Anther culture response of accessions of several hexaploid Avena species was studied with respect to requirement for auxin. The highest callus induction frequency was achieved withA. sativa L. cv. Stout (17.3%). The three most responsive genotypes, two of A. sativa and one of A. nuda L., had two-fold higher anther culture responses on growth regulator -free medium than on a medium containing 2,4-d. The A. sterilis L. accession CAV 2648 was the only genotype which consistently produced white, embryogenic structures (on solid MS+ 10% sucrose) and did so irrespective of the presence of 2,4-d. When transferred onto medium with lower sucrose concentration and an auxin transport inhibitor (TIBA), three green (two diploid [2t n=6t x=42] and one haploid [1t n=3t x=21]) and two albino plantlets were regenerated. The diploid regenerants set seed in the greenhouse. This first report of plants recovered from anther culture in the wild oat A. sterilis may provide an avenue to understand better and possibly overcome problems associated with androgenesis in cultivated oat. Morphological characteristics were studied in cytoplasmic male sterile (CMS) cybrids possessing the tobacco nuclear genome, Hyoscyamus niger plastome and recombinant mitochondria. After backcrosses with tobacco, new flower modifications were found, including: conversions of stamens into branched filamentous structures; alterations in the shape of petals and the corolla limb; and high degrees of reduction in most flower organs. Vegetative alterations (leaf elongation and stem branching) occurred in some cybrids. Results confirmed that a protoplast fusion-based alloplasmic cytoplasm transfer, followed by conventional backcrosses, is a useful tool for generating alternative CMS sources with novel nucleo-cytoplasmic compositions. These alterations in the genetic status were accompanied by modified floral and vegetative phenotypes.  相似文献

Somatic embryogenesis and regeneration of plantlets was achieved In callus cultures derived from cotyledonary leaf pieces of Hyoscyamus muticus L on MS medium enriched with 2 mg/l 2,4–0 and 0.5 mg/l BAP. For embryogenesis and organogenesis varying concentrations of NAA with or without BAP were added In the medium. Somaclone production in Antirrhinum majus plants by regeneration of plants from callus cultures has been achieved using three types of explant tissue. Regeneration from mature stem internode-derived callus was extremely poor. Callus derived from seedling shoot tips could be induced to form new shoots in six of seven cultivars tested. Regeneration was achieved in all seven cultivars when callus was produced from segments of hypocotyl and was most effective using agar-solidified medium containing 0.25 mgl-1 naphthoxyacetic acid + 10% coconut milk. In this case, five of the cultivars produced shoots directly, one produced leaves from the petioles of which new shoots emerged, and one regenerated plants chiefly through the production of embryoids.  相似文献

A genetic and physiological study of biennialism in the diploid selfer Hyoscyamus niger (black henbane), an obligate long-day plant, is described. Three annual and two biennial accessions that were homozygous for their respective growth habits were selected. The early-flowering trait of two annual accessions was dominant over the late-flowering trait of the third annual accession. The late-flowering annual accession, but not the early-flowering ones, responded to vernalization. Two biennial accessions remained vegetative after more than 1 year in soil and thus had an obligate vernalization requirement. Crosses between annual and biennial accessions showed that biennialism was conferred through a single dominant gene. However, plants containing only one copy of this dominant gene were transformed from biennials into very late-flowering winter-annual plants that responded more rapidly to vernalization than biennials. Taken together, these results indicated that there were allelic differences in photoperiod-specific flowering time genes and that biennialism was a dose-dependent trait with incomplete dominance. Models for flowering time regulation in henbane involving photoperiod-, vernalization-, and most likely gibberellin-specific pathways are discussed.  相似文献

Root and embryo derived callus tissues of rice grown on sucrose alone as carbon source lost their ability to organise shoots by 75 and 100 days in culture respectively. Along with 2% sucrose, either 3% sorbitol or 3% mannitol was found to be necessary in the growth medium for the callus to regenerate whole plants over a period of 1400 days without any decline in the shoot forming ability. Our results indicated that incorporation of sorbitol or mannitol in the callus proliferating medium provides long-term totipotent rice cultures with a high frequency (50–60%) of shoot differentiation.  相似文献

