Most estrous cycles in cows consist of 2 or 3 waves of follicular activity. Waves of ovarian follicular development comprise the growth of dominant follicles some of which become ovulatory and the others are anovulatory. Ovarian follicular activity in cows during estrous cycle was studied with a special reference to follicular waves and the circulating concentrations of estradiol and progesterone. Transrectal ultrasound examination was carried out during 14 interovulatory intervals in 7 cows. Ovarian follicular activity was recorded together with assessment of serum estradiol and progesterone concentrations. Three-wave versus two-wave interovulatory intervals was observed in 71.4% of cows. The 3-wave interovulatory intervals differed from 2-wave intervals in: 1) earlier emergence of the dominant follicles, 2) longer in length, and 3) shorter interval from emergence to ovulation. There was a progressive increase in follicular size and estradiol production during growth phase of each wave. A drop in estradiol concentration was observed during the static phase of dominant anovulatory follicles. The size of the ovulatory follicle was always greater and produced higher estradiol compared with the anovulatory follicle. In conclusion, there was a predominance of 3-wave follicular activity that was associated with an increase in length of interovulatory intervals. A dominant anovulatory follicle during its static phase may initiate the emergence of a subsequent wave. Follicular size and estradiol concentration may have an important role in controlling follicular development and in determining whether an estrous cycle will have 2 or 3-waves.  相似文献

Bovine viral diarrhea virus (BVDV) is a major cattle pathogen responsible for a spectrum of symptoms, including reproductive failure. This study was designed to establish the effects of BVDV infection on estradiol, progesterone and PGF2alpha secretion in the cow. Seven BVDV-free cows were challenged with non-cytopathogenic BVDV (strain Pe 515: 5x10(6) tissue culture infected dose50) so that peak viremia occurred during the initial phase of luteal development in a synchronized estrous cycle. Ovulation was also synchronized in 7 sham-infected animals. Within 2 wk of inoculation, viremia, leukopenia and serum neutralizing antibodies were recorded in all of the BVDV-infected cows but not the sham-infected animals. Between Day 4 and Day 9 post estrus the BVDV-infected cows had significantly (P<0.01) lower plasma estradiol levels than the sham-infected animals. However, the BVDV infection did not alter rectal temperatures, plasma progesterone concentrations or PGF2alpha secretion 17, 18 and 19 d post estrus. These data highlight a potential causal link between BVDV viremia, endocrine dysfunction and poor fertility in the cow.  相似文献

Folliculogenesis was studied daily in the 18 oestrous cycles in six prolific Olkuska ewes from October to December using transrectal ultrasonography to record the number and size of all ovarian follicles > or =2 mm in diameter. Blood samples were taken once a day and were analyzed for concentrations of FSH, LH, estradiol and progesterone. Follicular and hormonal data were analyzed for associations between different stages of development of the follicular waves and concentrations of FSH and estradiol. The first wave during which at least one follicle reached maximum diameter of > or =4 mm after ovulation, was defined as a wave 1, and the following waves were numbered sequentially. Waves 1, 2, 3, 4 and the ovulatory one emerged on days: -2 to 4, 4 to 8, 6 to 11, 10 to 12 and 11 to 15, respectively. The mean number of follicles per wave that reached diameter of > or =4 mm was 4.15 +/- 1.1 and 16.62 +/- 8.6 follicles per estrous cycle of a total 299 follicles were observed. Significantly more follicles (p> or =0.05) emerged on days 2, 8 and 13 than in other days. Serum FSH concentrations fluctuated from 0.11 ngml(-1) on day 2 to preovulatory maximum 1.81 ngml(-1) on day 17 of the estrous cycle. The emergence of follicular waves was associated with elevations of FSH concentrations in blood serum. The mean increase in FSH concentration was followed by the recruitment of follicles of the next wave. The mean daily FSH concentration and the mean number of follicles emerging each day were negatively correlated. The length of the interwave interval (4.4 +/- 1.6 days) did not differ significantly from the interval between pulses of FSH (4.8 +/- 0.3 days). The mean serum estradiol concentrations showed fluctuations until day 14 and then gradually increased from 5.47 +/- 0.3 pgml(-1) to reach a peak 13.14 +/- 0.2 pgml(-1) on the day before ovulation. To summarize, the growth of ovarian follicles during the estrous cycle in high fecundity Olkuska sheep exhibited a distinct wave-like pattern. Ovarian follicles emerged from the pool of 2 mm follicles. The preovulatory follicles originated from the large follicle population were present in the ovary at the time of luteal regression. The initial stages of the growth of the largest follicles appears to be controlled primarily by increases in FSH secretion.  相似文献

