Hydrocortisone (cortisol) increased the binding of thyrotropin-releasing hormone (TRH) to specific membrane receptors in 4 clonal strains of rat pituitary cells. At the highest effective cortisol concentration (3–5 × 10?6 M), the increase was observed within 6–8 hr and became maximal (140 to 160% of control binding) by 18–24 hr. Half-maximum stimulation occurred in serum-containing medium at 9 × 10?8 M cortisol, and a significant increase in TRH binding was seen at 3 × 10?8 M. Equilibrium binding studies showed that enhanced TRH binding was explained by an increase in receptor number with no change in affinity. Similar effects were seen with Dexamethasone, but no increase in TRH binding was noted when testosterone, methyltestosterone, progesterone, estradiol or the antiestrogen Lilly 88571 were added to the culture medium. Cortisol treatment did not cause the appearance of specific TRH binding sites in cell strains previously shown to lack receptors for the tripeptide (F4C1, GH12C1 and R5 cells). When added cortisol was removed from medium, receptor number decayed to control values with a T12 of about 30 hr. Previous studies have shown that TRH receptors in GH-cells can be down-modulated by TRH and thyroid hormones; the present findings demonstrate that glucocorticoid hormones can increase the number of TRH receptors in GH-cells.  相似文献

Aurintricarboxylic acid (ATA) was used as a low-molecular-weight chemical probe for the analysis of nuclear binding site(s) of avian progesterone receptor. The cytosol from the oviducts of white Leghorn hens or diethylstilbestrol-primed immature chicks was fractionated with (NH4)2SO4, redissolved and labeled with 3H-labeled R5020 for 2 h at 4 °C. An incubation with 0.1–0.5 mm ATA at 4 °C completely blocked the uptake of progesterone-receptor complex by isolated hen oviduct nuclei. The steroid binding properties of the receptor and the sedimentation characteristics of the progesterone-receptor complex appeared to remain intact under these conditions. The commercially available as well as laboratory-synthesized ATA has been known to be a mixture of several compounds. Eight different components of ATA were separated on silica gel (Kontes) by thin-layer chromatography using a solvent system containing chloroform, formic acid, and acetone (100/5/15, vv). The components were named according to their migration spot on the silica plates, 8 being the slowest and 1 being the fastest. Components 4–8 blocked the nuclear uptake by 40–70% at 0.1 mm whereas at higher concentration (1.0 mm) Components 7–8 completely inactivated the progesterone-receptor complex; Components 5–6 also blocked the nuclear uptake significantly. The effects of ATA appeared to be irreversible; dialysis of ATA-treated preparations, or an addition of divalent cations, BSA, or ATP did not remove its inhibitory influence. Further analysis on several compounds structurally related to ATA revealed that the unique aurintriphenylmethane ring system and the carboxylic acid groups are both necessary for its inhibitory effects on progesterone receptor.  相似文献

