Summary The removal of calcium ions from Murashige and Skoog culture medium induced a marked increase in the accumulation of cardenolides in cell suspension cultures of Digitalis thapsi. Cell viability was not affected although growth was slightly reduced. Strontium ions could substitute for calcium in inhibiting cardenolide production, this effect of calcium being reversed by the addition of LaCl3 or ethyleneglycol-bis-(-aminoethyl ether)-N,N-tetraacetic acid. The results suggest that calcium, apart from its general effects on growth, may play a role in the regulation of cardenolide metabolism in a concentration dependent manner.Abbreviations BA

The effects of the auxins 2,4-D, NAA and IAA either alone or in combination with kinetin or BA were investigated to assess the morphogenetic potential of leaf, root and hypocotyl explants of Digitalis thapsi. Calluses were obtained from the three explants in basal medium without the addition of growth regulators and in leaves, the calluses formed roots. Application of 2,4-D, NAA or BA increased callus formation. The presence of NAA induced root formation and that of BA induced shoot formation via callus interphase. Indole-3-acetic acid alone only induced the generation of roots in the hypocotyl callus. Kinetin was ineffective in all the explants tested. Combinations of NAA with kinetin or BA were more effective in inducing organogenesis in leaf explants. Optimum responses were obtained in hypocotyl and root explants by using IAA in combination with BA, the highest rate of shoot regeneration being observed in hypocotyl explants.Rooting of the differentiated shoots was readily achieved in media without growth regulators. Regenerated plantlets were transferred to soil and grew with a survival rate of 70%.Abbreviations BA

The effect of calcium-deprivation on growth and the production of cardenolides in two undifferentiated cell lines of Digitalis thapsi maintained under three different light regimes (16 h photoperiod, darkness, or continuous light) was investigated. Growth
was stimulated by continuous light in both cell lines cultured in complete medium. The light regime did not affect cardenolide
accumulation in the cells of the hypocotyl-derived line; by contrast, continuous light or darkness increased the production
in the leaf-derived line. The elimination of calcium favoured cardenolide production independently of the origin of the suspensions
and the light regime, this beneficial effect being predominantly manifested under continuous light. 相似文献

An in vitro propagation system for Artemisia vulgaris L., a traditional medicinal plant, has been developed. The best organogenic response, including adventitious shoot number
and elongation, was obtained when hypocotyl segments were cultured onto MS medium supplemented with 4.54 μM TDZ (N-phenyl-N′-(1,2,3-thidiazol-yl) urea). Up to 28 shoots formed per explant for an optimal duration of exposure of 48 days. Regenerated
shoots formed roots when subcultured onto a medium containing 8.56 μM IAA (indole-3-acetic acid). Healthy plantlets were transferred
to a garden soil:farmyard soil:sand (2:1:1) mixture for acclimatization, which was successful, and subsequent maturity was
achieved under greenhouse conditions over a six-month period. The survival rate of the plantlets varied under acclimatization.
The regeneration protocol developed in this study provides a basis for germplasm conservation and for further investigation
of medicinally active constituents of A. vulgaris. This optimized protocol has been successfully employed for genetic transformation studies in A. vulgaris, which are currently underway in our laboratory. 相似文献

A procedure is described for the routine establishment and multiplication of red clover, Trifolium pratense L., shoot tips which should be applicable to a wide genotypic background. The addition of CO2 to the culture vial or use of polypropylene closures enhanced shoot multiplication at high levels of benzyladenine (BA). Horizontal orientation of crown buds resulted in more efficient multiplication. Culture of nodes from flowering stems was unsuccessful. The cytokinin BA was most effective for shoot multiplication at 2.0 mg/l with maximum shoot production by four weeks. A comparison of several genotypic sources revealed a 10-fold range in response to the multiplication medium with no differences observed among agronomic type or ploidy level. Factors favouring pollen callus proliferation, induction of embryogenesis and plant regeneration from cultured anthers of Digitalis obscura L. were determined. The presence of auxins was essential for cell proliferation and morphogenesis, and incubation in darkness singificantlyincreased these responses. Callus proliferation usually preceded embryo development, although sometimes direct embryogenesis was observed. On the other hand, bud differentiation was achieved only when callus was transferred to media containing cytokinin or several auxin/cytokinin combinations. Summary We analyse the fruiting phase of the life cycleof Digitalis thapsi L. We studied the production and size of seeds and capsules, and the interpopulational variability and inter-relationships of these parameters. The capsule volume, number of seeds per capsule, of seeds per individual, and of capsules per individual were inter-related and fairly affected by the medium, whereas seed weight was found to bea more independent character. A germination study showed that the species has agreat germinative capacity and has positive photoblasty. It is non-dormant especially if incubated under experimental conditions close to the climatic conditions of the site of provenance of the samples. An increase in temperature speeds up the process without increasing germinability. The morphological and biometric characteristics of the first stages of development of the species following germination were analysed. It was found that seed weight is not positively correlated with cotyledon length nor with any of the other parameters con-sidered, and that the length of the fifth leaf is a useful parameter to express inter-populational genetic variability. Seedling production in spring seems to be influenced more by physical than by chemical factors. 相似文献

Regeneration and development of shoots from sunflower cotyledons were improved by optimizing explant age, plant growth regulator
concentrations, and duration of exposure to plant growth regulators during shoot initiation, development and rooting. Shoot
initiation required only a brief exposure to auxin and cytokinin, and minimizing the duration of exposure to high levels of
plant growth regulators improved shoot development. Rooting was improved in terms of both the number of shoots that rooted
and the time required for rooting by incorporating activated charcoal in the lower layer of a 2-layer rooting medium.