The objective was to examine follicular and luteal development and function in mature, lactating beef cows grazing endophyte free (E-) or endophyte infected (E+) tall fescue during the early postpartum period. Angus, Hereford, and Angus x Hereford cows were exposed to pasture for 37-39 days before synchronized estrus. Serum concentrations of prolactin were evaluated during the luteal phase before the synchronized estrus. Every Monday, Wednesday, and Friday for one estrous cycle ovaries were monitored by transrectal ultrasonography and blood was collected for determination of serum concentrations of progesterone and estradiol in cows that responded to synchronization. Signs of fescue toxicosis in E+ cows included decreased serum concentrations of prolactin (84.9+/-13.6 pg/ml versus 32.3+/-12.0 pg/ml; P < 0.009) measured during the luteal phase (day 37 of grazing) and decreased body condition of cows and weight of cows and calves (P < 0.001). Neither serum concentrations of progesterone or estradiol, nor diameter of the CL differed between treatments. Diameter of the largest follicle tended to be smaller for cows grazing E+ fescue, especially between days 8 and 12 of the estrous cycle (P < 0.08). Numbers of class 1 (3-5 mm) and class 3 (>10 mm) follicles were similar (P > 0.05) between treatments, but number of class 2 (6-9 mm) follicles was reduced in E+ cows for most of the cycle (days 10 through 20; P < 0.03). Length of synchronized estrous cycle, days open, calving interval, and pregnancy rate at 30, 45, 60, and 90 days post-breeding was similar (P > 0.05) among treatment groups. Even though follicular dynamics (diameter of the largest follicle and number of class 2 follicles) were altered in cows grazing E+ tall fescue, follicular function was apparently not affected by ergot alkaloids.  相似文献

Elephants express two luteinizing hormone (LH) peaks timed 3 wk apart during the follicular phase. This is in marked contrast with the classic mammalian estrous cycle model with its single, ovulation-inducing LH peak. It is not clear why ovulation and a rise in progesterone only occur after the second LH peak in elephants. However, by combining ovarian ultrasound and hormone measurements in five Asian elephants (Elephas maximus), we have found a novel strategy for dominant follicle selection and luteal tissue accumulation. Two distinct waves of follicles develop during the follicular phase, each of which is terminated by an LH peak. At the first (anovulatory) LH surge, the largest follicles measure between 10 and 19.0 mm. At 7 ± 2.4 days before the second (ovulatory) LH surge, luteinization of these large follicles occurs. Simultaneously with luteinized follicle (LUF) formation, immunoreactive (ir) inhibin concentrations rise and stay elevated for 41.8 ± 5.8 days after ovulation and the subsequent rise in progesterone. We have found a significant relationship between LUF diameter and serum ir-inhibin level (r(2) = 0.82, P < 0.001). The results indicate that circulating ir-inhibin concentrations are derived from the luteinized granulosa cells of LUFs. Therefore, it appears that the development of LUFs is a precondition for inhibin secretion, which in turn impacts the selection of the ovulatory follicle. Only now, a single dominant follicle may deviate from the second follicular wave and ovulate after the second LH peak. Thus, elephants have evolved a different strategy for corpus luteum formation and selection of the ovulatory follicle as compared with other mammals.  相似文献

The objective was to determine the effects of the duration of progesterone exposure during the ovulatory wave on fertility (pregnancy rate) in beef cattle. We tested the hypothesis that short-progesterone exposure during the growing and early-static phase of the ovulatory follicle (analogous to the ovulatory wave of 3-wave cycles) is associated with higher fertility than a longer duration of exposure (analogous to the ovulatory wave of 2-wave cycles). Three to 5 days after ovulation, beef heifers (n = 172) and suckled beef cows (n = 193) were given an intravaginal progesterone-releasing device (CIDR) and 2.5 mg estradiol - 17β +50 mg progesterone im to induce a new follicular wave. Cattle were allocated to short- or long-progesterone exposure groups (for 3 and 6 d after wave emergence, respectively) after which prostaglandin F was administered and CIDR were removed. Forty-eight hours later, all cattle were given 12.5 mg pLH and artificially inseminated (AI) with frozen-thawed semen. The diameter of the two largest follicles and the corpus luteum were measured by transrectal ultrasonography at CIDR removal, insemination, and 36 h after insemination. Pregnancy diagnosis was done ultrasonically 38 and 65 d post-AI. There was no difference in pregnancy rates in short- vs long-progesterone exposure in heifers (53 vs 47%, P = 0.44) or cows (63 vs 58%, P = 0.51). However, the diameter of the ovulatory follicle at CIDR removal and AI was smaller in short- than in long-progesterone groups (P < 0.02), and larger in cows than in heifers (P < 0.006). In conclusion, short-progesterone exposure during the growing and early-static phase of the ovulatory follicle (similar to 3-wave cycles) was not associated with higher fertility than a longer progesterone exposure (similar to 2-wave cycles).  相似文献