Two studies were conducted to synchronize breeding in cattle using PGF2α and LHRH/FSHRH analogs. In the first study, 60 mature lactating Angus cows were assigned at random to 4 treatment groups: saline and saline (SS); 30 mg PGF2α tham salt + saline (PS); saline + 2 mg D-ala6-des-GlyNH210 LHRH/FSHRH ethylamide (D-ala6) (SA); 30 mg PGF2α tham salt + 2 mg D-ala6 (PA). The first letter of the two-letter code for each group always indicates a dual injection at an 11-day interval. PGF2α or saline was administered intramuscular (im) twice at an 11 day interval. D-ala6 or saline was administered 48 hr after PGF2α treatment. In the SA group, the D-ala6 was administered at first signs of estrus, and cows were then artificially inseminated (AI) 24 hr later. All cows in the PS group were inseminated 72 hr after the second PGF2α injection. In the SS group, cows were inseminated 24 hr after first signs of estrus. An additional 6 mature lactating Angus cows were added equally to the PS and PA groups to evaluate changes in serum LH. The percent calf crops was: SS = 40% (frsol|6/15); PS = 47% (715); SA = 47% (715); PA = 53% (815). In the second study, 51 mature lactating Angus cows and 39 Holstein heifers were assigned at random to 3 treatment groups: saline + saline (SS); 33.4 mg PGF2α tham salt + saline (PS); 33.4 mg PGF2α tham salt + 1 mg D-leu6-des GlyNH210 LHRH/FSHRH ethylamide (D-leu6) (PL). PGF2α tham salt or saline was administered im twice at an 11 day interval. D-leu6 or saline was administered 68 hr following the second PGF2α treatment. Cows pretreated with PGF2α were inseminated 80 hr after the second PGF2α injection. In the SS group, cows were administered saline at the time of natural estrus and were artificially inseminated 12 hr later. Calving percent to the first AI was SS = 70% (2130); PS = 53% (1630) and PL = 40% (1230). An additional 10 mature lactating Angus cows were used to evaluate changes in serum LH. Five of the cows were assigned to the PS treatment and five to the PL treatment. Sequential blood samples were collected to monitor serum LH levels. Using the Chi-square test, there were no significant differences between calving percentages of the control and PGF2α treated cows in either study. These results indicate that the PGF2α treatments were successful in timed artificial insemination of cows without detection of estrus. The LHRH/FSHRH analogs did not improve the conception even though they appear to induce a pituitary release of LH simultaneously in all cows within 1 hr of treatment.  相似文献

This report presents an analysis of histone gene expression in the cleaving embryo of the sea urchin, Strongylocentrotus purpuratus, with emphasis on whether the regulatory site(s) in the pathway of gene expression change as development proceeds. The analysis focuses on the equation, dP1dt = M·f·n·At, where dP1dt = the absolute rate of histone synthesis; M = the mole quantity of histone messenger RNA; f = the fraction of histone mRNA in polysomes; n = the polysome size; and At = the rate of elongation of nascent histone polypeptide chains. The embryo solves this rate equation differently at different times. Measurements were made (at 15°C) of absolute rates of histone synthesis (dP1dt). The rate of histone synthesis increases at least 48-fold during the first 6 hr after fertilization from less than 0.5 to 24 pg embryo?1 hr?1; in the period from 6 to 12 hr, this rate rises to 182 pg embryo?1 hr?1, an additional 7.7-fold rise, resulting in an overall increase of 370-fold between the 1-cell and 200-cell stage. The fraction of newly synthesized (zygotic) histone messenger RNA that partitions into polysomes (fzygotic) has also been measured during the first 12 hr of development. This fraction increases from 0.2 in the 2-hr embryo to 0.8 in the 6-hr embryo (16-cell stage), increasing slowly thereafter to near unity by 12 hr. The size of histone-synthesizing polysomes (n) does not change substantially over the 12-hr interval, remaining constant at a weighted mean of 5 ribosomes per polysome (range 3 to 7). Utilizing the data on fzygotic and dP1dt, the rate of elongation of nascent histone polypeptide chains (At) during the first 6 hr of development was estimated; At remains constant at 1.11 codons per second. This calculated value is in fair agreement with a direct measurement of histone peptide elongation rate in the 12-hr embryo. It is proposed that histone gene expression in cleaving sea urchin embryos be divided into two phases, distinguished on the basis of their pivotal translational parameters: Phase I (0–6 hr), during which f is rate determining, and Phase II (6 hr on), during which M is the rate-determining parameter.  相似文献

Radioautographs of rabbit retinas fixed immediately after a 1 or 2 min exposure in vitro to 3H leucine revealed high rates of protein synthesis in receptor cell inner segments, perikarya of ganglion cells, and cells of the inner nuclear layer. If these brieflly labelled retinas were returned to unlabelled medium for periods of up to 6 hr, the radioautographs revealed a progressive dispersion of the labelled proteins from their sites of synthesis. This was largely completed by 112 hr and appeared, in one instance at least, to involve processes other than simple diffusion. Superimposed on the dispersive phenomenon was a process of concentration of the newly formed proteins at two sites quite distant from their synthesis, that was apparent after 112hr. One of these sites was the receptor cell outer segments, as has been previously described, the other was the outer plexiform layer.  相似文献