Numerous white surface proliferations appeared in cultures of Ceratonia siliqua L. grown three to four weeks on medium containing 0.5 mg l-1 BA and 0.1 mg l-1 IBA. It was histologically confirmed that these proliferations were hypertrophied lenticels. Proliferations appeared first at the basal shoot internode and gradually spread acropetally, covering eventually the whole shoot except the uppermost internodes. Increase of BA concentration in the medium increased both the number of hypertrophied lenticels per shoot and the shoot multiplication index.Abbreviations BA

Adventitious shoot regeneration from root, hypocotyl, cotyledon and primary leaf explants of safflower (Carthamus tinctorius L.) was studied. Shoot regeneration was promoted by benzyladenine (BA) + naphthaleneacetic acid (NAA), BA + indole-3-butyric
acid (IBA), kinetin + NAA and thidiazuron (TDZ) + NAA incorporated in Murashige and Skoog (MS) basal medium. High frequency
of shoot regeneration and high number of shoots per regenerating explant were obtained on a wide range of TDZ + NAA combinations.
Proliferated shoots were elongated in MS + 0.5 mg dm−3 kinetin and well-developed shoots were rooted in half strength MS + 0.5 mg dm−3 NAA. Rooted shoots were successfully acclimatized and established in soil. 相似文献

High frequency of shoot multiplication and bulblet formation of garlic in liquid cultures 总被引:4,自引:0,他引:4
In vitro shoot proliferation and bulblet production of garlic (Allium sativum L.) was studied in liquid cultures. Shoots grown in vitro were used as explants and were cultured in MS medium supplemented with 2% (w/v) sucrose and 0.5 mg l–1 2-iP. Three culture methods (semi-solid, liquid-immersion and raft) were compared for shoot proliferation. Explants in liquid (immersion) culture exhibited an increased multiplication rate and fresh weight of shoots after 3 weeks of culture as compared with the other treatments. Bulblet formation and growth were studied in liquid medium with different concentrations of sucrose (2–13%). MS medium containing 11% (w/v) sucrose was optimal for bulblet development and bulblets developed in this medium within 9 weeks in culture. The highest multiplication rate was (135 bulblets/explant) found when explants were cultured in bulbing medium (MS medium containing 0.1 mg l–1 NAA+11% (w/v) sucrose) supplemented with 10 M JA. Growth retardants CCC, B-9, ABA also promoted induction and growth of bulblets. Darkness promoted the bulblet induction and growth compared to light conditions (16-h photoperiod of 50 mol m–2 s–1). The dormancy of bulblets was broken by cold treatment at 4 °C for 8 weeks. 相似文献

Calli from hypocotyl and root explants of Digitalis obscura L. showed regeneration of adventitious shoots, roots and embryos when transferred to Murashige & Skoog medium supplemented with cytokinins alone or in combination with auxins. Optimum shoot-bud formation was achieved in the presence of IAA and BA, while roots mainly appeared either in absence of growth regulators or with IAA and Kn. Embryo formation took place only in those combinations that included Kn. Embryo development was influenced by the type of auxin, and precocious germination occurred in media with NAA. Mechanically isolated cells from hypocotyl- and root-derived calli were plated in MS medium supplemented with several IAA and BA combinations. Single cells were able to proliferate forming callus within 20–30 days in culture. In order to induce organogenesis, calli were transferred to various regeneration media. Shoot-bud differentiation efficiency depended on both callus origin and medium initially used for cell culture, best results being obtained in calli grown from hypocotyl-derived cells cultured in the presence of casein hydrolysate. A further subculture to medium containing coconut milk and lower concentrations of NH4NO3 and sucrose promoted shoot development. Rooting was readily achieved upon transferring shoots onto half-strength MS medium. Plantlets were ultimately established in soil.Abbreviations BA

Primordial shoot explants excised from buds of one Larix decidua tree, about 30 years old, produced more adventitious buds, elongating into shoots, when grown on half strength Litvay medium than when grown on other basal media. Thidiazuron and N6-benzyladenine (BA) were equally effective in adventitious bud induction. In a comparative study of 30-year-old L. decidua, L. leptolepis, L. eurolepis, and L. laricina trees, explants from L. eurolepis and L. decidua produced a high number of cultures with adventitious buds that elongated into shoots; those from L. leptolepis were less productive, and those from L. laricina failed to form adventitious buds. The highest response was obtained with material collected in August and September, and in March and April; the lowest response occurred in explants from the October collection. 相似文献

