Man is exposed to epoxides of fatty acids from a number of sources, yet their degradative metabolism is not well understood. In mouse liver the 100,000 g supernatant or the cytosolic fraction is the most active fraction in hydrating cis- and trans-epoxymethyl stearates with the oxirane ring opening in a trans manner to give the corresponding threo and erythro diols, respectively. Hydration was also observed in the microsomal, nuclei and cell debris, and mitochondrial fractions in decreasing order of specific activity.  相似文献

The chemical reaction between (±)-styrene oxide and N-acetylcysteine produces both positional isomers (1 and 2) as a mixture of diastereoisomers with a preference for the benzylic thioether isomer 1 (2 : 1). Synthesis of the mercapturic acid conjugates from either (+)- or (?)-styrene oxide produces only two of the four possible stereoisomers. The single diastereoisomers of 1 and 2 were separated by high pressure liquid chromatography (HPLC) and identified by 1H- and 13C-nuclear magnetic resonance (NMR). The relative stereochemistry at the benzylic carbon center of the mercapturic acid conjugates was assigned on the basis of the established chemical correlation between optically pure styrene oxide and its precursor mandelic acid, and considerations on the mechanism of ring opening of epoxides by sulfur nucleophiles. The stereochemical definition of the isomers 36 should prove useful in investigations of the biotransformation of the glutathione (GSH) conjugates of styrene oxide.  相似文献

The hydrolysis of (±)-trans-3-bromo-1,2-epoxycyclohexane in the presence of rabbit liver microsomes was investigated, and found to yield, beside c-3-bromocyclohexane-r-1,t-2-diol, 2,3-epoxycyclohexanol. It was demonstrated that the latter compound was the only product of the enzymatic reaction, whereas the diol resulted from a non enzymatic hydration in the reaction medium. These data provide the first direct proof for a general base catalysis in the enzymatic epoxide hydration, previously hypothesized on the basis of several lines of indirect evidence, and disprove alternative mechanisms involving protonation of the oxirane oxygen.  相似文献

The metabolism of the prostaglandin F analogues, 15-methyl-Δ4-cis-PGF and 16,16-dimethyl-Δ4-cis-PGF, has been investigated in the cynomolgus monkey and the human female. The two analogues, tritium labelled in the 9β-position, were administered by intramuscular injections into the monkeys and by subcutaneous injections into the human. Excretion of tritium labelled products were followed in urine (in both species) and feces (in monkeys only) and several metabolites were identified by GC/MS. The analogues were found to be resistant to the 15-hydroxy dehydrogenase and furthermore the degradation by β-oxidation was delayed. About 13% of the given dose of 15-methyl-Δ4-cis-PGF was excreted unchanged into urine and feces from the monkey. The corresponding figure for 16,16-dimethyl-Δ4-cis-PGF was about 20%. In addition, a large part of the metabolites had the carbon skeleton intact and were only metabolized by ω-oxidation. The relative resistance to degradation of these two analogues is likely to be the basis for their prolonged pharmacological activity.  相似文献

Styrene and 1,3-butadiene are important intermediates used extensively in the plastics industry. They are metabolized mainly through cytochrome P450-mediated oxidation to the corresponding epoxides, which are subsequently converted to diols by epoxide hydrolase or through spontaneous hydration. The resulting styrene glycol and 3-butene-1,2-diol have been suggested as biomarkers of exposure to styrene and 1,3-butadiene, respectively. Unfortunately, poor ionization of the diols within electrospray mass spectrometers becomes an obstacle to the detection of the two diols by liquid chromatography/electrospray ionization-mass spectrometry (LC/ESI-MS). We developed an LC/ESI-MS approach to analyze styrene glycol and 3-butene-1,2-diol by means of derivatization with 2-bromopyridine-5-boronic acid (BPBA), which not only dramatically increases the sensitivity of diol detection but also facilitates the identification of the diols. The analytical approach developed was simple, quick, and convincing without the need for complicated chemical derivatization. To evaluate the feasibility of BPBA as a derivatizing reagent of diols, we investigated the impact of diol configuration on the affinity of a selection of diols to BPBA using the established LC/ESI-MS approach. We found that both cis and trans diols can be derivatized by BPBA. In conclusion, BPBA may be used as a general derivatizing reagent for the detection of vicinal diols by LC/MS.  相似文献