Tests for calluses rich in tropane alkaloids were made with newly induced calluses of Atropa belladonna, Datura stramonium and Hyoscyamus niger. Only calluses of H. niger gave an alkaloid-positive test.A Hyoscyamus cell line had the highest total alkaloid content of all the calluses screened by the cell-squash alkaloid assay. Investigations were undertaken to achieve rapid multiplication and improvement of Origanum vulgare (a herbaceous, ornamental plant well known for its aromatic and medicinal value) through plant regeneration from callus. The explants (cotyledons, hypocotyl and root segments) excised from 15 d old aseptic seedlings were cultured on Gamborg's B5 medium supplemented with 2,4-D, NAA and BAP individually and in various combinations (at concentrations of 0,10–7,10–6 and 10–5 M). Best callus induction was noted on medium with 10–7 M 2,4-D alone. The cotyledonary expiants proved to be the best source for compact and nodulated callus. The subcultured cotyledonary calli showed shoot induction when transferred onto media supplemented with BAP alone orin combination with 10–7M or 10–6MNAA. However, 10–5M NAA completely suppressed the shoot inducing ability of BAP. In general, NAA promoted root induction from all explants used including cotyledonary callus. Best shoot induction was obtained on medium supplemented with 10–6M BAP+10–6MNAA. Both IBA and NAA at 10–6 M proved to be equally effective in induction of roots from the cut ends of 15–20 mm long shoots (excised from callus) in half-strength B5 liquid medium. Summary Normal root of Hyoscyamus niger was cultivated in various nutrient conditions to optimize root growth and scopolamine production. Nitrate or phosphate concentration had little effect on root growth, while the low levels of nitrate or phosphate enhanced specific scopolamine content by 44% and 39%, respectively. Glucose or fructose was not adequate for the root culture as a carbon source, while the root growth was markedly enhanced at high sucrose concentration (up to 9%). The growth rate (0.59g dw./L.d.) and scopolamine productivity (1.98 mg/L.d.) was increased greater in roots with 9% initial sucrose concentration as compared to 3% sucrose (0.28 g dw./L.d., 0.98 mg/L.d.)  相似文献

Callus and cell suspension cultures were established from Hyoscyamusniger seedlings and their growth requirements investigated.Stock suspensions were more aggregated in the earlier passagesbut became finer as the cultures were serially sub-cultured.On transfer to auxin-free medium, the cultures formed rootsand shoots were rarely noted. Callus initiated from differentparts of the seedlings showed no differences in alkaloid levels.Alkaloid contents were slightly increased on transfer to auxin-freemedium. Summary Two macromutants [unbranched (Ub) and restricted branched (Rb)], one point mutation [yellow flowerd (Ada)] and two flowering variants (early and late) were isolated in the M2 generation of gammairradiated progenies of black henbane (H. niger) — a medicinal plant belonging to family Solanaceae. These major gene changes were coupled with simultaneous changes in several polygenic systems conditioning continuous variation for flowering time, plant height, numbers of shoots and leaves, biomass and crude drug content. Such quantitative variation in qualitative mutants were measured for both per se means and cross-bred performance in terms of: (1) shifts in univariate means, (2) changes in pooled divergence, (3) alterations in gca effects, and (4) changes in heterotic capacity. The mutants were significantly divergent from the parental base for all the six metric traits, both individually as well as collectively. Divergence among mutants was wider (D2 = 1491 to 11 469) than that for mutants versus source parent (D2 = 117 to 7120). Conspicuous alterations in both the nature and the magnitude of the gca effects associated with all mutants, except L-2013 (late flowering), relative to the parental base were observed for all of the metric traits examined (gca - per se means). Similarly, the mutants manifested greater heterotic potential than the parental base. This is even more strongly reflected in mutant x mutant hybrids for all of the characters. The amount of economic heterosis for diverse traits followed the pattern: crude drug > biomass > flowering time - plant height > number of shoots - number of leaves.  相似文献

Rapid cycling Brassica species have great potential in plant genetic research because of their short life cycles and their minimal space requirements. Rapid cycling B. oleracea can be grown with up to six generations per year. Protoplast culture of this genotype can be applied for gene transfer by direct DNA uptake and by protoplast fusion. We here report on fast regeneration of flowering plants from protoplasts of rapid cycling B. oleracea. Regeneration frequencies of 27–65% were achieved with multiple shoots developing from individual calli. The regenerated plants were grown to maturity, and flowering and other morphological characteristics were monitored. The regenerants flowered within a similar time frame as plants grown from seeds. The ploidy level of regenerated and seed-grown plants was measured by flow cytometry. Many (20–45%) of the regenerants were tetraploid. A daylength extension with incandescent light is more effective in promoting flowering of long-day plants like Hyoscyamus niger than fluorescent light. A low phytochrome photoequilibrium (Pfr/Ptot), attained by a far-red irradiation at the close of long days under fluorescent light, also promotes flowering. Moreover, if flower initiation processes are initiated by several long days, a low phytochrome photoequilibrium at the end of short, postinduction photoperiods also enhances flowering. The initiation phase of flowering requires Pfr to be present whereas the development phase proceeds more rapidly in the absence of Pfr. Spectral dependence studies, therefore, could be misinterpreted if the initiation and development stages are combined into a single audit of flowering.  相似文献