Summary A protocol for micropropagation of Virginia-type peanut plants, an ancient crop of the New World, is reported. This study
was conducted to explore the effect of silver nitrate (AgNO3), alone or in combination with growth regulators, on multiple shoot formation from shoot tip culture. Incorporation of AgNO3 into the medium, without growth regulators, induced regeneration of the explants (which did not develop at all in the AgNO3-free medium), and stimulated the emergence of axillary shoots. When AgNO3 was added in combination with cytokinins and α-naphthaleneacetic acid (NAA), maximum average shoot number per regenerating
explant was recorded (6.3) in Murashige and Skoog (MS) medium containing 33 μM 6-benzyladenine, 5.3 μM NAA, and 23.54 μM AgNO3. Moreover, AgNO3 showed a positive and marked effect on both shoot elongation and the reduction of callus proliferation from the basal ends
of shoot tips. Following a period of elongation, the shoots were rooted in hormone-free Ms medium, showing no residual effects
due to the long-term culture in AgNO3-containing media. Acclimatization was easily obtained after plantlets were transferred to pots under greenhouse conditions,

In contrast to undifferentiated cell suspension cultures of Digitalis lanata, photomixotrophic shoot cultures of Digitalis purpurea accumulate cardiac glycosides in substantial concentrations. They are used to investigate enzymes of the cardenolide pathway. All cardenolides are 5-configurated. The progesterone 5-reductase and the 3-hydroxysteroid-5-oxidoreductase are present in shoot cultures but not in undifferentiated cell cultures. These enzymes provide precursors for cardenolides, whereas the presence of the progesterone 5-reductase, also present in shoot cultures, is discussed with regard to its role in phytosterol biosynthesis and may be attributed to the general steroid pathway. The progesterone 5-reductase had an activity maximum during the early growth period seven days after onset of cultivation, whereas the corresponding progesterone 5-reductase activity was highest on day 11. The maximum cardenolide accumulation was after 24 days. The enzyme activities present in crude extracts from shoot cultures were characterized with regard to their requirements for NADPH and NADH, pH-optimum, temperature optimum, affinity to their substrates and their localization in the cell. Uniform leaf pieces of egg-plant, Solanum melongena L., were cultured in Murashige and Skoog's medium containing 2 mg l-1 kinetin and varying sugar levels. Glucose or fructose at 44 mM was optimal in inducing shoot regeneration compared to sucrose. Sucrose at 11 and 22 mM induced more shoot organogenesis than at lower or higher levels. An additional 22 mM mannitol with 22 mM sucrose enhanced shoot regeneration significantly more than 22 mM sucrose alone. The dual role of sugar as carbon and osmotic source in shoot regeneration from leaf explants of egg-plant is discussed. 相似文献

In order to establish and optimize an in vitro micropropagation protocol of Venus fly trap (Dionaea muscipula Ellis), a carnivorous plant, the effects of medium type, MS medium concentration, pH, and cytokinin and auxin types on shoot proliferation and root formation were investigated using 3-month-old shoots. The shoot proliferation was most effective in 2.3 M kinetin-supplemented 1/3MS medium at pH 5.5. The best conditions for rooting were 1/3MS medium supplemented with 0.5 M IBA. All subcultured shoots produced extensive root systems after 5–6 weeks culture. When plantlets after rooting were planted in plastic pots filled with 1:1 peat moss and sand, the survival rate of plantlets was almost 100%, exhibiting normal development. With subculture every 8 weeks, hundreds of the plants were propagated from a single plant within a year. 相似文献

The influences of nitrogen sources, culture temperature and activated charcoal supplements were studied in relation to the rooting ability of V. faba cuttings. The interaction of these factors led to quantitative and qualitative modifications of the culture responses. Low temperatures (14–18°C) were suitable for in vitro culture, limiting the formation of phenolics in plant material and making activated charcoal supplement unnecessary. Nitrogen supplements contributed in modifying the different plant responses, in accordance with temperature. Multiple shoot formation was obtained from the cotyledonary node and from the stem nodes cultivated in the presence of 6-benzylaminopurine (BAP). BAP at 4 mg l-1 was the most effective concentration in promoting high rates of shoot development. The original position of stem nodes was found to determine the explant response to plant growth regulator treatments, possibly due to the effect of residual apical dominance. 相似文献

Artemisinin, an anti-malarial drug isolated from the annual wormwood Artemisia annua L., has a marked activity against chloroquine-resistant and chloroquine-sensitive strains of Plasmodium falciparum, and is useful in treatment of cerebral malaria. Shoot cultures of Artemisia annua L. were established on Murashige and Skoog basal medium which contained (per litre) 30 g sucrose, 0.5 mg 6-benzyladenine and 0.05 mg naphthaleneacetic acid. Using an optimized combination of sucrose (30 g/l), nitrate (45 mM), inorganic phosphate (200 mg/l), gibberellic acid (7 mg/l) and the ratio of NH4
+-N to NO3
–-N of 1:3, artemisinin production reached 26.7 mg/l after 30 days. This procedure provides a potential alternative for production of artemisinin from in vitro tissue cultures. 相似文献
