rRNA-targeted寡核苷酸探针识别活性污泥微生物群落结构与功能研究进展 总被引:2,自引:0,他引:2
活性污泥中微生物群落内部关系非常复杂 ,及时对活性污泥中优势菌群和群落内部关系进行监测是污水处理中采取正确措施的关键。历史研究表明传统培养方法经常导致活性污泥优势菌群检测的失败 ,而r RNA- targeted寡核苷酸探针作为一种快速原位监测活性污泥微生物群落结构和功能的新工具被引入 ,使我们对参与污水净化的微生物群落结构和优势菌群能有较全面的了解。就该方法在识别除磷污泥、脱氮污泥、污泥泡沫和膨胀污泥中微生物群落结构和功能的典型应用进行综述 ,分析了该方法存在的优点和缺点 ,并对目前已建立且应用于活性污泥微生物检测的 r RNA- targeted寡核苷酸探针进行了详细总结 相似文献

目的探讨地高辛标记寡核苷酸基因探针应用于微生态研究的可行性和实用性。方法制备双歧杆菌属和部分种的地高辛标记16S rRNA寡核苷酸探针,初步应用于微生态制剂鉴定和临床肠道微生态检测,评价寡核苷酸探针杂交在肠道微生态研究和检测中的应用价值。结果地高辛标记寡核苷酸探针具有较好的特异性与灵敏度:地高辛标记的双歧杆菌属和种的共6种寡核苷酸基因探针与标准菌株杂交后灵敏度和特异度分别为属探针95%、75%,青春双歧87.5%、90%,两歧双歧87.5%、87.5%,短双歧87.5%、92.5%,婴儿双歧75%、95%,长双歧75%、100%。结论寡核苷酸基因探针用于肠道细菌的鉴定显示出一定前景,加大探针的种类与扩大调查范围有可能使该技术替代现有细菌培养技术。 相似文献

总RNA和mRNA来源的探针与cDNA芯片杂交的差异研究 总被引:1,自引:0,他引:1
提取BEP2D细胞的总RNA并按两种方式进行cDNA芯片探针的标记,一种是将100μg BEP2D细胞的总RNA利用逆转录法直接标记成荧光探针,另一种是先从100μg BEP2D细胞的总RNA中分离出mRNA,然后再标记成荧光探针。将两份标记好的探针同时与含有230个基因的cDNA芯片杂交。杂交后的芯片经Axon4100B扫描仪扫描,发现两种方式标记探针的一致性为93.04%,并且mRNA来源探针杂交后的荧光信号值较总RNA的弱。探讨了这两种方法标记探针在基因芯片表达谱研究中的差异性,目的是为利用这两种方法标记探针进行基因表达谱研究提供一些依据。 相似文献

以水貂生长激素(GH)基因作为控制水貂皮张长度性状主基因的候选基因,以大兴安岭水貂养殖基地养殖的水貂群为试验材料,通过PCR-SSCP方法对GH基因进行多态性检测。在该基因内含子1中发现1处碱基突变:C→A,并检测到3种基因型(AA、AB、BB),BB基因型个体与AA基因型个体皮张长度有一定的差异(P0.05)。在外显子2中发现2处碱基突变:T→A、C→G,并由此检测到了3种基因型,分别命名CC、CD、DD,但3种基因型对水貂皮长的影响没有显著的差异(P0.05)。统计各基因型之间的组合给水貂皮长带来的影响时,发现多数组合基因型对所检测的水貂皮长有显著影响(P0.05)。 相似文献

目的:探讨臂踝脉搏波传导速度(brachio—anklepulsewavevelocity,baPWV)与冠状动脉粥样硬化狭窄程度的相关性。方法:选择198例行冠脉造影的患者为研究对象,根据冠状动脉管腔狭窄程度分为非冠心病组(对照组,50例)和冠心病组(148例);并将冠-22病组分为单支病变组(48例)、双支病变组(56例)及多支病变组(44例),所有患者均采用动脉硬化检测仪检测baPWV,观察各组baPWV值及Gensini积分的变化,并对其行相关性分析。结果:CAD组多支病变、双支病变、单支病变亚组及对照组冠状动脉Gensini积分分别为45.06±7.27、30.83±4.50、24.83±4.94和13.75±1.74,baPWV值分别为2.13±0.32、1.68±0.14、1.12±0.13和0.93±0.17m/s,组间差异具有统计学意义(P〈0.05)。冠心病组baPWV值与冠状动脉Gensini积分呈正相关。结论:baPWV值与冠状动脉粥样硬化的病变范围密切相关,可作为诊断冠心病和预测其病变严重程度的辅助指标。 相似文献

