not in sandy soil, Cd, Cu, Zn, and Pb concentrations in shoots remained below or close to the tolerated values according to EEC norms. The MSWC might be used as conditioner for clay soil, but not for sandy soil.  相似文献

The objective of this study was to identify and characterize other proteins than fimbrial proteins potentially involved in R. albus 20 adhesion to cellulose using an adhesion-related antiserum preparation (i.e. anti-Adh serum). From protein fractions of R. albus 20 grown on cellulose, the serum recognized at least 10 cellulose-binding proteins (CBPs), among which homologs of glycoside hydrolases (family 5, 9 and 48) of R. albus 8 (i.e. Cel5G, Cel9B and Cel48A) were identified by a proteomic approach. In strain 20, Cel9B and Cel48A were identified as two major CBPs and as bacterial cell-associated proteins. The anti-Adh serum was also shown to target the C-terminal family 37 carbohydrate-binding module (CBM37) of Cel9B and Cel48A, indicating that this module, unique to R. albus, may play a significant role in bacterial adhesion to cellulose as suggested previously for R. albus 8. Overall, our results support the hypothesis of an adhesion mechanism involving the CBM37 of Cel9B and Cel48A. This adhesion mechanism may not be restricted to these two enzymes but may also involve other CBM37-containing proteins such as Cel5G and the other uncharacterised proteins recognized by the anti-Adh serum. Biological soil crusts (biocrusts) are photosynthetic mats formed through an association of prokaryotic and eukaryotic microorganisms with soil particles. Biocrusts are found in virtually any terrestrial ecosystem where vascular plant coverage is abiotically limited, with drylands comprising the primary habitat for them. We studied the dynamics of the active bacterial community in two biocrusts from an arid and a hyperarid region in the Negev Desert, Israel, under light‐oxic and dark‐anoxic incubation conditions after simulated rainfall. We used H218O for hydrating the crusts and analysed the bacterial community in the upper and lower parts of the biocrust using an RNA‐stable isotope probing approach coupled with 454‐pyrosequencing. In both biocrusts, two distinct bacterial communities developed under each incubation condition. The active anaerobic communities were initially dominated by members of the order Bacillales which were later replaced by Clostridiales. The aerobic communities on the other hand were dominated by Sphingobacteriales and several Alphaproteobacteria (Rhizobiales, Rhodobacterales, Rhodospirillales and Rubrobacteriales). Actinomycetales were the dominant bacterial order in the dry crusts but quickly collapsed and accounted for < 1% of the community by the end of the incubation. Our study shows that biocrusts host a diverse community whose members display complex interactions as they resuscitate from dormancy.  相似文献

Echolocating insectivorous bats consummate prey captures using a distinct vocal motor pattern commonly known as the terminal or feeding buzz, which is widely considered a fixed motor pattern executed independently of auditory feedback influences. The Mexican free-tailed bat, Tadarida brasiliensis, offers an opportunity to explore the role of sensory feedback in buzzing because they emit similar buzzes both in flight during foraging and while stationary as communication sounds. Here we compared the spectral and temporal patterns of foraging and communication buzzes to address whether or not auditory feedback may influence buzz patterns. We found that while foraging buzzes uttered in open space were composed of generic FM calls, communication buzzes were composed of an adapted CF–FM call similar to the call type used by T. brasiliensis when navigating in confined spaces. This provides the first evidence that some bats can make significant context-dependent changes in the spectral parameters of calls within their buzz. We also found that inter-pulse intervals, but not call durations, were different within the two buzz types. These observations indicate that though a common pattern generator hierarchically organizes all buzzes, T. brasiliensis retains a significant capacity to adapt the spectral and temporal patterns of elements within its buzzes.  相似文献

