AIMS: Antifungal metabolites of Pseudomonas antimicrobica have previously been shown to inhibit conidial germination of the grey mould pathogen Botrytis cinerea. In this study, metabolites of the bacterium have been tested at different stages of Botrytis germination to determine their effects on germ tube production and extension. METHODS AND RESULTS: Metabolites were added to conidia that had been pre-incubated for either 120 or 255 min. Pseudomonas antimicrobica inhibited B. cinerea conidial germination and caused a significant reduction in germ tube extension, irrespective of the stage of germination. Abnormal germination and a reduction in the frequency of lateral branching of the germ tubes in the presence of the metabolites were also reported, suggesting interference with normal hyphal development. CONCLUSION: The bacterium can inhibit germination of conidia and extension of germ tubes at different stages of Botrytis development. SIGNIFICANCE AND IMPACT OF THE STUDY: The antagonistic activity of the bacterium has promising implications for its use as a biocontrol agent.  相似文献

This paper reports an experimental study of water dispersal potential and germination of the shingle beach plant Mertensia maritima in which we consider the effects of physical factors (cold treatment, mechanical wear of the pericarp and salt-water exposure) on the diaspores. Our approach also includes testing effects of different orders of the treatments, in contrast to most earlier studies of diaspore ecology. A cold period was necessary to break seed dormancy, and prolonged cold treatment (stratification at 2°C) enhanced germination. Mechanical wear of the pericarp before cold treatment did not affect germination, whereas mechanical wear after cold treatment increased germination significantly. Seeds exposed to 6 weeks of cold treatment before floating in salt water for 6 weeks did not germinate. In contrast, for seeds given the same cold treatment after floating, the germination was more than 50%. Most undamaged and slightly damaged nutlets stayed afloat throughout the dispersal experiment (9 weeks) in 3% salt water, whereas seeds that fell out of damaged nutlets sunk immediately. Thus, the results suggest that the potential for long-distance dispersal is high unless the diaspores (nutlets) are severely damaged, but the order of cold treatment and water dispersal seems to be of great importance for germination: seeds dispersed in autumn (before cold periods) have a much higher probability of germinating than seeds dispersed in winter or early spring (after a cold period). Similar effects of the relative timing of physical processes have hitherto only been reported for two other water-dispersed beach plants. Future studies in diaspore ecology should consider such timing effects as they may be important determinants of the distribution and abundance of plants.  相似文献

BACKGROUND AND AIMS: Germination studies of species from fire-prone habitats are often focused on the role that fire plays in breaking dormancy. However, for some plant groups in these habitats, such as the genus Leucopogon (Ericaceae), dormancy of fresh seeds is not broken by fire cues. In the field, these same species display a flush of seedling emergence post-fire. Dormancy and germination mechanisms therefore appear complex and mostly unknown. This study aimed to identify these mechanisms by establishing dormancy class and testing the effects of a set of typical germination cues, including those directly related to fire and entirely independent of fire. METHODS: To classify dormancy, we assessed seed permeability and embryo morphology, and conducted germination experiments at seasonal temperatures in incubators. To test the effects of fire cues on germination, factorial combinations of smoke, heat and dark treatments were applied. Ageing treatments, using burial and seasonal incubation, were also tested. Germination phenology was established. KEY RESULTS: Seeds were dormant at release and had underdeveloped embryos. Primary dormancy of the study species was classified as morphophysiological. Seasonal temperature changes overcame primary dormancy and controlled timing of germination. Fire cues did not break primary dormancy, but there was a trend for smoke to enhance germination once this dormancy was overcome. CONCLUSIONS: Despite the fact that fire is a predominant disturbance and that many species display a flush of emergence post-fire, seasonal temperatures broke the primary physiological dormancy of the study species. It is important to distinguish between fire being responsible for breaking dormancy and solely having a role in enhancing levels of post-fire germination for seeds in which dormancy has been overcome by other factors. Biogeographical evidence suggests that morphological and physiological factors, and therefore seasonal temperatures, are likely to be important in controlling the dormancy and patterns of post-fire germination of many species in fire-prone regions.  相似文献