NMR titration curves have been recorded for all the 13C resonances of cis and transN-acetyl-dl-proline in 2H2O. the measured pK2H values are 3.4 ± 0.8 and 4.13 ± 0.08 respectively; the free energy of ionization for the trans isomer being (3.8 kJ/mole) greater than for the cis. The ionization shifts of the two isomers differ significantly only at the acetyl carbonyl and Cγ positions. It is suggested that these are related to conformational changes which stabilize the trans form at low p2H.  相似文献

Metabolism of biosynthetic (?)-trans-(3R,4R)-dihydroxy-3,4-dihydrochrysene by liver microsomes from control, phenobarbital-treated and 3-methylcholanthrene-treated rats was investigated. Although previous studies of the metabolism of related benzo[a]pyrene and benzo[e]pyrene dihydrodiols which also prefer the diaxial conformation had indicated that diol epoxides were minor metabolites, the diastereomeric chrysene 3,4-diol-1,2-epoxides-1 and ?2 were major metabolites (66–90%). All three types of microsomes metabolized the chrysene 3,4-dihydrodiol at low but essentially similar rates (0.5–0.7 nmol substrate/nmol cytochrome P-450/min).  相似文献

Cytokinin-autonomous tobacco callus was incubated in defined mineral medium containing 3H-adenine for 60 minutes. Radioactivity was incorporated into the four predominant free cytokinins, ribosyl-trans-zeatin, trans-zeatin, N6-(Δ2-isopentenyl) adenosine and N6-(Δ2-isopentenyl) adenine. The bases were more abundant than their respective ribosides, N6-(Δ2-isopentenyl) adenine being the most abundant cytokinin. No discrete peaks of radioactivity could be detected on the HPLC column eluate corresponding to the elution volumes of cis-zeatin and ribosyl-cis-zeatin.  相似文献

The hydration properties of Escherichia coli lipids (phosphatidylglycerol, phosphatidylethanolamine) and synthetic 1,2-dioleoyl-sn-glycero-3-phosphocholine in H2O/2H2O mixtures (9:1, v/v) were investigated with 2H-NMR. Comparison of the 2H2O spin lattice relaxation time (T1) as a function of the water content revealed a remarkable quantitative similarity of all three lipid-H2O systems. Two distinct hydration regions could be discerned in the T1 relaxation time profile. (1) A minimum of 11–16 water molecules was needed to form a primary hydration shell, characterized by an average relaxation time of T1 ≈ 90 ms. (2) Additional water was found to be in exchange with the primary hydration shell. The exchange process could be described in terms of a two-site exchange model, assuming rapid exchange between bulk water with T1 = 500 ms and hydration water with T1 = 80–120 ms. Analysis of the linewidth and the residual quadrupole splitting (at low water content) confirmed the size of the primary hydration layer. However, each lipid-water system exhibited a somewhat different linewidth behavior, and a detailed molecular interpretation appeared to be preposterous.  相似文献

Both pairs of dl-ll-desoxy- and dl-13-cis-erythro-15, 16-dihydroxyprostaglandins have been synthesized via 1,4-conjugate additions of an appropriately functionalized cis-vinyl cuprate to the requisite cyclopentenone. A number of epoxides, including cis- and trans-stilbene oxides, were assayed as substrates for epoxide hydrolases (EHs) by gas-liquid chromatography. Radiolabeled stilbene oxides were prepared by sodium borotritide reduction of desyl chloride followed by ring closure with base treatment. Rapid radiometric assays for EHs were performed by differential partitioning of the epoxide into dodecane, while the product diol remained in the aqueous phase. Glutathione (GSH) transferase was similarly assayed by partitioning the epoxide and diol, if formed metabolically, into 1-hexanol, while the GSH conjugate was retained in the aqueous phase. The cytosolic EH rapidly hydrates the trans isomer while the cis is very poorly hydrated. In contrast, the cis is a better substrate for the microsomal EH than the trans. GSH transferase utilized both epoxides as substrates, but conjugation is faster with the cis isomer. Cytosolic EH activity is high in mouse but very low in rat and guinea pig. Microsomal EH activity, in contrast, is highest in guinea pig, intermediate in rat, and the lowest in mouse. GSH transferase activity, which is high in all three species, can be inhibited by chalcone, with an I50 of 3.1 × 10?5m. These assays facilitate the rapid evaluation and direct comparison of epoxide-metabolizing systems in cell homogenates used in short-term mutagenicity assays, cell or organ culture, and possibly in vivo.  相似文献