In vivo receptor binding was examined using 3H-spiperone and 3H-pimozide for dopamine receptors and 3H-LSD for serotonin receptors. Two strategies for improving total: nonspecific binding ratios were tested. The first was to deplete endogenous ligands by various pharmacological treatments prior to 3H-ligand administration in an attempt to increase specific receptor binding; the second was to perfuse the brain with ice-cold saline after 3H-ligand administration in an attempt to reduce nonspecific binding. Alteration of dopamine and serotonin by administering d-amphetamine, reserpine, alpha-methyl-paratyrosine or parachlorophenylalanine did not significantly elevate striatal: cerebellar or cortical: cerebellar (measures of total: nonspecific) bonding ratios. However, perfusion with ice-cold saline significantly improved the ratios for both dopamine and serotonin receptors. Thus, cold saline perfusion may be of value in reducing blank values in autoradiographic and other studies requiring invivo labelling of receptors.  相似文献

Experiments were designed to determine whether or not various antiestrogens in direct competition with estradiol-17β (E2) would inhibit the translocation of the estrogen receptor complex from the cytoplasm to nuclei in rat uterine tissue. Incubation of the antiestrogens CI-628, cis-clomiphene, U-11,100A and MER-25 with rat uteri caused the nuclear uptake of the antiestrogen receptor complex which was greatest for most antiestrogens at concentrations of 1 × 10?6 to 1 × 10?5M. At higher concentrations of CI-628, cis-clomiphene, and U-11,100A the nuclear binding of the antiestrogen receptor complex was greatly decreased. Incubation of the antiestrogens with E2 resulted in a dramatic inhibition of the nuclear uptake of the estrogen receptor. Trans-clomiphene, a weak estrogen, did not inhibit the movement of the uterine cytoplasmic receptor into the nuclear fraction.  相似文献

In homogenates of Macacamulatta (Rhesus) or Cebusapella amygdaloid nuclear complex, adenylate cyclase activity was approximately doubled by either 10μM dopamine or 8mM NaF. In the presence of morphine, the stimulation by dopamine was reduced. A 90–100% inhibition of the dopamine stimulation was obtained with 20μM, and a 50% inhibition, with 5μM morphine. The effects of 10μM morphine on dopamine stimulation were reversed by 10μM naloxone. Morphine itself did not significantly affect the basal adenylate cyclase activity, but in the presence of 10μM morphine the stimulation by 8mM NaF was reduced approxiamtely 50%. The data suggest an action of morphine at a receptor site which is distinct from the dopamine receptor, but which inhibits the dopamine-stimulated adenylate cyclase. In addition, the cyclic GMP content of Cebus amygdala slices was reduced by 50–75% during incubation for 5–20 minutes with morphine. Maximum effects on cyclic GMP were obtained with 10μM, and half-maximum effects, with 0.1μM morphine. The effect of morphine on amygdala cyclic GMP was not reversed by naloxone. Thus, this action of morphine may not be receptor mediated, or may involve the interaction of morphine with receptors other than the opiate receptor.  相似文献

The in vivo stimulation by thyrotropin of the synthesis of poly(A)-RNA and non-poly(A)-RNA in thyroid tissue was studied in 18 day old male rats. Each rat was injected with 0.25 ml of saline or of thyrotropin (0.25 unit) 4 hr or 8 hr before killing. Rats were injected with 3H-uridine 2 to 4 hr before sampling of thyroid tissue. Poly(A)-RNA and non-poly(A)-RNA were isolated by oligo (dT)-cellulose chromatography. Poly(A)-RNA accounts for about 3.5% of total cellular RNA; the specific activity of labeled poly(A)-RNA was 4–7 fold greater than that of non-poly(A)-RNA. A stimulation of about 40% and 90% over the control values was observed in the incorporation of 3H-uridine into poly(A)-RNA and non-poly(A)-RNA, respectively, in thyroid 4 to 8 hr after hormonal injection. The RNA contents of thyroid from hormone-treated rats did not change during the same time period. The stimulation of synthesis of poly(A)-RNA and non-poly(A)-RNA in thyroid was tissue specific insofar as these phenomena were not seen in liver or brain tissues.  相似文献