BACKGROUND AND AIMS: Plant functional trait responses to processes such as grassland management have been analysed frequently; however, the scaling-up from individual traits to the outcomes of vegetation dynamics has seldom been tested. In this experiment, germination success was studied with respect to the relationships between grassland management (mowing and grazing), as well as abandonment, and two traits that are relevant for seedling recruitment: seed mass and germination season. On the basis of discussions in the literature and indirect trait analyses in our previous studies, the following hypotheses are proposed: (1) with respect to seed mass, mowing and grazing favour the germination of small seeds, whereas after abandonment the germination success of larger seeds is higher; and (2) with respect to germination season, mowing and grazing favour autumn-germinating seeds, whereas succession promotes spring-germinating seeds. METHODS: The germination experiment took place in a semi-natural, dry grassland in north-east Germany. Seeds of eight herbaceous species that differ with respect to seed mass and germination season were sown in mown, grazed and abandoned plots. Germination success was documented during the following year. KEY RESULTS AND CONCLUSIONS: Contrary to the hypothesis, germination of small seeds was not promoted by mowing or grazing and they germinated relatively more often than expected in the abandoned plots. A relationship between abandonment and gaps of bare soil below the vegetation cover that favour germination of small seeds was likely, but could not be proved statistically. It is possible that the small seeds suffered less from predation. Mowing favoured autumn germination, which could be explained by the removal of biomass in late summer. Contrary to our expectation, there was relatively more spring germination after grazing than after mowing, yet vegetation height was smallest in spring. Generally, germination season was found to be related to the temporal occurrence of favourable light conditions.  相似文献

Storage oil breakdown plays an important role in the life cycle of many plants by providing the carbon skeletons that support seedling growth immediately following germination. 1-Butanol, a specific inhibitor of phospholipase D (PLD)-dependent production of the signalling molecule phosphatidic acid (PA), inhibited Arabidopsis seed germination. N-Acylethanolamines (NAEs), which have been shown to inhibits PLDα1 activity, have no effect on seed germination. However, mobilization profile of triacylglycerols (TAG) that induced by each compound has not been reported. To gain deeper insights into the mode of mobilization of TAG during NAE 12:0 or 1-butanol treatment, we conducted a detailed comparative analysis of the effect of NAE 12:0, DMSO, 1-butanol and tert-butanol on Arabidopsis seed germination and fatty acid composition, tert-butanol and DMSO served as the corresponding controls treatment respectively. Our data show that 1-butanol, but not the inactive tert-butanol isomer, inhibited Arabidopsis seed germination, which is accompanied by a with retardation of the mobilization of triacylglycerols (TAG). In contrast, NAE 12:0 did not affect mobilization of TAG, nor did it significantly delay seed germination as monitored by radicle and cotyledon emergence. 1-Butanol induced RNA degradation in seeds and seedlings. We speculate that the large-scale degradation of RNA under the induction of 1-butanol may lead to abnormal gene expression in genes necessary for seed germination, including the genes needed for the mobilization of oil bodies, and thus cause a delay of seed germination. To the best of our knowledge, we report for the first time that 1-butanol delays the mobilization of TAG.  相似文献

H(2)O(2) promotes seed germination of cereal plants such as barley, wheat and rice, and several mechanisms have been proposed for its action [Naredo et al. (1998) Seed Sci. Technol. 26: 675-689]. We investigated the role of H(2)O(2) in the germination of Zinnia elegans seeds. H(2)O(2) promoted seed germination in a dose-dependent manner as did respiratory inhibitors, indicating that H(2)O(2) itself possibly promotes seed germination rather than O(2). Seed germination was promoted by removal of pericarp from seeds or by removal of ethanol-soluble compounds from the seeds with pericarp. The ethanol-soluble compounds suppressed the germination of seeds having no pericarp, and this effect was reversed by H(2)O(2). These findings indicate that oxidation of the germination inhibitor(s) present in the pericarp by H(2)O(2) promotes seed germination. Antioxidants which are derivatives of well-known germination inhibitors suppressed seed germination in a dose-dependent manner, suggesting that, to initiate seed germination, a germination inhibitor(s) should be decomposed by an oxidant such as H(2)O(2).  相似文献

Biochemical events which occur during macroconidial germination have been studied in the dermatophyte Microsporum gypseum. The specific activity levels of various metabolic enzymes have been assayed during germination time periods. The accumulated levels of several of these enzymes, as a function of exogenous carbohydrate source, have been investigated. M. gypseum was found to possess a constitutive glyoxalate shunt, a constitutive glucokinase, a fructose phosphoenolpyruvate transferase, and a mannitol phosphoenolpyruvate transferase. The integration of endogenous reserve utilization during germination is discussed. The purification and properties of an alkaline phosphatase and its possible relationship to sporulation and spore germination also are described.  相似文献