The mechanism of enzymatic epoxide hydration by a cytosolic or 100,000 g soluble mammalian liver enzyme (in contrast to the microsomal enzymes) was examined by monitoring 18O distribution following chemical and enzymatic hydrations of 16O or 18O epoxide labeled (±) 1-(4′-ethylphenoxy)-3, 7-dimethyl-6, 7-epoxyoctane. Acid catalyzed hydration of the 18O epoxide in 16O water, and hydration of the 16O epoxide in 18O water, indicated that attack by water was predominantly on the tertiary carbon (C-7). Enzymatic epoxide hydration led to attack predominantly on secondary carbon (C-6). These data are consistent with water attacking as a nucleophile in the enzymatic reaction.  相似文献

A mutant strain (PL pT 1143) of Pseudomonasputida PL, has been isolated for its inability to growth with p-cymene as carbon source. The mutant oxidizes p-cymene (and p-cumate) to a compound (λmax 293 nm) which is readily converted to 3-hydroxy-p-cumate by acid. 4-Trifluoromethylbenzoate is oxidized by the mutant to an acid-stable intermediate (λmax 277nm) that has been crystallized. The spectral properties (u.v., i.r., NMR and mass) of this metabolite are consistent with those expected for a 2,3-dihydro-2,3-dihydroxy derivative of 4-trifluoromethylbenzoate. Further support of this structure was provided by elemental analysis and the properties of two derivatives of the metabolite, 4-trifluoromethyl-3-hydroxybenzoate and an acetonide formed with 2,2-dimethoxypropane. The stability of a product obtained by treatment of the dihydrodiol metabolite with triacetylosmate indicates that it is the cis-isomer.  相似文献

A cell-free extract prepared from Fusarium solani pisi grown on cutin, catalyzed the hydration of 18-hydroxy-9,10-epoxyoctadecanoic acid to 9,10,18-trihydroxyoctadecanoic acid while extracts from glucose-grown cells contained <6% of this activity. The product was identified by Chromatographic techniques and by radio gas-liquid chromatography of its periodate oxidation products. This epoxide hydrase activity had a pH optimum at 9.0 and it was located mainly in the 100,000g supernatant fraction. Rate of hydration of the epoxy acid was linear up to 15 min and up to a protein concentration of 30 μg/ml. This fungal epoxide hydrase has a molecular weight of 35,000, as determined by Sephadex G-100 gel filtration. It was partially purified by ammonium sulfate fractionation and gel filtration. The apparent Km and V of the enzyme was 2 × 10?4m and 222 nmoles/min/mg, respectively. Parachloromercuribenzoate strongly inhibited the enzyme, while N-ethylmaleimide was a less potent inhibitor. 1,1,1,-Trichloropropylene-2,3-oxide at 10?3m gave 50% inhibition of the hydration of 18-hydroxy-9,10-epoxyoctadecanoic acid. Kinetic analysis showed that trichloropropylene oxide was a competitive inhibitor. 18-Acetoxy-9,10-epox-yoctadecanoic acid, methyl 18-acetoxy-9,10-epoxyoctadecanoate, 9,10-epoxyoctadecanoic acid, and styrene oxide were not readily hydrated by this fungal epoxide hydrase showing that it has a stringent substrate specificity. Analysis of the enzymatic hydration product on boric acid-impregnated silica gel plates showed that the product obtained from the cis epoxide was exclusively erythro while acid hydrolysis of this epoxide gave rise to the expected threo product. This enzyme is novel in that it catalyzes cis hydration of epoxide while the other epoxide hydrases heretofore isolated catalyzed trans hydration of epoxides.  相似文献

A precise continuous photometric assay has been devised and utilized for mechanistic studies of chicken and rat liver microsomal epoxide hydrolase (EH). The assay is based on monitoring the hydration of p-nitrostyrene oxide (PNSO) at 310 nm. Rat liver EH hydrates S-(+)- and R-(?)-PNSO differentially, the Km and V values for the former being ca. four times those for the latter; in contrast, enantiomeric differences are negligible with chicken liver EH. With rat EH V increases slightly from pH 7 to 8 and then falls rapidly from pH 8 to 9.5; Km remains constant from pH 7 to 8 and then increases steadily from pH 8 to 9.5. In 86 mol% D2O the solvent isotope effect on V (H2OD2O) is 1.103 ± 0.015. Both rat and chicken EH show a 3% inverse isotope effect for the hydration of [7-2H]PNSO and a 4% normal isotope effect for the hydration of [8-2H2]PNSO. These observations are discussed in terms of the possible participation of acid as well as base catalysis in the enzymatic mechanism.  相似文献