The objective of this study was to determine the efficacy of a progesterone-releasing intravaginal silastic device (Controlled Internal Drug Release: CIDR) for inducing ovulation in beef cows with persistent ovarian cysts. Fifteen cows with cysts and abnormal cycles for over 40 days were randomly assigned to receive either a single CIDR (CIDR group, n=9), or a CIDR containing no progesterone (blank CIDR) (BLANK group, n=6) for about 14 days. Determination of plasma progesterone levels at the beginning of CIDR treatment indicated 4 of 6 BLANK cows with non-luteinized cysts and 5 of 9 CIDR cows with non-luteinized cysts. In 5 of 6 BLANK cows, one follicular wave appeared and newly emerged dominant follicles increased in size up to 20 mm in diameter and persisted during the experiment, while one cow experienced estrus with spontaneous ovulation. In contrast, during CIDR treatment, 2 or 3 waves, in which dominant follicles were from 7 to 15 mm in diameter, appeared approximately at 7-day intervals. Within 3 days after CIDR removal, estrous behavior was detected followed by ovulation of the dominant follicle in the last wave. All CIDR cows resumed normal cyclicity with 2 follicular waves for over 2 months. Insertion of a CIDR caused a rapid increase of about 2 ng/mL in plasma progesterone. The levels were greater than 1.3 ng/mL until removal of a CIDR, then dropped under 0.3 ng/mL. Concentrations of plasma estradiol in BLANK cows increased during growth of the cystic follicles, with high levels greater than 10 pg/mL for over 10 days. In 4 of 5 cows with non-luteinized cysts, with high plasma estradiol on the day of CIDR insertion, CIDR treatment resulted in rapid decline of estradiol levels. During placement of the CIDR, estradiol levels showed no increase in the growth phase of a newly appeared dominant follicle. After CIDR removal, however, estradiol significantly increased associated with the growth of ovulatory follicles in all 9 cows. A transient increase in plasma FSH levels preceded detection of each follicular or cyst wave in both BLANK and CIDR cows. Pulse frequency and mean concentration of LH in cows with non-luteinized cysts showed values corresponding to those in normal follicular phase. However, throughout CIDR treatment, these parameters reduced to levels found in the normal luteal phase. In cows with luteinized cysts, parameters of LH secretion were as low as in the normal luteal phase before and during CIDR treatment, then increased significantly after CIDR removal. Present results indicate that treatment with CIDR proved effective in restoring ovulation and reestablishing normal cyclicity in beef donor cows with cysts persistent for a long period. The CIDR reduced and maintained LH secretion at normal luteal levels, thereby, inducing atresia of estrogen-active cysts and preventing formation of cysts from the newly emerged follicles.  相似文献

The aim of this study was to determine if the duration of treatment with an intravaginal progesterone releasing insert (IVP(4)) after treatment with oestradiol benzoate (ODB) at the time of insertion and 24 h after removal would affect selected variables including: size of ovarian follicles at the time of removal of inserts, diameter of ovulatory follicles, plasma concentrations of progesterone following ovulation, and duration of the following oestrous cycle. Characteristics of oestrus at a synchronised and spontaneous oestrus were also monitored. Non-lactating Holstein cows were synchronised with an IVP(4) for 5 (n = 10), 7 (n = 10), 8 (n = 9) or 9 (n = 9) days together with injections of ODB at device insertion (2 mg) and 24 h after removal (1 mg). Ultrasonography showed no significant effect of treatment on the day of emergence of preovulatory follicles relative to the day of removal of inserts (overall mean = -4.22 +/- 0.58; P = 0.15) for cows that ovulated within 120 h insert removal (n = 36). Treatment with ODB and an IVP(4) for 5 days reduced the diameter of preovulatory follicles at the time of removal of inserts and for the following 2 days compared to cows treated for 7-9 days (mean difference 2.56 +/- 1.15 mm; P = 0.033) but did not reduce the diameter of the ovulatory follicle (P = 0.21). Day of emergence relative to removal of inserts was associated with the diameter of the ovulatory follicle (R2 = 0.69; P < 0.001). Concentrations of progesterone and the diameter of the corpus luteum following ovulation were not affected by treatment (P > 0.20), but were affected by the diameter of the ovulatory follicle (P < 0.01). Diameter of the ovulatory follicle did not affect interoestrous and interovulatory intervals (P > 0.40). We conclude that treatment with an IVP(4) for 5 compared to 7-9 days with ODB administered at device insertion, and 24 h after removal reduced the diameter of preovulatory follicles at the time of removal of the insert but did not reduce the diameter of the ovulatory follicle or concentrations of progesterone in plasma. Emergence of preovulatory follicles closer to the time of removal of inserts reduced the diameter of the ovulatory follicle when oestrus was induced with ODB. Ovulation of smaller follicles reduced concentrations of progesterone in plasma following ovulation but did not affect oestrous cycle duration.  相似文献