尖孢镰孢菌古巴专化型(Fusarium oxysporum f.sp.cubense)是香蕉枯萎病的病原菌,该菌是一种土壤习居菌,了解香蕉根区土壤中真菌多样性及镰孢菌属(Fusarium)真菌所占比例,对如何减少土壤中的病原菌、预防香蕉枯萎病的发生有重要的指导意义。该文通过采集不同宿根年限的香蕉健康植株和枯萎病植株的根区土壤,利用高通量测序技术测定土壤样品中的真菌种群。结果表明:(1)同一宿根年限的香蕉植株中,健康植株根区土壤中所获的reads及OTUs数量均高于枯萎病植株,说明健康植株根区土壤的真菌多样性丰富于枯萎病植株。(2)除了一年生香蕉枯萎病植株以担子菌门(Basidiomycota)为主外,其他土壤样品中均以子囊菌门(Ascomycota)为主,其中的丛赤壳科最高相对丰度来自三年生健康植株的根区土壤(26.02%),其次是五年生的枯萎病植株根区土壤(15.56%)。(3)在丛赤壳科中,镰孢菌属在三年生健康植株土壤中的相对丰度最高(2.54%),在其他样品中的相对丰度在0.1%~0.65%之间;在镰孢菌属中,腐皮镰孢菌(Fusarium solani)的相对丰度(0~1.59%之间)高于尖孢镰孢菌(F.oxysporum),尖孢镰孢菌仅占很小的比例(相对丰度0~0.08%之间)。可见,在不同香蕉植株的根区土壤中,健康植株的根区土壤真菌多样性高于枯萎病植株,无论是健康植株还是枯萎病植株的根区土壤中,作为香蕉枯萎病病原菌的镰孢菌属或尖孢镰孢菌的群体均不占主导地位。  相似文献

冬水田-水稻是川中丘陵区传统的稻田种植模式,冬水田种植模式转变是实现多熟种植及机械化的重要途径。为探究冬水田-水稻种植模式转旱作过程中作物季及休闲期土壤呼吸速率及其组分构成,试验设置冬水田-水稻转旱作(FTD)、冬水田-水稻(FR)和冬闲田-玉米(FM)3种不同种植模式,采用根排除法和静态明箱-气相色谱法原位取样测定作物季及季后休闲期土壤呼吸及其组分,并通过测算净生态系统生产力(NEP)进而判断冬水田-水稻转旱作过程的农田系统碳汇强度。结果表明:(1)FTD显著提高了土壤总呼吸速率及其自养和异养呼吸速率,从而提高了其累积排放量(P<0.05)。与FR相比,FTD的土壤总呼吸及其自养和异养呼吸的累积排放量分别提高了13.14倍、11.32倍和15.56倍(P<0.05);与FM相比,FTD的土壤总呼吸及其自养和异养呼吸的累积排放量分别提高了70.56%、40.83%和115.47%(P<0.05)。(2)与FR和FM相比,FTD均降低了土壤呼吸及其组分的温度敏感性(Q10),且土壤总呼吸的温度敏感性介于异养呼吸和自养呼吸之间。(3)FR,FM和FTD的净生态系统生产力(NEP)均为正值,其数值分别为7911.66 kg/hm2,5667.89 kg/hm2和1583.46 kg/hm2,均表现为大气CO2的碳汇,但与FR与FM相比,FTD显著降低了其净生态系统生产力,呈现出较弱的碳汇。  相似文献

BMI and percent body fat (%BF) are both related to height (Ht) in prepubertal children, so may misrepresent childhood adiposity, especially in tall or short children. We sought to construct replacement functions for BMI and %BF that are independent of Ht. Fat mass (FM) was measured using dual‐energy X‐ray absorptiometry, together with Ht and body mass (BM) in 746 healthy boys and girls aged 8 years (0.34 s.d.). Relationships between BM, FM, and Ht were measured and values of p and q derived such that the functions BM. Ht?p and FM.BM?q were unrelated to Ht. BM was not directly proportional to Ht2, BMI being significantly related to Ht in both boys and girls (P < 0.001). BM was proportional to Ht3, BM. Ht?3 being independent of Ht. Similarly, FM was not directly proportional to BM and %BF was significantly related to Ht (P < 0.001). While FM was proportional to BM2, FM.BM?1.5 was the function found to be independent of Ht. Using the 85th and 95th percentiles as the cutoffs for overweight and obesity respectively, 6.4% of the boys and 6.8% of the girls were classified differently by BMI and the Ht independent measure BM. Ht?3. Similarly, 10.1% boys and 13.7% girls were classified differently by %BF and the Ht independent measure FM.BM?1.5. We propose that improved diagnostic accuracy of body composition in 8‐year‐olds is provided by the BM function (BMF, BM. Ht?3) and FM function (FMF, FM.BM?1.5) replacing BMI and %BF, which both overestimate the adiposity of taller children and underestimate it in shorter children.  相似文献