The rate of lateral spread of clonal plants is considered a plastic trait that responds directly to environmental conditions. However, this response is likely constrained by evolutionary history and other species attributes. Here we ask how the relationships between lateral spread and the distribution of herbaceous plant species in different environments are influenced by the type of spacer (epigeogenous versus hypogeogenous rhizome), its persistence (integrator versus splitter), and the height and phylogeny of the plant. Analysis of spacer length of 367 species from temperate to arctic Europe revealed that other plant characteristics modulate the relationship between spacer length and environmental conditions. While plants with epigeogenous rhizomes, which have usually shorter spacers than hypogeogenous rhizomes, were associated with illuminated habitats, plants with hypogeogenous rhizomes were associated with warm habitats. These relationships were also shown within the specific rhizome type, as within the group of species with epigeogenous rhizomes, those having short spacers were associated with more light. The trend toward long spacers in warm environment was detected within both epigeogenous and hypogeogenous rhizome bearing species. Splitters were found to be associated with wet, nutrient rich habitats irrespective of rhizome type. When plant height was accounted for by using the ratio of spacer length to plant height (L:H) instead of absolute spacer length, no relationship with environmental variables was found until phylogenetic relationships among the species were taken into account. This implies that overall variability in L:H ratio in the dataset masked trends common for different taxonomic groups. Future comparative studies relating particular clonal growth forms or individual traits to environmental conditions should consider the constraints arising from other growth characteristics and evolutionary history of the subject species. 相似文献

A genetic map of rye, Secale cereale L., chromosome 1R covering 247 cM was constructed utilizing 27 RFLP and four C-band markers, including terminal C-bands. Genetic mapping of C-bands and the centromere, and in situ hybridization of three RFLP clones, allowed for the integration of the genetic and cytological maps. Eight contact points between the genetic and cytological maps revealed variation in the recombination distance to cytological distance ratio ranging between 0.25 and 1.95, a 7.8-fold difference. Recombination was found to be highest in the satellite region of 1RS and lowest in the most distal region of 1RL. 相似文献

In Palaearctic birds, tail length was more variable than wing and tarsus, but not bill lengths. Tails of the ornamental shapes pin, lyre and graduated varied more in length than did non-ornamental shapes. Log coefficient of variation (C V) tail length showed an overall U-shaped relationship with longtailedness, but although the CV for most tail shapes increased in short-tailed species, only in ornamental shapes was C V also high in long-tailed species. C V of fork depth was lowest at a fork depth of 2, and considerably higher in shallow forked tails. CV streamer lengths were similar to CV deep fork depths. The more deeply forked tails thus seem ornamental. Phylogenetically independent contrasts confirmed in males, but not females, that long-tailed species had greater CV than medium-tailed species, and the greater CV of graduated than square tails, but the CV of short- and medium-tailed species did not differ. These comparisons, however, did not control for tail shape. The greater elongation and CV of tails with ornamental shapes are consistent with an influence of sexual/signal selection on these tails, and the increase in CV with longtailedness suggests that Weber's law applies to the perceptual threshold for tail length. Sexual selection may have a widespread, but moderate, influence on tail traits in birds. 相似文献