The objective of this study was to examine changes in intrafollicular concentrations of inhibins and steroids in heifers during growth of dominant follicles. To obtain dominant ovulatory follicles, heifers received injections of prostaglandin (PG) on Day 9 of an estrous cycle and were ovariectomized (OVX) 0, 24, 48, 60, or 72 h after injection. To obtain dominant nonovulatory follicles, heifers were OVX on Day 3, 6, or 9 of a cycle. Follicular size was determined, follicular fluid (FF) was collected from follicles 6 mm or greater in diameter, and RIA was used to quantify concentrations of inhibins, estradiol, and progesterone in FF. During growth of dominant ovulatory follicles, concentrations of estradiol and progesterone increased, whereas inhibins decreased when compared with dominant follicles on Day 9 before PG treatment. Concentrations of inhibins were inversely correlated with size and concentrations of estradiol in dominant ovulatory follicles. As dominant nonovulatory follicles increased in size, concentrations of inhibins, estradiol, and progesterone increased. Concentrations of inhibins were positively correlated with size and with progesterone concentrations in dominant nonovulatory follicles. Concentrations of inhibins were greater in dominant nonovulatory follicles than in atretic follicles. In summary, intrafollicular concentrations of inhibins decreased during growth of dominant ovulatory follicles, but increased during growth of dominant nonovulatory follicles. Because of the well-known suppressive action of inhibins on FSH secretion, we hypothesize that inhibins are involved in growth and atresia of dominant follicles during the bovine estrous cycle.  相似文献

Nutritionally induced anovulatory and cyclic Angus x Hereford heifers were used to evaluate follicular growth and concentrations of hormones and metabolites during anovulation and resumption of ovulation. Anovulatory heifers were fed to gain 0.6 (LGAIN) or 1.5 (HGAIN) kg/day until resumption of ovulation, and heifers with normal estrous cycles were fed a maintenance diet (M). Follicles >/= 4 mm in diameter were measured by daily ultrasonography in HGAIN and LGAIN heifers during one follicular wave before realimentation (Wan) and in two waves (W-2, W-1) immediately before the wave resulting in first ovulation or luteinization (W0). Ovaries of M heifers were evaluated to determine the day of ovulation of the second-wave dominant follicle (DF). Resumption of ovulation after realimentation occurred 23 days earlier in HGAIN than in LGAIN. Maximum diameter, growth rate, and persistence of dominant follicles increased, while persistence of first subordinate follicles decreased between anovulation and resumption of ovulation in anovulatory heifers. Concentrations of LH in serum were similar for HGAIN and LGAIN and gradually increased during realimentation. The increase in estradiol before the first ovulation was less in realimented heifers compared with cyclic heifers. Concentrations of insulin-like growth factor-I (IGF-I) in HGAIN and LGAIN gradually increased during realimentation but were lower than concentrations of IGF-I in cyclic heifers at ovulation. Increased diameter, growth rate, and persistence of the DF were associated with increased concentrations of LH, estradiol, and IGF-I during the transition from nutritionally induced anovulation to resumption of ovulatory cycles.  相似文献