The effect of genotype and ploidy on RuBP carboxylase (EC 4.1.1.39) activity, chlorophyll content, leaf area, chloroplast ultrastructure and not photosynthesis among monoploid. diploid and tetraploid anther-derived plants of Solanum phureja Juz, and Buk. was studied. Within the monoploid group, RuBP carboxylase activity and concentration displayed a significant genotypic effect. For the diploids, variation among genotypes was significant for total protein content and maximum specific activity of RuBP carboxylase, and among the tetraploids for net photosynthesis and specific leaf weight. Ploidy effect was evident regarding net photosynthesis, leaf area and chlorophyll content. The different ploidy groups among the anther-derived plants surpassed the anther donor plant for all characteristics except maximum activity of RuBP carboxylase and net photosynthesis. For the latter only the tetra-ploid group was superior to the anther source plant. However, a monoploid genotype with an increase of 9% in maximum activity of RuBP carboxylase over the anther-donor plant was identified. Segregation of trails rind differential gene expression together with possible mutations during androgenesis are discussed as sources of variation.  相似文献

Hyoscyamine 6 beta-hydroxylase, a 2-oxoglutarate-dependent dioxygenase that catalyzes the hydroxylation of l-hyoscyamine to 6 beta-hydroxyhyoscyamine in the biosynthetic pathway leading to scopolamine [Hashimoto, T. & Yamada, Y. (1986) Plant Physiol. 81, 619-625] was purified 310-fold from root cultures of Hyoscyamus niger L. The enzyme has an average Mr of 41,000 as determined by gel filtration on Superose 12 and exhibited maximum activity at pH 7.8 l-Hyoscyamine and 2-oxoglutarate are required for the enzyme activity, with respective Km values of 35 microM and 43 microM. Fe2+, catalase and a reductant such as ascorbate significantly activated the enzyme. 2-Oxoglutarate was not replaced by any of ten other oxo acids tested, nor was Fe2+ by nine other divalent cations tested. The enzyme was inhibited moderately by EDTA, Tiron and various oxo acids and aliphatic dicarboxylic acids, and strongly by nitroblue tetrazolium and divalent cations Mn2+, Co2+, Ni2+, Cu2+, Zn2+, Cd2+ and Hg2+. Several pyridine dicarboxylates and o-dihydroxyphenyl derivatives inhibited the hydroxylase. Pyridine 2,4-dicarboxylate and 3,4-dihydroxybenzoate are competitive inhibitors with respect to 2-oxoglutarate with the respective Ki values of 9 microM and 90 microM. Several alkaloids with structures similar to l-hyoscyamine were hydroxylated by the enzyme at the C-6 position of the tropane moiety. The enzyme preparation also epoxidized 6,7-dehydrohyoscyamine, a hypothetical precursor of scopolamine, to scopolamine (Km 10 microM). This epoxidation reaction required the same co-factors as the hydroxylation reaction and the epoxidase activities were found in the same fractions with the hydroxylase activities during purification. Two possible pathways for scopolamine biosynthesis are discussed in the light of the hydroxylase and epoxidase activities found in the partially purified preparation of hyoscyamine 6 beta-hydroxylase.  相似文献

Summary Somatic hybrid plants have been regenerated following polyethylene glycol mediated fusion of leaf mesophyll protoplasts from tomato and protoplasts from Lycopersicon pennellii callus. Three different cultivars of tomato were used as sources of protoplasts: Early Girl, Manapal, and UC82B. Fusions were performed between protoplasts of these tomato cultivars and protoplasts of L. pennellii, and between protoplasts of the cultivars and protoplasts of L. pennellii that had been exposed to 3 or 6 krads of gamma radiation. Somatic hybrid plants were identified on the basis of heterozygous isozyme banding patterns, and leaf and flower morphology. Somatic hybrid plants were regenerated following fusion of tomato protoplasts with either untreated or irradiated L. pennellii protoplasts. All were heterozygous for isozyme loci on five different chromosomes. Regenerated somatic hybrids showed inheritance of either or both parental chloroplast genomes, but predominantly the L. pennellii mitochondrial genome. The regenerated somatic hybrid plants exhibited reduced fertility, less than 20% viable pollen. A total of 34 somatic hybrid calli were identified. Of these, 21 regenerated shoots, and 7 produced seed following manual pollinations.  相似文献

Coronilla varia L. (crownvetch) plants were regenerated from callus cultures through somatic embryogenesis. Callus cultures were initiated using hypocotyls excised from sterile seedlings. Cultures were then transferred from a modified Gamborg's B5 medium containing 2,4-D to a medium containing no plant growth regulators (basal B5). Formation of embryos was evident in 12 of 32 callus lines after transfer of callus to BOi2Y (modified Blayde medium supplemented with 100 mg inositol and 2 g yeast extract/L). Basal B5 supplemented with 10 mM asparagine or 20 mM NH4Cl could be substituted for BOi2Y. Embryos subsequently transferred to basal B5 developed roots and shoots.