Four seed lots of sorghum cultivar IS 34213 were obtained which had been harvested early or at full maturity and then dried either swiftly in the sun or slowly in the shade. The earlier harvested seeds were lighter, smaller and of lower density. Earlier harvested seeds had pericarps which permitted faster water uptake and greater leakage of electrolytes. Water uptake and electrolyte leakage were positively correlated. The method of seed drying had no effect on water uptake or leakage. Greater leakage was associated with lower final germination. The results are interpreted in relation to germination testing.  相似文献

Seed germination is regulated by several environmental factors, such as moisture, oxygen, temperature, light, and nutrients. Light is a critical regulator of seed germination in small-seeded plants, including Arabidopsis and lettuce. Phytochromes, a class of photoreceptors, play a major role in perceiving light to induce seed germination. Classical physiological studies have long suggested the involvement of gibberellin (GA) and abscisic acid (ABA) in the phytochrome-mediated germination response. Recent studies have demonstrated that phytochromes modulate endogenous levels of GA and ABA, as well as GA responsiveness. Several key components that link the perception of light and the modulation of hormone levels and responsiveness have been identified. Complex regulatory loops between light, GA and ABA signaling pathways have been uncovered.  相似文献

Scarification and aseptic germination of Musa balbisiana . Amer. Jour. Bot. 49(5): 515–520. Illus. 1962.—Methods for germinating seeds of Musa balbisiana Colla under aseptic conditions have been developed. Scarification was required for germination under these conditions, and mechanical was superior to chemical scarification. Various methods of mechanical scarification and cultural procedures were developed which facilitated the operation when large numbers of aseptic seedlings were required. Removing a chip from the lateral portion of the seed coat to expose the endosperm was the most effective method of scarification: germination percentages averaged 80%, and the time required for germination in sterile culture was shortened from the 3–6 weeks required for intact seeds in soil to 6–10 days. However, scarification did not shorten the time required for germination in soil, and seeds treated with some methods of mechanical scarification failed to germinate, as a result of their decomposition by microorganisms. The effectiveness of scarification in causing germination in aseptic culture is not presently understood, but, as the excised embryo exhibits no dormancy, the factors delaying germination apparently reside in other portions of the seed.  相似文献

ABA antagonists have potential application as growth regulators to improve germination and seedling growth at low temperatures for oilseeds and pulses grown in regions with short seasons such as those in western Canada. Towards development of practical ABA antagonists, a series of 3′-substituted ABA analogs were synthesized and screened in seed germination assays in canola (Brassica napus), lentil (Lens culinaris), and soybean (Glycine max) at low temperature and in overcoming exogenous ABA. The most promising analog, ABA 1009, was selected for further germination testing of dose responses in canola, lentil, and soybean. Analog ABA 1009 at 100 µM was effective in overcoming ABA (10 µM)-induced inhibition for canola, lentil, and soybean germination at ambient temperature, and also promoted germination at low temperature for canola (5 °C).

Developmental mutants affected in either sporulation or spore germination have been isolated from Microsporum gypseum with the aid of nitrosoguanidine or as spontaneously occurring mutants. The time course levels of several proteins temporally associated with conidial development have been assayed in the wild-type and mutant strains. The spore germination characteristics of two of the mutants are described. The relationship of alkaline protease accumulation to tyrosinase accumulation and spore germination is discussed.  相似文献

Effect of distillery effluent on seed germination in some vegetable crops   总被引:1,自引:0,他引:1  
A laboratory experiment was conducted to study the effect of different concentrations (0%, 5%, 10%, 15%, 20%, 25%, 50%, 75% and 100%) of distillery effluent (raw spent wash) on seed germination (%), speed of germination, peak value and germination value in some vegetable crops: tomato, chilli, bottle gourd, cucumber and onion. The distillery effluent did not show any inhibitory effect on seed germination at low concentration except in tomato, but in onion the germination was significantly higher (84%) at 10% concentration as against 63% in the control. Irrespective of the crop species, at highest concentrations (75% and 100%), complete failure of germination was observed. The speed of germination, peak value and germination value also followed a similar trend. We found that a concentration of 5% was critical for seed germination in tomato and bottle gourd, and 25% in the rest of the crops. Based on the tolerance to distillery effluent, the crops studied have been arranged in the following order: cucumber > chilli > onion > bottle gourd > tomato. We conclude that the effect of the distillery effluent is crop-specific and due care should be taken before using the distillery effluent for pre-sowing irrigation purposes.  相似文献