The objective of the experiment was to examine the interaction of endophyte-infected tall fescue and environmental temperature on follicular and luteal development and function in beef heifers. Heifers were fed endophyte-free or endophyte-infected tall fescue seed at thermoneutral or heat stress temperatures (n = 6/treatment) 4 wk before and 3 wk after synchronized ovulation. All heifers were subjected to thermoneutral conditions (19 degrees C, 50% relative humidity) from Days -7 to -2; temperature increased incrementally from Days -1 to 0 and cycled between 25 degrees C and 31 degrees C between Days 1 and 20 for heat-stressed heifers. Serum was collected and ovaries monitored every other day after induced luteolysis between Days 1 and 23 or until ovulation. Size and location of follicles >4 mm and corpora lutea were recorded. Serum concentrations of prolactin were reduced in heat-stressed heifers fed infected seed and both heat stress and infected seed decreased total cholesterol. Rectal temperature and respiration rate were greatest in heifers fed the infected seed when exposed to maximal temperatures. Heat stress led to reduced diameter of the corpus luteum and serum progesterone compared with thermoneutral conditions. Progesterone was reduced more so in heifers fed infected seed. The combination of infected seed and heat stress was associated with reduced diameter of the preovulatory dominant follicle, and consumption of infected seed led to fewer large follicles during the estrous cycle. Both stressors led to reduced serum estradiol. Impaired follicle function may explain reduced pregnancy rates commonly observed in heifers grazing infected tall fescue pasture.  相似文献

The pattern of ovarian follicle development in maiden cyclic lambs was characterized using the definition of a follicle wave as the changes in the number of follicles among the days of the estrous cycle, as originally defined in cattle by Rajakoski in 1960. We also examined the steroid content relationships among follicles on Days 5 (Wave 1) and 14 (Waves 2 and 3) of the estrous cycle. In Experiment 1, the ovaries of 20 cyclic lambs (40 to 45 kg) were examined daily using transrectal ultrasonography for 1 or 2 estrous cycles (n = 31 cycles). The number of small (2 and 3 mm in diameter), medium (4 and 5 mm) and large (> or = 6 mm) follicles were aligned with the beginning and end of the average length estrous cycle and then compared among days. Identified follicles were defined as those that grew to > or = 4 mm and remained at > or = 3 mm for > or = 3 d. The number of identified follicles emerging (retrospectively identified at 2 or 3 mm) per ewe per day was also aligned with the average length estrous cycle. In Experiment 2, ewe lambs were ovariectomized on Day 5 (n = 6) or 14 (n = 5) of the estrous cycle, then follicle diameters and follicular fluid concentrations of estradiol and progesterone were compared among follicles. Data were analyzed by repeated measures ANOVA and compared among days using Fisher's LSD. In Experiment 1, either 2 (n = 10 cycles), 3 (n = 20 cycles) or 4 (n = 1 cycle) periods of emergence of identified follicles occurred during individual cycles, with estrous cycle lengths of 15.6 +/- 1.6, 16.1 +/- 1.1 and 17 d respectively. In animals with 2 or 3 periods of emergence of identified follicles, the total number of small, medium and large follicles differed (P < 0.05) among days of the estrous cycle showing a wave-like pattern. In Experiment 2, a single follicle collected on each of Days 5 and 14 of the cycle (6.2 +/- 0.2 and 3.9 +/- 0.2 mm in diameter) had a higher (P < 0.05) concentration of follicular fluid estradiol (36.2 +/- 4.4 and 50.9 +/- 21.6 ng/mL) than other follicles collected on the same day (next largest follicle: 4.3 +/- 0.3 and 3.5 +/- 0.4 mm; 4.3 +/- 0.9 and 18.2 +/- 6.7 ng/mL estradiol). The results showed that 1) there was a synchronous emergence of follicles associated with fluctuations in the number and size of follicles during the estrous cycle; 2) within a wave there was a hierarchy among follicles for diameter and steroid content; 3) ovarian follicle growth in ewe lambs occurred in 2 or 3 organized waves during the estrous cycle.  相似文献