Three ninhydrin-positive lipids of Streptomyces sioyaensis were found. These lipids were called substance A, B and C, tentatively. Study on the distribution of these lipids in Actinomycetales has shown that substance A was common in all of the strains tested, and that substance B was found in the limited strains. Summary The rufous horseshoe bat, Rhinolophus rouxi, was trained to discriminate differences in target distance. During the discrimination trials, the bats emitted complex FM/CF/FM pulses containing first harmonic and dominant second harmonic components.Loud free running artificial pulses, simulating the CF/FM part of the natural echolocation components, interfered with the ability of the bat to discriminate target distance. Changes in the frequency or frequency pattern of the artificial pulses resulted in systematic changes in the degree of interference. Interference occurred when artificial CF/FM pulses were presented at frequencies near those of the bat's own first or second harmonic components.These findings suggest that Rhinolophus rouxi uses both the first and second harmonic components of its complex multiharmonic echolocation sound for distance discrimination. Decrease in fat mass (FM) is a one of the aims of pediatric obesity treatment; however, measurement techniques suitable for routine clinical assessment are lacking. The objective of this study was to validate whole‐body bioelectrical impedance analysis (BIA; TANITA BC‐418MA) against the three‐component (3C) model of body composition in obese children and adolescents, and to test the accuracy of our new equations in an independent sample studied longitudinally. A total of 77 white obese subjects (30 males) aged 5–22 years, BMI‐standard deviation score (SDS) 1.6–3.9, had measurements of weight, height (HT), body volume, total body water (TBW), and impedance (Z). FM and fat‐free mass (FFM) were calculated using the 3C model or predicted from TANITA. FFM was predicted from HT2/Z. This equation was then evaluated in 17 other obese children (5 males) aged 9–13 years. Compared to the 3C model, TANITA manufacturer's equations overestimated FFM by 2.7 kg (P < 0.001). We derived a new equation: FFM = ?2.211 + 1.115 (HT2/Z), with r2 of 0.96, standard error of the estimate 2.3 kg. Use of this equation in the independent sample showed no significant bias in FM or FFM (mean bias 0.5 ± 2.4 kg; P = 0.4), and no significant bias in change in FM or FFM (mean bias 0.2 ± 1.8 kg; P = 0.7), accounting for 58% (P < 0.001) and 55% (P = 0.001) of the change in FM and FFM, respectively. Our derived BIA equation, shown to be reliable for longitudinal assessment in white obese children, will aid routine clinical monitoring of body composition in this population.  相似文献

Slash‐and‐burn clearing of forest typically results in increase in soil nutrient availability. However, the impact of these nutrients on the soil microbiome is not known. Using next generation sequencing of 16S rRNA gene and shotgun metagenomic DNA, we compared the structure and the potential functions of bacterial community in forest soils to deforested soils in the Amazon region and related the differences to soil chemical factors. Deforestation decreased soil organic matter content and factors linked to soil acidity and raised soil pH, base saturation and exchangeable bases. Concomitant to expected changes in soil chemical factors, we observed an increase in the alpha diversity of the bacterial microbiota and relative abundances of putative copiotrophic bacteria such as Actinomycetales and a decrease in the relative abundances of bacterial taxa such as Chlamydiae, Planctomycetes and Verrucomicrobia in the deforested soils. We did not observe an increase in genes related to microbial nutrient metabolism in deforested soils. However, we did observe changes in community functions such as increases in DNA repair, protein processing, modification, degradation and folding functions, and these functions might reflect adaptation to changes in soil characteristics due to forest clear‐cutting and burning. In addition, there were changes in the composition of the bacterial groups associated with metabolism‐related functions. Co‐occurrence microbial network analysis identified distinct phylogenetic patterns for forest and deforested soils and suggested relationships between Planctomycetes and aluminium content, and Actinobacteria and nitrogen sources in Amazon soils. The results support taxonomic and functional adaptations in the soil bacterial community following deforestation. We hypothesize that these microbial adaptations may serve as a buffer to drastic changes in soil fertility after slash‐and‐burning deforestation in the Amazon region.  相似文献

The effect of changing feeds on individual feed intake and feeding and dominance ranks in groups of African catfish Clarias gariepinus was investigated. Following feeding on a commercial feed groups (n = 3) of six African catfish were either fed fish meal (FM42) or maize gluten (MG35) based feeds for 5 days before being switched to the other feed for 5 days. Energy intake was significantly lower on FM42 than on MG35, dry matter intake and protein intake were significantly lower on FM42 than on commercial feed and this occurred whether FM42 was fed first or second. There were no significant differences between intake of MG35 and commercial feed. Thus, the action of changing the feed on its own did not affect feed intake since the decrease was shown to be feed‐specific to FM42. Six types of agonistic behaviours were identified and used to assign dominance rank. There were no correlations between dominance and feeding ranks. This was due to non‐linear hierarchies with one dominant fish in each group. Feeding ranks were more stable when feeding MG35 than FM42. Feeding rank stability (Kendall's coefficient of concordance) was significant in five out of six groups fed MG35 (compared with three out of six fed FM42). Feeding rank stability was higher in five out of the six groups when they were fed MG35 than when the same group was fed FM42. The experiment provided evidence that the introduction of a novel feed can, but does not necessarily, alter feed intake and that feed can influence the stability of feeding ranks.  相似文献