Twenty-five biotically pollinated plants of the Chaco Serrano Forest (Córdoba, Argentina) were studied in order to analyze whether ‘flower functionality’ is related to the relationship between pollen size and pistil length. Because flower functionality may act on the respective mean values of pollen size and pistil length rather than on intraspecific variation in these traits, we expected (1) a high positive correlation between pollen size and pistil length in a set of sympatric species, independent of their degree of pollination specialization or generalization; and (2) no interspecific correlation between the coefficients of variation (CVs) of those traits. On the other hand, on the assumption that pollinators are influencing the variation in floral traits (e.g. in pistil length) we expected lower mean phenotypic variation of pollen size and pistil length in pollination-specialist plants than in pollination-generalist ones. A positive correlation between pollen size and pistil length was found for the set of species, but not between the CVs of these traits. This trend was maintained when pollination-specialist plants were analyzed separately, but no statistical significance was obtained for the correlation in pollination-generalist plants. Contrary to our expectations, pollination-specialist plants did not show less mean intraspecific variation in floral traits than pollination-generalist plants. Therefore, the relationship between pollen size and pistil length among species suggests that the pollination system may be of less importance as a selective force than flower functionality. 相似文献

The pulse rates of atmospherics at 10 and 28 kHz have been compared with the variations of outside air temperature up to a height of 7 km. In the 10 kHz band the pulse rate showed a strong statistically significant correlation with air temperature for all heights up to 7 km, the maximum correlation coefficient beingr=0.78 at a level of significanceP0.001. In the 28 kHz band the correlation is weaker: up to a height of 3 km the correlation coefficient ranges from 0.17 to 0.3 (P 0.001 and 0.02, respectively) and above 3 kmr ranges from 0.15 to 0.17 (P 0.02 and 0.05, respectively).As the outdoor absolute humidity depends mainly on the air temperature, it is now clear why previous investigations showed strong correlations between the difference (S) of the atmospherics pulse rates at 10 and 28 kHz, the absolute humidity (AH) outdoors and the diffusion time (T

Oligonucleotide chip-based assays can be a sample-thrifty, time-saving, routine tool for evaluation of chemical-induced DNA strand breaks. This article describes a novel approach using an oligonucleotide chip to determine photosensitizer-induced DNA single-strand breaks. Surface coverage of fluorophore-labeled oligonucleotides on silicon dioxide chip surfaces was determined on alkaline phosphatase digestion. Fluorescence maxima (at 520 nm) of the solutions were converted to molar concentrations of the fluorescein-modified oligonucleotide by interpolation from a predetermined standard linear calibration curve. The photosensitizing activity of chlorpromazine and triflupromazine toward DNA single-strand breaks was then studied at different drug doses and also as a function of photoirradiation time. Photoinduced single-strand breaks calculated using the method described here agreed with values predicted by theoretical extrapolation of the single-strand breaks obtained for plasmid DNAs from agarose gel electrophoresis, and thereby indirectly validated the chip-based assays. Under UV irradiation (93.6 kJ/m2) chlorpromazine (0.08 mM) was found to have significant photogenotoxicity. However, triflupromazine did not exhibit any (photo)genotoxicity over the concentration range studied (0.04–0.20 mM). The method developed will be useful for quantitative screening of drug genotoxicity in terms of induction of breaks in DNA. 相似文献

Summary DNA restriction endonuclease fragment analysis is used to examine the genetic organization, inheritance and linkage associations of the ribosomal DNA in pea. The substantial variation observed in the length of the intergenic spacer region is shown to segregate in Mendelian fashion involving two independent genetic loci, designated Rrn1 and Rrn2. Linkage between Rrn1 and two marker loci on chromosome 4 establishes the approximate location of this tandem array. Rrn2 shows linkage with a set of isozyme loci which assort independently of other markers on all seven chromosomes. Combining these observations with previous cytological data, we suggest that Rrn2 and the isozyme loci linked to it constitute a new linkage group on chromosome 7. The general absence of spacer length classes common to both rRNA loci in any of the lines we examined indicates that little or no genetic exchange occurs between the nonhomologous nucleolar organizer regions. 相似文献

A statistical reanalysis was performed on the data fecently reported on a 6-laboratory, collaborative cytogenetic study to measure and minimize interlaboratory variation. Three of the laboratories had mean values significantly different from the others on most of the 6 indexes of chemically-induced aberration; one laboratory with values higher and two with values lower. Furthermore, relative variability of the values around the means was consistently lower in one of the 6 participating laborabories. The results of the reanalysis of this collaborative study demonstrates that significant interlaboratory differences exist and that these should be adjusted or diminished before rat cytogenetic analysis can be an effective test system for evaluation of a compound for mutagenic potential. 相似文献