This is the first paper to describe ovarian changes associated with follicular growth, ovulation, and corpus luteum (CL) formation as monitored by ultrasonography in a multiovular primate, the marmoset monkey (Callithrix jacchus). Examinations were carried out transabdominally on unsedated females using a 10 MHz probe. Cycles were monitored by plasma progesterone and controlled by administration of prostaglandin F2α (PGF). The reliability of ultrasound was validated by comparing findings with direct observation of the ovaries at laparotomy. In eight females, 25 follicles were counted, of which 92% were depicted correctly by ultrasound. Of 14 CLs in five females, number and position were confirmed at laparotomy for 78%. Ultrasound examinations of ovaries throughout the follicular and luteal phase were performed in eight cycles and related to plasma profiles of luteinizing hormone (LH) and progesterone. One of these cycles was anovulatory. In the remaining seven cycles, 19 follicles were considered ovulatory follicles since they were seen on consecutive days and found again as CLs. Growth of individual follicles was monitored by measurements of follicle diameter from day 7 onward. Disappearance of follicles or changes in echogenicity were noted between days 9 and 11, preventing further measurements. Mean follicle size increased from 2.1 mm (range 1.6 mm–2.7 mm) on day 7 to 3.2 mm (range 2.7 mm–4.0 mm) on the day last seen. With one exception, the day follicles were last seen by ultrasound was consistent with the day of the preovulatory LH surge (day 9–11). The postovulatory rise in progesterone occurred 1–2 days later (day 11–13). These findings suggest that the day of ovulation as observed by ultrasound was characterized by either disappearance of follicles or increased follicular echogenicity. In conclusion, ultrasonography provides a reliable, noninvasive method for examinations of the ovarian cycle in the marmoset monkey. © 1996 Wiley-Liss, Inc.  相似文献

Color Doppler transrectal ultrasound was used to evaluate blood flow area in the wall of dominant anovulatory follicles versus ovulatory follicles in mares during the transition between anovulatory and ovulatory seasons. Daily examinations were done in 11 control mares toward the end of the anovulatory season. In 13 separate mares, follicular fluid was collected from 30-mm follicles, and blood flow areas from control mares were used as a basis for designating the sampled follicle as either anovulatory or ovulatory. Blood flow area in the controls ranged from 0.18 to 0.35 cm(2) in six mares on the day of a 30-mm anovulatory follicle and from 0.25 to 0.86 cm(2) in 11 mares on the day of a 30-mm ovulatory follicle; the ranges did not overlap except for one follicle. In the controls, mean blood flow area was lower (P < 0.05) in the anovulatory group than in the ovulatory group for each day beginning with the first Doppler examination at 25 mm. For plasma LH in controls, an effect of follicle group (P < 0.0001) and an interaction (P < 0.0001) of group by day reflected lower (P < 0.05) concentrations in the anovulatory group on Days -6, -2, and 5-8 (Day 0 = 30-mm follicle). For plasma FSH, an interaction (P < 0.0001) reflected higher (P < 0.05) concentrations in the anovulatory group on Days -3 and 1-4. More (P < 0.05) statistically identified FSH surges occurred in the anovulatory group during Days -7 to 8. In the sampled mares, follicular-fluid concentrations of estradiol, free insulin-like growth factor-1, inhibin-A, and vascular endothelial growth factor were lower (P < 0.05) in 30-mm designated anovulatory follicles than in 30-mm designated ovulatory follicles. Results were interpreted as follows: 1) The future anovulatory dominant-sized follicle developed under an LH deficiency, 2) the LH deficiency led to reductions in blood flow area and in concentrations of follicular-fluid factors, and 3) the reduction in follicle production of FSH suppressors resulted in higher plasma FSH concentrations.  相似文献

Transitions from the anovulatory to the ovulatory season (n=20) and ovulatory to anovulatory season (n=11), were monitored daily by transrectal ultrasonography in wapiti. In 17 of 20 observations, the first interovulatory interval (IOI) was short (9.1+/-0.3 days; mean+/-S.E.M.) compared with later in the ovulatory season (21.3+/-0.1) and the last IOI (21.2+/-0.6 days). With one exception, the short IOI were composed of only one wave of follicular development. Subsequent IOI were composed of two or three waves. Maximum diameters of the first two ovulatory follicles were similar (11.3+/-0.4 mm versus 11.3+/-0.2 mm), but both were larger (P<0.05) than the last two ovulatory follicles of the ovulatory season (10.3+/-0.3 and 10.1+/-0.4 mm). Multiple ovulations occurred in three hinds at the first ovulation of the season and in one hind at the second ovulation, but were not at any other time. Day-to-day profiles of CL diameter and plasma progesterone concentration were smaller (P<0.05) for short versus long IOI. Maximum diameter (12.8+/-0.6 mm versus 12.5+/-0.6 mm) and the diameter profile of the last CL of the season were not different from that of the previous CL. In summary, transition to regular ovulation occurred over a 3-week interval and was preceded by one short IOI (9 days). Multiple ovulations were detected only at the onset of the ovulatory season. The characteristics of the last IOI of the ovulatory season were similar to those reported during the rut. The wave pattern of follicle development was maintained throughout both fall and winter transition periods and follicular wave emergence was preceded by a surge in serum FSH concentrations. Transition to anovulation occurred over a 3-month interval and was marked by a failure of the dominant follicle to ovulate after a typical luteal phase.  相似文献