In vitro chromosome doubling was induced in octoploid (2n = 58) yacon using oryzalin and colchicine as mitotic spindle inhibitors. Nodal segments of in vitro cultured plants, 5–15 mm long, were exposed to 20, 25, or 30 μM oryzalin and 1, 3, or 5 mM colchicine for 24 or 48 h. The resulting ploidy level was determined by chromosome counting and flow cytometry. Out of 240 nodal segments, 3.33% hexadecaploid (2n = 116) plants were regenerated after the application of oryzalin. The greatest proportions of hexadecaploid plants (1.6%) were obtained after 48 h of 25 μM oryzalin treatment. With the colchicine treatment, only 0.42% hexadecaploid plants were detected and their survival rate was significantly lower in comparison with the oryzalin treatment. In hexadecaploid yacon, significantly higher levels of saccharides were detected (FOS 13.9 g/100 g FM, fructose 4.6 g/100 g FM and glucose 2.1 g/100 g FM) compared to the octoploid control (FOS 5.3 g/100 g FM, fructose 2.9 g/100 g FM and glucose 1.0 g/100 g FM). These results indicate that in vitro treatment of nodal segments with oryzalin solution could be an effective procedure for chromosome doubling and the polyploidy breeding can help to increase the FOS content in the tuberous roots.  相似文献

The bacterial community compositions in Chenopodium album and Stellaria media seeds recovered from soil (soil weed seedbank), from bulk soil, and from seeds harvested from plants grown in the same soils were compared. It was hypothesized that bacterial communities in soil weed seedbanks are distinct from the ones present in bulk soils. For that purpose, bacterial polymerase chain reaction denaturing gradient gel electrophoresis (PCR–DGGE) fingerprints, made from DNA extracts of different soils and seed fractions, were analyzed by principal component analysis. Bacterial fingerprints from C. album and S. media seeds differed from each other and from soil. Further, it revealed that bacterial fingerprints from soil-recovered and plant-harvested seeds from the same species clustered together. Hence, it was concluded that microbial communities associated with seeds in soil mostly originated from the mother plant and not from soil. In addition, the results indicated that the presence of a weed seedbank in arable soils can increase soil microbial diversity. Thus, a change in species composition or size of the soil weed seedbank, for instance, as a result of a change in crop management, could affect soil microbial diversity. The consequence of increased diversity is yet unknown, but by virtue of identification of dominant bands in PCR–DGGE fingerprints as Lysobacter oryzae (among four other species), it became clear that bacteria potentially antagonizing phytopathogens dominate in C. album seeds in soil. The role of these potential antagonists on weed and crop plant growth was discussed.  相似文献

The diversity and community structures of actinobacteria in saline sediments collected from Yunnan and Xinjiang Provinces, China, were investigated with cultivation and 16S rRNA gene analysis. A total of 163 actinobacterial isolates were obtained, and they were affiliated with the order Actinomycetales (distributed into five suborders: Streptosporangineae, Micrococcineae, Streptomycineae, Pseudonocardineae, and Glycomycineae). A total of 748 actinobacterial 16S rRNA gene clones were examined, and they could be classified into Actinomycetales, Acidimicrobiales, and unclassified actinobacteria. The Actinomycetales sequences were distributed into nine suborders: Streptosporangineae, Glycomycineae, Micromonosporineae, Pseudonocardineae, Corynebacterineae, Frankineae, Propionibacterineae, Streptomycineae, and Micrococcineae. The unclassified actinobacteria contained three new clusters at the level of subclass or order. Our 16S rRNA gene phylogenetic data indicated that actinobacterial communities were very diverse in the investigated saline sediments (salinity 0.4–11.6%) and some actinobacterial members may be halotolerant or halophilic. The actinobacterial community structures in the saline sediments were different from those in marine and freshwater environments. Our data have implications for a better understanding of the distribution of Actinobacteria in saline environments.