A heterogeneous particulate fraction of mouse brain homogenates binds NRDC 157 (3-phenoxybenzyl [1R,cis]-3-(2,2-dibromovinyl)-2,2- dimethylcyclopropanecarboxylate), a potent pyrethroid insecticide, stereospecifically and with high affinity. Stereospecific binding is a minor component of total binding (2.8%); the remainder of observed binding is predominantly nonspecific and unsaturable. Stereospecific binding is half-saturated at 4×10?8M and fully saturated at concentrations in excess of 1×10?7M. The stereospecific binding capacity of this preparation was 200–250 pmoles of NRDC 157 per gram equivalent of brain tissue (2.3–2.8 pmol/mg protein). This binding site may represent the neural receptor involved in the stereospecific toxic action of pyrethroids.  相似文献

3-Methylcholanthrene and five related dihydrodiols have been tested for microsome-mediated mutagenicity towards Salmonellatyphimurium TA100 and for the induction of mutation to 8-azaguanine resistance in V79 Chinese hamster cells and malignant transformation in M2 mouse fibroblasts. In both mutagenicity test systems, the 9,10-diol was considerably more active than either the parent hydrocarbon, the related cis-2α,3-diol, the trans-4,5-, the trans-7,8- or the trans-11,12-dihydrodiols. At a non-toxic concentration (1μg/ml medium), the 9,10-diol induced the formation of more transformed malignant foci in cultures of M2 cells than 3-methylcholanthrene and the other diols were either inactive or only weakly active in this test system. The results obtained indicate that the 9,10-dihydrodiol derived from 3-methylcholanthrene is involved, presumably following conversion into the corresponding vicinal diol-epoxide, 9,10-dihydro-9,10-dihydroxy-3-methylcholanthrene 7,8-oxide, in the metabolic activation of this carcinogenic polycyclic hydrocarbon.  相似文献

Behavioral comparisons of the stereoisomers of tetrahydrocannabinols   总被引:1,自引:0,他引:1  
The potencies of (?)-trans9-THC, (+)-trans9-THC, (+)-cis9-THC, (?)-trans8-THC and (+)-trans8-THC were compared in several different species. (?)-trans9-THC was 100 times more potent than (+)-trans9-THC in depressing schedule-controlled responding in monkeys. The (+)-trans isomers were less effective than their corresponding (?)-trans isomers in the dog static-ataxia test, but potency ratios could not be determined due to a lack of dose-responsiveness of the (+)-trans isomers. However, it appeared that their potency differed by at least ten fold. The potency of (+)-cis9-THC in the dog static-ataxia test was comparable to that of (+)-trans9-THC. The hypothermia in mice produced by the (?) isomers of trans9-THC and trans8-THC were 9.1 and 30.4 times greater than that produced by their respective (+)-isomers. Also, the potency ratio of the (+)- and (?)-trans9-THC was 5.6 as measured by depression of spontaneous activity in mice. The magnitude of the potency ratios of the THC stereo-isomers is dependent upon the species and the pharmacological test used.  相似文献

An unstable epoxide, leukotriene A4 (5(S)-trans-5,6-oxido-7,9-trans-11,14-cis-eicosatetraenoic acid), was earlier proposed to be an intermediate in the conversion of arachidonic acid into the slow reacting substance (SRS), leukotriene C4. In the present work synthetic leukotriene A4 was incubated with human leukocytes or murine mastocytoma cells. A lipoxygenase inhibitor, BW755C, was added in order to prevent leukotriene formation from endogenous substrate. Leukotriene C4 and 11-trans-leukotriene C4 were the main products with SRS activity. It was not established whether the 11-trans-compound was formed by isomerization at the leukotriene A4 or C4 stage.  相似文献

The stoichiometric reaction between d-TpGpGpCpCpA (d(T-G-G-C-C-A)) and cis-[Pt(NH3)2(H2O)2](NO3)2 (8.4 × 10?6 to 1.3 × 10?4M in water at pH 5.5–6) gives a single complex. High pressure gel permeation chromatography and pH-dependent 1H NMR analyses of the nonexchangeable base protons, show that it is a platinum chelate with the cis-PtII(NH3)2 moiety bound to the two N7 atoms of the adjacent guanines. A 3 × 10?3M reaction gives the same platinum chelate, via the formation of intermediate complexes, together with unsoluble adducts.  相似文献