A single injection of β-naphthoflavone dispersed in corn oil causes significant changes in rabbit liver polysome and polysomal poly(A+)mRNA driven in vitro protein synthesis. The changes occur between 6–18 hr and 30–36 hr after the injection. Our data indicate that the first effect is due to the β-naphthoflavone and the second effect is due to the oil vehicle. In vitro translation of rabbit liver polysomes obtained from treated rabbits followed by specific immunoprecipition and gel electrophoresis, showed that maximal levels of translatable cytochrome P-450 LM4 occurred 18–24 hr after β-naphthoflavone treatment.  相似文献

Ovariectomized guinea pigs were given estradiol benzoate (EB) followed 40 hr later by progesterone (P). Behavioral testing commenced 1 hr after P injection and continued at hourly intervals for 8 hr. This treatment activated lordosis in almost 100% of animals. Administration of the antiestrogen MER-25 (75 mg/kg body wt per injection) between 2 hr before and 6 hr after EB treatment did not cause a significant decline in proportion of animals displaying lordosis, but did cause a decrease in length of time the lordosis position was held (maximum lordosis, sec). In contrast, 1314 animals given MER-25 at 2 hr before and 2 hr after P and 810 animals given MER-25 simultaneously with and 2 hr after P, failed to show lordosis. Administration of supplementary EB at around the time of P injection, partially alleviated these behavior-blocking effects of MER-25. When MER-25 was given 2–6 hr after administration of P there was a significant decrease in duration of heat (hr). These results suggest that in addition to its early “triggering” effects, estrogen has important “maintenance” effects which determine the character of heat in guinea pigs. Continued presence of estrogen in the nervous system may be a requirement for the facilitatory actions of P on sexual behavior in guinea pigs, but such a requirement may not exist in other rodents such as rats.  相似文献

A heterogeneous particulate fraction of mouse brain homogenates binds NRDC 157 (3-phenoxybenzyl [1R,cis]-3-(2,2-dibromovinyl)-2,2- dimethylcyclopropanecarboxylate), a potent pyrethroid insecticide, stereospecifically and with high affinity. Stereospecific binding is a minor component of total binding (2.8%); the remainder of observed binding is predominantly nonspecific and unsaturable. Stereospecific binding is half-saturated at 4×10?8M and fully saturated at concentrations in excess of 1×10?7M. The stereospecific binding capacity of this preparation was 200–250 pmoles of NRDC 157 per gram equivalent of brain tissue (2.3–2.8 pmol/mg protein). This binding site may represent the neural receptor involved in the stereospecific toxic action of pyrethroids.  相似文献