Experiments with Arabidopsis have been developed for spaceflight studies in the European Space Agency's Biorack module. The Biorack is a multiuser facility that is flown on the United States Space Shuttle and serves as a small laboratory for studying cell and developmental biology in unicells, plants, and small invertebrates. The purpose of our spaceflight research was to investigate the starch-statolith model for gravity perception by studying wild-type (WT) and three starch-deficient mutants of Arabidopsis. Since spaceflight opportunities for biological experimentation are scarce, the extensive ground-based testing described in this paper is needed to ensure the success of a flight project. Therefore, the specific aims of our ground-based research were: (1) to modify the internal configuration of the flight hardware, which originally was designed for large lentil seeds, to accommodate small Arabidopsis seeds; (2) to maximize seed germination in the hardware; and (3) to develop favorable conditions in flight hardware for the growth and gravitropism of seedlings. The hardware has been modified, and growth conditions for Arabidopsis have been optimized. These experiments were successfully flown on two Space Shuttle missions in 1997.  相似文献

Aims Light requirements for cactus seed germination have been considered to be associated with their adult plant height and seed mass, but this has not been thoroughly studied for other succulent species. In order to understand seed photosensitivity from desert species belonging to Asparagaceae (subfamily Agavoideae) and Cactaceae, we performed a germination experiment with and without light for 12 species and 2 varieties from 1 species from the Southern Chihuahuan Desert. We also determined if adult growth is totally determined by seedling 'growth form' in cacti.Methods We performed a germination experiment using light and darkness for 13 species from Southern Chihuahuan Desert: 10 rosette species (Asparagaceae), as well as 1 globose, 1 columnar and 2 varieties from 1 depressed-globose species (Cactaceae). The response variables were seed germination percentage and relative light germination (RLG). In addition, in order to determine if adult-globose cacti could have cylindrical seedlings, we calculated the shape index (height/width ratio) for Coryphanta clavata and Mammillaria compressa .Important findings All species were considered neutral photoblastic. Eleven species had similar seed germination in both light and dark conditions, and three taxa (M. compressa and the two varieties of Ferocactus latispinus) showed higher germination with light than without it. Agave salmiana, M. compressa and the two varieties of F. latispinus had higher RLG than the other species. Seed mass was an important factor because with higher seed mass there was lower dependence to light. These findings support the hypothesis that small seed mass and light requirements have coevolved as an adaptation to ensure germination. One adult-globose cactus species, M. compressa, and one adult-columnar species, C. clavata, had small seeds and neutral fotoblasticism. Seedlings from these two species exposed to light were cylindrical and those under darkness conditions were columnar. Perhaps seeds from this species are able to germinate in the dark because they produce columnar seedlings with the ability to emerge from greater soil depths where sunlight cannot penetrate.  相似文献

Polypeptides synthesized in dissected embryos of Zea mays at different stages of embryogenesis and early germination have been characterized by their migration in two-dimensional gel electrophoresis. This analysis has been carried out with in vivo labeled polypeptides from excised embryos and with proteins synthesized in vitro in the rabbit reticulocyte system directed by poly(A+) RNA isolated from the different developmental stages. We have identified three main sets of expressed polypeptides: (a) embryonic set: this group of polypeptides is synthesized in young and mature embryos but not in early germination; (b) maturation set: this group of polypeptides is not present in young embryos and appears during the maturation period. Some of these polypeptides are still present in early germination while others disappear from stored mRNAs in dry embryos. One particular group from this set can be induced prematurely in young embryos by incubation with abscisic acid; and (c) germination set: this group of polypeptides is not expressed in the maturation period and appears after brief imbibition of the dry embryos.  相似文献

Aims: To determine germination triggers of Clostridium frigidicarnis, an important spoilage bacterium of chilled vacuum‐packed meat. Methods and Results: Germination of Cl. frigidicarnis spores in the presence of a range of potential nutrient and non‐nutrient germinants was tested by monitoring the fall in optical density and by phase‐contrast microscopy. The amino acid l ‐valine induced strong germination when paired with l ‐lactate in sodium phosphate under anaerobic conditions. Several other amino acids promoted germination when paired with l ‐lactate in sodium phosphate and the co‐germinants NaHCO3 and l ‐cysteine. Heat activation, while not necessary for germination, increased the rate of germination. Spore germination was not observed when spores were incubated aerobically. Conclusions: Spores of psychrotolerant Cl. frigidicarnis germinated in the presence of l ‐valine in combination with l ‐lactate in sodium phosphate buffer under anaerobic conditions. Significance and Impact of the Study: Anaerobic conditions, l ‐valine and l ‐lactate, have been identified as triggering germination in Cl. frigidicarnis, and are all present in packs of fresh, vacuum‐packaged, red meat. This new information adds to what is known about red meat spoilage by cold tolerant clostridia and can be used to develop intervention strategies to prevent meat spoilage.  相似文献