This study was conducted to test the hypothesis that supplementation of growing follicles with LH during the early spring transitional period would promote the development of steroidogenically active, dominant follicles with the ability to respond to an ovulatory dose of hCG. Mares during early transition were randomly assigned to receive a subovulatory dose of equine LH (in the form of a purified equine pituitary fraction) or saline (transitional control; n = 7 mares per group) following ablation of all follicles >15 mm. Treatments were administered intravenously every 12 h from the day the largest follicle of the post-ablation wave reached 20 mm until a follicle reached >32 mm, when an ovulatory dose of hCG (3000 IU) was given. Saline-treated mares during June and July were used as ovulatory controls. In a preliminary study, injection of this pituitary fraction (eLH) to anestrus mares was followed by an increase in circulating levels of LH (P < 0.01) but not FSH (P > 0.6). Administration of eLH during early transition stimulated the growth of the dominant follicle (Group x Day, P < 0.00001), which attained diameters similar to the dominant follicle in ovulatory controls (P > 0.1). In contrast, eLH had no effect on the diameter of the largest subordinate follicle or the number of follicles >10 mm during treatment (P > 0.3). The numbers of mares that ovulated in response to hCG in transitional control, transitional eLH and ovulatory control groups (2 of 2, 3 of 5 and 7 of 7, respectively) were not significantly different (P > 0.1). However, after hCG-induced ovulation, all transitional mares returned to an anovulatory state. Circulating estradiol levels increased during the experimental period in ovulatory controls but not in transitional eLH or transitional control groups (Group x Day, P = 0.013). In addition, although progesterone levels increased after ovulation in transitional control and transitional eLH groups, levels in these two groups were lower than in the ovulatory control group after ovulation (Group, P = 0.045). In conclusion, although LH supplementation of early transitional waves beginning after the largest follicle reached 20 mm promoted growth of ovulatory-size follicles, these follicles were developmentally deficient as indicated by their reduced steroidogenic activity.  相似文献

Reproductive performance is reportedly reduced in some rhesus monkeys during the summer months, even when environmental conditions are controlled. The mechanism(s) underlying this phenomenon remain unknown. We noted that the pattern of folliculogenesis appeared to be altered in rhesus monkeys that continued to exhibit ovulatory menstrual cycles during the "nonbreeding" season. This study was designed to investigate the effect of season on development of the dominant follicle (DF) and upon levels of serum gonadotropins and sex steroids in animals maintained in a controlled environment. Forty-four menstrual cycles were evaluated from October, 1982 to October, 1983. Animals were housed individually in controlled light (12L:12D) and temperature (22-25 degrees C). A DF was identified by laparoscopy on Day 6 of the cycle in only 45% of cycles during the months of May through September, compared with 87.5% the remainder of the year. No effect of season was detected on either the length of the menstrual cycle or luteal phase, mean follicular diameter, or the percentage of ovulatory cycles. During the follicular phase, amounts of follicle-stimulating hormone (FSH) in peripheral sera were depressed, whereas those of luteinizing hormone (LH) were consistently elevated. Amounts of circulating estradiol were similar between groups. However, serum concentrations of progesterone were markedly reduced in the summer. Development of the DF appeared to be delayed in the early follicular phase during the summer months in those rhesus monkeys that had ovulatory menstrual cycles. This delay was accompanied by an alteration in the FSH to LH ratio. Although most cycles were ovulatory, altered follicular development resulted in deficient luteal function.  相似文献

The reproductive tracts of 13 mature hinds were examined daily by transrectal ultrasonography and blood samples were taken daily from October to January to characterize follicular, luteal, and endocrine dynamics in wapiti during the estrous season. Follicle development occurred in waves characterized by regular, synchronous development of a group of follicles in temporal succession to a surge in serum FSH concentration. The mean interovulatory interval was 21.3 +/- 0.1 d, but was shorter in hinds exhibiting two follicular waves than in hinds exhibiting three and four waves (P < 0.05). The interwave interval was similar among waves in two-wave cycles and the first wave of three-wave cycles. All other interwave intervals in three- and four-wave cycles were shorter (P < 0.05). The maximum diameter of the dominant follicle of the first wave was similar among two-, three-, and four-wave cycles. For all other waves in three- and four-wave cycles, the maximum diameter was smaller (P < 0.05). Corpus luteum diameter and plasma progesterone concentrations were similar between two- and three-wave cycles, but the luteal phase was longer (P < 0.05) in four-wave cycles. The dominant follicle emerged at a diameter of 4 mm at 0.4 +/- 0.1 and 0.8 +/- 0.1 d before the largest and second largest subordinate follicles, respectively. The follicle destined to become dominant was larger (P < 0.05) than the largest subordinate follicle one day after emergence, which coincided with the first significant decrease in serum FSH concentration. We concluded that the estrous cycle in wapiti is characterized by two, three, or four waves of follicular development (each preceded by a surge in circulating FSH), that there is a positive relationship between the number of waves and the duration of the cycle, and an inverse relationship between the number of waves and the magnitude of follicular dominance (diameter and duration of the dominant follicle).  相似文献