The presence or absence of calcium ions during the isolation of nuclei from SV40-infected African green monkey kidney cells significantly affects the size of SV40 DNA synthesized in vitro. Twelve mature chronically-ovariectomized Brahman cows were randomly assigned to receive three of six different dosages of estradiol-17b (E2) at three different time periods such that at the termination of the trial six animals received each E2 dosage. The E2 was suspended in 0, 1, 2.5, 5, 10 and 20 milligrams. A two week period was maintained between injections. The cows were bled via coccygeal vessel puncture immediately before E2 injection, every 2 hr from 0 to 18 hr, every hr from 18 to 42 hr and every 2 hr from 42 to 48 hr postinjection. Blood was processed to yield serum and stored at ?20 Celsius. Serum luteinizing hormone (LH) was quantitated by validated radioimmunoassay. An LH surge was defined as a sustained elevation of LH at least two standard deviations above the level of LH prior to the rise and was observed in 06, 36, 56, 56, 56, and 66 cows administered 0, 1, 2.5, 5, 10, and 20 mg of E2, respectively. All animals injected with E2 responded with a significant initial decrease (independent of E2 dosage) in LH that persisted from 2 through 12 hr post E2 injection. No significant decrease in LH levels was recorded in animals injected with the corn oil vehicle. The time to the LH surge differed (P<.05) between 1 mg E2 (10 hr) vs 20 mg E2 (19.5 hr), 1 mg E2 vs 10 mg E2 (16.2 hr), and 2.5 mg E2 (12.4 hr) vs 20 mg estradiol-17β. Luteinizing hormone concentrations at the onset of the surge did not differ (P>.10) between E2 dosages. The elapsed time from E2 injection to the peak LH value differed (P<.05) between 1 mg E2 (20.3 hr) vs 20 mg E2 (26.8 hr), and 2.5 mg E2 (21.2 hr) vs 20 mg estradiol-17β. The peak LH value, the area under the LH curve and the duration of the LH surge did not differ (P>.10) with E2 dosage. The time to the end of the LH surge differed (P<.05) between 1 mg E2 (25.3 hr) vs 2.5 mg E2 (31.6 hr), 1 mg E2 vs 5 mg (34.4 hr), 1 mg E2 vs 10 mg E2 (34.8 hr), 1 mg E2 vs 20 mg E2 (37.3 hr), and 2.5 mg E2 vs 20 mg estradiol-17β. Luteinizing hormone values at the termination of the surge did not differ (P>.10) between dosages nor did the LH values at the termination of the surge differ (P>.10) from LH concentrations observed at the onset of the LH surge.  相似文献

Hepatocytes of 14-day-old rats have no detectable glucokinase activity invivo, but it was induced by insulin (10?8M) in primary cultures of these hepatocytes. The glucokinase induced by insulin was separated by electrophoresis on a cellulose acetate membrane and identified by its low affinity for glucose. This precocious induction of glucokinase was completely prevented by the presence of either actinomycin D or cycloheximide. Glucagon also inhibited its induction by insulin. Dexamethasone and testosterone, which alone had no inductive effect, strongly enhanced the induction by insulin. When hepatocytes of 14-day-old rats were cultured with 10?7M insulin, 10?6M dexamethasone and 10?7M testosterone for 48 hr, their glucokinase activity increased to the non-induced level in hepatocytes of adult rats. Estrogen, thyroxine or growth hormone did not induce glucokinase precociously. Testosterone did not enhance induction of glucokinase by insulin in cultured hepatocytes of adult rats.  相似文献

The effects of Cloprostenol administration on porcine luteal lipid and arachidonic acid accumulation were examined in relation to luteal in vitro progesterone and prostaglandin F synthesis in 18 mature gilts at day 12 of the estrous cycle. Basal and net in vitro release of progesterone from luteal tissue was depressed at 8 hr after treatment whereas net in vitro release of prostaglandin F was elevated at 8 hr. Inclusion of copper dithiothreitol or reduced glutathione in the incubation media resulted in minor alterations of in vitro release of progesterone and prostaglandin F and no changes in composition of luteal lipids or fatty acids. Luteal contents of triglyceride had increased by 8 hr after treatment whereas contents of free and esterified cholesterols had increased by 32 hr after Cloprostenol administration. Luteal contents of phospholipid and free fatty acids were not affected by Cloprostenol administration. At 32 hr after treatment, percentages and content of arachidonic acid had increased in luteal cholesterol esters and triglycerides. Although arachidonic acid percentages increased in luteal free fatty acids and phospholipids, calculated arachidonic acid contents did not change following Cloprostenol administration. Induced luteal regression was associated with decreased in vitro progesterone release, increased in vitro prostaglandin F release, and accelerated lipid and arachidonic acid accumulation within the corpus luteum. The effects of altered lipid metabolism on release of prostaglandin F could not be defined. However, availability of arachidonic acid did not appear to be rate-limiting in relation to luteal in vitro prostaglandin F synthesis.  相似文献