The mare is a good comparative model for study of ovarian follicles in women, owing to striking similarities in follicular waves and the mechanism for selection of a dominant follicle. Commonality in follicle dynamics between mares and women include: (1) a ratio of 2.2:1 (mare:woman) in diameter of the largest follicle at wave emergence when the wave-stimulating FSH surge reaches maximum, in diameter increase of the two largest follicles between emergence and the beginning of deviation between the future dominant and subordinate follicles, in diameter of each of the two largest follicles at the beginning of deviation, and in maximum diameter of the preovulatory follicle; (2) emergence of the future ovulatory follicle before the largest subordinate follicle; (3) a mean interval of 1 day between emergence of individual follicles of the wave; (4) percentage increase in diameter of follicles for the 3 days before deviation; (5) deviation 3 or 4 days after emergence; (6) 25% incidence of a major anovulatory follicular wave emerging before the ovulatory wave; (7) 40% incidence of a predeviation follicle preceding the ovulatory wave; (8) small but significant increase in estradiol and LH before deviation; (9) cooperative roles of FSH and insulin-like growth factor 1 and its proteases in the deviation process; (10) age-related effects on the follicles and oocytes; (11) approximate 37-hour interval between administration of hCG and ovulation; and (12) similar gray-scale and color-Doppler ultrasound changes in the preovulatory follicle. In conclusion, the mare may be the premier nonprimate model for study of follicle dynamics in women.  相似文献

Computer-assisted image analysis was used to evaluate ultrasound images of bovine ovarian follicles. The ovaries of 8 sexually mature heifers were examined daily by transrectal ultrasonography for 2 estrous cycles. Ultrasonographic examinations of the ovaries were then videotaped, and the dominant and subordinate follicles of successive waves were individually identified and monitored. Recorded images of the dominant anovulatory follicle of the first wave (n = 15) and the ovulatory follicle of the last wave (n = 15) of the estrous cycle were subsequently digitized for computer analysis of echotexture (mean pixel value and pixel heterogeneity). Regions of the image spanning the breadth of the follicle wall were selected, and image analysis revealed that mean pixel value of the dominant anovulatory follicle changed over time (P = 0.0005). Mean pixel value decreased (P = 0.0005) dramatically during the early static phase (Days 6 to 8, Day 0 = day of ovulation), increased (P = 0.0005) at the onset of the regressing phase (Day 12), and reached maximal levels (P = 0.0005) on Day 14. Similarly, image echotexture of the ovulatory follicle revealed a time-dependent effect (P = 0.0001) due to a rapid decrease in mean pixel values between 7 and 4 d before ovulation, followed by an increase until the day before ovulation. The echotexture of images of the follicular antrum were also evaluated and with regard to the dominant anovulatory follicle, a time-dependent effect was not detected for mean pixel value (P = 0.62) but was observed for pixel heterogeneity (P = 0.02). In addition, there was a positive correlation between mean pixel value and heterogeneity (r = 0.61, P = 0.0001). Heterogeneity initially decreased (P = 0.02) and remained low until the emergence of the second follicular wave (mean Day 9). Values subsequently increased and became variable during the late static and regressing phases (> Day 9). Mean pixel value of the antrum of the dominant ovulatory follicle increased (P = 0.0001) as the day of ovulation approached. Heterogeneity did not change (P = 0.14), nor was there any correlation between mean pixel value and heterogeneity for the antrum of the ovulatory follicle (r = 0.06, P = 0.49). We concluded that changes in echotexture (mean pixel value and heterogeneity) of bovine ovarian follicles assessed by computer analysis of ultrasound images were temporally related to functional status (i.e., anovulatory versus ovulatory; growing, static or regressing). The results were strongly supportive of the concept that ultrasonographically detected image attributes are a reflection of physiologic status.  相似文献