The time at which dosage compensation of an X-linked gene in the mouse is established has been estimated by measuring the activity levels of two glycolytic enzymes, phosphoglycerate kinase (EC 2.7.2.3) and triosephosphate isomerase (EC 5.3.1.1), during early development of embryos from XX and XO mice. During preimplantation development the level of phosphoglycerate kinase in embryos from XX mice was constant for the first 48 hr of development (2.55–2.71 nmoles/hr/embryo) and then dropped to one-half the earlier level (1.44 nmoles/hr/embryo) by 72 hr of development. The general developmental profile of phosphoglycerate kinase was similar in embryos from XO mice; however, the absolute level of enzyme activity was reduced to approximately 1.4 nmoles/hr/embryo during the first 48 hr of development and to 0.9 nmoles/hr/embryo by 72 hr of development. These differences in phosphoglycerate kinase levels between embryos from XX and XO mice disappeared between the blastocyst and egg cylinder stages (150 hr postplug) during which time the activity levels had increased to 183 and 193 nmoles/hr/egg cylinder for embryos from XO and XX mice, respectively.The activity level for triosephosphate isomerase in embryos from XX and XO mothers did not differ at any stage of development; this suggests that the gene coding for triosephosphate isomerase is autosomal. In addition the level of activity remained constant during preimplantation development (approximately 3 nmoles/hr/embryo) and then, like phosphoglycerate kinase, increased 100-fold between the blastocyst and egg cylinder stages.The frequency distribution of the activity ratio (triosephosphate isomerase to phosphoglycerate kinase) in the egg cylinder of individual embryos from both XX and XO mothers was determined in order to detect differences among XX, XO and XY embryos. These frequency distributions were unimodal, and hence these results coupled with the gene dosage differences observed during preimplantation development indicate that dosage compensation for an X-linked gene has been established in the 6-day mouse embryo.  相似文献

In the presence of the Na+-channel blocker amiloride, the short-circuit current across the skins of bullfrog tadpoles in metamorphic stages XIX–XXIV was subjected to fluctuation analysis. The resulting power spectra contained a Lorentzian component of which the plateau value (S0) decreased while the corner frequency (fc) increased as the mucosal amiloride concentration was increased from 0.5 to 24 μM. From the linear relationship between the fc values and the amiloride concentrations it was possible to determine the binding (k′01) and unbinding (k10) constants for amiloride to its receptor on the Na+ channel. With these parameters as well as short-circuit current and S0 values, the current through the individual Na+ channels (i) was calculated (average 0.58 pA). It did not increase significantly during late metamorphosis. The density of Na+ channels (M) in the apical membrane, on the other hand, increased significantly. It would appear that the increase in short-circuit current which occurs at this time is due primarily to an increase in amiloride-blockable Na+ channels. Unexpectedly, a Lorentzian component could be fitted to power spectra in amiloride-treated skins (stages XIX–XXI) which showed no amiloride-sensitive short-circuit current. Moreover, the typical increase in fc with the amiloride concentration did not occur in these animals.  相似文献

Rat serum is 2–5 fold more active than human serum in cleaving the three hypothalamic releasing hormones, luteinizing hormone releasing hormone (LH-RH), thyrotropin releasing hormone (TRH), and somatostatin. LH-RH was degraded by two distinct enzymatic mechanisms; 1) endopeptidase cleavage, 2) C-terminal cleavage. The C-terminal cleaving enzyme was active in rat serum but present only in trace levels in human. These mechanisms were substantiated by the use of suitably substituted analogs; D-Ala at position 6 of LH-RH prevented cleavage at the -Tyr5-D-Ala6-Leu7-site and the presence of ethylamide (C2H5NH2) at position 10 inhibited significantly the action of the second enzyme. These analogs have an enhanced biological activity invivo which correlates well with their decreased rate of degradation. Somatostatin was degraded by endopeptidase cleavage at one or more sites. D-Trip in position 8 blocked cleavage of the -Trp8-Lys9-bond, reducing significantly the rate of degradation. This also correlates well with the enhanced biological potency of the (D-Trp8)-somatostatin analog. TRH was degraded by cleavage of the pyroGlu-His and His-Pro.NH2 bonds with the release of free His and Pro. The analog (3-Me-His2)-TRH was degraded by a similar mechanism with the release of 3-Me-His.  相似文献
